Survey of Southeast U.S. beef veterinarians regarding methods for bull breeding soundness evaluation

The purpose of this survey was to describe veterinarians’ methods for performing bull breeding soundness evaluations (BSEs). Veterinarians in Mississippi, Louisiana and Arkansas were emailed a survey regarding their experience, demo­graphics, opinions and perceptions of client pref­erences regarding BSEs. Logistic regression was used to test respondent characteristics for associations with dichotomous outcomes including evaluating semen morphology as part of every BSE and detecting the diadem defect. Of 3,757 veterinar­ians solicited, 204 (5%) responded and 83 (2%) qualified for analysis. Of these, 10/73 (14%) indicated they do not evaluate morphology during every BSE. When shown an image of the diadem defect, 18/73 (25%) indicated seeing it often or some­times, and 55/73 (75%) indicated rarely or never. The only fac­tor associated with evaluating morphology as part of every BSE, was indicating belief that morphology was most predic­tive of bull fertility vs. believing motility was most predictive, or being unsure (OR = 11.2, 95% C.I. = 1.3-94.1). Respondents who always evaluate morphology with bright field microscopy at 1,000X (OR = 4.1, 95% C.I. = 1.2-13.3) or with phase contrast microscopy at ≥ 400X (OR = 5.4, 95% C.I. = 1.1-27.7) had higher odds of indicat­ing they detect the diadem defect sometimes/often. There was an interaction between always using 1000X magnification, or not, and always using 400X, or not, on the reported detection of the diadem defect. Veterinarians’ perceptions of which as­pects of the BSE are most predictive of fertility influence their methods for performing BSEs, and influence the frequency of detecting diadem defects.

Survey of Mississippi cow-calf producers regarding factors associated with veterinarians performing bull breeding soundness evaluations and/or pregnancy diagnosis

Reproductive efficiency is a major driver of profitability in cow-calf herds, yet many beef producers do not hire veterinar­ians to perform services intended to improve reproductive ef­ficiency such as bull breeding soundness evaluation (BSE) or pregnancy diagnosis. The objective of this study was to deter­mine reasons Mississippi cow-calf producers do or do not hire veterinarians to perform these services. Anonymous surveys were mailed to 1,500 randomly selected members of the Missis­sippi Cattlemen’s Association. Multivariable logistic regression was used to test respondent characteristics for association with outcome variables (e.g., respondents hire a veterinarian for BSEs and/or pregnancy diagnosis). Of 414 returned surveys, 397 (96%) qualified for analysis for an overall response rate of 26%. Of respondents that answered the respective ques­tions, 155/386 (40%) hire a veterinarian for BSEs, and 103/392 (26%) hire a veterinarian for pregnancy diagnosis. Of 396 re­spondents, 229 (58%) indicated they had some form of a defined breeding season. The most common reasons for not hiring a veterinarian were lack of time/help (82/202, 41%) for BSEs and cost (91/248, 37%) for pregnancy diagnosis. On in­ferential analysis, the factors positively associated with hiring a veterinarian for BSEs were having a defined breeding season (OR = 6.8, 95% C.I. = 4.1-11.1) and raising seedstock cattle (OR = 1.7, 95%C.I. = 1.0-13.3). The only factor positively associ­ated with hiring a veterinarian for pregnancy diagnosis was having a defined breeding season (OR = 4.2, 95%C.I. = 2.5-7.3). Management style and operation type influenced producers hiring a veterinarian for BSEs and pregnancy diagnosis.

Genetic parameters for yearling male reproduction traits in tropical composite cattle population.

