Influence of Perceptual Load on Attentional Orienting in Post-Stroke Fatigue: A Study of Auditory Evoked Potentials

Objective Increasing perceptual load alters behavioral outcomes in post-stroke fatigue (PSF). While the effect of perceptual load on top-down attentional processing is known, here we investigate if increasing perceptual load modulates bottom-up attentional processing in a fatigue dependent manner. Methods In this cross-sectional observational study, in 29 first-time stroke survivors with no clinical depression, an auditory oddball task consisting of target, standard, and novel tones was performed in conditions of low and high perceptual load. Electroencephalography was used to measure auditory evoked potentials. Perceived effort was rated using the visual analog scale at regular intervals during the experiment. Fatigue was measured using the fatigue severity scale. The effect of fatigue and perceptual load on behavior (response time, accuracy, and effort rating) and auditory evoked potentials (amplitude and latency) was examined using mixed model ananlysis of variances (ANOVA). Results Response time was prolonged with greater perceptual load and fatigue. There was no effect of load or fatigue on accuracy. Greater effort was reported with higher perceptual load both in high and low fatigue. p300a amplitude of auditory evoked potentials (AEP) for novel stimuli was attenuated in high fatigue with increasing load when compared to low fatigue. Latency of p300a was longer in low fatigue with increasing load when compared to high fatigue. There were no effects on p300b components, with smaller N100 in high load conditions. Interpretation High fatigue specific modulation of p300a component of AEP with increasing load is indicative of distractor driven alteration in orienting response, suggestive of compromise in bottom-up selective attention in PSF.

A structured ICA-based process for removing auditory evoked potentials

Transcranial magnetic stimulation (TMS)-evoked potentials (TEPs), recorded using electroencephalography (EEG), reflect a combination of TMS-induced cortical activity and multi-sensory responses to TMS. The auditory evoked potential (AEP) is a high-amplitude sensory potential—evoked by the “click” sound produced by every TMS pulse—that can dominate the TEP and obscure observation of other neural components. The AEP is peripherally evoked and therefore should not be stimulation site specific. We address the problem of disentangling the peripherally evoked AEP of the TEP from components evoked by cortical stimulation and ask whether removal of AEP enables more accurate isolation of TEP. We hypothesized that isolation of the AEP using Independent Components Analysis (ICA) would reveal features that are stimulation site specific and unique individual features. In order to improve the effectiveness of ICA for removal of AEP from the TEP, and thus more clearly separate the transcranial-evoked and non-specific TMS-modulated potentials, we merged sham and active TMS datasets representing multiple stimulation conditions, removed the resulting AEP component, and evaluated performance across different sham protocols and clinical populations using reduction in Global and Local Mean Field Power (GMFP/LMFP) and cosine similarity analysis. We show that removing AEPs significantly reduced GMFP and LMFP in the post-stimulation TEP (14 to 400 ms), driven by time windows consistent with the N100 and P200 temporal characteristics of AEPs. Cosine similarity analysis supports that removing AEPs reduces TEP similarity between subjects and reduces TEP similarity between stimulation conditions. Similarity is reduced most in a mid-latency window consistent with the N100 time-course, but nevertheless remains high in this time window. Residual TEP in this window has a time-course and topography unique from AEPs, which follow-up exploratory analyses suggest could be a modulation in the alpha band that is not stimulation site specific but is unique to individual subject. We show, using two datasets and two implementations of sham, evidence in cortical topography, TEP time-course, GMFP/LMFP and cosine similarity analyses that this procedure is effective and conservative in removing the AEP from TEP, and may thus better isolate TMS-evoked activity. We show TEP remaining in early, mid and late latencies. The early response is site and subject specific. Later response may be consistent with TMS-modulated alpha activity that is not site specific but is unique to the individual. TEP remaining after removal of AEP is unique and can provide insight into TMS-evoked potentials and other modulated oscillatory dynamics.

