Heavy Metal Remediation Using Phosphate-Solubilizing Fungi: From Bioprocess to Application

Heavy metal pollution has been a major environmental issue in recent years, seriously threatening land, water sources, agriculture, and human health. The remediation of heavy metal pollution has been a continuously vital issue for current research. Bioremediation is an effective and cost-efficient approach to reduce heavy metal toxicity. Phosphate-solubilizing fungi (PSF) have shown promise in heavy metal bioremediation due to their high tolerance and activity levels. However, the full potential of PSF in bioremediation needs further exploration. PSF activity, metabolite production, and environmental conditions can influence their efficiency in remediating heavy metals. These factors play a critical role in the practical application of PSF and necessitate improvement pathways. This article reviews potential strategies to enhance heavy metal remediation using PSF and optimizing bioprocesses and applications.

Tolerance pattern against the heavy metals and pesticides in Escherichia coli isolated from water samples.

In this study water samples were investigated for the presence of heavy metals and pesticide resistance in Escherichia coli (E. coli) collected from district Jhal Magsi. Identification of E. coli was carried out via Polymerase Chain Reaction (PCR) assays. Heavy metals and pesticide resistance were conducted by the Kirby-Bauer disk diffusion method. E. coli was confirmed by using uidA (623bp) and usp (515bp) genes primer. A relatively high resistance rate was observed for Cd and DDT (2, 4 D) with 100% at the concentration of 1600µl/mL. Samples showed the resistance pattern for Cr with 55% and sensitivity was 45% at 800µl/mL. Resistance and sensitive percentage of Pb, Co were observed 61, 63, and 39, 37% at 400µl/mL. Cyanazine resistance and the sensitive percentage were 51 and 49% at 800µg/mL. For Chlorpyrifos and Carbofuran, 54 and 65% resistance and sensitivity 46 and 35% were measured at a lower range of 400µl/mL. Excessive use of heavy metals and pesticide pollution in standing water near agriculture fields contributed to accelerating the abundance of multi-pollutant-resistant E. coli in water that could be useful in the bioremediation of pesticides and heavy metals.

Toxic Effect of Heavy Metal Poisoning on Living Organisms in Water and Health Risk: A Central India Study

The main causes of this environmental catastrophe are pollutants such as iron, zinc, lead, nickel, and chromium that have entered the water system as a result of uncontrolled industrial effluent discharge in the area. This study finds resistant bacteria and shows the toxicity of heavy metals in industrial effluent near the steel industry. Five separate locations were used for water sampling, and 16s RNA sequencing of the microorganisms that were found was done along with physicochemical parameter analysis and atomic absorption spectrophotometer use. Different water samples’ conductivity, TDS, salinity, DO, turbidity, and pH levels indicate changes in the quality of the water. In addition to having higher values for each of these characteristics, Sample D3’s water samples had greater concentrations of heavy metals, suggesting contamination. Aspergillus niger and Aspergillus terreus were identified in all samples. Both Gram-negative and Gram-positive Bacilli’s resistance to heavy metals has been found to be a sign of microbial adaptation to exposure to heavy metals. 16s rRNA sequencing of microorganisms revealed their roles in heavy metal bioremediation processes. The delicate balance between the flora and fauna in the affected water bodies has been upset by the devastating losses in aquatic life. In this study, we presented the biological methods for eliminating heavy metals, which comprised both metabolically dependent and independent techniques.

The molecular insights of cyanobacterial bioremediations of heavy metals: the current and the future challenges.

In recent years, overexplorations of ore and the growth of industries are the prime factors in the release of heavy metals in environments. As a result, the food crops and water bodies are contaminated with metals which may have several adverse effects on the health of humans and other living species. These metals and metalloids, such as Zn, Cu, Mn, Ni, Cr, Pb, Cd, and As, upset the biochemical pathways of metabolite synthesis in living organisms and contribute to the etiology of different diseases. Microorganisms include bacteria, archaea, viruses, and many unicellular eukaryotes, which can span three domains of life-Archaea, Bacteria, and Eukarya-and some microorganisms, such as cyanobacteria, have shown high efficiency in the biosorption rate of heavy metals. Cyanobacteria are suitable for bioremediation as they can grow in adverse environments, have a less negative impact on the surrounding environment, and are relatively cheaper to manage. The structure of cyanobacteria has shown no extensive internal-bound membranes, so it can directly employ the physiological mechanisms to uptake heavy metals from contamination sites. Such biochemical makeups are suitable for managing and bioremediating heavy metal concentrations in polluted environments. This review aims to explore the potential of cyanobacteria in the bioremediation of heavy metals and metalloids in water bodies. Additionally, we have identified the prospects for enhancing bioremediation effectiveness.

