Characterizing and identifying of miRNAs involved in berberine modulating glucose metabolism of Megalobrama amblycephala.

The present study, as one part of a larger project that aimed to investigate the effects of dietary berberine (BBR) on fish growth and glucose regulation, mainly focused on whether miRNAs involve in BBR's modulation of glucose metabolism in fish. Blunt snout bream Megalobrama amblycephala (average weight of 20.36 ± 1.44g) were exposed to the control diet (NCD, 30% carbohydrate), the high-carbohydrate diet (HCD, 43% carbohydrate) and the berberine diet (HCB, HCD supplemented with 50mg/kg BBR). After 10weeks' feeding trial, intraperitoneal injection of glucose was conducted, and then, the plasma and liver were sampled at 0h, 1h, 2h, 6h, and 12h. The results showed the plasma glucose levels in all groups rose sharply and peaked at 1h after glucose injection. Unlike the NCD and HCB groups, the plasma glucose in the HCD group did not decrease after 1h, while remained high level until at 2h. The NCD group significantly increased liver glycogen content at times 0-2h compared to the other two groups and then liver glycogen decreased sharply until at times 6-12h. To investigate the role of BBR that may cause the changes in plasma glucose and liver glycogen, miRNA high-throughput sequencing was performed on three groups of liver tissues at 2h time point. Eventually, 20 and 12 differentially expressed miRNAs (DEMs) were obtained in HCD vs NCD and HCB vs HCD, respectively. Through function analyzing, we found that HCD may affect liver metabolism under glucose loading through the NF-κB pathway; and miRNAs regulated by BBR mainly play roles in adipocyte lipolysis, niacin and nicotinamide metabolism, and amino acid transmembrane transport. In the functional exploration of newly discovered novel:Chr12_18892, we found its target gene, adenylate cyclase 3 (adcy3), was widely involved in lipid decomposition, amino acid metabolism, and other pathways. Furthermore, a targeting relationship of novel:Chr12_18892 and adcy3 was confirmed by double luciferase assay. Thus, BBR may promote novel:Chr12_18892 to regulate the expression of adcy3 and participate in glucose metabolism.

Characterization of type II IFNs and their receptors in a cyprinid fish, the blunt snout bream Megalobrama amblycephala

Type II interferons (IFNs) are a key class of molecules regulating innate and adaptive immunity in vertebrates. In the present study, two members of the type II IFNs, IFN-γ and IFNγ-rel, were identified in the blunt snout bream (Megalobrama amblycephala). The open reading frame (ORF) of IFN-γ and IFNγ-rel was found to have 564 bp and 492 bp, encoding 187 and 163 amino acids, with the first 26 and 24 amino acids being the signal peptide, respectively. IFN-γ and IFNγ-rel genes showed a high degree of similarity to their zebrafish homologues, being 76.9 % and 58.9 %, respectively. In the phylogenetic tree, IFN-γ and IFNγ-rel were clustered with homologous genes in cyprinids. In blunt snout bream, IFN-γ and IFNγ-rel were constitutively expressed in trunk kidney, head kidney, spleen, liver, heart, muscle, gill, intestine and brain and were significantly up-regulated by poly (I:C) induction in head kidney, spleen, liver, gill and intestine. Using recombinant proteins of IFN-γ and IFNγ-rel, the surface plasmon resonance (SPR) results showed that IFN-γ was bound to CRFB6, CRFB13 and CRFB17, but mainly to CRFB6 and CRFB13, whereas IFN-γrel bound mainly to CRFB17 and had no affinity with CRFB6. These results contribute to a better understanding on type II IFNs and their receptor usage in teleost fish.

Molecular characterization of nineteen cytokine receptor family B (CRFB) members, CRFB1, CRFB2, CRFB4-17, with three CRFB9 and two CRFB14 in a cyprinid fish, the blunt snout bream Megalobrama amblycephala

The class II cytokine receptor family members are receptors of class 2 helical cytokines in mammals, and are named cytokine receptor family B (CRFB) in fish. In zebrafish, sixteen members, including CRFB1, CRFB2 and CRFB4-17 were reported. With the availability of genome sequence, a total of nineteen CRFBs was identified in the blunt snout bream (Megalobrama amblycephala), including CRFB1, CRFB2, CRFB4-17 with the presence of three CRFB9 isoforms, and two CRFB14 isoforms. These CRFB molecules contain well conserved features, such as fibronectin type III (FNIII) domain, transmembrane and intracellular domains as other class II cytokine receptors, and are phylogenetically grouped into thirteen clades with their homologues from other species of fish. The CRFB genes were constitutively expressed in organs/tissues examined in the fish. The finding of more CRFB members in the bream may provide clues to understand possible receptor-ligand interaction and their diversity from an evolutionary point of view.

