Anti-inflammatory and antinociceptive effects, and safety toxicological profile of a new paracetamol analog, LQFM291.

In the scope of a research program with the goal of developing treatments for inflammatory diseases, the pharmacological evaluation of LQFM291, designed by molecular hybridization from butylated hydroxytoluene and paracetamol, was described. The antioxidant profile of LQFM291 was evaluated by electrochemical measurement. Also, acute or repeated treatments with equimolar doses to paracetamol were used to evaluate the antinociceptive and/or anti-inflammatory activities of LQFM291 in animal models. The toxicologic potential of LQFM291 was also evaluated and compared to paracetamol through biochemical and histopathological analysis after the repeated treatment schedule. As a result of the acute treatment, paracetamol showed a similar antinociceptive effect in formalin test compared to LQFM291. Whereas, after the repeated treatment, when carrageenan-induced hyperalgesia and edema tests were performed, paracetamol showed a delayed antinociceptive and anti-inflammatory effect compared to LQFM291. Furthermore, as other advantages the LQFM291 showed a high redox capacity, a gastroprotective activity and a safety pharmacological profile without any liver or kidney damage. These effects can be related to the prevention of oxidative stress by reduction of protein and lipid peroxidation in gastric tissue, maintenance of glutathione levels in hepatic homogenate, and a systemic reduction of pro-inflammatory cytokine levels, which may characterize the LQFM291 as a more viable and effective alternative to relief pain and inflammatory signs in patients with chronic disorders.

Anti-Arthritic Evaluation of Ginger, Colchicum and Detoxified Nux-Vomica combination for Poly Herbal Unani Formulation

Introduction: Arthritis (Wajaul Mafasil) is a condition which is growing worldwide due to lifestyle, environmental and genetic factors. In Unani literature, there are many herbs which are praised for treatment of Arthritis. So polyherbal formulation contains Ginger, Colchicum and Nux- Vomica is taken in combination for arthritis study. This combination is not previously reported but used by unani practitioners. Method: Three crude herbs (Ginger, Colchicum and Nux vomica) were extracted out in both aqueous and hydro-alcoholic solvent. LD-50 of all extracts (aqueous and hydro-alcoholic extract) was determined. Now respective extracts were mixed in effective dose ratio to obtain aqueous and hydro-alcoholic dosage form. Finally both effective combinations (aqueous and hydro-alcoholic) convert into tablet dosage form to determine its anti-arthritic activity by Carrageenan Induced Oedema Test, Cotton Pellet Induced Granuloma Test and Freund’s adjuvant Induced Arthritis Test. The efficacy of the Unani formulation was compared with reference drug (Diclofenac sodium). Result and Discussion: in Carrageenan Oedema Test, animals in high dose hydro-alcoholic, shows decrease in paw volume significantly after 3 hours of Carrageenan injection. In Cotton Pellet Induced Granuloma Test, animals in high dose hydro-alcoholic, shows reduction in granuloma formation significantly. In Freund’s Adjuvant Induced Arthritis Test, The significant reduction in paw volume was found in high dose hydro-alcoholic. Conclusion: Conclusively this study establishes anti-arthritic potential of polyherbal extract in unani literature. Thus these drugs possess synergistic anti-arthritic potential in combination against acute, sub-acute as well as chronic arthritis. So in future compatible dosages form may be prepared for treating arthritis.

Regulation of the redox signaling and inflammation by Terminalia myriocarpa leaves and the predictive interactions of it's major metabolites with iNOS and NF-ĸB

