Male Reproduction and Health after Recovery from COVID-19 Infection

Objective. To study the reproductive hormones, semen quality, psychology, and sexual function of childbearing age men after recovery from Corona Virus Disease-2019 (COVID-19) infection. Materials and Methods. We recruited 387 men of childbearing age who recovered from COVID-19 infection and underwent prepregnancy health checks at the Reproductive Center of Shandong Maternal and Child Health Hospital from January to March 2023 as study subjects. Routine semen analysis and reproductive hormones were performed. The clinical symptoms, anxiety, depression, erectile function, and premature ejaculation questionnaire were investigated during the COVID-19 infection period. A control group of 226 men of childbearing age who were not infected with COVID-19 between August 2022 and October 2022 were selected based on questionnaire responses, reproductive hormones, and semen parameters. Results. The main clinical symptoms during COVID-19 infection of the 387 research subjects were fever (342/387, 88.37%), muscle pain (155/387, 40.05%), anosmia (124/387, 32.04%), cough (106/387, 27.39%), throat pain (53/387, 13.70%), nausea and vomiting (16/387, 4.13%), and testicular discomfort (5/387, 1.29%). The rates of anxiety and depression were 11.37% (44/387) and 32.82% (127/387), respectively; erectile dysfunction (ED) and premature ejaculation (PE) of the research subjects were accounted for 32.30% (125/387) and 6.98% (27/387). The IEF-5 score (P<0.001) decreased significantly compared to the control group, while the PHQ-9 score (P<0.001), the rate of increased ED (P<0.001), and the rate of depression increased (P<0.001). There were statistically significant differences in liquefaction time (P<0.05), the progressive sperm motility (P<0.05), total sperm motility (P<0.05), the rates of progressive sperm motility <32% (P<0.05), and total sperm motility <40% (P<0.05) between the 1 month, the 2 months after recovery from COVID-19 and controls. Circular cell counts (P<0.05) at 2 and 3 months were lower than in the controls. No changes in reproductive hormones before and after recovery from infection. Conclusion. COVID-19 infection exacerbates depression and ED. It has a sustained effect on sperm motility and liquefaction time in men. The effects of COVID-19 infection on semen persist for approximately 2 months, with recovery occurring in the third month. Reproductive hormone levels do not appear to be continuously affected after recovery from COVID-19.

Lycopene supplement mitigates polystyrene microplastics (PS-MPs)-induced reproductive alteration in rats via modulation of steroidogenic enzymes, inhibition of apoptosis and oxido-inflammatory reaction

Increasing levels of polystyrene microplastics (PS-MPs) in the environment poses huge global health threat. Prolonged exposure to PS-MPs results in both male and female infertility, with a deleterious effect on reproductive health. In rats, the disruptive effects of chemotoxicants have been countered by lycopene (LYC). This study examined the protective benefits of lycopene (LYC) against spermatological damage and hormonal alterations brought on by polystyrene microplastics (PS-MPs) in male rats. Twenty Wistar rats were split into four groups of five rats, each consisting of: the control group, the PSMPs treatment group, the PS-MPs + LYC co-treatment group, and the LYC supplementation group. Severe testicular histological damage was seen to have resulted from exposure to PS-MPs. The PS-MPs treatment dramatically reduced the antioxidant enzyme activities while raising the levels of ROS and MDA, thus reducing sperm progressive motility. Also, spermatological damage and hormonal alterations brought on by polystyrene microplastics (PS-MPs) were also prevented by the effects of lycopene (LYC). In conclusion, LYC supplementation has the ability to reduce the harmful effects of PS-MPs on the male reproductive system. Accordingly, LYC prevented PS-MPs-induced spermatological damage and reproductive hormonal changes in rats via its anti-oxidant, androgenic, anti-apoptotic, anti-inflammatory properties.

