Development of microflow ultra high performance liquid chromatography-mass spectrometry metabolomic assays for analysis of mammalian biofluids

Introduction and objectivesThe application of untargeted metabolomics assays using ultra high performance liquid chromatography-mass spectrometry (UHPLC-MS) to study metabolism in biological systems including humans is rapidly increasing. In some of these studies there is a requirement to collect and analyse low sample volumes of biofluids (e.g. tear fluid) or low cell and tissue mass samples (e.g. tissue needle biopsies). The application of microflow, capillary or nano liquid chromatography (≤ 1.0 mm column internal diameter (i.d.)) theoretically should accomplish a higher assay sensitivity compared to analytical liquid chromatography (2.1–5.0 mm column internal diameter). To date, there has been limited research into microflow UHPLC-MS assays that can be applied to study samples of low volume or mass.MethodsThis paper presents three complementary UHPLC-MS assays (aqueous C18 reversed-phase, lipidomics C18 reversed-phase and Hydrophilic Interaction Liquid Chromatography (HILIC)) applying 1.0 mm internal diameter columns for untargeted metabolomics. Human plasma and urine samples were applied for the method development, with porcine plasma, urine and tear fluid used for method assessment. Data were collected and compared for columns of the same length, stationary phase and stationary phase particle size but with two different column internal diameters (2.1 mm and 1.0 mm).Results and conclusionsAll three assays showed an increase in peak areas and peak widths when applying the 1.0 mm i.d. assays. HILIC assays provide an advantage at lower sample dilutions whereas for reversed phase (RP) assays there was no benefit added. This can be seen in the validation study where a much higher number of compounds were detected in the HILIC assay. RP assays were still appropriate for small volume samples with hundreds of compounds being detected. In summary, the 1.0 mm i.d. column assays are applicable for small volume samples where dilution is required during sample preparation.

Comparison of experimental and simulated separation performance in capillary tube-in-manifold devices

A metal tube-in-manifold packed bed capillary column device, designed to overcome common limitations associated with capillary LC separations, is described. Experimental results of initial packing tests with sub-3 μm core-shell particles demonstrated efficiencies greater than 47,000 plates/m for a separation performed using the column device. Computational fluid dynamics (CFD) modeling of the multicomponent separation used for this work was validated against experimental LC results and the optimized model was able to effectively predict component peak retention times. However, the accuracy of predicted efficiencies requires further refinement. The tube-in-manifold design demonstrates that packed capillary columns with cylindrical cross-sectional channel geometry and ultrahigh pressure, low dead volume fluidic connections are achievable.

B-169 Combining the EXENT System and LC-MS for the Detection of Monoclonal Immunoglobulins at Low Levels

Abstract Background The EXENT® system is an automated platform designed to quantify monoclonal immunoglobulins in serum. EXENT® combines immunoprecipitation and MALDI-TOF mass spectrometry for the detection of monoclonal immunoglobulins at levels lower than traditional gel based methods. This study was designed to demonstrate if EXENT® prepared samples that were negative could be reflexed to a more sensitive method based on capillary flow Liquid Chromatography- Mass Spectrometry (LC-MS) without any modifications to the EXENT® prepared sample. Methods Patient samples containing a monoclonal immunoglobulin (ranging from 20 to 40 g/L) were diluted into pooled polyclonal serum and the samples were prepared and analyzed by EXENT®. Samples that were negative by EXENT® were reflexed by directly loading EXENT® eluates onto the LC-MS instrument. The abundance of the monoclonal immunoglobulin was defined by the signal-to-noise (S/N) of the light chain peak observed after deconvolution. Results Baseline samples were diluted down to two levels, 0.100 g/L and 0.001 g/L. LC-MS detected the same light chain from the monoclonal immunoglobulin as the EXENT® system in all reflexed samples that were negative by EXENT®. Conclusions LC-MS can detect monoclonal immunoglobulins that can no longer be detected using the EXENT® system by tracking the unique molecular mass of the monoclonal light chain. Reflexing samples to LC-MS did not require additional sample handling resulting in a faster time-to-result than current NGS, NGF, and clonotypic peptide approaches. Furthermore, monitoring the intact monoclonal light chain circumvents enzymatic cleavage to create a unique clonotypic peptide, alleviating a situation where no unique peptide can be generated as has been shown previously by Bergen et al. As a consequence, this reflex-methodology results in a consistent way of measuring the presence of malignant B-cells with high sensitivity.

