Melatonin enhances salt tolerance in sorghum by modulating photosynthetic performance, osmoregulation, antioxidant defense, and ion homeostasis.

Melatonin is a potent antioxidant that can prevent plant damage caused by adverse stresses. It remains unclear whether exogenous melatonin can mitigate the effects of salt stress on seed germination and seedling growth of sorghum (Sorghum bicolor (L.) Moench). The aim of this study was to decipher the protective mechanisms of exogenous melatonin (100 μmol/L) on sorghum seedlings under NaCl-induced salt stress (120 mmol/L). Plant morphological, photosynthetic, and physiological characteristics were analyzed at different timepoints after sowing. Results showed that salt stress inhibited seed germination, seedling growth, and plant biomass accumulation by reducing photosynthetic pigment contents, photosynthetic efficiency, root vigor, and mineral uptake. In contrast, seed priming with melatonin enhanced photosynthetic pigment biosynthesis, photosynthetic efficiency, root vigor, and K+ content under salt stress. Melatonin application additionally enhanced the activities of antioxidant enzymes (superoxide dismutase, catalase, ascorbate peroxidase, and glutathione reductase) and increased the levels of non-enzymatic antioxidants (reduced glutathione, ascorbic acid) in the leaves. These changes were accompanied by increase in the leaf contents of soluble sugars, soluble proteins, and proline, as well as decrease in hydrogen peroxide accumulation, malondialdehyde content, and electrolyte leakage. Our findings indicate that exogenous melatonin can alleviate salt stress-induced damage in sorghum seedlings through multifaceted mechanisms, such as improving photosynthetic performance and root vigor, facilitating ion homeostasis and osmoregulation, and promoting antioxidant defense and reactive oxygen species scavenging.

The influence of spermidine on the build-up of fucoxanthin in Isochrysis sp. Acclimated to varying light intensities

Spermidine is a type of important growth regulator, which involved in the biosynthesis of photosynthetic pigments, and has the function of promoting cell proliferation. In this study, Isochrysis sp. was selected as the research object to explore the effects of spermidine supplementation on the growth of algal cells and fucoxanthin synthesis under different light intensities. The results showed that the cell density (5.40 × 106 cells/mL) of algae were the highest at 11 days under the light intensity of 200 μmol·m−2·s−1 and spermidine content of 150 μM. The contents of diadinoxanthin (1.09 mg/g) and fucoxanthin (6.11 mg/g) were the highest when spermidine was added under low light intensity, and the growth of algal cells and fucoxanthin metabolism were the most significant. In the carotenoid synthesis pathway, PDS (phytoene desaturase) was up-regulated by 1.96 times and VDE (violaxanthin de-epoxidase) was down-regulated by 0.95 times, which may promote fucoxanthin accumulation.

Development of in-line anoxic small-angle X-ray scattering and structural characterization of an oxygen-sensing transcriptional regulator

Oxygen-sensitive metalloenzymes are responsible for many of the most fundamental biochemical processes in nature, from the reduction of dinitrogen in nitrogenase to the biosynthesis of photosynthetic pigments. However, biophysical characterization of such proteins under anoxic conditions can be challenging, especially at noncryogenic temperatures. In this study, we introduce the first in-line anoxic small-angle X-ray scattering (anSAXS) system at a major national synchrotron source, featuring both batch-mode and chromatography-mode capabilities. To demonstrate chromatography-coupled anSAXS, we investigated the oligomeric interconversions of the fumarate and nitrate reduction (FNR) transcription factor, which is responsible for the transcriptional response to changing oxygen conditions in the facultative anaerobe Escherichia coli. Previous work has shown that FNR contains a labile [4Fe-4S] cluster that is degraded when oxygen is present and that this change in cluster composition leads to the dissociation of the DNA-binding dimeric form. Using anSAXS, we provide the first direct structural evidence for the oxygen-induced dissociation of the E.coli FNR dimer and its correlation with cluster composition. We further demonstrate how complex FNR-DNA interactions can be studied by investigating the promoter region of the anaerobic ribonucleotide reductase genes, nrdDG, which contains tandem FNR-binding sites. By coupling size-exclusion chromatography-anSAXS with full-spectrum UV-Vis analysis, we show that the [4Fe-4S] cluster-containing dimeric form of FNR can bind to both sites in the nrdDG promoter region. The development of in-line anSAXS greatly expands the toolbox available for the study of complex metalloproteins and provides a foundation for future expansions.

