Tailor-Made Polysaccharides for Biomedical Applications.

Polysaccharides (PSAs) are carbohydrate-based macromolecules widely used in the biomedical field, either in their pure form or in blends/nanocomposites with other materials. The relationship between structure, properties, and functions has inspired scientists to design multifunctional PSAs for various biomedical applications by incorporating unique molecular structures and targeted bulk properties. Multiple strategies, such as conjugation, grafting, cross-linking, and functionalization, have been explored to control their mechanical properties, electrical conductivity, hydrophilicity, degradability, rheological features, and stimuli-responsiveness. For instance, custom-made PSAs are known for their worldwide biomedical applications in tissue engineering, drug/gene delivery, and regenerative medicine. Furthermore, the remarkable advancements in supramolecular engineering and chemistry have paved the way for mission-oriented biomaterial synthesis and the fabrication of customized biomaterials. These materials can synergistically combine the benefits of biology and chemistry to tackle important biomedical questions. Herein, we categorize and summarize PSAs based on their synthesis methods, and explore the main strategies used to customize their chemical structures. We then highlight various properties of PSAs using practical examples. Lastly, we thoroughly describe the biomedical applications of tailor-made PSAs, along with their current existing challenges and potential future directions.

The researcher examines the potential of smart materials as controlled drug release vehicles in tissue engineering, aiming to enable the localized regeneration of injured tissue by delivering precisely dosed drugs at precisely timed intervals.

Tuning the rheological properties of chitosan/alginate hydrogels for tissue engineering application

Biomaterials that mimic biological tissues are in great demand in tissue engineering. Hydrogels are rising as a conducive candidate in tissue engineering on account of their water-holding capacity, mechanical strength, and elasticity. Nevertheless, the development of hydrogels that mimic biological tissues with the desired stability and mechanical strength, remains a notable barrier. In this investigation, chitosan/alginate hydrogels were prepared by tuning the chitosan concentration from 0.5 % to 2.5 %w/v with a fixed 1 %w/v alginate concentration using 0.1 % glutaraldehyde as a gelling agent. The hydrogel system forms through covalent bonding between chitosan and alginate, mediated by aldehyde groups of the glutaraldehyde. The mechanical strength of the hydrogel was elucidated by probing its viscoelastic behavior through rheological analysis. The rheological investigations revealed that the prepared chitosan/alginate hydrogels are profoundly stable with shear-thinning characteristics. The increase in chitosan concentration enhanced the stability and viscoelastic attributes of the hydrogels. The yield stress values ranging from 0.93 kPa to 14.24 kPa indicate the possibility of using these hydrogels in bioadhesives and soft and bone tissue engineering. The complex modulus of the prepared chitosan/alginate hydrogels resembles the shear modulus of liver tissues, osteogenic cells, and articular cartilage, and hence, these hydrogels pose as suitable candidates in liver, bone, and cartilage tissue engineering. The chitosan/alginate hydrogels possess outstanding self-healing ability, along with biocompatibility, stability, and mechanical strength, rendering them highly advantageous for applications in biomedical tissue engineering and bioadhesives.

This critical review explores the application of biodegradable polymer biomaterials in tissue engineering, highlighting their potential to revolutionize regenerative medicine and tissue substitute. Biodegradable polymers, due to their ability to mimic the extracellular matrix, offer a sustainable alternative for the development of tissue scaffolds that degrade at a rate matching new tissue formation. This review systematically covers the evolution, types, and applications of those materials, addressing both natural and synthetic polymers. Special attention is given to the fabrication techniques, along with 3-d bioprinting and nano-fabrication, that allow the introduction of scaffolds tailored for unique tissue engineering packages. The evaluation discusses the contemporary demanding situations, together with the balance among mechanical properties and biodegradability, and the mixing of scaffolds with host tissues. furthermore, it delves into future directions, including the development of hybrid biomaterials and the incorporation of bioactive molecules to enhance tissue regeneration. The advancements in biodegradable polymer biomaterials constitute a massive step in the direction of the development of more effective and personalised processes to tissue engineering.

Cheminformatics-Based Design and Synthesis of Hydroxyapatite/Collagen Nanocomposites for Biomedical Applications.