Changes in Reproductive Hormones Levels Following Administration of Hydroethanolic Extract of Azanza garckeana Fruit in Cadmium Chloride Exposed Female Wistar Rats

There have been many claims of aphrodisiac and other beneficial attributes of Azanza garckeana fruit by both tradomedicinal practitioners and some early scientific investigations; the present study, thus, set out to investigate the changes in reproductive hormones following administration of hydroethanolic extract of azanza garckeana fruit (HEAGF) in cadmium chloride exposed female Wistar rats. Thirty five (35) female Wistar rats weighing 230±10 grams were divided into 7 groups of 5 rats each. Group 1 served as negative control that received 1ml of distilled water; Group 2 served as positive control and received oral dose of 10mg/kg body weight of CdCl2 (or Cd); Group 3 had Clomiphene citrate (0.3mg/ml) + Cd; Group 4 received Vitamin E (0.3mg/ml) + Cd; Group 5 received 250mg/kg HEAGF+ Cd; Group 6 received 500mg/kg HEAGF+ Cd and Group 7received 1000mg/kg HEAGF+ Cd. All treatments were done orally and daily for 28 days. Blood samples were collected, and serum levels of reproductive hormones including estrogen, progesterone, follicle-stimulating hormone (FSH), and luteinizing hormone (LH) were measured using ELISA and spectrophotometric technique. The results showed that CdCl2 exposure significantly (P<0.05) decreased FSH level but non-significantly (P>0.05) decreased LH level while increasing oestrogen and progesterone levels compared to the negative control group. However, administration of HEAGF, especially the 250mg/kg bw dose most appreciably restored the hormonal balance towards normal levels. In conclusion, although, the findings of the present study highlights the potential therapeutic value of HEAGF in improving the adverse effects of an environmental toxin―CdCl2 on female reproductive health indicators, further studies may be necessary to elucidate the underlying mechanisms of action prior to its clinical exploration and possible recommendation for supplementation in reproductive disorders associated with a heavy metal toxicity.

Maternal intake of paracetamol during pregnancy and biomarkers of male fecundity in young adult sons

Paracetamol is suggested to have endocrine disrupting properties possibly affecting fetal programming of reproductive health that might lead to impaired semen quality and changes in reproductive hormones. In this longitudinal study, we included 1058 young adult men born 1998-2000 into the Danish National Birth Cohort with follow-up at 18-21 years of age. The exposure, maternal intake of paracetamol, was modelled in three ways: dichotomized, trimester-specific, and as duration of exposure categorized into: short (1-2 weeks), medium (3-9 weeks) or long duration (>9 weeks) vs. no intake. Outcomes included semen characteristics, self-measured testis volume, and reproductive hormone levels. We used negative binominal regression to estimate the percentage difference and 95% confidence interval (CI) for each outcome. In total, 547 (48%) sons were prenatally exposed to paracetamol due to maternal intake at least once. Maternal intake of paracetamol during pregnancy was not associated with any of the biomarkers in the dichotomized or trimester-specific exposure models. For duration of exposure, sons of mothers with long duration of maternal intake of paracetamol showed tendencies towards lower semen concentration (-14% [95% CI: -31%; 8%]), a higher proportion of nonprogressive and immotile spermatozoa (8% [95% CI: -4%; 21%]) and higher DNA Fragmentation Index (16% [95% CI: -1%; 36%]) compared to son of mothers with no intake. Maternal intake of paracetamol during pregnancy was not clearly associated with biomarkers of male fecundity in adult sons. However, it cannot be ruled out that long duration of maternal intake of paracetamol might be associated with impaired semen characteristics.

Within-person changes in reproductive hormones and cognition in the menopause transition.

Most women complain of cognitive deficits in the menopause transition, though the cause is unclear. The current study investigated the role that within-person changes in reproductive hormones, particularly estradiol, may play in triggering such perimenopausal cognitive difficulties. Participants were 43 women aged 45-55years and currently in the menopause transition. Once a week for 12weeks, participants provided a urine sample for the measurement of estrone glucuronide (E1G), a urinary metabolite of estradiol. Every three weeks across the 12-week period, participants also underwent cognitive testing, including assessments of immediate and delayed memory, attention, and executive function, and completed questionnaires assessing subjective cognitive performance. Potential confounding variables including sleep quality, vasomotor symptoms, and depressive symptoms were also assessed. Within-person E1G was positively associated with objective measures of attention, particularly the ability to passively register auditory information on the first pass, as well as subjective measures of memory, specifically relating to a lower frequency of forgetting things in everyday life. Perimenopausal women with lower estimated levels of intellectual functioning furthermore exhibited a stronger relationship between E1G changes and objective cognitive performance. While depressive mood, poor sleep, and vasomotor symptoms were all negatively associated with at least one aspect of cognitive function, the E1G-cognition relationship was not explained by these factors. This study provides evidence that validates perimenopausal women's cognitive complaints but also suggests that cognitive deficits are generally mild and transient.

