Based on the compositeness condition, the coupling between X(4160) and the Ds∗D¯s∗\\documentclass[12pt]{minimal} \\usepackage{amsmath} \\usepackage{wasysym} \\usepackage{amsfonts} \\usepackage{amssymb} \\usepackage{amsbsy} \\usepackage{mathrsfs} \\usepackage{upgreek} \\setlength{\\oddsidemargin}{-69pt} \\begin{document}$$D_s^*\\bar{D}_s^*$$\\end{document} molecular state is determined. The other coupling constants used are determined phenomenologically. The calculation of the decay widths shows that the D∗D¯∗\\documentclass[12pt]{minimal} \\usepackage{amsmath} \\usepackage{wasysym} \\usepackage{amsfonts} \\usepackage{amssymb} \\usepackage{amsbsy} \\usepackage{mathrsfs} \\usepackage{upgreek} \\setlength{\\oddsidemargin}{-69pt} \\begin{document}$$D^*\\bar{D}^*$$\\end{document} decay mode has a larger branching ratio. These predictions can be seen as a test for the charmonium assignment of X(4160).

Mining expressed sequence tag (EST) microsatellite markers to assess the genetic differentiation of five Hynobius species endemic to Taiwan

Taiwan harbors five endemic species of salamanders (Hynobius spp.) that inhabit distinct alpine regions, contributing to population fragmentation across isolated “sky islands”. With an evolutionary history spanning multiple glacial-interglacial cycles, these species represent an exceptional paradigm for exploring biogeography and speciation. However, a lack of suitable genetic markers applicable across species has limited research efforts. Thus, developing cross-amplifying markers is imperative. Expressed sequence-tag simple-sequence repeats (EST-SSRs) that amplify across divergent lineages are ideal for species identification in instances where phenotypic differentiation is challenging. Here, we report a suite of cross-amplifying EST-SSRs from the transcriptomes of the five Hynobius species that exhibit an interspecies transferability rate of 67.67%. To identify individual markers exhibiting cross-species polymorphism and to assess interspecies genetic diversity, we assayed 140 individuals from the five species across 84 sampling sites. A set of EST-SSRs with a high interspecies polymorphic information content (PIC = 0.63) effectively classified these individuals into five distinct clusters, as supported by discriminant analysis of principal components (DAPC), STRUCTURE assignment tests, and Neighbor-joining trees. Moreover, pair-wise FST values > 0.15 indicate notable between-cluster genetic divergence. Our set of 20 polymorphic EST-SSRs is suitable for assessing population structure within and among Hynobius species, as well as for long-term monitoring of their genetic composition.

Population and spatial dynamics of desert bighorn sheep in Grand Canyon during an outbreak of respiratory pneumonia

IntroductionTerrestrial species in riverine ecosystems face unique constraints leading to diverging patterns of population structure, connectivity, and disease dynamics. Desert bighorn sheep (Ovis canadensis nelsoni) in Grand Canyon National Park, a large native population in the southwestern USA, offer a unique opportunity to evaluate population patterns and processes in a remote riverine system with ongoing anthropogenic impacts. We integrated non-invasive, invasive, and citizen-science methods to address questions on abundance, distribution, disease status, genetic structure, and habitat fragmentation.MethodsWe compiled bighorn sightings collected during river trips by park staff, commercial guides, and private citizens from 2000–2018 and captured bighorn in 2010–2016 to deploy GPS collars and test for disease. From 2011–2015, we non-invasively collected fecal samples and genotyped them at 9–16 microsatellite loci for individual identification and genetic structure. We used assignment tests to evaluate genetic structure and identify subpopulations, then estimated gene flow and recent migration to evaluate fragmentation. We used spatial capture-recapture to estimate annual population size, distribution, and trends after accounting for spatial variation in detection with a resource selection function model.Results and discussionFrom 2010–2018, 3,176 sightings of bighorn were reported, with sightings of 56–145 bighorn annually on formal surveys. From 2012–2016, bighorn exhibiting signs of respiratory disease were observed along the river throughout the park. Of 25 captured individuals, 56% were infected by Mycoplasma ovipneumoniae, a key respiratory pathogen, and 81% were recently exposed. Pellet sampling for population estimation from 2011–2015 yielded 1,250 genotypes and 453 individuals. We detected 6 genetic clusters that exhibited mild to moderate genetic structure (FST 0.022–0.126). The river, distance, and likely topography restricted recent gene flow, but we detected cross-river movements in one section via genetic recaptures, no subpopulation appeared completely isolated, and genetic diversity was among the highest reported. Recolonization of one large stretch of currently empty habitat appears limited by the constrained topology of this system. Annual population estimates ranged 536–552 (95% CrI range 451–647), lamb:ewe ratios varied, and no significant population decline was detected. We provide a multi-method sampling framework useful for sampling other wildlife in remote riverine systems.