Metal-Resistant PGPR Strain Azospirillum brasilense EMCC1454 Enhances Growth and Chromium Stress Tolerance of Chickpea (Cicer arietinum L.) by Modulating Redox Potential, Osmolytes, Antioxidants, and Stress-Related Gene Expression.

Heavy metal stress, including from chromium, has detrimental effects on crop growth and yields worldwide. Plant growth-promoting rhizobacteria (PGPR) have demonstrated great efficiency in mitigating these adverse effects. The present study investigated the potential of the PGPR strain Azospirillum brasilense EMCC1454 as a useful bio-inoculant for boosting the growth, performance and chromium stress tolerance of chickpea (Cicer arietinum L.) plants exposed to varying levels of chromium stress (0, 130 and 260 µM K2Cr2O7). The results revealed that A. brasilense EMCC1454 could tolerate chromium stress up to 260 µM and exhibited various plant growth-promoting (PGP) activities, including nitrogen fixation, phosphate solubilization, and generation of siderophore, trehalose, exopolysaccharide, ACC deaminase, indole acetic acid, and hydrolytic enzymes. Chromium stress doses induced the formation of PGP substances and antioxidants in A. brasilense EMCC1454. In addition, plant growth experiments showed that chromium stress significantly inhibited the growth, minerals acquisition, leaf relative water content, biosynthesis of photosynthetic pigments, gas exchange traits, and levels of phenolics and flavonoids of chickpea plants. Contrarily, it increased the concentrations of proline, glycine betaine, soluble sugars, proteins, oxidative stress markers, and enzymatic (CAT, APX, SOD, and POD) and non-enzymatic (ascorbic acid and glutathione) antioxidants in plants. On the other hand, A. brasilense EMCC1454 application alleviated oxidative stress markers and significantly boosted the growth traits, gas exchange characteristics, nutrient acquisition, osmolyte formation, and enzymatic and non-enzymatic antioxidants in chromium-stressed plants. Moreover, this bacterial inoculation upregulated the expression of genes related to stress tolerance (CAT, SOD, APX, CHS, DREB2A, CHI, and PAL). Overall, the current study demonstrated the effectiveness of A. brasilense EMCC1454 in enhancing plant growth and mitigating chromium toxicity impacts on chickpea plants grown under chromium stress circumstances by modulating the antioxidant machinery, photosynthesis, osmolyte production, and stress-related gene expression.

Co-chaperoning of chlorophyll and carotenoid biosynthesis by ORANGE family proteins in plants

Chlorophylls and carotenoids are essential photosynthetic pigments. Plants spatiotemporally coordinate the needs of chlorophylls and carotenoids for optimal photosynthesis and fitness in response to diverse environmental and developmental cues. However, how the biosynthesis pathways of these two pigments are coordinated, particularly at posttranslational level to allow rapid control, remains largely unknown. Here, we report that the highly conserved ORANGE (OR) family proteins coordinate both pathways via posttranslationally mediating the first committed enzyme in each pathway. We demonstrate that OR family proteins physically interact with magnesium chelatase subunit I (CHLI) in chlorophyll biosynthesis pathway in addition to phytoene synthase (PSY) in carotenoid biosynthesis pathway and concurrently stabilize CHLI and PSY enzymes. We show that loss of OR genes hinders both chlorophyll and carotenoid biosynthesis, limits light-harvesting complex assembly, and impairs thylakoid grana stacking in chloroplasts. Overexpression of OR safeguards photosynthetic pigment biosynthesis and enhances thermotolerance in both Arabidopsis and tomato plants. Our findings establish a novel mechanism by which plants coordinate chlorophyll and carotenoid biosynthesis and provide a potential genetic target to generate climate-resilient crops.

