Cold atmospheric pressure air plasma jet disinfection of table eggs: Inactivation of Salmonella enterica, cuticle integrity and egg quality

Eggshell cuticles are first lines of defense against egg-associated pathogens, such as Salmonella enterica serovar Enteritidis (SE). Infections from eggs contaminated with this strain remain a significant risk. In addition, changes in the cuticle are closely related to changes in egg safety. The emerging non-thermal atmospheric pressure plasma technology enables a high rate of microbial inactivation at near-ambient temperatures, making it ideal for food safety applications. This study examines the effects of a cold atmospheric pressure air plasma jet (CAAP-J) on eggshell cuticle and egg quality whilst inactivating SE. Shell eggs inoculated with SE (7 log10 cfu/egg) were used as the samples to test the decontamination performance of the device. The tests were conducted using an industrial CAAP-J with different power levels (600–800 W), exposure times (60–120 s), at a fixeddistance of 20 mm from the plasma jet and an air flow rate of 3600 L/h. It was found that the best results were obtained after 120 s at maximum plasma power (800 W). Subsequent to the implementation of this plasma procedure, it was determined that no viable cells could be detected. After CAAP-J treatment, the temperature remains below 50.5 °C, thereby minimizing the risk of altering egg quality. All specific measurements (egg white pH, yolk pH, yolk color, HU, and eggshell breaking strength) have shown that CAAP-J treatment has no negative effect on egg quality. No changes in eggshell cuticle quality after CAAP-J treatment was confirmed through scanning electron microscope (SEM).

Ozone Concentration Measurement of an Atmospheric Pressure Air Plasma Jet by Ultraviolet Absorption Spectroscopy

Atmospheric pressure plasma jets have great potentials for various biomedical applications due to abundance with active oxygen species. In this article, an air plasma jet with a geometry of microhollow cathode discharge has been developed, which operates in a pulsed mode or a continuous mode depending on the dissipated power of the plasma jet. There are some continuous spectra emitted from the plasma jet, whose integrated intensity indicates that electron density increases with the increase of dissipated power, while decreases with increasing air flow rate for both of the two modes. Moreover, the electron density in the continuous mode is higher than that in the pulsed mode. Gas temperature of the plasma jet is obtained by the best fitting of the simulated and the experimental optical emission spectrum of OH (A-X), and ozone concentration produced by the plasma jet is measured by means of ultraviolet absorption spectroscopy. Results indicate that both ozone concentration and gas temperature decrease with increasing distance from the jet nozzle. At the jet nozzle, both of them increase with increasing dissipated power. However, ozone concentration decreases, while gas temperature increases with increasing air flow rate. The results are very important for the biomedical application of plasma jet.

A Magnetic Field Induced Cold Atmospheric Pressure Air Plasma Jet

Cold atmospheric pressure plasma jets (APPJs) using air as working gas are of particular interest and desirable but with significant challenge. In this article, an air APPJ induced by an external radial magnetic field is reported. The experimental results show that with the external magnet, an air plasma jet can be obtained with a length of 7 mm and a width of 10 mm. While without the external magnet, the plasma only exists in the dielectric tube. The dynamic behavior shows that its propagation velocity is close to the gas flow velocity. The plasma jet appears no difference when it is in contact with a ground conductive object. On the other hand, optical emission spectrum shows that they are dominated by the N2 and <inline-formula xmlns:mml="http://www.w3.org/1998/Math/MathML" xmlns:xlink="http://www.w3.org/1999/xlink"> <tex-math notation="LaTeX">$\text{N}_{2}^{+}$ </tex-math></inline-formula> emission. The electrical characteristic shows that the peak of the discharge current and the breakdown voltage increase when the magnet is used. The gas temperature of the plasma jet is about 310 K. Laser-induced fluorescence measurement shows that high concentration of OH radicals appears along the plasma jet. Investigation of different solutions activated by air plasma jet and their anticancer effect

In the field of plasma biomedicine, research on a plasma-activated medium (PAM) has attracted increasing attention in recent years because of its excellent characteristics. In this study, we used an atmospheric pressure air plasma jet to treat four different solutions: de-ionized water, RPMI 1640 medium, phosphate buffered saline (PBS), and saline. In order to investigate the property differences of different PAM, we mainly analyzed the physical and chemical properties and liquid-phase active species of different PAM and evaluated the inactivation of A549 lung cancer cells. The results show that the concentrations of long-lived reactive species (H2O2, NO2−, and NO3−) in different PAM increased with increasing treatment time. Biological experiments showed that the antitumor effects were in the order of PBS &amp;gt; saline &amp;gt; RPMI 1640 medium, and the best inactivation effect of plasma-activated PBS for 12 min was 89%. Meanwhile, plasma-activated PBS effectively promoted apoptosis in A549 cells, and the highest apoptosis rate was 91.3%. Therefore, this study demonstrates the medical application of different PAM in killing cancer cells and promotes the understanding of plasma–liquid interaction.