Ethnopharmacological relevanceThe present study was designed to investigate the regulation of the redox signaling and inflammation by ethanolic leaf extract of Terminalia myriocarpaVan Heurck & Müller (ETM), inspired by the reported antioxidant potential of the plant bark and the anti-edema effect of the same genus. Materials and methodsHPLC-DAD dereplication study was conducted to detect the major polyphenolic secondary metabolites. In-vitro DPPH free radical scavenging assay, nitric oxide (NO) scavenging assay, Fe2+ ion chelating ability assay and reducing power assay were conducted to evaluate the antioxidant capacity. The molecular mechanism of anti-inflammation was investigated via assessing the NO and NF-ĸB inhibiting properties in different cell lines. In-vivo carrageenan and histamine-induced edema tests were conducted using established animal models. Pro-inflammatory proteins iNOS and NF-κB were docked against the major metabolites of ETM in the in-silico study. ResultsHPLC dereplication analysis revealed the presence of considerable amount of ellagic acid, where methyl-(S)-flavogallonate was previously reported in T. myriocarpa. Significant antioxidant activity was found in every in- vitro redox assay conducted. NO was reduced in RAW 264.7 cells, showing 83.67 ± 4.18% inhibitory activity at the highest tested concentration. TNF-α induced NF-κB was also observed to be reduced in 293/NF-кB-luc cells with an inhibitory activity of 66.23 ± 0.81% at the highest dose tested. In-vivo carrageenan-induced edema test demonstrated significant anti-inflammatory activity (p < 0.05; p < 0.01) at both doses of 250 and 500 mg/kg with 60.10% highest reduction in rat paw volume. Using same doses, histamine-induced edema test exhibited mentionable anti-inflammatory potential (p < 0.05; p < 0.01) with 67.91% highest reduction in rat paw volume. Moreover, ellagic acid and methyl-(S)-flavogallonate showed significant binding affinity with iNOS (−8.5 and −8.7 Kcal/moL, respectively) and NF-κB (−7.3 and −7.3 Kcal/moL, respectively). ConclusionMentionable basis was found on behalf of the anti-inflammatory and antioxidant potentials of ETM which might be correlated with its NF-ĸB inhibiting properties.

Phytochemical composition and pharmacological assessment of callus and parent plant of Asteriscus graveolens (Forssk.) Less. from Algerian Sahara

Purpose: To determine the phytochemical composition, acute toxicity, as well as analgesic and antiinflammatory properties of extracts of the aerial parts and callus cultures of Asteriscus graveolens.Methods: Different standard chemical tests were used for the screening of bioactive compounds in aqueous extracts of aerial parts of Asteriscus graveolens (PAE) and callus (CAE). High-performance liquid chromatography (HPLC) analysis was performed to identify the constituents of the glycosidic extract (GE). Acute oral toxicity test was carried out to ascertain the safety of the plant material. The analgesic and anti-inflammatory properties of the extracts were determined using acetic acid-induced writhing test and carrageenan-induced edema test, respectively.Results: Phytochemical analysis showed some differences in composition between the aerial parts and callus cultures. Results from HPLC analysis revealed the presence of 12 phenolic compounds in the GE of adult plants. In addition to being orally safe (no mortality), all extracts showed significant inhibition of acetic acid-induced chemical pain: PAE and GE produced 63 and 68 % reductions in pain, respectively, relative to 60.26 % pain reduction by aspirin. All extracts exerted potent anti-inflammatory effects, with PAE producing the highest effect (73 %) at a dose of 100 mg/mL.Conclusion: Asteriscus graveolens extracts possess potent pharmacological properties due to the presence of some phenolic compounds, especially flavonoids. These findings provide a scientific basis for the traditional uses of Asteriscus graveolens, and indicate that it is a potential source of antiinflammatory agents.&#x0D; Keywords: Acute toxicity, Anti-nociceptive, Anti-inflammatory, Aqueous extract, Callus, Flavonoids

Neo-clerodane diterpenic influence in the antinociceptive and anti-inflammatory properties of Salvia circinnata Cav.