Changes in physiological responses and immunity of blunt snout bream Megalobrama amblycephala from transport stress.

This study aimed to investigate the effects of transport stress on the physiological responses and immunity of Megalobrama amblycephala (blunt snout bream). Fish (109.67 ± 1.51g) were sampled at nine time points: before transport (control), at 0h, 1h, 3h, 6h, 12h, 24h, 3days, and 7days after 4h of medium-distance transportation, and four fish were sampled in each time point. The results showed that plasma cortisol, triiodothyronine (T3), complement component 3 (C3), complement component 4 (C4), immunoglobulin M (IgM) and nitrogen monoxide (NO) concentrations, and alternative complement pathway (ACH50), acid phosphatase (ACP), and myeloperoxidase (MPO) activities all reached the peak at 0h after transportation; C4 and NO concentrations as well as ACP and MPO activities returned to the control level after 1h, ACH50 activity as well as cortisol, T3, and IgM concentration returned to the control level after 12h, and C3 concentration returned to the control level after 24h respectively. Plasma glucose and total protein concentrations as well as lysozyme activity all reached the peak at 1h after transportation, total protein concentration and lysozyme activity returned to the control level after 3h, and glucose concentration returned to the control level after 6h (P < 0.05). Liver heat shock protein 70 expression reached the peak at 1h after transportation, and returned to the control level after 24h; liver heat shock protein 90 expression reached the peak at 0h after transportation and returned to the control level after 12h (P < 0.05). Overall, these findings suggested that 4h of medium-distance transportation caused stress response of blunt snout bream, and transport stress had a significant effect on plasma indicators. But the recovery of 24h after transport could return the physiological response, immune indexes, and the expression of heat shock protein to the normal level. This also provided data support for the medium-distance transportation of blunt snout bream in the future.

Hydroxytyrosol Attenuates High-Fat-Diet-Induced Oxidative Stress, Apoptosis and Inflammation of Blunt Snout Bream (Megalobrama amblycephala) through Its Regulation of Mitochondrial Homeostasis

The present study was conducted to investigate the effects of dietary hydroxytyrosol (HT) on oxidative stress, inflammation and mitochondrial homeostasis in blunt snout bream (Megalobrama amblycephala). Fish were fed a low-fat diet (LFD, 5% lipid), a high-fat diet (HFD, 15% lipid), an LFD supplementing 200 mg/kg HT, or an HFD supplementing 200 mg/kg HT. After 10-week feeding, significant reduction of growth was observed in fish fed HFD, compared with other groups. HFD caused oxidative stress and more apoptosis of hepatocytes, while HT addition resulted in significant decrease of ROS and MDA contents, and the apoptotic hepatocytes. Moreover, the expression of genes involving inflammation of HFD group were elevated. Supplementing HT to HFD can attenuate this. All the activities of complexes of mitochondria in the HFD group were decreased compared with those in the LFD group, while supplementing HT to HFD significantly increased complex I-III activities. Furthermore, HFD downregulated the expressions of Atg5 and NRF-1 which induced the failure of mitophagy and biogenesis, while, supplementing HT to HFD reversed these expressions involving mitochondrial autophagy and biogenesis. In summary, adding HT to HFD relieved oxidative stress, apoptosis and inflammation, likely due to its regulation of mitochondrial homeostasis.

Effects of Pantothenic Acid on Growth Performance, Oxidative Stress, and Lipid Content in High-Fat-Fed Megalobrama amblycephala

