D2 dopamine receptors and the striatopallidal pathway modulate L-DOPA-induced dyskinesia in the mouse

L-DOPA-induced dyskinesia (LID) remains a major complication of Parkinson's disease management for which better therapies are necessary. The contribution of the striatonigral direct pathway to LID is widely acknowledged but whether the striatopallidal pathway is involved remains debated. Selective optogenetic stimulation of striatonigral axon terminals induces dyskinesia in mice rendered hemiparkinsonian with the toxin 6-hydroxydopamine (6-OHDA). Here we show that optogenetically-induced dyskinesia is increased by the D2-type dopamine receptor agonist quinpirole. Although the quinpirole effect may be mediated by D2 receptor stimulation in striatopallidal neurons, alternative mechanisms may be responsible as well. To selectively modulate the striatopallidal pathway, we selectively expressed channelrhodopsin-2 (ChR2) in D2 receptor expressing neurons by crossing D2-Cre and ChR2-flox mice. The animals were rendered hemiparkinsonian and implanted with an optic fiber at the ipsilateral external globus pallidus (GPe). Stimulation of ChR2 at striatopallidal axon terminals reduced LID and also general motility during the off L-DOPA state, without modifying the pro-motor effect of low doses of L-DOPA producing mild or no dyskinesia. Overall, the present study shows that D2-type dopamine receptors and the striatopallidal pathway modulate dyskinesia and suggest that targeting striatopallidal axon terminals at the GPe may have therapeutic potential in the management of LID.

Substantia Nigra Pars Reticulata Projections to the Pedunculopontine Nucleus Modulate Dyskinesia.

Long-term use of levodopa for Parkinson's disease (PD) treatment is often hindered by development of motor complications, including levodopa-induced dyskinesia (LID). The substantia nigra pars reticulata (SNr) and globus pallidus internal segment (GPi) are the output nuclei of the basal ganglia. Dysregulation of SNr and GPi activity contributes to PD pathophysiology and LID. The objective of this study was to determine whether direct modulation of SNr GABAergic neurons and SNr projections to the pedunculopontine nucleus (PPN) regulates PD symptoms and LID in a mouse model. We expressed Cre-recombinase activated channelrhodopsin-2 (ChR2) or halorhodopsin adeno-associated virus-2 (AAV2) vectors selectively in SNr GABAergic neurons of Vgat-IRES-Cre mice in a 6-hydroxydopamine model of PD to investigate whether direct optogenetic modulation of SNr neurons or their projections to the PPN regulates PD symptoms and LID expression. The forepaw stepping task, mouse LID rating scale, and open-field locomotion were used to assess akinesia and LID to test the effect of SNr modulation. Akinesia was improved by suppressing SNr neuron activity with halorhodopsin. LID was significantly reduced by increasing SNr neuronal activity with ChR2, which did not interfere with the antiakinetic effect of levodopa. Optical stimulation of ChR2 in SNr projections to the PPN recapitulated direct SNr stimulation. Modulation of SNr GABAergic neurons alters akinesia and LID expression in a manner consistent with the rate model of basal ganglia circuitry. Moreover, the projections from SNr to PPN likely mediate the antidyskinetic effect of increasing SNr neuronal activity, identifying a potential novel role for the PPN in LID. © 2023 The Authors. Handling a mature calcium signature through optogenetics improves the differentiation of primary murine myotubes

Calcium takes part in numerous cellular processes such as proliferation, migration, differentiation, or cell death and plays a particular role in myogenesis of skeletal muscle. Indeed, intracellular calcium signaling participates, in a non-negligeable manner, to the “on” signal of muscle differentiation from undifferentiated cells to differentiated myotubes. Therefore, this differentiation can be modulated by controlling calcium activity with electrical or optogenetic stimulation approaches. In this study, we used the optogenetic tool channelrhodopsin 2 (ChR2) to control calcium activity and to modulate skeletal muscle differentiation. Using primary cultures of mouse myotubes, we showed that ChR2 stimulation was well-adapted to control intracellular calcium activity at the single cell or whole culture scale. To modulate the calcium-dependent myotube differentiation, we used an optical stimulation protocol based on GCAMP6s-decoded spontaneous calcium activity patterns of differentiated myotubes. The optical training of myotubes increased the fusion index and their contractile ability. This study demonstrates that handling a mature calcium signature with such optogenetic tool improves the differentiation of primary murine myotubes.