Effects of phosphate-solubilizing bacteria Aspergillus flavus AF-LRH1 on promoting phosphorus solubilization, wheat growth and soil heavy metal remediation

Increasing nutrient supply level and eliminating pollution risk are essential to promote farmland soil use. In the soil-plant nutrition cycle, phosphorus is a limiting nutrient, toxic metals such as Cd and Pb are stress threats. Utilizing microorganisms as a bio-amendment could be a viable approach to enhance the cycling of soil phosphorus nutrients and immobilize Cd and Pb in the soil. This study involved the isolation of Aspergillus flavus AF-LRH1, a phosphate solubilizing fungal strain from soil. The strain exhibited the capacity to solubilize calcium phosphate, attaining a phosphorus concentration of 220.73 mg L−1 in the medium. High performance liquid chromatography analysis showed that AF-LRH1 produced a combination of organic acids (tartaric acid 260.7 mg L−1, fumaric acid 0.8 mg L−1, citric acid 911.9 mg L−1) to dissolve Ca3(PO4)2 by providing H+ ions. AF-LRH1 also utilizes NH4+ for the biosynthesis of amino acids and releases protons, which facilitate the dissolution of insoluble Ca3(PO4)2 and calcium phytate. Through these processes, calcium phosphate is partly converted into hydroxyapatite because of biological solubilization. AF-LRH1 strain showed the capability to recover phosphorus from various types of sludge-based solid residue by increasing the solubility of insoluble phosphate compounds for plant nutrient supply. In addition, the combining addition of AF-LRH1 strain and phosphate-containing amendments (calcium phosphate, sludge ash and biochar) benefited the decontamination of Pb and Cd contaminated soil. This finding confirmed the AF-LRH1 could be a promising environmental-friendly biofertilizer suitable for agricultural applications and bioremediation of heavy metal contaminated soil.

Phycoremediation: a path towards heavy metal bioremediation from wastewater

AbstractHeavy metals (HMs) have emerged as a significant and complex water pollution problem globally. These pollutants are particularly concerning due to their teratogenic, mutagenic, poisonous and carcinogenic properties, as well as their non‐biodegradability. Traditional removal techniques often fall short in addressing these issues, leading to a search for more effective solutions. One promising alternative is the phycoremediation process, which employs microalgae to remove HMs from wastewater. This method is not only cost‐effective but also environmentally friendly, offering the additional benefits of nutrient recovery from polluted water and conversion into value‐added products. This review delves into the capabilities of microalgae in remediating HM‐polluted water, examining the various factors and methods that affect the process. Key improvements that can enhance the efficiency of phycoremediation include immobilizing microalgae to increase their stability and longevity, utilizing binary cultures to harness synergistic effects and implementing cellular and genetic modifications to boost HM uptake and resilience. Moreover, the integration of algorithms and artificial intelligence can optimize phycoremediation processes by predicting and controlling environmental conditions, enhancing the precision and effectiveness of HM removal. The combination of these advanced strategies holds promise for overcoming the limitations of conventional methods, positioning phycoremediation as a viable solution for mitigating HM contamination in water bodies. © 2024 The Author(s). Simultaneous removal of malachite green and lead from water by consortium dry-biomasses of Bacillus licheniformis AG3 and Bacillus cereus M116

