Sonochemical treatment of packaging materials for prolonging fresh produce shelf life

Packaging bags made of polyethylene (PE) were sonochemically coated with edible antibacterial nanoparticles of chitosan (CS). In this work, the nanoparticles (NPs) were deposited on the surface of PE packaging bags by applying sonication waves on an acetic solution of chitosan. The characterization of CS NPs and PE bags was conducted by physicochemical techniques. The results showed that the coated bags had longer freshness than the uncoated ones. Furthermore, the characterization of cucumber, mushroom, and garlic placed into coated and uncoated PE bags was conducted by monitoring various parameters such as mass loss, total soluble solids, pH, and visual inspection. The study revealed that the PE bags coated with CS NPs showed a noticeable result in extending the shelf life of fresh produce. Finally, the antibacterial activity of PE bags was evaluated against various bacterial species. Hence, the PE bags coated with CS NPs could be a promising candidate for elongating the shelf life of packaged fresh produce.

Biosynthesis and characterization of chitosan nanoparticles, determination of its antifungal and antiaflatoxigenic against Aspergillus flavus isolates

Aflatoxin is a mycotoxin generated mostly by the fungus Aspergillus flavus, which may be found in food and feed. It is a carcinogenic poison for both humans and animals. The goal of this investigation was to see if toxicogenic A. flavus strains might be found on nuts in the Kurdistan Region. A total of Fifty nut samples. The colony color of A. flavus and texture were used to identify macromorphological qualities, whereas the spore color, size, structure, conidiophore structure, and vesicle shape were used to assess micromorphological properties. Direct imaging of the UV fluorescence of A. flavus colonies on coconut agar media (CAM) was used to detect aflatoxin formation. Natural compounds, such as chitosan, have been suggested as a way to prevent fungal infection because of their extensive and well-known antibacterial action against a variety of microorganisms. The goal of this entry was to morphologically characterize and determine aflatoxigenic A. flavus isolates, characterize nanoparticles using UV–visible, X-ray diffraction (XRD), Energy-dispersive X-ray (EDX), and scanning electron microscope (SEM) analyses, and determine the formulations' in vitro antifungal activity on the treated fungus. Individual concentrations were created by altering the percentages of ingredients such chitosan solution, chitosan nanoparticles, glycerol, and canola oil. The final concentrations of chitosan solution and chitosan nanoparticles were 20% and 2% respectively. Chitosan nanoparticles completely inhibit the growth of A. flavus at 2%.

The Use of Chitosan Nanoparticles for Delivery of CpG ODN in Treatment of Allergic Balb/C Mice.

This study aims to prepare high stability chitosan nanoparticles (CNP) and examine the ability of CNP in CpG-ODN delivery when treating allergic mice model. Preparation and characterization of CNP were performed by ionic gelation, dynamic light scattering, and zeta sizer. The CNP cytotoxicity and activation ability of CpG ODN delivered with CNP were tested using a cell counting kit-8 and Quanti blue method. Allergic mice were injected intraperitoneal with 10 ug ovalbumin on day 0 and 7, and then treated with intranasal CpG ODN/CpG ODN, delivered with CNP/CNP, on the third week three times per week for three weeks. The ELISA method measured cytokine and IgE profiles in the allergic mice's plasma and spleen. CNP results have sizes 27.73 nm±3.67 dan 188.23 nm±53.47, spherical in shape and non-toxic, and did not alter the NF-κB activation of CpG ODN in RAW-blue cells. The application of CpG ODN delivered by chitosan nanoparticles shows no statistical difference between groups of IFN-γ, IL-10, and IL-13 in Balb/c mice's plasma and spleen, in contrast with IgE level. The results showed that using chitosan nanoparticles as a delivery system for CpG ODN has the potency to safely CpG ODN efficacy.

