Homeostatic Regulation of Motoneuron Properties in Development.

Homeostatic plasticity represents a set of compensatory mechanisms that are engaged following a perturbation to some feature of neuronal or network function. Homeostatic mechanisms are most robustly expressed during development, a period that is replete with various perturbations such as increased cell size and the addition/removal of synaptic connections. In this review we look at numerous studies that have advanced our understanding of homeostatic plasticity by taking advantage of the accessibility of developing motoneurons. We discuss the homeostatic regulation of embryonic movements in the living chick embryo and describe the spinal compensatory mechanisms that act to recover these movements (homeostatic intrinsic plasticity) or stabilize synaptic strength (synaptic scaling). We describe the expression and triggering mechanisms of these forms of homeostatic plasticity and thereby gain an understanding of their roles in the motor system. We then illustrate how these findings can be extended to studies of developing motoneurons in other systems including the rodents, zebrafish, and fly. Furthermore, studies in developing drosophila have been critical in identifying some of the molecular signaling cascades and expression mechanisms that underlie homeostatic intrinsic membrane excitability. This powerful model organism has also been used to study a presynaptic form of homeostatic plasticity where increases or decreases in synaptic transmission are associated with compensatory changes in probability of release at the neuromuscular junction. Further, we describe studies that demonstrate homeostatic adjustments of ion channel expression following perturbations to other kinds of ion channels. Finally, we discuss work in xenopus that shows a homeostatic regulation of neurotransmitter phenotype in developing motoneurons following activity perturbations. Together, this work illustrates the importance of developing motoneurons in elucidating the mechanisms and roles of homeostatic plasticity.

The Uniform and Nonuniform Nature of Slow and Rapid Scaling in Embryonic Motoneurons.

Neurons regulate the strength of their synapses in response to a perturbation to stabilize neuronal signaling through a form of homeostatic plasticity known as synaptic scaling. The process of scaling has the potential to alter all of a cell's miniature postsynaptic current (mPSC) amplitudes by a single multiplicative factor (uniform scaling), and in doing so could change action potential-dependent or evoked synaptic strength by that factor. However, recent studies suggest that individual synapses scale with different scaling factors (nonuniform). This could complicate the simple multiplicative transform from mPSC scaling to the evoked response. We have previously identified a slow AMPAergic and GABAergic synaptic scaling in chick embryo motoneurons following 2 d in vivo perturbations inhibiting neuronal activity or GABAAR function, and now show a rapid form of scaling following NMDAR blockade in vitro Slow GABAergic scaling appeared to be of a classical uniform pattern. Alternatively, other forms of rapid and slow scaling demonstrated a uniform and nonuniform component in their mPSC amplitude distributions. Slow and rapid AMPAergic scaling was mediated by insertion of GluA2-lacking AMPA receptors. The nonuniform pattern of scaling may contribute to the observed complexity of the changes in evoked responses. Scaling-induced changes in mPSC amplitudes were not associated with changes in probability of release (Pr). Together, our results demonstrate a new rapid form of scaling in embryonic motoneurons, that slow and rapid scaling is not purely uniform, and that upscaling does not translate to an increase in evoked responses in a simple manner.SIGNIFICANCE STATEMENT Different forms of homeostatic plasticity are thought to play a critical role in maintaining neural function. For example, altering the amplitudes of spontaneous currents through a form of homeostatic plasticity known as synaptic scaling could affect evoked transmission; however, this is rarely tested. Here we demonstrate two forms of scaling and show that in many cases synaptic strength scales differently for distinct synapses within an embryonic motoneuron. These results have functional consequences for evoked synaptic strength and suggest that, like Hebbian plasticity, scaling can change relative synaptic strengths within a cell. Furthermore, our results demonstrate how different forms of homeostatic plasticity influence neuronal communication as the nascent spinal network is first established in the embryonic period.