The impact of reproductive hormone changes on the immune response of patients with leukemia

Background & Objective: Leukemias have been associated with various immune and endocrine changes. Several studies have established a correlation between an imbalance in pituitary hormones and sex steroids and the initiation of illness in either one or both genders. The hormone changes can affect normal physiological homogeneity of the human body, thus effecting health as well as the course of unrelated diseases. We aimed to investigate the influence of alterations in reproductive hormones on the immune response of individuals diagnosed with leukemia.
 Material and Methods: In this case-control study, a total of 130 subjects, out of which 80 patients were known cases with hematological cancer, were enrolled at the Medical City Department/Hematology Center in Baghdad, Iraq, from February-October 2022. Out of 80 patients diagnosed with leukemia, 45 patients had acute myeloid leukemia (AML), and 35 patients had acute lymphoblastic leukemia (ALL). The study involved 50 healthy controls. Estradiol (E2), triiodothyronine (T3), follicle-stimulating hormone (FSH), and progesterone levels were measured using both ELISA and the immunofluorescence assay method in all participants. C-reactive protein (CRP), ferritin, and neutrophil-to-lymphocyte ratio (NLR) was also determined.
 Results: The findings of the study demonstrated a significant elevation in the levels of CRP, ferritin (P < 0.001), and NLR (P = 0.01) in the individual’s group when compared to the control group. Serum progesterone was increased in patients diagnosed with AML and ALL in comparison to the control group (P = 0.01). FSH was elevated (P = 0.02) in AML and decreased in ALL. This difference was found to be statistically significant. The levels of T3 and E2 were found to be significantly reduced in both AML and ALL patients when compared to the control group (P = 0.1 and 0.02 respectively).

 Conclusions: The findings of the study offer empirical support for the impact of hormones on the immune response in leukemia pathogenesis.

From menarche to menopause: psychoneuroendocrinology across the female reproductive lifespan

As reproductive hormones rise and begin to fluctuate with puberty and the beginning of the menstrual cycle (i.e., menarche), there is substantially increased risk of affective illness for females compared to males. Additional risk is associated with reproductive transition events characterized by dramatic hormone changes, including the decrease in progesterone and estradiol during the premenstrual phase, the abrupt withdrawal from high pregnancy hormone levels during the postpartum period, and the turbulent hormone flux that occurs during pubertal and menopausal transitions. As such, a differential sensitivity to normal changes in reproductive hormones during specific reproductive transition events may precipitate or exacerbate affective symptoms in susceptible women. Using a model of mood disorder susceptibility in which rapid and abrupt changes in reproductive hormones may trigger the onset of affective illness in vulnerable females, this symposium will present novel findings on psychosocial and biological mechanisms underlying hormone-related, transdiagnostic affective state change spanning the female reproductive lifespan – from menarche to menopause. Following a brief introduction by the Chair, Elizabeth Andersen, the first speaker, Hannah Klusmann, will present methods and recommendations for determining ovulation status in peripubertal females within one year of menarche and how mood sensitivity to hormone flux across the menstrual cycle differs by ovulation status. Katja Schmalenberger will present recent findings on the coupling of heart rate variability suppression and the cyclical increase in mood symptoms during the midluteal menstrual cycle phase. The third speaker, Crystal Schiller, will discuss findings from three studies that used a sex steroid manipulation in women with a history of postpartum depression to determine functional neuroimaging correlates of postpartum irritability symptoms. Danielle Swales’ presentation on the interaction of life stress and estradiol on reward processing during the menopause transition will conclude the symposium.

