Zero-valent iron (nZVI) nanoparticles mediate SlERF1 expression to enhance cadmium stress tolerance in tomato

Cadmium (Cd) pollution threatens plant physiological and biochemical activities and crop production. Significant progress has been made in characterizing how nanoparticles affect Cd stress tolerance; however, the molecular mechanism of nZVI nanoparticles in Cd stress remains largely uncharacterized. Plants treated with nZVI and exposed to Cd had increased antioxidant capacity and reduced Cd accumulation in plant tissues. The nZVI treatment differentially affected the expression of genes involved in plant environmental responses, including those associated with the ERF transcription factor. SlEFR1 was upregulated by Cd stress in nZVI-treated plants when compared with the control and the predicted protein-protein interactions suggested SlERF1 interacts with proteins associated with plant hormone signaling pathway and related to stress. Yeast overexpressing SlEFR1 grew faster after Cd exposure and significantly had higher Cd stress tolerance when compared with empty vector controls. These results suggest that nZVI induces Cd stress tolerance by activating SlERF1 expression to improve plant growth and nutrient accumulation. Our study reveals the molecular mechanism of Cd stress tolerance for improved plant growth and will support new research on overcoming Cd stress and improving vegetable crop production.

Pleiotropic melatonin-mediated responses on growth and cadmium phytoextraction of Brassica napus: A bioecological trial for enhancing phytoremediation of soil cadmium

Melatonin (MT) has recently gained significant scientific interest, though its mechanism of action in enhancing plant vigor, cadmium (Cd) tolerance, and Cd phytoremediation processes are poorly understood. Therefore, here we investigated the beneficial role of MT in improving growth and Cd remediation potential of rapeseed (Brassica napus). Plants, with or without MT (200 µM L−1), were subjected to Cd stress (30 mg kg1). Without MT, higher Cd accumulation (up to 99%) negatively affected plant growth and developmental feature as well as altered expression of several key genes (DEGs) involved in different molecular pathways of B. napus. As compared to only Cd-stressed counterparts, MT-treated plants exhibited better physiological performance as indicated by improved leaf photosynthetic and gaseous exchange processes (3–48%) followed by plant growth (up to 50%), fresh plant biomass (up to 45%), dry plant biomass (up to 32%), and growth tolerance indices (up to 50%) under Cd exposure. MT application enhanced Cd tolerance and phytoremediation capacity of B. napus by augmenting (1) Cd accumulation in plant tissues and its translocation to above-ground parts (by up to 45.0%), (2) Cd distribution in the leaf cell wall (by up to 42%), and (3) Cd detoxification by elevating phytochelatins (by up to 8%) and metallothioneins (by upto 14%) biosynthesis, in comparison to Cd-treated plants. MT played a protective role in stabilizing hydrogen peroxide and malondialdehyde levels in the tissue of the Cd-treated plants by enhancing the content of osmolytes (proline and total soluble protein) and activities of antioxidant enzymes (SOD, CAT, APX and GR). Transcriptomic analysis revealed that MT regulated 1809 differentially expressed genes (828 up and 981 down) together with 297 commonly expressed DEGs (CK vs Cd and Cd vs CdMT groups) involved in plant-pathogen interaction pathway, protein processing in the endoplasmic reticulum pathway, mitogen-activated protein kinase signaling pathway, and plant hormone signal transduction pathway which ultimately promoted plant growth and Cd remediation potential in the Cd-stressed plants. These results provide insights into the unexplored pleiotropic beneficial action of MT in enhancing in the growth and Cd phytoextraction potential of B. napus, paving the way for developing Cd-tolerant oilseed crops with higher remediation capacity as a bioecological trial for enhancing phytoremediation of hazardous toxic metals in the environment.

Soil applied silicon and manganese combined with foliar application of 5-aminolevulinic acid mediate photosynthetic recovery in Cd-stressed Salvia miltiorrhiza by regulating Cd-transporter genes.