Synthetic textile dye malachite green (MG) and heavy metals present in industrial wastewater are hazardous to the ecosystem. Bioremediation of dyes and heavy metals using dry-biomasses has advantages over chemical methods. This study screened an acclimatized, heavy metal-resistant, and dye-degrading Gram positive Bacillus licheniformis AG3 strain from the textile wastewater near Kolkata, West Bengal. The EDXRF analysis of this colored wastewater effluent showed 36.33 mg/L lead, significantly higher than the WHO recommendation. Previously, Bag et al. showed bioremediation of synthetic dyes using dry-biomass of Bacillus cereus M116 from an aqueous solution (Bag et al. Arch Microbiol 203(7):3811–3823, 2021). Here, a consortium of dry-biomasses of B.licheniformis AG3 and B. cereus M116 strains (1:1 w/w ratio) was prepared for the simultaneous removal of lead and MG from wastewater. Statistical optimization determines that the pH, initial concentration of contaminants, and dry-biomass concentrations are critical for bioremediation under batch procedures. Further, optimization using the response surface methodology showed that 0.01% consortium dry-biomasses eliminated a maximum of 99.35% MG and 96.01% lead (II) within 6 h. SEM–EDS and FTIR confirmed a strong surface biosorption. Furthermore, a fixed-bed biofilter column of the consortium dry-biomasses was prepared, which was able to remove 98.1% MG and 98.5% lead at the 0.5–1 mL/min flow rate. Together, this study developed a biofilter with a consortium dry biomasses of B. licheniformis AG3 and B. cereus M116 for the simultaneous removal of MG and lead from wastewater.

Bioremediation of heavy metals by an unexplored bacterium, Pseudoxanthomonas mexicana strain GTZY isolated from aerobic-biofilm wastewater system.

We prompted to characterize a wastewater bacterium, Pseudoxanthomonas mexicana GTZY, that efficiently transforms toxic mercury and arsenic, explores its bioremediation capability, and reveals their relevant gene resistance operons. The isolated strain was characterized by its phylogenetic, biochemical, and phenotypic properties. The strain GTZY potentially removed 84.3% of mercury and their mercury volatilization (Hg(II) to Hg(0)) was confirmed using the X-ray film method, and its respective merA gene was PCR amplified. In addition, strain GTZY efficiently removed arsenate (68.5%) and arsenite (63.2%), and showed resistance up to > 175 and > 55mM, respectively. Their genomic annotations disclosed the linkage of Tn2-transposon and int1 in both ends of mer operon (merAPTR). The co-existence of arsP and arsH proteins in its intrinsic ars operon (arsCPRH) was extremely diverse from its ancestral species. We believe that the mercury resistance-conferring mer operon of P. mexicana GTZY presumably derived horizontally from other species in the reactor, while the arsenic resistance-conferring intrinsic ars operon was highly diversified and evolved from its ancestral species. By considering the potential of the strain GTZY to transform heavy metals, this can be used to recover contaminated sites.

Improved microbial-plant soil bioremediation of PAHs and heavy metal through in silico methods

The combined pollution of polycyclic aromatic hydrocarbons (PAHs) and organic cadmium (Cd) in farmland soils, and the field controlling strategy need to be studied urgently. In this study, 5 PAHs, 5 Cd and 11 soil conditioners were selected to explore the co-exposure risk and remediation efficiency. Firstly, a significant combination Fl-alkylalkoxy cadmium was obtained using forward and reverse methods coupling variation coefficient methods (the combined pollution value was 0.173). Secondly, the interaction energy of microbial degradation / plant absorption of Fl under Cd stress, and microbial mineralization / plant absorption of alkylalkoxy cadmium under PAHs stress were characterized using factorial experimental design, molecular docking and molecular dynamics simulation. The combined pollution of alkylalkoxy cadmium and dialkyl cadmium, phenanthrene and Benzo [a] pyrene was significant (synergistic contribution rates were 17.58 % and 19.22 %, respectively). In addition, 6 soil conditioners with significant efficiency were selected to design Taguchi orthogonal experimental schemes, indicating the microbial degradation / mineralization and plant absorption were significantly effective (the maximum increase of remediation efficiency was 93.81 %) under the combinations (i.e., trratone, coumarol, fulvamic acid, potassium fertilizer and others, etc.). Finally, it was found that the soil conditioners affected the hydrophobic groups and forces, and the efficiency was proportional to the highest peak value and minimum distance in the RDF curve. This study identifies the risk characteristics of co-exposure of PAHs and Cd and screens effective soil conditioners, providing theoretical guidance for risk controlling.

