Chlorophyll fluorescence characteristics and H2O2 contents of Chinese tallow tree are dependent on population origin, nutrients and salinity

Abstract Plants from invasive populations often have higher growth rates than conspecifics from native populations due to better environmental adaptability. However, the roles of improved chlorophyll fluorescence or antioxidant defenses in helping them to grow better under adverse situations are insufficient, even though this is a key physiological question for elucidating mechanisms of plant invasion. Here, we conducted experiments with eight native (China) and eight introduced (USA) populations of Chinese tallow tree (Triadica sebifera). We tested how salinity, nutrients (overall amount or N:P in two separate experiments) and their interaction affected T. sebifera aboveground biomass, leaf area, chlorophyll fluorescence and antioxidant defenses. Plants from introduced populations were larger than those from native populations, but salinity and nutrient shortage (low nutrients or high N:P) reduced this advantage, possibly reflecting differences in chlorophyll fluorescence based on their higher PSII maximum photochemical efficiency (Fv/Fm) and PSI maximum photo-oxidizable P700 in higher nutrient conditions. Native population plants had lower Fv/Fm with saline. Except in high nutrients/N:P with salinity, introduced population plants had lower electron transfer rate and photochemical quantum yield. There were no differences in antioxidant defenses between introduced and native populations except accumulation of hydrogen peroxide (H2O2), which was lower for introduced populations. Low nutrients and higher N:P or salinity increased total antioxidant capacity and H2O2. Our results indicate that nutrients and salinity induce differences in H2O2 contents and chlorophyll fluorescence characteristics between introduced and native populations of an invasive plant, illuminating adaptive mechanisms using photosynthetic physiological descriptors in order to predict invasions.

Fructose Regulation of Stomatal Movement and its Effects on the Photosynthetic System in Vicia faba L.

Background: Vicia faba L. is a significant grain legume that is rich in protein and nutrients. It is widely cultivated for human consumption and animal feed in temperate regions. As a signaling molecule, fructose is involved in many metabolic and developmental processes. The objective of this study was to demonstrate whether fructose may increase reactive oxygen species (ROS) production in guard cells inducing stomatal closure and examine whether the exogenous application of fructose may regulate photosynthesis and chlorophyll fluorescence characteristics. Methods: After approximately 4 weeks of sowing the treated broad bean seeds, the fully inflated leaves were used for experimental data collection including stomatal movement, gas exchange, chlorophyll fluorescence and ROS production in guard cells. Result: Fructose reduced stomatal conductance (Gs), transpiration rate (Tr) and stomatal aperture of intact leaves, indicating that fructose can trigger stomatal closure in a dose- and time-dependent manner in the epidermal peels of the broad bean plant (Vicia faba L.). Meanwhile, fructose increased the photochemical efficiency, photosynthetic rate (Pn) and water use efficiency (WUE). Fructose-induced stomatal closure was associated with nitric oxide (NO), calcium ion (Ca2+), aquaporin and ROS.

Photosynthetic activity of sweet sorghum fertilised by sewage sludge

ABSTRACT Sewage sludge is a promising fertiliser for sweet sorghum growing on marginal land. We investigate the effects of different doses of sludge on the characteristics of chlorophyll fluorescence. Two types of sewage sludge were used: without flocculant and treated with flocculant. Sludge application helps to reduce the amount of inactive chlorophyll in the reaction centres, increases photosynthetic activity, and reduces the sensitivity of the photosynthetic apparatus to adverse environmental factors. Flocculated sludge had the more beneficial effect, particularly at the early stages of development; the optimal dose was 40-60t/ha.