Fertility is economically important but is hard to quantify and measure in breeding programs which has led extensive breeding programs to ignore fertility in their selection criteria. While female fertility traits have been extensively researched, male fertility traits have been largely ignored. It is estimated that 20% to 40% of bulls have sub-fertility, reducing the number of calves born and profits, highlighting the importance of investigating bull fertility. The most practical measure of male fertility is a bull breeding soundness evaluation (BBSE) which assesses structure as well as semen quality and quantity. Generally, traits recorded in a BBSE are neither genetically evaluated nor used for selection in breeding programs. All traits recorded during a BBSE were analyzed through a series of univariate and bivariate linear mixed models using a genomic relationship matrix to estimate genetic parameters. All genotype and phenotype data were obtained from a tropical composite commercial cattle population and imputed to 27,638 single-nucleotide polymorphisms (SNPs) with a total of 2,613 genotyped animals with BBSE records ranging from 616 to 826 animals depending on the trait. The heritabilities of the 27 traits recorded during a BBSE ranged from 0.02 to 0.49. Seven of the male fertility traits were recommended to be included in a breeding program based on their heritability and their phenotypic and genetic correlations. These traits are scrotal circumference, percent normal sperm, proximal droplets, distal midpiece reflex, knobbed acrosomes, vacuoles/teratoids, and sheath score. Using these seven traits in a breeding program would result in higher calving rates, increasing production and profitability.

Bull Breeding Soundness Assessment Using Artificial Neural Network-Based Predictive Models

For years, efforts have been devoted to establishing an effective bull breeding soundness evaluation procedure; usual research on this subject is based on bull breeding soundness examination (BBSE) methodologies, which have significant limitations in terms of their evaluation procedure, such as their high cost, time consumption, and administrative difficulty, as well as a lack of diagnostic laboratories equipped to handle the more difficult cases. This research focused on the creation of a prediction model to supplement and/or improve the BBSE approach through the study of two algorithms, namely, clustering and artificial neural networks (ANNs), to find the optimum machine learning (ML) approach for our application, with an emphasis on data categorization accuracy. This tool was designed to assist veterinary medicine and farmers in identifying key factors and increasing certainty in their decision-making during the selection of bulls for breeding purposes, providing data from a limited number of factors generated from a deep pairing study of bulls. Zebu, European, and crossbred bulls were the general groupings. The data utilized in the model’s creation (N = 359) considered five variables that influence improvement decisions. This approach enhanced decision-making by 12% compared to traditional breeding bull management. ANN obtained an accuracy of 90%, with precision rates of 97% for satisfactory, 92% for unsatisfactory, and 85% for bad. These results indicate that the proposed method can be considered an effective alternative for innovative decision-making in traditional BBSE.

A proteomic approach to identifying spermatozoa proteins in Indonesian native Madura bulls.

Proteins assist sperm mature, transit the female reproductive tract, and recognise sperm oocytes. Indigenous Indonesian bulls, Madura bulls, have not been studied for reproductive proteomics. As local Indonesian beef livestock, Madura cattle assist in achieving food security; hence, their number must be improved. Thus, the identification of molecular proteomics-based bull fertility biomarkers is needed. This study aimed to characterise the sperm fertility function of the superior Madura bull (Bos indicus × Bos Javanicus) spermatozoa proteome. Frozen semen from eight Madura superior bulls (Bos indicus × Bos javanicus) aged 4-8 years was obtained from the artificial insemination centre (AIC) in Singosari and Lembang. Madura superior bulls are those that have passed the bull breeding soundness evaluation. Frozen sperm were thawed and centrifuged at 3000 × g for 30 min. Proteins in sperm were characterised through proteomic analysis using liquid chromatography-tandem mass spectrometry (LC-MS/MS). The resulting gene symbols for each protein were then subjected to bioinformatics tools, including UniProt, DAVID, and STRING databases. Regarding sperm fertility, the analysis revealed that 15 proteins were identified in the sperm of Madura bulls. Amongst the identified proteins, the superior Madura bull sperm contained several motilities, energy-related proteins, and chaperone proteins. A substantial portion of characterised proteins are linked to metabolic pathways and the tricarboxylic acid (TCA) cycle, contributing to sperm energy production. In conclusion, the first in-depth proteome identification of sperm related to sperm quality and bull fertility of a unique indigenous Madura breed of Indonesia was performed using the LC-MS/MS proteomic method. These findings may serve as a reference point for further studies related to the functions of bovine sperm and biomarkers of fertility and sperm quality.

Incorporation of Testicular Ultrasonography and Hair Steroid Concentrations in Bull Breeding Soundness Evaluation.

