The Versatility and Diagnostic Potential of VOC Profiling for Noninfectious Diseases.

A variety of volatile organic compounds (VOCs) are produced and emitted by the human body every day. The identity and concentration of these VOCs reflect an individual's metabolic condition. Information regarding the production and origin of VOCs, however, has yet to be congruent among the scientific community. This review article focuses on the recent investigations of the source and detection of biological VOCs as a potential for noninvasive discrimination between healthy and diseased individuals. Analyzing the changes in the components of VOC profiles could provide information regarding the molecular mechanisms behind disease as well as presenting new approaches for personalized screening and diagnosis. VOC research has prioritized the study of cancer, resulting in many research articles and reviews being written on the topic. This review summarizes the information gained about VOC cancer studies over the past 10 years and looks at how this knowledge correlates with and can be expanded to new and upcoming fields of VOC research, including neurodegenerative and other noninfectious diseases. Recent advances in analytical techniques have allowed for the analysis of VOCs measured in breath, urine, blood, feces, and skin. New diagnostic approaches founded on sensor-based techniques allow for cheaper and quicker results, and we compare their diagnostic dependability with gas chromatography- and mass spectrometry-based techniques. The future of VOC analysis as a clinical practice and the challenges associated with this transition are also discussed and future research priorities are summarized.

The Potential for Urban Vegetation to Mitigate Ambient Air Pollution Threats to Public Health


 
 
 This paper assesses the threat that ambient air pollution poses to urban public health and the potential role of urban vegetation to mitigate those threats. Air pollution is a major global risk to health, especially in urban areas. In this paper, four major air pollutants were assessed: particulate matter, tropospheric ozone, nitrogen dioxide, and sulfur dioxide. These pollutants were found to have several adverse effects, including increasing mortality and respiratory morbidity. These pollutants come from a variety of sources, but a major contributor in urban areas is the burning of fossil fuels in automobiles. The adverse health effects of pollution are expected to grow as climate change worsens air quality. Research and case studies find that urban vegetation can filter air and remove pollutants through deposition and stomatal uptake. The effectiveness of air pollution removal is dependent upon specific variables, including leaf characteristics, type of vegetation, and seasons. Urban vegetation may worsen air quality in some cases due to slowing ventilation and producing biological volatile organic compounds. While urban vegetation has potential to mitigate ambient air pollution, conducting site specific research is needed when implementing greenspace policies.
 
 

The Potential Benefits of Therapeutic Treatment Using Gaseous Terpenes at Ambient Low Levels

Biological volatile organic compounds (BVOCs) are emitted abundantly from the flora. Among BVOCs, monoterpenes (MTs) have been used for aromatherapy (e.g., forest bathing) to treat human physiological and psychological conditions. However, relatively little is known about the therapeutic effects of MTs at low part per billion by volume (ppbv) levels. The effects of artificial MTs on human subjects were assessed by both olfactory and therapeutic parameters (brain alpha waves and stress index). Gaseous standards of three monoterpenes (i.e., α-pinene, β-pinene, and d-limonene) prepared at low ppbv levels were used individually and as mixtures. Fifty-nine healthy and non-asthmatics volunteer university students were selected for the test. All human subjects inhaled low ppbv levels of monoterpene in the testing room. Brain alpha waves and stress index were investigated during the inhalation time. Questionnaires were also used after testing. It was found that the detection threshold of MTs was close to 5 ppbv. When the MT levels increased from 0 to 20 ppbv, the mean values of brain alpha waves derived from all participants increased from 9.8 to 15.1. In contrast, the stress index values declined from 46.2 to 34.7. The overall results suggest that MTs have great potential to positively affect the relaxation state of subjects in a manner similar to forest bathing in terms of short-term effect. They can thus be applied as potential therapeutic media for mental health care.

Influence of physiological and environmental factors on the diurnal variation in emissions of biogenic volatile compounds from Pinus tabuliformis

Biological volatile organic compounds (BVOCs) have a large influence on atmospheric environmental quality, climate change and the carbon cycle. This study assesses the composition and diurnal variation in emission rates of BVOCs from Pinus tabuliformis, using an enclosure technique. Environmental parameters (temperature and light intensity) and physiological parameters (net photosynthetic rate, Pn; stomatal conductance, gs; intercellular CO2 concentration, Ci; and transpiration rate, Tr) that may affect emission behavior were continuously monitored. The 10 most abundant compound groups emitted by P. tabuliformis were classified by gas chromatography–mass spectrometry. The dominant monoterpenoid compounds emitted were α-pinene, β-myrcene, α-farnesene and limonene. The diurnal emission rate of BVOCs changed with temperature and light intensity, with dynamic analysis of BVOCs emissions revealing that their emission rates were more affected by temperature than light. The variation in monoterpene emission rates was consistent with estimates of Pn, gs and Tr. Basal emission rates (at 30 °C,) of the main BVOCs ranged from 0.006 to 0.273 μg −1/(hr g), while the basal ER standardization coefficients ranged from 0.049 to 0.144 °C−1. Overall, these results provide a detailed reference for the effective selection and configuration of tree species to effectively prevent and control atmospheric pollution.