Hypoxia-induced depression of synaptic transmission becomes irreversible by intracellular accumulation of non-excitatory amino acids

The intracellular accumulation of some amino acids (AAs), mainly glutamine, can contribute to brain edema observed during liver failure. We recently demonstrated that individual applications of high concentrations (10 mM) of some non-excitatory AAs increase the electrical resistance of hippocampal slices, indicating cell swelling. Therefore, we pondered whether an AA mixture's application might cause cell swelling at a physiological concentration range. In rat hippocampal slices, we carried out extra- and intracellular electrophysiological recordings and AAs analysis to address this question. We applied a mixture of 19 AAs at their plasmatic concentrations (Plasma solution: Ala, Gly, Gln, His, Ser, Tau, Thr, Arg, Leu, Met, Pro, Val, Asn, Cys, Phe, Ile, Lys, Tyr, and Trp). This solution was afterward divided into two according to the individual AAs at 10 mM concentration inducing synaptic potentiation (Plasma1, containing the first seven AAs of Plasma) or not (Plasma2, with the remaining AAs). Plasma application increased evoked field potentials requiring extracellular chloride. This effect was mimicked by the Plasma1 but not the Plasma2 solution. Plasma1-induced potentiation was independent of changes in release probability, basic electrophysiological membrane properties, and NMDAR activation. AAs in Plasma1 act cooperatively to accumulate intracellularly and to induce synaptic potentiation. In the presence of Plasma1, the reversible synaptic depression caused by a 40-min hypoxia period turned into an irreversible disappearance of synaptic potentials through an NMDAR-dependent mechanism. The presence of a system A transport inhibitor did not block Plasma1-mediated effects. These results indicate that cell swelling, induced by the accumulation of non-excitotoxic AAs through unidentified transporters, might foster deleterious effects produced by hypoxia-ischemia episodes.

Orexin facilitates GABAergic IPSCs via postsynaptic OX1 receptors coupling to the intracellular PKC signalling cascade in the rat cerebral cortex

Orexin has multiple physiological functions including wakefulness, appetite, nicotine intake, and nociception. The cerebral cortex receives abundant orexinergic projections and expresses both orexinergic receptor 1 (OX1R) and 2 (OX2R). However, little is known about orexinergic regulation of GABA-mediated inhibitory synaptic transmission. In the cerebral cortex, there are multiple GABAergic neural subtypes, each of which has its own morphological and physiological characteristics. Therefore, identification of presynaptic GABAergic neural subtypes is critical to understand orexinergic effects on GABAergic connections. We focused on inhibitory synapses at pyramidal neurons (PNs) from fast-spiking GABAergic neurons (FSNs) in the insular cortex by a paired whole-cell patch-clamp technique, and elucidated the mechanisms of orexin-induced IPSC regulation. We found that both orexin A and orexin B enhanced unitary IPSC (uIPSC) amplitude in FSN→PN connections without changing the paired-pulse ratio or failure rate. These effects were blocked by SB-334867, an OX1 receptor (OX1R) antagonist, but not by TCS-OX2-29, an OX2R antagonist. [Ala11, D-Leu15]-orexin B, a selective OX2R agonist, had little effect on uIPSCs. Variance-mean analysis demonstrated an increase in quantal content without a change in release probability or the number of readily releasable pools. Laser photolysis of caged GABA revealed that orexin A enhanced GABA-mediated currents in PNs. Downstream blockade of Gq/11 protein-coupled OX1Rs by IP3 receptor or protein kinase C (PKC) blockers and BAPTA injection into postsynaptic PNs diminished the orexin A-induced uIPSC enhancement. These results suggest that the orexinergic uIPSC enhancement is mediated via postsynaptic OX1Rs, which potentiate GABAA receptors through PKC activation.

Glucose and lactate as metabolic constraints on presynaptic transmission at an excitatory synapse.