IReenCAM: automated imaging system for kinetic analysis of photosynthetic pigment biosynthesis at high spatiotemporal resolution during early deetiolation.

Seedling de-etiolation is one of the key stages of the plant life cycle, characterized by a strong rearrangement of the plant development and metabolism. The conversion of dark accumulated protochlorophyllide to chlorophyll in etioplasts of de-etiolating plants is taking place in order of ns to µs after seedlings illumination, leading to detectable increase of chlorophyll levels in order of minutes after de-etiolation initiation. The highly complex chlorophyll biosynthesis integrates number of regulatory events including light and hormonal signaling, thus making de-etiolation an ideal model to study the underlying molecular mechanisms. Here we introduce the iReenCAM, a novel tool designed for non-invasive fluorescence-based quantitation of early stages of chlorophyll biosynthesis during de-etiolation with high spatial and temporal resolution. iReenCAM comprises customized HW configuration and optimized SW packages, allowing synchronized automated measurement and analysis of the acquired fluorescence image data. Using the system and carefully optimized protocol, we show tight correlation between the iReenCAM monitored fluorescence and HPLC measured chlorophyll accumulation during first 4h of seedling de-etiolation in wild type Arabidopsis and mutants with disturbed chlorophyll biosynthesis. Using the approach, we demonstrate negative effect of exogenously applied cytokinins and ethylene on chlorophyll biosynthesis during early de-etiolation. Accordingly, we identify type-B response regulators, the cytokinin-responsive transcriptional activators ARR1 and ARR12 as negative regulators of early chlorophyll biosynthesis, while contrasting response was observed in case of EIN2 and EIN3, the components of canonical ethylene signaling cascade. Knowing that, we propose the use of iReenCAM as a new phenotyping tool, suitable for quantitative and robust characterization of the highly dynamic response of seedling de-etiolation.

The uppermost monoterpenes improving Cinnamomum camphora thermotolerance by serving signaling functions.

Terpenes serve important functions in enhancing plant thermotolerance. Cinnamomum camphora mainly has eucalyptol (EuL), camphor (CmR), linalool (LnL) and borneol (BeL) chemotypes basing on the uppermost monoterpenes. To reveal the thermotolerance mechanisms of these uppermost monoterpenes (eucalyptol, camphor, linalool, and borneol) in C. camphora, we surveyed the ROS metabolism and photosynthesis in the 4 chemotypes fumigated with the corresponding uppermost monoterpene after fosmidomycin (Fos) inhibiting monoterpene synthesis under high temperature at 38°C (Fos+38°C+monoterpene), and investigated the related gene expression in EuL and CmR. Meanwhile, the thermotolerance differences among the 4 uppermost monoterpenes were analyzed. In contrast to normal temperature (28°C), ROS levels and antioxidant enzyme activities in the 4 chemotypes increased under 38°C, and further increased in the treatment with Fos inhibiting monoterpene synthesis at 38°C (Fos+38°C), which may be caused by the alterations in expression of the genes related with non-enzymatic and enzymatic antioxidant formation according to the analyses in EuL and CmR. Compared with Fos+38°C treatment, Fos+38°C+monoterpene treatments lowered ROS levels and antioxidant enzyme activities for the increased non-enzymatic antioxidant gene expression and decreased enzymatic antioxidant gene expression, respectively. High temperature at 38°C reduced the chlorophyll and carotenoid content as well as photosynthetic abilities, which may result from the declined expression of the genes associated with photosynthetic pigment biosynthesis, light reaction, and carbon fixation. Fos+38°C treatment aggravated the reduction. In contrast to Fos+38°C treatment, Fos+38°C+monoterpene treatments increased photosynthetic pigment content and improved photosynthetic abilities by up-regulating related gene expression. Among the 4 uppermost monoterpenes, camphor showed strong abilities in lowering ROS and maintaining photosynthesis, while eucalyptol showed weak abilities. This was consistent with the recovery effects of the gene expression in the treatments with camphor and eucalyptol fumigation. Therefore, the uppermost monoterpenes can enhance C. camphora thermotolerance as signaling molecules, and may have differences in the signaling functions.