Ethnopharmacological relevanceMexico is considered an ancestral center of diversity of Salvia species, however many of them lack scientific information. Salvia circinnata Cav. (syn. Salvia amarissima Ortega) is an endemic species used in traditional medicine to treat disorders attributed to a cold state like anxiety in the central nervous system, as well as gastrointestinal ailments and pain relief. Aim of the studyTo give preclinical evidence about the pharmacological properties of this species by investigating its antinociceptive and anti-inflammatory effects, the chemical nature of at least one metabolite, and a possible mechanism of action and adverse effects, using different experimental models of pain. Material and methodsDifferent crude extracts of Salvia circinnata Cav. aerial parts were prepared using increasing polarity and evaluated in the formalin test in mice. This screening allowed to select and evaluate an ethyl acetate extract (EtOAc), as the most bioactive extract, and a metabolite. Antinociceptive and anti-inflammatory activities were confirmed using the plantar test and carrageenan-induced edema. The antinociceptive effects of the extracts were compared to that observed with morphine (1 mg/kg), tramadol (20 mg/kg) or indomethacin (20 mg/kg) as reference drugs. Participation of opioids and TRPV1 receptors was investigated, as well as acute toxicity and adverse effects of sedation and gastric damage. ResultsEtOAc (0.1–10 mg/kg) of S. circinnata Cav. showed a dose-dependent and significant antinociceptive activity, associated in part with the presence of a neo-clerodane glycoside amarisolide A (0.01–1 mg/kg), in the neurogenic and inflammatory phases of the formalin test. Central action of both treatments was corroborated in the plantar test, whereas anti-inflammatory effects were confirmed with the extract (1 and 10 mg/kg) and amarisolide A (1 mg/kg) in the carrageenan-induced edema test. An opioid mechanism in both treatments, and the TRPV1 receptor modulation in the extract were involved. No acute toxicity and adverse effects were noticed with the extract and pure compound in comparison to the reference drugs. ConclusionThese results provide preclinical evidence of the ethnopharmacological antinociceptive S. circinnata Cav. properties, in which the neo-clerodane diterpene glycoside amarisolide A was partially responsible involving the participation of the opioid receptors, while TRPV1 receptor modulation was implicated in the anti-inflammatory activity may be because of the presence of other constituents. This information supports the use of this species in folk medicine for pain therapy.

Beneficial effects of (-)-epigallocatechin-3-O-gallate on diabetic peripheral neuropathy in the rat model.

Diabetic neuropathic pain is characterized by spontaneous pain with hyperalgesia and allodynia. We investigated whether (-)-epigallocatechin-3-O-gallate could improve diabetic neuropathic pain development through hypoglycemic, hypolipidemic, antioxidant, and anti-inflammatory effects. Diabetes was induced in rats by streptozotocin (55 mg/kg/once) and treated with (-)-epigallocatechin-3-O-gallate (25 mg/kg/orally/once/daily/5 weeks). Diabetic rats showed an increase in serum levels of glucose, nitric oxide, triglyceride, total cholesterol, and low-density lipoprotein-cholesterol with a decrease in high-density lipoprotein-cholesterol and body weight. Also, there was an elevation in brain malondialdehyde with a marked reduction in brain levels of glutathione, superoxide dismutase, catalase, glutathione peroxidase, and glutathione-S-transferase. Furthermore, diabetic rats showed a clear reduction in plasma levels of insulin and an increase in plasma cytokines (interleukin-6 and tumor necrosis factor-α). Moreover, diabetic rats exhibited hyperalgesia as indicated by a hot plate, tail immersion, formalin, and carrageenan-induced edema tests as well as brain histopathological changes (neuron degeneration, gliosis, astrocytosis, congestion and hemorrhage). (-)-Epigallocatechin-3-O-gallate treatment ameliorated alterations in body weight, biochemical parameters, pain sensation, and histopathological changes in brain tissue. (-)-Epigallocatechin-3-O-gallate offers promising hypoglycemic, hypolipidemic, antioxidant and anti-inflammatory effects, which can prevent the development and progression of diabetic neuropathic pain.

Evaluation of Antioxidant, Anti-inflammatory and Analgesic Activities of Leaves of Saba senegalensis (A.DC) Pichon (Apocynaceae)

Aims: To evaluate antioxidant, anti-inflammatory and analgesic activities of extracts of Saba senegalensis leaves.&#x0D; Study Design: In vitro antioxidant assay and in vivo anti-inflammatory, analgesic assay of Saba senegalensis extracts.&#x0D; Place and Duration of Study: Saba senegalensis leaves, were collected in the Centre Region of Burkina Faso, in June–July 2015. The experiments were conducted at the department of Medicine and Traditional Pharmacopeia-Pharmacy (MEPHATRA-PH) of Institute of Research in Health Science (IRSS).&#x0D; Methodology: The anti-oedematous tests with carrageenan and the analgesic with acetic acid and investigate effect on isolated organ were carried out. The standards were acetylsalicylic acid and paracetamol.&#x0D; Results: After five hours of carrageenan-induced edema test, aqueous decoction (AD) presented better inhibition on all measure. In fact, at the different doses of 200 mg/kg, 400 mg/kg, and 600 mg/kg it presented percentages of inhibitions respectively of 30.81%, 62.27% and 72.71%. For the analgesic test, the hydroethanol macerate (HEM) showed a better pain reduction compared to the AD with a maximum effect of 77.28% at 400 mg/kg. Antioxidant activity with AD and his fractions shows that AD showed a better activity for the DPPH assay with an IC50 of 1.74 ± 0.10 μg/mL and a reducing power of 59.53 ± 2.16 mmol ET/g Sample. For HEM and his fractions, the ethyl acetate fraction (FHEM-AcOEt) showed a better IC50 of 0.18 ± 0.01 μg/mL for the DPPH test and dichloromethane fraction (FHEM-DCM) a reducing power agent of 88.88 ± 2.65 mmol ET/g Sample. All fractions were endowed with antioxidant properties by both methods.&#x0D; Conclusion: The study findings suggest that the presence of phenolic and terpenoid compounds could explain the antioxidant, anti-inflammatory and analgesic properties of these extracts.