Salvia miltiorrhiza is an important medicinal plant that experiences significant growth and biomass losses when cultivated on cadmium (Cd) contaminated soils. High Cd accumulation in plant tissues also increases the risk of metal entry into the food chain. In this study, we proposed that Cd accumulation in S. miltiorrhiza can be restricted through plant growth regulators and nutrient management. Therefore, S. miltiorrhiza seedlings were transplanted into mixed nutrient soil for two weeks, then treated with 30 mg kg-1 CdCl2, 200 mg kg-1 Na2SiO3·9H2O, and 100 mg kg-1 MnSO4, and simultaneously sprayed with 10 mg L-1 ALA on the leaves one week later. This study showed that elevated Cd accumulation significantly reduced plant growth and biomass. This growth inhibition damaged photosynthetic machinery and impaired carbon assimilation. In contrast, 5-aminolevulinic acid (ALA) significantly promoted the biomass of S. miltiorrhiza, and the dry weight of plants treated with ALA combined with manganese (Mn)/silicon (Si) increased by 42% and 55% as compared with Cd+Mn and Cd+Si treatments. Exogenously applied ALA and Si/Mn significantly activated antioxidant enzymes and promoted the growth recovery of S. miltiorrhiza. Further, exogenous ALA also reduced the Cd concentration in S. miltiorrhiza, especially when combined with Si. Compared with the Cd+Si treatment, the Cd+Si+ALA treatment reduced the Cd concentration in roots and leaves by 59% and 60%, respectively. Gene expression analysis suggested that ALA and Si significantly up-regulated genes associated with Cd transport. Other genes related to heavy metal tolerance mechanisms are also regulated to cope with heavy metal stress. These results indicated that the combined action of ALA and Si/Mn could reduce Cd-toxicity by increasing chlorophyll content and changing oxidative stress and can also affect Cd accumulation by regulating gene expression.

Oxalic Acid Mitigates Cadmium Toxicity in Cicer arietinum L. Germinating Seeds by Maintaining the Cellular Redox Homeostasis

The present study aimed to investigate the potential protective role of oxalic acid (OA) against Cadmium (Cd) toxicity. Chickpea (Cicer arietinum L.) seeds were exposed to 200 µM Cd stress for 6 days or to co-treatment with 200 µM Cd + 100 µM OA for 3 days following to 3-day Cd stress. The application of OA ameliorated the growth of both roots and shoots of Cd-treated seedlings. This effect was mediated by the restriction of Cd accumulation in plant tissues. Besides, malondialdehyde (MDA) and carbonyl group contents decreased in OA-treated seedlings, suggesting that OA reversed the Cd-induced oxidative stress and its detrimental effect on cell membrane integrity. These results were further confirmed by the reduction by 2- and 1.7-fold of hydrogen peroxide (H2O2) accrual in roots and shoots, respectively, when compared to Cd-challenged seedlings. Moreover, OA corrected the Cd-imposed imbalance of the glutathione redox state, mainly via the restoration of the glutathione pool. This achievement seems to be the result of the modulation of glutathione peroxidase (GPX) and glutathione reductase (GR) activities. Likewise, OA counteracted the adverse effect of Cd on nicotinamides redox state reflected by the restoration of the balance between oxidized [NAD(P)] and reduced [NAD(P)H] forms. Taken together, our results suggest that the exogenous supply of OA to germinating seeds can be a promising alternative to improve plant tolerance to heavy metal stress.

Effects of Co-Contamination of Microplastics and Cd on Plant Growth and Cd Accumulation