The Effects of Chronic Asprosin Administration on Sense of Smell and Sexual Behavior in Female Rats

Introduction: Asprosin is an adipokine released from white adipose tissue during fasting and acts through the olfactory receptor. It is known that adipokines play roles in reproductive physiology in mammals. However, there are very few studies conducted on role of asprosin in reproductive functions. There are no studies on its relationship with sexual motivation. It was shown in the literature that administration of asprosin to male mice improves olfaction. It is also known that there is a strong correlation between smell and sexual desire. In view of this, it was hypothesized that chronic administration of asprosin would improve olfactory performance and increase sexual incentive motivation in female rats for male partners. Methods: This hypothesis was tested by applying the hidden cookie test, sexual incentive test, active research test, and sexual behavior test. The changes in serum hormone levels in female rats that chronically received asprosin were also measured and compared. Results: Chronic asprosin exposure increased olfactory performance, male preference ratio, male investigation preference ratio, activity index, and anogenital investigation behavior. Also, serum oxytocin and estradiol levels increased following chronic administration of asprosin in female rats. Conclusion: These data suggest that chronic administration of asprosin can result in increased sexual incentive motivation for opposite sex in female rats over increased olfactory performance and changes in reproductive hormones.

Polycystic Ovary Cyst Segmentation Using Adaptive K-means with Reptile Search Algorith

Polycystic ovary syndrome (PCOS) is a disorder in the female ovary caused because of reproductive age group hormonal changes. PCOS is a different follicle that is formed in the ovary and is termed an endocrine disorder. This disorder’s effects are often linked with clinical symptoms such as arteries, acne, hirsutism, diabetes, cardiovascular disease, and chronic infertility. It is mainly associated with type 2 diabetes, obesity with high cholesterol. This must be diagnosed at an earlier stage for avoiding other related diseases. To ensure infertility, various kinds of ovulatory failures must be significantly diagnosed and recognized. The physicians manually determine the PCOS using ultrasound images, but it is inefficient to declare whether it is a simple cyst, PCOS, or cancer cyst. This manual detection is prone to trying errors. In this paper, PCOS detection is performed through a series of processes such as preprocessing, segmentation, feature selection, and classification. The speckle noise is removed in preprocessing, and the images are enhanced for further processing. The proposed improved adaptive K-means with reptile search algorithm (IAKmeans-RSA) has been utilized for cyst segmentation and follicles recognition. The relevant features from the segmented images are extracted using a convolutional neural network (CNN). Finally, the classification is performed using the Deep Neural Network (DNN) approach. The proposed system efficiently diagnosed PCOS through cyst detection from the input images. The algorithm’s efficiency compared with existing methods shows that the proposed model is superior in segmenting and diagnosing PCOS.

IMPACT OF LIFESTYLE MODIFICATION ON THE IMPROVEMENT OF ADOLESCENT REPRODUCTIVE DISORDER WITH PCOS : A LITERATURE REVIEW

Polycystic ovarian syndrome (PCOS) is described as a symptom of disturbances in the aspects of reproduction and metabolism. Reproductive disorders usually manifest as hyperandrogenism, irregular ovulation, and polycystic ovaries, leading to decreased fertility. This was a literature review research that writes briefly, concisely and up to date on a topic from scientific publication articles. The purpose of this research was to solve the problem of reproductive disorders in young women with PCOS from the theory studied. Articles were compiled by conducting a library search through Google Scholar and Scientdirect from 2013-2022 with a total of 7 articles. Therapy for improving the body's metabolism is improved so that the function of reproductive hormones is also expected to function properly and normally, so that lifestyle changes become a support for therapy for women with PCO to reproductive hormonal changes related to successful ovulation in adolescent girls with PCOS.

Changes in women’s physical function in mid-life by reproductive age and hormones: a longitudinal study

BackgroundWhether women’s physical function in mid-life is related to their reproductive age is not known. The objectives of this study were to examine and compare changes in physical function in women by reproductive age, measured as time since final menstrual period (FMP), and chronological age, and to explore associations with repeatedly assessed levels of reproductive hormones.MethodsWe used data from 2319 UK women with up to three repeated measurements of physical function (median length of follow up: 2 years), focusing on changes occurring in women experiencing a natural menopausal transition. The main outcome was a composite physical function score that incorporated assessments of strength (grip strength), balance (one-leg stand) and cardiorespiratory fitness (timed chair rises). Associations with time since FMP, age, and time-updated measures of anti-Müllerian hormone, follicle-stimulating hormone and luteinizing hormone were assessed by multilevel models and generalised estimating equations models adjusted for the underlying effects of chronological age and confounding by education, age at first birth and smoking.ResultsThe results showed that, adjusted for these confounders, time since FMP (− 0.21 SD per 10 years, 95% CI − 0.37, − 0.06) and chronological age (− 0.31 SD per 10 years, 95% CI − 0.46, − 0.15) were inversely associated with the physical function composite score. Grip strength seemed to be the main contributor to the decline in the composite score by time since FMP. There was no strong evidence of associations between any of the three reproductive hormones and the composite score.ConclusionsPhysical function in women in mid-life declined with both chronological and reproductive age. The decline with reproductive age was independent of chronological age but did not seem to be driven by changes in reproductive hormones.