Innovative biosynthesis, artificial intelligence-based optimization, and characterization of chitosan nanoparticles by Streptomyces microflavus and their inhibitory potential against Pectobacterium carotovorum

Microbial-based strategy in nanotechnology offers economic, eco-friendly, and biosafety advantages over traditional chemical and physical protocols. The current study describes a novel biosynthesis protocol for chitosan nanoparticles (CNPs), employing a pioneer Streptomyces sp. strain NEAE-83, which exhibited a significant potential for CNPs biosynthesis. It was identified as Streptomyces microflavus strain NEAE-83 based on morphological, and physiological properties as well as the 16S rRNA sequence (GenBank accession number: MG384964). CNPs were characterized by SEM, TEM, EDXS, zeta potential, FTIR, XRD, TGA, and DSC. CNPs biosynthesis was maximized using a mathematical model, face-centered central composite design (CCFCD). The highest yield of CNPs (9.41 mg/mL) was obtained in run no. 27, using an initial pH of 5.5, 1% chitosan, 40 °C, and a 12 h incubation period. Innovatively, the artificial neural network (ANN), was used for validating and predicting CNPs biosynthesis based on the trials data of CCFCD. Despite the high precision degree of both models, ANN was supreme in the prediction of CNPs biosynthesis compared to CCFCD. ANN had a higher prediction efficacy and, lower error values (RMSE, MDA, and SSE). CNPs biosynthesized by Streptomyces microflavus strain NEAE-83 showed in-vitro antibacterial activity against Pectobacterium carotovorum, which causes the potato soft rot. These results suggested its potential application for controlling the destructive potato soft rot diseases. This is the first report on the biosynthesis of CNPs using a newly isolated; Streptomyces microflavus strain NEAE-83 as an eco-friendly approach and optimization of the biosynthesis process by artificial intelligence.

The targeted delivery of chitosan nanoparticles to treat indoxacarb: induced lung fibrosis in rats

BackgroundThis study evaluated the effects of chitosan nanoparticles (Ch-NPs) on indoxacarb (INDOX)-induced pulmonary fibrosis in in vivo and in vitro models. In in vivo studies, 40 male albino rats were randomly divided into four groups (10 rats/group): Group 1, normal control; Group 2, INDOX (600 mg/kg b.w.); Group 3, Ch-NPs (2 mg/kg b.w.); and Group 4, Ch-NPs + INDOX. Characterization of Ch-NPs was done measuring dynamic light scattering, zeta potential, Fourier-transform infrared spectroscopy, transmission electron microscopy, and antioxidant activity studies after various Ch-NPs treatments. From in vitro studies, the impact of Ch-NPs on A549 lung carcinoma cell proliferation was also examined.ResultsOur data indicated that INDOX provoked considerable lung damage as indicated by decreased antioxidant enzyme levels of superoxide dismutase and glutathione peroxidase, increased production of nitric oxide and malondialdehyde serum levels, elevated myeloperoxidase activity, increased hydroxyproline and cytokeratin-19 serum levels, and significantly upregulated matrix metallopeptidase-9 and microRNA-101 gene expression levels when compared with controls. Furthermore, histopathological and immunohistochemical investigations of cyclooxygenase-2 in the lung tissue revealed marked inflammation, severe fibrosis, and neutrophil infiltration. Critically, Ch-NPs treatment significantly reversed INDOX-induced changes in lung biochemical, histopathological, and immunohistochemical outcomes.ConclusionTherefore, Ch-NPs may function as potential therapeutic drugs for lung fibrosis owing to their antioxidant, anti-inflammatory, and antifibrotic activities with neutrophil infiltration.

Fabrication and characterization of chitosan nanoparticles using the coffee-ring effect for photodynamic therapy.

Biocompatible nanoparticles have been increasingly used in a variety of medical applications, including photodynamic therapy. Although the impact of synthesis parameters and purification methods is reported in previous studies, it is still challenging to produce a reliable protocol for the fabrication, purification, and characterization of nanoparticles in the 200-300 nm range that are highly monodisperse for biomedical applications. We investigated the synthesis of chitosan nanoparticles in the 200-300 nm range by evaluating the chitosan to sodium tripolyphosphate (TPP) mass ratio and acetic acid concentration of the chitosan solution. Chitosan nanoparticles were also crosslinked to rose bengal and incubated with human breast cancer cells (MCF-7) to test photodynamic activity using a green laser (λ = 532 nm, power = 90 mW). We established a simple protocol to fabricate and purify biocompatible nanoparticles with the most frequent size occurring between 200 and 300 nm. This was achieved using a chitosan to TPP mass ratio of 5:1 in 1% v/v acetic acid at a pH of 5.5. The protocol involved the formation of nanoparticle coffee rings that showed the particle shape to be spherical in the first approximation. Photodynamic treatment with rose bengal-nanoparticles killed ~98% of cancer cells. A simple protocol was established to prepare and purify spherical and biocompatible chitosan nanoparticles with a peak size of ~200 nm. These have remarkable antitumor activity when coupled with photodynamic treatment.

