Suppression of α-glucosidase gene by RNA interference can increase resistance of Culex quinquefasciatus to Bacillus sphaericus binary toxin

Bacillus sphaericus binary toxin displays a specific toxicity towards Culex spp. larvae. Resistance of Culex quinquefasciatus to B. sphaericus is associated with the reduction or absence of 66-kDa α-glucosidase, a binary toxin receptor, in larval midgut microvilli. In this study, long double-stranded RNA (dsRNA) targeting the α-glucosidase gene was used to suppress the expression of α-glucosidase in Cx. quinquefasciatus larvae and the consequences of the suppression was investigated. Three sets of dsRNAs, dsGRi1, dsGRi2, and dsGRi3, which are homologous to distinct regions of α-glucosidase gene, were delivered to mosquito larva by feeding with dsRNA–chitosan particles. Reverse transcriptase-polymerase chain reaction results demonstrated that the expression of α-glucosidase mRNA was reduced by 43% in larvae fed with dsGRi3–chitosan, compared to those fed with dsgfp–chitosan particles. Western blot analysis revealed a 7–17% reduction of α-glucosidase protein expression in larvae fed with dsGRi–chitosan. Mortality rates of larvae that received dsRNAs–chitosan particles were only 45–62% compared to those that received dsgfp–chitosan particles. These results indicate that α-glucosidase is essential for the toxicity of B. sphaericus binary toxin to Cx. quinquefasciatus.

The N-terminal third of the BinB subunit from the Bacillus sphaericus binary toxin is sufficient for its interaction with midgut receptors in Culex quinquefasciatus

Heterodimeric binary (Bin) toxin, the major insecticidal protein from Bacillus sphaericus, acts on Culex quinquefasciatus larvae through specific binding to the midgut receptor Cqm1, a role mediated by its 448-amino-acid-long BinB subunit. The molecular basis for receptor recognition is not well understood and this study attempted to identify protein segments and amino acid motifs within BinB that are required for this event. First, N- and C-terminally truncated constructs were evaluated for their capacity to bind to native Cqm1 through pull-down assays. These showed that residues N33 to L158 of the subunit are required for Cqm1 binding. Nine different full-length mutants were then generated in which selected blocks of three amino acids were replaced by alanines. In new pull-down assays, two mutants, in which residues (85) IRF(87) and (147) FQF(149) were targeted, failed to bind the receptor. Competition binding assays confirmed the requirements for the N-terminal 158 residues, and the (147) FQF(149) epitope, for the mutant proteins to compete with native Bin toxin when binding to membrane fractions from the insect midgut. The data from this work rule out the involvement of C-terminal segments in receptor binding, highlighting the need for multiple elements within the protein's N-terminal third for it to occur.

Stability of Culex quinquefasciatus resistance to Bacillus sphaericus evaluated by molecular tools

Bacillus sphaericus binary toxin action on Culex quinquefasciatus larvae relies on the binding to Cqm1α-glucosidases, which act as midgut receptors. Resistance of two laboratory-selected colonies is associated with the allele cqm1 REC that prevents Cqm1 expression as membrane-bound molecules. This study evaluated stability of resistance after the interruption of selection pressure and introduction of susceptible individuals in these colonies. Bioassays showed that frequency of resistant larvae did not decrease throughout 11 generations, under these conditions, and it was associated to a similar frequency of larvae lacking the Cqm1α-glucosidase receptor, detected by in gel enzymatic assays. Direct screening of the cqm1 REC allele, by specific PCR, showed that its frequency remained stable throughout 11 generations. Parental resistant colony did not display biological costs regarding fecundity, fertility and pupal weight and data from susceptibility assays, enzymatic assays and PCR screening showed that cqm1 REC was not disfavored in competition with the susceptible allele and persisted in the progenies, in the lack of selection pressure. Characterization of molecular basis of resistance is essential for developing diagnostic tools and data have relevant implication for the establishment of strategies for resistance management.