Monitoring low O2 stress in apples – What we can learn from chlorophyll fluorescence and respiratory characteristics

Dynamic controlled atmosphere (DCA) is becoming a storage method of choice for apple producers seeking to store long-term. There is limited prior work comparing DCA monitoring chlorophyll fluorescence yield (DCA-CF) and respiratory characteristics (DCA-RQ), particularly for the popular UK-grown apple cultivars ‘Gala’ and ‘Braeburn’. However, these techniques can give an insight into the physiological response of apples to low O2 storage, which could help to design more efficient and sustainable storage regimes. In this study the chlorophyll fluorescence (CF) and respiratory quotient (RQ) responses of ‘Gala’ and ‘Braeburn’ are compared in typical and exaggerated DCA conditions. Results show that not only do the two techniques differ in their detection of low O2 stress but the two cultivars studied also differ noticeably in their behaviour. CF increase was confirmed to be a reaction to a stress signal from inside the fruit. Respiration rate curves were found to reflect the internal structure of the fruit, with ‘Gala’’s more homogeneous tissues and greater gas permeability resulting in a strongly bimodal O2 consumption rate where the change in rate hypothetically corresponds with the majority of the fruit tissue switching to anaerobic respiration at the same time. CF and RQ showed noticeable differences between cultivars, agreeing with previous observations that ‘Braeburn’ is less tolerant of low O2 and elevated CO2 than ‘Gala’ (CF Lower O2 Limit (LOL) for Braeburn of approximately 0.75 kPa O2 relative to 0.5 kPa O2 for Gala). Additionally, the CF and RQ responses indicated different LOLs for ‘Gala’, at approximately 0.5 kPa O2 for CF and 0.25 kPa O2 for RQ. Elevated CO2 depressed both the CF and RQ response, with a strong depressive effect on the rate of CO2 evolution but little effect on the rate of O2 consumption.

Characteristics of Solar-Induced Chlorophyll Fluorescence in the Three River Headwaters Region, Qinghai-Tibetan Plateau during 2001 to 2020

The ecology of the Three River Headwaters Region (TRHR) is related to the long-term sustainable development of Qinghai Province and the whole of China. The change in chlorophyll fluorescence is an important index to measure the ecological environment. Therefore, it is of great significance to study the spatial and temporal distributions of Solar-induced chlorophyll fluorescence (SIF)and the related influencing factors in the TRHR. In this study, a high-resolution SIF dataset (2001–2020) was selected to be averaged on a time scale of years and months to investigate the annual and seasonal SIF characteristics, and the influencing climate factors were analyzed in combination with meteorological data by statistical method. The results showed that the SIF values ranged from 0.05 to 0.073 during 2001–2020, with a peak value of 0.073 in 2005 and 2009 and a minimum value of 0.05 in 2002. The averages of SIF values were higher in the source regions of the Yellow River source region (YR) and Langcang (Mekong) River source region (LCR) than in the source region of the Yangtze River source region (YZR). The SIF values of the TRHR in July, August and September were significantly higher than those in other months. The maximum value occurred in August at 0.11, and the minimum value was 0.008 in December. The precipitation had greater effect on the inter-annual variations in SIF. The monthly variation of SIF is influenced by precipitation, temperature and relative humidity. In addition, the influence of human activities and altitude on SIF should not be ignored. The results have certain reference value for protecting vegetation in the TRHR, and provide a reference for other regions to analyze the spatiotemporal changes and influencing factors by using SIF data.

Performance Evaluation of New Table Grape Varieties under High Light Intensity Conditions Based on the Photosynthetic and Chlorophyll Fluorescence Characteristics