Synapses have high energy demands which increase during intense activity. We show that presynaptic terminals can utilise extracellular glucose or lactate to generate energy to maintain synaptic transmission. Reducing energy substrates induces a metabolic stress: presynaptic ATP depletion impaired synaptic transmission through a reduction in the number of functional synaptic vesicle release sites and a slowing of vesicle pool replenishment, without a consistent change in release probability. Metabolic function is compromised in many pathological conditions (e.g. stroke, traumatic brain injury and neurodegeneration). Knowledge of how synaptic transmission is constrained by metabolic stress, especially during intense brain activity, will provide insights to improve cognition following pathological insults. The synapse has high energy demands, which increase during intense activity. Presynaptic ATP production depends on substrate availability and usage will increase during activity, which in turn could influence transmitter release and information transmission. We investigated transmitter release at the mouse calyx of Held synapse using glucose or lactate (10, 1 or 0mm) as the extracellular substrates while inducing metabolic stress. High-frequency stimulation (HFS) and recovery paradigms evoked trains of EPSCs monitored under voltage-clamp. Whilst postsynaptic intracellular ATP was stabilised by diffusion from the patch pipette, depletion of glucose increased EPSC depression during HFS and impaired subsequent recovery. Computational modelling of these data demonstrated a reduction in the number of functional release sites and slowed vesicle pool replenishment during metabolic stress, with little change in release probability. Directly depleting presynaptic terminal ATP impaired transmitter release in an analogous manner to glucose depletion. In the absence of glucose, presynaptic terminal metabolism could utilise lactate from the aCSF and this was blocked by inhibition of monocarboxylate transporters (MCTs). MCT inhibitors significantly suppressed transmission in low glucose, implying that lactate is a presynaptic substrate. Additionally, block of glycogenolysis accelerated synaptic transmission failure in the absence of extracellular glucose, consistent with supplemental supply of lactate by local astrocytes. We conclude that both glucose and lactate support presynaptic metabolism and that limited availability, exacerbated by high-intensity firing, constrains presynaptic ATP, impeding transmission through a reduction in functional presynaptic release sites as vesicle recycling slows when ATP levels are low.

SNAP-25 phosphorylation at Ser187 regulates synaptic facilitation and short-term plasticity in an age-dependent manner

Neurotransmitter release is mediated by the SNARE complex, but the role of its phosphorylation has scarcely been elucidated. Although PKC activators are known to facilitate synaptic transmission, there has been a heated debate on whether PKC mediates facilitation of neurotransmitter release through phosphorylation. One of the SNARE proteins, SNAP-25, is phosphorylated at the residue serine-187 by PKC, but its physiological significance has been unclear. To examine these issues, we analyzed mutant mice lacking the phosphorylation of SNAP-25 serine-187 and found that they exhibited reduced release probability and enhanced presynaptic short-term plasticity, suggesting that not only the release process, but also the dynamics of synaptic vesicles was regulated by the phosphorylation. Furthermore, it has been known that the release probability changes with development, but the precise mechanism has been unclear, and we found that developmental changes in release probability of neurotransmitters were regulated by the phosphorylation. These results indicate that SNAP-25 phosphorylation developmentally facilitates neurotransmitter release but strongly inhibits presynaptic short-term plasticity via modification of the dynamics of synaptic vesicles in presynaptic terminals.

Presynaptic D1 heteroreceptors and mGlu autoreceptors act at individual cortical release sites to modify glutamate release