Fungus mediated biogenic synthesis and characterization of chitosan nanoparticles and its combine effect with Trichoderma asperellum against Fusarium oxysporum, Sclerotium rolfsii and Rhizoctonia solani

In the present study, chitosan nanoparticles were synthesized from four different fungal sources viz., Fusarium oxysporum, Metarhizium anisopliae, Beauveria bassiana and Trichoderma viride and from one commercial source of chitosan.UV–VIS spectroscopy study of synthesized nanoparticles showed absorption peaks at the range of 310.02 to 342.00 nm. Functional groups of chitosan nanoparticle as OH, N–H, C–H, C=O, C–O, C–N and P=O were confirmed by FTIR study. Surface properties of the synthesized nanoparticles were found to be positively charged and were found stable in nature. Electron microscopy study showed that synthesized nanoparticles are nearly spherical in shape. DLS analysis showed the average size of the chitosan nanoparticles synthesized from F. oxysporum, M. anisopliae, B. bassiana, T. viride and commercial products as 273.20, 172.50, 78.36, 89.03 and 300.10 nm respectively. Compatibility study of chitosan nanoparticles with T. asperellum showed highly compatible reaction with chitosan nanoparticles at the tested concentration. In vitro assay against Fusarium oxysporum, Rhizoctonia solani and Sclerotium rolfsii showed combination of T. asperellum and chitosan nanoparticle as superior in suppression of mycelial growth of the pathogens as compared to Trichoderma alone and carbendazim @0.1%.

Alternative methods for the pilot-scale production and characterization of chitosan nanoparticles.

This work describes the production/characterization of low molar mass chitosan nanoparticles derived from waste shrimp shells (SSC), as well as from a commercial chitosan (CC). The production of low molar mass nanochitosan employed thermal shock, alternating between 100 °C and ambient temperature, followed by grinding the dry material (SSC and CC) in a ball mill, producing around 500 g of nanochitosan per batch. A highlight of the methodology employed is that it enables nanochitosan to be obtained even from a low quality commercial raw material. All particles had diameters smaller than 223 nm, with an average diameter below 25 nm (determined by DLS), while reductions of molar mass were between 8.4-fold and 13.5-fold. The depolymerization process resulted in a reduction in crystallinity of 38.1 to 25.4% and 55.6 to 25.9% in the CC and SSC samples, respectively. The production of nanochitosans was also confirmed by TEM through the observation of crystalline domains with diameters between 5 and 10 nm. This work perfectly reproduces the results on bench scale from previous research. The simple and inexpensive processes enable easy scale-up, representing an important advance in the production chain of biopolymers. Graphical abstract.

Characterization of Chitosan Nanoparticles from Milkfish Scales as an Alternative Preservatives of Fresh Pangas Catfish (Pangasius hypopthalmus)