The evaluation of photosynthetic characteristics of plants is important for the success rate of germplasm introduction. To select grape varieties with higher adaptability and trait performance, this experiment is aimed at evaluating and comparing the photosynthetic indices, chlorophyll fluorescence parameters, photosynthetic pigment content, and leaf characteristics of five Chinese hybrid varieties. The results showed that under high light intensity stress, the leaf growth of ‘Ruidu Cuixia’ was most affected and its specific leaf weight was the lowest, while ‘Jing Hongbao’ had the highest chlorophyll content. The maximum net photosynthetic rate (Pnmax), maximum light quantum yield (Fv/Fm), and apparent quantum efficiency (AQE) were different among varieties. It was reported that the ‘Ruidu Zaohong’ variety had the highest Pnmax. ‘Ruidu Wuheyi’ was found to have the highest Fv/Fm, while the highest AQE was recorded for ‘Ruidu Cuixia’, with intercellular CO2 concentration (Ci) and stomatal conductance (gs) at 292.56 μmol·mol−1, 766.56 mmol·m−2·s−1, and 66.8 μmol·m−2·s−1, respectively. The indices of ABS/CSm, TRo/CSm, and DIo/CSm were significantly different among varieties, and these indices of ‘Ruidu Zaohong’ were the highest. Pn was positively correlated with Ci and Tr, gs were positively correlated with Fv and TRo/CSm. The specific leaf area was negatively correlated with Fv/Fm and ΦDIo. The results of the principal component analysis and TOPSIS comprehensive evaluation showed that ‘Jing Hongbao’ and ‘Ruidu Cuixia’ performed best. Overall, the measurement of the photosynthetic characteristics of the plants during the growing period provided valuable data for the varietal introduction strategies. The better photosynthetic performance of ‘Jing Hongbao’ and ‘Ruidu Cuixia’ indicates more adaptability to the long day, high light intensity, and the high-temperature climate of Xinjiang.

Exploration of chlorophyll fluorescence characteristics gene regulatory in rice (Oryza sativa L.): a genome-wide association study.

Chlorophyll content and fluorescence parameters are crucial indicators to evaluate the light use efficiency in rice; however, the correlations among these parameters and the underlying genetic mechanisms remain poorly understood. Here, to clarify these issues, we conducted a genome-wide association study (GWAS) on 225 rice accessions. In the phenotypic and Mendelian randomization (MR) analysis, a weak negative correlation was observed between the chlorophyll content and actual quantum yield of photosystem II (). The phenotypic diversity observed in SPAD, , , and among accessions was affected by genetic background. Furthermore, the GWAS identified 78 SNPs and 17 candidate genes significantly associated with SPAD, , , , and . Combining GWAS on 225 rice accessions with transcriptome analysis of two varieties exhibiting distinct fluorescence characteristics revealed two potential candidate genes (Os03g0583000 from & traits and Os06g0587200 from trait), which are respectively associated with peroxisomes, and protein kinase catalytic domains might involve in regulating the chlorophyll content and chlorophyll fluorescence. This study provides novel insights into the correlation among chlorophyll content and fluorescence parameters and the genetic mechanisms in rice, and offers valuable information for the breeding of rice with enhanced photosynthetic efficiency.

Effects of Shading on the Growth and Photosynthetic Fluorescence Characteristics of Castanopsis hystrix Seedlings of Top Community-Building Species in Southern Subtropical China

Castanopsis hystrix is a major community-building species in the top communities of southern subtropical China, with a high natural regeneration capacity. However, excessive logging and the introduction of exotic tree species have substantially reduced the area of natural forest patches of Castanopsis hystrix, and seedling regeneration is essential for the long-term continuation of Castanopsis hystrix populations. To explore the effects of light intensity on the seedling emergence and early growth of Castanopsis hystrix, shading experiments were conducted under four shading treatments (0%, 40%, 60%, and 80%). The growth, biomass accumulation, and distribution, the quality index of seedlings, the morphology and structure of the root systems of seedlings, and the leaf chlorophyll content and chlorophyll fluorescence properties of seedlings under different shading treatments were analyzed. The results displayed the following: (1) Shade intensity impacts growth of Castanopsis hystrix seedlings and biomass allocation, with optimal results observed at 60% shade, leading to the promotion of organic matter production in leaves and the limitation of stem growth. (2) Using a multi-indicator composite index, it was determined that seedling quality for Castanopsis hystrix peaks at 60% shade intensity. (3) Shade significantly impacts the morphology and structure of Castanopsis hystrix’s root system, with most root characteristics peaking at 60% shade, indicating a substantial increase in root development compared to no-shade conditions. (4) The D-values indicated the most suitable shade intensity for seedling growth to be 60%, suggesting that Castanopsis hystrix seedlings are sensitive to light and excessive light can be detrimental to their growth. (5) The 60% shade treatment showed the maximum values of chlorophyll fluorescence characteristics and photochemical activity, with variations in energy conversion efficiency and dissipation reflected in parameters like photochemical burst coefficient (qP), photochemical burst coefficient (qN), the actual photometric yield of PSII under light acclimation (YII), and the maximum photosynthetic electron transport rate in photoinhibition (ETR), thereby supporting seedling growth and maintaining the normal function of photosynthetic organs. In conclusion, 60% shade treatment can effectively improve the growth and photosynthetic characteristics of Castanopsis hystrix seedlings and promote the accumulation of nutrient elements, ultimately promoting their growth.