Role of cysteine at positions 67, 161 and 241 of a Bacillus sphaericus binary toxin BinB

Binary toxin consisting of BinA and BinB from Bacillus sphaericus is toxic to mosquito larvae. BinB is responsible for specific binding to the larval gut cell membrane while BinA is crucial for toxicity. To investigate functional role of cysteine in BinB, three cysteine residues at positions 67, 161, and 241 were replaced by alanine or serine. Mutations at these positions did not affect protein production and overall structure of BinB. These cysteine residues are not involved in disulfide bond formation between BinB molecules. Mosquito-larvicidal assays revealed that C67 and C161 are essential for toxicity, whereas C241 is not. Mutations at C67 and C161 resulted in weaker BinA-BinB interaction. The loss of toxicity may be due to the reduction of interactions between BinA and BinB or BinB and its receptor. C67 and C161 could also play a part during conformational changes or internalization of the binary toxin into the target cell. [BMB reports 2010; 43(1): 23-28].

Identification and characterization of the receptor for the Bacillus sphaericus binary toxin in the malaria vector mosquito, Anopheles gambiae

The binary toxin (Bin) from Bacillus sphaericus exhibits a highly insecticidal activity against Culex and Anopheles mosquitoes. The cytotoxicity of Bin requires an interaction with a specific receptor present on the membrane of midgut epithelial cells in larvae. A direct correlation exists between binding affinity and toxicity. The toxin binds with high affinity to its receptor in its primary target, Culex pipiens, and displays a lower affinity to the receptor in Anopheles gambiae, which is less sensitive to Bin. Although the Bin receptor has previously been identified and named Cpm1 in C. pipiens, its structure in Anopheles remains unknown. In this study, we hypothesize that the Anopheles Bin receptor is an ortholog of Cpm1. By screening the Anopheles genomic database, we identified a candidate gene (Agm3) which is expressed primarily on the surface of midgut cells in larvae and which functions as a receptor for Bin. A Cpm1-like gene is also present in the Bin-refractory species Aedes aegypti. Overall, our results indicate that the three mosquito genes examined share a very similar organization and are strongly conserved at the amino acid level, in particular in the NH(2)-terminus, a region believed to contain the ligand binding site, suggesting that relatively few amino acids residues are critical for high affinity binding of the toxin.

Effects of a mosquitocidal toxin on a mammalian epithelial cell line expressing its target receptor

The spread of diseases transmitted by Anopheles and Culex mosquitoes, such as malaria and West Nile fever, is a growing concern for human health. Bacillus sphaericus binary toxin (Bin) is one of the few available bioinsecticides able to control populations of these mosquitoes efficiently. We previously showed that Bin binds to Cpm1, an alpha-glucosidase located on the apical side of Culex larval midgut epithelium. We analysed the effects of Bin by expressing a construct encoding Cpm1 in the mammalian epithelial MDCK cell line. Cpm1 is targeted to the apical side of polarized MDCK, where it is anchored by glycosylphosphatidylinositol (GPI) and displays alpha-glucosidase activity. Bin bound to transfected cells and induced a non-specific current presumably related to the opening of pores. The formation of these pores may be related to the location of the toxin/receptor complex in lipid raft microdomains. Finally, Bin promoted the time-dependent appearance of intracytoplasmic vacuoles but did not drive cell lysis. Thus, the dual functionality (enzyme/toxin receptor) of Cpm1 is fully conserved in MDCK cells and Cpm1 is an essential target protein for Bin cytotoxicity in Culex mosquitoes.

Crystallization of the mosquito-larvicidal binary toxin produced by Bacillus sphaericus.

The Bacillus sphaericus binary toxin is expressed during the early stages of sporulation and is composed of two separately encoded polypeptides: BinA (41.9 kDa) and BinB (51.4 kDa). The binary toxin forms microcrystalline inclusions inside the mother cell that, once ingested, are solubilized in the alkaline pH of the larval gut. B. sphaericus cultures were grown to complete sporulation, harvested and washed. The binary toxin was solubilized with 50 mM NaOH, purified using column chromatography and crystallized. Crystals were grown using the sitting-drop vapor-diffusion method in a glycine-buffered solution containing 0.5 M NaCl and 19.5% PEG 4000 as the precipitant. Native data were collected to a resolution of 2.7 A.

Characterization of Cyt2Bc toxin from Bacillus thuringiensis subsp. medellin.