Fish is a commodities that are very easy to decompose (Highly Perishable), therefore made efforts to extend the shelf life of fish by using preservatives. Preservatives that are not harmful to health are chitosan nanoparticles. The aim of this research are to know the characterization of chitosan nanoparticles from milkfish scales and their effect as fresh pangas catfish preservative by using concentration variation 1%, 2%, 3%, 4%, and 5% and optimization of storage time 12, 24, 36 , 48, 60, and 72 hours. Design in this research is experimental laboratory. Characterization of chitosan nanoparticles using FT-IT and SEM-EDX. While the effect of fresh pangas catfish preservation on chitosan nanoparticles is based on water, ash, protein, and ALT parameters. The results obtained on the characterization of chitosan nanoparticles on the identification of functional groups (FT-IR) showed absorption bands of OH groups at a peak 3450,99 cm-1, the CN snow absorption band was seen at the peak 2360,44 cm-1, the amine band of NH amine at the peak of 1639,20 cm-1 and the crystalline alcohol CO uptake band was observed at a peak of 1096,33 cm-1. The characterization results of SEM-EDX chitosan nanoparticles are round but clumped and clearly visible particle size changes. The experimental results were analyzed with two-way ANOVA statistic showing a significant effect between concentration variation and length of storage time to extend the durability of fresh pangas catfish. The best results of chitosan nanoparticles on fresh pangas catfish is at 5% concentrations at 72 hours of storage time with water value 16,4%, ash value 0,04%, protein value 0,04%, and ALT value 4,0 x 105cfu/g.

Synthesis and characterization of chitosan nanoparticles containing teicoplanin using sol–gel

Chitosan, as a natural polysaccharide, has a unique structure and multi-functional properties. One of the most prominent specifications of chitosan is high biocompatibility, good biodegradability, low toxicity and antibacterial and antiallergenicity properties. Chitosan has a high potential for controlled drug delivery. Therefore, investigating the loading capacity and release rate of chitosan at different conditions is important. By reducing particle size, chitosan has shown a high ability of teicoplanin loading due to its cationic property, which is important in this research. The aim of this study was to investigate chitosan nanoparticle potential for use in biomedical devices for drug delivery systems. Nanoparticles were prepared by ionic gelation with tripolyphosphate (TPP) ion, and the factors that affected chitosan nanoparticle size were investigated. The prepared samples were characterized using DLS, FTIR, TGA, DSC and XRD techniques. It is found at best condition with CS/TPP ratio of 2:1 nanoparticles were obtained at an average size of about 100 nm. The results confirmed that the drug (teicoplanin) loaded on the TPP cross-linked chitosan nanoparticles causes an increase in nanochitosan size and there was no interaction between teicoplanin and chitosan. Also, it is observed that 28.2% of teicoplanin was released in the first 10 h and the release is continued gradually to receive 37.4% in 100 h. Thus, it seems that chitosan nanoparticles mitigate the drug release and are suitable for sustained drug release.

Vitamin C is a very important module related to many biochemical functions of human body. Vitamin C is highly sensitive and can be lost if exposed to high temperature, humid air, light, and alkaline pH. In recent decades, use of chitosan nanoparticles as nanocarriers has received much attention due to their biocompatibility, biodegradability, and non-toxicity. Chitosan nanoparticles containing vitamins, flavors, enzymes, and antimicrobial agents can maintain their activity. Chitosan nanoparticles can stimulate the stabilization of vitamin C and improve controlled release. This review provides a summary of the preparation and characterization of chitosan nanoparticles and potential benefits of vitamin C-containing chitosan nanoparticles. In addition, biological functions, dietary restrictions, and mechanisms of vitamin C are reviewed. Various challenges and future scope for improving the properties of vitamin C using chitosan nanoparticles are also discussed.

Encapsulation of S-nitrosoglutathione into chitosan nanoparticles improves drought tolerance of sugarcane plants

The entrapment of NO donors in nanomaterials has emerged as a strategy to protect these molecules from rapid degradation, allowing a more controlled release of NO and prolonging its effect. On the other hand, we have found beneficial effects of S-nitrosoglutathione (GSNO) – a NO donor – supplying to sugarcane plants under water deficit. Here, we hypothesized that GSNO encapsulated into nanoparticles would be more effective in attenuating the effects of water deficit on sugarcane plants as compared to the supplying of GSNO in its free form. The synthesis and characterization of chitosan nanoparticles containing GSNO were also reported. Sugarcane plants were grown in nutrient solution, and then subjected to the following treatments: control (well-hydrated); water deficit (WD); WD + GSNO sprayed in its free form (WDG) or encapsulated (WDG-NP). In general, both GSNO forms attenuated the effects of water deficit on sugarcane plants. However, the encapsulation of this donor into chitosan nanoparticles caused higher photosynthetic rates under water deficit, as compared to plants supplied with free GSNO. The root/shoot ratio was also increased when encapsulated GSNO was supplied, indicating that delayed release of NO improves drought tolerance of sugarcane plants. Our results provide experimental evidence that nanotechnology can be used for enhancing NO-induced benefits for plants under stressful conditions, alleviating the negative impact of water deficit on plant metabolism and increasing biomass allocation to root system.

