Promiscuous Catalytic Activity of a Binuclear Metallohydrolase: Peptide and Phosphoester Hydrolyses.

In this study, chemical promiscuity of a binuclear metallohydrolase Streptomyces griseus aminopeptidase (SgAP) has been investigated using DFT calculations. SgAP catalyzes two diverse reactions, peptide and phosphoester hydrolyses, using its binuclear (Zn-Zn) core. On the basis of the experimental information, mechanisms of these reactions have been investigated utilizing leucine p-nitro aniline (Leu-pNA) and bis(4-nitrophenyl) phosphate (BNPP) as the substrates. The computed barriers of 16.5 and 16.8 kcal/mol for the most plausible mechanisms proposed by the DFT calculations are in good agreement with the measured values of 13.9 and 18.3 kcal/mol for the Leu-pNA and BNPP hydrolyses, respectively. The former was found to occur through the transfer of two protons, while the latter with only one proton transfer. They are in line with the experimental observations. The cleavage of the peptide bond was the rate-determining process for the Leu-pNA hydrolysis. However, the creation of the nucleophile and its attack on the electrophile phosphorus atom was the rate-determining step for the BNPP hydrolysis. These calculations showed that the chemical nature of the substrate and its binding mode influence the nucleophilicity of the metal bound hydroxyl nucleophile. Additionally, the nucleophilicity was found to be critical for the Leu-pNA hydrolysis, whereas double Lewis acid activation was needed for the BNPP hydrolysis. That could be one of the reasons why peptide hydrolysis can be catalyzed by both mononuclear and binuclear metal cofactors containing hydrolases, while phosphoester hydrolysis is almost exclusively by binuclear metallohydrolases. These results will be helpful in the development of versatile catalysts for chemically distinct hydrolytic reactions.

Fabrication of highly active phosphatase-like fluorescent cerium-doped carbon dots for in situ monitoring the hydrolysis of phosphate diesters.

Ce-Doped carbon dots (CeCDs) were fabricated via a one-step hydrothermal carbonization using Ce(NO3)3·6H2O and EDTA·2H2O as precursors, and various experimental techniques were employed to characterize the morphology, structure and composition of the as-obtained CeCDs. Using the disodium salt of bis(4-nitrophenyl)phosphate (BNPP) as a DNA model substrate, mimetic phosphatase activity toward phosphate ester hydrolysis cleavage was investigated. It was found that similar to the catalytic character of the natural enzyme, CeCDs as a hydrolase mimic can exhibit a good catalytic activity for promoting BNPP hydrolysis, in which metal Ce(iii) acts as a center for binding and activation of the metal-bound hydroxide complex as well as a source of nucleophilic metal hydroxides. Additionally, based on the Inner Filter Effect (IFE), fluorescence spectra can be used to monitor the hydrolysis of BNPP using CeCDs, which mimic phosphatase. Thus, the unique properties of the CeCD mimetic phosphatases as well as the IFE sensing strategy would provide an ideal platform to monitor the catalytic phosphate ester hydrolysis processes. Finally, to validate the availability of the established catalytic systems, the CeCDs were further applied for degradation of organophosphorus pesticide chlorpyrifos. The degradation efficiency was estimated to be a satisfactory value of 74.50%, exhibiting a potential application prospect for treatment of the pollutants in the soil and water.

New Zn(II) complexes based on biologically active substituted acetic acid and nitrogen donor ligands: synthesis, crystal structure and biological applications

The complexes [Zn(phenylacetato)2(2-aminopyridin)2] (3), [Zn(phenylacetato)2(1,10-phenanthroline)]·H2O (4), and [Zn(phenylacetato)2(2,9-dimethyl-1,10-phenanthroline)]·0.5 H2O (5) were prepared and characterized by IR-, UV–Visible, 1H and 13C NMR spectroscopy, and single crystal X-ray diffraction. BNPP hydrolysis of the complexes and their parent nitrogen ligands showed that the hydrolysis rate of bis-(4-nitrophenyl) phosphate (BNPP) was 1.7 × 105 L mol−1 s−1 for 3, 3.1 × 105 L mol−1 s−1 for 4 and 4.3 × 104 L mol−1 s−1 for 5. Antibacterial activities show the effect of complexation on activity against Gram-positive (S. epidermidis, S. aureus, E. faecalis, M. luteus and B. subtilis) and Gram-negative (K. pneumonia, E. coli, P. mirabilis and P. aeruginosa) bacteria using the agar well diffusion method. Complex 4 showed good activity against G− bacteria except P. aeruginosa, and against G+ bacteria except E. ferabis. Complex 5 showed no activity against G− bacteria, low activity against M. luteus and B. subtilis bacteria and high activity against S. epidemidis and S. aureus. Complex 3 did not show any activity against G− or G+ bacteria.

