Evaluation of deep eutectic solvents chiral selectors based on lactobionic acid in capillary electrophoresis.

Recently, deep eutectic solvents (DESs) have attracted considerable interest in analytical chemistry. This work described the enantioseparations of twenty amino alcohol drugs with several DESs based on lactobionic acid (LA) as the sole chiral selector in capillary electrophoresis (CE) firstly. Compared to the single LA system and the ionic liquid/LA synergistic system, the DES system exhibited considerably improved separations. The influences of some key parameters on separations were investigated in detail. This work also experimentally demonstrated that the carboxyl group was indispensable in the process of chiral recognition. The mechanisms of the improvements of DESs on enantioseparations were studied via ultraviolet spectroscopy. Furthermore, the proposed method was used to determine the enantiomeric purity of propranolol hydrochloride successfully. This is the first time that chiral DESs were utilized as the sole chiral selectors in CE, and this strategy has opened up a new prospect for the use of DESs in enantioseparation.

Novel supramolecular deep eutectic solvent (SUPRADES) as a sole chiral selector in capillary electrophoresis for the enantiomeric separation of fluorine-substituted amphetamine analogs

In this study, a novel supramolecular deep eutectic solvent consisting of sulfated-β-CD and citric acid (S-β-CD-CA) is reported for the first time. This innovative system was evaluated as a sole chiral selector in capillary electrophoresis for the enantioseparation of six fluorine-substituted amphetamine analogs, yielding remarkable outcomes. Baseline separations of all amphetamine analogs under study were achieved in less than 21.00 min using the S-β-CD-CA as the chiral selector. It was observed that the addition of 0.050 % v/v S-β-CD-CA into the background electrolyte resulted in the baseline separation of five out of the six fluorine-substituted amphetamine analogs, while in the case of the para-substituted amphetamine analog, 4-fluoramphetamine (4-FA), a higher percentage (0.15 % v/v) was required to achieve baseline enantioseparation. These findings emphasized the potential of this new supramolecular system in providing a class of solvents with promising chiral recognition properties.

Enhancement of chiral drugs separation by a novel adjustable gravity mediated capillary electrophoresis combined with sulfonic propyl ether β-CD polymer

A water-soluble negative sulfonic propyl ether β-CD polymer (SPE-β-CDP) to be used as chiral selector in capillary electrophoresis (CE) was polymerized. The sulfonic substitution degree of each β-CD in SPE-β-CDP was statistically homogenized. The only one negative peak in electrophoretogram with indirect ultraviolate method proved its uniformity of electrophoretic behavior. There were 7.12 sulfonic substitution in β-CD unit and 164 μmole β-CD units in each gram of SPE-β-CDP, which corresponded a molecular weight of 7000 or more. Compared with monomer, SPE-β-CDP was lower effect on electrical current of CE, indicating a high concentration of SPE-β-CDP could be added. Its separation ability was verified by 12 chiral drugs. SPE-β-CDP also showed advantages of good water solubility, easy preparation and recovery to reduce the overall cost. However, five of 12 chiral drugs were hardly to be fully separated which was normal for any kind of chiral selector. A newly adjustable gravity mediated capillary electrophoresis (AGM-CE) technology was proposed and combined with SPE-β-CDP to enhance the chiral separation efficiencies of propranolol, salbutamol, omeprazole, ofloxacin and phenoxybenzamine which were markedly improved to 3.02, 1.17, 7.63, 4.14, and 2.81, respectively. Furthermore, its gradient mode (AGMg-CE) was also used to improve resolution through utilizing the zero mobility point, at which the effective apparent mobility of one racemate was zero. Resolutions of five chiral drugs were significantly improved, especially resolution of carvedilol changed from 0.43 to 1.0. These indicated SPE-β-CDP as chiral selector, AGM-CE and AGMg-CE as new CE technologies had a great potential in chiral separation.

