Uniaxial Symmetry Breaking in Bacteriorhodopsin at the Thermal Phase Transition of Lipids of Purple Membranes.

The article correlates between symmetry breaking and phase transition. An analogy, extending from physics to biology, is known to exist between these two topics. Bacteriorhodopsin (bR) as a paradigm of membrane proteins has been used as a case study in the present work. The bR, as the sole protein embedded in what is called a purple membrane (PM), has attracted widespread interest in bionanotechnological applications. The lipids of PM have a crucial role in maintaining the crystal lattice of bR inside PM. For this reason, the present work has been concerned with elucidating the thermal phase transition properties of the PM lipids in orthogonal directions. The results indicated that the axial symmetry of bR exhibits considerable changes occurring at the thermal phase transition of lipids. These changes are brought by an anomaly observed in the time course of orthogonal electric responses during the application of thermal fields on PM. The observed anomaly may bear on symmetry breaking in bR occurring at the phase transition of lipids based on such analogy found between symmetry breaking and phase transition. Lipid-protein interactions may underlie the broken axial symmetry of bR at such lipid thermal transition of PM. Accordingly, thermally perturbed axial symmetry of bR may be of biological relevance relying on the essence of the crystal lattice of bR. Most importantly, a question has to be raised in the present study: Can bR, as a helical protein with broken axial symmetry, affect the symmetry breaking of helical light? This may be of potential technical applications based on a recent discovery that bR breaks the symmetry of helical light.

Encapsulation of Nanoparticles with Statistical Copolymers with Different Surface Charges and Analysis of Their Interactions with Proteins and Cells.

Encapsulation with polymers is a well-known strategy to stabilize and functionalize nanomaterials and tune their physicochemical properties. Amphiphilic copolymers are promising in this context, but their structural diversity and complexity also make understanding and predicting their behavior challenging. This is particularly the case in complex media which are relevant for intended applications in medicine and nanobiotechnology. Here, we studied the encapsulation of gold nanoparticles and quantum dots with amphiphilic copolymers differing in their charge and molecular structure. Protein adsorption to the nanoconjugates was studied with fluorescence correlation spectroscopy, and their surface activity was studied with dynamic interfacial tensiometry. Encapsulation of the nanoparticles without affecting their characteristic properties was possible with all tested polymers and provided good stabilization. However, the interaction with proteins and cells significantly depended on structural details. We identified statistical copolymers providing strongly reduced protein adsorption and low unspecific cellular uptake. Interestingly, different zwitterionic amphiphilic copolymers showed substantial differences in their resulting bio-repulsive properties. Among the polymers tested herein, statistical copolymers with sulfobetaine and phosphatidylcholine sidechains performed better than copolymers with carboxylic acid- and dimethylamino-terminated sidechains.

Comprehensive Review of Nanotechnology: Innovations and Multidisciplinary Applications

Nanotechnology, which involves the control of substances at the nanoscale, has emerged as a valuable discipline that holds the capacity to fundamentally transform numerous scientific sectors, such as materials science, healthcare, environmental remediation, and agriculture. This review article explores the uses and advancements of nanotechnology, emphasizing how it might improve crop yield, advance medical treatments, and provide solutions to environmental problems. This also examines revolutionary applications of nanobiotechnology, including CRISPR/Cas9 genome editing, targeted drug delivery systems, cancer therapy, and regenerative medicine. It highlights the potential of these technologies to enhance diagnostic capabilities and pharmaceutical results. It also looks at how nanotechnology is affecting agriculture, with a focus on improvements in food safety, pest and disease control, and crop yield. Additionally, environmental applications are covered, with an emphasis on the use of nanomaterials for sustainable resource management and pollution remediation. The significance of nanotechnology's contributions to green chemistry, its interdisciplinary character, and the challenges and potential for its incorporation into conventional applications are all emphasized in the review. The article provides an overview of the current status and future directions of nanotechnology by discussing the synthesis of nanoparticles, their commercialization, and the challenges surrounding nano-innovations.