This paper presents a novel cheminformatics approach for the design and synthesis of hydroxyapatite/collagen nanocomposites, which have potential biomedical applications in tissue engineering, drug delivery, and orthopedic and dental implants. The nanocomposites are synthesized by the co-precipitation method with different ratios of hydroxyapatite and collagen. Their mechanical, biological, and degradation properties are analyzed using various experimental and computational techniques. Attenuated total reflection-Fourier-transform infrared spectroscopy, thermogravimetric analysis, and X-ray diffraction unveil the low crystallinity and nanoscale particle size of hydroxyapatite (22.62 nm) and hydroxyapatite/collagen composites (14.81 nm). These findings are substantiated by scanning electron microscopy with energy-dispersive X-ray spectroscopy, confirming the Ca/P ratio between 1.65 and 1.53 and attesting to the formation of non-stoichiometric apatites in all samples, further validated by molecular simulation. The antimicrobial activity of the nanocomposites is evaluated in vitro against several bacterial and fungal strains, demonstrating their medical potential. Additionally, in silico analyses are performed to predict the absorption, distribution, metabolism, and excretion properties and the bioavailability of the collagen samples. This study paves the way for the development of novel biomaterials using chemoinformatics tools and methods, facilitating the optimization of design and synthesis parameters, as well as the prediction of biological outcomes. Future research directions should encompass the investigation of in vivo biocompatibility and bioactivity of the nanocomposites, while exploring further applications and functionalities of these innovative materials.

SiO2/SiC Nanowire Surfaces as a Candidate Biomaterial for Bone Regeneration

Tissue engineering (TE) and nanomedicine require devices with hydrophilic surfaces to better interact with the biological environment. This work presents a study on the wettability of cubic silicon-carbide-based (SiC) surfaces. We developed four cubic silicon-carbide-based epitaxial layers and three nanowire (NW) substrates. Sample morphologies were analyzed, and their wettabilities were quantified before and after a hydrogen plasma treatment to remove impurities due to growth residues and enhance hydrophilicity. Moreover, sample biocompatibility has been assessed with regard to L929 cells. Our results showed that core–shell nanowires (SiO2/SiC NWs), with and without hydrogen plasma treatment, are the most suitable candidate material for biological applications due to their high wettability that is not influenced by specific treatments. Biological tests underlined the non-toxicity of the developed biomaterials with regard to murine fibroblasts, and the proliferation assay highlighted the efficacy of all the surfaces with regard to murine osteoblasts. In conclusion, SiO2/SiC NWs offer a suitable substrate to develop platforms and membranes useful for biomedical applications in tissue engineering due to their peculiar characteristics.

Halal Perspectives on Collagen-Based Biomaterials in Tissue Engineering Applications

The halal status of biomedical applications has recently been under intense demand due to the utilization of critical ingredients in developing the products. One of the disciplines in biomedical applications, uses various types of biomaterials is tissue engineering. Due to its promising biocompatibility, low antigenicity, and biodegradability, collagen is well known and frequently employed as biomaterials for tissue engineering applications. Most of collagen come from sources including bovine, porcine, marine, and other mammal species. These huge sources end up being one of the most important factors in determining the halal status. Therefore, the purpose of this review is to emphasize collagen's application as biomaterials from the halal standpoint. Additionally, several collagen sources and their status as halal biomaterials for neural, skin, bone, and tissue engineering were covered, with an emphasis on the potential implementation of collagen from halal sources.

Fabrication and Characterization of Electrospun Chitosan/Polylactic Acid (CH/PLA) Nanofiber Scaffolds for Biomedical Application.

The present study demonstrates a strategy for preparing porous composite fibrous materials with superior biocompatibility and antibacterial performance. The findings reveal that the incorporation of PEG into the spinning solutions significantly influences the fiber diameters, morphology, and porous area fraction. The addition of a hydrophilic homopolymer, PEG, into the Ch/PLA spinning solution enhances the hydrophilicity of the resulting materials. The hybrid fibrous materials, comprising Ch modified with PLA and PEG as a co-solvent, along with post-treatment to improve water stability, exhibit a slower rate of degradation (stable, moderate weight loss over 16 weeks) and reduced hydrophobicity (lower contact angle, reaching 21.95 ± 2.17°), rendering them promising for biomedical applications. The antibacterial activity of the membranes is evaluated against Staphylococcus aureus and Escherichia coli, with PEG-containing samples showing a twofold increase in bacterial reduction rate. In vitro cell culture studies demonstrated that PEG-containing materials promote uniform cell attachment, comparable to PEG-free nanofibers. The comprehensive evaluation of these novel materials, which exhibit improved physical, chemical, and biological properties, highlights their potential for biomedical applications in tissue engineering and regenerative medicine.