Microdialysis-integrated HPLC system with dual-electrode detection using track-etched membrane electrodes for in vivo monitoring of dopamine dynamics

A capillary high-performance liquid chromatography (HPLC) system equipped with a dual-electrode detector utilizing track-etched membrane electrodes (TEMEs) was combined with a microdialysis sampling setup. The electrochemical detector benefits from the high electrolysis efficiency of TEMEs, allowing for calibration-free coulometric detection and simplifying data analysis to determine the dopamine recovery through a dialysis probe. Additionally, this system was used for in vivo monitoring of dopamine in the right striatum of a mouse brain. Temporal changes in dopamine levels, including an exponential decay immediately after the dialysis probe insertion and an excess release of dopamine induced by a high concentration of potassium ions, confirmed the system’s proper operation. Furthermore, subsequent measurements following the intraperitoneal injection of mirtazapine showed no increase in dopamine levels in the right dorsal striatum. The dual-electrode system displayed characteristic dopamine detection behavior, with anodic and cathodic peak pairs indicative of reversible electrochemical reactions. This capability facilitated the identification of the dopamine peak within the complex chromatogram of the mouse brain dialysate. The consistency between dopamine collection efficiency from standard solutions and dialysate indicated the absence of interfering electroactive substances overlapping with the dopamine peak in the chromatogram. This integrated analysis system successfully tracked temporal fluctuations in dopamine concentration within the mouse brain.

A stationary phase of tetraethylene glycol-modified silica for separation of phenolic acids

AbstractSilica, as a stationary phase, has low separation efficiency accompanied by overlapping, broadened, and tailed peaks, so it needs to be modified to improve its efficiency. This study aims to develop a silica-based stationary phase modified by tetraethylene glycol (TEG) to separate phenolic compounds. Silica was modified by a chemical bond between silanol groups on the silica surface and TEG through a 3-glycidyloxypropylmethoxysilane reaction. The modified silica was packed into a capillary column and used to separate simple phenolic compounds consisting of phenol, pyrocatechol, and pyrogallol. A sample of 0.2 µL was injected into the capillary liquid chromatography and the mobile phase employed was acetonitrile 98% with a flow rate of 3 μL min−1. Elution was also done isocratically in this process and detection was carried out at a wavelength of 254 nm. The mixture of simple phenolic compounds was successfully separated in less than 7 min. The asymmetry factor and resolution were 1.43–2.12 and 1.72–5.43 respectively. The number of the theoretical plates ranged from 213 to 7,857. Columns containing Si-TEG stationary phase also separate phenolic compounds, which consist of gallic acid, syringic acid, ferulic acid, and caffeic acid. A sample of 0.2 µL was injected into the capillary liquid chromatography and successfully separated the mixture in less than 12 min. The samples were eluted isocratically using a mixture of methanol and 50 mM phosphate buffer pH 2.5 (8:92) with a flow rate of 3 μL min−1. The phenolic acids compounds were detected at a wavelength of 280 nm. The chromatogram showed four separate peaks. The asymmetry factor and resolution were 1.53–1.63 and 1.14–1.74, respectively, but the number of the theoretical plates was low, ranging from 190 to 796.