Degradation of bis-p-nitrophenyl phosphate using zero-valent iron nanoparticles

Phosphate esters are employed in some agrochemical formulations and have long life time in the Environment. They are neurotoxic to mammals and it is very difficult to hydrolyze them. It is easy to find papers in the literature dealing with transition metal complexes used in the hydrolysis processes of organophosphorous compounds. However, there are few reports related with degradation of phosphate esters with inorganic nanoparticles. In this work bis-4-nitrophenyl phosphate (BNPP) was used as an agrochemical agent model. The BNPP interaction with zero-valent iron nanoparticles (ZVI NPs), in aqueous media, was searched. The concentration of BNPP was 1000 times higher than the ZVI NPs concentration. The average size of the used iron nanoparticles was 10.2 ± 3.2 nm. The BNPP degradation process was monitored by means of UV-visible method. Initially, the BNPP hydrolysis happens through the P-O bonds breaking-off under the action of the ZVI NPs. Subsequently, the nitro groups were reduced to amine groups. The overall process takes place in 10 minutes. The reaction products were identified employing standard substances in adequate concentrations. The iron by-products were isolated and characterized by X-RD. These iron derivatives were identified as magnetite (Fe3O4) and/or maghemite (γ-Fe2O3) and lepidocrocite (γ-FeOOH). A suggested BNPP degradation mechanism will be discussed.

Effective cleavage of phosphodiester promoted by the zinc(II) and copper(II) inclusion complexes of β-cyclodextrin

To construct the model of metallohydrolase, two inclusion complexes [MLCl2(β-CD)] (1, M=Zn(II); 2, M=Cu(II); L=N,N′-bis(2-pyridylmethyl)amantadine; β-CD=β-cyclodextrin) were synthesized by mixing β-CDs with the pre-synthesized complexes G1, [ZnLCl2] and G2, [CuLCl2]. Structures of G1, G2, 1 and 2 were characterized by X-ray crystallography, respectively. In solution, two chloride anions of G1 and G2 underwent ligand exchange with solvent molecules according to ESI-MS analysis. The chemical equilibrium constants were determined by potentiometric pH titration. The kinetics of bis(4-nitrophenyl) phosphate (BNPP) hydrolysis catalyzed by G1, G2, 1 and 2 were examined at pHs ranging from 7.50 to 10.50 at 308±0.1K. The pH profile of rate constant of BNPP hydrolysis catalyzed by 1 exhibited an exponential increase with the second-order rate constant of 2.68×10−3M−1s−1 assigned to the di-hydroxo species, which was approximately an order of magnitude higher than those of reported mono-Zn(II)-hydroxo species. The high reactivity was presumably hydroxyl-rich microenvironment provided by β-CDs, which might effect in stabilizing either the labile zinc-hydroxo species or the catalytic transition state.

Phosphodiester cleavage by trivalent lanthanides in the presence of native cyclodextrins

Testing phosphodiesterase activity of Eu(III) in the presence of native cyclodextrins revealed capacity of β-cyclodextrin (β-CD) to stabilize catalytically active metal hydroxocomplexes in mildly basic solutions. Kinetics of the hydrolysis of bis(4-nitrophenyl) phosphate (BNPP) and transesterification of 2-hydroxypropyl 4-nitrophenyl phosphate (HPNP) as models of DNA and RNA respectively has been studied with La(III), Pr(III), Nd(III), Eu(III), Gd(III) and Dy(III) cations in the presence of β-CD in the range of pH 7.0–9.0. The overall catalytic effect with 2mM lanthanide–β-CD complexes was up to 105 for HPNP and 108 for BNPP at pH 8 demonstrating the highest catalytic activity among so far reported artificial phosphodiesterases. Analysis of concentration and pH-dependences of observed rate constants for different lanthanides showed that active species are binuclear polyhydroxocomplexes of general type [M2(β-CD)(OH)n]6−n with n=3–5. The metal–β-CD and phosphodiester–β-CD interactions were studied by 1H NMR spectroscopy. Mechanistic implications of much higher catalytic efficiency in BNPP hydrolysis as compared to HPNP transesterification are discussed.