The aim of this work was to study release of glutamic acid (GLU) from one-axon terminal or bouton at-a-time using cortical neurons grown in vitro to study the effect of presynaptic auto- and heteroreceptor stimulation. Neurons were infected with release reporters SypHx2 or iGluSnFR at 7 or 3 days-in-vitro (DIV) respectively. At 13–15 DIV single synaptic boutons were identified from images obtained from a confocal scanning microscope before and after field electrical stimulation. We further stimulated release by raising intracellular levels of cAMP with forskolin (10µM). Forskolin-mediated effects were dependent on protein kinase A (PKA) and did not result from an increase in endocytosis, but rather from an increase in the size of the vesicle readily releasable pool. Once iGluSnFR was confirmed as more sensitive than SypHx2, it was used to study the participation of presynaptic auto- and heteroreceptors on GLU release. Although most receptor agonizts (carbamylcholine, nicotine, dopamine D2, BDNF) did not affect electrically stimulated GLU release, a significant increase was observed in the presence of metabotropic D1/D5 heteroreceptor agonist (SKF38393 10µM) that was reversed by PKA inhibitors. Interestingly, stimulation of group II metabotropic mGLU2/3 autoreceptors (LY379268 50nM) induced a decrease in GLU release that was reversed by the specific mGLU2/3 receptor antagonist (LY341495 1µM) and also by PKA inhibitors (KT5720 200nM and PKI14-22 400nM). These changes in release probability at individual release sites suggest another level of control of the distribution of transmitter substances in cortical tissue.

Model-based prediction of maximum pool size in the ribbon synapse

The synaptic ribbon is a specialized structure in photoreceptor neurons that tethers vesicles prior to release (Figure ​(Figure1A).1A). When a cell is stimulated, vesicles are released from the ribbon and later replenished from the population of mobile vesicles in the synaptic terminal. A train of depolarizing pulses causes the ribbon to alternate between periods of release (lasting Δt = 25 ms) and replenishment (lasting T = 50ms), which occur on estimated timescales of τr = 5 ms (for release) and τa = 815 ms (for replenishment). After the first few pulses, the system approaches a limit cycle, and the amount of vesicles released on each pulse converges to a limiting value, R (Figure 1B). This can be used to determine the maximum available pool size on the ribbon, A. The standard method for estimating A is to measure the rate of replenishment in the limit, and then back-extrapolate from the cumulative release plot to obtain the available pool size at the start of the pulse train [1]. When comparing pulse trains of different strengths, this method yields substantially different values for A, a somewhat paradoxical result. Back-extrapolation assumes, however, that the replenishment rate is constant, even though it is thought to be proportional to the available space on the ribbon [2]. Figure 1 A) The synaptic ribbon. Table 1 Maximum pool size predictions from pulse train data We developed a model-based approach to estimate A from the limiting release R. We modeled the rate of release (resp. replenishment) to simply be proportional to the number of vesicles on the ribbon (resp. vacant ribbon sites), and using the measured timescale τr (resp. τa). By solving the alternating differential equations, we derived a recurrence relation for the release during each pulse, Ri, which we then solved to obtain a closed form expression for Ri and the limiting release R. Specifically, we found that A = cR, where c is a function of τr,τa,Δt,T, and p, with p a release constant that captures the stimulus dependence of release probabilities, and can be estimated from the first release, R1. In contrast to the back-extrapolation method, our model-based estimate for A was similar across stimulus types (Table 1), while p was much smaller for the weaker stimulus. This suggests that available pool size does not change with stimulus strength; instead, differences in release result from changes in release probability.

Modeling and measurement of vesicle pools at the cone ribbon synapse: Changes in release probability are solely responsible for voltage-dependent changes in release.

Postsynaptic responses are a product of quantal amplitude (Q), size of the releasable vesicle pool (N), and release probability (P). Voltage-dependent changes in presynaptic Ca(2+) entry alter postsynaptic responses primarily by changing P but have also been shown to influence N. With simultaneous whole cell recordings from cone photoreceptors and horizontal cells in tiger salamander retinal slices, we measured N and P at cone ribbon synapses by using a train of depolarizing pulses to stimulate release and deplete the pool. We developed an analytical model that calculates the total pool size contributing to release under different stimulus conditions by taking into account the prior history of release and empirically determined properties of replenishment. The model provided a formula that calculates vesicle pool size from measurements of the initial postsynaptic response and limiting rate of release evoked by a train of pulses, the fraction of release sites available for replenishment, and the time constant for replenishment. Results of the model showed that weak and strong depolarizing stimuli evoked release with differing probabilities but the same size vesicle pool. Enhancing intraterminal Ca(2+) spread by lowering Ca(2+) buffering or applying BayK8644 did not increase PSCs evoked with strong test steps, showing there is a fixed upper limit to pool size. Together, these results suggest that light-evoked changes in cone membrane potential alter synaptic release solely by changing release probability.