Optogenetic Manipulations of Amygdala Neurons Modulate Spinal Nociceptive Processing and Behavior Under Normal Conditions and in an Arthritis Pain Model.

The amygdala is an important neural substrate for the emotional–affective dimension of pain and modulation of pain. The central nucleus (CeA) serves major amygdala output functions and receives nociceptive and affected–related information from the spino-parabrachial and lateral–basolateral amygdala (LA–BLA) networks. The CeA is a major site of extra–hypothalamic expression of corticotropin releasing factor (CRF, also known as corticotropin releasing hormone, CRH), and amygdala CRF neurons form widespread projections to target regions involved in behavioral and descending pain modulation. Here we explored the effects of modulating amygdala neurons on nociceptive processing in the spinal cord and on pain-like behaviors, using optogenetic activation or silencing of BLA to CeA projections and CeA–CRF neurons under normal conditions and in an acute pain model. Extracellular single unit recordings were made from spinal dorsal horn wide dynamic range (WDR) neurons, which respond more strongly to noxious than innocuous mechanical stimuli, in normal and arthritic adult rats (5–6 h postinduction of a kaolin/carrageenan–monoarthritis in the left knee). For optogenetic activation or silencing of CRF neurons, a Cre–inducible viral vector (DIO–AAV) encoding channelrhodopsin 2 (ChR2) or enhanced Natronomonas pharaonis halorhodopsin (eNpHR3.0) was injected stereotaxically into the right CeA of transgenic Crh–Cre rats. For optogenetic activation or silencing of BLA axon terminals in the CeA, a viral vector (AAV) encoding ChR2 or eNpHR3.0 under the control of the CaMKII promoter was injected stereotaxically into the right BLA of Sprague–Dawley rats. For wireless optical stimulation of ChR2 or eNpHR3.0 expressing CeA–CRF neurons or BLA–CeA axon terminals, an LED optic fiber was stereotaxically implanted into the right CeA. Optical activation of CeA–CRF neurons or of BLA axon terminals in the CeA increased the evoked responses of spinal WDR neurons and induced pain-like behaviors (hypersensitivity and vocalizations) under normal condition. Conversely, optical silencing of CeA–CRF neurons or of BLA axon terminals in the CeA decreased the evoked responses of spinal WDR neurons and vocalizations, but not hypersensitivity, in the arthritis pain model. These findings suggest that the amygdala can drive the activity of spinal cord neurons and pain-like behaviors under normal conditions and in a pain model.

Optogenetics Design of Mechanistically-Based Stimulation Patterns for Cardiac Defibrillation

Current rescue therapies for life-threatening arrhythmias disregard the pathological electro-anatomical substrate and base their efficacy on a generalized electrical discharge. Here, we developed an all-optical platform to examine less invasive defibrillation strategies. An ultrafast wide-field macroscope operating at 2 KHz (100 x 100 pixel) was developed to optically map action potential propagation with a red-shifted voltage sensitive dye (di-4-ANBDQPQ) in whole mouse hearts. Control of the electrical activity was achieved by employing transgenic mouse hearts expressing Channel Rhodopsin-2 (ChR2). In order to draw arbitrarily-chosen ChR2 stimulation patterns with sub-millisecond temporal resolution, the macroscope was implemented with a random-access scanning head based on acousto-optic deflectors (AODs). AODs rapidly scan the laser beam across the whole field of view exciting different volume with a commutation time of few μs. At the end of one cycle the AODs return to the initial position and repeat the stimulation cycle. Alternatively, a simpler optical solution based on digital micromirror device (DMD) in combination with a high power LED was used to manipulate light positioning in a real simultaneous manner. We employed the macroscope to study the mechanistic features of ventricular tachycardia and we designed mechanistically-based cardioversion/defibrillation patterns exploiting the transient refractoriness of myocardium produced by the ChR2 stimulation. Multiple regions of conduction block revealed to efficiently defibrillate arrhythmic hearts but with lower energy requirements as compared to whole ventricle interventions. To confirm that the cardioversion efficiency is rigorously dependent on the mechanistic-based design, we positioned multiple regions of conduction block regardless of the re-entry arrhythmic wavefront obtaining a dramatically reduced cardioversion rate. In conclusion, this work demonstrates that defibrillation energies can be substantially reduced by applying discrete stimulation patterns and promotes the investigation of new anti-arrhythmic strategies.