Advancements in bionanotechnological applications for climate-smart agriculture and food production

Due to the significant challenges of growing global population and climate change, the agriculture and food industries are facing a continued demand for improving productivity. The integration of nanotechnology in food and agriculture has been envisioned to supply abundant alternative avenues for the crop improvement and food security. To meet these challenges, nanotechnology has made remarkable advancements in agricultural sciences, resulting in numerous beneficial impacts on crop yield and productivity. The key areas of nano-based agriculture aim to enhance food quality, optimized fertilizer doses, reduced agricultural inputs, and improved nutrient uptake by plants from the soil. Additionally, employing minimal hazardous agrochemicals, lowering fertilizer losses, solidification of soil and water quality, and managing nutrient supply efficiently are attributed to the advantages of nanomaterials. In future, the continued efforts and convergence of agriculture and nanotechnology offers promising benefits such as equipping plants with cutting-edge nanotools to combat abiotic (drought, salt, and temperature) and biotic (insects and diseases) stresses, precise and rapid diagnostics, and maximizing utilization of available resources. The ultimate goal in agricultural sciences is to achieve sustainable food production, which forms the backdrop for harnessing the properties of nanomaterials to enhance crop productivity and address widening growing climate challenges. In summary, the extensive applications of nanotechnology in agriculture are anticipated to support climate-smart crop cultivation.

Bacteriophages of Thermophilic 'Bacillus Group' Bacteria-A Systematic Review, 2023 Update.

Bacteriophages associated with thermophiles are gaining increased attention due to their pivotal roles in various biogeochemical and ecological processes, as well as their applications in biotechnology and bionanotechnology. Although thermophages are not suitable for controlling bacterial infections in humans or animals, their individual components, such as enzymes and capsid proteins, can be employed in molecular biology and significantly contribute to the enhancement of human and animal health. Despite their significance, thermophages still remain underrepresented in the known prokaryotic virosphere, primarily due to limited in-depth investigations. However, due to their unique properties, thermophages are currently attracting increasing interest, as evidenced by several newly discovered phages belonging to this group. This review offers an updated compilation of thermophages characterized to date, focusing on species infecting the thermophilic bacilli. Moreover, it presents experimental findings, including novel proteomic data (39 proteins) concerning the model TP-84 bacteriophage, along with the first announcement of 6 recently discovered thermophages infecting Geobacillus thermodenitrificans: PK5.2, PK2.1, NIIg10.1, NIIg2.1, NIIg2.2, and NIIg2.3. This review serves as an update to our previous publication in 2021.

Zinc-Induced Luminescent l-Valine-Based Pseudopeptidic Soft Bioinspired Materials for Precise Tuning of Nanoassembly.

Pseudopeptide-based bioinspired materials are emerging for selective recognition of biologically significant analytes and are applicable in the modern nanoscience field. Therefore, we have developed novel multifunctional C2-symmetric soft pseudopeptides by amino acid l-valine and salicylaldehyde fragments using a series of aliphatic linkers. They are highly selective and sensitive to Zn (II) ions under physiological conditions and reveal significant fluorescence enhancement with the PET mechanism. The molecular self-assembly shows zinc-induced morphological transformation of the rod-shaped assembly into a chain-like morphology. Such a metal-induced hierarchical nano-assembly may have relevance for specific nanobiotechnology applications.

Synthesis of Photoluminescent 2D Self-Assembled Silver Thiolate Nanoclusters for Sensors and Biomolecule Support.

Silver thiolate nanoclusters (Ag NCs) show distinctive optical properties resulting from their hybrid nature, metallic and molecular, exhibiting size-, structure-, and surface-dependent photoluminescence, thus enabling the exploitation of Ag NCs for potential applications in nanobiotechnology, catalysis, and biomedicine. However, tailoring Ag NCs for specific applications requires achieving long-term stability and may involve modifying surface chemistry, fine-tuning ligand composition, or adding functional groups. In this study, we report the synthesis of novel Ag NCs using 2-ethanephenylthiolate (SR) as a ligand, highlight critical points addressing stability, and characterize their optical and structural properties. A preliminary electrical characterization revealed high anisotropy, well suited for potential use in electronics/sensing applications. We also present the synthesis and characterization of Ag NCs using 10-carboxylic 2-ol thiolate (SR'COOH) having a terminal carboxylic group for conjugation with amine-containing molecules. We present a preliminary assessment of its bioconjugation capability using bovine serum albumin as a model protein indicating its prospective application as a biomolecule support.