Hybrid organic monolithic column containing MIL-68(Al) for the separation of small molecules by capillary HPLC

A hybrid organic monolithic column made of poly(lauryl methacrylate-co-1,6-hexanediol dimethacrylate) and the metal-organic framework MIL-68(Al) was prepared for the first time. The column was used in capillary liquid chromatography, both in isocratic and gradient elution modes. Separation performance towards small molecules of different chemical nature (polycyclic aromatic hydrocarbons, alkylbenzenes, phenols, etc.) was studied. Monte Carlo simulations were made to both select the proper precursors to obtain empty metal-organic framework micropores in the monolithic polymer and also, to analyze the potential free access of the studied analytes into the micropores (necessary to improve mass transfer and column efficiency). The hereby synthesized metal-organic framework microcrystals allowed obtaining homogeneous hybrid monolithic columns. Adding of MIL-68(Al) (1030 m2 g-1 BET specific surface area) increased the surface area from 3.9 m2 g-1 for the parent monolith to 18.2 m2 g-1 for the hybrid column containing 8 mg mL-1 of the microcrystals. Chromatographic performance of this new column was evaluated by studying retention factors, resolution, and plate counts at room temperature. Different compounds, not completely resolved in the parent monolith, were partially or completely separated after metal-organic framework addition. Using the monolithic column with only 2 mg mL-1 of MIL-68(Al), five alkylbenzenes were completely separated with very symmetrical peak shapes, resolution factors up to 3.60 and plate counts of 4300 plates m-1 for n-hexylbenzene. This value is higher than those obtained by other authors who used organic monolithic columns with embedded metal-organic frameworks to perform separations at room temperature. Additionally, nine polycyclic aromatic hydrocarbons were partially or completely resolved in gradient elution mode. The hybrid monolithic columns exhibited very good intra-day (%RSD=1.9), inter-day (%RSD=2.6), and column-to-column (%RSD=4.3) reproducibility values. Easy and fast column preparation, and versatility to efficiently separate several compounds of different chemical nature in isocratic and gradient mode, makes this new hybrid column a very good option for the analysis of small molecules in capillary (or nano) HPLC.

Development of a capillary electrophoresis method with laser-induced fluorescence detection for the characterization of oligosaccharides in human milk

Breast milk is the most appropriate food for an infant in the first months of life - it provides the essential nutrients required for proper growth, with breast milk oligosaccharides (HMOs) being the third most abundant component. HMO concentrations vary based on factors like lactation stage and maternal health. They play crucial roles in infant health, acting as prebiotics, antimicrobials, and immune modulators. Analytical methods like capillary electrophoresis and liquid chromatography are the most often used in HMOs qualitative and quantitative analysis. Continuing research aims to enhance analytical techniques for comprehensive HMOs analysis, this work presents the development of a capillary electrophoresis method with laser-induced fluorescence detection along with derivatization and solid-phase extraction steps for the characterisation of major oligosaccharides in colostrum samples. Validation parameters, such as linearity (0.9949–0.9989), limits of detection (5.49–16.40 ng cm−3) and quantification (18.30–54.67 ng cm−3), intra- and inter-day precision (2.41–9.78% and 1.27—14.5%, respectively), trueness (− 8.4 to − 2.0%), and recovery (70.02–113.5%) and repeatability (2.22–10.58%) for solid-phase extraction stage were assessed. The method was evaluated using RGB additive colour model regarding the analytical and practical effectiveness and greenness of the method (achieving a brilliance score of 67.1%). Moreover, the developed method was successfully applied to determine three oligosaccharides (DSLNT, 3’SL, and 6’SL) in colostrum samples. Considering its effectiveness, the method has promise for this type of application.Graphical abstract

Design and fabrication of fluorous monoliths with tunable surface property for capillary liquid chromatography