Comparison of Bovine- and Porcine-Derived Decellularized Biomaterials: Promising Platforms for Tissue Engineering Applications.

Animal-derived xenogeneic biomaterials utilized in different surgeries are promising for various applications in tissue engineering. However, tissue decellularization is necessary to attain a bioactive extracellular matrix (ECM) that can be safely transplanted. The main objective of the present study is to assess the structural integrity, biocompatibility, and potential use of various acellular biomaterials for tissue engineering applications. Hence, a bovine pericardium (BP), porcine pericardium (PP), and porcine tunica vaginalis (PTV) were decellularized using a Trypsin, Triton X (TX), and sodium dodecyl sulfate (SDS) (Trypsin + TX + SDS) protocol. The results reveal effective elimination of the cellular antigens with preservation of the ECM integrity confirmed via staining and electron microscopy. The elasticity of the decellularized PP (DPP) was markedly (p < 0.0001) increased. The tensile strength of DBP, and DPP was not affected after decellularization. All decellularized tissues were biocompatible with persistent growth of the adipose stem cells over 30 days. The staining confirmed cell adherence either to the peripheries of the materials or within their matrices. Moreover, the in vivo investigation confirmed the biocompatibility and degradability of the decellularized scaffolds. Conclusively, Trypsin + TX + SDS is a successful new protocol for tissue decellularization. Moreover, decellularized pericardia and tunica vaginalis are promising scaffolds for the engineering of different tissues with higher potential for the use of DPP in cardiovascular applications and DBP and DPTV in the reconstruction of higher-stress-bearing abdominal walls.

Aminosilane Functionalized Aligned Fiber PCL Scaffolds for Peripheral Nerve Repair.

Silane modification is a simple and cost-effective tool to modify existing biomaterials for tissue engineering applications. Aminosilane layer deposition has previously been shown to control NG108-15 neuronal cell and primary Schwann cell adhesion and differentiation by controlling deposition of ─NH2 groups at the submicron scale across the entirety of a surface by varying silane chain length. This is the first study toreport depositing 11-aminoundecyltriethoxysilane (CL11) onto aligned Polycaprolactone (PCL) scaffolds for peripheral nerve regeneration. Fibers are manufactured via electrospinning and characterized using water contact angle measurements, atomic force microscopy (AFM), and X-ray photoelectron spectroscopy (XPS). Confirmed modified fibers are investigated using in vitro cell culture of NG108-15 neuronal cells and primary Schwann cells to determine cell viability, cell differentiation, and phenotype. CL11-modified fibers significantly support NG108-15 neuronal cell and Schwann cell viability. NG108-15 neuronal cell differentiation maintains Schwann cell phenotype compared to unmodified PCL fiber scaffolds. 3D ex vivo culture of Dorsal root ganglion explants (DRGs) confirms further Schwann cell migration and longer neurite outgrowth from DRG explants cultured on CL11 fiber scaffolds compared to unmodified scaffolds. Thus, a reproducible and cost-effective tool is reported to modify biomaterials with functional amine groups that can significantly improve nerve guidance devices and enhance nerve regeneration.

Synthesis and Characterization of Cellulose Based Injectable Polyurethane Gels

This article describes the synthesis and characterization of cellulose-based injectable polyurethane gels for tissue engineering applications. The gels were prepared by combining a cellulose derivative with a polyurethane precursor and crosslinking agent to form a stable gel under physiological conditions. The resulting gels were evaluated for swelling behavior.&#x0D; The addition of cellulose to polyurethane is a promising approach to improve the properties of polyurethane. Cellulose is an abundant, environmentally friendly and inexpensive polysaccharide polymer widely used in hydrogels, pharmaceuticals and agriculture. The obtained cellulose-based polyurethane composites were characterized in detail using various techniques such as Fourier transform infrared spectroscopy (FTIR), Thermogravimetric Analyzer (TGA), Differential Scanning Calorimeter (DSC), Differential Thermal Analysis (DTA), swelling test measurements and optical microscopy. The results showed that the obtained composites are suitable for the production of injectable insulating materials. The paper also briefly discusses the various uses of polyurethanes in different fields, including furniture manufacturing, medical devices such as hospital beds, catheters, injection-coated devices, wound dressings and surgical dressings, as well as marine, air and land vehicles. In conclusion, this paper provides valuable insights into the development of cellulose-based injectable polyurethane gels for tissue engineering applications and highlights the potential of cellulose as an additive to enhance the properties of polyurethane. The results of this study may contribute to the development of new and improved biomaterials for tissue engineering applications.

