TLR activation, immune response and viral protection elicited in cattle by a commercial vaccine against Bovine Herpesvirus-1

The innate and acquired immune response induced by a commercial inactivated vaccine against Bovine Herpesvirus-1 (BoHV-1) and protection conferred against the virus were analyzed in cattle. Vaccination induced high levels of BoHV-1 antibodies at 30, 60, and 90 days post-vaccination (dpv). IgG1 and IgG2 isotypes were detected at 90 dpv, as well as virus-neutralizing antibodies. An increase of anti-BoHV-1 IgG1 in nasal swabs was detected 6 days post-challenge in vaccinated animals. After viral challenge, lower virus excretion and lower clinical score were observed in vaccinated as compared to unvaccinated animals, as well as BoHV-1-specific proliferation of lymphocytes and production of IFNγ, TNFα, and IL-4. Downregulation of the expression of endosome Toll-like receptors 8–9 was detected after booster vaccination. This is the first thorough study of the immunity generated by a commercial vaccine against BoHV-1 in cattle.

Phylogenetic analysis and characterization of bovine herpesvirus-1 in cattle of China, 2016–2019

Bovine herpesvirus type 1 (BoHV-1) is one of the most critical pathogens in cattle and is prevalent in China. BoHV-1 is divided into two gene types, BoHV-1.1 and 1.2, which are further differentiated into two subtypes, BoHV-1.2a and 1.2b. However, the phylogenetic analysis of BoHV-1 isolates has not been reported in China. To perform a molecular epidemiological survey based on isolates from cattle in China, 102 lung tissue samples of calves under ten months of age with respiratory disease (BRD) that died from 2016 to 2019 in China were used to isolate BoHV-1 with Madin-Darby bovine kidney (MDBK) cells. Part of the BoHV-1 isolates were applied to the phylogenetic analysis based on the region of the glycoprotein C (gC) gene of BoHV-1. Thirty BoHV-1 isolates were obtained, and the gC gene of 13 isolates was amplified by polymerase chain reaction (PCR) methods and sequenced. The result of the phylogenetic analysis according to the 451-nucleotide portion of the gC gene found that all of 13 isolates belonged to the BoHV-1.2b gene subtype, but these isolates had located two different phylogenetic tree branches. The gC gene sequence homology of isolates in group1 was higher with a reference strain of BoHV-1.2b EVI14 up to 98.0–100%, while in group 2, this was higher with reference strain BoHV-1.2b B589 up to 97.8–99.8%. The deduced amino acid sequence of gC from isolates in group 2 had two amino acid mutations with interference strain BoHV-1.2b K22 or BoHV-1.1 COOPER. The cytopathic effects (CPEs) of BoHV-1 isolates in group 2 were ulcered on the centration like a volcano on MDBK cell, and different from traditional CPEs of BoHV-1. Overall, BoHV-1.2b seems to be the primary strain of BoHV-1 in cattle in China and is also a critical cause of BRD. These BoHV-1.2b isolates had significant genetic variations.

Stress due to vaccination may induce re-activation of latent bovine herpesvirus-1 in cattle

This study evidences that, stress due to vaccination can cause re-activation and clinical outbreak of bovine herpesvirus-1 in latently infected cattle. Three to four days following mass-vaccination of cattle against Foot and Mouth Disease in Karnataka of India, symptoms of acute respiratory distress, conjunctivitis and vulvo-vaginitis were observed in vaccinated cattle in many villages of the state. Nasal and ocular swabs were collected from 25 ailing cattle in six villages of Hassan district of Karnataka. Upon bacteriological analysis, the samples were found negative for Pasteurella multocida. The serum samples from ailing animals were positive for bovine herpesvirus- 1 (BoHV-1) antibodies by Indirect-ELISA. The swab samples were found for BoHV-1 by PCR targeting glycoprotein- C gene of BoHV-1. The swab samples when subjected to virus isolation in MDBK cells yielded characteristic CPE of bunch of grape like clustering of cells by fifth passage. PCR targeting conserved region on glycoprotein-C gene on DNA extracted from cell-culture supernatants showing CPE confirmed the presence of BoHV-1. Nucleotide sequencing of the PCR amplicon showed that the BoHV-1 isolated during this study shared 100% sequence identity with BoHV-1 isolates from India, Switzerland and Brazil. This study emphasizes that stress due to vaccination can cause re-activation and clinical outbreak of bovine herpesvirus-1 in latently infected cattle.