A pre-docking source for the power-law behavior of spontaneous quantal release: application to the analysis of LTP.

In neurons, power-law behavior with different scaling exponents has been reported at many different levels, including fluctuations in membrane potentials, synaptic transmission up to neuronal network dynamics. Unfortunately in most cases the source of this non-linear feature remains controversial. Here we have analyzed the dynamics of spontaneous quantal release at hippocampal synapses and characterized their power-law behavior. While in control conditions a fractal exponent greater than zero was rarely observed, its value was greatly increased by α-latrotoxin (α-LTX), a potent stimulator of spontaneous release, known to act at the very last step of vesicle fusion. Based on computer modeling, we confirmed that at an increase in fusion probability would unmask a pre-docking phenomenon with 1/f structure, where α estimated from the release series appears to sense the increase in release probability independently from the number of active sites. In the simplest scenario the pre-docking 1/f process could coincide with the Brownian diffusion of synaptic vesicles. Interestingly, when the effect of long-term potentiation (LTP) was tested, a ~200% long-lasting increase in quantal frequency was accompanied by a significant increase in the scaling exponent. The similarity between the action of LTP and of α-LTX suggests an increased contribution of high release probability sites following the induction of LTP. In conclusion, our results indicate that the source of the synaptic power-law behavior arises before synaptic vesicles dock to the active zone and that the fractal exponent α is capable of sensing a change in release probability independently from the number of active sites or synapses.

Nitric oxide activity-dependent regulator compensates synaptic depression and enhances metabolic efficiency in the auditory brainstem

Neuronal computations come with a certain metabolic cost. Consequently, the brain’s limited energy supply imposes constraints on its information processing capability. Neural modulators, such as nitric oxide (NO), alter neural network properties in response to activity, and consequently will likely affect energy usage as well as function. Based on experimental data of the effects of NO in the auditory brainstem, we use ac omputational model to address the question as to what extent neuronal modulation by NO could change the efficiency or the metabolic cost of neuronal transmission. We have built a three component model (pre-synaptic, post-synaptic and action potential initiation zone) of the calyx of Held and its target, the medial nucleus of the trapezoid body (MNTB) principle neuron, to estimate the metabolic efficiency in this model system. Model parameters are fit such that data from whole cell recordings is reproduced, and models are compared between a baseline condition with low levels of NO, and following conditioning with evoked activity that increases NO levels. Raised NO modulates both synaptic short term depression and cell excitability through re-balancing of K+ conductances. The synapse model exhibits short term depression, which was implemented using a standard depletion model. Differences between the unconditioned (control) and conditioned (NO) model were implemented as changes in release probability and vesicle recycling rate. The MNTB neuron is represented by as ingle compartment, Hodgkin-Huxley type model. The ionic conductances were identified from ventral cochlear nucleus bushy cells [1] and MNTB principal neurons [2]. The channel densities were set to reproduce the control and NO-modulated conditions in the MNTB neurons [3]. For each compartment, metabolic cost was scaled to match the data from synaptic activity or action potential firing during very low frequency stimulation [4] and calculated in the same way for the higher frequency activity commonly seen at this synapse. The efficiency of action potentials was calculated based on the overlap of the underpinning sodium and potassium currents and the total area of the sodium current [5]. Our results show that in the case of NO modulation, the MNTB neuron model has a higher efficiency for AP generation (58.5%) than in control conditions (49.5%). For high frequency stimulation, NO modulation very slightly increases the energy cost per vesicle released, (49.5*10 3 ATP molecules), compared to the control case (48.6*10 3 ATP molecules). Depression is less under NO, so that more vesicles are released at the end of a high frequency train than in control conditions. We have quantified the higher energetic cost implied by the higher frequency at which the principle neuron is able to fire for the NO case.