Assessing the integrity of auditory processing and sensory memory in adults with cystinosis (CTNS gene mutations)

BackgroundCystinosis, a rare lysosomal storage disease, is characterized by cystine crystallization and accumulation within tissues and organs, including the kidneys and brain. Its impact on neural function appears mild relative to its effects on other organs, but therapeutic advances have led to substantially increased life expectancy, necessitating deeper understanding of its impact on neurocognitive function in adulthood. We previously demonstrated intact auditory sensory processing, accompanied by mild sensory memory difficulties, in children and adolescents with cystinosis.MethodsWe investigated whether further progressive decrements in these processes would be observed in adults with cystinosis, comparing high-density auditory-evoked potential (AEP) recordings from adults with cystinosis (N = 15; ages: 19–38 years) to those of age-matched controls (N = 17). We employed a duration oddball paradigm with different stimulation rates, in which participants passively listened to regularly occurring standard tones interspersed with infrequently occurring deviant tones. Analyses focused on AEP components reflecting auditory sensory-perceptual processing (N1 and P2), sensory memory (mismatch negativity, MMN), and attentional orienting (P3a).ResultsOverall, adults with cystinosis produced highly similar sensory-perceptual AEP responses to those observed in controls suggesting intact early auditory cortical processing. However, significantly increased P2 and P3a amplitudes and reduced MMN at slower stimulation rates were observed, suggesting mild-to-moderate changes in auditory sensory memory and attentional processing. While cognitive testing revealed lower scores on verbal IQ and perceptual reasoning in cystinosis, these did not correlate with the AEP measures.ConclusionsThese neurophysiological data point to the emergence of subtle auditory processing deficits in early adulthood in cystinosis, warranting further investigation of memory and attentional processes in this population, and of their consequences for perceptual and cognitive function.

Noninvasive scalp recording of the middle latency responses and cortical auditory evoked potentials in the alert common marmoset

The common marmoset (Callithrix jacchus), a New World monkey, serves as a useful animal model in clinical and basic neuroscience. The present study recorded scalp auditory evoked potentials (AEP) in non-sedated common marmoset monkeys (n = 4) using a noninvasive method similar to that used in humans, and aimed to identify nonhuman primate correlates of the human AEP components. A pure tone stimulus was presented while electroencephalograms were recorded using up to 16 disk electrodes placed on the scalp and earlobes. Candidate homologues of two categories of the human AEP, namely, the middle latency responses (MLR; Na, Pa, Nb, and Pb) and the cortical auditory evoked potentials (CAEP; P1, N1, P2, N2, and the sustained potential, SP) were identified in the marmoset. These waves were labeled as CjNa, CjPa, CjNb, CjPb, CjP1, CjN1, CjP2, CjN2, and CjSP, where Cj stands for Callithrix jacchus. The last MLR component, CjPb, was identical to the first CAEP component, CjP1, similar to the relationship between Pb and P1 in humans. The peak latencies of the marmoset MLR and CAEP were generally shorter than in humans, which suggests a shorter integration time in neural processing. To our knowledge, the present study represents the first scalp recorded MLR and CAEP in the alert common marmoset. Further use of these recording methods would enable valid species comparisons of homologous brain indices between humans and animals.

Informational Masking Effects of Similarity and Uncertainty on Early and Late Stages of Auditory Cortical Processing

