Effects of selection for litter size variability on ovarian folliculogenesis, ovarian cell proliferation, apoptosis, and production of regulatory peptides in rabbits

The present study aimed to identify the novel mechanisms regulating rabbit fecundity. For this purpose, the association between litter size variability, fecundity, ovarian morphology, and markers of proliferation, apoptosis, steroidogenesis, and the presence of regulatory proteins were examined in ovarian cells from two rabbit lines divergently selected for low (LL) and high (HL) variability in litter size throughout sixteen generations. Ovaries and uteri were isolated from multiparous non-lactating female rabbits from each line at sixteen generations of selection. One part of the ovary was subjected to histomorphometric analysis of folliculogenesis. From the rest of ovary, ovarian granulosa cells (OGCs) were isolated, cultured, and cell viability, proliferation (accumulation of PCNA and of cyclin B1, and BrdU-positive cells), and apoptosis (accumulation of caspase 3, bax and DNA fragmentation) were evaluated by the Trypan blue exclusion test and BrdU, quantitative immunocytochemistry, and cell death detection assays. Furthermore, OGCs were subjected to proteomic analysis by using the nano HPLC-Chip-MS/MS method. The release of progesterone and oestradiol was measured by ELISA. The LL had more than one kit per litter than the HL (7.6 kits vs. 6.5 kits, p ≤ 0.05). No differences were found in the diameter of primordial and primary ovarian follicles, theca, and granulosa thickness, but the diameter of oocytes in the primary and secondary follicles was higher in the LL than in the HL (88.49 µm in the LL vs. 77.86 µm in the HL for oocytes of primary follicles, p ≤ 0.05; 122.10 µm in the LL vs. 109.87 µm in the HL for oocytes of secondary follicles, p ≤ 0.05). Preovulatory follicles were presented only in the ovaries of the LL. The LL had higher incorporation of BrdU and reduced accumulation of bax within OGCs (0.92% in the LL vs. 0.44% in the HL for incorporation of BrdU, p ≤ 0.05; 41% in the LL vs. 48% in the HL for accumulation of bax, p ≤ 0.05). Ovarian fragments from the LL produced less progesterone and oestradiol than those of the HL (12 ng/mg tissue/day in the LL vs, 45 ng/mg tissue/day in the HL for progesterone, p ≤ 0.05; 11 ng/mg tissue/day in the LL vs. 30 ng/mg tissue/day in the HL for oestradiol, p ≤ 0.05). Besides, the OGCs from the LL produced a higher number of specific regulatory proteins involved in cell differentiation, proliferation, and adhesion than the HL (50 vs 38, p ≤ 0.05). In conclusion, higher prolificacy in the LL line would be caused by: (1) the selection of growing primordial ovarian follicles; (2) better transformation to preovulatory follicles; (3) increased cytoplasmic maturation of oocytes; (4) increased DNA synthesis and decreased cytoplasmic apoptosis in OGCs; (5) changes in ovarian steroidogenesis; and (6) changes in the number of peptides involved in cell differentiation, proliferation, and adhesion. HIGHLIGHTS Assessing fecundity in two divergently rabbit lines for variability in litter size. Fecundity and its variability are related to the diameter and quality of oocytes. Selection and better growth of preovulatory follicles have been related to fecundity.