This study was to elucidate the effects of dietary pantothenic acid (PA) on growth, lipid metabolism, and antioxidant status in juvenile blunt snout bream Megalobrama amblycephala. Fish were fed with four experimental diets containing two lipid levels (5% and 11%) and two PA supplementation levels (24 mg kg-1 and 240 mg kg-1) for eight weeks. The results showed that diets containing 11% lipid significantly increased condition factor, mesenteric fat index, triacylglycerol (TG), and cholesterol (TCH) in the liver as well as total lipid content and TCH in the muscle ( P &lt; 0.05 ), but significantly decreased specific growth rate and whole-body protein ( P &lt; 0.05 ). As the dietary lipid level increased, hepatic antioxidant enzyme activities were significantly increased ( P &lt; 0.05 ) as well as malondialdehyde content ( P &lt; 0.05 ). Besides, cytosolic cytochrome c concentration was also significantly ( P &lt; 0.05 ) increased, while mitochondrial protein, cytochrome c, and Na+-K+-ATPase activities were significantly decreased ( P &lt; 0.05 ). There were no significant changes on growth performance and body composition of PA supplementation. Then, significantly negative ( P &lt; 0.05 ) interactions were observed between dietary lipid and PA supplementation on EPA, DHA, and EPA + DHA content of the muscle. Saturated fatty acid (SFA) and polyunsaturated fatty acid (PUFA) contents in the liver were significantly increased ( P &lt; 0.05 ) with dietary lipid level increased, while liver monounsaturated fatty acid (MUFA) was significantly decreased ( P &lt; 0.05 ). Muscle SFA, DHA, and EPA + DHA contents were significantly decreased ( P &lt; 0.05 ) when dietary fat level increased. Meanwhile, EPA, DHA, and EPA + DHA content was significantly decreased ( P &lt; 0.05 ) with dietary high PA supplementation. On the whole, a high PA supplementation level of 240 mg kg-1 could alleviate lipid content accumulated and oxidative damage induced by a high-fat diet.

Transcriptome Analysis Reveals Sexual Disparities between Olfactory and Immune Gene Expression in the Olfactory Epithelium of Megalobrama amblycephala.

The olfactory organ is an important chemoreceptor in vertebrates. However, the sexual disparities in gene expression patterns in the olfactory organ in fish remain unstudied. Here, we conducted a transcriptome analysis of the olfactory epithelium (OE) of male and female blunt snout bream (Megalobrama amblycephala) to identify the differences. The histological analysis showed that there were 22 leaf-like olfactory lamellaes on one side of the OE of the adult blunt snout bream. The sensory area of OE is enriched with ciliated receptor cells and microvilli receptor cells. The transcriptome analysis showed that only 10 out of 336 olfactory receptor genes (224 ORs, 5 V1Rs, 55 V2Rs, and 52 TAARs) exhibited significant expression differences between males and females, and most of the differentially expressed genes were related to the immune system. We also validated these results using qPCR: 10 OR genes and 6 immunity-related genes significantly differed between males and females. The FISH analysis results indicated that the ORs were mainly expressed at the edge of the olfactory lamellae. Collectively, our study reveals that gender is not an important factor influencing the expression of olfactory receptors, but the expression of immune genes varies greatly between the genders in blunt snout bream.

Nutrient metabolism in the liver and muscle of juvenile blunt snout bream (Megalobrama amblycephala) in response to dietary methionine levels

A 75-day rearing trial was designed to study the response of juvenile Megalobrama amblycephala to dietary methionine (Met) levels. Three practical diets with graded Met levels (0.40%, 0.84% and 1.28% dry matter) were prepared to feed the juvenile fish. The results showed that the 0.84% Met diet significantly improved the growth compared with 0.40% diets. Compared with 0.84% and 1.28% Met, 0.40% Met significantly increased the hepatic lipid content, while decreasing the muscular lipid and glycogen contents. 0.40% Met decreased the protein levels of phospho-Eukaryotic initiation factor 4E binding protein-1 (p-4e-bp1), 4e-bp1 and Ribosomal protein S6 kinase 1 in the liver, compared with 0.84% diet, while an increasing trend was observed in the muscle. Met supplementation tended to decrease and increase lipid synthesis in the liver and muscle, respectively, via changing mRNA levels of sterol regulatory element-binding protein 1, fatty acid synthetase and acetyl-CoA carboxylase. 1.28% dietary Met promoted fatty acid β-oxidation and lipolysis in both the liver and muscle by increasing carnitine palmitoyl transferase 1, peroxisome proliferator activated receptor alpha, lipoprotein lipase and lipase mRNA levels. Compared with 0.40% and 0.84% dietary Met, 1.28% Met enhanced the mRNA levels of hepatic gluconeogenesis related genes phosphoenolpyruvate carboxykinase (pepck), and glucose-6-phosphatase, and muscular glycolysis related genes phosphofructokinase (pfk), and pyruvate kinase (pk). The mRNA levels of hepatic pfk, pk and glucokinase were markedly downregulated by 1.28% Met compared with 0.84% level. Muscular pepck, glycogen synthase, and hepatic glucose transporters 2 mRNA levels were induced by 1.28% Met. Generally, deficient Met level decreased the growth of juvenile Megalobrama amblycephala, and the different nutrient metabolism responses to dietary Met were revealed in the liver and muscle.