Waterlogging Tolerance of Actinidia valvata Dunn Is Associated with High Activities of Pyruvate Decarboxylase, Alcohol Dehydrogenase and Antioxidant Enzymes.

Kiwifruit (Actinidia spp.) is susceptible to waterlogging stress. Although abundant wild germplasm resources exist among Actinidia plants for improving the waterlogging tolerance of kiwifruit cultivars, the underlying mechanisms remain largely unknown. Here, a comparative study was undertaken using one wild germplasm, Maorenshen (A. valvata Dunn, MRS), and one cultivar, Miliang-1 (A. chinensis var. deliciosa (A.Chev.) A.Chev. cv. Miliang-1, ML). Under stress, the ML plantlets were seriously damaged with wilted chlorotic leaves and blackened rotten roots, whereas the symptoms of injury in the MRS plantlets were much fewer, along with higher photosynthetic rates, chlorophyll fluorescence characteristics and root activity under stress conditions. However, neither aerenchyma in the root nor adventitious roots appeared in both germplasms upon stress exposure. The activities of pyruvate decarboxylase (PDC) and alcohol dehydrogenase (ADH), as well as their transcript levels, were constitutively higher in MRS than those in ML under both normal and stress conditions. Waterlogging stress significantly enhanced the PDC and ADH enzyme activities in both germplasms, which were 60.8% and 22.4% higher in the MRS roots than those in the ML roots under waterlogging stress, respectively. Moreover, MRS displayed higher activities of antioxidant enzymes, including SOD, CAT, and APX, as well as DPPH-radical scavenging ability, and decreased H2O2 and MDA accumulation under both normal and stress conditions. Our findings suggest that the waterlogging tolerance of the wild A. valvata germplasm was associated with high PDC and ADH, as well as antioxidant ability.

Do various levels of salinity change chlorophyll fluorescence, nutrient uptake, and physiological characteristics of Mentha ecotypes?

