Prospective fecal microbiomic biomarkers for chronic wasting disease.

Chronic wasting disease (CWD) is a naturally occurring prion disease in cervids that has been rapidly proliferating in the United States. Here, we investigated a potential link between CWD infection and gut microbiome by analyzing 50 fecal samples obtained from CWD-positive animals of different sexes from various regions in the USA compared to 50 CWD-negative controls using high throughput sequencing of the 16S ribosomal RNA and targeted metabolomics. Our analysis reveals promising trends in the gut microbiota that could potentially be CWD-dependent, including several bacterial taxa at each rank level, as well as taxa pairs, that can differentiate between CWD-negative and CWD-positive deer. Through machine-learning, these taxa and taxa pairs at each rank level could facilitate identification of around 70% of both the CWD-negative and the CWD-positive samples. Our results provide a potential tool for diagnostics and surveillance of CWD in the wild, as well as conceptual advances in our understanding of the disease.IMPORTANCEThis is a comprehensive study that tests the connection between the composition of the gut microbiome in deer in response to chronic wasting disease (CWD). We analyzed 50 fecal samples obtained from CWD-positive animals compared to 50 CWD-negative controls to identify CWD-dependent changes in the gut microbiome, matched with the analysis of fecal metabolites. Our results show promising trends suggesting that fecal microbial composition can directly correspond to CWD disease status. These results point to the microbial composition of the feces as a potential tool for diagnostics and surveillance of CWD in the wild, including non-invasive CWD detection in asymptomatic deer and deer habitats, and enable conceptual advances in our understanding of the disease.

Assessing Different Chronic Wasting Disease Training Aids for Use with Detection Dogs.

Chronic wasting disease (CWD) is a highly infectious, fatal prion disease that affects cervid species. One promising method for CWD surveillance is the use of detection dog-handler teams wherein dogs are trained on the volatile organic compound signature of CWD fecal matter. However, using fecal matter from CWD-positive deer poses a biohazard risk; CWD prions can bind to soil particles and remain infectious in contaminated areas for extended periods of time, and it is very difficult to decontaminate the affected areas. One solution is to use noninfectious training aids that can replicate the odor of fecal matter from CWD-positive and CWD-negative deer and are safe to use in the environment. Trained CWD detection dogs' sensitivity and specificity for different training aid materials (cotton, GetXent tubes, and polydimethylsiloxane, or PDMS) incubated with fecal matter from CWD-positive and CWD-negative deer at two different temperatures (21 °C and 37 °C) for three different lengths of time (6 h, 24 h, and 48 h) were evaluated. Cotton incubated at 21 °C for 24 h was identified as the best aid for CWD based on the dogs' performance and practical needs for training aid creation. Implications for CWD detection training and for training aid selection in general are discussed.

Expanding CWD disease surveillance options using environmental contamination at deer signposts

Abstract Environmental surveillance can allow early detection of diseases, which increases management options and can improve disease trajectories. Chronic wasting disease (CWD) in cervids is a significant prion disease that has been spreading across North America since the 1960s, leading to cervid population declines and concern from hunters and state wildlife agencies. White‐tailed deer have a unique breeding season behaviour called scraping, where they deposit urine and saliva at shared sites. Since both these fluids can contain CWD prions, scrape sites have the potential to serve as sentinel sites for environmental surveillance of CWD. To examine this potential, we used camera traps to monitor deer behaviour and collected environmental samples from 105 scrape sites. The 48 km2 study site was located at the centre of the CWD zone in southwestern Tennessee, where CWD prevalence is ~50%. We also sampled scrapes in northern Mississippi at the leading edge of the same CWD distribution to test the potential for early CWD detection using scrape sampling. From camera data, we identified 218 unique bucks visiting 105 scrapes, with a mean of 12.2 ± 7.5 bucks per scrape (mean ± SD, range 1–39) and individual bucks visiting a mean of 5.9 ± 4.6 monitored scrapes each (range 1–23). Using real‐time quaking‐induced conversion (RT‐QuIC), we detected prion seeding activity in 20% of the soil and 41% of the licking branches of the scrape sites within the CWD study area, and in 25% of the soil and 11% of the licking branches of scrape sites sampled at the edge of the known CWD distribution. Our data show there is environmental prion contamination at scrape sites. This supports the idea that scrapes could serve as early warning sentinel sites for CWD surveillance through testing soil and licking branches for prion seeding activity, especially in areas with limited access to harvested deer samples.