We cloned and sequenced a new cytolysin gene from Bacillus thuringiensis subsp. medellin. Three IS240-like insertion sequence elements and the previously cloned cyt1Ab and p21 genes were found in the vicinity of the cytolysin gene. The cytolysin gene encodes a protein 29.7 kDa in size that is 91.5% identical to Cyt2Ba from Bacillus thuringiensis subsp. israelensis and has been designated Cyt2Bc. Inclusions containing Cyt2Bc were purified from the crystal-negative strain SPL407 of B. thuringiensis. Cyt2Bc reacted weakly with antibodies directed against Cyt2Ba and was not recognized by an antiserum directed against the reference cytolysin Cyt1Aa. Cyt2Bc was hemolytic only upon activation with trypsin and had only one-third to one-fifth of the activity of Cyt2Ba, depending on the activation time. Cyt2Bc was also mosquitocidal against Aedes aegypti, Anopheles stephensi, and Culex quinquefasciatus, including strains resistant to the Bacillus sphaericus binary toxin. Its toxicity was half of that of Cyt2Ba on all mosquito species except resistant C. quinquefasciatus.

Resistance in a Laboratory Population of Culex Quinquefasciatus (Diptera: Culicidae) to Bacillus Sphaericus Binary Toxin is Due to a Change in the Receptor on Midgut Brush-Border Membranes

Direct binding experiments with isolated brush border membrane fractions (BBMF) from larvae of a susceptible laboratory strain of Culex quinquefasciatus Say, indicated the presence of a single class of Bacillus sphaericus binary toxin receptors. The dissociation constant (Kd) was approximately 11 nM and the maximum binding capacity (Bmax) approximately 8 pmol/mg BBMF protein. Similar binding experiments with a field population of C. quinquefasciatus that had been selected in the laboratory to more than 100,000-fold resistance to B. sphaericus binary toxin failed to reveal the presence of any specific binding. Thus this resistant strain had lost the functional receptor for B. sphaericus toxin. The binding characteristics of BBMF from the F1 larval progeny (susceptible females x resistant males) were very close to those of the parental susceptible strain, consistent with the resistance being recessive.

Binding of Bacillus sphaericus binary toxin to a specific receptor on midgut brush-border membranes from mosquito larvae.

The presence of specific receptors for Bacillus sphaericus binary toxin on brush-border membrane fractions (BBMF) from Culex pipiens larvae midgut cells was demonstrated by an in vitro binding assay. Both activated and radiolabelled polypeptides from the 51-kDa and 42-kDa binary toxin of B. sphaericus 1593 specifically bound to BBMF. Direct binding and homologous competition experiments indicated a single class of B. sphaericus toxin receptors, with a dissociation constant (Kd) of approximately 20 nM and a maximum binding capacity (Bmax) of approximately 7 pmol/mg BBMF protein. The sugars GalNAc, GlcNAc and N-acetyl neuraminic acid had no detectable inhibitory effect on toxin binding to C. pipiens BBMF. Binding experiments with the non-susceptible mosquito species Aedes aegypti failed to detect significant binding of B. sphaericus binary toxin to A. aegypti BBMF.

Binding of purified Bacillus sphaericus binary toxin and its deletion derivatives to Culex quinquefasciatus gut: elucidation of functional binding domains.

Highly larvicidal strains of Bacillus sphaericus produce a binary toxin composed of 51 and 42 kDa proteins which binds to sharply delineated regions of the gastric caecum and posterior midgut of susceptible larvae of the mosquito Culex quinquefasciatus. To investigate the role of the individual subunits and the organization of functional binding regions within the toxin, plasmids were constructed for the expression in Escherichia coli of the toxin proteins and their NH2- and COOH-terminal deletion derivatives as fusions with glutathione S-transferase (GST). Toxin proteins were purified by affinity chromatography followed by cleavage from the GST carrier with thrombin. The LC50 values for the purified toxin proteins and their deletion derivatives were determined. The binding patterns of fluorescently labelled toxin suggested that the 51 kDa protein is the primary binding component of the toxin and mediates the regional binding and internalization of the 42 kDa protein. Examination of the toxin deletion derivatives revealed that the NH2-terminal region of the 51 kDa protein was required for binding to the larval gut, whilst the COOH-terminal region was responsible for interacting with the 42 kDa protein. Toxicity was strongly correlated with the subsequent internalization of the toxin, probably by endocytosis.