The potential of the use of deep eutectic solvents and amino acid-based ionic liquids to enhance the chiral discrimination ability of different chiral selectors in capillary electrophoresis

The effect of the combined use of amino acid-based ionic liquids (AAILs) and deep eutectic solvents (DESs) with either cyclodextrin- (CD) or cyclofructan- (CF) based chiral selectors for the chiral separation of amphetamine derivatives was investigated in the present study. A non-significant improvement in enantiomeric separation of target analytes was observed when AAILs were combined with either CF or CD. On the other side, a markedly improved chiral separation of enantiomers was obtained using the dual carboxymethyl-β-cyclodextrin/DES system, highlighting the existence of a synergistic effect. After the addition of 0.5% v/v of choline chloride-ethylene glycol, the resolution of the enantiomers of amphetamine, methamphetamine and 3-fluorethamphetamine, increased from 1.4, 1.1, 1.0 to 1.8, 1.8, and 1.5 min, and the analysis times increased from 19.54, 20.48, 18.71 to 35.71, 35.78 and 32.90 min, respectively. This was not the case for the CF/DES dual system, in which the separation of amphetamines worsened, indicating an antagonistic effect. In conclusion, DESs are a very promising additive in capillary electrophoresis that can improve the separation of chiral molecules in combination with CDs but not CFs.

Evaluation of kasugamycin as a chiral selector in capillary electrophoresis.

The discovery of novel chiral selectors always fascinates us. This work describes the chiral separation performances of a new chiral selector (kasugamycin, KAS) in capillary electrophoresis (CE) for six pairs of stereoisomers, including ephedrine and pseudoephedrine, quinine and quinidine, cinchonine and cinchonidine, and amlodipine, promethazine and ofloxacin enantiomers. Kasugamycin, an aminoglycoside antibiotic in agriculture, shows significant biological activity against rice blast with low toxicity. As it turns out, this new chiral selector possesses good CE compatibility and stereoselectivity towards model analytes. In this work, we systematically investigated several separation parameters including kasugamycin concentration, buffer pH, separation voltage and the composition of the buffer solution. A detailed discussion about the chiral recognition mechanism was made based on Statistical Product and Service Solution (SPSS) analysis, NMR experiments (1D and 2D) and molecular modeling. This is the first time that kasugamycin is utilized as a chiral selector in CE, and the development of new chiral selectors from agricultural or veterinary antibiotics deserves more attention.

NMR investigation of counterion binding to undecyl LL-leucinevalanate micelles

NMR spectroscopy was used to investigate the binding of cationic counterions to anionic micelles formed by undecyl LL-leucinevalaninate (und-LV). Amino acid-based surfactants like und-LV are green alternatives to commercial surfactants. Monomeric and polymeric forms of und-LV micelles have also been used as chiral selectors in capillary electrophoresis (CE) separations. The counterions investigated were Na+, the tetraethylammonium ion, L-Lysine, 1,4-diaminobutane, 1,6-diaminohexane, and cis and trans isomers of 1,2-diaminocyclohexane and 1,4-diaminocyclohexane. NMR measurements of counterion and micelle diffusion coefficients were used to calculate micelle radii and the mole fraction of counterion molecules bound to the micelles. Two-dimensional NMR experiments were used to investigate the structures of the counterion: micelle complexes. The mole fraction of bound counterions did not change with pH in solutions containing Na+ or tetraethylammonium counterions. With all other counterions, however, the mole fraction of micelle-bound counterions was higher below pH 10 when counterions had a + 1 or +2 charge, and lower above pH 10–10.5 when the counterions were neutral or zwitterionic. Changes in micelle radii with pH and two-dimensional NMR experiments suggested that L-Lysine, 1,4-diaminobutane, and 1,6-diaminohexane bound parallel to the micelle surface with their amine functional groups interacting with different surfactant monomers. In contrast, the diaminocyclohexane isomers were found to bind perpendicular to the micelle surface. The mole fractions of micelle-bound cis-1,4-diaminocyclohexane and trans-1,4-diaminocyclohexane were very similar to one another. However, with 1,2-cyclohexanediamine, at low pH the mole fraction of micelle-bound counterions was larger for the cis isomer than for the trans isomer.

The Use of Antibiotics as Chiral Selectors in Capillary Electrophoresis: A Review.