Heterogeneity of pathological prion protein accumulation in the brain of moose (Alces alces) from Norway, Sweden and Finland with chronic wasting disease

Prion diseases are a group of neurodegenerative, transmissible, and fatal disorders that affect several animal species. They are characterized by the conformational conversion of the cellular prion protein (PrPC) into the pathological prion protein (PrPSc). In 2016, chronic wasting disease (CWD) gained great importance at European level due to the first disease detection in a wild reindeer (Rangifer tarandus) in Norway. The subsequent intensive CWD surveillance launched in cervids resulted in the detection of CWD in moose (Alces alces), with 11 cases in Norway, 3 in Finland and 4 in Sweden. These moose cases differ considerably from CWD cases in North American and reindeer in Norway, as PrPSc was detectable in the brain but not in lymphoid tissues. These facts suggest the occurrence of a new type of CWD. Here, we show some immunohistochemical features that are clearly different from CWD cases in North American and Norwegian reindeer. Further, the different types of PrPSc deposits found among moose demonstrate strong variations between the cases, supporting the postulation that these cases could carry multiple strains of CWD.

Hunter Willingness-to-Pay for Disease Testing: Evidence from Chronic Wasting Disease in White-Tailed Deer

AbstractUsing the case of chronic wasting disease (CWD) in white-tailed deer, we estimated hunters’ willingness-to-pay (WTP) for testing their game and determined how their perception of disease risk, trust, and confidence in wildlife agency affected hunter participation in ongoing disease surveillance. The average WTP for testing was $23.75 per deer, and it was positively related to trust and confidence in the wildlife agency and the perception of risk about deer populations in the declining area and the pathogen spreading to other areas. These findings imply that implementing active outreach programs can improve hunter participation in user-paid systems for CWD surveillance.

Impact of landcover composition and density of localized deer culling sites on chronic wasting disease prevalence

Chronic Wasting Disease (CWD), the prion disease of the Cervidae family, has been managed in Illinois deer since it was first detected in the Fall of 2002. Management uses a state-sponsored localized focus culling (LFC) program, implemented as close as possible to previously identified CWD-infected locations (TRSs (township/range/section)). We used hunter-harvest and LFC deer from 4621 and 435 unique TRSs, respectively, over 16 years of surveillance and management (2003–2017). We divided the study area into groups of TRSs with similar landcover types (SPLT) to assess CWD hunter-harvest prevalence at LFC and non-LFC sites by landcover composition. We also evaluate the importance of the month when LFC was implemented and determine whether the density of LFC sites or the total number of deer removed by LFC predicts hunter-harvest CWD prevalence. The percentage of CWD positive samples from hunters was lower than for LFC samples (for the study area and SPLTs). The probability of CWD increased by 5.24% for all the SPLT groups combined in the study area, by 4.6% from areas without an LFC nearby, and by 1.21% for areas with a prior LFC nearby. For all the TRS in the study area, low CWD odds (<1) in hunter-harvest deer were found in three SPLTs, in two SPLTs within TRSs with non-LFC, and five in five SPLTs within TRSs with LFC. The results suggest the importance of accounting for landcover composition to implement and sustain management in habitats with a higher risk of CWD. Our findings support that hunter-harvest alone cannot control CWD and the critical need for continued LFC intervention. For the whole study area—regardless of landcover composition—LFC in January was more important in decreasing hunter-harvest CWD prevalence than when LFC was conducted in March. However, the LFC conducted in January, February, and March were equally important when evaluating the month per habitat. Furthermore, the density of LFC sites in proximity to known infected areas is a better predictor of CWD than the number of deer removed by LFC, suggesting that increasing the density of LFC sites has a greater impact on CWD. The proximity of LFC to infected areas helps control CWD. Ultimately, landowners’ and hunters’ collaborations with the CWD surveillance and management programs are critical to protecting the Illinois wild deer herd; this study demonstrates their ongoing and valuable contributions to protecting this natural and public resource.