Purification of recombinant bacterial collagens containing structural perturbations.

Streptococcus pyogenes-derived recombinant bacterial collagen-like proteins (CLPs) are emerging as a potential biomaterial for biomedical research and applications. Bacterial CLPs form stable triple helices and lack specific interactions with human cell surface receptors, thus enabling the design of novel biomaterials with specific functional attributes. Bacterial collagens have been instrumental in understanding collagen structure and function in normal and pathological conditions. These proteins can be readily produced in E. coli, purified using affinity chromatography, and subsequently isolated after cleavage of the affinity tag. Trypsin is a widely used protease during this purification step since the triple helix structure is resistant to trypsin digestion. However, the introduction of Gly→X mutations or natural interruptions within CLPs can perturb the triple helix structure, making them susceptible to trypsin digestion. Consequently, removing the affinity tag and isolating collagen-like (CL) domains containing mutations is impossible without degradation of the product. We present an alternative method to isolate CL domains containing Gly→X mutations utilizing a TEV protease cleavage site. Protein expression and purification conditions were optimized for designed protein constructs to achieve high yield and purity. Enzymatic digestion assays demonstrated that CL domains from wild-type CLPs could be isolated by digestion with either trypsin or TEV protease. In contrast, CLPs containing Gly→Arg mutations are readily digested by trypsin while digestion with TEV protease cleaved the His6-tag, enabling the isolation of mutant CL domains. The developed method can be adapted to CLPs containing various new biological sequences to develop multifunctional biomaterials for tissue engineering applications.

The Potential of Duplex Stainless Steel Processed by Laser Powder Bed Fusion for Biomedical Applications: A Review

The austenitic stainless steels utilized in the production of osteosynthesis devices are susceptible to crevice corrosion. Several studies have compared the corrosive behavior of austenitic and duplex stainless steels (DSS), both of which are recognized as viable biomaterials for tissue engineering applications. All of the in vitro and in vivo studies on animals and clinical results reported to date indicate that austeno-ferritic duplex stainless steel can be recommended as a suitable alternative to ASTM F138 steel, since it is resistant to crevice corrosion in the human body and presents superior mechanical properties. The use of DSS for biomedical applications is still under discussion, mainly due to the lack of knowledge of its behavior in terms of device heating or induced movement when exposed to magnetic fields, a potentially harmful effect for the human body. As a breakthrough production technology, additive manufacturing (AM) has demonstrated significant benefits for the fabrication of metal devices with patient-specific geometry. Laser powder bed fusion has particularly been used to manufacture DSS-based components. A fine control of the processing conditions allows for an understanding of DSS microstructural evolution, which is essential for selecting processing parameters and estimating performance, including mechanical properties and corrosion resistance. Furthermore, scientific investigation is necessary for determining the relationships among material, process, and magnetic properties, in order to establish the underlying principles and critical responses. The purpose of this review is to highlight the key performances of DSS for biomedical applications and to point out the relevant role of advanced processing technologies such as additive manufacturing.

Recent Advances in Collagen Antimicrobial Biomaterials for Tissue Engineering Applications: A Review.

Biomaterial-based therapies have been receiving attention for treating microbial infections mainly to overcome the increasing number of drug-resistant bacterial strains and off-target impacts of therapeutic agents by conventional strategies. A fibrous, non-soluble protein, collagen, is one of the most studied biopolymers for the development of antimicrobial biomaterials owing to its superior physicochemical, biomechanical, and biological properties. In this study, we reviewed the different approaches used to develop collagen-based antimicrobial devices, such as non-pharmacological, antibiotic, metal oxide, antimicrobial peptide, herbal extract-based, and combination approaches, with a particular focus on preclinical studies that have been published in the last decade.