FRI390 The Effects Of Chronic Asprosin Administration On Ovarian Tissue

Purpose: Asprosin is a novel glucogenic and orexigenic hormone produced by the fibrillin 1 (FBN1) gene that is secreted by white adipose tissue during fasting. Fibrillins (FBNs) are structural and signalling proteins found mainly in the ovarian stroma, tunica albuginea and theca layers. The aim of this study was to determine the effects of asprosin on ovarian tissue in the female rats. Materials and methods: Twenty-four female Sprague-Dawley rats were randomly divided into 2 groups (n=12) as control and asprosin. Asprosin and control groups were given intraperitoneal saline at doses of 500 ng/kg and 1 ml/kg, respectively, at 14.00 every day for eight weeks. After the application, decapitation was performed, and the rapidly removed ovarian tissues were placed in a 10% formaldehyde solution for fixation. Formalin-fixed ovaries were dehydrated in a graded alcohol series and graded ethanol, respectively After that, all tissues were embedded in paraffin wax. The ovaries were cut into 5-µm sections and stained with hematoxylin and eosin (H&E) and also Masson trichrome staining. Ovarian follicular reserve and fibrosis were evaluated by light microscopy. The follicles were histologically classified as primordial, primary, secondary, and Graaf and all follicular structures were counted in each section. The fibrosis was evaluated with Masson’s trichrome staining and scored from 0 to 3 semi-quantitatively (0 = no fibrosis, 1 = low fibrosis, 2 = intermediate fibrosis, 3 = severe fibrosis). Results: In the asprosin group, unlike the control group, occasional congestion was observed in the vessels located in the ovarian medulla. Apart from this, no histopathological changes were found. An increase in fibrosis was detected in the ovarian sections of the asprosin group when compared to the control group (p = 0,035). In the light microscopic evaluation of the ovarian sections of the rats in the asprosin group, the primary and secondary follicle numbers were significantly higher than those in the control group (p = 0,008 and p = 0,047 respectively). However, the difference between the control and asprosin groups in terms of the primordial follicle, Graafian follicle and corpus luteum numbers were not statistically significant. Conclusion: Our results have shown that chronic asprosin administration causes an increase in the number of primary and secondary follicles, suggesting that changes in ovarian steroidogenesis and folliculogenesis depending on the application may lead to positive effects on female fertility. Seasonal changes in endoplasmic reticulum stress and steroidogenesis in the ovary of the wild ground squirrels (Citellus dauricus Brandt)

The development of the follicle is accompanied by steroidogenesis and secretion, the endoplasmic reticulum (ER) requires significant synthesis of relevant proteins to support changes in the follicular microenvironment. The aim of this study was to investigate whether seasonal changes in gonadotropins and ovarian steroid hormones in the wild ground squirrels induce endoplasmic reticulum stress (ERS) and changes in ERS-mediated unfolded protein response (UPR) signaling. There were significant seasonal differences in ovarian mass, with values higher in the breeding season and relatively low in the non-breeding season. Histological observations revealed that ovaries in the breeding season had germ cells including primordial follicles, primary follicles, secondary follicles, tertiary follicles, and the corpus luteal, whereas ovaries consisted mainly of primary and secondary follicles in the non-breeding season. Analysis of ovarian transcriptome data showed that 1298 genes were up-regulated in expression and 1432 genes were down-regulated in expression during both periods. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis indicated that these genes were mainly enriched in estrogen signaling pathways, ovarian steroidogenesis and endoplasmic reticulum protein processing pathways. The expression levels of steroidogenic enzymes (P450scc, P450c17, 3β-HSD, and P450arom) and gonadotropin receptor (FSHR and LHR) were significantly increased during the breeding season compared to the non-breeding season. GRP78 and UPR signaling factors (ATF4, ATF6, XBP1s) associated with ERS were expressed in both seasons. The mRNA expressions of Atf6 and Xbp1s were higher in the breeding season than those of the non-breeding season. Conversely, Atf4 and its downstream homologous protein (Chop) exhibited higher expression during the non-breeding season. In addition, follicle stimulating hormone (FSH), luteinizing hormone (LH), estradiol-17β, and progesterone of serum were significantly higher in the breeding season than those of the non-breeding season. These results suggested that UPR signaling, associated with seasonal changes in ovarian steroidogenesis, was activated during the breeding season and that ERS might be involved in regulating seasonal changes in ovarian steroidogenesis in the wild ground squirrels.

Seasonal changes in endoplasmic reticulum stress and ovarian steroidogenesis in the muskrats (Ondatra zibethicus).