Chirality is becoming an essential issue in modern pharmaceutical research as regulatory agencies emphasize the safety and efficiency of enantiomers in drug development. The development of efficient and reliable chiral separation methods became a necessity in the last 30 years, and capillary electrophoresis (CE), due to its relatively low costs and “green” features, is attracting increased attention. Cyclodextrin (CD) and their derivatives are the most frequently used chiral selectors (CSs) in CE, however, the use of antibiotics as CSs represents an interesting alternative. Various classes of antibiotics (aminoglycosides, ansamycins, glycopeptides, lincosamides, macrolides, tetracyclines) have been used more or less successfully for the enantio-separation of pharmaceuticals. Antibiotics offer the possibility of a multitude of potential interactions (electrostatic, inclusion, hydrogen bonding, etc.) due to their chemical diversity, allowing the enantio-separation of analytes with a wide range of structural characteristics. This article aims to review the application of various classes of antibiotics in the CE enantio-separation of pharmaceuticals. Antibiotic physiochemical characteristics, variables impacting enantio-separation, advantages, and disadvantages when certain antibiotics are used as CSs in CE are also explored.

Evaluation of Poly(Glycidyl Methacrylate) Nanocoating for Chiral Separation with Glu-β-CD as Chiral Selector in Capillary Electrophoresis

In this paper, a capillary coated with poly(glycidyl methacrylate) nanoparticles (PGMA NPs) was prepared and applied to construct a capillary electrophoresis (CE) enantioseparation system with glucosyl-β-cyclodextrin (Glu-β-CD) as a chiral selector. The PGMA NP coating can be easily introduced through a simple ring-opening reaction. Two basic drugs were used as models to evaluate the separation performance of the PGMA coating. Under the optimal conditions selected, the separation of the two enantiomers was obtained.

Single Isomer N-Heterocyclic Cyclodextrin Derivatives as Chiral Selectors in Capillary Electrophoresis.

In order to better understand the chiral recognition mechanisms of positively charged cyclodextrin (CD) derivatives, the synthesis, the pKa determination by 1H nuclear magnetic resonance (NMR)-pH titration and a comparative chiral capillary electrophoretic (CE) study were performed with two series of mono-substituted cationic single isomer CDs. The first series of selectors were mono-(6-N-pyrrolidine-6-deoxy)-β-CD (PYR-β-CD), mono-(6-N-piperidine-6-deoxy)-β-CD (PIP-β-CD), mono-(6-N-morpholine-6-deoxy)-β-CD (MO-β-CD) and mono-(6-N-piperazine-6-deoxy)-β-CD (PIPA-β-CD), carrying a pH-adjustable moiety at the narrower rim of the cavity, while the second set represented by their quaternarized, permanently cationic counterparts: mono-(6-N-(N-methyl-pyrrolidine)-6-deoxy)-β-CD (MePYR-β-CD), mono-(6-N-(N-methyl-piperidine)-6-deoxy)-β-CD (MePIP-β-CD), mono-(6-N-(N-methyl-morpholine)-6-deoxy)-β-CD (MeMO-β-CD) and mono-(6-N-(4,4-N,N-dimethyl-piperazine)-β-CD (diMePIPA-β-CD). Based on pH-dependent and selector concentration-dependent comparative studies of these single isomer N-heterocyclic CDs presented herein, it can be concluded that all CDs could successfully be applied as chiral selectors for the enantiodiscrimination of several negatively charged and zwitterionic model racemates. The substituent-dependent enantiomer migration order reversal of dansylated-valine using PIP-β-CD contrary to PYP-β-CD, MO-β-CD and PIPA-β-CD was also studied by 1H- and 2D ROESY NMR experiments.

Evaluation of Poly(glycidyl methacrylate)-Coated Column for Enantioseparation with Azithromycin Lactobionate and Clindamycin Phosphate as Chiral Selectors in Capillary Electrophoresis

In this study, a poly(glycidyl methacrylate) nanoparticle (PGMA NP)-coated column system with two antibiotics as selector was constructed for enantioseparation. The PGMA NP coating inside the capillary columns could be easily introduced by a simple ring open reaction. Five basic drugs were used as the model to evaluate the enantioselectivity of Azithromycin Lactobionate (AL) and Clindamycin phosphate (CP)-based separation system. Factors that influence the chiral separation resolution were systematically investigated, such as chiral selector concentration, background electrolyte (BGE) pH, applied voltage and system temperature. Under the optimized conditions, improved enantioseparations were obtained for all enantiomers and the maximum column efficiency of model drugs could reach 120,000 plates/m. The repeatability of relative standard deviations for EOF representing run-to-run, day-to-day and column-to-column was less than 4.32%. In addition, molecular modeling with AutoDock was applied to elucidate the explored potential mechanism of AL/CP for recognizing racemic drugs.