Mint (Lamiaceae) is an economically important crop with many uses in the herbal, ornamental, culinary, pharmaceutical and food industries. To expand global cultivation, a better understanding of the growth and physiological responses of mint to soil salinity conditions is increasingly a strategic necessity. This study was aimed to investigate the NaCl (0, 2.5, 5, and 7.5 dS m−1) salinity stress effects on chlorophyll fluorescence, nutrient uptake, and physiological characteristics of different mint species [(18 ecotypes(E)] under the greenhouse environment. Based on the results obtained, the highest and the lowest soluble sugar contents (93.41 and 39.61 mg g−1 dry matter, respectively) were found in E15 at 7.5 dS m−1 salinity and in E6 under non-saline conditions that was raised by 57.5% upon 7.5 dS m−1 NaCl treatment over the control. The leaf relative water content was decreased in response to increasing salinity levels with the lowest values occurring at the highest salinity treatment by 42.2%, where ecotype 16 had the highest relative water content (53.2%) in the same salinity levels. Although, stomatal conductivity decreased as salinity intensity increased, the amount of this increase varied among ecotypes. At the highest level of salinity, E16 and E6 showed maximum and minimum stomatal conductivity, 7.23 and 4.13 mmol (H2O) m−2 s−1, respectively, which was a 42.8% difference between them. By increasing salinity levels, the nitrogen content of the plants was significantly decreased, which varied from 0.74 mg g−1 dry matter for ecotype 1–0.98 mg g−1 DW for ecotype 16. In term of 7.5 dS m−1 NaCl, the highest level of phosphorus (0.25 mg g−1 dry matter) was observed in ecotype 18. As salinity levels increased, the amount of potassium in shoot was decreased in a dose dependent manner in the mint ecotypes tested. At the highest salinity level, E8 and E15 had the highest, and E6 and E16 had the lowest level of potassium in shoot (1.34 mg g−1 dry matter and 1.24 mg g−1 dry matter, respectively). Overall, this research indicates that the mint E14, E10, and E18 are the most tolerant ecotypes displaying a high Fv/Fm index at all employed NaCl salinity levels. Thus, saline conditions may change the physiological processes and defense pathway resulting in dramatic changes in plant nutrients, photosynthesis efficiency and essential oil content of Mentha species.

Effects of Seasonal Changes on Chlorophyll Fluorescence and Physiological Characteristics in the Two Taxus Species.

Taxus is a rare and endangered woody plant worldwide with important economic and ecological values. However, the weak environmental adaptability of Taxus species, in particular the unstable photosynthetic activity in different seasons, always affects its normal growth and development and limits its conservation and exploitation. To improve the survival of Taxus trees in cultivated areas, the seasonal dynamics of chlorophyll fluorescence (CF) and key physiological parameters were comprehensively investigated in T. media and T. mairei. The results demonstrated that the photosynthetic activity of both Taxus species was sensitive to local summer and winter environmental conditions, with the heterogeneity of fluorescence signatures intuitively presented on the needle surface by CF-Imaging detection, while images of maximum quantum efficiency of PSII photochemistry (Fv/Fm) demonstrated values below 0.7 in the blue-green sectors in winter. The distribution of light energy was regulated by the photosynthetic apparatus in both Taxus species to maintain a stable actual quantum yield of PSII photochemistry (φPSII), which was around 0.4-0.5. Based on a redundancy discriminant analysis, the interpretation rate of light intensity and air temperature ranked as the top two in both Taxus species, which were considered the main environmental factors affecting the photosynthetic performance of Taxus by disturbing the electron transport chain. In the winter, T. mairei exhibited weaker electron transport activity than T. media, thus caused lower photochemistry and more severe photosynthetic damages. Interestingly, both Taxus species demonstrated consistent response patterns, including diverse energy dissipation strategies and enhancement of osmoregulatory substances and antioxidative activities, thus maintaining stable photosynthetic functions in response to environmental changes.

Light spectrum effects on rocket and lamb's lettuce cultivated in a vertical indoor farming system