Microplastics (MPs) occur widely in terrestrial ecosystems. However, information on the interaction of MPs with metals in terrestrial ecosystems is lacking in the literature. The present study investigated the effects of two types of MPs (high-density polyethylene (HDPE) and polystyrene (PS)) with different dosages (i.e., 0, 0.1%, 1%, and 10%) on the uptake and effects of Cd in maize plants grown in an agricultural soil. Results showed that addition of Cd at a 5 mg/kg caused inhibited plant growth and resulted in high Cd accumulation in plant tissues. Polyethylene alone showed no significant phytotoxic effects, but a high-dose of HDPE (10%) amplified Cd phytotoxicity. Polystyrene negatively affected maize growth and phytoxicity further increased in the presence of Cd. Both HDPE and PS caused soil diethylenetriaminepentaacetic acid (DTPA)-extractable Cd concentrations to increase but did not significantly affect Cd uptake into plant tissues. In the soil without Cd addition, HDPE decreased soil pH, while PS did not significantly alter soil pH. However, in the soil spiked with Cd, both HDPE and PS increased pH. Overall, impacts on plant growth and Cd accumulation varied with MP type and dose, and PS induced substantial phytotoxicity. In conclusion, co-occurring MPs can change Cd bioavailability, plant performance, and soil traits. Our findings highlight the ecological impacts that could occur from the release of MPs into soil.

Interactions of microplastics and cadmium on plant growth and arbuscular mycorrhizal fungal communities in an agricultural soil

Microplastics (MPs) as emerging contaminants have attracted attention worldwide, but little is known on their interactions with metallic contaminants in soil-plant systems. Here, we investigated the interactions between MPs, i.e., polyethylene (PE) and polylactic acid (PLA), and cadmium (Cd) on plant performance and arbuscular mycorrhizal fungal community in an agricultural soil. PE showed no noticeable phytotoxicity, while 10% PLA decreased maize biomass and chlorophyll content in leaves. A significant interaction on root biomass occurred between PE and Cd, but not between PLA and Cd. Both PE and PLA caused increase in soil pH and DTPA-extractable Cd concentrations, but no alterations in Cd accumulation in plant tissues. Different numbers of endemic and total OTUs were observed in various treatments. The relative abundance of arbuscular mycorrhizal fungi (AMF) genera highly varied with MPs and Cd. MPs altered AMF community structure and diversity, depending on their type and dose. Coexisting Cd produced slight but significant interactions with MPs on the dominant AMF genera. Overall, plant growth and AMF community varied with MPs type and dose, Cd, and their interactions, and the high dose of PLA produced stronger phytotoxicity. In conclusion, coexisting MPs and Cd can jointly drive shifts in plant performance and root symbiosis, thereby posing additional risks for agroecosystems and soil biodiversity.

Integration of earthworms and arbuscular mycorrhizal fungi into phytoremediation of cadmium-contaminated soil by Solanum nigrum L

Arbuscular mycorrhizal fungi (AMF) and earthworms independently enhance plant growth, heavy metal (HM) tolerance, and HM uptake, thus they are potential key factors in phytoremediation. However, few studies have investigated their interactions in HM phytoextraction by hyperaccumulators. This study highlights the independent and interactive effects of earthworms and AMF on Solanum nigrum. Plants inoculated with either AMF or earthworms exhibited ameliorated growth via enhancement of productivity, metal tolerance, and phosphorus (P) acquisition. Co-inoculation with both had more pronounced effects on plant biomass and P acquisition in shoots, but not in roots, and in Cd-polluted soils it significantly promoted (P < 0.05) shoot biomass (20.7–134.6 %) and P content (20.4–112.0 %). AMF and earthworms increased Cd accumulation in plant tissues, but only AMF affected Cd partitioning between shoots and roots. Although AMF decreased root-to-shoot translocation of Cd at high Cd levels, this was counterbalanced by earthworms. Both AMF and its combination with earthworms enhanced Cd phytoavailability by altering Cd chemical fractions and decreasing pH. Co-inoculation increased Cd removal amounts up to 149.3 % in 120 mg kg−1 Cd-spiked soils. Interactions between the two organisms were synergistic in Cd phytoextraction. Thus, earthworm-AMF-plant symbiosis potentially plays an essential role in phytoremediation of HM-polluted soils.