The role of dietary chromium supplementation in relieving heat stress of juvenile blunt snout bream Megalobrama amblycephala

The present study assessed the role of dietary chromium (Cr) supplementation in relieving heat stress (HS) of juvenile blunt snout bream Megalobrama amblycephala. The supplemented Cr contents by chromium picolinate (Cr-Pic) was 0 mg/kg (control group), 0.4 mg/kg, 1.6 mg/kg and 12.0 mg/kg, respectively. The fish continued to be fed four diets at suitable temperatures (26 °C) for 2 weeks, and then the temperature was then heated up to 33 °C through thermo-regulated system. The results showed that Cr supplementation had no significant effect on the immune indices and antioxidant indices before HS (P > 0.05). However, Cr supplementation played an important role in relieving HS. After HS, compared with the control group, 1.6 mg/kg and 12.0 mg/kg Cr supplementation groups significantly lowered the plasma glucose level and aspartate transaminase (AST) activity (P < 0.05), and 0.4 mg/kg and 1.6 mg/kg Cr supplementation groups significantly lowered alanine aminotransferase (ALT) activity (P < 0.05). 0.4 mg/kg and 1.6 mg/kg supplementation groups significantly improved hepatic total superoxide dismutase (T-SOD) activity (P < 0.05). Furthermore, 0.4mg/kg-12.0 mg/kg Cr supplementation groups significantly improved the activities of hepatic glutathione peroxidase (GPx) and catalase (CAT) and lowered hepatic malondialdehyde (MDA) level in liver (P < 0.05). The mRNA levels of hepatic copper zinc superoxide dismutase (Cu/Zn-SOD), CAT and GPx were significantly improved in 0.4mg/kg-12.0 mg/kg supplementation Cr groups (P < 0.05), however, there was no significant variation of hepatic manganese superoxide dismutase (Mn-SOD) mRNA levels under different levels of supplementation (P > 0.05). Significantly lower mRNA levels of hepatic pro-inflammatory cytokines observed in 0.4mg/kg-12.0 mg/kg Cr supplementation groups including tumour necrosis factor-α (TNF-α), interleukin 1β (IL-1β) and interleukin 8 (IL-8) (P < 0.05), and 0.4mg/kg-12.0 mg/kg Cr supplementation significantly improved the relative expressions of hepatic heat shock protein 70 (HSP70) and heat shock protein 90 (HSP90) (P < 0.05). The present study indicated that dietary Cr supplementation might have no significant effect on immune capacity and antioxidant capacity under normal physiological conditions, whereas it played an important role in relieving HS.

Utilization of pelleted and extruded feed by blunt snout bream Megalobrama amblycephala: Insights from growth performance, health status and feed cost.

The present study was conducted to evaluate the utilization of both pelleted feed (PF) and extruded feed (EF) by blunt snout bream Megalobrama amblycephala based on growth performance, stress responses, innate immunity and disease resistance. Both the PF and EF were prepared with the same formula. Fish were divided randomly into 2 groups, including one fed the PF continuously and one offered the EF continuously. The whole feeding trial lasted 8weeks, after which fish were subjected to Aeromonas hydrophila infection. The results showed that the feed intake, feed conversion ratio, hepatic total superoxide dismutase activity and glutathione content, plasma complement 3 and complement 4 levels as well as myeloperoxidase activity of the EF group were all significantly lower than those of the PF group, while the opposite was true for the condition factor, the viscera index, the abdominal fat percentage, nitrogen and energy retention, hepatic malondialdehyde content, plasma levels of cortisol, glucose, lactate, total protein and globulin as well as the activities of plasma alanine aminotransferase and aspartate aminotransferase. In addition, the EF group also obtained relatively low activities of hepatic glutathione peroxidase and plasma acid phosphatase as well as high cumulative mortality rates at 24-96h after Aeromonas hydrophila challenge. Furthermore, the feed cost of culturing this species with EF is lower than that with PF. These findings indicated that compared with PF, EF could increase the feed utilization and economic benefits of blunt snout bream, but reduce its anti-stress ability, non-specific immunity, A. hydrophila resistance and feed cost.