Development and characterization of touchable air plasma jet device for inactivation of oral bacteria

A compact low-temperature and atmospheric-pressure air plasma jet source is developed for the inactivation of oral bacteria. Electrical and optical measurements are carried out to understand the fundamental characteristics of the custom-built system. The adjustable operating parameters are optimized to regulate the length of the plasma plume and control the generation of the reactive species. Results show that the temperature of the plasma jet is around 40–45 °C during 5 min plasma-on, without inducing any thermal effects on bacterial cells. Importantly, the safety of the device is estimated by evaluating the current through the human body via an equivalent electrical circuit. Antibacterial studies indicate that the plasma jet can effectively inactivate E. coli, P. aeruginosa and S. aureus within 20 s. Further analysis on the morphology of bacterial cells and the intracellular DNA leakage is performed to evaluate the integrity of the cell. This study suggests that the developed portable air plasma jet can effectively deactivate oral bacteria and has great potential in future dental or clinical medical applications.

Treatment modalities for peri-implantitis: A review of literature

Introduction: Implant Dentistry was created to show reliable treatment approaches for restoring the oral cavity’s esthetic and function. However, while dental implants have a high long-term success rate, dental implants are exposed to mechanical or biological complications. Objective: The therapeutic approaches will be addressed in this review, which is the main objective of this study. Materials and Methods: Data sources: The PubMed/MEDLINE, SCOPUS, and Web of Science databases were used to conduct the literature search from June to September 2021. Study selection: Non-surgical and surgical peri-implantitis treatment modalities are the main themes of this study. Result: Implant dentistry was created to show reliable treatment approaches for restoring the oral cavity’s esthetic and function. However, while dental implants have a high long-term success rate, dental implants are exposed to mechanical or biological complications. Conclusion: Peri-implantitis appears to be a multifactorial disease including the patient’s host/microbe response, implant characteristics, soft tissue and the hard conditions surrounding the implant, and the dentist’s surgical and prosthetic part experience. Different treatment modalities are present today, all are used to treat peri-implantitis, such as surgery, laser therapy, and innovative treatments such as cold atmospheric pressure air plasma jet (CAPAJ), enamel matrix derivative, and PRF gel.

Decontamination of Escherichia coli on the surface of soybean seeds using plasma activated water

The objective of the study was the evaluation of the efficacy of plasma activated water (PAW) for Escherichia coli inoculated on soybean seed surface. PAW was prepared by high-power atmospheric pressure air plasma jet. The inactivation kinetics, explained by Bigelow log-linear and Weibull models, showed the significant antibacterial potential of PAW in comparison to the control (sterile distilled water). PAW treatment for 25 min let to log10 CFU/g reduction of E. coli from 6.8 to 3.6 and its antibacterial effect persisted further 44 h (2.7 log10 CFU/g). Decimal reduction time (D-value) for tested bacteria was 7 min. Furthermore, the influence on PAW on the polyphenolics content and antioxidant activity was evaluated. To identify the PAW chemistry, physical properties, including pH, redox potential, and conductivity were estimated. The levels of H2O2, nitrate (NO3−) and nitrite anions (NO2−) as long-lived species were measured and compared to SDW without plasma activation.

Cold atmospheric-pressure air plasma jet: Physics and opportunities

Cold atmospheric-pressure plasma jet generates rich reactive species including reactive oxygen species and reactive nitrogen species with gas temperature close to or at room temperature, which is very attractive for applications such as plasma medicine. However, under one atmospheric pressure, due to the high electron–neutral particles collision frequency (1011–12/s), it is difficult to generate atmospheric pressure plasma while keeping the gas temperature close to or at room temperature. Furthermore, when air rather than noble gases is used as working gas, due to the low energy levels of rotational and vibrational states of nitrogen and oxygen, it becomes extremely challenging to generate cold atmospheric pressure air plasma jet (CAAP-J) with gas temperature close to or at room temperature. Fortunately, after decades of research, several CAAP-Js have been reported. In this review, the state-of-the-art of the development of CAAP-Js is presented. The CAAP-Js are grouped into six categories based on their electrode configuration. A brief discussion on each group of the CAAP-Js is presented. Moreover, the physics of CAAP-Js is discussed, including the dynamics, the striation phenomenon, the temporal behavior of plasma parameters, and the nonequilibrium characteristic of CAAP-Js. Furthermore, the measurements of the reactive species generated by CAAP-Js are briefly reviewed. Finally, discussions and perspective of future research on CAAP-Js are presented.

