Cell wall metabolism during the growth of peach fruit: Association of hardness with cell wall pectins

In this study, molecular composition of cell wall polysaccharides of peach during growth was investigated to elucidate influencing mechanism of hardness which regulated fruit quality. Results revealed that as fruit grew stage I to V (almost 36 to 103 day after flowering), hardness firstly increased and then decreased, reaching its peak in stage II. Microstructure indicated an increase in cell wall thickness and a transition from compact to loose adhesion between cell walls from stage Ⅰ to Ⅱ. Moreover, molecular weight (Mw), monosaccharide composition and fourier transform infrared spectra (FTIR) of peach polysaccharides extracted by water (WPP), chelating agent (CPP) and alkali (APP) indicated Mw of WPP and APP showed a downward trend and reaching the highest in stage II for CPP. Similar tendency appeared in the galacturonic acid (GalA) content. Content of galactose and arabinose in all polysaccharide decreased. Degree of esterificationa (DE) of CPP was the lowest in stage II. An increasing trend appeared in WPP. Finally, β-Galactosidase, α-l-arabinofuranosidase, pectin methyl esterase, polygalacturonase and their associated genes were analyzed to be evidence of the change of molecular composition. Above results implied Mw, GalA content and DE of CPP related with hardness during growth.

How do the carbon and nitrogen sources affect the synthesis of β-(1,3/1,6)-glucan, its structure and the susceptibility of Candida utilis yeast cells to immunolabelling with β-(1,3)-glucan monoclonal antibodies?

BackgroundThe need to limit antibiotic therapy due to the spreading resistance of pathogenic microorganisms to these medicinal substances stimulates research on new therapeutic agents, including the treatment and prevention of animal diseases. This is one of the goals of the European Green Deal and the Farm-To-Fork strategy. Yeast biomass with an appropriate composition and exposure of cell wall polysaccharides could constitute a functional feed additive in precision animal nutrition, naturally stimulating the immune system to fight infections.ResultsThe results of the research carried out in this study showed that the composition of Candida utilis ATCC 9950 yeast biomass differed depending on growth medium, considering especially the content of β-(1,3/1,6)-glucan, α-glucan, and trehalose. The highest β-(1,3/1,6)-glucan content was observed after cultivation in deproteinated potato juice water (DPJW) as a nitrogen source and glycerol as a carbon source. Isolation of the polysaccharide from yeast biomass confirmed the highest yield of β-(1,3/1,6)-glucan after cultivation in indicated medium. The differences in the susceptibility of β-(1,3)-glucan localized in cells to interaction with specific β-(1,3)-glucan antibody was noted depending on the culture conditions. The polymer in cells from the DPJW supplemented with glycerol and galactose were labelled with monoclonal antibodies with highest intensity, interestingly being less susceptible to such an interaction after cell multiplication in medium with glycerol as carbon source and yeast extract plus peptone as a nitrogen source.ConclusionsObtained results confirmed differences in the structure of the β-(1,3/1,6)-glucan polymers considering side-chain length and branching frequency, as well as in quantity of β-(1,3)- and β-(1,6)-chains, however, no visible relationship was observed between the structural characteristics of the isolated polymers and its susceptibility to immunolabeling in whole cells. Presumably, other outer surface components and molecules can mask, shield, protect, or hide epitopes from antibodies. β-(1,3)-Glucan was more intensely recognized by monoclonal antibody in cells with lower trehalose and glycogen content. This suggests the need to cultivate yeast biomass under appropriate conditions to fulfil possible therapeutic functions. However, our in vitro findings should be confirmed in further studies using tissue or animal models.

