Monoclonal proliferation of B-cells with two integration sites of bovine leukemia virus proviral DNA in cattle with enzootic bovine leukosis.

The present study analyzed B-cell clonality and bovine leukemia virus (BLV) provirus integration sites in cattle with enzootic bovine leukosis (EBL) having BLV proviral copy numbers less or greater than the number of bovine nucleated cells. EBL cattle with BLV copy numbers less than the number of bovine nucleated cells showed monoclonal and biclonal proliferation of B-cells with one BLV provirus integration site. On the other hand, EBL cattle with BLV copy numbers greater than the number of bovine nucleated cells showed monoclonal proliferation of B-cells with two BLV provirus integration sites. These results suggest that superinfection of BLV can occur in EBL cattle.

No evidence of bovine leukemia virus proviral DNA and antibodies in human specimens from Japan

BackgroundThe potential risk and association of bovine leukemia virus (BLV) with human remains controversial as it has been reported to be both positive and negative in human breast cancer and blood samples. Therefore, establishing the presence of BLV in comprehensive human clinical samples in different geographical locations is essential.ResultIn this study, we examined the presence of BLV proviral DNA in human blood and breast cancer tissue specimens from Japan. PCR analysis of BLV provirus in 97 Japanese human blood samples and 23 breast cancer tissues showed negative result for all samples tested using long-fragment PCR and highly-sensitive short-fragment PCR amplification. No IgG and IgM antibodies were detected in any of the 97 human serum samples using BLV gp51 and p24 indirect ELISA test. Western blot analysis also showed negative result for IgG and IgM antibodies in all tested human serum samples.ConclusionOur results indicate that Japanese human specimens including 97 human blood, 23 breast cancer tissues, and 97 serum samples were negative for BLV.

Identification of novel integration sites for bovine leukemia virus proviral DNA in cancer driver genes in cattle with persistent lymphocytosis

Enzootic bovine leukosis is one of the unsolved problems of cattle breeding in many countries. The etiological agent of the disease is the bovine leukemia virus (BLV) – an oncogenic retrovirus, that infects B-lymphocytes in cattle. The number and genetic content of BLV provirus integration sites in the bovine genome were reported to can be used as an early diagnostic sign of leukemogenesis in the infected cattle, but patterns of BLV provirus integration into the bovine genome and associations between genomic features of the integration sites and development of lymphocytosis and B-cell lymphomas remain poorly elucidated. Here we present data on five novel BLV provirus integration sites in the genome of cattle with persistent lymphocytosis. Two of these sites were located in introns of scfd2 and pgpep1 genes, which have been recognized as cancer driver genes. Three of the rest integration sites were found in the intergenic spaces between ctps1 and cited4, nampt and ccdc71, skp2 and lmbrd2 genes, from which cited4 and skp2 also possess oncogenic properties. These data support previous findings of the association between localization of BLV proviral DNA near cancer driver genes and leukemogenesis in the BLV-infected cattle.

Molecular Characterization of Bovine Leukemia Virus with the Evidence of a New Genotype Circulating in Cattle from Kazakhstan.

Bovine leukemia virus (BLV) is a retrovirus that causes enzootic bovine leukosis (EBL) and has worldwide distribution. Infections with BLV have been reported in cattle from Kazakhstan but the virus has not yet been thoroughly characterized. In this study, we detect and estimate the level of BLV proviral DNA by qPCR in DNA samples from 119 cattle naturally infected with BLV, from 18 farms located in four different geographical regions of Kazakhstan. Furthermore, we conducted the phylogenetic and molecular analysis of 41 BLV env-gp51 gene sequences from BLV infected cattle. Phylogenetic analysis showed the affiliation of sequences to two already known genotypes G4 and G7 and also to a new genotype, classified as genotype G12. In addition, a multivariate method was employed for analysis of the association between proviral load and different variables such as the geographical location of the herd, cattle breeds, age of animals, and the presence of particular BLV genotypes. In summary, the results of this study provide the first evidence on molecular characterization of BLV circulating in cattle from Kazakhstan.

Potential Risk Factors Associated with Infection with Bovine Leukaemia Virus in Dairy and Beef Cattle in Taiwan.