Phytoremediation potential of Acorus calamus in soils co-contaminated with cadmium and polycyclic aromatic hydrocarbons

Phytoremediation is a promising technology for the remediation of sites co-contaminated with inorganic (heavy metal) and organic pollutants. A greenhouse experiment was conducted to investigate the independent and interactive effects of cadmium (Cd) and polycyclic aromatic hydrocarbons (PAHs) on the growth of the wetland plant Acorus calamus and its ability to uptake, accumulate, and remove pollutants from soils. Our results showed that growth and biomass of A. calamus were significantly influenced by the interaction of Cd and PAHs after 60 days of growth. The combined treatment of low Cd and low PAHs increased plant biomass and Cd accumulation in plant tissues, thus enhancing Cd removal. Dissipation of PAHs from soils was not significantly influenced by Cd addition or by the presence of plants. Correlation analysis also indicated a positive relationship between residual concentrations of phenantherene and pyrene (PAHs), whereas enzyme activities (dehydrogenase and polyphenol oxidase) were negatively correlated with each other. Cluster analysis was used to evaluate the similarity between different treatments during phytoremediation of Cd and PAHs. Our results suggest that A. calamus might be useful for phytoremediation of co-contaminated soil.

Brassinosteroids mitigate cadmium toxicity in cowpea plants

Anthropogenic activities and improper uses of phosphate fertilizers have led to an increase in cadmium concentrations in agricultural soils. Brassinosteroids are steroid hormones that are rapidly assimilated and metabolised with beneficial roles in physiological and biochemical processes in plants. Our aim was to ascertain whether exogenous treatment with 24-epibrassinolide (EBR) can mitigate the Cd toxicity, and whether this substance can reduce the Cd accumulation in plant tissues. Furthermore, the dose response to EBR was determined following exposure to Cd in Vigna unguiculata. The experiment was a completely randomised factorial design with two concentrations of Cd (0 and 500 μM) and three concentrations of EBR (0, 50, and 100 nM). Spraying plants exposed to Cd with EBR significantly reduced the concentrations of Cd and increased nutrient contents in all tissues. The EBR treatment caused significant enhancements in leaf, root, and total dry matter. Foliar application of EBR reduced the negative effects of Cd toxicity on chlorophyll fluorescence and gas exchange parameters. Pretreatment with EBR also increased contents of pigments in plants exposed to Cd, compared with the identical treatments without EBR. Cd elevated contents of oxidant compounds, inducing cell damages, while EBR significantly decreased the concentrations of these compounds. We confirmed that EBR mitigated the negative effects related to Cd toxicity, reduced the absorption and transport of Cd, and increased the contents of essential elements. In plants exposed to Cd, the most apparent dose response was found for 100 nM EBR, with beneficial repercussions on growth, gas exchange, primary photosynthetic processes, and photosynthetic pigments, which were intrinsically connected to lower production of oxidant compounds and cell damage.

The Combination of DGT Technique and Traditional Chemical Methods for Evaluation of Cadmium Bioavailability in Contaminated Soils with Organic Amendment.

Organic amendments have been proposed as a means of remediation for Cd-contaminated soils. However, understanding the inhibitory effects of organic materials on metal immobilization requires further research. In this study colza cake, a typical organic amendment material, was investigated in order to elucidate the ability of this material to reduce toxicity of Cd-contaminated soil. Available concentrations of Cd in soils were measured using an in situ diffusive gradients in thin films (DGT) technique in combination with traditional chemical methods, such as HOAc (aqua regia), EDTA (ethylene diamine tetraacetic acid), NaOAc (sodium acetate), CaCl2, and labile Cd in pore water. These results were applied to predict the Cd bioavailability after the addition of colza cake to Cd-contaminated soil. Two commonly grown cash crops, wheat and maize, were selected for Cd accumulation studies, and were found to be sensitive to Cd bioavailability. Results showed that the addition of colza cake may inhibit the growth of wheat and maize. Furthermore, the addition of increasing colza cake doses led to decreasing shoot and root biomass accumulation. However, increasing colza cake doses did lead to the reduction of Cd accumulation in plant tissues, as indicated by the decreasing Cd concentrations in shoots and roots. The labile concentration of Cd obtained by DGT measurements and the traditional chemical extraction methods, showed the clear decrease of Cd with the addition of increasing colza cake doses. All indicators showed significant positive correlations (p < 0.01) with the accumulation of Cd in plant tissues, however, all of the methods could not reflect plant growth status. Additionally, the capability of Cd to change from solid phase to become available in a soil solution decreased with increasing colza cake doses. This was reflected by the decreases in the ratio (R) value of CDGT to Csol. Our study suggests that the sharp decrease in R values could not only reflect the extremely low capability of labile Cd to be released from its solid phase, but may also be applied to evaluate the abnormal growth of the plants.