Hierarchically porous monoliths with satisfactory properties have been employed in diverse fields, especially separation. In this study, pentafluorophenyl acrylate (PFPA), pentaerythritol tetraacrylate (PETA) and trimethylolpropane tris(3-mercaptopropionate) (TTMP) were selected as precursors to fabricate a novel monolithic column by thermally initiated polymerization in the presence of a binary porogenic system containing tetrahydrofuran and 1-propanol. The fabricated poly(PFPA-co-PETA-co-TTMP) monolithic column revealed excellent permeability and mechanical stability. Additionally, baseline separation of the mixture of small molecules can be achieved, involving alkylbenzene and fluorobenzene in chromatographic assessment, and the theoretical plate number is up to 60,500 plates/m for butylbenzene with a linear velocity of 0.14 mm/s. Tryptic digest of HeLa as an analyte was used to investigate the possibility of the poly(PFPA-co-PETA-co-TTMP) monolith in biological separation by cLC-MS/MS. Moreover, benefiting from the existence of pentafluorophenyl groups, the cucurbit[8]uril (CB[8]) could be modified on the prepared poly(PFPA-co-PETA-co-TTMP) monolith through host-guest interaction to obtain poly(PFPA-co-PETA-co-TTMP)-CB[8] monolith. It could be observed that significant changes in retention behavior of analytes appeared after immobilizing CB[8] on the monolith. It offered an innovative approach by utilizing host-guest interaction to fabricate monolithic columns with different chromatographic behaviors.

Construction of adamantane-based monolithic column with three-dimensionally porous structure for small molecules separation and biosample analysis

BackgroundThe fabrication technique of capillary column is the key to the development and application of capillary liquid chromatography (cLC) to improve separation efficiency for analytes. The capillary monolithic column possessed three-dimensionally connected porous or channel structures. Unique porous structure endows excellent permeability and high performance in diverse fields, especially in separation. Thereinto, organic monolithic columns have attracted widespread attention due to their advantages of simple preparation and excellent biocompatibility. However, their separation selectivity needs to be further developed and regulated to apply the separation of more diverse samples. ResultsA novel polymeric monolithic column was prepared via thermally initiated in situ copolymerization of 2-methyladamantan-2-yl acrylate (MADA) with ditrimethylolpropane tetraacrylate (DTTA) in fused silica. The prepared poly(MADA-co-DTTA) monolith showed adjustable permeability, developed porous structure and high thermal stability. Consequently, it exhibited excellent separation capability of small molecules (alkylbenzenes and polycyclic aromatic hydrocarbons). Especially, when acetonitrile/water (60/40, v/v) was used as the mobile phase, the theoretical plate numbers reached 84,000 plates m−1 for butylbenzene at a linear velocity of 0.5 mm s−1. Most importantly, the hydrophobicity of the poly(MADA-co-DTTA) monolithic column was regulated via host-guest interaction between adamantyl group and cucurbit [7]uril (CB[7]). Additionally, the poly(MADA-co-DTTA) monolith was further adopted for the analysis of the tryptic digest of proteins from HeLa by cLC-MS/MS. The 33,783 unique peptides and 5,299 proteins were identified on the monolith, which exhibited great separation ability for complex samples. Significance and noveltyDue to abundant pore structure and good chemical properties, the poly(MADA-co-DTTA) monolithic column exhibited high performance for the separations of small molecules and biological sample. Meanwhile, owing to the existence of adamantyl-group, CB[7] was immobilized on the poly(MADA-co-DTTA) monolithic column to fabricate poly(MADA-co-DTTA)-CB[7] by host-guest interaction. It is possible to adjust the surface chemistry of the monolithic materials to accommodate more complex analytes.

First Detection of Algal Caribbean Ciguatoxin in Amberjack Causing Ciguatera Poisoning in the Canary Islands (Spain).