Aquatic Ecological Risk Evaluation of Chiral Triazole Fungicide Prothioconazole and Its Metabolite Prothioconazole-Desthio on Lemna minor

The potential risk posed by the chiral triazole fungicide prothioconazole and its metabolite, prothioconazole-desthio to aquatic ecosystems has attracted attention. At present, the aquatic toxicity of prothioconazole is focused on aquatic animals, and the study of aquatic plants is limited. In this work, the acute toxicity of prothioconazole (PTZ) and its metabolite, prothioconazole-desthio (PTD), to the aquatic plant Lemna minor (L. minor) was evaluated at the enantiomer level. The effects of the prothioconazole and its metabolite enantiomer on the physiological and biochemical indices, including growth rate, photosynthetic pigment content, and antioxidant-defense-enzymes activity, of L. minor were measured to evaluate the potential risk. The results showed that prothioconazole and prothioconazole-desthio possessed obvious stereoselective toxicity to Lemna minor with an LC50 (7 days) of 0.76–5.63 mg/L. The toxicity order was S-PTD > Rac-PTD > S-PTZ > R-PTD > Rac-PTZ > R-PTZ. The S-PTZ, which had the highest toxicity, obviously inhibited the biosynthesis of photosynthetic pigments and the activity of antioxidant-defense enzymes (malondialdehyde, catalase and superoxide dismutase), leading to an increase in MDA content and oxidative damage. The results further confirmed that the metabolism of PTZ in aquatic ecosystems increased its exposure risk, providing data support and a theoretical basis for the risk assessment of PTZ.

ANÁLISE MULTIVARIADA DE GRAVIOLEIRA SOB ESTRESSE SALINO E APLICAÇÃO EXÓGENA DE PERÓXIDO DE HIDROGÊNIO

ABSTRACT The objective of this study was to evaluate, through multivariate data analysis, the effect of exogenous application of hydrogen peroxide on the photosynthetic pigments, gas exchange and growth of soursop seedlings under salt stress. The study was conducted in a greenhouse, at Federal University of Campina Grande - Paraíba. The assay was carried out from May to October 2018. The treatments were distributed in a randomized block design, in a 5 × 5 factorial arrangement, corresponding to five levels of irrigation water electrical conductivity - ECw (0.6-control, 1.2, 1.8, 2.4, and 3.0 dS m-1) and five concentrations of hydrogen peroxide - H2O2 (0, 10, 20, 30, and 40 μM), with two plants per plot and four replicates. Irrigation water salinity from 1.2 dS m-1 negatively affected the biosynthesis of photosynthetic pigments, gas exchange and growth of soursop. Application of hydrogen peroxide at the concentration of 20 μM resulted in attenuation of salt stress effects on the biosynthesis of photosynthetic pigments, gas exchange and growth of soursop. Hydrogen peroxide concentrations above 30 μM intensified the deleterious effect of irrigation water salinity on the photosynthetic pigments, gas exchange and growth of soursop.

Effects of Lead, Copper and Cadmium on Bioaccumulation and Translocation Factors and Biosynthesis of Photosynthetic Pigments in Vicia faba L. (Broad Beans) at Different Stages of Growth