Internet-enabled lab-on-a-chip technology for education

Despite many interventions, science education remains highly inequitable throughout the world. Internet-enabled experimental learning has the potential to reach underserved communities and increase the diversity of the scientific workforce. Here, we demonstrate the use of lab-on-a-chip (LoC) technologies to expose Latinx life science undergraduate students to introductory concepts of computer programming by taking advantage of open-loop cloud-integrated LoCs. We developed a context-aware curriculum to train students at over 8000 km from the experimental site. Through this curriculum, the students completed an assignment testing bacteria contamination in water using LoCs. We showed that this approach was sufficient to reduce the students’ fear of programming and increase their interest in continuing careers with a computer science component. Altogether, we conclude that LoC-based internet-enabled learning can become a powerful tool to train Latinx students and increase the diversity in STEM.

Large-scale assessment of genetic structure to assess risk of populations of a large herbivore to disease.

Chronic wasting disease (CWD) can spread among cervids by direct and indirect transmission, the former being more likely in emerging areas. Identifying subpopulations allows the delineation of focal areas to target for intervention. We aimed to assess the population structure of white-tailed deer (Odocoileus virginianus) in the northeastern United States at a regional scale to inform managers regarding gene flow throughout the region. We genotyped 10 microsatellites in 5701 wild deer samples from Maryland, New York, Ohio, Pennsylvania, and Virginia. We evaluated the distribution of genetic variability through spatial principal component analysis and inferred genetic structure using non-spatial and spatial Bayesian clustering algorithms (BCAs). We simulated populations representing each inferred wild cluster, wild deer in each state and each physiographic province, total wild population, and a captive population. We conducted genetic assignment tests using these potential sources, calculating the probability of samples being correctly assigned to their origin. Non-spatial BCA identified two clusters across the region, while spatial BCA suggested a maximum of nine clusters. Assignment tests correctly placed deer into captive or wild origin in most cases (94%), as previously reported, but performance varied when assigning wild deer to more specific origins. Assignments to clusters inferred via non-spatial BCA performed well, but efficiency was greatly reduced when assigning samples to clusters inferred via spatial BCA. Differences between spatial BCA clusters are not strong enough to make assignment tests a reliable method for inferring the geographic origin of deer using 10 microsatellites. However, the genetic distinction between clusters may indicate natural and anthropogenic barriers of interest for management.

Population assignment of migratory Westslope Cutthroat Trout (WCT) in the Clark Fork–Pend Oreille River basin

AbstractObjectiveThe Clark Fork–Pend Oreille River basin of northeastern Washington and the Idaho Panhandle historically supported a robust metapopulation of the Westslope Cutthroat Trout (WCT) Oncorhynchus lewisi, a western native salmonid of high cultural and economic value. The construction of impassible hydroelectric dams and smaller instream barriers has prevented the return of migratory WCT to spawning tributaries, leading to the fragmentation of this metapopulation over the past 100 years. One such impassible barrier is Albeni Falls Dam (AFD) near Newport, Washington, which was completed without fish passage capabilities in 1955. We sought to examine large‐scale genetic patterns in the study area and determine the most likely spawning tributary of origin for migratory WCT captured below AFD.MethodsWe created a genetic baseline representative of populations within the Clark Fork–Pend Oreille River basin from upstream and downstream of the dam using 191 biallelic single‐nucleotide polymorphism genetic markers. Our data set included 124 collections, which allowed for an examination of population structure and hatchery influence across the study area and provided a robust tool for population assignment. Population assignment tests were conducted using the program RUBIAS.ResultPopulation assignment tests were successful for all pure WCT of unknown origin despite potential influence from hatchery lineages across the study area. Of 83 migratory WCT captured below AFD, approximately 80% were assigned to tributaries upstream of AFD with a posterior assignment probability of at least 90%. Only one fish was assigned to a tributary downstream of AFD.ConclusionOur results indicate that AFD disrupts the natural metapopulation dynamics of WCT populations in the basin. Passage for WCT at this barrier would reestablish metapopulation connectivity within the basin by allowing migratory individuals to make genetic contributions to populations upstream of the dam.