Bioremoval of tannins and heavy metals using immobilized tannase and biomass of Aspergillus glaucus

BackgroundThe presence of inorganic pollutants and heavy metals in industrial effluents has become a serious threat and environmental issues. Fungi have a remarkable ability to exclude heavy metals from wastewater through biosorption in eco-friendly way. Tannase plays an important role in bioconversion of tannin, a major constituent of tannery effluent, to gallic acid which has great pharmaceutical applications. Therefore, the aim of the current study was to exploit the potential of tannase from Aspergillus glaucus and fungal biomass waste for the bioremediation of heavy metals and tannin.ResultsTannase from A. glaucus was partially purified 4.8-fold by ammonium sulfate precipitation (80%). The enzyme was optimally active at pH 5.0 and 40 °C and stable at this temperature for 1 h. Tannase showed high stability at different physiological conditions, displayed about 50% of its activity at 60 °C and pH range 5.0–6.0. Immobilization of tannase was carried out using methods such. as entrapment in Na-alginate and covalent binding to chitosan. The effects of Na-alginate concentrations on the beads formation and enzyme immobilization revealed that maximum immobilization efficiency (75%) was obtained with 3% Na-alginate. A potential reusability of the immobilized enzyme was showed through keeping 70% of its relative activity up to the fourth cycle. The best bioconversion efficiency of tannic acid to gallic acid by immobilized tannase was at 40 °C with tannic acid concentration up to 50 g/l. Moreover, bioremediation of heavy metal (Cr3+, Pb2+, Cu2+, Fe3+, and Mn2+) from aqueous solution using A. glaucus biomass waste was achieved with uptake percentage of (37.20, 60.30, 55.27, 79.03 and 21.13 respectively). The biomass was successfully used repeatedly for removing Cr3+ after using desorbing agent (0.1 N HCl) for three cycles.ConclusionThese results shed the light on the potential use of tannase from locally isolated A. glaucus in the bioremediation of industrial tanneries contained heavy metals and tannin.

High acidity organic waste degradation and the potential to bioremediation of heavy metals in soil by an acid-tolerant Serratia sp.

Highly acidic citrus pomace (CP) is a byproduct of Pericarpium Citri Reticulatae production and causes significant environmental damage. In this study, a newly isolated acid-tolerant strain of Serratia sp. JS-043 was used to treat CP and evaluate the effect of reduced acid citrus pomace (RACP) in passivating heavy metals. The results showed that biological treatment could remove 97.56% of citric acid in CP, the organic matter in the soil increased by 202.60% and the catalase activity in the soil increased from 0 to 0.117Ug-1. Adding RACP into soil can increase the stabilization of Cu, Zn, As, Co, and Pb. Specifically, through the metabolism of strain JS-043, RACP was also involved in the stabilization of Zn and Pb, and Residual Fraction in the total pool of these metals increased by 10.73% and 10.54%, respectively. Finally, the genome sequence of Serratia sp. JS-043 was completed, and the genetic basis of its acid-resistant and acid-reducing characteristics was preliminarily revealed. JS-043 also contains many genes encoding proteins associated with heavy metal ion tolerance and transport. These findings suggest that JS-043 may be a high-potential strain to improve the quality of acidic organic wastes that can then be useful for soil bioremediation.

Environmental Importance of Mulberry: A Review

Mulberry is a woody, deciduous tree that is economically important. It is regarded as a distinctive plant on the planet due to its widespread geological distribution across continents, ability to be cultivated in various forms, multiple uses of leaf foliage and positive impact in environmental safety approaches such as ecorestoration of degraded lands, bioremediation of polluted sites, water conservation, soil erosion prevention, and enhancement of air quality through carbon sequestration. Mulberry has a robust root system. Mulberry root systems can significantly improve soil shear strength and anti-erosive capacity. Mulberry plantations are extremely effective in suppressing sand storms and conserving water and soil. The review investigates the role of mulberry trees in carbon sequestration, ecorestoration, soil and water conservation, bioremediation of heavy metals and afforestation.

Impact of biochar produced at different pyrolysis conditions on heavy metal contaminated soil.