Enantioselective Study on the Biodegradation of Verapamil and Cytalopram by Chiral Capillary Electrophoresis

Many of the currently available drugs are chiral compounds that are marketed as racemates or, to a lesser extent, in the form of one of the enantiomers since a pair of enantiomers may have different toxicological and ecotoxicological properties compared to each other. The evaluation of enantioselectivity in biodegradation processes is essential for environmental risk assessment. The objective of this research is to study the enantioselectivity in the biodegradation of two common chiral drugs, citalopram and verapamil, using highly sulphated-γ-cyclodextrin (HS-γ-CD) as chiral selector in Capillary Electrophoresis. Biodegradation experiments were performed in batch mode using a minimal salt medium inoculated with an activated sludge and supplemented with the corresponding enantiomeric mixture. The cultures were incubated at 20 °C for 28 days. Abiotic degradation of verapamil and citalopram enantiomers was also assessed. The concentration of the enantiomers of verapamil and citalopram were monitored using 0.7% and 0.1% m/v HS-γ-CD solutions as chiral selector, respectively. Separations were carried out using the complete filling technique. The results of biodegradability tests indicate that citalopram could be considered potentially persistent while verapamil is presumed to be a non-persistent compound. No evidence of enantioselectivity was observed in any of the biodegradation processes.

L-Histidinium Chiral Ionic Liquid Functionalized β-Cyclodextrin as Chiral Selector in Capillary Electrophoresis.

Nowadays, ionic liquids (ILs) functionalized cyclodextrins (CDs) have drawn increasing attention in chiral separation. Herein, a novel β-CD derivative functionalized by L-histidinium IL, mono-6-deoxy-6-L-histidinium-β-cyclodextrin chloride (L-HMCDCl), was synthesized for the first time and utilized for enantioseparation of nefopam and chlorphenamine in capillary electrophoresis. The L-HMCDCl exhibited superior enantioselectivity compared with native β-CD. The effect of some key parameters such as chiral selector concentration, buffer pH and applied voltage on the enantioseparation was investigated in detail. In the interest of the chiral discrimination mechanism and the enhanced enantioselectivity of L-HMCDCl, molecular modeling with AutoDock was employed to study the interaction, which was in good agreement with experimental results.

Cyclodextrins as mobile phase additives in open-tubular admicellar electrochromatography for achiral and chiral separations

Native cyclodextrins (α-, β- and γ-CD) which are traditional chiral selectors in capillary electrophoresis were studied in open-tubular admicellar electrochromatography (OT-AMEC) using cetyltrimethylammonium bromide (CTAB) as dynamic stationary pseudophase. The initial mobile phase was 0.2 mM CTAB in 25 mM sodium tetraborate (pH 9.2), which forms admicelles (bilayer) at the solid surface/liquid interface inside the capillary. Different molar ratios of [CD]:[CTAB], i.e., 0:1 to 2:1 for [α-CD]:[CTAB] and [β-CD]:[CTAB] and 0:1 to 20:1 for [γ-CD]:[CTAB] in the mobile phase were investigated. An increase in the CD concentration decreased the magnitude of the electroosmotic flow (EOF). This suggests that CDs increase the critical surface aggregation concentration of CTAB, reducing the amount of free CTAB available to aggregate at the solid surface/liquid interface. At a molar ratio of 1.5:1 for α-CD, 2:1 for β-CD and 20:1 for γ-CD, the EOF changed from anodic to cathodic in direction. This suggests that no admicelles were formed under these conditions. Consequently, the retention factors (k) of the tested analytes (dichlorprop, fenoprop, ibuprofen, ketoprofen, propranolol, verapamil, hexanophenone, verapamil and valerophenone) decreased until k = 0 (no admicelles or stationary pseudophase). We observed a change in the migration orders of the analytes because of the resulting change in the effective electrophoretic mobility. Finally, we achieved chiral separation of mecoprop and dichlorprop in OT-AMEC with α- or β-CD in the mobile phase, which was not possible in electrokinetic chromatography using the same α- or β-CD concentration in the background solution. The chiral separation was aided by the retention caused by the dynamic stationary pseudophase.