Many studies have shown roles for endoplasmic reticulum stress (ERS)/unfolded protein response (UPR) signaling cascades with ovarian folliculogenesis, and oocyte maturation. In this study, we investigated seasonal changes in ERS and ovarian steroidogenesis in the muskrats (Ondatra zibethicus) during the breeding season (BS) and non-breeding season (NBS). There were noticeable seasonal variations in the weight and size of muskrat ovaries with values higher in the BS than that in NBS. The circulating luteinizing hormone (LH), follicle-stimulating hormone (FSH), 17β-estradiol, and progesterone of the female muskrats were higher during the BS. The RNA-seq data of ovaries during different seasons revealed 2580 differentially expressed genes, further analysis showed a prominent enrichment of ERS-related pathways and ovarian steroidogenesis pathway. Immunohistochemical results showed that GRP78 and steroidogenic enzymes (P450scc, 3β-HSD, P450c17, and P450arom) existed in the various kinds of cells in muskrat ovaries during the BS and NBS. In ovaries from the BS, the mRNA levels of P450scc, P450arom, P450c17, and 3β-HSD were considerably higher. Furthermore, the expression levels of oxidative stress-related genes (SOD2, CAT, and GPX1) and UPR signal genes (Bip/GRP78, ATF4, ATF6, and XBP1s) were increased strikingly higher during the BS in comparison with the NBS. However, the mRNA levels of CCAAT-enhancer-binding protein homologous protein (CHOP) and caspase-3 had no considerable difference between the BS and NBS. Taken together, these results suggested that UPR signaling associated with the seasonal changes in ovarian steroidogenesis is activated in the BS and the delicate balance in redox regulation is important for seasonal reproduction in the muskrats.

An Ovarian Steroid Metabolomic Pathway Analysis in Basal and Polycystic Ovary Syndrome (PCOS)-like Gonadotropin Conditions Reveals a Hyperandrogenic Phenotype Measured by Mass Spectrometry

Prior work has demonstrated that murine ovarian explants and isolated ovarian follicles can recapitulate human-like 28-day cycles in vitro with normal patterns of estradiol and progesterone secretion in response to gonadotropin stimulation. The objective of this study was to manipulate the gonadotropin stimulation protocol to mimic polycystic ovary syndrome (PCOS) and assess the resulting changes in ovarian steroidogenesis. A secondary aim of the study was to develop a high-throughput, sensitive, and specific liquid chromatography with tandem mass spectrometry (LC-MS/MS) assay to measure seven steroid hormones (estrone, estradiol, progesterone, testosterone, androstenedione, dehydroepiandrosterone, and dihydrotestosterone) in conditioned culture media. Ovaries were harvested from 12-day-old CD-1 mice and cultured for 28 days, with ovulation induction on culture day 14. Media were supplemented human chorionic gonadotropin (hCG, a luteinizing hormone analog) and follicle stimulating hormone (FSH) at ratios of 1:0 (standard media), 1:1 (physiologic ratio), and 3:1 (PCOS-like ratio). Ovaries cultured in PCOS-like media displayed hyperandrogenism and impaired ovulation, two key features of a PCOS-like phenotype. Taken together, this first-of-its-kind presentation of hormone levels from single tissues creates a map of the enzymatic steps most acutely affected by gonadotropin dysregulation and may provide opportunities for assessing other potential insults in PCOS pathogenesis.

Physiological and Pathological Androgen Actions in the Ovary.

Androgens, although traditionally thought to be male sex steroids, play important roles in female reproduction, both in healthy and pathological states. This mini-review focuses on recent advances in our knowledge of the role of androgens in the ovary. Androgen receptor (AR) is expressed in oocytes, granulosa cells, and theca cells, and is temporally regulated during follicular development. Mouse knockout studies have shown that AR expression in granulosa cells is critical for normal follicular development and subsequent ovulation. In addition, androgens are involved in regulating dynamic changes in ovarian steroidogenesis that are critical for normal cycling. Androgen effects on follicle development have been incorporated into clinical practice in women with diminished ovarian reserve, albeit with limited success in available literature. At the other extreme, androgen excess leads to disordered follicle development and anovulatory infertility known as polycystic ovary syndrome (PCOS), with studies suggesting that theca cell AR may mediate many of these negative effects. Finally, both prenatal and postnatal animal models of androgen excess have been developed and are being used to study the pathophysiology of PCOS both within the ovary and with regard to overall metabolic health. Taken together, current scientific consensus is that a careful balance of androgen activity in the ovary is necessary for reproductive health in women.