Cell wall polysaccharides, phenolic extractability and mechanical properties of Aleatico winegrapes dehydrated under sun or in controlled conditions

Aleatico is an Italian red winegrape variety traditionally used partially dehydrated to produce Passito wines. The aim of this research is to assess the effect of two different off-vine withering techniques (natural sun-exposed: SUN, and controlled temperature and humidity conditions: CTR) on grapes physicochemical modifications, focusing on mechanical properties, phenolic profile, and cell-wall polysaccharide structures. The composition and extractability of polyphenols were studied before and after the two withering processes, differentiating the tissues (skin, pulp, seeds). A stronger effect on cell-wall polysaccharides composition pectin was highlighted by chemical and immunochemical approaches in SUN skins at the level of pectin chains and de-esterification, affecting grapes mechanical properties, especially for skin rigidity (<Esk). Sun-dehydrated grapes showed the highest pH (about 0.3 units more than FRESH and CTR), and the lowest acidity (−0.71 g/L vs FRESH) and anthocyanin content in wine-like solution (−41%), whereas these parameters were better preserved in controlled conditions. Instead, seeds tannins were higher in SUN than other samples, with a lower oligomers/polymers ratio. Indeed, after withering the phenolic extractability decreased in skins (total phenolics: from 60% for FRESH to 40% for SUN; anthocyanins: from 72% for FRESH to 46% for SUN) and increased in seeds (total phenolics: from 30% for FRESH to 41% for SUN) compared with fresh samples, in both cases more markedly for SUN than CTR. Skin phenolic composition after 10 days of simulated-maceration was affected by the combination of the loss/concentration balance, together with their modified release in the wine-like solution. As regards seeds, instead, the differences highlighted by the extractable profiles were due to the modified extractability as consequence of the different withering conditions and not to chemical quantitative/qualitative modifications of phenolic compounds caused by dehydration.

Enzyme-Assisted Extraction for the Recovery of Food-Grade Chlorophyll-Based Green Colorant.

The aim of the study was to develop a biotechnological approach for the green recovery of chlorophyll from spinach, to be used as a natural food colorant. The plant matrix was characterized in terms of cell wall polysaccharide composition, and a tailored enzymatic mix based on cellulase (40%) xylanase (41%) and polygalacturonase (19%) was formulated. The process variables (temperature (°C), time (h), enzyme mix dose (U/g), zinc concentration (ppm), and buffer/substrate ratio (B/S)) and their interactions were studied by response surface methodology. The overlay plot made it possible to identify the process conditions (T: 25 °C, Zn: 150 ppm e B/S: 17.5, t: <2 h and enzyme mix dose between 12 and 45 U/g) to maximize the amount of chlorophyll, and concurrently, the quality of the green color of the extract. Finally, the novel colorant was applied in the production of a real food.

Immunochemical Identification of the Main Cell Wall Polysaccharides of the Early Land Plant Marchantia polymorpha.

Plant primary cell walls are composite structures surrounding the protoplast and containing pectins, hemicelluloses, and cellulose polysaccharides, as well as proteins. Their composition changed during the evolution of the green lineage from algae to terrestrial plants, i.e., from an aquatic to a terrestrial environment. The constraints of life in terrestrial environments have generated new requirements for the organisms, necessitating adaptations, such as cell wall modifications. We have studied the cell wall polysaccharide composition of thalli of Marchantia polymorpha, a bryophyte belonging to one of the first land plant genera. Using a collection of specific antibodies raised against different cell wall polysaccharide epitopes, we were able to identify in polysaccharide-enriched fractions: pectins, including low-methylesterified homogalacturonans; rhamnogalacturonan I with arabinan side-chains; and hemicelluloses, such as xyloglucans with XXLG and XXXG modules, mannans, including galactomannans, and xylans. We could also show the even distribution of XXLG xyloglucans and galactomannans in the cell walls of thalli by immunocytochemistry. These results are discussed with regard to the cell wall proteome composition and in the context of the evolution of the green lineage. The cell wall polysaccharides of M. polymorpha illustrate the transition from the charophyte ancestors of terrestrial plants containing xyloglucans, xylans and mannans as hemicelluloses, and embryophytes which do not exhibit mannans as major primary cell wall polysaccharides.