Ciguatera Poisoning (CP) is an illness associated with the consumption of fish contaminated with potent natural toxins found in the marine environment, commonly known as ciguatoxins (CTXs). The risk characterization of CP has become a worldwide concern due to the widespread expansion of these natural toxins. The identification of CTXs is hindered by the lack of commercially available reference materials. This limitation impedes progress in developing analytical tools and conducting toxicological studies essential for establishing regulatory levels for control. This study focuses on characterizing the CTX profile of an amberjack responsible for a recent CP case in the Canary Islands (Spain), located on the east Atlantic coast. The exceptional sensitivity offered by Capillary Liquid Chromatography coupled with High-Resolution Mass Spectrometry (cLC-HRMS) enabled the detection, for the first time in fish contaminated in the Canary Islands, of traces of an algal ciguatoxin recently identified in G. silvae and G. caribeaus from the Caribbean Sea. This algal toxin was structurally characterized by cLC-HRMS being initially identified as C-CTX5. The total toxin concentration of CTXs was eight times higher than the guidance level proposed by the Food and Drug Administration (0.1 ng C-CTX1/g fish tissue), with C-CTX1 and 17-hydroxy-C-CTX1 as major CTXs.

Carla Beatriz Grespan Bottoli holds a degree in Industrial Chemistry from the Federal University of Santa Maria (1996), as well as a Ph.D. in Chemistry (2002), and a postdoctoral degree in Chemistry (2002-2004), both from the State University of Campinas (Unicamp). In 2005, she undertook an internship at the Institute of Nuclear Sciences of the National Autonomous University of Mexico (UNAM) to study radiolysis in vitamins using gamma radiation from a source of Cobalt 60. In 2017, she presented her habilitation thesis (associate professor's thesis) at Unicamp. Currently, she is Associate Professor and Postgraduate Advisor at the Institute of Chemistry at Unicamp (IQ-Unicamp), where she works in the area of analytical chemistry, with emphasis on separation methods, mainly liquid chromatography. Prof. Bottoli coordinates the NovaCrom – Liquid Chromatography Laboratory (https://novacromlab.wixsite.com/novacrom), which is a research group that has been working on the development of new monolithic stationary phases for capillary liquid chromatography. Research is also focused on the development of analytical methods that employ liquid chromatography coupled with mass spectrometry to study the translocation of pesticides in plants such as coconut, palm, and soybean. Between 2019 and 2021, Prof. Bottoli was an associate coordinator of the Research Commission of the IQ-Unicamp and is currently a member of the Research Integrity Commission at Unicamp. She is a member of the Brazilian Chemical Society and a researcher at the National Institute of Science and Technology in Bioanalytics (INCTBio). Carla Bottoli is the mother of Murilo, 13 and Mateus, 10.

Histidine tag modified magnetic beads for analysis of arsenic binding proteins

BackgroundMany proteins with thiol groups can bind with trivalent arsenic which are termed as arsenic binding proteins, thus change their physiological functions. Therefore, it is vital to analyze the arsenic binding proteins in cells. The Pull-Down strategy based on biotinylated phenylarsenic acid (Bio-PAO(III)) probes is an effective way for analysis of arsenic binding proteins. In this strategy, streptavidin magnetic beads (SA-MBs) was applied to capture the arsenic binding proteins conjugating with Bio-PAO(III) probe. However, strong interaction between SA and biotin makes the elution of arsenic binding proteins not easy. ResultsWe developed a novel affinity separation strategy to address the challenge of eluting arsenic binding proteins, a key issue with the existing Bio-PAO(III) Pull-Down method. By employing magnetic beads modified with Nα-Bis(carboxymethyl)-l-lysine (NTA-Lys), polyhistidine-tag (His6-Tag), and SA (MB-NTA(Ni)-His6-SA), we established a more efficient purification process. This innovative approach enables selective capture of arsenic binding proteins in HepG2 cells labeled by Bio-PAO(III) probes, facilitating gentle digestion by trypsin for precise identification through capillary high performance liquid chromatography (Cap HPLC)-electrospray ionization (ESI)-tandem mass spectrometry (MS/MS). What is more, the magnetic beads can be regenerated by using imidazole as the eluent, and the obtained MB-NTA(Ni) can be reloaded with His6-SA for next use. Our method successfully identified 41 arsenic binding proteins, including those involved in cytoskeletal structure, heat shock response, transcriptional regulation, DNA damage repair, redox state regulation, mitochondrial dehydrogenase function, and protein synthesis and structure. SignificanceThis work contributes to a more comprehensive understanding of the toxic mechanisms of arsenic, potentially providing valuable insights for the prevention or treatment of arsenic-related diseases.