Fast online two-dimensional analysis of monoterpenes using multi-capillary column high-pressure photoionization time-of-flight mass spectrometry

One of the most abundant biological volatile organic compounds (BVOCs) in the atmosphere, monoterpene, is characterized by its short lifetime, low concentration, fast temporal and spatial variations, and wide variety of isomers. In this study, a multi-capillary column (MCC) was combined with high-pressure photoionization time-of-flight mass spectrometry (HPPI-TOF MS) and employed to develop an MCC-HPPI-TOF MS combination instrument as an online two-dimensional gas chromatography-mass spectrometry (GC-MS) method for the rapid qualitative and quantitative analysis of monoterpene isomers. As a result, six monoterpene isomers, α -pinene, β -pinene, α -terpinene, γ -terpinene, 3-carene, and limonene, were successfully isolated in 180 s with limits of detection (LODs) as low as 6 μg/m3 without sample pre-enrichment. This method was successfully applied to the rapid online analysis of monoterpenes released from the branches and leaves of Cedrus atlantica and Sabina chinensis, which shows the capability and potential application of the method for the online detection of complex sample mixtures in environmental monitoring, process analysis, and other fields.

Canine Detection of the Volatilome: A Review of Implications for Pathogen and Disease Detection.

The volatilome is the entire set of volatile organic compounds (VOC) produced by an organism. The accumulation of VOC inside and outside of the body reflects the unique metabolic state of an organism. Scientists are developing technologies to non-invasively detect VOC for the purposes of medical diagnosis, therapeutic monitoring, disease outbreak containment, and disease prevention. Detection dogs are proven to be a valuable real-time mobile detection technology for the detection of VOC related to explosives, narcotics, humans, and many other targets of interests. Little is known about what dogs are detecting when searching for biological targets. It is important to understand where biological VOC originates and how dogs might be able to detect biological targets. This review paper discusses the recent scientific literature involving VOC analysis and postulates potential biological targets for canine detection. Dogs have shown their ability to detect pathogen and disease-specific VOC. Future research will determine if dogs can be employed operationally in hospitals, on borders, in underserved areas, on farms, and in other operational environments to give real-time feedback on the presence of a biological target.

Progress on the analytical methodology for biological volatile organic compounds

Biological terminal metabolites are volatile organic compounds (VOCs) with strong volatility, containing important bio-information related with biological metabolism. As a crucial research precondition, analytical methodology for biological VOCs has attracted much attention to achieve the adequate composition information of biological VOCs at different physiological status and metabolism phases. In this article, the analytical methodology for biological VOCs is summarized, focusing on the latest advances on sampling and analytical techniques followed by the preliminary application of distilling potential, but crucial, bio-information. Systematic study of the relationship among biological VOCs, biological VOC characteristics and corresponding biomarkers would result in a potential but promising research field, bioodoromics. The concept of bioodoromics is described, and the developing trend of bioodoromics in the future is proposed in this paper. It is believed that bioodoromics possesses the power to describe potential but crucial bio-information related with metabolism pathway. The study of biological VOCs and potential bioodoromics would facilitate relevant fields such as insect prevention, disease diagnosis, criminal track-down, agricultural product quality control and food safety, etc..

Preparation of polypyrrole composite solid-phase microextraction fiber coatings by sol–gel technique for the trace analysis of polar biological volatile organic compounds

Two novel polypyrrole (PPy) composite solid-phase microextraction (SPME) fiber coatings involving polypyrrole β-naphthalenesulfonic acid (PPy/β-NSA) and polypyrrole graphene (PPy/GR) composite SPME fiber coatings were prepared by a simple sol-gel technique for selectively sampling relatively polar biological volatile organic compounds (VOCs). Crucial preparation conditions of the PPy composite SPME fiber coatings were optimized and are discussed in detail. Physical tests suggested that the PPy composite SPME fiber coatings possessed a porous surface morphology, stable chemical and thermal properties. Due to the inducing polar functional groups in the PPy molecule, the PPy composite SPME fiber coatings achieved a higher extraction capacity and special selectivity for the polar biological VOCs with conjugate structures, compared with commercial SPME fiber coatings. Enrichment factors of most of the VOCs by the PPy/β-NSA and PPy/GR SPME fibers were much higher than those achieved by common commercially available SPME fiber coatings. Finally, the PPy/β-NSA and PPy/GR SPME fiber coatings were applied for the trace analysis of typical polar VOCs from ant and coriander samples coupled with gas chromatography/mass spectrometry (GC/MS) detection, respectively. It was satisfactory that the average contents of 4-heptanone, 4-heptanol, 4-nonanone and methyl 5-methylsalicylate from ant samples were actually found to be 28.0, 58.7, 3.0 and 0.6 μgg(-1), and the average contents of nonane, decanal, undecanal and dodecanal from coriander samples were actually found to be 0.79, 0.13, 0.06 and 0.21 μgg(-1). The results suggested that PPy composite SPME coatings will be a potentially excellent sampling technique for the trace analysis of polar biological VOCs.