Disruption of a DUF247 Containing Protein Alters Cell Wall Polysaccharides and Reduces Growth in Arabidopsis.

Plant cell wall biosynthesis is a complex process that requires proteins and enzymes from glycan synthesis to wall assembly. We show that disruption of At3g50120 (DUF247-1), a member of the DUF247 multigene family containing 28 genes in Arabidopsis, results in alterations to the structure and composition of cell wall polysaccharides and reduced growth and plant size. An ELISA using cell wall antibodies shows that the mutants also exhibit ~50% reductions in xyloglucan (XyG), glucuronoxylan (GX) and heteromannan (HM) epitopes in the NaOH fraction and ~50% increases in homogalacturonan (HG) epitopes in the CDTA fraction. Furthermore, the polymer sizes of XyGs and GXs are reduced with concomitant increases in short-chain polymers, while those of HGs and mHGs are slightly increased. Complementation using 35S:DUF247-1 partially recovers the XyG and HG content, but not those of GX and HM, suggesting that DUF247-1 is more closely associated with XyGs and HGs. DUF247-1 is expressed throughout Arabidopsis, particularly in vascular and developing tissues, and its disruption affects the expression of other gene members, indicating a regulatory control role within the gene family. Our results demonstrate that DUF247-1 is required for normal cell wall composition and structure and Arabidopsis growth.

Exploring the polysaccharide composition of plant cell walls in succulent aloes

Societal Impact StatementAloes are iconic succulent plants native to Africa, Madagascar and the Arabian Peninsula. The succulent leaf mesophyll of aloes has been used extensively as a herbal product for centuries, contributing to their overexploitation. Health benefits are attributed to their polysaccharide content. We present a comprehensive comparison of the polysaccharide composition of succulent tissues from 93 Aloe species. We found polysaccharide composition primarily related to leaf morphology in alignment with the broad range of Aloe species used medicinally. All aloes except Aloe ferox and Aloe vera are endangered raising concern about over‐harvesting of wild species.Summary Aloes are iconic succulent plants native to Africa, Madagascar and the Arabian Peninsula. All aloes except the commercially grown Aloe ferox and Aloe vera are protected according to the Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES). Major factors contributing to their over‐exploitation are their ornamental value and medicinal use with more than 25% of Aloe species being utilised. The succulent inner leaf mesophyll of aloes is used in traditional medicine, with the healing effect ascribed to the properties of their structural polysaccharides. To explore the correlation between Aloe polysaccharide profiles and other biologically relevant traits across the genus, we (1) extracted polysaccharides and created profiles for nearly 100 representative species using carbohydrate microarrays and molecular probes. We targeted six major plant cell wall polysaccharide groups using 27 different molecular probes. We (2) tested for phylogenetic signal in the polysaccharide profiles and (3) assembled an exhaustive database from literature on the geographic region, level of endemism, altitude, habitat, habit, medicinal use and leaf morphology of the individual species of Aloe. In the absence of phylogenetic signal of polysaccharide profiles, multivariate linear modelling without phylogenetic correction was used and showed that polysaccharide composition primarily correlated with leaf morphology, highlighting the fundamental role of polysaccharides as the building blocks of plants. No correlations between polysaccharide composition of commercial and non‐commercial species were found. We found polysaccharide composition to primarily relate to leaf morphology emphasising the fundamental and structural role of polysaccharides.

A multimodal and multiscale investigation of factors affecting the juice yield of cider apples

Apple cider juice yield at harvest and after 15 and 30 days of storage durations was studied by analyzing the mechanical properties of fresh and plasmolyzed flesh, water distribution, cell wall polysaccharide composition and organization of the apples; in this study, the apple varieties used were Avrolles, Douce coetligne, Douce moen, Judor, Petit jaune. Juice yield mainly depended on the apple variety and the storage duration. Cellulose organization and cell wall pectin hydration were affected by ripening and are related to fruit firmness. Flesh viscoelastic mechanical properties were not general indications of juice yields. However, these properties helped distinguish the varieties according to flesh damage caused by ice crystals upon freezing. Cell encapsulation of the juice in the flesh contributed to lower yields. The apple variety and harvesting mode are recommended as a means to better control juice yield variations.