Solution Speciation of the Dinuclear ZrIV‐Substituted Keggin Polyoxometalate [{α‐PW11O39Zr(μ‐OH)(H2O)}2]8– and Its Reactivity towards DNA‐Model Phosphodiester Hydrolysis

AbstractThe solution speciation of the ZrIV‐substituted Keggin polyoxometalate (Et2NH2)8[{α‐PW11O39Zr(μ‐OH)(H2O)}2]·7H2O (ZrK 2:2) was fully determined under different pD, temperature, and concentration conditions. Subsequently, phosphodiester bond hydrolysis of the DNA model substrate bis(4‐nitrophenyl) phosphate (BNPP) promoted by ZrK 2:2 was studied in detail. In the presence of ZrK 2:2, phosphoester bond hydrolysis in BNPP proceeded with a rate constant of kobs = (4.75 ± 0.25) × 10–6 s–1 at pD 6.4 and 60 °C, which represented a 320‐fold rate enhancement relative to the spontaneous hydrolysis of BNPP. The pD dependence of kobs exhibits a bell‐shaped profile, with the fastest rate observed at pD 6.4. An activation energy (Ea) of 60.16 kJ mol–1, enthalpy of activation (ΔH#) of 57.44 kJ mol–1, entropy of activation (ΔS#) of –173.16 J mol–1 K–1, and Gibbs activation energy (ΔG#) of 111.12 kJ mol–1 at 37 °C were calculated. The influence of the concentration of ZrK 2:2 on the reaction rate constant was studied in the concentration range 0.5 to 6.0 mM. The results showed that ZrK 2:2 is able to hydrolyze an excess amount of BNPP, thus demonstrating that ZrK 2:2 acts as a catalyst for phospho(di)ester bond hydrolysis. In addition, the influence of ionic strength and the inhibitor diphenyl phosphate on BNPP hydrolysis were examined.

Preparation of a new metallomicelle catalyst for the hydrolysis of bis(4-nitrophenyl) phosphate

AbstractA new metallomicellar system containing cerium(III), a macrocylic polyamine ligand, and the nonionic surfactant Brij35(polyoxyethylene(23) lauryl ether) was prepared and used as a catalyst in the hydrolysis of bis(4-nitrophenyl) phosphate (BNPP). Catalytic rate of the BNPP hydrolysis was measured kinetically using the UV-VIS spectrophotometric method. The results indicate that the metallomicellar system has relatively high stability and excellent catalytic function in the BNPP hydrolysis; also, the reaction rate of the BNPP catalytic hydrolysis increased by a factor of ca. 1 × 1010 compared to the BNPP spontaneous hydrolysis due to the catalytic effect of the active species and the local concentration effect of the micelles in the metallomicellar system. Experimental results also showed that the mono-hydroxy complex containing the macrocyclic polyamine ligand and cerium(III) is the real active species in the BNPP catalytic hydrolysis, and that the micelles provide a useful catalytic environment for the reaction. On basis of the research results, the reaction mechanism of BNPP catalytic hydrolysis has been proposed.

Rapid hydrolysis of phosphate ester promoted by Ce(iv) conjugating with a β-cyclodextrin monomer and dimer

Two N-donor ligands (L(1) and L(2)) derived from a β-cyclodextrin (βCD) monomer and dimer were employed to mediate the hydrolytic activity and stability of the Ce(IV) ion in aqueous solution. Complexes Ce(IV)-L(1) and Ce(IV)-L(2) were prepared in situ and characterized by means of UV-vis and NMR measurements. Ce(IV)-L(1) catalyzed the hydrolysis of a DNA model, bis(4-nitrophenyl)phosphate (BNPP) with k(cat) = 5.2 × 10(-3) s(-1) (half-life t(1/2) ≈ 2 minutes) under mild conditions, which represented an approximate 130 million-fold acceleration with respect to the spontaneous hydrolysis of BNPP. The dinuclear species, [Ce(2)L(1)(2)(OH)(5)](3+), contributed splendidly to the catalytic efficiency which echoed the active species postulation of [Ce(2)(OH)(7)](+) in the literature. Ce(IV)-L(2) exhibited efficient binding with BNPP giving 1/K(M) = 2.1 × 10(5) M(-1) which exceeded other Ce(IV) species, e.g. [Ce(4)(OH)(15)](+), by 2 orders of magnitude, which highlighted the hydrophobicity effect of βCDs. Such a highly binding affinity leads to the second-order rate constant, k(cat)/K(M) = 2.3 × 10(2) M(-1) s(-1), which probably ranks as the highest in the non-enzymatic cleavage of BNPP under similar conditions. Additionally, Ce(IV)-L(2) showed favorable tolerance to basic aqua owing to the bulky protection of double βCD pendants.