A genetic circuit on a single DNA molecule as an autonomous dissipative nanodevice

Realizing genetic circuits on single DNA molecules as self-encoded dissipative nanodevices is a major step toward miniaturization of autonomous biological systems. A circuit operating on a single DNA implies that genetically encoded proteins localize during coupled transcription-translation to DNA, but a single-molecule measurement demonstrating this has remained a challenge. Here, we use a genetically encoded fluorescent reporter system with improved temporal resolution and observe the synthesis of individual proteins tethered to a DNA molecule by transient complexes of RNA polymerase, messenger RNA, and ribosome. Against expectations in dilute cell-free conditions where equilibrium considerations favor dispersion, these nascent proteins linger long enough to regulate cascaded reactions on the same DNA. We rationally design a pulsatile genetic circuit by encoding an activator and repressor in feedback on the same DNA molecule. Driven by the local synthesis of only several proteins per hour and gene, the circuit dynamics exhibit enhanced variability between individual DNA molecules, and fluctuations with a broad power spectrum. Our results demonstrate that co-expressional localization, as a nonequilibrium process, facilitates single-DNA genetic circuits as dissipative nanodevices, with implications for nanobiotechnology applications and artificial cell design.

A New Era for Gold Nitrate Nanoparticle Applications

Gold nanoparticles are included in the group of metallic nanoparticles, and their application has become possible recently because of new optical equipment, electronic devices, and probes capable of manufacturing, detecting, and identifying biomolecules of medical interest. Among the range of nanoparticles, one stands out: the gold nanoparticle. It doesn't possess toxic characteristics and can be administered in vivo. In this study, the structural, optical and electronic properties of gold nitrate were investigated using the density functional theory formalism, considering the gradient generalized approximation. Finally, we observe its application in nanobiotechnology, specifically photothermal applications.

ZnO based 0-3D diverse nano-architectures, films and coatings for biomedical applications.

Thin-film nano-architecting is a promising approach that controls the properties of nanoscale surfaces to increase their interdisciplinary applications in a variety of fields. In this context, zinc oxide (ZnO)-based various nano-architectures (0-3D) such as quantum dots, nanorods/nanotubes, nanothin films, tetrapods, nanoflowers, hollow structures, etc. have been extensively researched by the scientific community in the past decade. Owing to its unique surface charge transport properties, optoelectronic properties and reported biomedical applications, ZnO has been considered as one of the most important futuristic bio-nanomaterials. This review is focused on the design/synthesis and engineering of 0-3D nano-architecture ZnO-based thin films and coatings with tunable characteristics for multifunctional biomedical applications. Although ZnO has been extensively researched, ZnO thin films composed of 0-3D nanoarchitectures with promising thin film device bio-nanotechnology applications have rarely been reviewed. The current review focuses on important details about the technologies used to make ZnO-based thin films, as well as the customization of properties related to bioactivities, characterization, and device fabrication for modern biomedical uses that are relevant. It features biosensing, tissue engineering/wound healing, antibacterial, antiviral, and anticancer activity, as well as biomedical diagnosis and therapy with an emphasis on a better understanding of the mechanisms of action. Eventually, key issues, experimental parameters and factors, open challenges, etc. in thin film device fabrications and applications, and future prospects will be discussed, followed by a summary and conclusion.

Bioinspired Amino Acid Based Materials in Bionanotechnology: From Minimalistic Building Blocks and Assembly Mechanism to Applications.

Inspired by natural hierarchical self-assembly of proteins and peptides, amino acids, as the basic building units, have been shown to self-assemble to form highly ordered structures through supramolecular interactions. The fabrication of functional biomaterials comprised of extremely simple biomolecules has gained increasing interest due to the advantages of biocompatibility, easy functionalization, and structural modularity. In particular, amino acid based assemblies have shown attractive physical characteristics for various bionanotechnology applications. Herein, we propose a review paper to summarize the design strategies as well as research advances of amino acid based supramolecular assemblies as smart functional materials. We first briefly introduce bioinspired reductionist design strategies and assembly mechanism for amino acid based molecular assembly materials through noncovalent interactions in condensed states, including self-assembly, metal ion mediated coordination assembly, and coassembly. In the following part, we provide an overview of the properties and functions of amino acid based materials toward applications in nanotechnology and biomedicine. Finally, we give an overview of the remaining challenges and future perspectives on the fabrication of amino acid based supramolecular biomaterials with desired properties. We believe that this review will promote the prosperous development of innovative bioinspired functional materials formed by minimalistic building blocks.