Network analysis of mesoscale optical recordings to assess regional, functional connectivity.

With modern optical imaging methods, it is possible to map structural and functional connectivity. Optical imaging studies that aim to describe large-scale neural connectivity often need to handle large and complex datasets. In order to interpret these datasets, new methods for analyzing structural and functional connectivity are being developed. Recently, network analysis, based on graph theory, has been used to describe and quantify brain connectivity in both experimental and clinical studies. We outline how to apply regional, functional network analysis to mesoscale optical imaging using voltage-sensitive-dye imaging and channelrhodopsin-2 stimulation in a mouse model. We include links to sample datasets and an analysis script. The analyses we employ can be applied to other types of fluorescence wide-field imaging, including genetically encoded calcium indicators, to assess network properties. We discuss the benefits and limitations of using network analysis for interpreting optical imaging data and define network properties that may be used to compare across preparations or other manipulations such as animal models of disease.

The effects of optical, electrical and chemical stimulation on serotonin release from median raphe and hippocampus of mice

The present study has examined several characteristics of the release of [3H]5-HT from the median raphe nucleus (MRN) and hippocampus in terms of its dependence of nerve impulse. We used electrical stimulation and the sodium channel opener veratridine, which excite all of the neuronal processes in the stimulation field, and optogenetics to selectively stimulate those terminals which express channelrhodopsin-2 (ChR2) and compared 5-HT release evoked by electrical and chemical depolarization and by light. We injected an adeno-associated virus containing DNA construct encoding ChR2 into the MRN of mice and investigated [3H]5-HT release from MRN and hippocampal slices. Serotonergic nerve terminals was locally stimulated with 473 nm light (blue laser diode) and electrically by bipolar electrode and by veratridine and transmitter release was monitored by collecting the effluent in a fraction collector. Electrical filed stimulation and veratridine resulted in a significantly increase in the efflux of 5-HT, whereas optical stimulation of ChR2 expressing nerve terminals at various frequencies (10, 20, 50, 100 Hz) elicited only a negligible increase in 5-HT release either from the hippocampus or from the MRN itself. The electrically induced release of radioactive neurotransmitter was completely inhibited by perfusion with tetrodotoxin. We have also applied the 5-HT transporter inhibitor, fluoxetine and the GABAA blocker bicuculline to relieve released 5-HT from re-uptake and any endogenous inhibition. Nevertheless, the effect of optical stimulation remained closed to the detection limit under these condition. In conclusion, whereas our method is suitable to detect [3H]5-HT efflux in response to ongoing neuronal sodium channel activity its sensitivity is too low to detect transmitter efflux evoked by focal optogenetic stimulation. The most likely reason for the failure of detection of 5-HT efflux is that ChR2 is expressed only by a small subpopulation of nerve terminals.

Optogenetic control of nerve growth.