Carboxy and diphosphate ester hydrolysis promoted by di- or tri-nuclear zinc(II) complexes based on β-cyclodextrin

A new ligand (L), 6-mono-(2-(2-hydroxy-3-(hydroxymethyl)-5-methyl benzylamino)-ethylamino)-β-cyclodextrin, based on β-cyclodextrin derivatives with dinucleating units was synthesized and used to prepare a trimetallic bis-ligands zinc complex (Zn 3(L 2−) 2). The esterase activity of the complex was investigated by the hydrolysis of two carboxylic acid esters, bis(4-nitrophenyl)carbonate (BNPC) and 4-nitrophenyl acetate (NA), and a DNA model bis(4-nitrophenyl)phosphate (BNPP) as a phosphate ester. The catalytic rate for BNPC was very high, which was found to be a 5.63 × 10 3-fold rate enhancement over uncatalyzed hydrolysis and 1.62 × 10 2-fold rate enhancement over uncatalyzed hydrolysis for NA hydrolysis at pH = 7.0. For the catalytic hydrolysis of BNPP, the initial first-order rate constant of 0.1 mM catalyst was 5.85 × 10 −8 s −1 at pH = 8.50 and 35 °C, which is a 731-fold acceleration over uncatalyzed hydrolysis. The second rate constant ( k BNPP) was found to be 1.22 × 10 −3 M −1 s −1 at pH = 10.0. According to the potentiometric titration study, the zinc complex exists in a dinuclear single ligand coordinated mode and a trinuclear bis-ligands system at pH ≥ 7.0. The ester hydrolysis activity was attributed to the cooperative interaction of the two metal centers and the hydrophobic cavity of β-cyclodextrin with substrates.

Ester catalytic hydrolysis by a tridentate N,N′,N″-copper bridged cyclodextrin dimer

Abstract A new pyridine bridged cyclodextrin dimer mono-copper complex (CuL) was synthesized and characterized. The hydrolysis of carboxylic acid esters, bis(4-nitrophenyl)carbonate (BNPC) and 4-nitrophenyl acetate (NA), and phosphate ester, a DNA model bis(4-nitrophenyl)phosphate (BNPP), promoted by CuL has been investigated. The resulting hydrolysis rate constants showed that CuL had a very high rate of catalysis for BNPC hydrolysis, yielding a 2.73 × 103-fold rate enhancement over uncatalyzed hydrolysis at pH 7.00, compared to only a 78.2-fold rate enhancement for NA hydrolysis. The initial first-order rate constant of catalytic hydrolysis for BNPP was 1.01 × 10−7 s−1 at pH 8.5, 35 °C and 0.1 mM catalyst concentration, about 1260-fold acceleration over uncatalyzed hydrolysis. The second rate constant kBNPP of BNPP hydrolysis promoted by CuL was found to be 5.94 × 10−4 M−1 s−1.

Ester Hydrolysis by a Cyclodextrin Dimer Catalyst with a Tridentate N,N′,N′′‐Zinc Linking Group