Multivesicular Release Underlies Short Term Synaptic Potentiation Independent of Release Probability Change in the Supraoptic Nucleus

Magnocellular neurons of the supraoptic nucleus receive glutamatergic excitatory inputs that regulate the firing activity and hormone release from these neurons. A strong, brief activation of these excitatory inputs induces a lingering barrage of tetrodotoxin-resistant miniature EPSCs (mEPSCs) that lasts for tens of minutes. This is known to accompany an immediate increase in large amplitude mEPSCs. However, it remains unknown how long this amplitude increase can last and whether it is simply a byproduct of greater release probability. Using in vitro patch clamp recording on acute rat brain slices, we found that a brief, high frequency stimulation (HFS) of afferents induced a potentiation of mEPSC amplitude lasting up to 20 min. This amplitude potentiation did not correlate with changes in mEPSC frequency, suggesting that it does not reflect changes in presynaptic release probability. Nonetheless, neither postsynaptic calcium chelator nor the NMDA receptor antagonist blocked the potentiation. Together with the known calcium dependency of HFS-induced potentiation of mEPSCs, our results imply that mEPSC amplitude increase requires presynaptic calcium. Further analysis showed multimodal distribution of mEPSC amplitude, suggesting that large mEPSCs were due to multivesicular glutamate release, even at late post-HFS when the frequency is no longer elevated. In conclusion, high frequency activation of excitatory synapses induces lasting multivesicular release in the SON, which is independent of changes in release probability. This represents a novel form of synaptic plasticity that may contribute to prolonged excitatory tone necessary for generation of burst firing of magnocellular neurons.

Maternal Loss of Ube3a Produces an Excitatory/Inhibitory Imbalance through Neuron Type-Specific Synaptic Defects

Angelman syndrome (AS) is a neurodevelopmental disorder caused by loss of the maternally inherited allele of UBE3A. AS model mice, which carry a maternal Ube3a null mutation (Ube3a(m-/p+)), recapitulate major features of AS in humans, including enhanced seizure susceptibility. Excitatory neurotransmission onto neocortical pyramidal neurons is diminished in Ube3a(m-/p+) mice, seemingly at odds with enhanced seizure susceptibility. We show here that inhibitory drive onto neocortical pyramidal neurons is more severely decreased in Ube3a(m-/p+) mice. This inhibitory deficit follows the loss of excitatory inputs and appears to arise from defective presynaptic vesicle cycling in multiple interneuron populations. In contrast, excitatory and inhibitory synaptic inputs onto inhibitory interneurons are largely normal. Our results indicate that there are neuron type-specific synaptic deficits in Ube3a(m-/p+) mice despite the presence of Ube3a in all neurons. These deficits result in excitatory/inhibitory imbalance at cellular and circuit levels and may contribute to seizure susceptibility in AS.

Pull-Push Neuromodulation of LTP and LTD Enables Bidirectional Experience-Induced Synaptic Scaling in Visual Cortex

Neuromodulatory input, acting on G protein-coupled receptors, is essential for the induction of experience-dependent cortical plasticity. Here we report that G-coupled receptors in layer II/III of visual cortex control the polarity of synaptic plasticity through a pull-push regulation of LTP and LTD. In slices, receptors coupled to Gs promote LTP while suppressing LTD; conversely, receptors coupled to Gq11 promote LTD and suppress LTP. In vivo, the selective stimulation of Gs- or Gq11-coupled receptors brings the cortex into LTP-only or LTD-only states, which allows the potentiation or depression of targeted synapses with visual stimulation. The pull-push regulation of LTP/LTD occurs via direct control of the synaptic plasticity machinery and it is independent of changes in NMDAR activation or neuronal excitability. We propose these simple rules governing the pull-push control of LTP/LTD form a general metaplasticity mechanism that may contribute to neuromodulation of plasticity in other cortical circuits.