Assembly Requirements for the Construction of Large-Scale Binary Protein Structures.

The precise assembly of multiple biomacromolecules into well-defined structures and materials is of great importance for various biomedical and nanobiotechnological applications. In this study, we investigate the assembly requirements for two-component materials using charged protein nanocages as building blocks. To achieve this, we designed several variants of ferritin nanocages to determine the surface characteristics necessary for the formation of large-scale binary three-dimensional (3D) assemblies. These nanocage variants were employed in protein crystallization experiments and macromolecular crystallography analyses, complemented by computational methods. Through the screening of nanocage variant combinations at various ionic strengths, we identified three essential features for successful assembly: (1) the presence of a favored crystal contact region, (2) the presence of a charged patch not involved in crystal contacts, and (3) sufficient distinctiveness between the nanocages. Surprisingly, the absence of noncrystal contact mediating patches had a detrimental effect on the assemblies, highlighting their unexpected importance. Intriguingly, we observed the formation of not only binary structures but also both negatively and positively charged unitary structures under previously exclusively binary conditions. Overall, our findings will inform future design strategies by providing some design rules, showcasing the utility of supercharging symmetric building blocks in facilitating the assembly of biomacromolecules into large-scale binary 3D assemblies.

Thermally induced Fe2O3 spikes decorated Ag/Fe2O3 nanocomposite fabrication for anti-bacterial and anti-cancer activities

In the context of anti-cellular catalytic permutations, the development of thermally-induced nanotechnology is of great importance. In this study, iron oxides (Fe2O3) decorated silver (Ag) core–shell nanocomposite was prepared using a green thermal decomposition process without using any additional chemicals. The nanocomposite was characterized for its composition, phase interactions, morphology, and stability using spectroscopic, thermogravimetric, and microscopic techniques. The resultant nanocomposite were also investigated against different types of bacteria and cancer cell lines. Both Gram-positive bacteria (Staphylococcus aureus) and Gram-negative bacteria (Klebsiella spp., E. coli, and Pseudomonas) growth was inhibited by the Ag/Fe2O3 nanocomposite. The highest 19 mm zone of inhibition (ZOI) was found for Staphylococcus aureus by the combined effect of Ag and Fe2O3. The antibiofilm efficacy of the prepared nanocomposites showed biofilm destruction of 82.56% Staphylococcus aureus and 51.06% Klebsiella spp. Furthermore, the nanocomposite resulted in 80–90% death of Hela and BHK-21 cells but displayed lower cell toxicity in the case of the Vero cell line. This pathway of nanocomposites preparation with particle surface engineering would open new doors in the fields of nanobiotechnology and nanobiomedical applications.

Phytofabrication, characterization, and evaluation of novel bioinspired selenium–iron (Se–Fe) nanocomposites using Allium sativum extract for bio-potential applications

Abstract Green nano-chemistry is an advanced research route covering eco-friendly fabrication approaches for synthesizing bimetallic nanocomposites (NCs) to enhance their therapeutic properties. The current study aims to phytofabrication, characterization, and bio-potential evaluation of novel selenium–iron (Se–Fe) NCs by utilizing garlic extract. The morphological and physicochemical features of Se–Fe NCs were evaluated by UV–visible spectroscopy, scanning electron microscopy, energy-dispersive X-ray, Fourier transform infrared, X-ray diffraction, and Zeta potential analysis. The findings showed that garlic cloves extract was a promising capping and reducing agent for the formulation of the NC. To explore the antioxidant potential of a bioinspired Se–Fe NC, 2,2-diphenyl-1-picrylhydrazyl and reducing power assays were performed. Furthermore, antioxidant efficacy was confirmed through antimicrobial activities against clinical pathogens. Phytosynthesized Se–Fe NCs (25, 50, 75, and 100 ppm) showed a dose-dependent response. Higher concentrations of Se–Fe NCs impose a more potent antioxidant and antimicrobial potential. The astonishing findings suggest that phytochemicals in Allium sativum extract are useful reducing agents in the formulation of well-defined Se–Fe NCs, and such NCs could act as competitive inhibitors against pathogens. To the extent of our understanding, Se–Fe NC is the first time synthesized and demonstrates the distinctiveness of green chemistry and will give multifunctional applications in nano-biotechnology.