Atmospheric-Pressure Air Plasma Jet and Its Striation Discharge Mode for Treatment of Thermally Sensitive Materials

A hollow inner electrode covered with a dielectric tube was inserted into an outer electrode and airflow was fed through the inner electrode. The electrodes were then connected to a transformer operated at an alternating voltage with sinusoidal waveform at a frequency of 20 kHz. The resulting discharge is ejected out of the outer electrode from a 0.7 mm hole in the form of a plasma jet into ambient air. The attributes of the discharged plasma jet were evaluated by monitoring the voltage and current behavior and by investigating the optical emissions. The discharge patterns in the atmospheric-pressure air plasma jet in the form of striations could be observed by the naked eye. Furthermore, we reported the striation mechanism by ion acoustic wave propagation by utilizing a simple calculation.

Enhancement Propagation of Protocorms in Orchid (Cymbidium tracyanum L. Castle) by Cold Atmospheric Pressure Air Plasma Jet

Propagation of orchid (Cymbidium tracyanum L. Castle) treated by using an air cold atmospheric pressure plasma jet at room temperature was investigated. The peak-to-peak voltage discharge and flow rate of the air atmospheric pressure plasma jet were set at 10.5 kV and 10 L/min, respectively. The constituent particles of the air cold atmospheric pressure plasma jet were revealed by optical emission spectroscopy. At the end of the jet nozzle, the constituent particles were observed, such as hydroxyl radical ˙OH, N2 first negative system (N2 1 −), N2s positive system (N2 2 +), and atomic oxygen. The average size, average number of buds, fresh and dry weights simultaneously increased by cold air plasma treatment compared with the control ones. The highest positive effects of treated protocorms on average size, average number of buds, fresh and dry weight were significantly improved by 238.2, 475.9, 85.6, and 152.5%, respectively. The transmission electron microscope showed the cytoplasmic cell wall in orchid protocorm disturbed by air cold plasma. Thus, the effects of constituent particles of air plasma might attack in the cell wall of protocorm leading to cell loosening, which improved elongation and released bud dormancy. These results indicated that the propagation of orchid protocorms depended on the effects of air cold plasma treatment with different plasma time durations.

Influence of Nonthermal Atmospheric Plasma-Activated Water on the Structural, Optical, and Biological Properties of Aspergillus brasiliensis Spores

Plasma-activated water (PAW) has emerged as a platform for sterilizing fungal pathogens. In this study, we investigated the influence of PAW on black melanized spores of Aspergillus brasiliensis to explore the mechanism of fungal spore inactivation. PAW was prepared by activating deionized water with a nonthermal atmospheric pressure air plasma jet (soft plasma jet). The concentrations of H2O2 and NOx in the PAW treated by the soft plasma jet for 3 min were 50 μM and 1.8 mM, respectively, and the pH of the PAW was 3.10. The reactive oxygen and nitrogen species (RONS) in the PAW increased with longer plasma activation time. After being treated for 30 min in the PAW with a plasma activation time of 3 min, the spore viability dramatically dropped to 15%. The viabilities of 0.3% H2O2- and 0.3% HNO3-treated spores were 22% and 42%, respectively. The breakage of the spore cell wall by the PAW was revealed in scanning electron microscope images and flow cytometry measurements. Disruption of cell wall integrity provides a path for intracellular components to escape and RONS of the PAW can attack intracellular components directly. Degradation of high molecular genomic DNA was also observed by agarose gel electrophoresis. These results suggest that long-lived reactive species generated in the PAW play an important role in the inactivation of melanized fungal spores. Consequently, PAW produced by a soft plasma jet can be applied to sterilize bioprotective walled fungal spores in a relatively large volume.