Heterogeneous release probabilities and activity-dependent short-term synaptic depression.

Synaptic transmission is a major mechanism by which neurons communicate with each other. Basic steps in neurotransmitter release are similar in all synapses. However, many properties of release vary between synapses and reflect specific structural and functional requirements, endowing synapses with specialized functions. Recently, Gelman et al.1 described properties of release and short-term depression at specialized nicotinic synapses in the brainstem of goldfish, Carassius auratus (Linnaeus). These axo-axonic synapses between the Mauthner cell collaterals and their targets, cranial relay neurons (CRNs), exhibit strong short-term depression, even at stimulation frequencies as low as 0.33 Hz. In short, amplitudes of post-synaptic responses, evoked by presynaptic trains of action potentials, were depressed with a time course approximated by a sum of two exponential functions. Initially, fast depression reduced the amplitude of EPSP(2) (response after the second stimulus), to less than 50% of EPSP(1) (response after the first stimulus). This was followed by a slow component of depression that produced an additional 10-30% amplitude reduction over a time-span of tens to hundreds of seconds. Interestingly, depressed EPSPs exhibited longer latencies than that of the "undepressed" EPSP1. Additionally, fast and slow calcium chelators (BAPTA and EGTA), injected pre-synaptically, were equally potent in reducing release. These data are consistent with a previously proposed general mechanism that assumes a change in release probability after the initial release. However, in an alternative interpretation the results could be coherently explained by postulating two releasable pools of vesicles, with high and low release probabilities, and a generally accepted depletion scheme. This latter interpretation will be discussed in this article.

These axo-axonic synapses between the Mauthner cell collaterals and their targets, cranial relay neurons (CRNs), exhibit strong short-term depression, even at stimulation frequencies as low as 0.33 Hz. In short, amplitudes of post-synaptic responses, evoked by pre-synaptic trains of action potentials, were depressed with a time course approximated by a sum of two exponential functions. Initially, fast depression reduced the amplitude of EPSP2 (response after the second stimulus), to less than 50% of EPSP1 (response after the first stimulus). This was followed by a slow component of depression that produced an additional 10-30% amplitude reduction over a time-span of tens to hundreds of seconds. Interestingly, depressed EPSPs exhibited longer latencies than that of the ‘undepressed’ EPSP1. Additionally, fast and slow calcium chelators (BAPTA and EGTA), injected pre-synaptically, were equally potent in reducing release. These data are consistent with a previously proposed general mechanism that assumes a change in release probability after the initial release. However, in an alternative interpretation the results could be coherently explained by postulating two releasable pools of vesicles, with high and low release probabilities, and a generally accepted depletion scheme. This latter interpretation will be discussed in this article.

Homeostatic Synaptic Plasticity through Changes in Presynaptic Calcium Influx

Chronic perturbations of electrical activity within neural circuits lead to compensatory changes in synaptic strength collectively termed homeostatic synaptic plasticity. The postsynaptic mechanisms underlying these modifications have been characterized in some detail, but the presynaptic mechanisms that alter the efficiency of evoked neurotransmitter release are less clear. To investigate the role of presynaptic calcium influx, we have combined the use of two fluorescent proteins in cultured hippocampal neurons: a calcium reporter localized to synaptic vesicles, SyGCaMP2, and a reporter of vesicle fusion, SypHy. We find that a decrease in the activity of the network causes an increase in the amount of calcium entering the synaptic bouton in response to an action potential and an increase in the probability of vesicle fusion. Homeostatic changes in release probability varied as the third power of calcium influx. These results indicate that changes in the number and/or function of presynaptic calcium channels are major determinants of homeostatic changes in synaptic strength.