Utilization of raw and gelatinized starch by blunt snout bream Megalobrama amblycephala as evidenced by the glycolipid metabolism, glucose tolerance and mitochondrial function

This study aimed to evaluate the influences of dietary raw and gelatinized wheat starch and their ratios on the glycolipid metabolism, glucose tolerance and mitochondrial function of blunt snout bream Megalobrama amblycephala. Three diets incorporating 100% raw starch (R100), 100% gelatinized starch (G100) and 75% raw starch +25% gelatinized starch (R75/G25) were randomly assigned to 15 cages of fish for 10 weeks, after which fish were sampled. Then the remaining fish within each treatment were combined and later split into 6 tanks for a time series sampling in a glucose tolerance test. Daily gain index, relative feed intake and plasma levels of glucose, triglyceride, pyruvate and lactate all showed little difference among all the treatments. The R75/G25 group obtained the highest feed efficiency and tissue (namely liver and the abdominal fat tissue) glycogen contents. However, the highest tissue (namely muscle, liver and the abdominal fat tissue) lipid contents and plasma total cholesterol levels were both observed in the G100 group. In addition, the G100 group obtained significantly higher transcriptions of glucose transporter 2, glucokinase, peroxisome proliferator activated receptor γ and acetyl-CoA carboxylase α, but relatively low transcripts of the genes involved in gluconeogenesis and fatty acid β-oxidation, compared with the R100 group. The R100 group showed the highest glucose clearance rate, while the area under the glucose curve showed no statistical difference. Furthermore, the R100 group had relatively low activities of mitochondrial complex I, II, III and V as well as the transcriptions of cytochrome c oxidase 2, cytochrome b and NADH dehydrogenase 1 and ATP synthase 6 compared with other groups. Overall, our results suggested that high dietary inclusions of gelatinized starch could enhance the glucose transportation, glycolysis, lipogenesis, lipid deposition, glucose tolerance and mitochondrial function of blunt snout bream, while depress the gluconeogenesis and fatty acid oxidation.

Molecular Characterization of PGC-1β (PPAR Gamma Coactivator 1β) and its Roles in Mitochondrial Biogenesis in Blunt Snout Bream (Megalobrama amblycephala)

This study aimed at achieving the molecular characterization of peroxisome proliferator-activated receptor-gamma coactivator 1β (PGC-1β) and exploring its modulatory roles in mitochondria biogenesis in blunt snout bream (Megalobrama amblycephala). A full-length cDNA of PGC-1β was cloned from liver which covered 3110 bp encoding 859 amino acids. The conserved motifs of PGC-1β family proteins were gained by MEME software, and the phylogenetic analyses showed motif loss and rearrangement of PGC-1β in fish. The function of PGC-1β was evaluated through overexpression and knockdown of PGC-1β in primary hepatocytes of blunt snout bream. We observed overexpression of PGC-1β along with enhanced mitochondrial transcription factor A (TFAM) expression and mtDNA copies in hepatocytes, and its knockdown led to slightly reduced NRF1 expression. However, knockdown of PGC-1β did not significantly influence TFAM expression or mtDNA copies. The alterations in mitochondria biogenesis were assessed following high-fat intake, and the results showed that it induces downregulation of PGC-1β. Furthermore, significant decreases in mitochondrial respiratory chain activities and mitochondria biogenesis were observed by high-fat intake. Our findings demonstrated that overexpression of PGC-1β induces the enhancement of TFAM expression and mtDNA amount but not NRF-1. Therefore, it could be concluded that PGC-1β is involved in mitochondrial biogenesis in blunt snout bream but not through PGC-1β/NRF-1 pathway.

Screening of Biomarkers Related to Ovarian Maturation and Spawning in Blunt Snout Bream (Megalobrama amblycephala) Based on Metabolomics and Transcriptomics.

In fish breeding practices, gamete maturity of females is vital to reproductive success. For some species, it is possible to estimate the female maturation status based on abdomen observation, but quite difficult for some species which mature at big size. To screen out the potential biomarker in fish blood relating to female maturation, we employed the approach integrating the UPLC-MS/MS and RNA-seq techniques to investigate the metabolites and genes reflecting the sexual maturation and spawning of female blunt snout bream Megalobrama amblycephala. The study included four groups, 1-year-old immature female individuals, 2-year-old immature female individuals, 2-year-old sexually mature female individuals, and 2-year-old sexually mature female individuals after 24h of successful spawning. The upregulated metabolites in mature females were involved in "steroid hormone biosynthesis," "metabolic pathways," "glycerophospholipid metabolism," etc. compared with those of immature individuals. As the key intermediate of steroid hormone biosynthesis, 17α-hydroxypregnenolone exhibited the highest level in 2-year-old mature females than in the immature females. Meanwhile, the metabolites (i.e., dodecanoic acid and myristic acid) participating in fatty acid synthesis exhibited much lower levels in the females after spawning than those before spawning. In addition to the metabolites, the genes involved in ovarian steroidogenesis were significantly upregulated in the 2-year-old immature females compared to the 1-year-old immature females, indicating that the ovarian steroidogenesis plays important roles in ovarian development of M. amblycephala at the early stages. The significant upregulation of genes (i.e., itpr1, camk2, and mekk2) involved in the "GnRH signaling pathway" was observed in the mature females compared with the immature females, which indicated that the estrogen levels increased after female maturation in M. amblycephala. Moreover, many genes (e.g., gck, creb1, tf2-9, ryr2, asgr1, and creb1) regulating insulin secretion and thyroid hormone synthesis were significantly downregulated after female spawning. The dynamics of gene expression and metabolites observed in this study provide novel cues for guiding fish practical artificial reproduction.