Meta-analysis of prevalence of bovine herpes virus 1 in cattle in Mainland China

Bovine herpesvirus 1 (BHV-1), an important pathogen of cattle, can cause severe clinical syndromes including respiratory disease, genital disease, and late-term abortions, as well as neurological and systemic disease in cattle. For assessing the prevalence of BHV-1 infection in mainland China, a systematic review and meta-analysis was conducted. In this systematic review and meta-analysis, we searched English and Chinese literature databases for published paper regarding the prevalence of BHV-1 in cattle in China from inception to May 20, 2018. Search strings included if they reported the cattle samples of more than 30 cattle and provided information that allowed us to establish the prevalence of BHV-1. Moreover, we excluded repeated studies, reviews, other hosts studies, as well as studies with inconsistent data, incomplete information or only provided prevalence data, and out of mainland China data. We extracted how many cattle have BHV-1 infection from the obtained studies, moreover, and calculated pooled prevalence of BHV-1 infection in cattle. The data of 41 articles (including data on 43,441 cattle) are compliant with the standards. The pooled prevalence of BHV-1 in cattle in China was 40%, the pooled prevalence of BHV-1 in cattle from Northeast China (24%) was significant lower than those from other regions. In addition, the prevalence of BHV-1 was associated with publication time of paper, detection methods, age of cattle, and clinical symptoms (pneumonia, abortion etc.).Our findings suggest that BHV-1 is common in cattle in mainland China. It is necessary to monitor the prevalence of BHV-1 in cattle and the powerful and effective regulatory measures should be taken out to prevent the spread of BHV-1.

Prevalence of Bovine Herpesvirus-1 in cattle and buffaloes in Punjab

Aim: The aim of the present study was to identify the prevalence of Bovine Herpesvirus-1 (BHV-1) in cattle and buffaloes in the Punjab using PCR as diagnostic tool. Materials and Methods: A total of 63 samples (Semen- 57, placental cotyledons-1, vaginal secretions-1, foetal stomach contents-1 and tracheal swabs-3) from cattle and buffaloes were processed for identification of BHV-1 using PCR Results: From January 2007 to December 2010 (Semen- 57, placental cotyledons-1, vaginal secretions-1, foetal stomach contents-1 and tracheal swabs-3) from cattle and buffaloes were collected. The DNA was extracted from a total of 63 samples and subjected to PCR revealed that none of the sample positive for the BHV-1 infection. Conclusion: From the study it was concluded that the farms screened were free from BHV-1 infection.

Prevalence of serum antibodies to bovine herpesvirus-1 in cattle in Galicia (NW Spain)

Infectious bovine rhinotracheitis (IBR) is a highly infectious disease of cattle caused by bovine herpesvirus 1 (BHV-1) that causes considerable economic losses to the dairy and beef industry. This report describes the results of two surveys in which prevalence of serum antibodies against BHV-1 in Galicia (NW Spain) was determined. The first was performed on the entire cattle population of the region (year 2000) and the second on the herds included in a voluntary livestock health programme, which comprises IBR, conducted by a group of farms designated ADSG (Health Defence Groups) in this same region (year 2004). Anti-BHV-1 antibodies were determined by anti-gB ELISA in serum samples from all animals aged over one year in the area rsquo;s dairy and beef herds. Mean individual animal and herd prevalence recorded in 2000 were 38.4% (ranging 37.3%-39.5%; 43.2% dairy, 26.8% beef) and 50.4% (ranging 46.4%- 54.4%; 58.3% dairy, 48.1% beef), respectively. Mean true prevalence by herd and animal for the ADSG farms were 47.2% (44.9%-49.5%; 51.5% dairy, 45.2% beef, 42.3% mixed) and 35.7% (35.3%-36.1%; 37.8% dairy, 33.1% beef, 25.5% mixed), respectively. However, the use of non-maker vaccines make difficult to develop a control and eradication program based on serological analysis.

Quantification of the transmission of bovine herpesvirus 1 among red deer (Cervus elaphus) under experimental conditions

Bovine herpesvirus 1 (BHV1) is endemically present in a cattle population that lives in a nature reserve in the Netherlands. Red deer (Cervus elaphus), living in the same nature reserve, can come into contact with the BHV1-infected cattle and could then become infected with BHV1. For the eradication of BHV1 in cattle, it is, therefore, important to know whether red deer alone can play a role in the transmission of BHV1. For that reason, we quantified the transmission of BHV1 among farmed red deer under experimental conditions. Two groups of ten animals were formed. In each group, five of these animals were inoculated with BHV1 and the other five served as contact animals. Three inoculated animals in each transmission experiment became infected and none of the contact animals became infected. The one-sided 95% confidence interval for R [0.0-0.94] showed that limited transmission might occur among red deer. Based on these results, we would expect only minor outbreaks of BHV1 to occur in red deer populations. We concluded that BHV1 will probably not survive longer than a few decades (several times the mean deer lifetime) in red deer populations. Consequently, it is not necessary for the eradication of BHV1 in cattle to eradicate BHV1 in red deer populations as well.