Polysaccharide composition during cotton seed fibre development: temporal differences between species and in different seasons

BackgroundCotton seed fibres are long single-celled epidermal trichomes that first appear on the surface of the ovule at anthesis and then elongate rapidly over a period of 15–25 days until a secondary cell wall (SCW) begins to develop through a rapid increase in the deposition of microfibrillar cellulose between the plasma membrane and the primary cell wall that eventually terminates elongation. Quantitative measurements of the different polysaccharide components in both wall types over time and how they influence fibre quality can direct studies involved in enhancing fibre properties for yarn quality through cell wall manipulation or molecular breeding.ResultsA detailed chemical analysis of cell wall composition by differential solvent fractionation was used to identify the range of polysaccharides present in mature cotton fibres and used to validate a simpler total cell wall monosaccharide linkage analysis protocol for wall compositional analysis. Analysis of fibres from 5 days post-anthesis through maturity for three cultivated species, Gossypium hirsutum, G. barbadense, and G. arboreum, showed the dynamic nature of cell wall polysaccharide composition through fibre development and that it progressed differently for each species. Plants grown in the glasshouse during either autumn to winter or spring to summer and within each species had fibre qualities and temporal aspects of cell wall development that were different for each season. Notably, the timing of the deposition of the SCW was delayed in winter grown plants and appeared to influence key fibre quality properties.ConclusionsThese results suggest that the temporal aspects of cell wall polysaccharide biogenesis during fibre development influence final fibre quality, and this timing is determined by both genetic and environmental factors. The onset of SCW synthesis appears to be a critical factor coinciding with termination of fibre elongation and specifying the duration of wall thickening that then affects fibre length and other wall-associated quality parameters that ultimately determine yarn quality.

Investigating the potential of Andean lupin as a lignocellulosic feedstock for Europe: First genome‐wide association study on Lupinus mutabilis biomass quality

AbstractThe development of multipurpose crops will drive the transformation of agricultural “waste” into added‐value products, helping to meet biomass demands without competing with food production or increasing environmental pressure. Lupinus mutabilis, has been proposed not only as a valid source of protein and oil for Europe but also as a possible source of lignocellulosic feedstock for the biorefinery industry. In this study, the quality of L. mutabilis lignocellulosic biomass and its genetic architecture are investigated for the first time, using a panel of 223 accessions planted across three locations in two different European cropping conditions. Biomass quality was evaluated based on the estimation of neutral detergent fiber, cellulose, hemicellulose and acid detergent lignin fractions, and on the basis of the monosaccharide composition of cell wall polysaccharides. The broad variation in yield and composition of biomass encountered in the panel confirms the potential of L. mutabilis as lignocellulosic feedstock and points out the value of this panel as a breeding tool for the improvement of biomass quality. A genome‐wide association study was conducted to identify single‐nucleotide polymorphisms (SNPs) associated with biomass quality, both across locations and per specific location. Scanning of 16,781 SNPs across the whole genome identified 46 unique quantitative trait loci for biomass quality, 4 of which were detected as common either among traits or GWAS models. For each of the traits analyzed, between 3 and 10 SNPs were detected, explaining 2.7%–15.9% of the phenotypic variation. Underlying these loci, 28 genes were proposed as candidate genes for biomass quality. Important genes involved in cellulose and sucrose synthesis (CESA4, SPP1,WRKY33, GONST2), monolignol biosynthesis (SKIP31, WAT1, CCR‐SNL6) and pectin degradation (RAV1, PE) were identified and will require validation to confirm their value for application in L. mutabilis breeding.