Tetraalkylammonium-l-tartrate ionic liquids as sole chiral selectors in capillary electrophoresis

In the field of separation science, the use of tartaric acid-based ionic liquids (TTILs) for chiral separation has not yet been reported. In this work, six novel mono- and di-tetraalkylammonium l-tartrate ILs (TAA-l-TTs) with different alkane chain length were evaluated for their potential as sole chiral selectors in capillary electrophoresis (CE). In comparison with conventional l-tartaric acid (l-TT) chiral selector, these IL-type chiral selectors exhibit remarkably improved enantioseparation performance, which is even better than most previously reported TT ester-type chiral selectors. Major factors influencing CE separation were investigated, including the background electrolyte (BGE) composition, ILs concentration, type and proportion of organic modifier, buffer pH, etc. The results show that the l-TT-based IL-type chiral selectors prefer a weak acid buffer (150 mM boric acid at pH 4.5) with a high proportion of methanol (80%-90%). The best resolutions (Rs) were obtained when the IL concentration was set at 60 mM. We further carried out a series of comparative experiments and a molecular simulation study to explore the enantiorecognition mechanism of TTILs. The result indicates that the TAA-l-TTs cannot be considered as a simple combination of TAA salt and l-TT. The dissociation degree of TTILs may be an important factor influencing the enantiorecognition process.

Single isomer cyclodextrins as chiral selectors in capillary electrophoresis.

Since decades, cyclodextrins are one of the most powerful selectors in chiral capillary electrophoresis for the enantioseparation of diverse organic compounds. This review concerns papers published over the last decade (from 2009 until nowadays), dealing with the capillary electrophoretic application of single isomer cyclodextrin derivatives in chiral separations. Following a brief overview of their synthetic approaches, the inventory of the neutral, negatively and positively charged (including both permanently ionic and pH-tunable ionizable substituents) and zwitterionic CD derivatives is presented, with insights to underlying structural aspects by NMR spectroscopy and molecular modeling. CE represents an ideal tool to study the weak, non-covalent supramolecular interactions. The published methods are reviewed in the light of enantioselectivity, enantiomer migration order and the fine-tuning of enantiodiscrimination by the substitution pattern of the single entity selector molecules, which is hardly possible for their randomly substituted counterparts. All the reviewed publications herein support that cyclodextrin-based chiral capillary electrophoresis seems to remain a popular choice in pharmaceutical and biomedical analysis.

Use of Gamithromycin as a Chiral Selector in Capillary Electrophoresis

In this short communication, we report the use of a second-generation macrolide antibiotic, gamithromycin (Gam), as a novel chiral selector for enantioseparation in capillary electrophoresis (CE). A preliminary analysis of the experiment results shows that Gam is especially suitable for the separation of chiral primary amines. Factors influencing enantioseparations were systematically investigated including the composition of the background electrolyte (BGE), concentration of Gam, the type and proportion of organic solvents, applied voltage, etc. In particular, N-Methylformamide (NMF) was successfully used as a non-aqueous solvent for Gam, and shown to be extremely effective for the separation of primaquine (PMQ) and 1-aminoindan (AMI) when used alone or mixed with other commonly used non-aqueous solvents (e.g. methanol). To our knowledge this was also the first application of NMF as a non-aqueous solvent for antibiotic chiral selectors in CE. The best separations were obtained with 100 mM Tris, 125 mM H3BO3 and 80 mM Gam in methanol/NMF (25:75) solvent for PMQ and AMI, or 80-100 mM Gam in methanol for the other model analytes. Among the analytes, the resolution (Rs) of amlodipine (AML) reached up to 15.65, which is to our knowledge the highest value ever reported in CE studies for this compound (except for using molecularly imprinted polymers technique).