High-Throughput Capillary Liquid Chromatography Using a Droplet Injection and Application to Reaction Screening.

The cycle time of a standard liquid chromatography (LC) system is the sum of the time for the chromatographic run and the autosampler injection sequence. Although LC separation times in the 1-10 s range have been demonstrated, injection sequences are commonly >15 s, limiting throughput possible with LC separations. Further, such separations are performed on relatively large bore columns requiring flow rates of ≥5 mL/min, thus generating large volumes of mobile phase waste when used for large scale screening and increasing the difficulty in interfacing to mass spectrometry. Here, a droplet injector system was established that replaces the autosampler with a four-port, two-position valve equipped with a 20 nL internal loop interfaced to a syringe pump and a three-axis positioner to withdraw sample droplets from a well plate. In the system, sample and immiscible fluid are pulled alternately from a well plate into a capillary and then through the injection valve. The valve is actuated when sample fills the loop to allow sequential injection of samples at high throughput. Capillary LC columns with 300 μm inner diameter were used to reduce the consumption of mobile phase and sample. The system achieved 96 separations of 20 nL droplet samples containing 3 components in as little as 8.1 min with 5-s cycle time. This system was coupled to a mass spectrometer through an electrospray ionization source for high-throughput chemical reaction screening.

Small-Scale Serial Size Exclusion Chromatography (s3SEC) for High Sensitivity Top-Down Proteomics of Large Proteoforms.

Top-down-mass spectrometry (MS)-based proteomics has emerged as a premier technology to examine proteins at the proteoform level, enabling characterization of genetic mutations, alternative splicing, and post-translational modifications. However, significant challenges that remain in top-down proteomics include the analysis of large proteoforms and the sensitivity required to examine proteoforms from minimal amounts of sample. To address these challenges, we have developed a new method termed "small-scale serial Size Exclusion Chromatography" (s3SEC), which incorporates a small-scale protein extraction (1 mg of tissue) and serial SEC without postfractionation sample handling, coupled with online high sensitivity capillary reversed-phase liquid chromatography tandem MS (RPLC-MS/MS) for analysis of large proteoforms. The s3SEC-RPLC-MS/MS method significantly enhanced the sensitivity and reduced the proteome complexity across the fractions, enabling the detection of high MW proteoforms previously undetected in one-dimensional (1D)-RPLC analysis. Importantly, we observed a drastic improvement in the signal intensity of high MW proteoforms in early fractions when using the s3SEC-RPLC method. Moreover, we demonstrate that this s3SEC-RPLC-MS/MS method also allows the analysis of lower MW proteoforms in subsequent fractions without significant alteration in proteoform abundance and equivalent or improved fragmentation efficiency to that of the 1D-RPLC approach. Although this study focuses on the use of cardiac tissue, the s3SEC-RPLC-MS/MS method could be broadly applicable to other systems with limited sample inputs.

