Manganese Induced Alterations in the Blood Pyruvate Levels of a Freshwater Indian Catfish Heteropneustes fossilis

Blood pyruvate levels of a freshwater Indian Catfish, Heteropneustes fossilis, were investigated following sublethal manganese intoxication (3440 mg/l MnSO4·H2O; 0.8 of LC50 96 h). Blood pyruvic acid levels registered a statistically significant elevation at all exposure periods. Maximum elevation of 53.33% was observed at 96 h.

Study of blood pyruvic acid levels in pellagrins with and without neurological manifestations and response to nicotinic acid therapy

In 25 cases of pellagra and 10 healthy controls, the blood pyruvic acid levels in the fasting stage and after 60 and 90 minutes of glucose load were estimated by the technique of Friedmann & Haugen (1943) . The blood pyruvic acid levels after 60 and 90 minutes of glucose load were significantly higher in pellagrins as compared to controls. The following conclusions were drawn from this study: (i) that there is impairment of pyruvic acid metabolism in cases of pellagra, which is more marked in pellagrins with neurological manifestations than in those without; (ii) that after administration of nicotinic acid alone for 15 days the pyruvic acid levels returned to normal, thereby indicating that nicotinic acid deficiency is the cause of deranged pyruvate metabolism; (iii) that there is significant improvement in neurological status after nicotinic acid therapy.

Effects of intravenous injection of xylitol on blood sugar, blood pyruvic acid and plasma insulin levels in the dog.

Xylitol was injected intravenously in nine normal dogs and five alloxanized dogs with diabetes. Dosage of xylitol administered was 0.4 g per kg body weight. Changes of blood sugar, blood pyruvic acid and plasma insulin were as follows: 1. Significant decrease of blood pyruvic acid level was shown in all of the nine normal dogs receiving xylitol, and the lowest level of blood pyruvic acid was found at 5 min after the injection. 2. Remarkable increase of plasma insulin was observed in the normal dogs following xylitol injection, and the highest peak of plasma insulin concentration was at 20 min after the injection. The increase of plasma insulin was much higher than that produced by injection of the equivalent dosage of glucose. 3. Hypoglycemia following administration of xylitol occurred in the normal dogs. The lowest level was shown at 30 min after injection of xylitol. 4. These characteristic responses of blood pyruvic acid, blood sugar and plasma insulin to xylitol were not found in all of the five diabetic dogs. 5. These results suggested that the hypoglycemia produced by xylitol might be caused by the hypersecretion of endogenous insulin from pancreatic beta cells.

CLINICAL STUDIES ON ATP (ADENOSINE TRIPHOSPHATE)

In diabetic patients (41 adults), the effect of ATP injection on the carbohydrate metabolism was compared with insulin action. The results were summarized as follows. 1) By administration of ATP, blood sugar levels in diabetic patients were usually reduced, but these levels in normal subjects were unchanged by ATP administration. 2) In many cases, alimentary hyperglycemia in diabetic patients was controlled by administration of ATP, and hypoglycemic effect of insulin was raised when ATP was injected with insulin. 3) Serum inorganic phosphorus were reduced by administration of ATP and the reducing-ratio of phosphorus by intravenous injection of glucose was intensified by administration of ATP. 4) Blood pyruvic acid levels were raised by insulin injection. On the contrary, blood pyrubic acid levels were reduced by ATP injection, and the elevation of blood pyruvic acid levels by intravenous injection of glucose was also controlled by ATP injection. 5) Blood lactic acid levels were reduced by ATP injection and the elevation of lactic acid levels by intravenous injection of glucose was controlled by ATP.

Thioctic acid content and pyruvate oxidation in ethionine-damaged rat livers.

Terminal blood pyruvic acid level was elevated in rats with liver lesions induced by feeding a diet containing ethionine. Pyruvate oxidation by mitochondria from livers of these rats diminished. The hepatic content of thioctic acid was decreased relative to control animals; the thioctic acid content of liver mitochondria was also reduced. In vitro addition of thioctic acid did not stimulate the diminished hepatic mitochondrial pyruvic acid oxidation. Undiminished urinary excretion of thioctic acid, failure of thioctic acid injection to increase the diminished hepatic mitochondrial thioctic acid content, and normal kidney thioctic acid content suggest that total body thioctic acid synthesis is unimpaired but that hepatic thioctic acid “binding” is impaired, in rats fed a diet containing ethionine.

SummaryA simultaneous measurement has been made of lactic acid and pyruvic acid changes in the blood of neuropsychiatric patients before and after extreme muscular excitaion induced by electroshock therapy (EST). The blood lactic acid level of all patients following SST rose sharply to a peak (six times pre-shock level) within 3 to 5 minutes and then returned to the normal level after 1 to 11/2 hours. The blood pyruvic acid level of all patients rose immediately following EST for thirty seconds (4-6 mg %), then fell to a minimum (2-3 mg %) within 3 to 5 minutes and rose again to a peak three times the pre-shock level 20 min after EST.