Evaluation of Pharmacological Activities of Leaves and Bark of Myristica fatua var. magnifica Extracts

Myristica fatua var. magnifica (Beddome) Sinclair belongs to the family Myristicaceae. It is a tree used as an important plant to cure various diseases. In the present study the Petroleum ether, Ethyl acetate and Methanolic extracts of Myristica fatua was evaluated for its toxicity, potential analgesic, depressant and in vitro and in vivo anti-inflammatory activity. The extracts at different concentration from 100 to 2000 mg/kg were used to evaluate the toxicity in albino rats. For evaluating the analgesic, depressant, in vitro and in vivo anti-inflammatory activity, tail flick, actophotometer, rota-rod, HRBC membrane and carrageenan induced edema test in selected animals were done at the dose 200 and 400 mg/kg. Acute toxicity studies revealed that the extracts of Myristica fatua var. are nontoxic at the studied concentration. The appropriate dose range for preclinical study was found to be 200 and 400 mg/kg. Oral administration of different extracts at the dose of 200 and 400 mg/kg exhibited dose dependent analgesic, depressant and in vivo anti-inflammatory activity in selected animals. In vitro anti-inflammatory studies showed dose dependent activity with percentage of stabilization was found to be 79.66%, 80.63% and 85.22% at concentration of 200 mg/ml of M. fatua leaves, bark and standard respectively. Hence present investigation established some pharmacological evidences to support the folklore claim that Myristica fatua var. magnifica is used as analgesic, depressant and anti-inflammatory agent.

Preventive treatment with dizocilpine attenuates oedema in a carrageenan model of inflammation: the interaction of glutamatergic and nitrergic signaling

Dizocilpine is a highly selective and potent non-competitive antagonist of the N-methyl-D-aspartate (NMDA) glutamate receptor. It is well known that dizocilpine has different neuroprotective effects in animal models of pain, epilepsy and oedema during trauma. The search for alternative antiinflammatory drugs is ongoing. We investigated the anti-oedematous effects of dizocilpine and the probable mechanism of action in a rat model that mimics local and persistent inflammation without tissue injury or damage. Male Wistar rats were injected with 100μL of 0.5% carrageenan to the plantar surface of the hind paw. Anti-oedematous activity was assessed in the carrageenan-induced paw inflammatory oedema test with a plethysmometer. To assess possible mechanisms of dizocilpine action, we examined the effects of the selective inhibitor of neuronal [N-ω-propyl-L-arginine hydrochloride (L-NPA)] and inducible [S-methylisothiourea (SMT)] nitric oxide synthase (NOS). Dizocilpine after systemic (0.0005, 0.005 and 0.02mg/kg, subcutaneous (s.c.)), but not after local peripheral administration, reduced the paw inflammatory oedema. The effect is not dose dependent, and the highest decrease by about 47% at the time of maximally developed oedema was achieved with 0.005mg/kg. Intraperitoneally (i.p.) administered L-NPA (0.5, 1 and 2mg/kg) or SMT (0.005, 0.01 and 0.015mg/kg) before dizocilpine abolished or reduced the anti-oedematous effect of dizocilpine by about 70-85%. An acute single dose of dizocilpine administered before inducing oedema systemically reduced the development of inflammatory oedema. The mechanism of the anti-oedematous effect includes, at least partially, an increase in nitric oxide (NO) production.