Trace elements in the environmental media contribute to toxicities of different types. Their presence in the arable pedosphere is a human-health risk factor. This study focused on Vicia faba represented by two Tunisian varieties of bean (Mamdouh) and faba bean (Badii). The objective was to analyze the effects of lead, copper and cadmium on their growth, chlorophyll-content and carotenoids-content, as well as the bioaccumulation and translocation factor, at different stages of growth. For each metal, the concentrations the plants were subjected to were 6, 0.3 and 0.03 mg/L of the metal in the compound for lead nitrate, copper nitrate and cadmium acetate, respectively. The analysis was carried out using an atomic absorption spectrophotometer (ICP-MS), encompassing all the parts of the plant. The authors detected a perceptible decrease in the fresh weight of roots and shoots, as well as a drop in the chlorophyll and carotenoid, for all the three heavy metals. Cadmium turned out to be the most toxic of the three metals and copper (which is incidentally an essential micronutrient for plant growth) the least. As far as the bioaccumulation factor was concerned, bean and faba bean exhibited different behaviours, both with regard to the growth stages and the heavy metal absorbed. During the vegetative growth stage, both were accumulators of all the three heavy metals (a translocation factor less than unity). However, in the flowering stage, faba bean was a hyper-accumulator of copper (TF > 1); while the bean plants accumulated a lot of lead in the pods-stage (TF > 1). It is worthwhile to pose new research questions and try to answer them in this study, if legumes are accumulator or hyper accumulator plants in which stage and in where organ accumulate more HMs.

24-Epibrassinolide Simultaneously Stimulates Photosynthetic Machinery and Biomass Accumulation in Tomato Plants under Lead Stress: Essential Contributions Connected to the Antioxidant System and Anatomical Structures

Several toxic pollutants are released into the atmosphere through human activities. Among these pollutants, lead (Pb) is a non-biodegradable element that can cause reduced cell division, impact negatively on the biosynthesis of photosynthetic pigments, and lower biomass accumulation, which can lead to plant death. 24-epibrassinolide (EBR) is a plant growth regulator with broad benefits on physiological functions and biochemical responses, conferring tolerance to plants against several biotic and abiotic stresses. The experiment was randomized with four treatments, two lead concentrations (0 and 200 µM Pb, described as −Pb and +Pb, respectively) and two EBR (0 and 100 nM EBR, described as −EBR and +EBR, respectively). We detected a negative impact of Pb stress in tomato plants; however, the exogenous application of EBR induced protection on leaf anatomy and photosynthetic apparatus, mitigating the Pb impacts on growth. This steroid enhances the root and leaf structures (in root tissue, the epidermis thickness; and in the leaf, palisade parenchyma, and spongy parenchyma), improving the membrane selectivity, light energy absorption, and CO2 fixation. Applying 200 µM Pb and 100 nM EBR caused an increase in superoxide dismutase, catalase, ascorbate peroxidase, and peroxidase activity (by 26%, 18%, 25%, and 20%, respectively). Moreover, the improvements obtained on photosynthetic pigments, electron transport rate, the effective quantum yield of photosystem II photochemistry, and net photosynthetic rate prove the benefits and protection of photosynthetic apparatus, resulting in increased biomass accumulation, with increases of 95%, 115%, 74%, and 92% in leaf, root, stem, and the whole plant, respectively. Taken together, our findings confirm that EBR alleviates the damages provoked by Pb stress in tomatoes.

Light quality as a driver of photosynthetic apparatus development.

Light provides energy for photosynthesis and also acts as an important environmental signal. During their evolution, plants acquired sophisticated sensory systems for light perception and light-dependent regulation of their growth and development in accordance with the local light environment. Under natural conditions, plants adapted by using their light sensors to finely distinguish direct sunlight and dark in the soil, deep grey shade under the upper soil layer or litter, green shade under the canopy and even lateral green reflectance from neighbours. Light perception also allows plants to evaluate in detail the weather, time of day, day length and thus the season. However, in artificial lighting conditions, plants are confronted with fundamentally different lighting conditions. The advent of new light sources - light-emitting diodes (LEDs), which emit narrow-band light - allows growing plants with light of different spectral bands or their combinations. This sets the task of finding out how light of different quality affects the development and functioning of plants, and in particular, their photosynthetic apparatus (PSA), which is one of the basic processes determining plant yield. In this review, we briefly describe how plants perceive environment light signals by their five families of photoreceptors and by the PSA as a particular light sensor, and how they use this information to form their PSA under artificial narrow-band LED-based lighting of different spectral composition. We consider light regulation of the biosynthesis of photosynthetic pigments, photosynthetic complexes and chloroplast ATP synthase function, PSA photoprotection mechanisms, carbon assimilation reactions and stomatal development and function.