This study aimed to assess the effectiveness of urban derived biochars such as Sugarcane bagasse (SB), Brinjal Stem (BS), and Citrus Peel (CP) produced at two different pyrolysis conditions (450 and 600°C for 60min) for soil heavy metal bioremediation potential. An ex-situ study was conducted to remediate single heavy metal-contaminated SoilRite with lead (Pb), copper (Cu), chromium (Cr) and cadmium (Cd), with biochars applied at different rates. Heavy metal status in soilrite was evaluated using various extraction methods (water-soluble, exchangeable, TCLP (Toxicity Characteristic Leaching Procedure), and PBET (Physiologically Based Extraction Tests)) to determine the biochar treatments' efficacy. The findings show that SB biochar at 450-60 are more effective in immobilizing heavy metals in water-soluble (Cd-100% Pb and Cu-70%), exchangeable (Pb:91%, Cd and Cu by 70-80%) and PBET-extracted forms (Cd-91%, Pb-80%, and Cu-75%), whereas biochar derived from BS (84%) and CP (90%) at 600-60 are more effective in immobilizing TCLP-extracted form of Pb and Cu. Urban derived biochars significantly reduced the toxicity of Pb, Cu, and Cd in various extractable forms and can stabilize and convert them into less accessible forms except for Cr. These extraction methods aid in evaluating environmental risks and influencing remediation strategies for soil heavy metal pollution. Urban biochar, as a cost-effective and eco-friendly solution, significantly solves this issue, facilitating sustainable waste management.

Genomic and proteomic analyses reveal the reduction mechanism of hexavalent chromium by the culturing supernatant of strain Pediococcus acidilactici 13-7

This study used lactic acid bacteria with high antioxidative properties to screen for strains capable of reducing hexavalent chromium [Cr (VI)] in their culturing supernatants. The strain Pediococcus acidilactici 13–7 exhibited potent Cr (VI)-reducing capability and remarkable resistance to Cr (VI) even at concentration as high as 24 mM. Comparative genomics analysis revealed a unique gene, ChrR, associated with Cr (VI) reduction in this strain, distinguishing it from four reference strains of P. acidilactici. The proteomic investigation identified proteins linked to the ChrR gene, such as nqo1, frdA, and gshR, indicating significant enrichment in redox-related functions and oxidative phosphorylation pathways. These findings suggest that P. acidilactici 13–7 possesses superior electron transfer capacity compared to other strains, making it more adaptable under highly oxidative conditions by modulating the external environment to mitigate oxidative stress. Collectively, the results demonstrated the potential application of this lactic acid bacterial strain for bioremediation of heavy metals by its ability to reduce Cr (VI), and shed light on the molecular mechanisms underlying Cr (VI) reduction of the strain P. acidilactici 13–7.

Mechanism of Biological Transport and Transformation of Copper, Cadmium, and Zinc in Water by Chlorella

This study investigates the effectiveness of Chlorella vulgaris in treating copper, cadmium, and zinc in aqueous solutions; the aim of this study was to examine the effects of various factors on the adsorption capacity of Chlorella in water. This study explored the intra- and extracellular adsorption and accumulation patterns of copper (Cu(II)), cadmium (Cd(II)), and zinc (Zn(II)), revealing their molecular response mechanisms under the most suitable conditions. The adsorption capacity of Chlorella to Cu(II), Cd(II), and Zn(II) in water was 93.63%, 73.45%, and 85.41%, respectively. The adsorption mechanism for heavy metals is governed by both intracellular and extracellular diffusion, with intracellular absorption serving as a supplement and external uptake predominating. XRD, XPS, FTIR, SEM-EDX, and TEM-EDX analyses showed that there would be the formation of precipitates such as Cu2S, CuS2, CdS, and ZnSO4. The adsorption of Cu(II) involves its simultaneous reduction to Cu(I). Moreover, specific functional groups present on the cellular surface, such as amino, carboxyl, aldehyde, and ether groups, interact with heavy metal ions. In view of its efficient heavy metal adsorption capacity and biosafety, this study recommends Chlorella as a potential biosorbent for the bioremediation and environmental treatment of heavy metal contaminated water in the future.