Spawning performance and plasma levels of GnRHa and sex steroids in cultured female Senegalese sole ( Solea senegalensis) treated with different GnRHa-delivery systems

The aquaculture of Senegalese sole ( Solea senegalensis) is limited by the poor spawning performance of the F1 generation cultured broodstock. The present study compared the efficiency of two sustained-release delivery systems loaded with gonadotropin-releasing hormone agonist (GnRHa) and a single GnRHa injection on the stimulation of oocyte maturation (OM), sex steroid hormone secretion and spawning. Analysis with an ELISA showed that the administered GnRHa was no longer detectable in the plasma 3, 7 and 14 d after administration via a single injection, microspheres and an implant, respectively. All GnRHa treatments induced a transient elevation of testosterone (T) and 17β estradiol (E 2) plasma levels after 1–3 d. Only three spawns were obtained from controls on days 35, 41 and 42 of the study and ovarian biopsies showed abundant fully vitellogenic oocytes, but little OM. Fish given a GnRHa injection spawned sporadically with the first spawn at 20 days post-treatment (dpt). In contrast, GnRHa implants and microspheres stimulated OM and daily spawning starting at 4 and 5 dpt, respectively, and lasting for a period of 28 d. Total fecundity in the broodstock treated with GnRHa-delivery systems was 15–22 times greater than controls and 3–5 times greater than GnRHa-injected fish. Although egg quality appeared to be high, no fertilization was obtained. The results suggest that GnRHa-delivery systems are highly efficient on stimulating OM and spawning in Senegalese sole, presumably through the long-term elevation of pituitary luteinizing hormone release, which in turn stimulated the appropriate changes in ovarian steroidogenesis. However, the effectiveness of the treatment was negated by possible dysfunctions of the male breeders or of the reproductive behavior of the broodstock.

Human Chorionic Gonadotropin (hCG) Up-Regulates wnt5b and wnt7b in the Mammary Gland, and hCGβ Transgenic Female Mice Present with Mammary Gland Tumors Exhibiting Characteristics of the Wnt/β-Catenin Pathway Activation

Transgenic (TG) mice expressing human chorionic gonadotropin (hCG) beta-subunit under the ubiquitin C promoter, presenting with a moderately elevated level of LH/hCG bioactivity develop multiple neoplasms secondary to the endocrine abnormalities, including mammary gland tumors after the age of 9 months. The increased levels of circulating estradiol, progesterone, and prolactin of the TG females after puberty boost the lobuloalveolar development in the mammary gland resulting ultimately in the formation of estrogen and progesterone receptor-negative, malignant tumors. These tumors have a similar histopathology with those observed in TG mice with activated wnt/beta-catenin pathway, showing increased expression of beta-catenin, also a common finding in human breast tumors. Transdifferentiation is observed in mammary tumors of the hCGbeta TG mice, accompanied by abnormal expression of the Wnt genes in the tumorous and nontumorous mammary gland tissue. Specifically we found increased expression of Wnt5b in the TG mammary glands at the age of 3 months and up-regulation of Wnt7b and -5b in the subsequently appearing tumors. Importantly, hCG was found to up-regulate these wnt ligands in mouse mammary gland, independent of the changes in ovarian steroidogenesis. Thus, the hCGbeta-overexpressing TG mice represent a novel model that links enhanced hCG action to dysregulated wnt signaling in the mammary gland, resulting in beta-catenin-stabilizing mammary tumorigenesis. The novel finding of hCG up-regulating wnt7b and wnt5b could contribute to pregnancy-induced breast cancer in humans.