Testicular ultrasonography and steroid concentrations (cortisol, dehydroepiandrosterone sulfate (DHEA-S), cortisol/DHEA-S ratio, testosterone) in hair were examined for their utility in the bull breeding soundness evaluation (BBSE). Beef and dairy bulls (n = 16; 2.7 ± 0.4 years old; body condition score 3.2 ± 0.1) of five breeds were maintained under the same conditions at an accredited semen collection center. Bulls underwent routine semen collection twice weekly for 12 weeks and semen was processed and cryopreserved. Ultrasonography and hair sampling were undertaken at the last semen collection. Bulls with homogeneous testicular parenchyma (n = 8) had a higher (p < 0.05) percentage of motile sperm post-thawing compared with bulls with heterogeneous parenchyma (n = 8). There were no differences (p > 0.05) in the hair concentrations of cortisol, DHEA-S, and testosterone between bulls with homogeneous and heterogeneous parenchyma. In bulls with homogeneous parenchyma, hair DHEA-S concentration was positively correlated with percentage motile sperm (R2 = 0.76), progressively motile sperm (R2 = 0.70), and motility yield (R2 = 0.71). The findings indicate that the integration of testicular ultrasonography and hair DHEA-S status in the BBSE could provide a more comprehensive assessment of indicative fertility in bulls. Additionally, ultrasonography can be used in the BBSE when the evaluation of semen parameters is not available.

Aniline blue test for sperm protamination-a valuable add-on to the bull breeding soundness evaluation.

Bull breeding soundness evaluation (BBSE) is the most common procedure used to predict bull potential fertility. However, the use of traditional methods for semen evaluation can affect its reliability. The inclusion of additional advanced test in BBSE may increase its accuracy. This study aimed to investigate the correlation between the degree of sperm protamination and BBSE main parameters of scrotal circumference (SC), progressive motility (PM), morphologically normal sperm (NS), and different categories of morphological defects. In addition, to determine the correlation between the three methods used for protamine assessment, five Brangus bulls were subjected to the BBSE. Semen samples were collected via electro-ejaculation and evaluated using traditional methods. Three different methods were used to determine the degree of sperm protamination: aniline blue (AB) staining, chromomycin A3 staining with fluorescent microscope (CMA3-FLM), and CMA3 with flow cytometry (CMA3-FCM). Sperm protamine deficiency assessed using the three methods exhibited significant differences among bulls according to their classification by BBSE, and showed significant negative correlation with semen quality parameters of NS and PM. A significant positive correlation was found between AB positivity and morphological abnormalities. The three methods used for protamine assessment also revealed significant positive correlations. Among the three tests, AB staining was the cheapest and easiest test that offers an objective assessment method for sperm protamination. Hence, it can be concluded that the assessment of protamination using AB staining test might serve as an additional valuable parameter or a replacement whenever detail sperm motility and morphology analyses in conducting BBSE to predict bull fertility are not possible.

Bull breeding soundness and cryopreservation of semen

The Society for Theriogenology (SFT) adopted the current form for the bull breeding soundness evaluation (BSE) in 2018. The BSE consists of four parts: a physical exam, a minimal scrotal circumference by the age of the bull, minimum progressive motility of 30%, and minimum morphology of 70% normal cells. A bull deemed to be a satisfactory potential breeder should unequivocally meet or exceed these four requirements. These standards as set forth by the SFT, give the veterinar­ian an objective approach to evaluating a bull. It has been said that “rarely are bulls infertile, but there are a lot of sub-fertile bulls”. Identification of these sub-fertile bulls by veterinarians, allows cattle producers to negate a potential negative impact on the overall productivity of the herd due to poor reproductive efficiency.&#x0D; Each part of the exam should be of equal importance to the vet­erinarian and no part should be omitted if an accurate assess­ment of the bull is to be determined. Carson et al in review of BSE trends for the past 20 years, deemed that overwhelmingly the most common cause of unsatisfactory or deferred classi­fication of bulls was due to unacceptable semen morphology. The preparation and interpretation of the morphology slide must be proficient and consistent for the results of the exam to be valid.&#x0D; The challenges that food animal practitioners encounter re­garding remaining relevant in the livestock industry continue to surface, and the profession must find means to maintain their position in a changing world. Promoting ourselves as ex­perts and the benefits of a professional exam by a veterinarian are essential to suppressing emerging imitators that provide a mediocre version of this exam sometimes referred to as the “se­men check”.