A new beta-cyclodextrin dimer, 2,6-dimethylpyridine-bridged-bis(6-monoammonio-beta-cyclodextrin) (pyridyl BisCD, L), is synthesized. Its zinc complex (ZnL) is prepared, characterized, and applied as a catalyst for diester hydrolysis. The formation constant (log K(ML)=7.31+/-0.04) of the complex and deprotonation constant (pK(a1)=8.14+/-0.03, pK(a2)=9.24+/-0.01) of the coordinated water molecule were determined by a potentiometric pH titration at (25+/-0.1) degrees C, indicating a tridentate N,N',N''-zinc coordination. Hydrolysis kinetics of carboxylic acid esters were determined with bis(4-nitrophenyl)carbonate (BNPC) and 4-nitrophenyl acetate (NA) as the substrates. The resulting hydrolysis rate constants show that ZnL has a very high rate of catalysis for BNPC hydrolysis, yielding an 8.98x10(3)-fold rate enhancement over uncatalyzed hydrolysis at pH 7.00, compared to only a 71.76-fold rate enhancement for NA hydrolysis. Hydrolysis kinetics of phosphate esters catalyzed by ZnL are also investigated using bis(4-nitrophenyl)phosphate (BNPP) and disodium 4-nitrophenyl phosphate (NPP) as the substrates. The initial first-order rate constant of catalytic hydrolysis for BNPP was 1.29x10(-7) s(-1) at pH 8.5, 35 degrees C and 0.1 mM catalyst concentration, about 1600-fold acceleration over uncatalyzed hydrolysis. The pH dependence of the BNPP cleavage in aqueous buffer was shown as a sigmoidal curve with an inflection point around pH 8.25, which is nearly identical to the pK(a) value of the catalyst from the potentiometric titration. The k(BNPP) of BNPP hydrolysis promoted by ZnL is found to be 1.68x10(-3) M(-1) s(-1), higher than that of NPP, and comparatively higher than those promoted by its other tridentate N,N',N''-zinc analogues.

Copper(II) Complexes of N‐Methylated Derivatives of ortho‐ and meta‐Xylyl‐Bridged Bis(1,4,7‐triazacyclononane) Ligands: Synthesis, X‐ray Structure and Reactivity as Artificial Nucleases

AbstractTwo new binucleating ligands, 1,3‐bis[(4,7‐dimethyl‐1,4,7‐triazacyclononan‐1‐yl)methyl]benzene (Lmemx) and 1,2‐bis[(4,7‐dimethyl‐1,4,7‐triazacyclononan‐1‐yl)methyl]benzene(Lmeox), have been prepared from 1,4,7‐triazatricyclo[5.2.1.04,10]decane and applied in the synthesis of the corresponding copper(II) complexes, [Cu2(μ‐AcO)2(Lmemx)](ClO4)2·H2O (Cm) and [Cu2(η2‐AcO)2(Lmeox)](ClO4)2 (Co). The X‐ray crystal structures confirmed the dinuclear nature of the complexes, with pairs of copper(II) centres residing in distorted square‐pyramidal coordination environments. The nitrogen donors on the ligands occupy three facially disposed coordination sites about each copper(II) centre, with two acetate ligands completing the copper(II) coordination spheres. For Cm, both acetate groups bridge the two metal centres, whilst in Co each acetate chelates to a single metal centre. To assess their utility as simple nuclease mimics, the metal complexes were tested for their phosphate ester cleavage ability using the model phosphodiester, bis(4‐nitrophenyl)phosphate (BNPP). It was found that Co exhibited the fastest rate of BNPP hydrolysis (kobsd. = 2.4 × 10–5 s–1), this rate being five times faster than that of the CuII complex of the non‐methylated analogue. Ester Hydrolysis by a Cyclodextrin Dimer Catalyst with a Metallophenanthroline Linking Group

A novel beta-cyclodextrin dimer, 1,10-phenanthroline-2,9-dimethyl-bridged-bis(6-monoammonio-beta-cyclodextrin) (phenBisCD, L), was synthesized. Its zinc complex (ZnL) has been prepared, characterized, and applied as a new catalyst for diester hydrolysis. The formation constant (logK(ML)=9.56+/-0.01) of the complex and deprotonation constant (pK(a)=8.18+/-0.04) of the coordinated water molecule were determined by a potentiometric pH titration at (298+/-0.1) K. Hydrolytic kinetics of carboxylic acid esters were performed with bis(4-nitrophenyl) carbonate (BNPC) and 4-nitrophenyl acetate (NA) as substrates. The obtained hydrolysis rate constants showed that ZnL has a very high rate of catalysis for BNPC hydrolysis, giving a 3.89x10(4)-fold rate enhancement over uncatalyzed hydrolysis at pH 7.01, relative to only a 42-fold rate enhancement for NA hydrolysis. Moreover, the hydrolysis second-order rate constants of both BNPC and NA greatly increases with pH. Hydrolytic kinetics of a phosphate diester catalyzed by ZnL was also investigated by using bis(4-nitrophenyl) phosphate (BNPP) as the substrate. The pH dependence of the BNPP cleavage in aqueous buffer shows a sigmoidal curve with an inflection point around pH 8.11, which was nearly identical to the pK(a) value from the potentiometric titration. The k(cat) of BNPP hydrolysis promoted by ZnL was found to be 9.9x10(-4) M(-1) s(-1), which is comparatively higher than most other reported Zn(II)-based systems. The possible intermediate for the hydrolysis of BNPP, BNPC, and NA catalyzed by ZnL is proposed on the basis of kinetic and thermodynamic analysis.