Rocket and lamb's lettuce are new leafy vegetables whose consumption in salads is increasing, especially as ready-to-eat products. These plants are often cultivated in greenhouses under artificial light, and are very important due to their pleasant taste, texture, and nutritional value to humans. The present study analyzes the effects of light quality provided by light emitting diodes (LEDs) on the yield and quality of different leafy vegetable species. Lamb's lettuce and rocket plants were grown in the same conditions under 4 light spectrums color fractions (NS-12, AP57, AP673L, and G2) for 27 and 37 days, respectively. Then fresh and dry weight, chlorophyll fluorescence, and chromatic characteristics were determined. The leaf quality of lamb's lettuce and rocket was characterized by measurements of the contents of carbohydrates, organic acids, and amino acids in leaves. Vegetative growth was significantly influenced by light quality and was species-dependent, and treatment with the highest proportion of Red:Blue light (G2) produced the highest fresh weight and maximum efficiency of PSII. The results of the chlorophyll fluorescence parameters were more influenced by the species rather than by the spectrum lights, except for the efficiency of PSII, which was higher in treatments with the lowest Red: Far-red ratio (AP67 and G2). G2 light spectrum reduces chlorophyll and carotenoid content compared to other treatments. Among all the primary metabolites analyzed, non-structural carbohydrates were the most dependent on light quality. Generally, the lights with the greatest effect on the parameters analyzed were those with higher Red:Blue ratios and a higher far-red fraction. No effect of light spectra on glutamate concentration was observed. However, a concentration increase was observed in treatments with a higher proportion of far-red light. Regarding organics acids, the different light spectrums did not affect the concentration of the majority of acids (citrate and malate) in rocket and lamb´s lettuce plants. The spectral composition of light has an influence on the quality and quantity of lamb's lettuce and rocket production. Therefore, each species studied responds differently to the light spectrums.

Effects of Lead Pollution on Photosynthetic Characteristics and Chlorophyll Fluorescence Parameters of Different Populations of Miscanthus floridulus

This study was conducted in order to study the effect of different concentrations of lead pollution on the photosynthetic characteristics and growth of Miscanthus floridulus, and to reveal its photosynthetic adaptability to lead stress. The differences of gas exchange parameters, chlorophyll fluorescence characteristics and photosynthetic pigment of two Miscanthus floridulus populations, one population from Boluo an uncontaminated site, and another population from Dabaoshan, a mine site, were compared and studied through nutrient solution culture experiments treated with heavy metal lead (Pb) in green house. The results showed that (1) under Pb stress, the net photosynthetic rate (Pn), transpiration rate (Tr), stomatal conductance (Gs), intercellular carbon dioxide concentration (Ci), and chlorophyll content (Chl) of the leaves of the two populations decreased in different amplitude. Under moderate and severe Pb stress (80 mg·L−1, 120 mg·L−1, 240 mg·L−1), the plant biomass of non-mining population and mining population plants were 54.5%, 39.7%, 29.4% and 70.4%, 54.7%, 50.9% of the control, respectively. (2) Stomatal restriction was the main factor for the Pn’s decrease in the leaves of the non-mine population under light Pb stress, while the non-stomatal restriction was the main factor for Pn’s decrease in the leaves of the non-mine population under middle and high Pb stresses. (3) Under Pb stress, the maximum photochemical efficiency (Fv/Fm) and potential activity (Fv/Fo) of PS II reaction centers in the two populations of M. floridulus decreased. However, Fv/Fm and Fv/Fo showed a smaller decrease, but the capability to utilize light and the potential to activate PSII of the mine population remained higher than that of the non-mine population. The changes of photochemical quenching coefficient (qP) and non-photochemical quenching coefficient (NPQ) of PSⅡ showed that qP value decreased and NPQ value increased in the two populations under Pb stress. On the whole, the resistance mining area population had a low qP reduction and a large increase in NPQ. Electronic transfer rate (ETR) and PSII actual fluorescence efficiency (ΦPSII) of the mine population changed slightly under Pb stress. These results indicated that when under Pb stress, the electron transport activity and photosynthetic apparatus were damaged less in the mine population with high resistance than in the non-mine population with low resistance. Conclusion: the mining population of M. floridulus has strong tolerance to Pb, which is suitable for the pioneer species of gramineae in vegetation restoration construction in metal mining area.