Programming assembly of biomimetic exosomes: An emerging theranostic nanomedicine platform.

Exosomes have emerged as a promising cell-free therapeutic approach. However, challenges in large-scale production, quality control, and heterogeneity must be overcome before they can be used clinically. Biomimetic exosomes containing key components of natural exosomes have been assembled through extrusion, artificial synthesis, and liposome fusion to address these limitations. These exosome-mimetics (EMs) possess similar morphology and function but provide higher yields, faster large-scale production, and similar size compared to conventional exosomes. This article provides an overview of the chemical and biological properties of various synthetic exosome systems, including nanovesicles (NVs), EMs, and hybrid exosomes. We highlight recent advances in the production and applications of nanobiotechnology and discuss the advantages, limitations, and potential clinical applications of programming assembly of exosome mimetics.

Degree-based topological indices of boron nanotubes

In the past two decades, boron nanotubes have received significant attention from researchers and scientists due to their wide-ranging applications in electronics, nanodevices, optical engineering, nanobiotechnology, and cosmetics. These nanotubular structures composed of boron present exceptional electrical and mechanical properties, making them highly potential nanomaterials. In this article, we study the molecular structure of significant classes of boron nanotubes, namely, trihexagonal boron nanotubes, triangular boron nanotubes, and boron-α nanotubes. Furthermore, we calculate various topological indices for these nanotubes, including the augmented Zagreb index, Sombor index, reduced Sombor index, sum-connectivity index, and arithmetic–geometric index. These indices hold substantial importance in assessing the physical, chemical, and biological characteristics of boron nanotubes.

Single-molecule analysis of osmolyte-mediated nanomechanical unfolding behavior of a protein domain

The small organic molecules, known as osmolytes being ubiquitously present in different cell types, affect protein folding, stability and aggregation. However, it is unknown how the osmolytes affect the nanomechanical unfolding behavior of protein domain. Here, we show the osmolyte-dependent mechanical unfolding properties of protein titin immunoglobulin-27 (I27) domain using an atomic force microscopy (AFM)-based single-molecule force spectroscopy. We found that amines and methylamines improved the mechanical stability of I27 domain, whereas polyols had no effect. Interestingly, glycine betaine (GB) or trimethylamine-N-oxide (TMAO) increased the average unfolding force of the protein domain. The kinetic parameters analyzed at single-molecule level reveal that stabilizing effect of osmolytes is due to a decrease in the unfolding rate constant of I27, which was confirmed by molecular dynamics simulations. Our study reveals different effects that diverse osmolytes have on the mechanical properties of the protein, and suggests the potential use of osmolytes in modulating the mechanical stability of proteins required for various nano-biotechnological applications.

QD-based fluorescent nanosensors: Production methods, optoelectronic properties, and recent food applications.

Food quality and safety are crucial public health concerns with global significance. In recent years, a series of fluorescence detection technologies have been widely used in the detection/monitoring of food quality and safety. Due to the advantages of wide detection range, high sensitivity, convenient and fast detection, and strong specificity, quantum dot (QD)-based fluorescent nanosensors have emerged as preferred candidates for food quality and safety analysis. In this comprehensive review, several common types of QD production methods are introduced, including colloidal synthesis, self-assembly, plasma synthesis, viral assembly, electrochemical assembly, and heavy-metal-free synthesis. The optoelectronic properties of QDs are described in detail at the electronic level, and the effect of food matrices on QDs was summarized. Recent advancements in the field of QD-based fluorescent nanosensors for trace level detection and monitoring of volatile components, heavy metal ions, food additives, pesticide residues, veterinary-drug residues, other chemical components, mycotoxins, foodborne pathogens, humidity, and temperature are also thoroughly summarized. Moreover, we discuss the limitations of the QD-based fluorescent nanosensors and present the challenges and future prospects for developing QD-based fluorescent nanosensors. As shown by numerous publications in the field, QD sensors have the advantages of strong anti-interference ability, convenient and quick operation, good linear response, and wide detection range. However, the reported assays are laboratory-focused and have not been industrialized and commercialized. Promising research needs to examine the potential applications of bionanotechnology in QD-based fluorescent nanosensors, and focus on the development of smart packaging films, labeled test strips, and portable kits-based sensors.