Bovine leukaemia virus (BLV), which is classified as a Deltaretrovirus, is the aetiologic agent of enzootic bovine leukosis (EBL), a chronic lymphoproliferative disorder with a worldwide distribution. EBL is widespread in dairy herds and causes a direct economic impact due to reduced milk production and the early culling of BLV-infected cattle. The BLV infection status in Taiwan remains largely unknown; a high prevalence of BLV in dairy cows was recently revealed. The present study further investigated BLV infections in beef cattle. Surprisingly, the prevalence of BLV proviral DNA was as low as 11.8% (23/195), which is significantly lower than that noted in dairy cows, which was 42.5% (102/240) (p < 0.001). Factors associated with BLV infections were subsequently investigated. Due to the differences in herd management, an analysis of risk factors for a BLV infection was independently conducted in these two sectors. Several factors associated with a BLV infection were identified. Age was significantly associated with BLV infection status in dairy cows (p < 0.001) but not in beef cattle. A high prevalence of BLV was observed in cattle >15.5 months old (57.8%) compared with those ≤15.5 months old (11.4%). Moreover, after stratification analysis, based on the critical age of 15.5 months, as determined by the receiver operating characteristic (ROC) curve, a significantly higher BLV prevalence was demonstrated in lactating dairy cows, cattle undergoing bull breeding, heifers at older ages, and those undergoing routine rectal palpation. Due to the high prevalence of BLV in Taiwan, the development of an effective control program, based on the identified risk factors, is important for interrupting the routes of BLV transmission within herds.

Absence of bovine leukemia virus in the buffy coats of breast cancer cases from Alabama, USA

Breast cancer is reported as one of the most common and deadly cancers among females. Recent findings have suggested that bovine leukemia virus (BLV), a highly prevalent bovine virus worldwide, might be linked to human breast cancer. However, the involvement of BLV as a risk factor for breast cancer remains controversial. In this study, BLV FRET-PCR was carried out on 238 blood-derived DNA samples from breast cancer patients from the Alabama Hereditary Cancer Cohort. In addition, randomly selected samples (n = 20) were evaluated by WGS for the presence of BLV genome. No BLV proviral DNA was detected in any of 238 samples assayed by FRET-qPCR in this study. Similarly, the WGS analysis did not detect the presence of the BLV genome in the DNA of the buffy coats from 20 randomly selected patients with breast cancer. This study did not support the findings of suggesting an association between BLV and breast cancer. Notably, nearly all the studies using in situ PCR and immunohistochemistry demonstrated positive associations while other studies using whole-genome sequencing and other methods failed to identify the BLV association with breast cancer. Further studies including all reported BLV detection techniques/methods on the same breast cancer sample sets would appear to be the most likely way of resolving the current contradictory evidence.

A New Approach to the Diagnosis of Enzootic Leukosis by Genetic Markers of Bovine Leukemia Virus

In this paper, we consider the gp51 and p24 genes of the bovine leukemia virus (BLV) as "DNA-targets" for the detection of the BLV proviral DNA. We found that the gp51 and p24 genes of BLV are sufficiently specific to be used as marker sequences in PCR. The bioinformatic analysis of intraspecies specificity was conducted for each primer combination, and as a result, we found that they are complementary to most of the full-genome nucleotide sequences of BLV isolates displayed at NCBI online resources. We were able to show that the designed primer combinations possess high sensitivity, reaching 15 cells/μl in both variants. Furthermore, we compared the PCR method with the suggested primer combinations against other laboratory diagnostic methods, namely BLV-AGID and ELISA. Regarding BLV detection, the PCR method with each primer combination applied to genes gp51 and p24 separately was more effective than both AGID and ELISA; it surpassed the former by 13.5% and 14.4%, respectively, and the latter by 6.1% and 7.0%, respectively. The simultaneous use of both primer combinations on a single amplified mixture showed the largest number of BLV-infected cows (53 animals) compared to the separate amplification of each genetic marker.