Photosynthetic and Antioxidant Responses of Gymnocarpos przewalskii to Simulated Rainfall Changes

Gymnocarpos przewalskii is a rare Tertiary relict species, mainly distributed in desert areas of northwestern China. Changes in rainfall have a significant impact on the physiological characteristics of desert plants. In the present study, the effects of five simulated rainfall levels on the gas exchange parameters, chlorophyll fluorescence characteristics, and antioxidant system of G. przewalskii were studied. The results show that with increased rainfall the net photosynthetic rate (Pn) and transpiration rate increase significantly. The reduction in Pn is caused by stomatal and non-stomatal limitations under decreased rainfall. Decreased rainfall markedly improves the instantaneous water-use efficiency of leaves. With increased rainfall, the maximum photosynthetic rate, apparent quantum efficiency, and light utilization range significantly increase. Under reduced rainfall, the chlorophyll content, maximum photochemical efficiency of PSII, and steady-state optical quantum efficiency decrease and photoinhibition is caused in the PSII system. A rainfall reduction of 30% leads to massive production of superoxide anions and hydrogen peroxide, causing obvious peroxidation damage. Meanwhile, superoxide dismutase, peroxidase, and catalase in the leaves are significantly enhanced to remove excess reactive oxygen species and alleviate the injury to photosynthetic apparatus. Our study reveals the effect of rainfall changes on the photosynthetic characteristics and antioxidant system of G. przewalskii, and can improve understanding of the adaptive strategies of desert plants under future precipitation changes in northwestern China.

Natural variations of chlorophyll fluorescence and ion transporter genes influenced the differential response of japonica rice germplasm with different salt tolerances.

Soil salinity seriously restricts rice growth, development, and production globally. Chlorophyll fluorescence and ion content reflect the level of injury and resistance of rice under salt stress. To understand the differences in the response mechanisms of japonica rice with varying degrees of salt tolerance, we analyzed the chlorophyll fluorescence characteristics and ion homeostasis of 12 japonica rice germplasm accessions by comprehensive evaluation of phenotype, haplotype, and expression of salt tolerance-related genes. The results revealed that salt-sensitive accessions were rapidly affected by the damage due to salinity. Salt tolerance score (STS) and relative chlorophyll relative content (RSPAD) were extremely significantly reduced (p<0.01), and chlorophyll fluorescence and ion homeostasis were influenced by various degrees under salt stress. The STS, RSPAD, and five chlorophyll fluorescence parameters of salt-tolerant accessions (STA) were significantly higher than that of salt-sensitive accessions (SSA). Principal component analysis (PCA) with 13 indices suggested three principal components (PCs), with a cumulative contribution rate of 90.254%, which were used to screen Huangluo (typical salt-tolerant germplasm) and Shanfuliya (typical salt-sensitive germplasm) based on the comprehensive evaluation D-value (DCI ). The expression characteristics of chlorophyll fluorescence genes (OsABCI7 and OsHCF222) and ion transporter protein genes (OsHKT1;5, OsHKT2;1, OsHAK21, OsAKT2, OsNHX1, and OsSOS1) were analyzed. The expressions of these genes were higher in Huangluo than in Shanfuliya under salt stress. Haplotype analysis revealed four key variations associated with salt tolerance, including an SNP (+1605 bp) within OsABCI7 exon, an SSR (-1231 bp) within OsHAK21 promoter, an indel site at OsNHX1 promoter (-822 bp), and an SNP (-1866 bp) within OsAKT2 promoter. Variation in OsABCI7 protein structure and differential expression of these three ion-transporter genes may contribute to the differential response of japonica rice to salt stress.

Optimized border irrigation delays winter wheat flag leaf senescence and promotes grain filling.