Graft Copolymers of Polysaccharide: Synthesis Methodology and Biomedical Applications in Tissue Engineering.

A polymer is a macromolecule that has a significant number of repeating units. It is possible to modify the architecture of a polymer via grafting, bridging, mixing, or generating composites. There are several uses for using natural polymers in culinary and medicinal applications. Polymeric materials became appealing because of their low density and ability to incorporate properties of their constituent constituents. High-energy accelerated electrons from the plasma induce chemical bond breaking in the polymeric structure, resulting in the generation of macromolecule radicals and graft copolymerization. Polymer grafting has become an important aspect of the formulation development process. When polymer functional groups are changed, a wide variety of desirable and unwanted properties can be added or removed. It can be concluded from the findings of the literature survey that graft copolymers of polysaccharides have significant biomedical applications including drug delivery and tissue engineering applications.

Bacterial Cellulose-Based Blends and Composites: Versatile Biomaterials for Tissue Engineering Applications.

Cellulose of bacterial origin, known as bacterial cellulose (BC), is one of the most versatile biomaterials that has a huge potential in tissue engineering due to its favourable mechanical properties, high hydrophilicity, crystallinity, and purity. Additional properties such as porous nano-fibrillar 3D structure and a high degree of polymerisation of BC mimic the properties of the native extracellular matrix (ECM), making it an excellent material for the fabrication of composite scaffolds suitable for cell growth and tissue development. Recently, the fabrication of BC-based scaffolds, including composites and blends with nanomaterials, and other biocompatible polymers has received particular attention owing to their desirable properties for tissue engineering. These have proven to be promising advanced materials in hard and soft tissue engineering. This review presents the latest state-of-the-art modified/functionalised BC-based composites and blends as advanced materials in tissue engineering. Their applicability as an ideal biomaterial in targeted tissue repair including bone, cartilage, vascular, skin, nerve, and cardiac tissue has been discussed. Additionally, this review briefly summarises the latest updates on the production strategies and characterisation of BC and its composites and blends. Finally, the challenges in the future development and the direction of future research are also discussed.

Self-Healing Hydrogels: Development, Biomedical Applications, and Challenges.

Polymeric hydrogels have drawn considerable attention as a biomedical material for their unique mechanical and chemical properties, which are very similar to natural tissues. Among the conventional hydrogel materials, self-healing hydrogels (SHH) are showing their promise in biomedical applications in tissue engineering, wound healing, and drug delivery. Additionally, their responses can be controlled via external stimuli (e.g., pH, temperature, pressure, or radiation). Identifying a suitable combination of viscous and elastic materials, lipophilicity and biocompatibility are crucial challenges in the development of SHH. Furthermore, the trade-off relation between the healing performance and the mechanical toughness also limits their real-time applications. Additionally, short-term and long-term effects of many SHH in the in vivo model are yet to be reported. This review will discuss the mechanism of various SHH, their recent advancements, and their challenges in tissue engineering, wound healing, and drug delivery.

Honey: An Advanced Antimicrobial and Wound Healing Biomaterial for Tissue Engineering Applications.

Honey was used in traditional medicine to treat wounds until the advent of modern medicine. The rising global antibiotic resistance has forced the development of novel therapies as alternatives to combat infections. Consequently, honey is experiencing a resurgence in evaluation for antimicrobial and wound healing applications. A range of both Gram-positive and Gram-negative bacteria, including antibiotic-resistant strains and biofilms, are inhibited by honey. Furthermore, susceptibility to antibiotics can be restored when used synergistically with honey. Honey’s antimicrobial activity also includes antifungal and antiviral properties, and in most varieties of honey, its activity is attributed to the enzymatic generation of hydrogen peroxide, a reactive oxygen species. Non-peroxide factors include low water activity, acidity, phenolic content, defensin-1, and methylglyoxal (Leptospermum honeys). Honey has also been widely explored as a tissue-regenerative agent. It can contribute to all stages of wound healing, and thus has been used in direct application and in dressings. The difficulty of the sustained delivery of honey’s active ingredients to the wound site has driven the development of tissue engineering approaches (e.g., electrospinning and hydrogels). This review presents the most in-depth and up-to-date comprehensive overview of honey’s antimicrobial and wound healing properties, commercial and medical uses, and its growing experimental use in tissue-engineered scaffolds.