The use of molecular diagnostics of BLV in the study of various biological material

It is known that milk is one of the factors in the transmission of susceptible cattle to retroviral infection, in this case, induced bovine leukemia virus (BLV). On the other hand, one of the most important aspects of studying the biological properties of BLV is the potential ability of the virus to infect xenogenic species of animals, including humans, who consume animal products and are in close contact with cattle. Milk may contain leukocytes with integrated proviral DNA, which produce mature BLV virions and the mature virions themselves, which have come from the bloodstream. The use of molecular diagnostics of this biological material will make it possible to more effectively track contaminated dairy products on farms. However, the detection of BLV proviral DNA in milk is important not only for human health, but also for animals. Since some of the products are used directly on farms, for example for feeding young animals. In regard to this, we have undertaken optimization of the methodology for the study of milk for the presence of proviral DNA of BLV. The article presents the results of the analysis of milk from 5 seropositive cows of the Holstein breed by the method of molecular diagnostics real-time polymerase chain reaction (PCR-RT). When optimizing the method of sample preparation of milk, casein and other impurities that are inhibitors of the amplification reaction were removed, in order to reduce their influence, the samples were centrifuged under various conditions. Several nucleic acid isolation techniques were used to extract proviral DNA. Based on the results of the studies, a positive amplification result was recorded in 1 out of 5 animals. At the same time, this positive result was found in test tubes with complementary DNA (cDNA) samples, it means that RNA was isolated both from the "white ring" fraction with white blood cells and monocytes, and from the sediment cells.

Of tumor diseases in the global animal husbandry industry, the greatest danger is bovine leukemia. Since the last century, this neoplastic disease has remained relevant for veterinary medicine and, in addition, more and more questions arise concerning the potential threat of the Bovine leukemia virus (BLV) for humans. The problem is being discussed (G.Yu. Kosovskiy et al., 2016) which additionally stimulates both fundamental research of the pathogen and the pathology, as well as the methods for detection to improve dairy cattle breeding and veterinary safety. A number of publications are devoted to BLV prevalence, peculiarities, the prospects for improving animal health and welfare based on selection and vaccines developed in the world (S.G. Hopkins et al., 1997; M.A. Juliarena et al., 2017). In our report, we retrospectively compare the experience of BLV eradication in the USSR, Russia and abroad, additionally focusing on the possible role of this pathogen in the occurrence of cancer in humans. In veterinary practice, serological tests, i.e., immunodiffusion (ID) assay in agar gel and enzyme-linked immunosorbent assay (ELISA) are mostly used to detect the BLV infection. G.C. Buehring et al. (2019) reported the detection of BLV proviral DNA in blood leukocytes in 38 % of patients examined by PCR and DNA sequencing. IgG antibodies to BLV were detected in 32 %, IgM — in 58 %, and IgA — in 32 % of the samples tested. Accumulated data indicate metabolic changes in the BLV-positive animals, e.g., disorder in the metabolism of tryptophan, a critical essential acid, leading to the accumulation of dangerous endogenous metabolites in the body. Particularly, free tryptophan, indole and anthranilic acid increased in level 4-8-fold are deposited in the organs of the immune and hematopoietic system (lymph nodes, spleen, liver), in the mammary gland, lungs, and kidneys. It has been established that the milk of leukemic animals differs from the milk of healthy cows in terms of physicochemical, bacteriological (lysozyme), technological parameters and mineral composition. In C57 mice fed with pasteurized milk and heat-treated meat from ID-positive cows, the blood cell profile changed. Available data drive to the unambiguous conclusion that the products derived from BLV-infected cattle pose an increased risk to humans. Despite the lack of convincing evidence of the BLV pathogenicity for humans, concerns about the etiological role of this virus in the occurance of cancer in humans additionally necessitates continuing research on the control and eradication of this common oncogenic retro-virus in livestock farms.

Detection of genotype 1 bovine leukemia virus from a C.schultzei pool: Do Culicoides spp. have a role on the transmission of bovine leukemia virus?

Bovine leukemia virus (BLV) is known as the etiological agent of Enzootic bovine leukosis (EBL), which is the most common neoplastic disease of cattle. While the major route of virus transmission is believed to be iatrogenic, BLV proviral DNA has been identified in biological materials, including nasal secretions, saliva, milk, colostrum, and semen, and in several insect species, including horses flies. However, insects' role in the natural transmission of BLV has not been clearly demonstrated. This study assessed the possible role of midges – Culicoides spp. – in BLV transmission. BLVs were genetically characterized and BLV infection seroprevelance was determined in 224 cattle sampled from 27 different small family herds in five different districts in Hatay province, southern Turkey. Out of the 25 Culicoides spp. pools, one (4.0%; 1/25) was a C.schultzei pool while 2.67% (6/224) of the sampled cattle were positive for BLV nucleic acid. The seroprevalance rates for the sampled herds and all sampled cattle were 7.40% (2/27) and 1.33% (3/224), respectively. According to the phylogenetic analysis, the sequences of the BLVs from the cattle (n = 6) and the one BLV-positive C.schultzei pool clustered on genotype 1 (G1) BLVs. Although these results do not reveal the exact role of Culicoides spp. or other midges flies in BLV transmission, the simultaneous presence of same substitions in BLVs from both cattle and a C.schultzei pool is noteworthy. Further studies on the env gene and other BLV gene regions detected from cattle and C.schultzei pools are ongoing to understand the possible epidemiological relationship between cattle and flies.