Organic-silica hybrid monolithic sorbents for sample preparation techniques: A review on advances in synthesis, characterization, and applications

Organic-silica hybrid monolithic materials have attracted considerable attention as potential stationary phases in separation science. These materials combine the advantages of organic polymer and silica-based monoliths, including easy preparation, lower back pressure, high permeability, excellent mechanical strength, thermal stability, and tunable surface chemistry with high surface area and selectivity. The outstanding chromatographic efficiency as stationary phase of hybrid monolithic capillary columns for capillary liquid chromatography and capillary electrochromatography has been reported in many papers. Organic-silica hybrid monolithic materials have also been extensively used in the field of sample preparation. Owing to their surface functionalities, these porous sorbents offer unique selectivity for pre-concentration of different analytes in the most complex matrixes by fast dynamic transport. These sorbents not only improve the analytical method sensitivity, but also introduce novelties in terms of extraction devices and instrument coupling strategies. The current review covers the period spanning from 2017 to 2023 and describes the properties of organic-inorganic hybrid monolithic materials, the present status of this technology and summarizes recent developments in their use as innovative sorbents for microextraction sample preparation techniques (solid phase microextraction with pipette tip, offline in-tube SPME, in-tube SPME online with LC, and in-tube SPME directly coupled with mass spectrometry). Aspects such as the synthesis methods (sol–gel process, one-pot approach, and polyhedral oligomeric silsesquioxanes-based procedure), characterization techniques, and strategies to improve extraction efficiency in various applications in different areas (environmental, food, bioanalysis, and proteomics) are also discussed.

Development, validation and application of a capillary microsampling LC–MS/MS method for the quantification of BIIB131 (SMTP-7) in rat plasma

Capillary microsampling (CMS) is a technique that can significantly reduce the blood collection volume compared to conventional sampling methods, and thus is much preferred for studies in rats and mice. BIIB131 (SMTP-7) is a novel thrombolytic drug candidate currently under Phase 2 clinical development for the treatment of acute ischemic stroke. To support the safety studies in rats, an accurate and reliable CMS LC–MS/MS assay for the quantification of BIIB131 in rat plasma was developed and validated. This method utilized stable-isotope labeled [13C515N2]-BIIB131 as the internal standard. The samples were extracted using acid-assisted liquid–liquid extraction with methyl tert-butyl ether (MTBE) and formic acid. The chromatographic separation was achieved on an ACE Excel 3 Super C18 analytical column (2.1 mm × 50 mm, 3.0 µm) using a gradient elution. The mass spectrometric detection of BIIB131 and its internal standard was achieved using positive ion electrospray multiple reaction monitoring (MRM). The standard curve ranged from 0.50 to 300 ng/mL for BIIB131 and was fitted to a 1/x2 weighted linear regression model. For regular QCs, the intra-assay precision was 1.7–6.1 % CV, the inter-assay precision was 2.7–11.0 % CV, and the intra-assay and inter-assay accuracy (%Bias) were –20.0–10.6 % and –7.8–6.3 %, respectively. For CMS QCs, the intra-assay and inter-assay precision were 2.2–13.6 % and 6.7–12.9 % CV, and the intra-assay and inter-assay accuracy (%Bias) were –13.2–15.0 % and –7.8–4.2 %, respectively. The validated CMS LC–MS/MS method has been successfully applied to a safety study in rats.

The analysis of semi-products at different stages of production and finished medicinal products is an integral part of an effective quality assurance system for pharmaceutical production. The traditional methods used in the pharmaceutical field, such as capillary electrophoresis, gas and liquid chromatography, are quite versatile and effective. However, they are distinguished by the high cost of instrumentation and component materials, the need to use high-purity solvents, the lack of mobility and the possibility of in situ studies. Such shortcomings can be overcome by using the digital colorimetry method. Its significant popularization has been facilitated by the development of portable and personal electronic devices, among which the smartphone can be distinguished. The aim of the work was to develop a fast and simple method for the determination of a number of polypeptide antibiotics in drugs by digital colorimetry using a smartphone as a color recording device. To implement the solid phase fluorimetric determination of polypeptide antibiotics, HPTLC plates based on silica gel with an aluminum substrate were used as a matrix. Under these conditions, the fluorescence of actinomycin D, virginiamycin M1, virginiamycin S1, and novobiocin was observed when exposed to monochromatic radiation in the UV part of the spectrum (365 nm). The measurement of fluorescence intensity on the surface of the matrices was carried out using a smartphone. Calibration characteristics were constructed for the determination of polypeptide antibiotics in the concentration range of 32–500 μg/mL. The method for assessing the quality of finished drugs was tested on commercial products “Piostacin”, “Cosmegen”, “Dactinomycin” purchased in retail chains. The relative standard deviation of the analysis results does not exceed 0.05. The duration of the analysis was 10–15 min. Based on the results of the studies, the paper presents a method for determining some antibiotics of the polypeptide series by the intrinsic fluorescence of deposited solutions on a solid substrate during treatment with monochromatic UV radiation. The experimental delivery conditions used allow the analysis of drugs based on actinomycin D, virginiamycin M1, virginiamycin S1, and novobiocin.