Understanding speech in a background of other people talking is a difficult listening situation for hearing-impaired individuals, and even for those with normal hearing. Speech-on-speech masking is known to contribute to increased perceptual difficulty over nonspeech background noise because of informational masking provided over and above the effects of energetic masking. While informational masking research has identified factors of similarity and uncertainty between target and masker that contribute to reduced behavioral performance in speech background noise, critical gaps in knowledge including the underlying neural-perceptual processes remain. By systematically manipulating aspects of acoustic similarity and uncertainty in the same auditory paradigm, the current study examined the time course and objectively quantified these informational masking effects at both early and late stages of auditory processing using auditory evoked potentials (AEPs). Thirty participants were included in a cross-sectional repeated measures design. Target-masker similarity was manipulated by varying the linguistic/phonetic similarity (i.e., language) of the talkers in the background. Specifically, four levels representing hypothesized increasing levels of informational masking were implemented: (1) no masker (quiet); (2) Mandarin; (3) Dutch; and (4) English. Stimulus uncertainty was manipulated by task complexity, specifically presentation of target-to-target interval (TTI) in the auditory evoked paradigm. Participants had to discriminate between English word stimuli (/bæt/ and /pæt/) presented in an oddball paradigm under each masker condition pressing buttons to either the target or standard stimulus. Responses were recorded simultaneously for P1-N1-P2 (standard waveform) and P3 (target waveform). This design allowed for simultaneous recording of multiple AEP peaks, as well as accuracy, reaction time, and d' behavioral discrimination to button press responses. Several trends in AEP components were consistent with effects of increasing linguistic/phonetic similarity and stimulus uncertainty. All babble maskers significantly affected outcomes compared to quiet. In addition, the native language English masker had the largest effect on outcomes in the AEP paradigm, including reduced P3 amplitude and area, as well as decreased accuracy and d' behavioral discrimination to target word responses. AEP outcomes for the Mandarin and Dutch maskers, however, were not significantly different across any measured component. Latency outcomes for both N1 and P3 also supported an effect of stimulus uncertainty, consistent with increased processing time related to greater task complexity. An unanticipated result was the absence of the interaction of linguistic/phonetic similarity and stimulus uncertainty. Observable effects of both similarity and uncertainty were evidenced at a level of the P3 more than the earlier N1 level of auditory cortical processing suggesting that higher-level active auditory processing may be more sensitive to informational masking deficits. The lack of significant interaction between similarity and uncertainty at either level of processing suggests that these informational masking factors operated independently. Speech babble maskers across languages altered AEP component measures, behavioral detection, and reaction time. Specifically, this occurred when the babble was in the native/same language as the target, while the effects of foreign language maskers did not differ. The objective results from this study provide a foundation for further investigation of how the linguistic content of target and masker and task difficulty contribute to difficulty understanding speech-in-noise.

Do visual and auditory stimulus-specific response modulation reflect different mechanisms of neocortical plasticity?

Stimulus response modulation (SRM) of sensory evoked potentials represents a promising method as a non-invasive index of long-term potentiation (LTP)-like synaptic plasticity in the human sensory cortices. As of today, however, no consensus exists regarding which experimental parameters elicit the most robust SRM response. The aim of the current study was twofold; firstly, we aimed to replicate former studies demonstrating visual SRM in healthy adults. Second, we integrated visual and auditory stimuli within the same SRM recording session to assay potential cross-modal associations. Such an association between modalities would strengthen the assumption that the SRM effect reflects common mechanisms underlying synaptic plasticity rather than reflecting modality-specific phenomena. A replication of previous findings showing robust potentiation of the visual evoked potential was evident, supporting the majority of previous work using similar paradigms, lending further support to the notion that high-frequent visual stimulation is a viable probe into LTP-like synaptic plasticity in the human visual cortex. The auditory evoked potentials (AEPs) did not, however, fully replicate previous work, though a significant increase of temporally later AEP components was found. In contrast to our hypothesis, there were no significant within-subject cross-modality correlations between the visual and auditory SRM. This lack of significant association might suggest that auditory and visual SRM depend on different mechanisms, and that further SRM studies on non-invasive LTP-like synaptic plasticity should focus on optimizing paradigms within the visual modality.

Latency-dependent filtering and compact representation of the complete auditory pathway response.

Auditory evoked potentials (AEPs) include the auditory brainstem response (ABR), middle latency response (MLR), and cortical auditory evoked potentials (CAEPs), each one covering a specific latency range and frequency band. For this reason, ABR, MLR, and CAEP are usually recorded separately using different protocols. This article proposes a procedure providing a latency-dependent filtering and down-sampling of the AEP responses. This way, each AEP component is appropriately filtered, according to its latency, and the complete auditory pathway response is conveniently represented (with the minimum number of samples, i.e., without unnecessary redundancies). The compact representation of the complete response facilitates a comprehensive analysis of the evoked potentials (keeping the natural continuity related to the neural activity transmission along the auditory pathway), which provides a new perspective in the design and analysis of AEP experiments. Additionally, the proposed compact representation reduces the storage or transmission requirements when large databases are manipulated for clinical or research purposes. The analysis of the AEP responses shows that a compact representation with 40 samples/decade (around 120 samples) is enough for accurately representing the response of the complete auditory pathway and provides appropriate latency-dependent filtering. MatLab/Octave code implementing the proposed procedure is included in the supplementary materials.