Anti-nociceptive and Anti-inflammatory Activities of Crotalaria pallida Aiton (Fam: Fabaceae) Leaves

The present study was designed to evaluate the anti-nociceptive and anti-inflammatory properties of the ethanolic extract of Crotalaria pallida Aiton (Family: Fabaceae) leaves based on its traditional uses in the treatment of urinary disorders, external application as a poultice to treat painful swelling of joints and to reduce fever. The anti-nociceptive effect was assessed in mice using acetic acidinduced writhing, formalin-induced paw licking and tail immersion assays. Anti-inflammatory activity was evaluated by carrageenan induced paw oedema in rats. In acetic acid-induced writhing test, the extract at different doses (50 mg/kg and 100 mg/kg bw) significantly (p&lt;0.001) and dose-dependently reduced pain by 37.50% and 47.70%, respectively. The extract also significantly inhibited formalininduced paw licking in mice at both the early and late phases. In the tail immersion test, the extract caused a significant inhibition of pain (64.91% inhibition after 4 hrs) at a dose of 100 mg/kg. C. pallida exhibited a considerable inhibition of paw oedema development in the carrageenan-induced oedema test. The findings of the study suggested that C. pallida leaves have significant anti-nociceptive and antiinflammatory effects, confirming the traditional uses of this plant in the treatment of various diseases associated with pain and inflammation.Bangladesh Pharmaceutical Journal 20(2): 165-171, 2017

Evaluation of the efficacy of Hylotelephium purpureum gel in the treatment of experimental periodontitis.

Local drug delivery systems have been proposed for the treatment of periodontitis, and Hylotelephium purpureum may be a suitable agent for use in such a system. The present study aimed to formulate a Hylotelephium purpureum gel (HPG) and evaluate the anti-inflammatory activity and antinociceptive effects of Hylotelephium purpureum and to assess the duration of action and efficacy of HPG in the treatment of experimental periodontitis in a KM mouse model. First, an acute toxicity study was performed, and secondly, xylene-induced mouse-ear edema, acetic acid-induced mouse peritoneal capillary permeability and carrageenan-induced hind-paw edema tests were used to investigate the anti-inflammatory activity of the gel. The acetic acid-induced writhing response and hot-plate tests were used to evaluate the antinociceptive activity. The therapeutic effects of HPG in experimental periodontitis were evaluated and minimum inhibitory concentration of the gel was determined. The results showed that intragastrically administration of 80 ml/kg of HPG produced no toxic effects. There were statistically significant changes in gingival indexes and sulcus bleeding indexes in the high-dose HPG-treated group. The serum levels of superoxide dismutase and glutathione peroxidase were significantly heightened, while the level of malondialdehyde was decreased. HPG exhibited 32.7% inhibition of edema, and altered the peritoneal capillary permeability in mice. The gel had relatively good bacteriostatic and bactericidal effects. It also exhibited antinociceptive activity, as demonstrated by the acetic acid-induced writhing response test and hot-plate test. In summary, the present study demonstrates that HPG is effective in the treatment of experimental periodontitis.

Evaluation of Analgesic and Anti-Inflammatory Activities of Water Extract of Galla Chinensis In Vivo Models.

Aim Pain and inflammation are associated with many diseases in humans and animals. Galla Chinensis, a traditional Chinese medicine, has a variety of pharmacological properties. The purpose of this study was to evaluate analgesic and anti-inflammatory activities of Galla Chinensis through different animal models. Method The analgesic activities were evaluated by hot-plate and writhing tests. The anti-inflammatory effects were assessed by ear edema, capillary permeability, and paw edema tests. The contents of cytokines (NO, iNOS, PGE2, and IL-10) in serum of rats in paw edema test were inspected by ELISA assays. Results In the hot-plate test, Galla Chinensis could significantly extend pain threshold when compared to control group. The inhibitory rates of writhes ranged from 36.62% to 68.57% in Galla Chinensis-treated mice. Treatment with Galla Chinensis (1 and 0.5 g/kg) could significantly inhibit ear edema (47.45 and 36.91%, resp.; P < 0.01). Galla Chinensis (1 g/kg) had significant (P < 0.05) anti-inflammatory activity in capillary permeability test (29.04%). In carrageenan-induced edema test, the inhibitory rates were 43.71% and 44.07% (P < 0.01) at 1 h and 2 h after administration of Galla Chinensis (1 g/kg), respectively, and the levels of proinflammatory cytokines were significantly reduced. Conclusion These results suggest that Galla Chinensis has analgesic and anti-inflammatory effects, which may be a candidate drug for the treatment of inflammation and pain.