Atmospheric air plasma jet for improvement of paint adhesion to aluminium surface in industrial applications

Improvement of paint adhesion to aluminium surfaces is one of the main challenges in many industrial applications. In this paper, we introduce the atmospheric pressure air plasma jet as an appropriate candidate for preparation of 5052 aluminium surface alloy to improve paint adhesion in the industrial level. The employed plasma jet can promote paint adhesion to aluminium surface at the treatment velocity of 2 m/min and plasma size of 10 mm. Based on the cross-cut test, adhesion of polyurethane paint to the surface greatly increases from 1B to 5B level due to the plasma treatment. According to the results, the surface wettability increases under the influence of the plasma treatment so that water droplet contact angle reduces from 79.0°±2.0°–27.5°±2.0° after the treatment. Dyne test ink also denotes the increment of surface energy to the greater than 72 mN/m. Besides, we employ various analytical methods to investigate the physical and chemical changes arise from the plasma processing to the surface. Atomic force microscopy (AFM) results show a twofold increase in the roughness parameters of plasma treated surface which can result in a stronger paint and surface interlocking. Chemical analysis of the surface reveals that plasma treatment of the aluminium surface leads to the surface cleaning and formation of hydrophilic functional groups that attract much more water towards the surface and improves the paint adhesion.

Interaction of Plasma Jet With Methanol as Fundamental Processes in Solution

Plasma depollution is a relatively new field that investigates potential applications of atmospheric pressure plasma in environmental engineering, such as microorganism inactivation and degradation of organic molecules in water. In order to enunciate mechanisms of degradation of organic matter at molecular or atomic levels, the interaction between He, N2, O2, and air plasma jet and methanol in water was analyzed by gas chromatography–mass spectrum and Fourier transform infrared spectroscopy. Results show that the efficiency of methanol degradation is strongly dependent on the working gas and plasma treatment time. Compared to atmospheric pressure N2, O2, and air plasma jet, the He plasma jet may be utilized to degrade methanol more efficiently in aqueous solution. The efficiencies of degradation of methanol in water can be well described by using the chemical reaction rate model, where reactive oxygen and hydroxyl radical species play crucial roles in the degradation process. This paper provides an insight into the mechanism of degradation of organic pollutant in water by plasma.

Spore Viability and Cell Wall Integrity of Cordyceps pruinosa Treated with an Electric Shock-Free, Atmospheric-Pressure Air Plasma Jet

Atmospheric-pressure Ar plasma jets are known to be detrimental to Cordyceps pruinosa spores. However, it is not clear what kinds of reactive species are more effective with regard to fungal cell death. Herein, we study which reactive species plays pivotal roles in the death of fungal spores using an electric shock-free, atmospheric-pressure air plasma jet, simply called soft plasma jet. Plasma treatment significantly reduced the spore viability and damaged fungal DNA. As observed from the circular dichroism spectra, scanning electron microscope images, and flow cytometric measurements, cell wall integrity was decreased by reactive oxygen and nitrogen species (RONS) from the plasma itself and the plasma-activated water. Consequently, degradation of the spore cell wall allows RONS from the plasma to reach the intracellular components. Such plasma-induced intracellular RONS can attack spore DNA and other intracellular components, as confirmed by electrophoresis analysis and phosphorylated histone measurement. In addition, weakening of the spore cell wall allowed for the loss of intracellular components, which can lead to cell death. Plasma radicals were investigated by measuring the optical emission spectrum of the soft plasma jet, and intracellular reactive oxygen species were confirmed by measuring the fluorescence of 2′, 7′-dichlorodihydrofluorescein-diacetate (H2DCF-DA)-stained spores. The soft plasma jet generated considerable amounts of H2O2 and NOx but a very small number of OH radicals as compared to the atmospheric-pressure Arplasma jet; this indicates that plasma-induced long-lived reactive species (H2O2 and NOx) play an important role in the weakening of spore cell walls and cell death.

Effect of novel atmospheric-pressure jet pretreatment on the drying kinetics and quality of white grapes.

Raisin is a popular snack and a common constituent of many foods owing to its good flavor and nutritional value. Conventional drying of grapes can be a slow and energy-consuming process as their waxy surface hinders efficient moisture migration. A drying pretreatment that disrupts the waxy cuticle is usually applied to increase the drying rate. The application of an atmospheric-pressure air plasma jet to the grape surface could effectively enhance the drying kinetics and decrease the drying time by more than 20%. Through etching of the waxy cuticle, the air plasma jet optimally improves the quality of the final product. Although the surface hydrophilicity was increased by 40%, the physical appearance, color, and texture of plasma-treated raisins were similar to the product from untreated control and chemical-treated groups. A more than twofold increase in the total phenolic content and antioxidant capacity was observed when compared to other experimental groups. The results indicate that atmospheric plasma could be a better option than using chemicals to pretreat grapes before drying since it leaves no toxic residue, while successfully preserving the product quality. This work shows the great potential for the application of atmospheric air plasma in the drying of food materials. © 2019 Society of Chemical Industry.