Gut microbiota: Linking nutrition and perinatal depression.

Perinatal depression is a mood disorder that is reported in women during pregnancy (prenatal) and after childbirth (postnatal). The onset of perinatal depression is associated with changes in reproductive hormones, stress hormones and neurosteroids. These chemical compounds can be modulated by the gut microbiota, which may affect maternal mental health during the perinatal period via the gut-brain-axis. Recent studies suggest that nutritional and dietary interventions (vitamin D, ω-3 fatty acids, iron, and fiber) effectively prevent or mitigate maternal depression and anxiety, but their efficacy is confounded by various factors, including the gut microbiota. Probiotics are efficacious in maintaining microbiota homeostasis, and thus, have the potential to modulate the development of perinatal mood disorders, despite no evidence in human. Therefore, clinical trials are warranted to investigate the role of probiotic supplementation in perinatal depression and behavioral changes. This article reviews the interplay between nutrition, gut microbiota and mood and cognition, and the evidence suggesting that probiotics affect the onset and development of perinatal depression.

Disruption of Sleep Continuity During the Perimenopause: Associations with Female Reproductive Hormone Profiles.

Nocturnal vasomotor symptoms (nVMS), depressive symptoms (DepSx), and female reproductive hormone changes contribute to perimenopause-associated disruption in sleep continuity. Hormonal changes underlie both nVMS and DepSx. However, their association with sleep continuity parameters resulting in perimenopause-associated sleep disruption remains unclear. We aimed to determine the association between female reproductive hormones and perimenopausal sleep discontinuity independent of nVMS and DepSx. Daily sleep and VMS diaries, and weekly serum assays of female reproductive hormones were obtained for 8 consecutive weeks in 45 perimenopausal women with mild DepSx but no primary sleep disorder. Generalized estimating equations were used to examine associations of estradiol, progesterone, and follicle stimulating hormone (FSH) with mean number of nightly awakenings, wakefulness after sleep onset (WASO) and sleep-onset latency (SOL) adjusting for nVMS and DepSx. Sleep disruption was common (median 1.5 awakenings/night, WASO 24.3 and SOL 20.0 minutes). More awakenings were associated with estradiol levels in the postmenopausal range (β = 0.14; 95% CI, 0.04 to 0.24; P = 0.007), and higher FSH levels (β [1-unit increase] = 0.12; 95% CI, 0.02 to 0.22; P = 0.02), but not with progesterone (β [1-unit increase] = -0.02; 95% CI, -0.06 to 0.01; P = 0.20) in adjusted models. Female reproductive hormones were not associated with WASO or SOL. Associations of more awakenings with lower estradiol and higher FSH levels provide support for a perimenopause-associated sleep discontinuity condition that is linked with female reproductive hormone changes, independent of nVMS and DepSx.

Microdissection Testicular Sperm Extraction Versus Multiple Needle-pass Percutaneous Testicular Sperm Aspiration in Men with Nonobstructive Azoospermia: A Randomized Clinical Trial