Postnatal Development of Dendrodendritic Inhibition in the Mammalian Olfactory Bulb

The mitral–granule cell (MC–GC) reciprocal synapse is an important source of auto- and lateral-inhibition in the olfactory bulb (OB), and this local inhibition is critical for odor discrimination. We may gain insight into the role of MC autoinhibition in olfaction by correlating the functional development of the autoinhibition with the postnatal development of olfactory function. We have studied the functional development of the MC–GC reciprocal synapse using whole-cell patch-clamp recordings from MCs and GCs in acute OB slices from 3- to 30-day-old rats. The magnitude of dendrodendritic inhibition (DDI) measured by depolarizing a single MC and recording recurrent inhibition in the same cell increased up to the fifteenth day of life (P15), but dropped between P15 and P30. The initial increase and later decrease in DDI was echoed by a similar increase and decrease in the frequency of miniature inhibitory post-synaptic currents, suggesting an accompanying modulation in the number of synapses available to participate in DDI. The late decrease in DDI could also result, in part, from a decrease in GC excitability as well as an increase in relative contribution of N-methyl d-aspartate (NMDA) receptors to γ-amino butyric acid (GABA) release from GC synapses. Changes in release probability of GABAergic synapses are unlikely to account for the late reduction in DDI, although they might contribute to the early increase during development. Our results demonstrate that the functional MC–GC circuit evolves over development in a complex manner that may include both construction and elimination of synapses.

The Pre/Post LTP Debate

The pre/post debate involves the question of whether long-term potentiation (LTP) is mediated by enhancement of release, enhancement of postsynaptic receptors, or both. Recent papers have presented evidence for purely postsynaptic or purely presynaptic changes, and a paper by Ahmed and Siegelbam (in this issue of Neuron) suggests a mechanism by which release is enhanced. This debate is increasingly constrained by technical advances that allow central synapses to be studied with increasing precision. A possible of way of reconciling conflicting evidence is suggested.

Developmental changes in short‐term facilitation are opposite at temporoammonic synapses compared to Schaffer collateral synapses onto CA1 pyramidal cells

CA1 pyramidal neurons receive two distinct excitatory inputs that are each capable of influencing hippocampal output and learning and memory. The Schaffer collateral (SC) input from CA3 axons onto the more proximal dendrites of CA1 is part of the trisynaptic circuit, which originates in Layer II of the entorhinal cortex (EC). The temporoammonic (TA) pathway to CA1 provides input directly from Layer III of the EC onto the most distal dendrites of CA1 pyramidal cells, and is involved in spatial memory and memory consolidation. We have previously described a developmental decrease in short-term facilitation from juvenile (P13-18) to young adult (P28-42) rats at SC synapses that is due to feedback inhibition via synaptically activated mGluR1 on CA1 interneurons. It is not known how short-term changes in synaptic strength are regulated at TA synapses, nor is it known how short-term plasticity is balanced at SC and TA inputs during development. Here we describe a novel developmental increase in short-term facilitation at TA synapses, which is the opposite of the decrease in facilitation occurring at SC synapses. Although short-term facilitation is much lower at TA synapses when compared with SC synapses in juveniles, short-term plasticity at SC and TA synapses converges at similar levels of paired-pulse facilitation in the young adult rat. However, in young adults CA3-CA1 synapses still exhibit more facilitation than TA-CA1 synapses during physiologically-relevant activity, suggesting that the two pathways are each poised to uniquely modulate CA1 output in an activity-dependent manner. Finally, we show that there is a developmental decrease in the initial release probability at TA synapses that underlies their developmental decrease in facilitation, but no developmental change in release probability at SC synapses. This represents a fundamental difference in the presynaptic function of the two major inputs to CA1, which could alter the flow of information in hippocampus during development.