Fabrication and characterization of chitosan nanoparticles and collagen-loaded polyurethane nanocomposite membrane coated with heparin for atrial septal defect (ASD) closure.

Atrial septal defect (ASD) constitutes 30-40% of all congenital heart diseases in adults. The most common complications in the treatment of ASD are embolization of the device and thrombosis formation. In this research, an occluding patch was developed for ASD treatment using a well-known textile technology called electrospinning. For the first time, a cardiovascular occluding patch was fabricated using medical grade polyurethane (PU) loaded with bioactive agents namely chitosan nanoparticles (Cn) and collagen (Co) which is then coated with heparin (Hp). Fourier transform infrared spectrum showed characteristic vibrations of several active constituents and changes in the absorbance due to the inclusion of active ingredients in the patch. The contact angle analysis demonstrated no significant decrease in contact angle compared to the control and the composite patches. The structure of the electrospun nanocomposite (PUCnCoHp) was examined through scanning electron microscopy. A decrease in nanofiber diameter between control PU and PUCnCoHp nanocomposite was observed. Water uptake was found to be decreased for the PUCnCoHp nanocomposite against the control. The hemocompatibility properties of the PUCnCoHp ASD occluding patch was inferred through in vitro hemocompatibility tests like activated partial thromboplastin time (APTT), prothrombin time (PT) and hemolysis assay. It was found that the PT and APTT time was significantly prolonged for the fabricated PUCnCoHp ASD occluding patch compared to the control. Likewise, the hemolysis percentage was also decreased for the PUCnCoHp ASD patch against the control. In conclusion, the developed PUCnCoHp patch demonstrates potential properties to be used for ASD occlusion.

Synthesis and characterization of chitosan nanoparticles and their effect on Fusarium head blight and oxidative activity in wheat

The main aim of present study was to prepare chitosan (CS) and chitosan nanoparticles (CS/NPs) to evaluate their antifungal and oxidative activity. CS/NPs were prepared based on the ionic gelation of CS with tripolyphosphate (TPP) anions by using centrifugation and pH change. The obtained nanoparticles (NPs) were characterized by size and zeta potential analysis. The antifungal activity of the CS and CS/NPs were evaluated on the Fusarium graminearum, which causes Fusarium head blight (FHB) on wheat by the method of spraying on the Potato dextrose agar (PDA) medium. The Dynamic light scattering (DLS) indicated that particle diameter (z-average) was approximately 180.9±35.5–339.4±50.9 and 225.7±42.81–595.7±81.7nm for NPs prepared from CS with different molecular weights by using centrifugation and pH change methods, respectively. Different concentrations of CS and NPs were tested to know the inhibitory effect of F. graminearum. Low molecular weight (LMW) CS and its NPs had high potential of antifungal activity on suppress of fungus growth. The maximum percentage of growth reduction was 68.18%, and 77.5% by CS and its NPs at concentrations of 1000 and 5000ppm, respectively. In greenhouse trials, at 28days after inoculation (dpi), the area under the disease progress curve (AUDPC) from 7 dpi to 28 dpi of control plants treated with acetic acid aqueous solution and distilled water was almost up to 7.36 and 7.7, respectively, while plants treated with CS and NPs only had approximately 3.61 and 3.34, respectively. Results revealed that H2O2 accumulations displayed a different pattern during the activation of plant defense systems, it had brownish sites on the infected palea. Since 24h post inoculation (hpi), the H2O2 accumulations were shown in both CS and NPs, and the elevated H2O2 accumulation appeared in 72 hpi in both treatments. CS and NPs at high concentration increased the degree of tissue and cell injury. The obtained results clearly suggest that CS and its NPs have remarkable potential for further field screening towards crop protection.