Macroelemental composition of cadmium stressed lettuce plants grown under conditions of intensive sulphur nutrition

Lettuce (Lactuca sativa L.) is moderately sensitive to cadmium (Cd) and shows high accumulation of this metal. Thus, this species is considered to be a good model for both identifying determinants controlling Cd accumulation in plant tissues and for developing breeding strategies aimed at limiting the accumulation of this metal in edible tissues. Simultaneously, lettuce is characterised by medium requirements for sulphur (S) – a macronutrient whose role is associated not only with proper growth and development, but also with stress tolerance. The common use of NPK fertilizers without sulphates (S-SO4) together with the progressive process of reducing emissions of S compounds to the natural environment may lead to deficiency of this element in plants. The present study evaluated the changes in macronutrient content and accumulation in Cd-stressed lettuce ‘Justyna’ supplied with different S doses. Four concentrations of Cd (0, 0.0002, 0.02 or 0.04 mM) and three levels of S applied in the form of S-SO4 (2, 6 or 9 mM S) were used. Cd exposure impaired the macronutrient balance and accumulation in lettuce. Intensive S nutrition to some extent alleviated Cd-induced toxicity. High S doses, especially 6 mM S, partially improved macronutrient status and restored the macronutrient balance. In Cd-stressed plants supplemented with additional S, an increase in root and shoot biomass and in the content of N, K and Mg was found, without significant changes in the Ca content. Simultaneously, the P and S contents in the biomass of both above- and underground organs remained unchanged. In the leaves, as opposite to the roots, intensive S nutrition reduced the accumulation of Cd. However, the foliar Cd concentration still exceeded the acceptable limits established for consumption. All the obtained results concerning the content of macronutrients and their ratios were referred, inter alia, to the standards i.e. the Diagnosis and Recommendation Integrated System (DRIS) norms.

Effects of drought on cadmium accumulation in peanuts grown in a contaminated calcareous soil.

This study aimed to investigate the effects of drought stress on cadmium (Cd) accumulation in peanut (Arachis hypogaea L.) grown in contaminated calcareous soils. Five peanut cultivars were grown in a calcareous soil spiked with 4 mg Cd kg(-1) soil (dry weight) under well-watered, mild drought, and severe drought conditions. The biomass production, gas exchange, spectral reflectance, and Cd accumulation in plant tissues were determined. The five cultivars significantly differed from each other in biomass production, gas exchange, spectral reflectance, and Cd accumulation. The effect of drought on Cd accumulation in peanuts varies with plant tissues, cultivars, and developmental stages. Drought decreased root Cd concentrations in seedlings of the two high Cd-accumulating cultivars (Haihua 1 and Zhenghong 3), which is associated with increasing leaf active Fe content. However, for the mature plants, drought stress caused an increase in Cd accumulation in roots, pod walls, and seeds depending on peanut cultivars. Negative correlations were found between seed Cd concentration and biomasses in both preflowering seedlings and mature plants. The seed Cd concentration in mature plants was also observed to be positively correlated with the shoot Cd concentration in preflowering seedlings. The increased Cd concentration in seeds of drought-stressed peanut plants grown in Cd-contaminated calcareous soils might be attributed to the drought-induced decrease of biomass production.