Chronic Wasting Disease Monitoring in Italy 2017-2019: Neuropathological Findings in Cervids.

Chronic wasting disease (CWD) is a prion disease that affects cervids; it is classified under transmissible spongiform encephalopathies (TSEs). CWD is particularly contagious, making its eradication in endemic areas very difficult and creating serious problems for cervid conservation and breeding. It has recently become an emerging public health risk to be managed by health authorities. Starting in 2017, active CWD surveillance in Italy has intensified with the monitoring of wild and farmed cervids. The present study summarizes findings from a histopathological survey of the brains from wild ruminants collected via CWD monitoring between 2017 and 2019. A total of 113 brains from 62 red deer (Cervus elaphus) and 51 roe deer (Capreolus capreolus) were submitted for analysis at the National Reference Center for Animal Encephalopathies (CEA) to determine major patterns of neuropathological lesions and correlated pathogens. Brain lesions were detected in 20 animals, 10 brain samples were unsuitable for examination, and 83 presented no lesions. Neuropathological examination revealed non-suppurative encephalitis or meningoencephalitis in most cases (15/20). This brain study revealed evidence for the absence of CWD in Italy and provided a reference spectrum of neuropathological lesions for differential diagnosis in cervids.

COMPARISON OF CHRONIC WASTING DISEASE DETECTION METHODS AND PROCEDURES: IMPLICATIONS FOR FREE-RANGING WHITE-TAILED DEER (ODOCOILEUS VIRGINIANUS) SURVEILLANCE AND MANAGEMENT.

Throughout North America, chronic wasting disease (CWD) has emerged as perhaps the greatest threat to wild cervid populations, including white-tailed deer (WTD; Odocoileus virginianus). White-tailed deer are the most sought-after big game species across North America with populations of various subspecies in nearly all Canadian provinces, the contiguous US, and Mexico. Documented CWD cases have dramatically increased across the WTD range since the mid-1990s, including in Minnesota, US. CWD surveillance in free-ranging WTD and other cervid populations mainly depends upon immunodetection methods such as immunohistochemistry and enzyme-linked immunosorbent assay (ELISA) on medial retropharyngeal lymph nodes and obex. More recent technologies centered on prion protein amplification methods of detection have shown promise as more sensitive and rapid CWD diagnostic tools. Here, we used blinded samples to test the efficacy of real-time quaking-induced conversion (RT-QuIC) in comparison to ELISA for screening tissues collected in 2019 from WTD in southeastern Minnesota, where CWD has been routinely detected since 2016. Our results support previous findings that RT-QuIC is a more sensitive tool for CWD detection than current antibody-based methods. Additionally, a CWD testing protocol that includes multiple lymphoid tissues (e.g., medial retropharyngeal lymph node, parotid lymph node, and palatine tonsil) per animal can effectively identify a greater number of CWD detections in a WTD population than a single sample type (e.g., medial retropharyngeal lymph nodes). These results show that the variability of CWD pathogenesis, sampling protocol, and testing platform must be considered for the effective detection and management of CWD throughout North America.