Nitric oxide-releasing chitosan nanoparticles alleviate the effects of salt stress in maize plants

Nitric oxide (NO) is a signaling molecule involved in plant response to various abiotic stresses. However, the application of NO donors in agriculture is hampered by the instability of these compounds. Despite the successful uses of NO-releasing nanoparticles for biomedical purposes and the variety of nanomaterials developed as carrier systems of agrochemicals, the potential applications of nanocarriers for NO delivery in plants have not yet been tested. Herein, we report the synthesis and characterization of chitosan nanoparticles (CS NPs) containing the NO donor S-nitroso-mercaptosuccinic acid (S-nitroso-MSA). The efficiency of these NO-releasing NPs in mitigating the deleterious effects of salinity on maize plants was compared to that of the non-encapsulated NO donor. The NPs were synthesized through ionotropic gelation process, and mercaptosuccinic acid (MSA), the NO donor precursor, was encapsulated into CS NPs (91.07% encapsulation efficiency). Free thiol groups of MSA-CS NPs were nitrosated, leading to S-nitroso-MSA-CS NPs (NO-releasing NPs). The incorporation of S-nitroso-MSA into CS NPs allowed a sustained NO release. Treatments of salt-stressed maize plants with S-nitroso-MSA-CS NPs resulted in a higher leaf S-nitrosothiols content compared to that of free S-nitroso-MSA. Moreover, S-nitroso-MSA-CS NPs were more efficient than was the free NO donor in the amelioration of the deleterious effects of salinity in photosystem II activity, chlorophyll content and growth of maize plants because the protective action of the nanoencapsulated S-nitroso-MSA was achieved at lower dosages. Overall, these results demonstrate the positive impact of S-nitroso-MSA nanoencapsulation in increasing NO bioactivity in maize plants under salt stress.

Natural extracts into chitosan nanocarriers for rosmarinic acid drug delivery

Context: Nanotechnology can be applied to deliver and protect antioxidants in order to control the oxidative stress phenomena in several chronic pathologies. Chitosan (CS) nanoparticles are biodegradable carriers that may protect antioxidants with potent biological activity such as rosmarinic acid (RA) in Salvia officinalis (sage) and Satureja montana (savory) extracts for safe and innovative therapies.Objective: Development and characterization of CS nanoparticles as a stable and protective vehicle to deliver RA for medical applications using natural extracts as sage and savory.Materials and methods: Antioxidant-CS based nanoparticles were prepared by ionic gelation with sodium tripolyphosphate (TPP), at pH 5.8 with a mass ratio of 7:1 (CS:TPP), with a theoretical antioxidant-CS loading of 40–50%. The nanoparticles were then characterized by different methods such as photon correlation spectroscopy, laser Doppler anemometry, scanning electron microscopy (SEM), differential scanning calorimetry (DSC), Fourier-transform infrared (FTIR), high-performance liquid chromatographic (HPLC), association efficiency, and antioxidant activity.Results and discussion: Individual and small sizing nanoparticles, around 300 nm, were obtained. SEM confirmed smooth and spherical nanoparticles after freeze-drying. No chemical interactions were found between antioxidants and CS, after encapsulation, by DSC and FTIR. The association efficiency was 51.2% for RA (with 40% loading) and 96.1 and 98.2% for sage and savory nanoparticles, respectively (both with 50% loading). Antioxidant activity values were higher than 0.0348 eq [Asc. Ac.] g/L/g extract and 0.4251 µmol/eq Trolox/g extract.Conclusion: The extracts under study are promising vehicles for RA drug delivery in CS nanocarriers.

The Characterization of Chitosan Nanoparticles by Raman Spectroscopy

To study the performance of chitosan nanoparticles systematically, we characterized MMA, chitosan and synthesized chitosan nanoparticles by Raman spectroscopy. Through analyzing the characteristic peaks of each substance, we found MMA grafted chitosan by opening its carbon-carbon double bond. So Raman spectroscopy is a very effective way in terms of the characterization of nanomaterials .