Dairy cattle with bovine leukaemia virus RNA show significantly increased leukocyte counts

Infection with bovine leukaemia virus (BLV), a retrovirus, causes dysfunction of the immune system and can have a marked economic impact on dairy industries due to decreased milk production and reduced lifespan in affected dairy cattle. The presence of proviral DNA has been the major diagnostic indicator of BLV infection. However in the course of BLV infection, the viral genome can be dormant, without detectable gene expression, resulting in limited impact on infected animals. At present, there is limited knowledge regarding haematological indices in dairy cattle that could indicate activation of the BLV genome and suggest reactivated BLV infection. In this study, BLV infection and BLV genome reactivation were evaluated based on the presence of BLV DNA and BLV env gene transcripts, respectively.BLV RNA transcription was confirmed. Among 93 whole blood samples obtained from asymptomatic dairy cattle, the prevalence of BLV proviral DNA and transcripts was 93.5% (n = 87/93) and 83.9% (n = 78/93), respectively. Between groups with and without BLV, the mean counts of white blood cells and lymphocytes in whole blood were significantly associated with the presence of BLV RNA (P < 0.05), but not with BLV proviral DNA. These results shed light on the activation status of the BLV genome and should be taken into account when evaluating the possible impact of BLV on cattle.

Detection and genotyping of bovine leukemia virus (BLV) in Vietnamese cattle.

Bovine leukemia virus (BLV) belongs to the genus, Deltaretrovirus of the family, Retroviridae and it is the causative agent of enzootic bovine leukosis. The prevalence of BLV in three provinces in the Red River Delta Region in the North of Vietnam, Hanoi, Vinhphuc and Bacninh was studied from April 2017 to June 2018. A total of 275 blood samples collected from cattle were used for serum isolation and DNA extraction. Of these samples, 266 sera were subjected to ELISA test for detecting antibody against BLV gp51 protein and 152 DNA samples were used to detect the 444 bp fragment corresponding to a part of the gp51 region of the env by nested PCR. The results showed that 16.5% (n=44) and 21.1% (n=32) of samples were positive for BLV gp51 antibody and BLV proviral DNA, respectively. Phylogenetic analysis of the partial (423 bp) and complete (913 bp) BLV env-gp51 gene indicated that Vietnamese strains were clustered into genotypes 1, 6 and 10 (G1, G6 and G10). Of those genotypes, G1 genotype was dominant; G6 strains were designated as G6e and G6f subgenotypes; the existence of genotype 10 was confirmed for the first time in Vietnam. The present study provides important information regarding the prevalence of BLV infection and genetic characteristics of BLV strains identified in Vietnam, contributing to promote the establishment of disease control and eradication strategies in Vietnam.

Absence of bovine leukemia virus proviral DNA in Japanese human blood cell lines and human cancer cell lines.

Bovine leukemia virus (BLV) infects cattle worldwide and causes B-cell lymphoma in cattle. BLV has been identified in human breast and lung cancer and in blood, but the association of BLV and human cancer is controversial. In this study, we investigated the existence of BLV in 145 Japanese human blood cell lines and 54 human cancer cell lines, using a new highly sensitive PCR assay that can amplify even one copy of BLV using LTR primers different from those in previous studies on BLV provirus in breast cancer. All samples were found negative for BLV provirus, suggesting that BLV is unlikely to infect humans.