Relationship between biomass, seed components and seed Cd concentration in various peanut (Arachis hypogaea L.) cultivars grown on Cd-contaminated soils

Peanuts (Arachis hypogaea L.) exhibit high genotypic variations in seed Cd accumulation, but the mechanism remains unclear. This study aimed to reveal the main factors that determine Cd concentration in peanut seeds. The biomasses and Cd accumulation in plant tissues as well as the Cd distribution in the seeds of 15 peanut cultivars were analyzed in a pot experiment at 4mgkg−1 Cd (treatment) and 0mgkg−1 Cd (control). Peanuts exhibited large variations among cultivars in terms of Cd accumulation and distribution at the whole-plant and seed levels. The peanut cultivars were divided into three groups based on [Cd]embryos as follows: (i) high Cd accumulators (Zhenghong 3 and Haihua 1), (ii) low Cd accumulators (Qishan 208, Luhua 8, and Yuhua 15), and (iii) intermediate Cd accumulators (10 remaining cultivars). [Cd]embryos was significantly correlated with [Cd]testae and [Cd]oils at control conditions, whereas in the 4mgkg−1 Cd treatment, [Cd]embryos was negatively correlated with plant biomass, total Cd and its proportion in vegetative organs, and seed oil contents. [Cd]embryos was positively correlated with protein contents, [Cd]oils, and proportion of Cd in protein extracts at 4mgkg−1 Cd treatments. The attenuation of Cd by high biomass of vegetative tissues and Cd-binding proteins in seeds mainly determined the Cd concentration in peanut seeds.

Uptake and Bioaccumulation of Pentachlorophenol by Emergent Wetland Plant Phragmites australis (Common Reed) in Cadmium Co-contaminated Soil

Despite many studies on phytoremediation of soils contaminated with either heavy metals or organics, little information is available on the effectiveness of phytoremediation of co-occurring metal and organic pollutants especially by using wetland species. Phragmites australis is a common wetland plant and its potential for phytoremediation of cadmium pentachlorophenol (Cd-PCP) co-contaminated soil was investigated. A greenhouse study was executed to elucidate the effects of Cd (0, 10, and 20 mg kg−1) without or with PCP (0, 50, and 250 mg kg−1) on the growth of the wetland plant P. australis and its uptake, accumulation and removal of pollutant from soils. After 75 days, plant biomass was significantly influenced by interaction of Cd and PCP and the effect of Cd on plant growth being stronger than that of PCP. Coexistence of PCP at low level lessened Cd toxicity to plants, resulting in improved plant growth and increased Cd accumulation in plant tissues. The dissipation of PCP in soils was significantly influenced by interactions of Cd, PCP and plant presence or absence. As an evaluation of soil biological activities after remediation soil enzyme was measured.

Studies of moleculargenetic mechanisms of Cd tolerance and accumulation in plants are the theoretical basis for the development of technologies for phytoremediation of soils contaminated with heavy metals and provide for the breeding of crop varieties with reduced Cd accumulation.

Polynuclear aromatic hydrocarbons (PAHs) mediate cadmium toxicity to an emergent wetland species

Growth and pollutant removal by emergent wetland plants may be influenced by interactions among mixed pollutants in constructed wetlands. A glasshouse experiment was conducted to investigate interactive effects of cadmium (Cd) × polynuclear aromatic hydrocarbons (PAHs) × plant treatments on growth of Juncus subsecundus, Cd and PAH removal from soil and the total number of microorganisms in soil. Growth and biomass of J. subsecundus were significantly influenced by interaction of Cd and PAHs, significantly decreasing with either Cd or PAH additions, but with the effect of Cd on plant growth being stronger than that of PAHs. The mixture of low Cd and low PAH lessened Cd toxicity to plants, resulting in improved plant growth and increased Cd accumulation in plant tissues, thus enhancing Cd removal by plants. The dissipation of PAHs in soils was significantly influenced by interactions of Cd, PAH and plant presence or absence. The total number of microorganisms in soils was significantly increased by the PAH additions. The interactive effect of Cd and PAHs on plant growth may be linked to the changes in the abundance of microorganisms in the rhizosphere, probably via a positive effect of PAH metabolites and/or phytohormones produced by microorganisms on plant growth.