Changes in ovarian steroidogenesis in insulin-resistant, type 2 diabetic Goto–Kakizaki rats after thyroidectomy and gonadotropin treatment

The present study used thyroidectomized insulin-resistant, type 2 diabetic Goto–Kakizaki (GK) rats to assess whether insulin resistance and hypothyroidism modulate ovarian physiology. Animals were treated with daily injections of 5 IU equine chorionic gonadotropin for 5 days starting 1 week after thyroidectomy. Control groups included rats of GK and control (Wistar) strains treated only with equine chorionic gonadotropin or thyroidectomy, or with no treatment (intact). In Wistar rats, equine chorionic gonadotropin injections tended to increase the serum concentrations of luteinizing hormone (LH) and testosterone more in the thyroidectomy group than in intact rats. Similar changes in LH and testosterone were observed in the thyroidectomy + equine chorionic gonadotropin and equine chorionic gonadotropin groups of GK rats, but the LH and testosterone levels in the thyroidectomy + equine chorionic gonadotropin group were significantly higher in GK rats. Expression of ovarian LH receptor messenger RNA (mRNA) was enhanced by thyroidectomy. The LH receptor mRNA levels were significantly higher in the thyroidectomy + equine chorionic gonadotropin group of GK rats than in the corresponding group of control rats. These results indicate that hypothyroidism in animals with insulin resistance and type 2 diabetes promotes LH and testosterone secretions, and suggests that the enhanced-testosterone levels is partially mediated by the enhancement of LH receptor expression and an increase in the serum level of LH.

Seasonal Changes in the Production of Two Novel and Abundant Ovarian Steroids in the Channel Catfish (Ictalurus punctatus)

Timely and appropriate changes in steroid plasma titers are necessary for successful reproduction in all vertebrates. Gonadal steroidogenesis of the most intensively cultured teleost species in North America, the channel catfish (Ictalurus punctatus), is poorly understood so a year-long study was conducted to investigate seasonal changes in ovarian steroidogenesis. Incubations of ovarian tissue were conducted monthly with [3H]pregnenolone and the medium was analyzed by high-performance liquid chromatography (HPLC) with radioactivity detection. The suite of steroids produced by the catfish ovary included the expected sex steroids (estradiol and testosterone) and 18 additional ovarian metabolites, including five steroids that have yet to be identified. Androstenedione, 20β-dihydroprogesterone, 5|P-dihydrotestosterone, estriol, 11β-hydroxyandrostenedione, 17-hydroxypregnenolone, 17-hydroxyprogesterone, 11β-hydroxytestosterone, and progesterone were characterized by a combination of HPLC and thin-layer chromatography. Two of the most abundant steroids were isolated and analyzed by gas chromatography coupled with mass spectrometry (GC-MS). One of the steroids, 7α-hydroxypregnenolone (7P5), is a novel steroid in teleosts and was produced late in vitellogenic growth of the oocyte. Evidence suggests that the enzyme responsible for converting pregnenolone to 7P5, 7α-hydroxylase, is a cytochrome P450. The second abundant steroid metabolite was partially characterized by GC-MS as an hydroxylated form of 17-hydroxy-pregnenolone (χ,17P5). This steroid was most abundant when the ovary was regressed and during early vitellogenesis and rapidly decreased prior to spawning. In mammals, 7P5 has been identified as an important neurosteroid; however, the reproductive significance of 7P5 and χ,17P5 in catfish is unknown.

Mating stimulates estradiol production by ovaries of the musk shrew (Suncus murinus).