A novel cold atmospheric pressure air plasma jet for peri-implantitis treatment: An in vitro study.

Peri-implantitis is difficult to treat in clinical settings; this is not only because it is a site-specific infectious disease but also because it impedes osseointegration. In this study, a novel cold atmospheric pressure air plasma jet (CAPAJ) was applied to study the treatment of peri-implantitis in vitro. CAPAJ treated the samples for 2, 4 and 6 min, respectively. To evaluate the titanium surface characteristics, the surface elemental composition (X-ray photoelectron spectroscopy [XPS]), roughness and hydrophilicity were evaluated in each group. Concurrently, the sterilization and osseointegration effect of CAPAJ were also examined. Results revealed that after CAPAJ modification, roughness and hydrophilicity of titanium surfaces were significantly increased. Moreover, XPS results demonstrated that the C1s peak was reduced and N1s and O1s peaks were obviously improved. More importantly, CAPAJ showed favorable sterilization and bone formation effects. CAPAJ seemed a simpler and more efficient strategy for the peri-implantitis treatment.

A cost- and time-saving strategy of spraying TiO2 self-cleaning coatings in tubular substrates by air cold plasma

In this study, using an atmospheric pressure air plasma jet generated by a dielectric barrier structure with hollow electrodes (HEDBS), we developed an ultrafast process for spraying TiO2 self-cleaning films inside tubular substrates. Importantly, SEM images showed that the TiO2 particles were dispersed evenly in the tubular substrates. Furthermore, Raman and XRD pattern indicated the anatase structure of the HEDBS-spayed TiO2 coating after heating at 270°C. Further results of the self cleaning test suggested that the proposed cost- and time-saving HEDBS approach with air working gas could provide a feasible way for synthesizing thin TiO2 nanofilms.

Two-photon absorption laser induced fluorescence measurement of atomic oxygen density in an atmospheric pressure air plasma jet

Atomic oxygen number density [O] is measured in an air atmospheric pressure plasma jet (APPJ) using two-photon absorption laser induced fluorescence (TALIF). Gas flow is fixed at 8 slpm, the RF power coupled into the plasma jet varied between 5 W and 20 W, and the resulting changes in atomic oxygen density measured. Photolysis of molecular oxygen is employed to allow in situ calibration of the TALIF system. During calibration, O2 photo-dissociation and two-photon excitation of the resulting oxygen atoms are achieved within the same laser pulse. The atomic oxygen density produced by photolysis is time varying and spatially non-uniform which needs to be corrected for to calibrate the TALIF system for measurement of atomic oxygen density in plasma. Knowledge of the laser pulse intensity I0(t), wavelength, and focal spot size allows correction factors to be determined using a rate equation model. Atomic oxygen is used for calibration and measurement, so the laser intensity can be increased outside the TALIF quadratic laser power dependence region without affecting the calibration reliability as the laser power dependence will still be the same for both. The atomic O density results obtained are not directly benchmarked against other known density measurement techniques. The results show that the plasma jet atomic oxygen content increases as the RF power coupled into the plasma increases.

Effects of a Nonthermal Atmospheric Pressure Plasma Jet on Human Gingival Fibroblasts for Biomedical Application.

Nonthermal atmospheric pressure plasma jets (APPJ) have been developed and applied in biomedical research as a cancer treatment or bacterial sterilization. However, the drawback of APPJ on normal oral cells during plasma treatment and underlying cell death mechanisms have not been studied and clearly explained, although there is known to be an influence from reactive oxygen species (ROS). Hence, this study investigates whether and how a nonthermal atmospheric pressure air plasma jet kills human normal gingival cells using immortalized human gingival fibroblasts (hTERT-hNOF cells). In this study, a set of physicochemical or biological methods were used to illuminate the killing mechanisms. It was found that ROS were induced intracellularly without a breakdown of the cell wall and apoptosis was involved in cell death when an air APPJ treatment was performed on the cells directly without media; the air treatment only supported a detachment of the cells without increase of ROS. It was also revealed that a correlation between intracellular ROS concentration and cells viability existed. These results indicated that the direct air APPJ treatment possibly raises safety issue to normal tissue and thereby APPJ application in biomedical field needs more in vitro and in vivo study to optimize it.