1,4,7,10-Tetraazacyclododecane Metal Complexes as Potent Promoters of Phosphodiester Hydrolysis under Physiological Conditions

Previously reported mono- and dinuclear Zn(II), Cu(II), and Ni(II) complexes of 1,4,7,10-tetrazacyclododecane ([12]aneN4 or cyclen) with different heterocyclic spacers (triazine, pyridine) of various lengths (bi- and tripyridine) or an azacrown-pendant have been tested for the hydrolysis of bis(4-nitrophenyl)phosphate (BNPP) under physiological conditions (pH 7-9, 25 degrees C). All Zn(II) complexes promote the hydrolysis of BNPP under physiological conditions, while those of Cu(II) and Ni(II) do not have a significant effect on the hydrolysis reaction. The hydrolysis kinetics in buffered solutions (0.05 M Bis/Tris, TRIS, HEPES, or CHES, I=0.1 M, NaCl) at 25 degrees C were determined by the initial slope method (product conversion<5%). Comparison of the second-order pH-independent rate constants (kBNPP, M(-1) s(-1)) for the mononuclear complexes ZnL1, ZnL3, and ZnL6, which are 6.1x10 (-5), 5.1x10(-5), and 5.7x10(-5), respectively, indicate that the heterocyclic moiety improves the rate of hydrolysis up to six times over the parent Zn([12]aneN4) complex (kBNPP=1.1x10(-5) M(-1) s(-1)). The reactive species is the Zn(II)-OH- complex, in which the Zn(II)-bound OH- acts as a nucleophile. For dinuclear complexes Zn2L2, Zn2L4, and Zn2L5, the rate of reaction is defined by the degree of cooperation between the metal centers, which is determined by the spacer length. Zn2L2 and Zn2L4 possessing shorter spacers are able to hydrolyze BNPP 1 to 2 orders of magnitudes faster than Zn2L5. The second-order rate constants k of Zn2L4 and Zn2L2 at pH 7, 8, and 9 are significantly higher than those of previously reported related complexes. The high BNPP hydrolytic activity may be related to pi-stacking and hydrophobic interactions between the aromatic spacer moieties and the substrate. Complexes Zn2L4 and Zn2L2 show hydrolytic activity at pH 7 and 8, which allows for the hydrolysis of activated phosphate esters under physiological conditions.

Simplified Speciation and Improved Phosphodiesterolytic Activity of Hydroxo Complexes of Trivalent Lanthanides in Aqueous DMSO

Potentiometric titrations of La(III), Nd(III), and Eu(III) perchlorates by Me 4N(OH) in 80% vol aq DMSO revealed formation of predominantly mononuclear complexes M(OH)n(3- n) (n = 1, 2, or 3) and a single binuclear complex M2(OH)(5+). Kinetics of the cleavage of two phosphate diesters, bis (4-nitrophenyl) phosphate (BNPP) and 2-hydroxypropyl 4-nitrophenyl phosphate (HPNPP), and a triester, 4-nitrophenyl diethyl phosphate (paraoxon), were studied as a function of metal and Me4N(OH) concentrations in the same medium. Rate of BNPP cleavage is second-order in metal and is proportional to the product of concentrations of M(OH)2(+) and M(OH)3 species. Rate of HPNPP cleavage is proportional to [M(OH)3](3) for La(III) and Nd(III) and to [M(OH)3](2) for Eu(III). Proposed mechanism for BNPP hydrolysis involves formation of M2(OH)5(diester) intermediate followed by intramolecular nucleophilic attack of hydroxide anion on the phosphoryl group of the substrate. Proposed mechanism for HPNPP cleavage involves formation of M3(OH)9(diester)(-) or M2(OH)6(diester)(-) intermediates followed by the general base-assisted intramolecular cyclization of HPNPP. The latter mechanism is supported by observation of the solvent kinetic isotope effect k H/kD = 2.9 for Eu(III) catalyzed HPNPP cleavage. The efficiency of catalysis in 80% DMSO is much higher than in water. The reaction rate observed in the presence of 1 mM metal in neutral solution surpasses the rate of background hydrolysis by a factor of 10(12)-10(13) for BNPP and 10(10) for HPNPP. The increased catalytic activity is attributed principally to the preferable solvation of lanthanide ions by DMSO, which creates an anhydrous microenvironment favorable for reaction in the coordination sphere of the catalyst. The catalytic activity of lanthanides in paraoxon hydrolysis is much lower with the estimated efficiency of catalysis about 10(5) for 1 mM La(III).