Anti-inflammatory and antioxidant potential of Dichrostachys cinerea root bark, an Ivorian anti-asthmatic herbal

Objectives : root bark of Dichrostachys cinerea is used in Ivorian folk for the treatment of asthma. This study aimed to evaluate anti-inflammatory and antioxidant activities of this plant. Methods : an aqueous-alcoholic extract was obtained from the root bark. Two tests i.e. rat's paw irritation with formalin and rat's paw oedema induced by carrageenan were used to appreciate the anti-inflammatory activity. The antioxidant activity was performed using ABTS (2.2-azinobis 3 ethyl-benzothiazoline-6-sulfonic acid), DPPH (1.1 diphenyl 1-2-picrylhydrazyl), and FRAP (Ferric Reducing Antioxidant Power) tests. Results : the extract, only at 1000 mg/kg b.wt, reduced significantly the licking time (22.03 1.08 s) in the second period of formalin-induced irritation test. Concerning the carrageenan-induced oedema test, oedema was weakly reduced 1, 2, and 3 hours after injection of carrageenan. Indeed, levels of inhibition were respectively 1.71 0.56 %, 1.45 0.70%, 4.5 1.91% for the extract at 10 mg/kg b.wt; 3.71 0.33%, 6.50 0.10%, 10.75 0.42% for the extract at 100 mg/kg b.wt. and 3.47 0.40%, 10.54 0.48%, 13.50 0.93 for the extract at 1000 mg/kg b.wt. The extract did not exert any antioxidant activity through both ABTS and DPPH methods, but with FRAP one, low doses showed a high antioxidant activity (86.33 1.78%) similar to that of Trolox (90.99 6.47%) used as reference. Conclusion : the herbal lacks anti-inflammatory activity. However, it induced an antioxidant activity which could be recommended to neutralize the free radicals involved in the bronchial hyperactivity phenomena during acute asthma.

Antinociceptive, anti-inflammatory and anxiolytic-like effects of the ethanolic extract, fractions and Hibalactone isolated from Hydrocotyle umbellata L. (Acariçoba) – Araliaceae

Hydrocotyle umbellata Linn. (Araliaceae) is specie used in the treatment of inflammatory diseases. Crude extract (E-HU) was prepared from H. umbellata subterraneous parts and fractionated by liquid-liquid partition, resulting hexane fraction (HF-HU), dichloromethane fraction (DF-HU), ethyl acetate fraction (EAF-HU) and aqueous fraction (AF-HU). The hibalactone (HU-1) was isolated from the DF-HU and its structure was elucidated by 1H NMR and 13C NMR Spectroscopy, mass spectrometry and crystallographic x-ray analysis. The formalin-induced nociception was used to evaluate antinociceptive activity; carrageenan-induced edema and pleurisy tests to evaluate anti-inflammatory activity and light-dark box to evaluate anxiolytic-like activity in mice. The acute oral treatments with E-HU (1000mg/kg), DF-HU (150mg/kg), EAF-HU (400mg/kg) and HU-1 (33mg/kg) decreased the licking time in both phases of the formalin test. In the carrageenan-induced inflammation models, the treatment with the same doses of E-HU, DF-HU, EAF-HU and HU-1 reduced the paw edema formation and leukocytes account into pleural cavity. In silico findings suggest that hibalactone present anti-inflammatory activity by interacting with the enzymes 5-lipoxygenase and cyclooxygenase-2. In the light dark box, the treatments with DF-HU, EAF-HU and HU-1 revealed an anxiolytic like effect. Thus, the E-HU and fractions of H. umbellata showed antinociceptive, anti-inflammatory and anxiolytic like activities, as also hibalactone, a possible phytoconstituent responsible for the biological effects of this specie.