Auditory Processing of Speech and Tones in Children With Tuberous Sclerosis Complex.

Individuals with Tuberous Sclerosis Complex (TSC) have atypical white matter integrity and neural connectivity in the brain, including language pathways. To explore functional activity associated with auditory and language processing in individuals with TSC, we used electroencephalography (EEG) to examine basic auditory correlates of detection (P1, N2, N4) and discrimination (mismatch negativity, MMN) of speech and non-speech stimuli for children with TSC and age- and sex-matched typically developing (TD) children. Children with TSC (TSC group) and without TSC (typically developing, TD group) participated in an auditory MMN paradigm containing two blocks of vowels (/a/and/u/) and two blocks of tones (800 Hz and 400 Hz). Continuous EEG data were collected. Multivariate pattern analysis (MVPA) was used to explore functional specificity of neural auditory processing. Speech-specific P1, N2, and N4 waveform components of the auditory evoked potential (AEP) were compared, and the mismatch response was calculated for both speech and tones. MVPA showed that the TD group, but not the TSC group, demonstrated above-chance pairwise decoding between speech and tones. The AEP component analysis suggested that while the TD group had an increased P1 amplitude in response to vowels compared to tones, the TSC group did not show this enhanced response to vowels. Additionally, the TD group had a greater N2 amplitude in response to vowels, but not tones, compared to the TSC group. The TSC group also demonstrated a longer N4 latency to vowels compared to tones, which was not seen in the TD group. No group differences were observed in the MMN response. In this study we identified features of the auditory response to speech sounds, but not acoustically matched tones, which differentiate children with TSC from TD children.

Functional Change of Brain Serotonergic Activity and Free Tryptophan in the Plasma of Depressed Women

The main objective was to show the decrement of serotoninergic brain activity in depressed women, through the analyses of the slope amplitude of N1/P2 components of the auditory-evoked potentials (AEP), and the measurement of the L-tryptophan free fraction in plasma (FFT). This cross-sectional study was carried out in 60 women, 30 depressed and 30 normal controls. Both groups were measured FFT, glucose, and neutral amino acids (NAA) levels; besides performing AEP to analyses the N1/P2 slope amplitude. It was found a lengthening in the slope amplitude of N1/P2 components of AEP in the group of depressed women, and despite that the level of FFT was low, there were no changes between bound fraction and the total L-Trp. The former suggests a decrease in serotonergic brain activity in the group of depressed women. Otherwise, since the auditory cortex response to sound is regulated by serotonergic innervation, it was expected a change in the behavior of AEP in the group of depressed patients. Thus, the slope amplitude of N1/P2 components of the AEP and the measurement of FFT have proved to be a good clinical indicators of the serotonergic neurotransmission state in the brain of depressed patients, and in another clinical conditions where brain serotonin is involved.

A putative electrophysiological biomarker of auditory sensory memory encoding is sensitive to pharmacological alterations of excitatory/inhibitory balance in male macaque monkeys.