A disjunct eddy accumulation system for the measurement of BVOC fluxes: instrument characterizations and field deployment

Abstract. Biological volatile organic compounds (BVOCs), such as isoprene and monoterpenes, are emitted in large amounts from forests. Quantification of the flux of BVOCs is critical in the evaluation of the impact of these compounds on the concentrations of atmospheric oxidants and on the production of secondary organic aerosol. A disjunct eddy accumulation (DEA) sampler system was constructed for the measurement of speciated BVOC fluxes. Unlike traditional eddy covariance (EC), the relatively new technique of disjunct sampling differs by taking short, discrete samples that allow for slower sampling frequencies. Disjunct sample airflow is directed into cartridges containing sorbent materials at sampling rates proportional to the magnitude of the vertical wind. Compounds accumulated on the cartridges are then quantified by thermal desorption and gas chromatography. Herein, we describe our initial tests to evaluate the disjunct sampler including the application of vertical wind measurements to create optimized sampling thresholds. Measurements of BVOC fluxes obtained from DEA during its deployment above a mixed hardwood forest at the University of Michigan Biological Station (Pellston, MI) during the 2009 CABINEX field campaign are reported. Daytime (09:00 a.m. to 05:00 p.m. LT) isoprene fluxes, when averaged over the footprint of the tower, were 1.31 mg m−2 h−1 which are comparable to previous flux measurements at this location. Speciated monoterpene fluxes are some of the first to be reported from this site. Daytime averages were 26.7 μg m−2 h−1 for α-pinene and 10.6 μg m−2 h−1 for β-pinene. These measured concentrations and fluxes were compared to the output of an atmospheric chemistry model, and were found to be consistent with our knowledge of the variables that control BVOCs fluxes at this site.

Fabrication of novel nanoporous array anodic alumina solid-phase microextraction fiber coating and its potential application for headspace sampling of biological volatile organic compounds

In the study, nanoporous array anodic alumina (NAAA) prepared by a simple, rapid and stable two-step anodic oxidization method was introduced as a novel solid-phase microextraction (SPME) fiber coating. The regular nanoporous array structure and chemical composition of NAAA SPME fiber coating was characterized and validated by scanning electron microscopy and energy dispersive spectroscopy, respectively. Compared with the commercial polydimethylsiloxane (PDMS) SPME fiber coating, NAAA SPME fiber coating achieved the higher enrichment capability (1.7–4.7 folds) for the mixed standards of volatile organic compounds (VOCs). The selectivity for volatile alcohols by NAAA SPME fiber coating demonstrated an increasing trend with the increasing polarity of alcohols caused by the gradually shortening carbon chains from 1-undecanol to 1-heptanol or the isomerization of carbon chains of some typical volatile alcohols including 2-ethyl hexanol, 1-octanol, 2-phenylethanol, 1-phenylethanol, 5-undecanol, 2-undecanol and 1-undecanol. Finally, NAAA SPME fiber coating was originally applied for the analysis of biological VOCs of Bailan flower, stinkbug and orange peel samples coupled with gas chromatography–mass spectrometry (GC–MS) detection. Thirty, twenty-seven and forty-four VOCs of Bailan flower, stinkbug and orange peel samples were sampled and identified, respectively. Moreover, the contents of trace 1-octanol and nonanal of real orange peel samples were quantified for the further method validation with satisfactory recoveries of 106.5 and 120.5%, respectively. This work proposed a sensitive, rapid, reliable and convenient analytical method for the potential study of trace and small molecular biological VOCs by the novel NAAA SPME fiber coating.

Fast Detection of Volatile Organic Compounds from Bacterial Cultures by Secondary Electrospray Ionization-Mass Spectrometry

We propose a novel application of secondary electrospray ionization-mass spectrometry (SESI-MS) as a real-time clinical diagnostic tool for bacterial infection. It is known that volatile organic compounds (VOCs), produced in different combinations and quantities by bacteria as metabolites, generate characteristic odors for certain bacteria. These VOCs comprise a specific metabolic profile that can be used for species or serovar identification, but rapid and sensitive analytical methods are required for broad utility. In this study, the VOC profiles of five bacterial groups from four genera, Pseudomonas aeruginosa, Staphylococcus aureus, Escherichia coli, Salmonella enterica serovar Typhimurium, and Salmonella enterica serovar Pullorum, were characterized by SESI-MS. Thirteen compounds were identified from these bacterial cultures, and the combination of these VOCs creates a unique pattern for each genus. In addition, principal component analysis (PCA) was applied for the purpose of species or serovar discrimination. The first three principal components exhibit a clear separation between the metabolic volatile profiles of these five bacterial groups that is independent of the growth medium. As a first step toward addressing the complexity of clinical application, in vitro tests for mixed cultures were conducted. The results show that individual species or serovars in a mixed culture are identifiable among a biological VOC background, and the ratios of the detected volatiles reflect the proportion of each bacterium in the mixture. Our data confirm the utility of SESI-MS in real-time identification of bacterial species or serovars in vitro, which, in the future, may play a promising clinical role in diagnosing infections.