BackgroundSideroxylon obtusifolium (Roem. & Schult.) T.D. Penn., Sapotaceae family, is a medicinal species native to the Brazilian Northeastern region. The plant is popularly used as an anti-inflammatory and hypoglycemic. PurposeTo evaluate the anti-inflammatory properties of the N-methyl-(2S,4R)-trans-4-hydroxy-l-proline (NMP) from S. obtusifolium leaves in models of inflammation and to clarify its action mechanisms. MethodsMale Swiss mice were distributed intocontrols and groups treated with NMP (25, 50 and 100mg/kg, p.o.), indomethacin or morphine (reference drugs). The animals were subjected to the formalin, carrageenan-induced edema and peritonitis tests. Furthermore, peritoneal lavage and slices from edematous paws were used for histological and immunohistochemical (iNOS, TNF-alpha, COX-2 and NF-kB) assays. ResultsDecreases in licking time, in the 1st and mainly in the 2nd phases of the formalin test, were shown after NMP treatments. In addition, decreases (around 50%) in paw edema were noticed at the 3rd h. The HE staining of paw slices demonstrated a complete reversion of the increased PMN cell numberafter NMP treatment. Similarly, decreases higher than 70% were also demonstrated in PMN cells, in the peritoneal fluid. Furthermore, NMP significantly decreased iNOS, TNF-alpha, COX-2 and NF-kB immunoreactivities. ConclusionsWe showed that S. obtusifolium presents a potent anti-inflammatory activity, due to the presence of the N-methyl-(2S,4R)-trans-4-hydroxy-l-proline(NMP) in the plant extract. This action is related to the inhibition by NMP of TNF-alpha and inflammatory enzymes.

ANTINOCICEPTIVE, ANTI-INFLAMMATORY EFFECTS AND SAFETY OF ZIZIPHUS MISTOL FRUITS

Ziziphus mistol Griseb. (Rhamnaceae), popularly known as “mistol,” is widely distributed throughout Perú, Bolivia, Paraguay and Argentina. Its fruit is consumed in different forms in several argentinean communities and used against biliary colic, dysentery, cold stomach and diseases of the respiratory system characterized by pain and inflammation. The present study was carried out to investigate the medicinal properties and safety of Ziziphus mistol (mistol) fruits ethanol and aqueous extracts and arrope. Antinociceptive activity was assessed using the formalin, acetic acid-induced writhing and tail-flick tests in rats. Anti-inflammatory effects were determined through carrageenan induced edema test and cotton pellet-induced granuloma formation, in rats. The safety was evaluated with test of acute toxicity (48 hours) and sub-chronic toxicity (91 days). All extracts (1,000 mg/kg b.w.) showed significant inhibition (P &lt;0.05) in the three model of pain experimentally induced in comparison to control. In a combination test using naloxone, diminished analgesic activity of aqueous extract and arrope were observed, indicating that their antinociceptive activity is connected with the opioid receptors. At dose 1000 mg/kg bw, the aqueous extract and arrope showed higher anti-inflammatory activity than the ethanol extract, in carrageenan and cotton pellet granuloma model used. In the acute toxicity study, a single dose of 4000 and 8000 mg/kg b.w., produced no mortality and no clinical signs of disease were observed after 48 hours. In the sub-chronic toxicity study the extracts no caused significant visible signs of toxicity, nor mortality for 91 consecutive days of treatment. Extracts and arrope of Z. mistol fruits could be good source of antinociceptive and anti-inflammatory agents because of its good activity and safety.

Bombax buonopozense is used in traditional medicine in Nigeria for the treatment of pains and feverish conditions. This study was orally carried out to establish the analgesic, anti-inflammatory and antipyretic properties of an aqueous stem bark extract of Bombax buonopozense in experimental animals. The analgesic activity was measured using the acetic acid-induced abdominal constriction and water tail imgmersion tests. The anti-inflammatory activity was assayed using the xylene and carrageenan-induced oedema tests, while the antipyretic activity was measured using the brewer&rsquo;s yeast and 2, 4 dinitrophenol-induced pyrexia. The stem bark extract (50, 100 and 200 mg/kg) at all doses used, was found to have significant (P&lt;0.05) analgesic, anti-inflammatory and anti-pyretic activities. Data shows that aqueous stem bark extract of B. buonopozense possesses constituents with therapeutic potential against pains and feverish conditions, thus supporting the use of the stem bark of this plant for similar ailments in traditional medicine. Evaluation of analgesic, anti-inflammatory, thrombolytic and hepatoprotective activities of roots of Premna esculenta (Roxb).