Due to the limited regenerative ability of neural tissue, a diverse set of biochemical and biophysical cues for increasing nerve growth has been investigated, including neurotrophic factors, topography, and electrical stimulation. In this report, we explore optogenetic control of neurite growth as a cell-specific alternative to electrical stimulation. By investigating a broad range of optical stimulation parameters on dorsal root ganglia (DRGs) expressing channelrhodopsin 2 (ChR2), we identified conditions that enhance neurite outgrowth by three-fold as compared to unstimulated or wild-type (WT) controls. Furthermore, optogenetic stimulation of ChR2 expressing DRGs induces directional outgrowth in WT DRGs co-cultured within a 10 mm vicinity of the optically sensitive ganglia. This observed enhancement and polarization of neurite growth was accompanied by an increased expression of neural growth and brain derived neurotrophic factors (NGF, BDNF). This work highlights the potential for implementing optogenetics to drive nerve growth in specific cell populations.

Study of the Spatial Correlation Between Neuronal Activity and BOLD fMRI Responses Evoked by Sensory and Channelrhodopsin-2 Stimulation in the Rat Somatosensory Cortex

In this work, we combined optogenetic tools with high-resolution blood oxygenation level-dependent functional MRI (BOLD fMRI), electrophysiology, and optical imaging of cerebral blood flow (CBF), to study the spatial correlation between the hemodynamic responses and neuronal activity. We first investigated the spatial and temporal characteristics of BOLD fMRI and the underlying neuronal responses evoked by sensory stimulations at different frequencies. The results demonstrated that under dexmedetomidine anesthesia, BOLD fMRI and neuronal activity in the rat primary somatosensory cortex (S1) have different frequency-dependency and distinct laminar activation profiles. We then found that localized activation of channelrhodopsin-2 (ChR2) expressed in neurons throughout the cortex induced neuronal responses that were confined to the light stimulation S1 region (<500 μm) with distinct laminar activation profile. However, the spatial extent of the hemodynamic responses measured by CBF and BOLD fMRI induced by both ChR2 and sensory stimulation was greater than 3 mm. These results suggest that due to the complex neurovascular coupling, it is challenging to determine specific characteristics of the underlying neuronal activity exclusively from the BOLD fMRI signals.

Optogenetic Excitation of the Pancreatic β-Cell

The β-cell is a subunit of the Islet of Langerhans, a micro-organ of the pancreas which maintains glucose homeostasis. This endocrine cell senses changes in blood glucose levels and releases the hormone insulin. An electrophysiological process governing insulin release occurs through initial uptake of blood glucose and generation of ATP which binds the K-ATP channel causing inhibition and subsequent membrane depolarization. Influx of calcium from voltage-gated channels then interact with insulin secretory vesicles to promote exocystosis into the blood stream. In order to directly influence calcium dynamics, insulin release, and to study the electrophysical properties of the β-cell we used Channelrhodopsin-2 (ChR2), a light activated cation channel capable of creating membrane depolarization with light stimulation. Transient transfection of the MIN6 β-cell line using ChR2-YFP was carried out followed by calcium imaging using Fura2-AM. Following ChR2 stimulation at 490nm we observed local depolarization events in positively transfected cells. We demonstrated the creation of new oscillatory behavior at high glucose levels that can be set at specified frequencies and demonstrate that ChR2 can be used to overcome disruption in the K-ATP channel. Next, we isolated intact islets from a mouse model with β-cell specific expression of ChR2-YFP. We demonstrated global depolarization events upon ChR2 stimulation and performed insulin secretion assays to demonstrate enhanced insulin secretion. In addition, a computational multi-cellular islet model including ChR2 was used for comparison between in-vitro and in-silico experiments. This work demonstrates the ability of cell-specific electrophysiological control in the islet of Langerhans which will give further insight into β-cell behavior. Future work will include analyzing local depolarization on global islet behavior, examining the effect of ChR2 on islet-islet coupling, and developing an in-vivo model; which will include testing the feasibility for reversing diabetes.