BackgroundSurgical extraction of testicular spermatozoa is needed in men with nonobstructive azoospermia (NOA) who wish to become biological fathers. Based on available uncontrolled studies with unspecific patient selection, microdissection testicular sperm extraction (mTESE), having a sperm retrieval rate (SRR) of 50%, is considered the most efficient sperm retrieval procedure. However, no randomized clinical trials for comparison of different sperm retrieval procedures exist. Testicular sperm aspiration (TESA) is simple and commonly used, and we hypothesized that this technique using multiple needle passes would give similar SRRs to mTESE. ObjectiveTo compare mTESE and multiple needle-pass TESA in men with NOA. Design, setting, and participantsA randomized clinical trial was performed between June 2017 and April 2021, with inclusion of 100 men with NOA from four centers in Denmark and Sweden. All participants received treatment at the same institution. InterventionParticipants were randomized to mTESE (n = 49) or multiple needle-pass TESA (n = 51). Patients with failed multiple needle-pass TESA proceeded directly to salvage mTESE. Outcome measurements and statistical analysisThe primary outcome was SRR. Secondary outcomes included complications and changes in reproductive hormones after surgery. Results and limitationsSpermatozoa were retrieved in 21/49 (43%) men after mTESE and in 11/51 (22%) men after multiple needle-pass TESA (rate difference –0.21; 95% confidence interval –0.39 to –0.03; p = 0.02). The combined SRR for multiple needle-pass TESA + salvage mTESE was 15/51 (29%). No complications occurred after multiple needle-pass TESA only, while 5/89 (6%) men having mTESE experienced a complication requiring surgical intervention. Overall, no statistically significant differences in reproductive hormones were observed between groups after 6 mo. Limitations include the low number of patients in secondary outcome data. ConclusionsIn direct comparison, SRR was higher in mTESE than in multiple needle-pass TESA. Patient summaryMen with azoospermia need surgical extraction of spermatozoa to become biological fathers. In this randomized trial, we compared two surgeries (microdissection testicular sperm extraction [mTESE] and testicular sperm aspiration [TESA]) and found that mTESE gives a higher sperm retrieval rate than multiple needle-pass TESA.

Multifactor Analysis and Intervention Study on Menstrual Disorders of Female Athletes in the Context of the Winter Olympic Games: A Case-Control Study Based on a Large Sample.

A case-control study was conducted to explore the multifactor analysis and intervention of menstrual disorders in female athletes under the background of the Winter Olympic Games, which is based on a large sample. For this purpose, from January 2020 to September 2021, 381 female athletes in long-term ice and snow sports were investigated by random sampling. All of them promoted gynecological examination and counted the incidence of menstrual disorders. The subjects were assigned into two groups according to their menstrual status: abnormal (n = 163) and normal menstrual state groups (n = 218). The basic and clinical data of the two groups were compared, and univariate analysis and multivariate logistic regression analysis were employed to explore the risk factors of menstrual disorders in female athletes. According to the random number table method, the menstrual disorder group was again assigned into the intervention group and the control group. The intervention group received health education and glucose supplement intervention to correct EAMDs, while the control group only received health education. The improvement of patients' ability balance and the changes of reproductive hormones were compared after intervention. The results of univariate analysis indicated that there exhibited no significant differences in age, menarche age, smoking history, drinking history, grade, sexual life history, abortion history, BMI, and location of household registration, but there were significant differences in family history, sleep quality, diet regularity, and mental health status (P < 0.05). The results of univariate analysis indicated that there exhibited no significant differences in age, menarche, smoking, drinking, grade, sexual life history, abortion history, family history, sleep quality, diet regularity, and mental health status. Logistic regression analysis indicated that family history of menstrual disorders, poor sleep quality, irregular diet, and mental health status all affected women's menstrual disorders (OR: 1.411, 95% CI: 1.378∼1.444; OR: 1.501, 95% CI: 1.030∼2.187; OR: 1.554, 95% CI: 1.086∼2.225; OR: 1.383, 95% CI: 1.018∼1.877, respectively) independent risk factors. According to the comparison of menstrual cycle, in the intervention group, 12 patients had menstrual cycle 21–28 days, 12 patients had menstrual cycle 28–38 days, and 58 patients were irregular and had no amenorrhea, while in the control group, 36 patients had menstrual cycle 21–28 days, 24 patients had 28–38 days, 12 patients had amenorrhea, and 11 patients had irregular menstruation, and there exhibited no significant difference (P > 0.05). There exhibited no significant difference in energy balance before and after intervention (P > 0.05); after intervention, the ability balance of the two groups was significantly promoted, and the degree of improvement in the study group was better (P < 0.05). The indexes of reproductive hormones in the follicular phase were compared before and after glucose supplement intervention, and there exhibited no significant difference before intervention (P > 0.05); after intervention, the serum LH and GnRH of the two groups decreased, while FSH and P increased. The improvement degree of the intervention group was better than that of the control group, but there exhibited no significant difference (P > 0.05). Before intervention, there exhibited no significant difference in the serum E2 level in the follicular phase (P > 0.05); after the intervention, the serum E2 of the two groups increased significantly, and the improvement of the intervention group was better (P < 0.05). Before intervention, there exhibited no significant difference in the serum E2 level in the follicular phase (P > 0.05); after the intervention, the serum E2 of the two groups increased significantly, and the improvement of the intervention group was better (P < 0.05). Female athletes have a high rate of menstrual disorders. Family history of menstrual disorders, poor sleep quality, irregular diet, and poor mental health are the main risk factors of menstrual disorders. Health education and sugar supplement intervention measures for female athletes play a positive role in the improvement of their ability balance and the regulation of reproductive hormones.