The amplitude of the auditory evoked N1 component that can be derived from noninvasive electroencephalographic recordings increases as a function of time between subsequent tones. N1 amplitudes in individuals with schizophrenia saturate at a lower asymptote, thus giving rise to a reduced dynamic range. Reduced N1 dynamic range is a putative electrophysiological biomarker of altered sensory memory function in individuals with the disease. To date, it is not clear what determines N1 dynamic range and what causes reduced N1 dynamic range in individuals with schizophrenia. Here we test the hypothesis that reduced N1 dynamic range results from a shift in excitatory/inhibitory (E/I) balance toward an excitation-deficient or inhibition-dominant state. We recorded auditory-evoked potentials (AEPs) while 4 macaque monkeys passively listened to sequences of sounds of random pitch and stimulus-onset asynchrony (SOA). Three independent experiments tested the effect of the N-methyl-d-aspartate receptor channel blockers ketamine and MK-801 as well as the γ-aminobutyric acid (GABA) A receptor-positive allosteric modulator midazolam on the dynamic range of a putative monkey N1 homologue and 4 other AEP components. Ketamine, MK-801 and midazolam reduced peak N1 amplitudes for the longest SOAs. Other AEP components were also affected, but revealed distinct patterns of susceptibility for the glutamatergic and GABA-ergic drugs. Different patterns of susceptibility point toward differences in the circuitry maintaining E/I balance of individual components. The study used systemic pharmacological interventions that may have acted on targets outside of the auditory cortex. The N1 dynamic range may be a marker of altered E/I balance. Reduced N1 dynamic range in individuals with schizophrenia may indicate that the auditory cortex is in an excitation-deficient or inhibition-dominant state. This may be the result of an incomplete compensation for a primary deficit in excitatory drive.

Noise attenuation analysis on auditory evoked potential based on maximum length sequence

The maximum length sequence (m-sequence) has been successfully used to study the linear/nonlinear components of auditory evoked potential (AEP) with rapid stimulation. However, more study is needed to evaluate the effect of the m-sequence order in terms of the noise attenuation performance. This study aimed to address this issue using response-free electroencephalogram (EEG) and EEGs with nonlinear AEPs. We examined the noise attenuation ratios to evaluate the noise variation for the calculations of superimposed averaging and cross-correlation, respectively, which constitutes the main process in the deconvolution method using the dataset of spontaneous EEGs to simulate the cases of different orders (order 5 to 12) of m-sequences. And an experiment using m-sequences of order 7 and 9 was performed in true cases with substantial linear and nonlinear AEPs. The results demonstrate that the noise attenuation ratio is well agreed with the theoretical value derived from the properties of m-sequences on the random noise condition. The comparison of waveforms for AEP components from two m-sequences showed high similarity suggesting the insensitivity of AEP to the m-sequence order. This study provides a more comprehensive solution to the selection of m-sequences which will facilitate the feasible application on the nonlinear AEP with m-sequence method.

Role of inter-trial phase coherence in atypical auditory evoked potentials to speech and nonspeech stimuli in children with autism

ObjectiveThis autism study investigated how inter-trial phase coherence (ITPC) drives abnormalities in auditory evoked potential (AEP) responses for speech and nonspeech stimuli. MethodsAuditory P1-N2 responses and ITPCs in the theta band (4–7 Hz) for pure tones and words were assessed with EEG data from 15 school-age children with autism and 16 age-matched typically developing (TD) controls. ResultsThe autism group showed enhanced P1 and reduced N2 for both speech and nonspeech stimuli in comparison with the TD group. Group differences were also found with enhanced theta ITPC for P1 followed by ITPC reduction for N2 in the autism group. The ITPC values were significant predictors of P1 and N2 amplitudes in both groups. ConclusionsAbnormal trial-to-trial phase synchrony plays an important role in AEP atypicalities in children with autism. ITPC-driven enhancement as well as attenuation in different AEP components may coexist, depending on the stage of information processing. SignificanceIt is necessary to examine the time course of auditory evoked potentials and the corresponding inter-trial coherence of neural oscillatory activities to better understand hyper- and hypo- sensitive responses in autism, which has important implications for sensory based treatment.