Spatio-temporal changes in chronic wasting disease risk in wild deer during 14 years of surveillance in Alberta, Canada

Disease risk modeling is a key first step to understand the spatio-temporal dynamics of wildlife disease and to direct cost-effective surveillance and management. In Alberta, active surveillance for chronic wasting disease (CWD) in wild cervids began in 1998 with the first case detected in free-ranging cervids in 2005. Following the detection, a herd reduction program was implemented during 2005–2008 and in 2006 the ongoing hunter-based CWD Surveillance Program became mandatory in high-risk Wildlife Management Units (WMU). We used data collected during the CWD surveillance program to 1) document growth in sex-specific CWD prevalence (proportion of deer in sample that is CWD-positive) in hunter-harvest deer in 6 WMUs consistently monitored from 2006 to 2018, 2) document landscape features associated with where CWD-positive compared to CWD-negative deer were removed during hunter harvest and herd reduction in an early (2005–2012) and in a late period (2013–2017), and 3) to map the spatial risk of harvesting a deer infected with CWD in the prairie parklands of Alberta. In the 6 continuously monitored WMUs, risk of a harvested deer being CWD positive increased from 2006 to 2018 with CWD prevalence remaining highest in male mule deer whereas overall growth rate in CWD prevalence was greater in female mule deer, but similar to male white-tailed deer. We found no evidence that the 3-year herd reduction program conducted immediately after CWD was first detected affected the rate at which CWD grew over the course of the invasion. Risk of deer being CWD-positive was the highest in animals taken near small stream drainages and on soils with low organic carbon content in the early period, whereas risk became highest in areas of agriculture especially when far from large river drainages where deer often concentrate in isolated woody patches. The change in the influence of proximity to known CWD-positive cases suggested the disease was initially patchy but became more spatially homogeneous over time. Our results indicate that a targeted-removal program will remove more CWD positive animals compared to hunter harvest. However, the discontinuation of targeted removals during our research program, restricted our ability to assess its long term impact on CWD prevalence.

Hofmeister Effect in RT-QuIC Seeding Activity of Chronic Wasting Disease Prions.

Chronic wasting disease (CWD) is a transmissible spongiform encephalopathy (TSE) that causes a fatal neurodegenerative disease in cervids. Cases of CWD are rapidly increasing in North America among wild and farmed cervid populations, and potential for zoonotic transmission is not yet determined. Therefore, in order to manage the disease, it is imperative to devise a system that can detect CWD during its early phases to prevent spread to new captive herds through introduction of CWD-affected animals into otherwise CWD-free herds. Real-time quaking–induced conversion (RT-QuIC) assays have been applied to detect the presence of disease-associated prions from various samples in both animals and humans. In this study, we have tested the use of five Hofmeister anions that range from weakly hydrating to strongly hydrating: Na3citrate, Na2SO4, NaCl, NaI, and NaClO4 in RT-QuIC reactions for CWD seeding activity using different recombinant prion proteins as substrates. This work shows how the ionic environment of the RT-QuIC reaction can enhance or diminish the seeding activity. The use of Na2SO4 or NaI as the sodium salt for RT-QuIC using bank vole recombinant prion substrate for the detection of CWD using brain samples reduces the lag time to detect with reasonable specificity. For detection of the CWD in fecal samples, only NaI showed comparable reduction in lag time relative to NaCl but required reduced temperature to alleviate spontaneous fibril formation in negative control samples. Selection of the proper ion environment and recombinant prion protein substrate will make RT-QuIC a powerful diagnostic tool for early detection of CWD prions, further supporting CWD surveillance in wild and captive cervids.