Investigation of fusidic acid as a chiral selector in capillary electrophoresis

In this short communication, we report the use of a steroid antibiotic, Fusidic Acid (Fus), as a novel chiral selector for enantioseparation in capillary electrophoresis (CE). Fus can be classified into the antibiotic chiral selector family but does not belong to any of the previously reported subfamilies. Factors influencing Fus in CE enantioseparations were systematically investigated including the composition of the background electrolyte (BGE), buffer pH, type and proportion of organic modifier, concentration of Fus, etc. The best separations were obtained with 50 mM phosphate buffer (pH 8.0) containing 60–70 mM Fus and 50% methanol. A preliminary analysis on the separation mechanism showed that Fus is especially suitable for the chiral analytes containing a rigid planar structure. The development of Fus as a novel chiral selector can be considered as a valuable work not only because of its unique enantioselectivity, but also because there are a number of steroids in nature that are structurally similar to Fus. This work indicates that the screening of high-efficient chiral selectors from these steroids is an attractive and promising research area.

Synthesis, application and molecular modeling study of ionic liquid functionalized lactobionic acid, 3-methyl-1-(3-sulfopropyl)-1H-imidazol-3-ium lactobionate, as a chiral selector in capillary electrophoresis.

Recently, ionic liquids have been widely used for chiral separation. However, several papers have reported the use of ionic liquids as the sole chiral selector. In this work, a novel capillary electrophoresis method using the ionic liquid 3-methyl-1-(3-sulfopropyl)-1H-imidazol-3-ium lactobionate (MSI-LA) as a chiral selector was developed for the enantioseparation of propranolol, metoprolol, bisoprolol, esmolol, atenolol, sotalol, terbutaline and clenbuterol. Method optimization was conducted and the optimized method (40 mM borax buffer, v/v 30% methanol, pH 8.5, 160 mM MSI-LA; +10 kV voltage; capillary temperature, 15 °C; 50 mbar/5 s injection; detection wavelength, 230 nm) was successful for the baseline separation of the eight basic drugs. The LA (lactobionic acid) system was compared. The possible mechanisms of enantioseparation were investigated using molecular modeling. Both the cation and anion of MSI-LA provided interactions between the chiral selector and analytes, and finally enhanced chiral recognition capability compared with LA.

Synthesis and application of tetramethylammonium-carboxymethylated-β-cyclodextrin: A novel ionic liquid in capillary electrophoresis enantioseparation

In this study, a carboxymethylated-β-cyclodextrin-based chiral ionic liquid (CIL), tetramethylammonium-carboxymethylated-β-cyclodextrin (TMA-CM-β-CD) was successfully designed and synthesized. This cyclodextrin-based ionic liquids (ILs) was used as the sole chiral selector in capillary electrophoresis, and it is very interesting to find that the chiral separation capability can be remarkably improved when a conventional cyclodextrin chiral selector evolved into an IL chiral selector. The ionic liquid showed satisfactory separation performance towards twelve tested drugs. A series of parameters affecting the enantioseparation, such as the type and proportion of organic modifier, buffer pH, chiral selector concentration, as well as applied voltage were systematically investigated. Additionally, the molecular docking program Autodock was applied to further demonstrate the mechanism of chiral recognition and the enhanced enantioselectivity of TMA-CM-β-CD, which kept in agreement with our experimental results.

Synthesis of a chiral ionic liquid, cholinium-clindamycin phosphate, as sole chiral selector in capillary electrophoresis

However, only several papers have reported the use of chiral ILs as the sole chiral selector. In this paper, a chiral ionic liquid, cholinium-clindamycin phosphate (Ch-CP), was synthesized and employed as a sole chiral selector in capillary electrophoresis (CE). A series of parameters affecting the separation were optimized, including chiral selector concentration, buffer pH, proportion of organic modifier, as well as the applied voltage. Under the optimal conditions, compared to clindamycin phosphate (CP), the IL selector showed better enantioseparation capability and improved peak shapes for five racemic drugs. In addition, Molecular docking program Autodock was employed to elucidate the chiral recognition mechanism of Ch-CP, the computing results conformed to the experimental results.