Auditory brainstem, middle and late latency responses to short gaps in noise at different presentation rates

Objective: The effects of rate on auditory-evoked potentials (AEP) to short noise gaps (12 ms) recorded at high sampling rates using wide-band filters were investigated. Design: Auditory brainstem (ABR), middle latency (MLR), late latency (LLR) and steady-state (ASSR) responses were simultaneously recorded in adult subjects at four gap rates (0.5, 1, 5 and 40 Hz). Major components (V, Na, Pa, Nb, Pb, N1 and P2) were identified at each rate and analysed for latency/amplitude characteristics. Gap responses at 40 Hz were recovered from Quasi-ASSRs (QASSR) using the CLAD deconvolution method. Study Sample: Fourteen right ears of young normal hearing subjects were tested. Results: All major components were present in all subjects at 1 Hz. P1 (P50) appeared as a low-pass filtered component of Pa and Pb waves. At higher rates, N1 and P2 disappeared completely while major ABR-MLR components were identified. Peak latencies were mostly determined by noise onsets slightly delayed by offset responses. Conclusions: Major AEP components can be recorded to short gaps at 1 Hz using high sampling rates and wide-band filters. At higher rates, only ABR and MLRs can be recorded. Such simultaneous recordings may provide a complete assessment of temporal resolution and processing at different levels of auditory pathways.

Loudness- and time-dependence of auditory evoked potentials is blunted by the NMDA channel blocker MK-801

Amplitudes of auditory evoked potentials (AEP) increase with the intensity/loudness of sounds (loudness-dependence of AEP, LDAEP), and the time between adjacent sounds (time-dependence of AEP, TDAEP). Both, blunted LDAEP and blunted TDAEP are markers of altered auditory function in schizophrenia (SZ). However, while blunted LDAEP has been attributed to altered serotonergic function, blunted TDAEP has been linked to altered NMDA receptor function. Despite phenomenological similarities of the two effects, no common pharmacological underpinnings have been identified. To test whether LDAEP and TDAEP are both affected by NMDA receptor blockade, two rhesus macaques passively listened to auditory clicks of 5 different intensities presented with stimulus-onset asynchronies ranging between 0.2 and 6.4s. 8 AEP components were analyzed, including the N85, the presumed human N1 homolog. LDAEP and TDAEP were estimated as the slopes of AEP amplitude with intensity and the logarithm of stimulus-onset asynchrony, respectively. On different days, AEPs were collected after systemic injection of MK-801 or vehicle. Both TDAEP and LDAEP of the N85 were blunted by the NMDA blocker MK-801 and recapitulate the SZ phenotype. In summary, LDAEP and TDAEP share important pharmacological commonalities that may help identify a common pharmacological intervention to normalize both electrophysiological phenotypes in SZ.

Modulation of mGlu2 Receptors, but Not PDE10A Inhibition Normalizes Pharmacologically-Induced Deviance in Auditory Evoked Potentials and Oscillations in Conscious Rats.

Improvement of cognitive impairments represents a high medical need in the development of new antipsychotics. Aberrant EEG gamma oscillations and reductions in the P1/N1 complex peak amplitude of the auditory evoked potential (AEP) are neurophysiological biomarkers for schizophrenia that indicate disruption in sensory information processing. Inhibition of phosphodiesterase (i.e. PDE10A) and activation of metabotropic glutamate receptor (mGluR2) signaling are believed to provide antipsychotic efficacy in schizophrenia, but it is unclear whether this occurs with cognition-enhancing potential. The present study used the auditory paired click paradigm in passive awake Sprague Dawley rats to 1) model disruption of AEP waveforms and oscillations as observed in schizophrenia by peripheral administration of amphetamine and the N-methyl-D-aspartate (NMDA) antagonist phencyclidine (PCP); 2) confirm the potential of the antipsychotics risperidone and olanzapine to attenuate these disruptions; 3) evaluate the potential of mGluR2 agonist LY404039 and PDE10 inhibitor PQ-10 to improve AEP deficits in both the amphetamine and PCP models. PCP and amphetamine disrupted auditory information processing to the first click, associated with suppression of the P1/N1 complex peak amplitude, and increased cortical gamma oscillations. Risperidone and olanzapine normalized PCP and amphetamine-induced abnormalities in AEP waveforms and aberrant gamma/alpha oscillations, respectively. LY404039 increased P1/N1 complex peak amplitudes and potently attenuated the disruptive effects of both PCP and amphetamine on AEPs amplitudes and oscillations. However, PQ-10 failed to show such effect in either models. These outcomes indicate that modulation of the mGluR2 results in effective restoration of abnormalities in AEP components in two widely used animal models of psychosis, whereas PDE10A inhibition does not.