Risk-based surveillance of chronic wasting disease in semi-domestic reindeer

Reindeer pastoralism is a widespread practise across Fennoscandia and Russia. An outbreak of chronic wasting disease (CWD) among wild reindeer (Rangifer tarandus) poses a severe threat to the semi-domestic reindeer herding culture. Establishing surveillance is therefore key, but current models for surveillance of CWD are designed for wild cervids and rely on samples obtained from recreational hunters. Targeting animal groups with a higher infection probability is often used for more efficient disease surveillance. CWD has a long incubation period of 2–3 years, and the animals show clinical signs in the later stages of the infection i.e. 1−4 months prior to death. The semi-domestic reindeer are free-ranging most of the year, but during slaughtering in late fall, herders stress the animals in penned areas. This allows removal of animals with deviant behaviour or physical appearance, and such removals are likely to include animals in the clinical stages of CWD if the population is infected. In Norway, the semi-domestic reindeer in Filefjell is adjacent to a previously CWD infected wild population. We developed a risk-based surveillance method for this semi-domestic setting to establish the probability of freedom from infection over time, or enable early disease detection and mitigation. The surveillance scheme with a scenario tree using three risk categories (sample category, demographic group, and deviations in behaviour or physical appearance) was more effective and less invasive as compared to the surveillance method developed for wild reindeer. We also simulated how variation in susceptibility, incubation period and time for onset of clinical signs (linked to variation in the prion protein gene, PRNP) would potentially affect surveillance. Surveillance for CWD was mandatory within EU-member states with reindeer (2018–2020). The diversity of management systems and epidemiological settings will require the development of a set of surveillance systems suitable for each different context. Our surveillance model is designed for a population with a high risk of CWD introduction requiring massive sampling, while at the same time aiming to limit adverse effects to the populations in areas of surveillance.

Landscape Features Fail to Explain Spatial Genetic Structure in White‐Tailed Deer Across Ohio, USA

ABSTRACTLandscape features influence wildlife movements across spatial scales and have the potential to influence the spread of disease. Chronic wasting disease (CWD) is a fatal prion disease affecting members of the family Cervidae, particularly white‐tailed deer (Odocoileus virginianus), and the first positive CWD case in a wild deer in Ohio, USA, was recorded in 2020. Landscape genetics approaches are increasingly used to better understand potential pathways for CWD spread in white‐tailed deer, but little is known about genetic structure of white‐tailed deer in Ohio. The objectives of our study were to evaluate spatial genetic structure in white‐tailed deer across Ohio and compare the support for isolation by distance (IBD) and isolation by landscape resistance (IBR) models in explaining this structure. We collected genetic data from 619 individual deer from 24 counties across Ohio during 2007–2009. We used microsatellite genotypes from 619 individuals genotyped at 11 loci and haplotypes from a 547‐base pair fragment of the mitochondrial DNA control region. We used spatial and non‐spatial genetic clustering tests to evaluate genetic structure in both types of genetic data and empirically optimized landscape resistance surfaces to compare IBD and IBR using microsatellite data. Non‐spatial genetic clustering tests failed to detect spatial genetic structure, whereas spatial genetic clustering tests indicated subtle spatial genetic structure. The IBD model consistently outperformed IBR models that included land cover, traffic volume, and streams. Our results indicated widespread genetic connectivity of white‐tailed deer across Ohio and negligible effects of landscape features. These patterns likely reflect some combination of minimal resistive effects of landscape features on white‐tail deer movement in Ohio and the effects of regional recolonization or translocation. We encourage continued CWD surveillance in Ohio, particularly in the proximity of confirmed cases. © 2021 The Wildlife Society. Real-Time Quaking-Induced Conversion Detection of PrPSc in Fecal Samples From Chronic Wasting Disease Infected White-Tailed Deer Using Bank Vole Substrate.

Chronic wasting disease (CWD) is a transmissible spongiform encephalopathy (TSE) that is fatal to free-range and captive cervids. CWD has been reported in the United States, Canada, South Korea, Norway, Finland, and Sweden, and the case numbers in both wild and farmed cervids are increasing rapidly. Studies indicate that lateral transmission of cervids likely occurs through the shedding of infectious prions in saliva, feces, urine, and blood into the environment. Therefore, the detection of CWD early in the incubation time is advantageous for disease management. In this study, we adapt real-time quacking-induced conversion (RT-QuIC) assays to detect the seeding activity of CWD prions in feces samples from clinical and preclinical white-tailed deer. By optimizing reaction conditions for temperature as well as the salt and salt concentration, prion seeding activity from both clinical and preclinical animals were detected by RT-QuIC. More specifically, all fecal samples collected from 6 to 30 months post inoculation showed seeding activity under the conditions of study. The combination of a highly sensitive detection tool paired with a sample type that may be collected non-invasively allows a useful tool to support CWD surveillance in wild and captive cervids.