Cortical Neurovascular Coupling Driven by Stimulation of Channelrhodopsin-2

While functional imaging is widely used in studies of the brain, how well the hemodynamic signal represents the underlying neural activity is still unclear. And there is a debate on whether hemodynamic signal is more tightly related to synaptic activity or action potentials. This study intends to address these questions by examining neurovascular coupling driven by pyramidal cells in the motor cortex of rats. Pyramidal cells in the motor cortex of rats were selectively transduced with the light sensitive cation channel channelrhodopsin-2 (ChR2). Electrophysiological recordings and optical intrinsic signal imaging were performed simultaneously and synchronously to capture the neural activity and hemodynamics induced by optical stimulation of ChR2-expressing pyramidal cells. Our results indicate that both synaptic activity (local field potential, LFP) and action potentials (multi-unit activity, MUA) are tightly related to hemodynamic signals. While LFPs in γ band are better in predicting hemodynamic signals elicited by short stimuli, MUA has better predictions to hemodynamic signals elicited by long stimuli. Our results also indicate that strong nonlinearity exists in neurovascular coupling.

Optogenetic activation during detector “dead time” enables compatible real-time fluorescence imaging

Optogenetic tools, such as channelrhodopsin2 (ChR2), have enabled the behavior of whole organisms by light-mediated manipulation of neuronal activities. Fluorescent indicators have been used to aid in the understanding of what is happening in living cells. To date, optogenetic stimulation and imaging acquisition were sequentially performed during detector “live time.” However, there is a problem with interrupting acquisition time sequences because such stimulation invades the time territory of fluorescent imaging. Here, our purpose was to show that optogenetic stimulation can be performed within the “dead time” of the charge-coupled device camera, the short interval of data transfer between frames. We show the kinetic measurement of Ca2+ dynamics in neuron-like cells upon ChR2 stimulation, by which we reveal the biphasic property of the Ca2+ increase in response to optical stimulation.

In vivo Large-Scale Cortical Mapping Using Channelrhodopsin-2 Stimulation in Transgenic Mice Reveals Asymmetric and Reciprocal Relationships between Cortical Areas

We have mapped intracortical activity in vivo independent of sensory input using arbitrary point channelrhodopsin-2 (ChR2) stimulation and regional voltage sensitive dye imaging in B6.Cg-Tg (Thy1-COP4/EYFP)18Gfng/J transgenic mice. Photostimulation of subsets of deep layer pyramidal neurons within forelimb, barrel, or visual primary sensory cortex led to downstream cortical maps that were dependent on synaptic transmission and were similar to peripheral sensory stimulation. ChR2-evoked maps confirmed homotopic connections between hemispheres and intracortical sensory and motor cortex connections. This ability of optogentically activated subpopulations of neurons to drive appropriate downstream maps suggests that mechanisms exist to allow prototypical cortical maps to self-assemble from the stimulation of neuronal subsets. Using this principle of map self-assembly, we employed ChR2 point stimulation to map connections between cortical areas that are not selectively activated by peripheral sensory stimulation or behavior. Representing the functional cortical regions as network nodes, we identified asymmetrical connection weights in individual nodes and identified the parietal association area as a network hub. Furthermore, we found that the strength of reciprocal intracortical connections between primary and secondary sensory areas are unequal, with connections from primary to secondary sensory areas being stronger than the reciprocal.

Opioid-Sensitive GABA Inputs from Rostromedial Tegmental Nucleus Synapse onto Midbrain Dopamine Neurons

Opioids increase dopamine release in the brain through inhibition of GABA-A IPSCs onto dopamine cells. Immunolabeling indicates that GABA neurons in the rostromedial tegmental nucleus (RMTg), also known as the tail of the ventral tegmental area, send a dense projection to midbrain dopamine neurons stain for μ-opioid receptors. There is however, little functional evidence that these neurons play a role in the opioid-dependent increase in dopamine neuron activity. The present study used retrograde tracers injected into the ventral tegmental area and substantia nigra (VTA/SN) to identify RMTg neurons that project to the VTA/SN. Whole-cell current-clamp and cell-attached recordings from labeled RMTg neurons were performed in sagittal slices from rat. The rhythmic spontaneous firing rate of RMTg neurons was decreased and the membrane potential was hyperpolarized in response to application of μ-opioid agonist DAMGO. Agonists that act at κ- and δ-opioid receptors (U69593 and DPDPE) failed to hyperpolarize RMTg neurons. Whole-cell recordings made in dopamine neurons revealed rhythmic, large amplitude spontaneous IPSCs that had a similar frequency, pattern and opioid sensitivity to the firing of RMTg neurons. In addition, electrical and channelrhodopsin-2 stimulation within the RMTg evoked GABA-A IPSCs in dopamine neurons that were inhibited by μ-opioid agonists DAMGO, but not κ- and δ-opioid agonists. Thus, this study demonstrates functional connection from the RMTg to the VTA/SN mediated by a dense, opioid-sensitive GABA innervation, and that the RMTg is a key structure in the μ-opioid receptor-dependent regulation of dopamine neurons.