Hydrolysis of BNPP Catalyzed by the Crowned Schiff Base Co(II) Complex Containing Benzoaza‐15‐Crown‐5 in Micellar Solution

It has been reported that three aza crowned Schiff base cobalt (II) complexes were synthesized and characterized, and the metallomicelle made up of the cobalt (II) complexes and surfactants(Brij35, CTAB, LSS), as mimic hydrolytic metalloenzyme, was used in catalytic hydrolysis of bis(4‐nitrophenyl) phosphate (BNPP). The analysis of specific absorption spectrums of the hydrolytic reaction systems indicated that key intermediates made up of BNPP and Co (II) complexes are formed in the reaction process of BNPP catalytic hydrolysis. The mechanism of BNPP catalytic hydrolysis proposed is based on the analytic result of specific absorption spectra. Based on the mechanism proposed, a kinetic mathematical model for the calculation of the kinetic parameter of BNPP catalytic hydrolysis has been established. The acid effect of reaction system, structure effect of the complexes, effect of temperature and effect of micelles on the rate of BNPP hydrolysis catalyzed by the complexes have been discussed.

Studies on BNPP Cleavage by Schiff Base Complexes Containing Benzoaza‐15‐Crown‐5 in DHAB Micellar Solution

Two novel benzoaza‐crown Schiff base cobalt (II) and manganese (III) complexes were synthesized and characterized. The hydrolysis of bis(4‐nitrophenyl) phosphate (BNPP) catalyzed by the two complexes was studied in buffer solution containing dihexadecyldimethylammonium bromide (DHAB) at 25°±0.1°C and different pH values. The kinetic mathematical model of BNPP hydrolysis was proposed, and the effects of different reaction conditions on BNPP hydrolysis were discussed. The results indicate that the two complexes (MnLCl and CoL) can efficiently accelerate the catalytic cleavage of BNPP in DHAB micellar solution. The pseudo‐first‐order rate constants (k obsd) of BNPP hydrolysis catalyzed by the metallomicelles of MnLCl/DHAB and CoL/DHAB are 2.32×107 times and 1.45×107 times higher than that of the BNPP spontaneous hydrolysis, respectively. Possible reasons for the huge rate accelerations include the lower critical micelle concentration (cmc) of DHAB and formation of metallomicelles made of complexes and DHAB. Furthermore, the BNPP cleavage catalyzed only by the two complexes was investigated in buffer solution. It was found that the hydrolytic rates of BNPP catalyzed only by the two complexes were about 1% of those catalyzed by MnLCl/DHAB and CoL/DHAB systems at 25°C, pH=7.00, and [BNPP]=2.0×10−4 mol · dm−3.

Rapid Phosphodiester Hydrolysis Catalyzed by Lanthanum(III)

AbstractThe catalytic hydrolysis of bis(4‐nitrophenyl)phosphate (BNPP) by lanthanum(III) ion in the presence of amino‐alcoholic ligands: diethanolamine (DEA) and triethanolamine (TEA), was investigated kinetically at 30 °C. The results indicated that the dinuclear dihydroxo complexes formed by lanthanum(III) ion with aminoalcoholic ligands might be the catalytically active species which catalyze the hydrolysis of BNPP to different extents and the catalytic mechanism was believed to involve the synergism of double Lewis acid activation of the substrate and an intramolecular nucleophilic attack of a bridging oxo ligand.