Getting in Front of Chronic Wasting Disease: Model-Informed Proactive Approach for Managing an Emerging Wildlife Disease.

Continuing geographic spread of chronic wasting disease (CWD) poses a serious threat to the sustainable future of cervids and hunting in North America. Moreover, CWD has been detected in captive cervids in South Korea and, in recent years, in free-ranging reindeer in Europe (Norway). Management of this disease is limited by logistical, financial, and sociopolitical considerations, and current strategies primarily focus on reducing host densities through hunter harvest and targeted culling. The success of such strategies in mitigating the spread and prevalence of CWD only upon detection is questionable. Here, we propose a proactive approach that emphasizes pre-emptive management through purposeful integration of virtual experiments (simulating alternate interventions as model scenarios) with the aim of evaluating their effectiveness. Here, we have used a published agent-based model that links white-tailed deer demography and behavior with CWD transmission dynamics to first derive a CWD outbreak trajectory and then use the trajectory to highlight issues associated with different phases of the CWD outbreak (pre-establishment/transition/endemic). Specifically, we highlight the practical constraints on surveillance in the pre-establishment phase and recommend that agencies use a realistic detection threshold for their CWD surveillance programs. We further demonstrate that many disease introductions are “dead ends” not leading to a full epidemic due to high stochasticity and harvesting in the pre-establishment phase of CWD. Model evaluated pre-emptive (pre-detection) harvest strategies could increase the resilience of the deer population to CWD spread and establishment. We conclude it is important to adaptively position CWD management ahead of, rather than behind, the CWD front.

An agent-based framework for improving wildlife disease surveillance: A case study of chronic wasting disease in Missouri white-tailed deer

Epidemiological surveillance for important wildlife diseases often relies on samples obtained from hunter-harvested animals. A problem, however, is that although convenient and cost-effective, hunter-harvest samples are not representative of the population due to heterogeneities in disease distribution and biased sampling. We developed an agent-based modeling framework that i) simulates a deer population in a user-generated landscape, and ii) uses a snapshot of the in silico deer population to simulate disease prevalence and distribution, harvest effort and sampling as per user-specified parameters. This framework can incorporate real-world heterogeneities in disease distribution, hunter harvest and harvest-based sampling, and therefore can be useful in informing wildlife disease surveillance strategies, specifically to determine population-specific sample sizes necessary for prompt detection of disease. Application of this framework is illustrated using the example of chronic wasting disease (CWD) surveillance in Missouri’s white-tailed deer (Odocoileus virginianus) population. We show how confidence in detecting CWD is grossly overestimated under the unrealistic, but standard, assumptions that sampling effort and disease are randomly and independently distributed. We then provide adjusted sample size recommendations based on more realistic assumptions. Wildlife agencies can use these open-access models to design their CWD surveillance. Furthermore, these models can be readily adapted to other regions and other wildlife disease systems.

Partnering with Taxidermists for Improved Chronic Wasting Disease Surveillance.

Simple SummaryChronic wasting disease (CWD) is a contagious neurological disease affecting deer, moose, elk, and reindeer. CWD is predominantly found in North America and has a higher prevalence in older male deer. To increase the submission of samples from older male deer, the Taxidermy Partnership Program (TPP) was implemented in New York State (NYS). Prior to 2013, CWD surveillance in New York State focused on testing samples of convenience, by collecting deer heads from meat processors. However, this sampling was biased because many of the heads from older male deer were taken to taxidermists to be mounted. In 2013, the Taxidermy Partnership Program (TPP) was created to train taxidermists to collect CWD samples, and to increase the proportion of older male deer submitted for CWD testing. Added benefits include improved communication with taxidermists and increased awareness about CWD. Trained taxidermists were able to successfully collect and submit tissue samples with few errors. Participating taxidermists were paid for viable samples. Currently, there is a stable number of taxidermists that participate each year. This program has proven to be a valuable resource for obtaining high-value CWD samples for the wildlife agency, requiring a minimal amount of funding and time.