Photoperiod affects blunt snout bream (Megalobrama amblycephala) growth, diel rhythm of cortisol, activities of antioxidant enzymes and mRNA expression of GH/IGF-I.

Juvenile blunt snout bream Megalobrama amblycephala were reared under three photoperiods, 8 L: 16D, 12 L: 12D and 16 L: 8D (L: light; D: dark) for over 8 weeks. The results showed that growth performance was significantly enhanced by long photoperiod. Contrary to feed conversion ratio, final length, final weight, weight gain and feed intake increased significantly as illumination time increased from 8 h daily to 16 h daily. Low size heterogeneity and whole-body lipid content in fish exposed to long photoperiod were also observed. Both relative mRNA expression level of GH and IGF-I increased as illumination time increased from 8 h to 16 h daily. The lowest value of plasma cortisol was observed at the middle the photophase while the highest value was observed at the transition between the day and night span. Hepatic MDA content significantly increased as illumination time increased from 8 h daily to 16 h daily. The activities of hepatic catalase and glutathione peroxidase were lowest in fish exposed to 16 L: 8D photoperiod and significantly lower than that in fish exposed to 8 L: 16D. These results indicate that photoperiod manipulation may not only improve growth performance but also reduce size heterogeneity. However, prolonged photoperiod could cause chronic stress since plasma cortisol level was higher in the long photoperiod group, leading to an increasing oxidative stress.

Comparative expression analysis of let-7 microRNAs during ovary development in Megalobrama amblycephala.

As a critical regulator of gene expression, let-7 family miRNAs have been reported to be involved in multiple physiological processes. In this study, in order to elucidate the putative regulatory effect of let-7 miRNAs during fish gonadal development and to identify which member is crucial for this regulation, the expression of ten members including let-7a/b/c/d/e/f/g/h/i/j were quantified in ovary, pituitary, and brain tissues during the different ovarian developmental stages of blunt snout bream Megalobrama amblycephala. According to the data from analysis of expression patterns, let-7a showed the highest expression value in almost all the tested ovaries, pituitaries, and brains, with let-7b and let-7d moderately expressed, following by other let-7 miRNAs. In terms of the differential expression levels of ten let-7 miRNAs at each developmental stage, the results showed that let-7a/b/d/f/h expression gradually increased during the ovary development from stage I to V and dropped significantly at the phase VI in ovary tissues. However, the expression of let-7a/b/e/f in pituitary increased during the ovary development from stage I to IV and declined at stage V. Among all the let-7 miRNAs, let-7a/b/d had the highest expression and their expression patterns were consistent with the gonad development of M. amblycephala. Furthermore, the mostly predicted target genes of let-7 miRNAs are significantly enriched in signaling pathways closely related to gonadal development through KEGG enrichment analysis. These results indicate that let-7 miRNA members, especially let-7a/b/d, may play important roles in the regulation of ovary development in M. amblycephala through negatively regulating expression of their target genes.

Effects of the total fish meal replacement by soybean meal on growth parameters, serum biochemistry, and hepatic and intestinal histology of juvenile blunt snout bream (Megalobrama amblycephala)

ABSTRACTFish meal is the main protein source in fish diet, but due to its unsustainability, it has to be replaced with more sustainable protein sources, such as soybean meal (SBM), in fish feeds. As SBM often produces negative effects on fish growth and health, we conducted an eight-week trial of complete fish meal replacement with SBM in the diet of juvenile (48.2 ± 0.55 g) blunt snout bream (Megalobrama amblycephala), a herbivorous cyprinid fish native to the Yangtze basin (China). SBM-fed fish exhibited: a worsened growth performance; improved survival rate (P > 0.05); elevated serum triglycerides and total cholesterol but reduced low-density lipoprotein in plasma (P < 0.05); mild lipid droplet accumulation and swelling of the liver; elevated goblet cell counts and mild signs of enteritis in the intestines. With negligible effects on growth and mild effects on health, M. amblycephala appears to be comparatively tolerant of SBM, but total fish meal replacement is not recommended. Future studies should aim to identify the SBM tolerance threshold in long-term growth trials.