Border irrigation is still the main irrigation method in the Huang-Huai-Hai Plain of China (HPC), and the suitable irrigation border length for water saving and high yield under traditional irrigation is still unclear. Therefore, a 2-year traditional border irrigation experiment (2017-2019) was conducted on the HPC. Four border lengths were tested: 20m (L20), 30m (L30), 40m (L40), and 50m (L50). These treatments were given supplementary irrigation at jointing and anthesis. An exclusively rainfed condition formed the control treatment. Compared with other treatments, the activities of superoxide dismutase antioxidant and sucrose phosphate synthetase, and the contents of sucrose and soluble proteins after anthesis were higher in the L40 and L50 treatments, while the content of malondialdehyde content was lower. Therefore, the L40 treatment effectively delayed the decrease in the soil plant analysis development (SPAD) value and chlorophyll fluorescence characteristics, promoted grain filling, and achieved the highest thousand-grain weight. Compared with the L40 treatment, the grain yields of the L20 and L30 treatment were significantly reduced, while the water productivity of the L50 treatment was significantly reduced. These findings suggest that 40m was the optimal border length for both high yield and water saving in this experiment. This study provides a simple and low-cost water-saving irrigation method for winter wheat in the HPC under traditional irrigation, which can help alleviate the pressure of agricultural water use.

Effects of Heat Stress during Seed Filling Stage on Brassica napus Seed Oil Accumulation and Chlorophyll Fluorescence Characteristics

As global temperature rise, the threat of heat stress to rapeseed production is becoming more obvious. Exploring the response characteristics of two important biological pathways, oil accumulation and photosynthesis, to heat stress during B. napus seed filling is helpful in the genetic improvement of heat-tolerant rapeseed. The effects of heat stress on seed oil accumulation and chlorophyll fluorescence characteristics of 29 B. napus germplasms with different oil content and environmental sensitivity, including 6 rapeseed varieties which exhibited environment-sensitive/insensitive and with high, medium or low oil content, were tested by whole plant heat stress or the in vitro silique culture system. Both assay exhibited similar trend on oil content of the rapeseed germplasms. The heat effect on the chlorophyll fluorescence kinetic parameters Fv/Fm, ETR and Y(II) were also consistent. Heat stress significantly decreased oil content, although there was abundant genetic variation on heat tolerance among the genotypes. Correlation analysis showed that the decrease rate of Fv/Fm of silique heat-stressed B. napus developing seed was positive correlative to the decrease rate of mature seed oil content of the whole plant heat-stressed rapeseed (R = 0.9214, P-value < 0.01). Overall, the results indicated that heat stress inhibited oil accumulation and photosynthesis in B. napus developing seed. The decrease rate of chlorophyll fluorescence parameter Fv/Fm of heat-stressed developing seed could be used as the index of heat tolerant rapeseed identification. Further, two heat insensitive rapeseed varieties with high oil content were identified.

Peanut is among the main oil crops in India with huge economic importance. The unpredictable rainy season during the growing time of peanuts causes waterlogging in peanut fields. Waterlogging triggers major environmental limitations that negatively affect the growth, physiology, and development of peanuts. Thus, the export and production of peanuts are severely affected by waterlogging. Therefore, the understanding of metabolic mechanisms under waterlogging is important to future water-stress tolerance breeding in peanuts. This study aimed to evaluate how peanuts responded to various waterlogging conditions in terms of their development, metabolic processes, and chlorophyll fluorescence characteristics. The evaluations were carried out at different stages of peanut variety DH-86 treated with waterlogging. The peanut plants were subjected to different waterlogging periods of 20, 40, 60, 80, and 100 days. The growth parameters including total dry mass, total leaf area, and total leaves number were calculated in all treatments. The phenomenological and specific energy fluxes and maximum photosystem II efficiency (FV/Fm) were also determined. The measurements were done statistically using PCA, G-Means clustering, and correlation analysis to explore the interaction between different physiological parameters. The waterlogging for 100 days caused a significant reduction in the total number of leaves, dry mass, and total leaf area. The most sensitive parameters are specific and phenomenological energy fluxes and Fv/Fm, which notably decreased as waterlogging duration increased. The results indicated the growth and physiological performance of the peanut cv. DH-86 was affected significantly due to waterlogging and the interaction between all these parameters in waterlogging. This research focused on how peanuts respond to waterlogging stress and provides the basis for future plant breeding efforts to improve peanut waterlogging tolerance, especially in rainy regions. This will improve the sustainability of the entire peanut industry.