Asian musk shrews (Suncus murinus) are induced ovulators, but exhibit no cyclic changes in reproductive structures or in sexual behavior. Mating behavior is induced by contact with a male. To determine if mating induces changes in ovarian steroidogenesis, ovaries removed from unmated animals and at 3, 10, 15, and 36 h after mating were cultured for 4 h in the presence or absence of gonadotropins (LH + FSH, 1 microgram/ml). Histological analysis revealed no obvious changes in follicular size or appearance at the end of culture in ovaries cultured at 3 and 10 h post-mating, as compared with ovaries from unmated shrews, and mating did not stimulate any discernable changes in steroid secretion in these two groups. However, at the end of the culture period, ovulation had occurred or was occurring in ovaries from 35% of the animals ovariectomized at 15 h after mating, and corpora lutea (CLs) were present in 39% of ovarian pairs obtained 36 h after mating. At 15 h post-mating, ovaries with ovulations secreted three times more estradiol than did ovaries that showed no evidence of stimulation by mating, but there were no differences in testosterone or progesterone production. In contrast, ovaries isolated 36 h post-mating with CLs secreted dramatically more of all three steroids than ovaries without CLs (23, 13, and 52 times more estradiol, testosterone, and progesterone, respectively). These data are consistent with plasma concentrations of estradiol at the time of ovariectomy, which were twice as high at both 15 and 36 h after mating, in animals whose ovaries showed evidence of ovarian stimulation.(ABSTRACT TRUNCATED AT 250 WORDS)

Steroidogenesis during postnatal development in the mouse ovary

In the mouse, follicular formation and development is largely postnatal. Changes in ovarian steroidogenesis during early postnatal life are likely, therefore, to reflect changes in follicular activity during early folliculogenesis. In this study, basal progesterone and androstenedione production and responsiveness to gonadotrophins, dibutyryl cyclic AMP (dbcAMP) and 22R-hydroxycholesterol have been measured following short-term in-vitro incubations of ovaries from mice aged 1, 3, 5, 7, 10 or 15 days. On the day of birth (day 1), basal and gonadotrophin-stimulated progesterone and androstenedione production were undetectable although a response to dbcAMP and a low level of cholesterol side-chain cleavage (CSCC) activity (measured using 22R-hydroxycholesterol) were present. On day 3 progesterone and androstenedione production were undetectable under all conditions. On day 5 only a low level of CSCC activity was detectable but on day 7 there was an increase in ovarian steroid production. Basal progesterone and androstenedione were detectable and LH, but not FSH, increased the production of both steroids. These changes were associated with a marked increase of more than 80-fold in CSCC activity. Basal steroid output increased from days 7 to 15 and LH continued to stimulate progesterone accumulation although no effect on androstenedione was seen. Addition of FSH had no effect on steroidogenesis on day 10 but significantly increased progesterone production on day 15. Ovaries from the mice used in these studies contained primordial follicles and stromal tissue on day 1. By day 5 primary and secondary follicles were present and the major increase in steroid production between days 5 and 7 was associated with an increase in secondary follicles and increased differentiation of the thecal tissue.(ABSTRACT TRUNCATED AT 250 WORDS)

Changes in ovarian steroidogenesis in vitro associated with final maturation of Atlantic croaker oocytes

Changes in ovarian steroidogenesis during final oocyte maturation (FOM) in the Atlantic croaker ( Micropogonias undulatus) were investigated in vitro by incubation of oocytes with tritiated pregnenolone, followed by chromatographic separation of the radioactive steroid products. Testosterone and estradiol-17β were the major tritiated steroids produced by the ovary before the onset of FOM. Induction of FOM by human chorionic gonadotropin, both in vivo and in vitro, coincided with the appearance of 17α,20β,21-trihydroxy-4-pregnen-3-one (20β-S) as the predominant steroid product. The production of 17α,20β-dihydroxy-4-pregnen-3-one and 11-deoxycorticosterone, maturation-inducing steroids previously proposed for other teleost species, could not be demonstrated in Atlantic croaker ovaries during FOM. The timely synthesis of 20β-S in croaker at the onset of oocyte maturation provides further evidence for a physiological role of 20β-S in the induction of FOM in this species.