Resveratrol Improves the Energy Sensing and Glycolipid Metabolism of Blunt Snout Bream Megalobrama amblycephala Fed High-Carbohydrate Diets by Activating the AMPK-SIRT1-PGC-1α Network.

This study investigated the effects of resveratrol on the growth performance, energy sensing, glycolipid metabolism and glucose and insulin load of blunt snout bream Megalobrama amblycephala fed high-carbohydrate diets. Fish (39.44 ± 0.06 g) were randomly fed three diets: a control diet (30% carbohydrate), a high-carbohydrate diet (HC, 41% carbohydrate), and the HC diet supplemented with 0.04% resveratrol (HCR) for 12 weeks. Fish fed the HC diet had significantly high values of nitrogen and energy retention efficiency, hepatosomatic index, intraperitoneal fat ratio, whole-body lipid content and intraperitoneal fat glycogen and lipid contents compared to the control group, but showed little difference with the HCR treatment. Liver and muscle lipid contents and plasma levels of glucose, glycated serum protein, advanced glycation end products and total cholesterol of fish fed the HC diet were significantly higher than those of the control group, whereas the opposite was found with resveratrol supplementation. Fish fed the HC diet obtained significantly low values of plasma insulin levels and hepatic adenosine monophosphate (AMP) contents and NAD+/NADH ratio compared to HCR treatment, but showed little difference with the control group. The opposite was found for hepatic adenosine triphosphate (ATP) contents and the ATP/AMP ratio. In addition, fish fed the HC diet showed significantly high transcriptions of glucose transporter 2 (GLUT2), glucose-6-phosphate dehydrogenase, glycogen synthase, fatty acid synthetase (FAS), acetyl-CoA carboxylase α (ACCα), peroxisome proliferator-activated receptor γ and PPARα compared to the control group, whereas the opposite was found for protein levels of AMP-activated protein kinase α (t-AMPKα), phosphorylated AMP-activated protein kinase α (p-AMPKα), sirtuin-1 (SIRT1), and p-AMPKα/t-AMPKα ratio as well as the transcriptions of AMPKα1, AMPKα2, SIRT1, PPARγ coactivator-1α (PGC-1α), phosphoenolpyruvate carboxykinase, fructose-1,6-bisphosphatase (FBPase), glucose-6-phosphatase, carnitine palmitoyl transferase I (CPT I) and acyl-CoA oxidase. Resveratrol supplementation significantly up-regulated the protein levels of t-AMPK, p-AMPK, and SIRT1, p-AMPK/t-AMPK ratio as well as the transcriptions of AMPKα1, AMPKα2, SIRT1, PGC-1α, GLUT2, FBPase, and CPT I compared to HC group, while the opposite was found for sterol regulatory element-binding protein-1, FAS and ACCα. Furthermore, resveratrol improved glucose and insulin tolerance of fish fed the HC diet after glucose and insulin load.

Regulation of mitochondrial biosynthesis and function by dietary carbohydrate levels and lipid sources in juvenile blunt snout bream Megalobrama amblycephala

This study aimed to investigate the effects of dietary non-protein energy adjustments on the mitochondrial biosynthesis and function of juvenile Megalobrama amblycephala. Fish (average weight: 37.98 ± 0.07 g) were fed eight diets containing two dietary carbohydrate levels (30% and 43%) and four lipid sources (fish oil, soybean oil, palm oil and the mixed oil) for 11 weeks. Liver mitochondrial respiratory chain complex V activity and ATP (adenosine triphosphate) content both increased significantly with increasing dietary carbohydrate levels, whereas the opposite was true for the AMP (adenosine 5′-monophosphate)/ATP ratio, hepatic transcripts of AMP-activated protein kinase α1 (AMPKα1), AMPKα2, peroxisome proliferators γ-activated receptor coativator-1α (PGC-1α), NADH dehydrogenase 1 and cytochrome c oxidase 1 (COX1) as well as the activities of Na+-K+-ATPase, succinate dehydrogenase (SDH), citrate synthase (CS) and mitochondrial respiratory chain complex I, III and IV. Additionally, hepatic ATP content, the transcripts of AMPKα, COX1 and ATP6 and the activities of Na+-K+-ATPase, SDH, CS and mitochondrial respiratory chain complex III were all significantly affected by lipid sources. Furthermore, an interaction between dietary carbohydrate levels and lipid sources was also observed in the activities of liver mitochondrial Na+-K+-ATPase and respiratory chain complex III as well as the transcripts of ATP6 and PGC-1α. Overall, these findings suggested that dietary carbohydrate levels and lipid sources remarkably affected the mitochondrial biosynthesis and function of M. amblycephala. A diet containing 30% carbohydrate and FO could boost its mitochondrial biosynthesis, while that of 30% carbohydrate and SO could enhance the mitochondrial function.