Bull sperm morphology assessment varied by evaluator

Assessment of sperm morphology is an important part of bull breeding soundness evaluation (BBSE). Whereas the effects ofevaluator experience and evaluation method on the sperm morphology estimation were assessed in other species, no such studywas conducted for bulls. Our objectives were to assess the effects of evaluator experience and number of sperm assessed on BBSEoutcomes. A single eosin-nigrosin sperm morphology slide from individual semen samples, collected from 35 yearling bullswas used. In Experiment I, 6 individuals (3 board-certified theriogenologists [DACT] and 3 fourth-year veterinary students [VS])evaluated 100 sperm from 35 slides twice (at least 1 week between evaluations). In Experiment 2, 3 DACT evaluated 100, 200,and 400 sperm from the same 5 sperm morphology slides to determine if assessing a higher number of sperm would increase theagreement of morphologic characteristics. In Experiment 1, there was a difference (p &lt; 0.0001) in the percent of sperm classifiedas morphologically normal between evaluator types (VS versus DACT). Furthermore, variation among evaluators affected spermmorphology assessments and bull breeding soundness evaluation classifications. Whereas the time needed to evaluate slides increased(p = 0.96) with increasing number of sperm assessed, there was no effect (Experiment 2) of number of sperm evaluatedon percent normal sperm, indicating that evaluating more than 100 sperm may not be justifiable. Further investigation on slidepreparation, microscope use, assessor experience, and continuing education/training is important to ensure the repeatability andvalidity of evaluating bovine sperm morphology.

Libido and serving skills of Zebu cattle bulls

The goal of this study was to evaluate the rejection rates of Zebu bulls during behavioral assessment (libido and serving ability) and the importance of this step in bull breeding soundness evaluation (BBSE). A total of 1,534 bulls reared in the state of Rio Grande do Sul, Brazil, of Nelore (1,340), Brahman (76), Guzerá (98), and Tabapuã (210) breeds were evaluated. The evaluation consisted of four steps. Step I was the general physical examination, Step II genital tract examination, Step III semen evaluation, and Step IV behavioral assessment of libido and the ability to complete copulation. To evaluate the relationship between the binary variables of approved or rejected during different steps of the BBSE, and age groups (young and mature bulls), chi-square tests and logistic regression were used. The general rejection rates in the BBSE were 5.64% and 15.20% for young and mature Zebu bulls, respectively. Problems with the sexual behavior (libido and/or serving ability) of bulls accounted for the rejection of 0.46% and 2.29% of young and mature Zebu bulls, respectively. The lack of libido and serving ability was shown to be an important problem concerning the total number of possible causes of Zebu bull rejection during the BBSE. This result suggests that the behavioral step should be performed regularly, rather than being complementary to the standard BBSE in Zebu cattle.

Scrotal circumference, bodyweight and semen characteristics in growing dairy-breed natural-service bulls in Tasmania, Australia

ABSTRACTAims: To provide herd managers with a set of decision rules allowing them to predict the likelihood that a juvenile bull is ready for Bull Breeding Soundness Evaluation (BBSE), or breeding, if bodyweight and scrotal circumference are known.Methods: This was a longitudinal study following two groups of young pasture-fed Holstein and Jersey bulls from northwest Tasmania, Australia. Individual scrotal circumference, bodyweight and semen characteristics were recorded at 6–8 weekly intervals, from 6–18 months of age. Classification and regression tree analyses were used to predict the probability that a bull had ≥70% normal sperm morphology based on scrotal circumference and bodyweight measurements.Results: Overall 1,661 scrotal circumference and bodyweight measurements were obtained, and 518 semen samples from 356 bulls were assessed for sperm morphology, from 16 examination sessions that took place between 29 May 2015 and 17 August 2016. Classification and regression tree analyses generated a decision tree for Holstein bulls with four node endpoints, and for Jersey bulls with three node endpoints. Diagnostic test performance showed that for Holstein bulls, using the node endpoints of scrotal circumference ≥27 cm and bodyweight ≥349 kg, 98% had ≥70% normal sperm (positive likelihood ratio 10.4; 95% CI = 2.7–41), and using the node endpoints of scrotal circumference ≥27 cm and bodyweight between 282–349 kg, 89% had ≥70% normal sperm (positive likelihood ratio 1.6; 95% CI = 0.9–2.6). For Jersey bulls, using the node endpoints of bodyweight ≥259 kg and scrotal circumference ≥29 cm, 88% had ≥70% normal sperm (positive likelihood ratio 3.4; 95% CI = 1.6–7.0).Conclusions: This study provides a set of relatively simple decision rules based on bodyweight and scrotal circumference measurements that allows herd managers to assess the likelihood that juvenile bulls are ready for BBSE or breeding.Abbreviations: BBSE: Bull breeding soundness evaluation; BRT: Boosted regression tree