The Role of Cell Wall Polysaccharides Disassembly and Enzyme Activity Changes in the Softening Process of Hami Melon (Cucumis melo L.).

To investigate the physiological and molecular properties relating to cell wall carbohydrate metabolism in fruit, the ultrastructure and polysaccharides compositions of the cell wall, as well as the fruit quality and activities of enzymes relating to fruit softening, were studied for three Hami melon varieties (‘Xizhoumi 17’, ‘Jinhuami 25’, and ‘Chougua’) representing three different storability levels. The results showed that ‘Chougua’ maintained a higher firmness on day 18, with the lowest decay incidence (0%). ‘Chougua’ showed a better storage quality and intact cell wall structure. The molecular weight and monosaccharide composition of cell wall polysaccharides for Hami melons underwent great changes during storage, and the degradation of pectin polysaccharides was obvious, involving the depolymerization of macromolecular polymers accompanied by the production of new macromolecular polymers and composition changes in pectin monosaccharides (glucose, galactose, and arabinose) during the softening process of the Hami melons. Polygalacturonase, pectin methylesterase, xyloglucan endo-transglycosylase/hydrolase, α-arabinofuranosidase, β-galactosidase, and cellulase were associated with fruit softening at different stages of storage. There were similar softening mechanisms in the three Hami melons. This study will provide reference for further study on the fruit softening mechanisms of Hami melons.

Toward bioeconomy of a multipurpose cereal: Cell wall chemistry of Sorghum is largely buffered against stem sugar content

AbstractBackground and ObjectivesSorghum (Sorghum bicolor L. Moench) is a multipurpose crop with high potential for the bioeconomy. Ten sweet, grain and dual‐purpose sorghum genotypes were compared with respect to sugar‐related traits and cell wall composition.FindingsThe dual‐purpose hybrids Ganymed, Zerberus, and Tarzan performed better with respect to plant height, cane weight, and juice volume, but the sweet genotype KIT1 produced the highest sugar concentration (°Brix) in the stem. Analytical sugar data demonstrate genotypic differences in terms of sugar composition and concentrations, with sucrose being dominant. The monosaccharide composition of the stem cell wall polysaccharides showed surprisingly little genotypic variability. Variations in contents of lignin and cell wall‐bound hydroxycinnamate monomers were moderate.ConclusionsSweet, grain and dual‐purpose sorghum genotypes differ widely in terms of sugar‐related morphological parameters but are comparable with respect to their cell wall chemistry.Significance and NoveltyThe use of sorghum as a bioeconomy crop has mainly focused on the extraction of sugar for bioethanol production. However, besides cell wall polymers, the potential usage of hydroxycinnamates as platform molecules for the chemical industry may improve the valorization of the residues after sugar extraction. This application appears to be fairly independent of genotype, further increasing the potential of sorghum for the bioeconomy.

Profiling the cell walls of seagrasses from A (Amphibolis) to Z (Zostera)

BackgroundThe polyphyletic group of seagrasses shows an evolutionary history from early monocotyledonous land plants to the marine environment. Seagrasses form important coastal ecosystems worldwide and large amounts of seagrass detritus washed on beaches might also be valuable bioeconomical resources. Despite this importance and potential, little is known about adaptation of these angiosperms to the marine environment and their cell walls.ResultsWe investigated polysaccharide composition of nine seagrass species from the Mediterranean, Red Sea and eastern Indian Ocean. Sequential extraction revealed a similar seagrass cell wall polysaccharide composition to terrestrial angiosperms: arabinogalactans, pectins and different hemicelluloses, especially xylans and/or xyloglucans. However, the pectic fractions were characterized by the monosaccharide apiose, suggesting unusual apiogalacturonans are a common feature of seagrass cell walls. Detailed analyses of four representative species identified differences between organs and species in their constituent monosaccharide composition and lignin content and structure. Rhizomes were richer in glucosyl units compared to leaves and roots. Enhalus had high apiosyl and arabinosyl abundance, while two Australian species of Amphibolis and Posidonia, were characterized by high amounts of xylosyl residues. Interestingly, the latter two species contained appreciable amounts of lignin, especially in roots and rhizomes whereas Zostera and Enhalus were lignin-free. Lignin structure in Amphibolis was characterized by a higher syringyl content compared to that of Posidonia.ConclusionsOur investigations give a first comprehensive overview on cell wall composition across seagrass families, which will help understanding adaptation to a marine environment in the evolutionary context and evaluating the potential of seagrass in biorefinery incentives.