Portable capillary LC for in-line UV monitoring and MS detection: Comparable sensitivity and much lower solvent consumption.

Pharmaceutical development currently relies on quality separation methods from early discovery through to line-of-site manufacturing. There have been significant advancements made regarding the column particle packing, internal diameter, length connectivity, the understanding of the impact key parameters like void volume, flow rate, and temperature all that affects the resultant separation quality, that is, resolution, peak shape, peak width, run time, and signal-to-noise ratio. There is however a strong need to establish better alternatives to large bulky high-performance liquid chromatography racks either for process analytical reaction monitoring or mass spectrometry analysis in establishing product quality. Compact, portable high-pressure liquid chromatography can be a more efficient alternative to traditional ultra-high pressure liquid chromatography and traditional liquid chromatography. The compact versatile instrument evaluated here allows good separation control with either the on-board column with fixed ultra-violet wavelength cartridge or for use with a high-resolution mass spectrometry. Significant space reduction results in greener lab spaces with improved energy efficiency for smaller labs with lower energy demands. In addition, this compact liquid chromatography was used as a portable reaction monitoring solution to compare forced degradation kinetics and assess portable liquid chromatography-mass spectrometry capability for the analyses required for pharmaceutical drug product testing.

High-performance peptide and disulfide mapping by direct injection of intact proteins using on-line coupled UV-liquid chromatography microdroplet mass spectrometry (UVLC-MMS)

Microdroplet mass spectrometry (MMS), achieving ultra-fast enzyme digestion in the ionization source, holds great promises for innovating protein analysis. Here, in-depth protein characterization is demonstrated by direct injection of intact protein mixtures via on-line coupling MMS with capillary C4 liquid chromatography (LC) containing UV windows (UVLC-MMS) through an enzyme introduction tee. We showed complete sets of peptides of individual proteins (hemoglobin, bovine serum albumin, and ribonuclease A) in a mixture could be obtained in one injection. Such full (100%) sequence coverage, however, could not be achieved by conventional nanoLC-MS method using bottom-up approach with single enzyme. Moreover, direct injection of a chaperone α-crystalline (α-Cry) complex yielded identification of post-translational modifications including novel sites and semi-quantitative characterization including 3:1 stoichiometry ratio of αA- and αB-Cry sub-units and ∼1.4 phosphorylation/subunit on S45 (novel site) and S122 (main site) of αA-Cry, ∼0.7 phosphorylation/subunit on S19 (main site) and S45 of αB-Cry, as well as 100% acetylation on both N-termini of each subunits by matching the mass and retention time of the intact and its digested peptides. Furthermore, trifluoroacetic acid was able to be used in the mobile phase with UVLC-MMS to improve the separation of differentially reduced intact species and detectability of the droplet-digested products. This allowed us to completely map four disulfide linkages of ribonuclease A based on collision-induced dissociation of disulfide clusters, some of which would otherwise not be detected, preventing scrambling or shuffling errors arising from lengthy bulk solution digestion by the bottom-up approach. Integration of UVLC and MMS greatly improves droplet digestion efficiency and MS detection, enabling highly efficient workflow for in-depth and accurate protein characterization.