Evaluation Of Zinc-Bearing Palygorskite Effects on Growth Performance, Nutrient Retention, Meat Quality, and Zinc Accumulation in Blunt Snout Bream Megalobrama Amblycephala

AbstractZinc (Zn) is widely known as an essential trace element for fish and new ways to supply it to them are needed. Palygorskite (Pal) is a natural silicate clay mineral and the palygorskite structure contains nano-channels, which are filled with water and exchangeable ions. Zn-bearing palygorskites (Zn-Pal) prepared using ion exchange have attracted attention due to the durable antibacterial properties that limit pathogens and as a potential new Zn source for livestock. The present study was conducted to evaluate the effects of Zn-Pal supplementation on the growth performance, nutrient retention, meat quality, Zn accumulation, and intestinal Zn transporter protein gene expression in blunt snout bream Megalobrama amblycephala. The fish were fed a basal diet without an exogenous Zn source and the basal diet was supplemented with 125 mg/kg Zn as Zn sulfate (ZnSO4) or 35, 80, or 125 mg/kg Zn as Zn-Pal. Each diet was tested using three replicates for 7 weeks. The results showed that dietary Zn-Pal supplementation quadratically (P&lt;0.05) increased growth performance, nutrient retention, total and Cu/Zn superoxide dismutase activity, Zn content in scales, and intestinal Zn transporter protein gene expression. The muscular cooking loss in blunt snout bream decreased with the optimum Zn-Pal Zn level of 35 mg/kg. Compared to the fish treated with ZnSO4, the fish supplemented with 35 mg/kg as Zn-Pal exhibited similar growth performance and nutrient retention (P&gt;0.05), increased mRNA expression of the metal-response element-binding transcription factor-1 in the intestine (P&lt;0.05), and decreased cooking loss of muscle (P&lt;0.05).The results suggested that 35 mg/kg Zn supplementation as Zn-Pal could improve the growth performance and body composition, increase nutrient retention and tissue Zn concentrations, enhance the muscle water-holding capacity, and enhance antioxidant status in blunt snout bream. The Zn-Pal was more efficient and could be used as an alternative Zn source to ZnSO4 in the diet of blunt snout bream.

Lysine supplement benefits the growth performance, protein synthesis, and muscle development of Megalobrama amblycephala fed diets with fish meal replaced by rice protein concentrate.

This study aimed to investigate the effects of lysine supplement on the growth performance of blunt snout bream Megalobrama amblycephala fed diets with fish meal (FM) replaced by rice protein concentrate (RPC) with the potential mechanisms characterized. Fish were fed three diets, including the FM diet (containing FM), the RPC diet (FM replaced by RPC), and the MRPC diet (the RPC diet supplemented with lysine) for 8weeks. Weight gain, protein efficiency ratio, and nitrogen and energy utilization of fish fed the FM diet were all significantly higher than those of the RPC treatment, but they showed no statistical difference with those of the MRPC group. Fish fed the RPC diet showed shorter villi length of the distal intestine than that of the other treatments. No significance was found in whole-body composition and intestinal and hepatic cell proliferation among all the treatments. However, fish fed the RPC diet obtained relatively low transcriptions of growth hormone (GH), GH receptor, insulin-like growth factor-I (IGF-I), target of rapamycin (TOR), ribosomal protein S6 kinase 1, myoblast determination protein, myogenic factor 5, and myostatin a (MSTNa) but high levels of eukaryotic translation initiation factor 4E-binding protein 2 (4E-BP2) than those of the other groups. Furthermore, little difference was found in the transcriptions of 4E-BP2, myogenin, muscle-specific regulatory 4, and MSTNb in muscle. Overall, these results showed that dietary supplement of lysine benefits the growth performance of blunt snout bream fed FM-free diets through the mediation of the GH-IGF-I axis, TOR signaling pathway, myogenic regulatory factors, and MSTN.