Toxic mechanism of two cyanobacterial volatiles β-cyclocitral and β-ionone on the photosynthesis in duckweed by altering gene expression

Volatile organic compounds (VOCs) promote cyanobacteria dominating eutrophicated waters, with aquatic plant decrease and even disappearance. To uncover the toxic mechanism of cyanobacterial VOCs on aquatic plants, we investigated the growth, photosynthetic pigment levels, photosynthetic abilities and related gene expression in duckweed treated with β-cyclocitral and β-ionone, 2 main components in the VOCs. The levels of chlorophylls and carotenoids gradually declined with raising the concentration of the 2 compounds and prolonging the treatment time. Their decline should result from the down-regulation of 8 genes associated with photosynthetic pigment biosynthesis and up-regulation of 2 genes involved in carotenoid degradation. The reduction was also found in the photosystem II (PSII) efficiency and O2 evolution rate, which should result from the lowered photosynthetic pigment levels and down-regulation of 38 genes related with photosynthetic process. The frond numbers, total frond area and fresh weight gradually decreased with raising the 2 compound concentration, which may result from the lowered photosynthetic abilities as well as down-regulated expression of 7 genes associated with growth-promoting hormone biosynthesis and signal transduction. It can be speculated that cyanobacterial VOCs may poison aquatic plants by lowering the photosynthesis and growth through altering related gene expression.

High Nitric Oxide Concentration Inhibits Photosynthetic Pigment Biosynthesis by Promoting the Degradation of Transcription Factor HY5 in Tomato.

Photosynthetic pigments in higher plants, including chlorophyll and carotenoid, are crucial for photosynthesis and photoprotection. Previous studies have shown that nitric oxide (NO) plays a dual role in plant photosynthesis. However, how pigment biosynthesis is suppressed by NO remains unclear. In this study, we generated NO-accumulated gsnor mutants, applied exogenous NO donors, and used a series of methods, including reverse transcription quantitative PCR, immunoblotting, chromatin immunoprecipitation, electrophoretic mobility shift, dual-luciferase, and NO content assays, to explore the regulation of photosynthetic pigment biosynthesis by NO in tomato. We established that both endogenous and exogenous NO inhibited pigment accumulation and photosynthetic capacities. High levels of NO stimulated the degradation of LONG HYPOCOTYL 5 (HY5) protein and further inactivated the transcription of genes encoding protochlorophyllide oxidoreductase C (PORC) and phytoene synthase 2 (PSY2)—two enzymes that catalyze the rate-limiting steps in chlorophyll and carotenoid biosynthesis. Our findings provide a new insight into the mechanism of NO signaling in modulating HY5-mediated photosynthetic pigment biosynthesis at the transcriptional level in tomato plants.

Differential Regulation of an OsIspH1, the Functional 4-Hydroxy-3-Methylbut-2-Enyl Diphosphate Reductase, for Photosynthetic Pigment Biosynthesis in Rice Leaves and Seeds.

The methylerythritol 4-phosphate (MEP) pathway is responsible for providing common precursors for the biosynthesis of diverse plastidial terpenoids, including chlorophylls, carotenoids, and phytohormones, in plants. In rice (Oryza sativa), the last-step genes encoding 4-hydroxy-3-methylbut-2-enyl diphosphate reductase [HDR/isoprenoid synthesis H (IspH)] have been annotated in two genes (OsIspH1 and OsIspH2) in the rice genome. The spatial transcript levels indicated that OsIspH1 is highly expressed in all tissues at different developmental stages, whereas OsIspH2 is barely expressed due to an early stop in exon 1 caused by splicing error. OsIspH1 localized into plastids and osisph1, a T-DNA inserted knockout mutant, showed an albino phenotype, indicating that OsIspH1 is the only functional gene. To elucidate the role of OsIspH1 in the MEP pathway, we created two single (H145P and K407R) and double (H145P/K407R) mutations and performed complementation tests in two hdr mutants, including Escherichia coli DLYT1 strains and osisph1 rice plants. The results showed that every single mutation retained HDR function, but a double mutation lost it, proposing that the complementary relations of two residues might be important for enzyme activity but not each residue. When overexpressed in rice plants, the double-mutated gene, OsIspH1MUT, reduced chlorophyll and carotenoid biosynthesis in the leaves and seeds. It confirmed the crucial role of OsIspH1 in plastidic terpenoid biosynthesis, revealing organ-specific differential regulation of OsIspH1 in rice plants.