Transcriptomic Changes of Photoperiodic Response in the Hypothalamus Were Identified in Ovariectomized and Estradiol-Treated Sheep.

Accurate timing of seasonal changes is an essential ability for an animal’s survival, and the change in the photoperiod is the key factor affecting reproductive seasonality in mammals. Emerging evidence has suggested that multiple hypothalamic genes participate in the photoperiod-induced regulation of reproductive activities in sheep, but the mechanism is still unclear. In this study, we initially examined the plasma level of two major reproductive hormones, namely, follicle-stimulating hormone (FSH) and prolactin (PRL), under different photoperiods in ovariectomized and estradiol-treated (OVX + E2) sheep using radioimmunoassay (RIA). Of the two hormones, the concentration of PRL significantly increased with the extension of the photoperiod, while FSH showed the opposite trend. Subsequently, an examination of the transcriptomic variation between the short photoperiod (SP) and long photoperiod (LP) was conducted. Differential expression analyses and functional annotation showed that several key genes in the insulin secretion (VAMP2, PRKACB, PRKCG, and PLCB1), GnRH (MAPK13, CGA, CDC42, ATF4, and LHB) pathways, and circadian entrainment (KCNJ5, PER1, GNB2, MTNR1A, and RASD1), as well as numerous lncRNAs, including XR_173257.3, XR_173415.3, XR_001435315.1, XR_001024596.2, and XR_001023464.2, were shown potentially vital for the hypothalamic photoperiodic response. Four of the differentially expressed mRNAs and lncRNAs were validated by qPCR. The constructed mRNA–mRNA interaction networks further revealed that transcripts potentially participated in hypothalamic thyroid hormone synthesis, endocrine resistance, and neuroactive ligand–receptor interactions. The interactome analysis of lncRNAs and their targets implied that XR_173257.3 and its target arylalkylamine N-acetyltransferase (AANAT) and XR_173415.3 and its target TH might participate in the regulation of seasonal reproduction. Together, the changes in reproductive hormones and transcriptome will help to determine the important photoperiod-induced lncRNAs and mRNAs and provide a valuable resource for further research on reproductive seasonality in sheep.

Dynamic Changes of Reproductive Hormones in Male Minipuberty: Temporal Dissociation of Leydig and Sertoli Cell Activity.

The male hypothalamic-pituitary-gonadal (HPG) axis is transiently active during the first months of life with surging serum concentrations of reproductive hormones. This period, termed minipuberty, appears to be essential for priming testicular function. Despite the central role for male reproductive function, longitudinal data on HPG axis activation in infancy is sparse. To explore the dynamics of HPG hormone activity in healthy male infants, to assess the association of HPG axis activity and testicular volume, and to establish reference curves for serum levels of reproductive hormones. Prospective, longitudinal birth cohort (the COPENHAGEN Minipuberty Study, 2016-2018, 1-year follow-up). Population-based. Healthy, male, term, singleton newborns were followed from birth on with repeated clinical examinations including blood sampling during a 1-year follow-up. A total of 128 boys contributed to this study, while 119 participated in the postnatal follow-up. Serum reproductive hormone concentrations and testicular volume. Reproductive hormone concentrations showed marked dynamics during the first 6 months of age. Gonadotropins, total testosterone, and insulin-like factor 3 peaked at around 1 month of age. Inhibin B, anti-Müllerian hormone, and testicular volume peaked at around 4 to 5 months. Correlations largely recapitulated typical HPG axis pathways but also differed significantly from adult men. We demonstrate a temporal dissociation of Leydig and Sertoli cell activity during male minipuberty and provide reference curves for reproductive hormones.