Na/K-ATPase and Regulation of Sperm Function.

A standard bull breeding soundness evaluation (BBSE) identifies bulls with semen that is grossly abnormal. Nonetheless, semen samples classified as satisfactory based on these traditional approaches differ in fertility; perhaps there are submicroscopic differences in sperm characteristics affecting fertility. Therefore, a better understanding of molecular regulation of sperm function could promote development of novel, evidence-based approaches to predict male fertility. Recently the α4 isoform of Na/K-ATPase (ATP1A4) has received considerable attention, due to its testis- specific expression in post-meiotic germ cells and mature sperm, in addition to its regulation of sperm motility and capacitation. Using fresh bull sperm, we determined that ATP1A4 resided in specialized microdomains (raft and non-raft) of the sperm plasma membrane and activated specific signaling (caveolin-1, EGFR, Src, ERK1/2) molecules during sperm capacitation. Furthermore, ATP1A4 was the predominant isoform responsible for total Na/K-ATPase activity in capacitated sperm. Despite the widely accepted dogma of transcriptional/translational quiescence, bovine sperm translated ATP1A4 mRNA on mitochondrial or mitochondrial-type ribosomes, increasing their content and activity during capacitation. Proteomic analysis of raft and non-raft fractions revealed a significant interaction between ATP1A4 and plakoglobin, a member of the β-catenin family of proteins involved in cell adhesion, in the equatorial segment of capacitated sperm, suggesting a potential role in sperm-oolemma fusion. In frozen-thawed sperm, ATP1A4 content and activity was greater in high- versus low-fertility bulls. Additionally, ATP1A4-induced increases in ROS, calcium, actin polymerization and tyrosine phosphorylation were also involved in regulating post-thaw sperm function in these bulls. Overall, results demonstrated that ATP1A4 had unique roles in controlling several aspects of sperm physiology, acting through well-established enzyme activity and signaling functions. Consequently, isoforms of Na/K-ATPase are potential biomarkers for male fertility.

Testis-specific isoform of angiotensin-converting enzyme (tACE) as a candidate marker for bull fertility.

Although a traditional bull breeding soundness evaluation is designed to identify bulls that are grossly abnormal, bulls classified as satisfactory potential breeders still vary in fertility, implying submicroscopic differences in sperm characteristics. Testis-specific isozyme of angiotensin-converting enzyme (tACE) is involved in the regulation of sperm function. Therefore, the aim of the present study was to determine tACE content, activity and localisation in bull spermatozoa and their associations with fertility. Semen from low-fertility (LF) and high-fertility (HF) Holstein bulls (n=20) with known FERTSOL rates, which represents the 56-day non-return rate, were used. There was greater tACE content (P<0.05) and tACE activity (P<0.01) in HF versus LF spermatozoa. Based on immunolocalisation, tACE was either in the acrosomal or postacrosomal region of the sperm head, with HF bulls having a higher proportion of spermatozoa with tACE in the acrosomal region than LF bulls (P<0.05). tACE content, activity, localisation to the acrosomal region and progressive motility were significantly correlated with fertility and, based on regression analysis, tACE content was predictive of fertility. tACE content and activity in semen were similar between yearling (10-13 months old) and mature (3-4 years old) bulls. Therefore, tACE has potential as a marker of field fertility in bulls at their earliest possible age.