The response decrease of auditory evoked potentials by repeated stimulation – Is there evidence for an interplay between habituation and sensitization?

ObjectivesTo assess whether the response decrement of auditory evoked potentials (AEPs) after stimulus repetition is affected by an interplay between sensitization and habituation. MethodsAEPs were recorded in 18 healthy participants. Stimulation consisted of trains with eight tones. The 6th stimulus of each train was a frequency deviant. The N100 amplitude to the 1st stimulus of the train was quantified in each trial. Trials with initially strong N100 responses and with initially weak N100 responses were averaged separately. ResultsFor the total trial sample, the N100 and P200 amplitudes decreased from the 1st to the 2nd stimulus of the train but not thereafter. Trials with an initially strong N100 response were qualified by likewise larger N100 amplitudes to the 2nd stimulus, as compared to trials with initially weak N100 responses, and were characterized by a pronounced N100 amplitude decrease from standards to deviants. ConclusionOur findings are difficult to reconcile with the view that the response decrement of AEP components after stimulus repetition is modulated by sensitization and habituation, as no evidence for either of these two processes could be obtained. Auditory-evoked potentials as a tool for follow-up of fibromyalgia

In this study, we assessed the value of auditory-evoked potentials (AEPs) as objective measurable reproducible tests for the follow-up of patients with fibromyalgia (FM) in response to pharmacologic and rehabilitative therapy. This study included 30 female FM patients and 10 age-matched female controls. All participants underwent a clinical examination, a psychiatric and functional assessment (sleep score, Fibromyalgia Impact Questionnaire, and Hospital Anxiety and Depression Scale) and measurement of AEPs elicited by tones of increasing intensity (60, 70, 80, and 90 dB) known as late cortical responses and cognitive auditory potentials (P300). Patients were subdivided into three equal groups. Group 1 received pregabalin, group 2 received fluoxetine, and group 3 included patients who performed a graded aerobic exercise program. Assessment was repeated at the end of the 8-week treatment period. Patients had significantly shorter N1 latencies at 60 and 70 dB, significantly shorter P2 latencies at all the studied intensities, and significantly higher N1P2 amplitudes at 90 dB.There was a statistically significant decrease in amplitude and a significant increase in P300 latency when compared with controls. Changes in AEP values before and after treatment were closely associated with the changes in psychiatric and functional assessment parameters. Improvement in the clinical assessment of the different symptoms of FM goes hand in hand with the improvement in the late cortical and cognitive components of AEPs, which provides evidence of the value of AEP as a simple, noninvasive, objective, and reproducible follow-up tool for assessment of hypervigilance and cognitive function in FM patients.

Repetition suppression of the rat auditory evoked potential at brief stimulus intervals

An important prerequisite for the development of animal models of human auditory evoked potentials (AEP) is the accurate identification of homology. Prior research has revealed some remarkably similar response properties between rat and human AEPs, although there remains little consensus regarding the nature or validity of this correspondence. In the present study we seek to extend this research by examining the response properties of rat AEP as a function of stimulus repetition and interval. The aim being to determine whether rat AEP components show the same paradoxical reversal of repetition suppression observed for the human N100 AEP component at brief stimulus intervals. To achieve this, AEPs were recorded epidurally at the vertex in the freely moving rat in response to acoustic stimuli presented at random stimulus intervals between 50 and 5000ms. Using stimulation and analysis techniques to remove AEP waveform distortion due to overlapping AEP responses, the present results show that rat AEP components can be successfully resolved at intervals as brief as 50ms. The results also demonstrate several fundamental types of correspondence between human and rat AEP components in terms of the sensitivity to stimulus interval and acoustic stimulus type. However the results found no evidence that rat AEP components show the reversal of repetition suppression at brief, relative to long, stimulus intervals as demonstrated for the N100 component in humans. The results are discussed in terms of EEG recording and AEP analysis procedures that provide promising avenues for future research.