Beneficial effects of irisin in experimental paroxetine-induced hyperprolactinemia.

It is known that selective serotonin reuptake inhibitors (SSRIs), represent an important and effective treatment of depression and other psychological disorders, these medications can increase prolactin levels mainly through activation of the serotonergic pathway. In this study, we aimed to determine the beneficial effects of irisin on paroxetine, a SSRI, induced hyperprolectinemia and in some other reproductive hormonal changes associated with hyperprolactinemia. Thirty two male Spraque-Dawley rats were used and divided into four groups including sham-operated control (vehicle), irisin (100 ng/kg/day for 28 days with mini-osmotic pumps), paroxetine (treated with 20 mg/kg paroxetine by oral gavage), irisin and paroxetine+irisin groups (n = 8). Serum prolactin (PRL), kisspeptin, luteinizing hormone (LH), follicle-stimulating hormone (FSH), testosterone and 5-alpha reductase levels were determined with enzyme-linked immunosorbent analysis (ELISA). In animals treated with paroxetine, PRL level increased and testosterone level decreased significantly (p < 0.05). Serum LH level was significantly increased in the group, but no significant changes were observed in the FSH, kisspeptin and 5-alpha reductase levels. Serum prolactin levels was significantly decreased in the group treated with irisin. While no significant difference was observed in kisspeptin, FSH and 5-alpha reductase levels, an increase in serum LH and testosterone levels with irisin administration (p < 0.05). In conclusion, chronic irisin exposure may reverse paroxetine-induced hyperprolactinemia. These results indicate that irisin may have the potential to be used as a therapeutic agent by primarily affecting paroxetine-induced increased prolactin and decreased testosterone levels.

Impact of tartrazine and curcumin on mineral status, and thyroid and reproductive hormones disruption in vivo

Endocrine disrupters (EDCs) are naturally occurring or man-made substances that either mimic or obstruct the functions of oestrogens and androgens, thyroid hormones, as well as microminerals in the body. The present work aimed to evaluate the effects of oral administration of tartrazine and curcumin, synthetic and natural dyes, respectively, on thyroid hormones (T3, T4, and TSH), female reproductive hormones (oestrogen, progesterone, LH, and FSH), and minerals (iron, copper, zinc, sodium, potassium, and chloride) in plasma, liver, and kidney of female rats after 15, 30, and 45 d of treatment. The rats were treated with admissible daily intake (ADI) and 10× ADI (9.6 and 96 mg/kg/body weight for tartrazine, 3.85 and 38.5 mg/kg/body weight for curcumin, respectively). Results showed significant changes in thyroid and female reproductive hormones, especially, in the tartrazine-treated groups as compared to the control. Low and high doses of tartrazine and curcumin significantly (p &lt; 0.05) decreased iron, copper, and zinc concentrations in plasma, whereas, the concentrations of sodium and copper in liver and kidney increased. Both tartrazine and curcumin, at ADI and 10× ADI, resulted in lower LH levels after 30 and 45 d of treatment. After 30 d, low and high dose of tartrazine significantly decreased T4, oestrogen, and FSH levels; whereas, progesterone level increased. The results demonstrated that hormone secretion and mineral content in tissues are severely affected at ADI and higher concentrations of tartrazine and curcumin. These observations suggested that lower doses of these dyes might be a safer option for their usage in foods and pharmaceuticals.

Mindful Menopause

Menopause is a part of natural aging process that causes a variety of physiological and psychological changes in the life of women. Various psycho social factors like lifestyle, health, culture, previous mood disorders all impact on menopause transition. Rapid changes in female reproductive hormones are implicated to cause fear, anxiety, depression and mood swings. However, insufficient coping abilities and poor adaptation to stress predisposes vulnerable women to emotional instability. Menopause is an unavoidable stage of life and acceptability of this fact depends on awareness, attitude, practices and health of women. Along with appropriate pharmacological treatment; lifestyle modifications and psycho-social support would go a long way to help women to steer through this phase of life with a sound mind.