A mutation in the latency-related gene of bovine herpesvirus 1 disrupts the latency reactivation cycle in calves.

Bovine herpesvirus 1 (BHV-1) is an important pathogen of cattle, and infection is usually initiated via the ocular or nasal cavity. Following acute infection, the primary site for BHV-1 latency is the sensory neuron. Reactivation from latency occurs sporadically, resulting in virus shedding and transmission to uninfected cattle. The only abundant viral transcript expressed during latency is the latency-related (LR) RNA, suggesting that it mediates some aspect of latency. An LR mutant was constructed by inserting three stop codons near the beginning of the LR-RNA, suggesting that expression of LR proteins would be altered. The LR mutant grew with wild-type (wt) efficiency in bovine kidney cells (MDBK). When calves were infected with the LR mutant, a dramatic decrease (3 to 4 logs) in ocular, but not nasal, viral shedding occurred during acute infection relative to the wt or the LR-rescued virus (M. Inman, L. Lovato, A. Doster, and C. Jones, J. Virol. 75:8507-8515, 2001). In this study, we examined the latency reactivation cycle in calves infected with the LR mutant and compared these results to those from calves infected with wt BHV-1 or the LR-rescued virus. During acute infection, lower levels of infectious virus were detected in trigeminal ganglion homogenates from calves infected with the LR mutant. As judged by in situ hybridization, BHV-1-positive neurons were detected in trigeminal ganglia of calves infected with the wt but not the LR mutant. Although LR-RNA was detected by reverse transcription-PCR in calves latently infected with the LR mutant, a semiquantitative PCR analysis revealed that lower levels of viral DNA were present in trigeminal ganglia of calves infected with the LR mutant. Dexamethasone treatment of calves latently infected with wt BHV-1 or the LR-rescued virus, but not the LR mutant, consistently induced reactivation from latency, as judged by shedding of infectious virus from the nose or eyes and increases in BHV-1-specific antibodies. In summary, this study demonstrates that wt expression of LR gene products plays an important role in the latency reactivation cycle of BHV-1 in cattle.

Suppository-Mediated DNA Immunization Induces Mucosal Immunity against Bovine Herpesvirus-1 in Cattle

Mucosal surfaces are the primary sites for the transmission of infectious agents including viruses, so effective vaccines generally should induce mucosal immunity. Furthermore, noninvasive delivery is desirable because of the ease of application, the high degree of patient compliance, and the improved safety for patients and clinicians due to the elimination of needles. Unfortunately, most of the conventional vaccines are parenterally administered and result in systemic rather than mucosal immunity. Here we present the first report of mucosal immunity by noninvasive DNA immunization in a target species. As an approach to induce mucosal immunity against bovine herpesvirus-1, cows were immunized intravaginally with suppositories containing plasmid coding for glycoprotein D. Serum IgG, as well as IgA both in the serum and in the nasal fluids, were detected, which supports the contention of a common mucosal immune system. This level of immunity was of sufficient magnitude to minimize weight loss and significantly reduce the duration of virus shedding after intranasal viral challenge, which demonstrates the efficacy of suppository-based administration of DNA vaccines to target species. As this is a very practical method of delivery, it has great potential to be applied as vaccine or therapy in a variety of species.

Modelling the effect of surveillance programmes on spread of bovine herpesvirus 1 between certified cattle herds

For the eradication of an infectious agent, like bovine herpesvirus 1 (BHV-1), surveillance and certification can be used to reduce the transmission between herds. The goal of surveillance is that a certified herd that becomes infected is detected timely so that infection of several other certified herds is prevented. What counts is whether the reproduction ratio R, i.e. the average number of certified herds infected by one infected certified herd can be kept below 1. To support policy makers in making decisions about the minimal demands for a surveillance programme in an eradication campaign of BHV-1 in cattle, two mathematical models were investigated. With these models, the basic reproduction ratio between herds was calculated. The surveillance programmes were characterised with sample size, sampling frequency, test sensitivity, herd size, vaccination status, and contacts between herds. When R between herds is below 1, then the surveillance programme is sufficiently good to prevent spread of infection, provided that R is estimated well. In the model based on bulk milk testing sample size was replaced by a threshold at which bulk milk can be found positive. The R between herds was mainly influenced by the vaccination status, sampling frequency, and contacts between herds. Herd size moderately affected the outcome. Test sensitivity and sample size, however, were of minor importance. If herds of 50 cows became free of BHV-1 without vaccination, then spread of infection between herds might be prevented when animals within herds are sampled once a year (milk or blood samples). This frequency needs to be intensified, being twice a year, for larger herds and/or herds with extensive contacts with other herds. When bulk milk is sampled instead, sampling should be done at least every 5 months and more intensively, being each month, with larger herd sizes and more contacts between herds.