Increases in Intracellular Calcium Triggered by Channelrhodopsin-2 Potentiate the Response of Metabotropic Glutamate Receptor mGluR7

The metabotropic glutamate receptor 7a (mGluR7a), a heptahelical Galphai/o-coupled protein, has been shown to be important for presynaptic feedback inhibition at central synapses and certain forms of long term potentiation and long term depression. The intracellular C terminus of mGluR7a interacts with calmodulin in a Ca2+-dependent manner, and calmodulin antagonists have been found to abolish presynaptic inhibition of glutamate release in neurons and mGluR7a-induced activation of G-protein-activated inwardly rectifying K+ channel (GIRK) channels in HEK293 cells. Here, we characterized the Ca2+ dependence of mGluR7a signaling in Xenopus oocytes by using channelrhodopsin-2 (ChR2), a Ca2+-permeable, light-activated ion channel for triggering Ca2+ influx, and a GIRK3.1/3.2 concatemer to monitor mGluR7a responses. Application of the agonist (S)-2-amino-4-phosphonobutanoic acid (l-AP4) (1-100 microm) caused a dose-dependent inward current in high K+ solutions due to activation of GIRK channels by G-protein betagamma subunits released from mGluR7a. Elevation of intracellular free Ca2+ by light stimulation of ChR2 markedly increased the amplitude of L-AP4 responses, and this effect was attenuated by the calcium chelator 1,2-bis(2-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid tetrakis (acetoxymethyl ester). l-AP4 responses were potentiated by submembranous [Ca2+] levels within physiological ranges and with a threshold close to resting [Ca2+]i values, as determined by recording the endogenous Xenopus Ca2+-activated chloride conductance. Together, these results show that L-AP4-dependent mGluR7a signaling is potentiated by physiological levels of [Ca2+]i, consistent with a model in which presynaptic mGluR7a acts as a coincidence detector of Ca2+ influx and glutamate release.

Escape Behavior Elicited by Single, Channelrhodopsin-2-Evoked Spikes in Zebrafish Somatosensory Neurons

Somatosensory neurons in teleosts and amphibians are sensitive to thermal, mechanical, or nociceptive stimuli [1, 2]. The two main types of such cells in zebrafish--Rohon-Beard and trigeminal neurons--have served as models for neural development [3-6], but little is known about how they encode tactile stimuli. The hindbrain networks that transduce somatosensory stimuli into a motor output encode information by using very few spikes in a small number of cells [7], but it is unclear whether activity in the primary receptor neurons is similarly efficient. To address this question, we manipulated the activity of zebrafish neurons with the light-activated cation channel, Channelrhodopsin-2 (ChR2) [8, 9]. We found that photoactivation of ChR2 in genetically defined populations of somatosensory neurons triggered escape behaviors in 24-hr-old zebrafish. Electrophysiological recordings from ChR2-positive trigeminal neurons in intact fish revealed that these cells have extremely low rates of spontaneous activity and can be induced to fire by brief pulses of blue light. Using this technique, we find that even a single action potential in a single sensory neuron was at times sufficient to evoke an escape behavior. These results establish ChR2 as a powerful tool for the manipulation of neural activity in zebrafish and reveal a degree of efficiency in coding that has not been found in primary sensory neurons.