Foliar Application of Leaf Extracts of Glycyrrhiza uralensis Increases Growth and Nutritional Value of Chinese Flowering Cabbage Plants under Field Conditions

This study was conducted to explore the effects of ethanolic extracts of leaves of Glycyrrhiza uralensis on the growth and nutritional quality of Chinese flowering cabbage (Brassica rapa subsp. parachinensis) under field conditions. Preliminary greenhouse experiments were carried out with different concentrations of G. uralensis extracts to select the suitable dose for field studies. An extract concentration of 12.5 g/L was selected based on relative growth rate analysis and increase in photosynthetic pigment biosynthesis. Shoot length, shoot fresh weight and dry weight, number of leaves, and marketable value of Chinese flowering cabbage plants were significantly increased in the field trials with foliar application of G. uralensis leaves extracts at a concentration of 12.5 g/L. This biotic elicitor also enhanced the total phenolic and flavonoid contents, with optimal values increasing by 18.76% and 22.43%, respectively, compared with the control under field conditions. The total glucosinolate content was effectively increased (from 11.21 to 15.37 μmol·g−1 DW sinigrin equivalents), particularly 4-methoxyglucobrassicin (from 4.31 to 6.72 μmol·g−1 DW sinigrin equivalents) and glucoalyssin (from 0.14 to 0.19 μmol·g−1 DW sinigrin equivalents) compared with the control plants in field trials. Overall, foliar application of leaf extracts of G. uralensis can markedly increase the growth of Chinese flowering cabbage and enhance its medicinal and nutritional quality in the fields.

A comprehensive study on the exposure of nanoplastics to constructed wetland ecological systems: Macrophyte physiology and microbial enzymology, community composition and metabolic functions

The increasing emissions of nanoplastics inevitably pose a potential threat to constructed wetland (CWs) ecosystems. However, to our knowledge, the studies on the impact of nanoplastics on CWs were mainly for the effect on nitrogen removal, but comprehensive research on wetlands is still lacking. Thus, we revealed the overall effects of polystyrene nanoparticles (PSNPs) on wastewater treatment performance, and separate exploration of macrophytes and microbes. Results showed that although 1 and 10 mg/L PSNPs had little influences on COD removal, they caused serious suppression in TN removal, and 10 mg/L PSNPs negatively affected the NH4+-N and TP removal. These suppressions were due to the accumulation of>97% PSNPs attached to the sand biofilm, which proved that CWs could effectively remove nanoplastics. As for macrophyte physiology, PSNPs significantly affected the activities of superoxide dismutase, catalase, peroxidase, and the content of malondialdehyde in each phase. The major photosynthetic pigment biosynthesis was inhibited with 10 mg/L PSNPs. Furthermore, the soil catalase activities and the relative abundances of main antioxidant enzymes were obviously impacted. Caused by excess nanoplastic-induced ROS, almost all activities of dehydrogenase, phosphatase, ammonia monooxygenase, nitrate reductase were significantly inhibited. As for metagenomics analysis, taxonomy annotation revealed that PSNPs negatively affected the major functional bacteria involved in global geochemical nitrogen, phosphorus and sulfur cycle. Functional annotation analysis revealed that some indispensable metabolism pathways (oxidative phosphorylation, pyruvate metabolism, ABC transporter, two-component system, biofilm formation, etc.) were also inhibited. Additionally, nanoplastics reduced the overall pattern of genes related to nitrogen metabolism and partial genes in glucose degradation. Overall, our results comprehensively indicate that nanoplastic is a potential danger to wetland ecosystems and should receive more attention.