Synthetic α-Helical Nanopore Reactor for Chemical Sensing.

The use of nanopores for the single-molecule sensing of folded proteins and biomacromolecules has recently gained attention. Here, we introduce a simplified synthetic α-helical transmembrane pore, pPorA, as a nanoreactor and sensor that exhibits functional versatility comparable to that of engineered protein and DNA nanopores. The pore, built from the assembly of synthetic 40-amino-acid-long peptides, is designed to contain cysteine residues within the lumen and at the pore terminus for site-specific chemical modification probed using single-channel electrical recordings. The reaction of the pore with differently charged activated thiol reagents was studied, wherein positively charged reagents electrophoretically driven into the pore resulted in pore blocking in discrete steps upon covalent bond formation. The asymmetric blockage patterns resulting from cis and trans-side addition of reagents reveal the pore orientation in the lipid membrane. Furthermore, activated PEG thiols covalently blocked the pores over a longer duration in a charge-independent manner, establishing the large diameter and orientation of the formed pores. While the covalent binding of thiol reagents caused a drop in the pore conductance, cationic cyclic octasaccharides produced time-resolved translocation events, confirming the structural flexibility and tunability of the pores. The ability of the pore to accommodate large analytes and the considerable current amplitude variation following bond formation events are promising for developing platforms to resolve multistep chemical reactions at the single-molecule level for applications in synthetic nanobiotechnology.

Silver and gold nanoparticles: Eco-friendly synthesis, antibiofilm, antiviral, and anticancer bioactivities

For the first time in this study, silver nanoparticles (AgNPs) and gold nanoparticles (AuNPs) were green synthesized by the cost-effective and eco-friendly procedure using Cotton seed meal and Fodder yeast extracts. The biosynthesized NPs were characterized by UV–Vis spectroscopy, dynamic light scattering analysis (DLS), transmission electron microscopy (TEM), selected area electron diffraction (SAED), and fourier-transform infrared (FTIR) spectroscopy. Furthermore, the biosynthesized NPs were tested in vitro against biofilm formation by some pathogenic negative bacteria (Escherichia coli, Proteus mirabilis, Klebsiella sp., Salmonella sp., and Pseudomonas aeruginosa) and negative bacteria (staphylococcus aureus) as well as against human denovirus serotype 5 (HAdV-5) and anticancer activity using HepG2 hepatocarcinoma cells. UV–Vis absorption spectra of reaction mixture of AgNPs and AuNPs exhibited maximum absorbance at 440 nm and 540 nm, respectively. This finding was confirmed by DLS measurements that the highest intensity of the AgNPs and AuNPs were 84 nm and 73.9 nm, respectively. FTIR measurements identified some functional groups detected in Cotton seed meal and Fodder yeast extracts that could be responsible for reduction of silver and gold ions to metallic silver and gold. The morphologies and particle size of AgNPs and AuNPs were confirmed by the TEM and SAED pattern analysis. Biosynthesized AgNPs and AuNPs showed good inhibitory effects against biofilms produced by Escherichia coli, Proteus mirabilis, Klebsiella sp., Salmonella sp., Pseudomonas aeruginosa, and Staphylococcus aureus. In addition, they showed anticancer activities against hepatocellular carcinoma (HepG-2) and antiviral activity against human adenovirus serotype 5 infection in vitro. Finally, the results of this study is expected to be extremely helpful to nano-biotechnology, pharmaceutical, and food packing applications through developing antimicrobial and/or an anticancer drugs from ecofriendly and inexpensive nanoparticles with multi-potentiality.