Zizania latifolia Cell Wall Polysaccharide Metabolism and Changes of Related Enzyme Activities during Postharvest Storage.

The metabolism of polysaccharides in the Zizania latifolia cell wall helps maintain the postharvest quality during storage. Fresh Z. latifolia was stored at 4 °C and 25 °C to evaluate the hardness, cell wall polysaccharide composition, cell wall structure, active ingredients, and cell wall metabolism-related enzyme activities. The results showed that hardness declined concomitantly with an increase in water-soluble pectin content during storage, as well as with a decrease in propectin and cellulose contents. Correlation analysis showed that lower activities of cell wall-degrading enzymes, such as polygalacturonase, cellulase, and β-galactosidase in Z. latifolia stored at 4 °C, were associated with lighter fiberization and greater hardness, compared with those stored at 25 °C. Additionally, the results of infrared spectroscopy showed that texture softening may be attributed to a decrease in the degree of esterification of water-soluble polysaccharides at 25 °C compared to that at 4 °C.

Subcellular coordination of plant cell wall synthesis.

Organelles of the plant cell cooperate to synthesize and secrete a strong yet flexible polysaccharide-based extracellular matrix: the cell wall. Cell wall composition varies among plant species, across cell types within a plant, within different regions of a single cell wall, and in response to intrinsic or extrinsic signals. This diversity in cell wall makeup is underpinned by common cellular mechanisms for cell wall production. Cellulose synthase complexes function at the plasma membrane and deposit their product into the cell wall. Matrix polysaccharides are synthesized by a multitude of glycosyltransferases in hundreds of mobile Golgi stacks, and an extensive set of vesicle trafficking proteins govern secretion to the cell wall. In this review, we discuss the different subcellular locations at which cell wall synthesis occurs, review the molecular mechanisms that control cell wall biosynthesis, and examine how these are regulated in response to different perturbations to maintain cell wall homeostasis.

Cell Wall Polysaccharide Composition of Grafted 'Liberty' Watermelon With Reduced Incidence of Hollow Heart Defect.

Grafting watermelon scions to interspecific squash hybrids has been found to increase fruit firmness. Triploid (seedless) watermelon are prone to hollow heart (HH), an internal fruit disorder characterized by a crack in the placental tissue expanding to a cavity. Although watermelon with lower tissue firmness tend to have a higher HH incidence, associated differences in cell wall polysaccharide composition are unknown. Grafting “Liberty” watermelon to “Carnivor” (interspecific hybrid rootstock, C. moschata × C. maxima) reduced HH 39% and increased tissue firmness by 3 N. Fruit with and without severe HH from both grafted and non-grafted plants were analyzed to determine differences in cell wall polysaccharides associated with grafting and HH. Alcohol insoluble residues (AIR) were sequentially extracted from placental tissue to yield water soluble (WSF), carbonate soluble (CSF), alkali soluble (ASF), or unextractable (UNX) pectic fractions. The CSF was lower in fruit with HH (24.5%) compared to those without HH (27.1%). AIRs were also reduced, hydrolyzed, and acetylated for GC-MS analysis of monosaccharide composition, and a portion of each AIR was methylated prior to hydrolysis and acetylation to produce partially methylated alditol acetates for polysaccharide linkage assembly. No differences in degree of methylation or galacturonic and glucuronic acid concentrations were found. Glucose and galactose were in highest abundance at 75.9 and 82.4 μg⋅mg–1 AIR, respectively, followed by xylose and arabinose (29.3 and 22.0 μg⋅mg–1). Mannose was higher in fruit with HH (p < 0.05) and xylose was highest in fruit from grafted plants (p < 0.05). Mannose is primarily found in heteromannan and rhamnogalacturonan I side chains, while xylose is found in xylogalacturonan or heteroxylan. In watermelon, 34 carbohydrate linkages were identified with galactose, glucose, and arabinose linkages in highest abundance. This represents the most comprehensive polysaccharide linkage analysis to date for watermelon, including the identification of several new linkages. However, total pectin and cell wall composition data could not explain the increased tissue firmness observed in fruit from grafted plants. Nonetheless, grafting onto the interspecific hybrid rootstock decreased the incidence of HH and can be a useful method for growers using HH susceptible cultivars.