Annual Report of the Scientific Network on BSE‐TSE 2018

The EFSA Scientific Network on bovine spongiform encephalopathies and other transmissible spongiform encephalopathies (BSE-TSE) held its 13th meeting on 15-16 October 2018 in Parma. The meeting served as an opportunity to exchange scientific information on BSE-TSE related issues among EU Member States, countries from the European Free Trade Association (EFTA), EFSA, the European Commission and ad hoc participants. In this occasion, ad hoc representation included the World Animal Health Organisation (OIE), the Department of Food Safety, Nutrition and Veterinary Public Health of the Istituto Superiore di Sanità (Italy), and the Animal and Plant Health Agency, UK. The topics discussed included: an update on BSE-TSE-related activities at national and international level and results from recent TSE-related research projects and risk assessments in several EU countries. A method that accounts for differential detectability in mixed samples of long‐term infections with applications to the case of chronic wasting disease in cervids

Abstract Surveillance of wildlife diseases is logistically difficult, and imperfect detection is a recurrent challenge for disease estimation. Using citizen science can increase sample sizes, but it is associated with a cost in terms of the anatomical type and quality of the sample. Additionally, biological tissue samples from remote areas lose quality due to autolysis. These challenges are faced in the case of emerging chronic wasting disease (CWD) in cervids. Here, we develop a stochastic scenario tree model of diagnostic sensitivity, allowing for a mixture of tissue sample types (lymph nodes and brain) and qualities while accounting for different detection probabilities during the CWD infection, lasting 2–3 years. We apply the diagnostic sensitivity in a Bayesian framework, enabling estimation of age‐class‐specific true prevalence, including the prevalence in latent, recently infected stages. We provide a simulation framework to estimate the sensitivity of the surveillance system (i.e., the probability of detecting the infection in a given population), when detectability varies among individuals due to different disease progression. We demonstrate the utility of our framework by applying it to the recent emergence of CWD in a European population of reindeer. We estimated apparent CWD prevalence at 1.2% of adults in the infected population of wild reindeer, while the true prevalence was 1.6%. The sensitivity estimation of the CWD surveillance was performed in an adjacent small (c. 500) and a large (c. 10,000) reindeer population, demonstrating low certainty of CWD absence. Our method has immediate application to the mandatory testing for CWD in EU countries commencing in 2018. Similar approaches that account for latent stages and a serial disease progression in various tissues with a temporal pattern of diagnostic sensitivity may enhance the estimation of the prevalence of wildlife diseases more generally.

Polymorphisms in the Prion Protein Gene, Associated with Chronic Wasting Disease, in the Korean Water Deer (Hydropotes inermis argyropus)

Prion diseases are fatal neurodegenerative diseases that affect humans and non-human mammals. Different alleles of the prion protein gene (PRNP) of humans and sheep are associated with varying susceptibilities to transmissible spongiform encephalopathies. Chronic wasting disease (CWD) is a prion disease of cervids, and polymorphisms at specific codons in the PRNP gene are associated with this disease. To assess the susceptibility of free-ranging deer to CWD, polymorphisms in PRNP were examined in the Korean water deer (Hydropotes inermis argyropus), focusing on codons that are potentially associated with CWD susceptibility (95, 96, 116, 132, 225, and 226). CWD surveillance was conducted by an antigen ELISA of tissue samples from 545 Korean water deer collected in eight provinces of the Republic of Korea. No prion protein associated with CWD was detected in any of the samples. These results suggest that PRNP of the Korean water deer shows low variation and the species has not been infected with CWD.