An Arabidopsis nonhost resistance gene, IMPORTIN ALPHA 2 provides immunity against rice sheath blight pathogen, Rhizoctonia solani.

There is neither resistant rice cultivar nor any control measure against Rhizoctonia solani AG-1 IA (RS), causal of sheath blight and a major threat to global rice production. Rice is a host and Arabidopsis is a nonhost with underlying nonhost resistance (NHR) gene which is largely untested. Using approaches of forward genetics and tools, cytology, and molecular biology, we identified homozygous mutants in Arabidopsis, mapped the NHR gene, and functionally characterized it in response to RS. Rss1 was mapped on Ch 4 between JAERI18 and Ch4_9.18 (844.6 Kb) and identified IMPORTIN ALPHA 2 as the candidate RSS1 gene. We found that breach of immunity in rss1 by RS activates defense responses whereas photosynthetic pigment biosynthesis and developmental processes are negatively regulated. In addition, a gradual decrease in PR1 by 3 dpi revealed that RSS1 positively regulated early SA-mediated resistance. Whereas increased expression of PDF1.2 by 3 dpi supported switching to necrotrophy, SA-mediated defense in Col-0 leading to immune response. Enhanced expression of ATG8a in rss1 supported autophagic cell death. IMPA2, IMPA1, and RAN1 function together to provide NHR against RS. These findings demonstrate that IMPA2 provides NHR against RS in Col-0 that evoke SA-mediated early immunity with boulevard for potential biotechnological application.

Engineering purple bacterial carotenoid biosynthesis to study the roles of carotenoids in light-harvesting complexes.

Carotenoids are important photosynthetic pigments that play key roles in light harvesting and energy transfer, photoprotection, and in the folding, assembly, and stabilization of light-harvesting pigment-protein complexes. The genetically tractable purple phototrophic bacteria have been useful for investigating the biosynthesis and function of photosynthetic pigments and cofactors, including carotenoids. Here, we give an overview of the roles of carotenoids in photosynthesis and of their biosynthesis, focusing on the extensively studied purple bacterium Rhodobacter sphaeroides as a model organism. We provide detailed procedures for manipulating carotenoid biosynthesis, and for the preparation and analysis of the light-harvesting and photosynthetic reaction center complexes that bind them. Using appropriate examples from the literature, we discuss how such approaches have enhanced our understanding of the biosynthesis of carotenoids and the photosynthesis-related functions of these fascinating molecules.

Photobleaching and Recovery of Symbiodiniaceae Effrenium voratum SCS01 Reveals Life Form Transformation Under Thermal Stress

Dinoflagellates in the family Symbiodiniaceae contain a number of species and play an important role in the establishment of coral reef ecosystems in oligotrophic marine waters. Effrenium voratum is likely an exclusively free-living and heterotrophic species of Symbiodiniaceae. How this species responds and acclimates to warming is largely unknown. The present study experimentally established the phenotypic landscapes related to the photobleaching and recovery processes of Effrenium voratum SCS01 following thermal stress. We found that thermal stress bleached the plastids of E. voratum SCS01 and caused the cells to become lighter in color. Thereafter, the bleached cells recovered rapidly when they returned to the optimal temperature. The dominant life form of E. voratum SCS01 shifted from mastigote cells to coccoid cells then returned to mastigote cells. Transcriptome analysis revealed that the photobleaching of E. voratum SCS01 was due to increased degradation and decreased biosynthesis of photosynthetic pigments. The thermally induced life form changes were related to the downregulation of genes for cell motility. Our results revealed the mechanism of photobleaching in E. voratum SCS01 and indicated life form transformation as a newly identified survival strategy of Effrenium voratum SCS01 under thermal stress.