Biochemical and Morphological Characterization of Heterotrophic Crypthecodinium cohnii and Chlorella vulgaris Cell Walls.

Microalgae are attractive for the food and cosmetic industries because of their nutrient composition. However, the bioaccessibility and extractability of nutrients in microalgae are limited by the rigid and indigestible cell wall. The goal of this study is to explore the cell wall polysaccharides (CWPSs) composition and morphology in heterotrophic Crypthecodinium cohnii and Chlorella vulgaris biomasses during growth. Our results showed that glucose was the major component of CWPSs and exopolysaccharides in C. cohnii. C. vulgaris CWPSs have a similar sugar profile in exponential and stationary phases, essentially composed of rhamnose and galactose. C. vulgaris cell wall thickness increased from 82 nm in the exponential phase to 114 nm in the stationary phase and consisted of two main layers. C. cohnii's cell wall was 133 nm thick and composed of several membranes surrounding thecal plates. Understanding of the microalgae cell wall helps developing a more efficient and targeted biorefinery approach.

A small molecule inhibits cell elongation by modulating cell wall polysaccharide composition in Arabidopsis

The plant primary cell wall is comprised of pectin, cellulose and hemicelluloses, whose dynamic interactions play essential roles in plant cell elongation. Through a chemical genetics screening, we identified a small molecule, named cell wall modulator (CWM), which disrupted cell growth and deformed cell shape in etiolated Arabidopsis hypocotyl. A pectin defective mutant qua2, identified from screening an Arabidopsis EMS mutant library, showed a reduced sensitivity to CWM treatment. On the other hand, pectinase treatment suppressed the CWM induced phenotype. Furthermore, cellulose content was decreased in response to CWM treatment, while the cellulose synthesis mutants ixr1 and ixr2 were hypersensitive to CWM. Together, the study identified a small molecule CWM that induced a modification of the cell wall in elongating cells, likely through interfering with pectin modification. This molecule may be used as a tool to study cell wall remodeling during plant growth.

A Novel Process for the Recovery of Betalains from Unsold Red Beets by Low-Temperature Enzyme-Assisted Extraction.

Food waste management plays a central role in the circular economy. To our knowledge, only a few studies have investigated the use of unsold fruit and vegetables from supermarkets as a substitute source for the extraction of natural colorants. Thus, the aim of this paper was to suggest a green, tailored protocol that avoids the use of organic solvents for the recovery of betalains from unsold red beets for use as a food colorant. The recovery of such pigments was carried out by a tailored enzymatic mix, blended considering the polysaccharide composition of the beetroot cell wall; thus, it consisted of: cellulase (37%), xylanase (35%), and pectinase (28%). The enzyme-assisted extraction protocol was optimized, and the most suitable conditions (in terms of pigment yield and color attributes) for the recovery of betalains from unsold beets appeared to be: 25 U/g total dose of enzymatic mix, temperature 25 °C, and processing time 240 min.