Traditional bull breeding soundness evaluation (BBSE) eliminates bulls that are grossly abnormal; however, bulls classified as satisfactory potential breeders still vary in field fertility, implying submicroscopic differences in sperm characteristics. The testis-specific isoform of Na/K-ATPase (ATP1A4) is involved in regulation of sperm motility and capacitation in bulls through well-established enzyme activity and signaling functions. The objective was to determine ATP1A4 content, activity and their relationship to post-thaw sperm function and field fertility, using semen samples from low-fertility (LF) and high-fertility (HF) Holstein bulls (n=20 each) with known FERTSOL rates (measure of field fertility, based on non-return rate). Frozen-thawed sperm from HF bulls had increased ATP1A4 content and activity compared to LF bulls. Furthermore, post-thaw sperm from HF bulls had increased tyrosine phosphorylation, ROS, F-actin content, and low intracellular calcium compared to LF bulls. Subsequent incubation of HF bull sperm with ouabain (a specific ligand of Na/K-ATPase) further augmented the post-thaw increase in tyrosine phosphorylation, ROS production, and F-actin content, whereas the increase in intracellular calcium was still low compared to LF bull sperm. ATP1A4 content and activity, ROS, F-actin and calcium were significantly correlated with fertility. In conclusion, we inferred that ATP1A4 content and activity differed among dairy bulls with satisfactory semen characteristics and that ATP1A4 may regulate sperm function through mechanisms involving ROS, F-actin and calcium in frozen-thawed sperm of HF and LF dairy bulls.

The value of trans-scrotal ultrasonography at bull breeding soundness evaluation (BBSE): The relationship between testicular parenchymal pixel intensity and semen quality

Bull breeding soundness evaluation (BBSE) is commonly undertaken to identify bulls that are potentially unfit for use as breeding sires. Various studies worldwide have found that approximately 20% of the bulls fail their routine prebreeding BBSE and are therefore considered subfertile. Multiple articles describe the use of testicular ultrasound as a noninvasive aid in the identification of specific testicular and epididymal lesions. Two previous studies have hypothesized a correlation between ultrasonographic testicular parenchymal pixel intensity (PI) and semen quality; however to date, no published studies have specifically examined this link. The aim of this study, therefore, was to assess the relationship between testicular parenchymal PI (measured using trans-scrotal ultrasonography) and semen quality (measured at BBSE), and the usefulness of testicular ultrasonography as an aid in predicting future fertility in bulls, in particular those that are deemed subfertile at the first examination. A total of 162 bulls from 35 farms in the South East of Scotland were submitted to routine BBSE and testicular ultrasonography between March and May 2014, and March and May 2015. Thirty-three animals failed their initial examination (BBSE1) due to poor semen quality, and were re-examined (BBSE2) 6 to 8 weeks later. Computer-aided image analysis and gross visual lesion scoring were performed on all ultrasonograms, and results were compared to semen quality at BBSE1 and BBSE2. The PI measurements were practical and repeatable in a field setting, and although the results of this study did not highlight any biological correlation between semen quality at BBSE1 or BBSE2 and testicular PI, it did identify that gross visual lesion scoring of testicular images is comparable to computer analysis of PI (P < 0.001) in identifying animals suffering from gross testicular fibrosis.

An assessment of dairy herd bulls in southern Australia: 2. Analysis of bull- and herd-level risk factors and their associations with pre- and postmating breeding soundness results.

In pasture-based, seasonally calving dairy herds of southern Australia, the mating period usually consists of an initial artificial insemination period followed by a period of natural service using herd bulls. The primary objective of this study was to identify associations between individual bull- and herd-level management factors and bull fertility as measured by a pre- and postmating bull breeding soundness evaluation (BBSE). Multivariable mixed effects logistic regression models were used to identify factors associated with bulls being classified as high risk of reduced fertility at the premating and postmating BBSE. Bulls older than 4 yr of age at the premating BBSE were more likely to be classified high risk compared with bulls less than 4 yr of age. Bulls that were in herds in which concentrates were fed before mating were more likely to be classified as high risk at the postmating BBSE compared with bulls that were in herds where concentrates were not fed.