Interlaboratory Comparison of Six Real-Time PCR Assays for Detection of Bovine Leukemia Virus Proviral DNA

Quantitative real-time PCR (qPCR) is increasingly being used for the detection of bovine leukemia virus (BLV) proviral DNA. Nevertheless, quality control for the validation and standardization of such tests is currently lacking. Therefore, the present study was initiated by three Office International des Epizooties (OIE) reference laboratories and three collaborating laboratories to measure the interlaboratory variability of six already developed and available BLV qPCR assays. For that purpose, an international panel of 58 DNA samples reflecting the dynamic range of the majority of the assays was distributed to six testing centers. Based on qualitative results, the overall agreement among all six laboratories was moderate. However, significant variability in the measurement of the BLV proviral DNA copy number was observed among different laboratories. Quantitative PCR assays, even when performed by experienced staff, can yield large variability in BLV proviral DNA copy numbers without harmonization. Further standardization of different factors (i.e., utilization of unified protocols and unique calibrators) should increase interlaboratory agreement.

On-site detection of bovine leukemia virus by a field-deployable automatic nucleic extraction plus insulated isothermal polymerase chain reaction system

Bovine leukemia virus (BLV) is a contagious, oncogenic deltaretrovirus of cattle with a worldwide distribution. In the US, over 40% of dairy cows are infected with the virus, and evidence of its economic impact is growing. This study evaluated the performance of a field-deployable automatic nucleic acid-extraction/insulated isothermal PCR (iiPCR) system for on-site BLV-proviral DNA detection in dairy cows compared with a conventional laboratory real-time PCR (rt-PCR). Assay performance was verified in parallel tests of 36 archived blood samples with 100% agreement (κ = 1.0; n = 36) between the iiPCR and conventional rt-PCR systems, and the limit of detection of the iiPCR assay was estimated to be 4 copies (genome equivalent) per reaction. The field-deployable iiPCR system was subsequently used on-farm to test freshly collected blood samples, and showed 100% agreement (κ = 1.0; n = 32) with the laboratory rt-PCR system. Fresh blood samples were collected on a second farm and tested on both systems, also with 100% agreement (κ = 1.0; n = 34). The field-deployable iiPCR/POCKIT™ combo system performs as well as a conventional laboratory-based rt-PCR system for detection of BLV proviral DNA in whole blood and may be a useful tool for on-farm evaluation of BLV-infection status in dairy cattle.

Quantification of bovine leukemia virus proviral DNA using a low-cost real-time polymerase chain reaction

The detection of bovine leukemia virus (BLV) proviral DNA is an important tool to address whether an animal is infected with BLV. Compared with serological assays, real-time PCR accounts for greater sensitivity and can serve as a confirmatory test for the clarification of inconclusive or discordant serological test results. However, the high cost related to real-time PCR assays has limited their systematic inclusion in BLV surveillance and eradication programs. The aim of the present study was to validate a low-cost quantitative real-time PCR. Interestingly, by using SYBR Green detection dye, we were able to reduce the cost of a single reaction by a factor of 5 compared with most common assays based on the use of fluorogenic probes (i.e., TaqMan technology). This approach allowed a highly sensitive and specific detection and quantification of BLV proviral DNA from purified peripheral blood leukocytes and a milk matrix. Due to its simplicity and low cost, our in-house BLV SYBR quantitative real-time PCR might be used either as a screening or as a confirmatory test in BLV control programs.

Breeding beef bulls as a source of BLV transmission

Bovine leukosis is a chronic lymphoproliferative disorder caused by bovine leukemia virus (BLV) that leads to economic losses in the beef and dairy industries. The USDA NAHMS Beef 1997 study estimated that 38% of cow-calf beef herds and 10.3% of individual adult cows in the US are BLV seropositive. Most BLV infected animals are asymptomatic carriers of the virus while a proportion of infected cattle develop lymphocytosis (30-40%) and lymphosarcoma (5-10%). The major route of virus transmission is iatrogenic through the transfer of blood from infected cattle. BLV proviral DNA has also been identified in nasal secretions, saliva, milk, colostrum, and semen, which could serve as potential sources for virus transmission. In the US, natural service accounts for 90% of the breeding in beef cow-calf operations. During natural breeding, minor genital tract trauma resulting from copulation may lead to blood transfer and transmission of BLV. Alternatively, BLV transmission may occur via transfer of infected secretions including semen and smegma during copulation. Little is known about the prevalence of BLV in breeding bulls and few studies have evaluated semen or smegma as a potential route of BLV transmission. The objectives of this study were to 1) determine the prevalence of BLV in breeding bulls presented for breeding soundness exams (BSE) at Michigan State University (MSU), 2) compare lymphocyte counts in BLV positive and negative bulls, and 3) evaluate the presence of BLV proviral DNA in bull smegma and semen.