Steroidogenesis during induced oocyte maturation and ovulation in the African catfish,Clarias gariepinus

Changes in ovarian steroidogenesis accompanying oocyte maturation and ovulation were studied in the African catfish,Clarias gariepinus. Laboratory-reared females with postvitellogenic ovaries were treated with pimozide and LHRH-analogue. The plasma gonadotropin levels were determined by means of a homologous radioimmunoassay, the condition of the ovaries was studied by histological examination of the follicles, and the steroidogenetic capacity of the ovaries was analyzed byin vitro incubation of tissue fragments for 3 h with [(3)H]-pregnenolone and [(3)H]androstenedione as precursors. Data were collected at regular intervals between 0 and 16 h after pimozide-LHRH analogue administration.Until 4 h after the beginning of the experiments the plasma gonadotropin levels did not rise above 25 ng/ml, the ovaries remained in the stage of postvitellogenesis, and testosterone was the main end product of steroidogenesis. Four hours later the gonadotropin concentration in the blood had risen to more than 150 ng/ml, and the ovaries had entered the stage of germinal vesicle migration. At the same time steroidogenesis shifted towards the production of 17α,20β-dihydroxy-4-pregnen-3-one, 5β-pregnane-3α, 17α-diol-20-one, 5β-pregnane-3α,6α,17α-triol-20-one, 5β-pregnane-3α,17α,20β-triol and 5β-pregnane-3α,6α,17α,20β-tetrol. During the subsequent stage of germinal vesicle breakdown the plasma gonadotropin level remained high, and the synthesis of the C21-steroids showed a further increase. Simultaneously, the production of some C19-steroid glucuronides was enhanced. The preovulation and especially the postovulation stages were accompanied by a gradual decrease in steroidogenic capacity of the ovaries, even though the plasma gonadotropin level remained high. It is concluded that the prematuration surge of gonadotropin influences the activity of enzymes involved in steroidogenesis, leading to a reduced C17,20-lyase and to an augmented activity of the enzymes 20β-hydroxysteroid dehydrogenase (HSD), 5β-reductase, 3α-HSD, 6α-hydroxylase and UDP-glucuronosyltransferase. During ovulation the activity of all steroidogenic enzymes, including such key enzymes as 3β-HSD and 17α-hydroxylase, gradually decreases.Not only 17α,20β-dihydroxy-4-pregnen-3-one, but also the 5β-reduced pregnanes may be involved in inducing oocyte maturation and/or ovulation. The very polar triol and tetrol products may function, together with the steroid glucuronides as sex pheromones.

Changes in ovarian steroidogenesis in prepuberal rats induced to ovulate by low doses of human chorionic gonadotrophin.

To determine whether the decrease in ovarian 5 alpha-reduced androgen production before first ovulation might be caused by an increase in serum LH, prepuberal female rats were injected at 28-31 days of age with low doses of human chorionic gonadotrophin (hCG) (0.05-0.075 i.u., four times daily). This treatment resulted in ovulation of six to ten ova per rat on day 32 in all animals. Treatment with hCG resulted in a gradual decrease in ovarian content and production (i.e. content in ovary and medium after 4h of incubation) of 5 alpha-dihydrotestosterone (DHT) and 5 alpha-androstane-3 alpha, 17 beta-diol. The ovarian content of DHT and the production of 5 alpha-androstane-3 alpha, 17 beta-diol decreased within 24 h after the first injection of hCG. Oestradiol content and production increased between 24 and 48 h after the start of treatment and was maximal on day 31 (day of pro-oestrus). Activities of 5 alpha-reductase and aromatase were measured in ovarian homogenates obtained on days 29-31. Activity of 5 alpha-reductase in hCG-treated rats was lower than that in control rats on all days studied. Aromatase activity in hCG-treated rats increased between days 29 and 31. It was concluded that multiple injections of low doses of hCG, which may induce ovulation, cause a decrease in 5 alpha-reduced androgen production, which is probably due to a decrease in 5 alpha-reductase activity. The subsequent increase in oestradiol production corresponds with an increase in aromatase activity.(ABSTRACT TRUNCATED AT 250 WORDS)