The objective of this study was to investigate the antinociceptive, anti-inflammatory, thrombolytic and hepatoprotective activities of root extracts of Premna esculenta (family: Verbenaceae). The analgesic activity was evaluated using the acetic-acid-induced writhing test in mice and radiant heat tail-flick method in rats. The anti-inflammatory activity was investigated by carrageenan-induced rat's paw edema, while the thrombolytic activity was evaluated by in vitro clot lysis model. The hepatoprotective activity was investigated against carbon-tetrachloride-induced liver damage in rats. In acetic-acid-induced writhing test, chloroform and ethyl acetate fraction of ethanolic extract at a dose of 200 mg/kg showed a significant (p<0.001) reduction in the number of writhes with 85.96% and 61.98% of inhibition, respectively. In radiant heat tail-flick method, the ethanolic extract produced 88.49% (p<0.001) elongation of tail-flicking time at 90 min after oral administration at same dose level. In the carrageenan-induced edema test, the ethanolic extract at a dose of 200 mg/kg showed a significant inhibition of paw edema with 22.68% and 17.24% inhibition after the first and third hours of the study period, respectively. In clot lysis model, the ethanolic extract at 5 mg/mL induced a significant clot lysis activity (37.69%, p<0.001). Oral administration of ethanolic extract at the dose of 400 mg/kg/day for 7 days significantly (p<0.001) reduced the elevated levels of serum glutamic pyruvic transaminase, serum glutamyl oxaloacetate transaminase and alkaline phosphatase compared to the CCl4-treated control group. The results of the study demonstrated the antinociceptive, anti-inflammatory, thrombolytic and hepatoprotective activities of roots of P. esculenta.

Evaluation of in vitro and in vivo anti-inflammatory and toxicity studies of methanolic extract of Rumex vesicarius Linn

Rumex vesicarius L. belongs to the family polygonaceae and is popularly known as “bladder dock”. It is a green vegetable traditionally used as an important plant to cure many diseases. In the present study the methanol extract of Rumex vesicarius L. was evaluated for its toxicity and potential in vitro and in vivo anti-inflammatory activity. The extract at different concentration from 100 to 2000 mg/kg bw was used to evaluate the toxicity on albino rat. For evaluating the in vivo anti-inflammatory activity, carrageenan induced edema test in rats were done at the dose 100 mg/kg bw and 200 mg/kg bw. The biochemical, hematological and histopathological studies were also estimated as supportive studies. Effect of methanol extract on protein denaturation and HRBC membrane were estimated to detect invitro anti-inflammatory activity. Acute toxicity and short term toxicity studies revealed that the methanol extract of Rumex vesicarius L. is nontoxic at the studied concentration. The appropriate dose range for preclinical study was found to be 100 mg/kg bw and 200 mg/kg bw. Oral administration of the extract at the dose of 100 and 200 mg/kg bw exhibited dose dependent and significant anti-inflammatory activity in carrageenan induced edema rats. The hematological and biochemical parameters were found normal. Invitro anti-inflammatory studies showed dose dependent activity with the IC50 value of 476.06 μg/ml and 491.355 μg/ml for inhibition of protein denaturation and stabilization of HRBC membrane respectively. Hence present investigation established some pharmacological evidences to support the folklore claim that Rumex vesicarius L. is used as anti-inflammatory agent.

Evaluation of Anti-inflammatory activity of Qurs-e-Mafasil Jadeed

With an increased incidence of drug toxicity and resistance to allopathic drugs, plant drug formulations could be an interesting alternative. Some plant drugs and phytochemicals are known to be anti-inflammatory properties, and can be of great significance in treatment of inflammatory disorders. These considerations require the scientific evaluation of the most important and commonly used traditional herbal formulations. A study has been done to find anti-inflammatory activity of Qurs-e-Mafasil Jadeed containing Colchicun luteum, Curcuma longa and gum of Acacia Arabica. In this proposed work 2 % aqueous suspension of qurs/tablet powder in gum acacia was used to determine its anti-inflammatory activity by carrageenin induced oedema test and cotton pellet induced granuloma test. Efficacy of this Unani formulation was compared with standard referent drug, Diclofenac sodium. The obtained results using carrageenin oedema test showed decrease in left hind paw volume significantly after 3 hours of carrageenin injection. In cotton pellet induced granuloma test, animals in all the test and standard drug tested groups, shows reduction in granuloma formation significantly. Thus, our results clearly indicate that this test formulation possesses significant anti-inflammatory activity in both acute and sub-acute phase.