Efficacy of a live glycoprotein E-negative bovine herpesvirus 1 vaccine in cattle in the field

To assess the efficacy of a live glycoprotein E-negative bovine herpesvirus 1 (BHV1) vaccine to reduce transmission of BHV1 in cattle, a randomised, double-blind, placebo-controlled field trial including 84 herds was conducted in the Netherlands. The incidence of BHV1 infections during 17 months was monitored by detecting antibodies against BHV1 glycoprotein E. In the placebo-treated group 214 seroconversions in 3985 paired sera, and in the vaccinated group 67 seroconversions in 3601 paired sera were detected. Based on these data, the transmission ratio R 0 was estimated for each treatment, using the maximum likelihood approach and the martingale approach. In placebo-treated herds R 0 was 2.5 (CI 1.4–3.1) using maximum likelihood and 2.8 (S.E. 0.4) using the martingale approach. In the vaccinated group these estimations were 1.2 (CI 0.5–1.5) and 1.5 (S.E. 0.4) respectively. The vaccinated and placebo-treated group differed significantly in transmission of BHV1. These results suggest that the use of this live gE-negative BHV1 vaccine will reduce the incidence and transmission of BHV1 infections in the field.

Prevalence of infectious bovine rhinotracheitis in southern India.

The authors describe a serological survey on the prevalence of infectious bovine rhinotracheitis (IBR) among cattle and buffalo (Bubalus bubalis) in three southern states of India. A local isolate of bovine herpesvirus 1 (BHV-1) from an outbreak of the respiratory form of IBR was used as a source of virus antigen in avidin-biotin enzyme-linked immunosorbent assay (ELISA). The overall prevalence of antibodies to BHV-1 in cattle was 50.9%, and in buffalo was 52.5%. Among breeding bulls, 114/120 samples from Tamil Nadu (95%) and 41/99 samples from Karnataka (41.4%) were seropositive. The possible association of IBR with bovine abortions was recorded in 31/56 samples (55.4%) from aborted crossbred cows. However, virus isolation was not performed on these animals. The authors also highlight the economic importance of IBR to the rapidly developing livestock industry in India.

A subunit gIV vaccine, produced by transfected mammalian cells in culture, induces mucosal immunity against bovine herpesvirus-1 in cattle

A truncated version of bovine herpesvirus-1 (BHV-1) glycoprotein IV (tgIV) was produced in a novel, non-destructive expression system based upon regulation of gene expression by the bovine heat-shock protein 70A (hsp70) gene promoter in Madin Darby bovine kidney (MDBK) cells. In this system, up to 20 micrograms ml-1 of secreted tgIV, which is equivalent to the yield from 4 x 10(6) cells, was produced daily over a period of up to 18 days. Different doses of tgIV were injected intramuscularly into seronegative calves. Virus-neutralizing antibodies were induced by all doses of tgIV, both in the serum and in the nasal superficial mucosa. However, the low dose (2.3 micrograms) induced significantly (p < 0.05) lower antibody titres than the medium (7 micrograms) and high (21 micrograms) doses. The medium and high doses of tgIV conferred protection from BHV-1 infection, as demonstrated by a significant (p < 0.05) reduction in clinical signs of respiratory disease and virus shedding in the nasal secretions postchallenge. However, the 2.3 micrograms group, although partially protected, was not significantly (p > 0.05) different from the placebo group. This study demonstrated the potential of an intramuscularly administered tgIV subunit vaccine to induce mucosal immunity to BHV-1 using an economic protein production system and an acceptable vaccine formulation. In addition, a strong correlation was observed between neutralizing antibodies in the serum and nasal superficial mucosa, virus shedding and clinical disease. Thus, serum neutralizing antibody levels in tgIV-immunized animals may be a good prognosticator of protection from BHV-1 infection and disease.

Bovine herpesvirus 1: rapid diagnosis of infection by direct filter hybridization.

Direct filter hybridization (DFH) was applied as a simple method of nucleic acid hybridization to diagnose bovine herpesvirus 1 (BHV-1) infection without previous purification of nucleic acids from the specimens. The DNA of BHV-1 was cleaved with the restriction endonuclease Pst I and randomly cloned into pKH47 plasmids. The clones were labelled with 32P or biotin and selected on uninfected and infected cells for the highest specific activity to detect BHV-1 infection. Two clones, which detected about 10 infected cells, were selected for the diagnosis of BHV-1 in cattle. On specimens collected during experimental and natural disease, the DFH showed to be in concordance with the standard method of virus isolation. This simple hybridization technique proved to be a sensitive and rapid alternative to virus isolation. Specific diagnosis of BHV-1 infection can be made even in simply equipped laboratories within 10 h.