Genetics, geography, and subspecies status of American mink in Florida, with an emphasis on Neogale vison evergladensis

Historical records of American mink (Neogale vison) in Florida suggest they occur in four populations, each currently recognized as a separate subspecies. The Everglades mink (N. v. evergladensis) is a state-listed threatened subspecies, and the Atlantic salt marsh mink (N. v. lutensis) and Gulf salt marsh mink (N. v. halilimnetes) are both Species of Greatest Conservation Need, but support for their taxonomic status is limited. We acquired 62 samples of mink for genetic analysis using mtDNA and microsatellite data with the objective of assessing the subspecies status of mink in Florida with an emphasis on evergladensis. We also compared historical and current range estimates of mink subspecies in Florida using museum specimens, public sightings, and detection records from scientific studies. All estimates of FST were significant when evergladensis was paired with other subspecies and both assignment tests and the Principal Coordinate Analysis separated evergladensis from other subspecies. The single mtDNA haplotype from evergladensis was not found elsewhere in Florida but was present in a subspecies outside of Florida (vulgivaga). Genetic diversity was lowest in evergladensis and highest for vulgivaga from Louisiana. We found support for lutensis as a separate taxonomic unit and assignment tests found some support for the genetic distinctiveness of halilimnetes. Sample sizes limited analyses in halilimnetes and prevented any conclusions about the subspecies status of mink in the Panhandle. Based on current range estimates, mink from evergladensis, lutensis, and halilimnetes are geographically isolated from one another and supports the genetic analysis. Using a more modern definition of subspecies that includes genetics, distribution, and ecology, we conclude that the data supports multiple subspecies of mink in Florida.

Young teens at the age of 12-14 years old are in the pseudo-naturalism development phase of drawing. At the range of the respective ages they are already able to draw by having attention to details, understanding the concept of space, and producing drawings that look realistic. As a matter of fact, the eight graders’ drawing works at SMP Negeri 12 Kota Bima have not yet demonstrated the result of the expected target on that phase. Classroom action research was conducted to fnd out about the problem and to help students increase the quality of their drawing. The improvement of students’ learning achievement in drawing using multifunction puzzle media in the learning process was investigated in the current classroom action research. The subjects of the research were the eight graders of Class VIII-A in SMP Negeri 12 Kota Bima in the academic year of 2016/2017. Data were collected using assignment test method. The learning achievement in drawing increases quantitatively if students can achieve the minimum class average score of 75. The result of the research demonstrates that students’ scores increase significantly after experiencing the learning process using multifunction puzzle media. The average students’ score increased from 72.58 in cycle 1 to 78.33 in cycle 2

The weak genetic structure of Melolontha melolontha (L.) and Melolontha hippocastani (Fabr.), two important forest pests, indicates their large population sizes and effective gene flow

Abstract Cockchafers Melolontha spp. are polyphagous root pests and outbreak species of considerable economic importance to forestry in Europe. Knowledge of genetic diversity within and between cockchafer populations is largely unexplored but could lead to more effective monitoring and management strategies of these pests. In this study, we fill several gaps in the knowledge of the genetic population structure of Melolontha melolontha and Melolontha hippocastani. Using microsatellite loci and over 200 individuals per species, we examined the genetic structure of Melolontha spp. populations collected from three primary outbreak areas in central and southeastern Poland. We demonstrated that our focal species share some common genetic patterns, such as considerable genetic diversity within subpopulations (within sampling sites) with minor effects of past bottlenecks possibly masked by current population sizes. The estimates of effective population sizes, although subject to wide credibility intervals, indicated a large number of individuals contributing to further generations of the population. The migration rates exceed the threshold of one per generation required for gene flow and maintenance of population equilibrium, indicating limited impact from forest management practices on either cockchafer species. While genetic differentiation was observed in M. melolontha populations, supported by various indices (fixation index, principal coordinate analysis, assignment tests), the M. hippocastani populations were genetically similar. This disparity may hint at the potential historical divergence between these species. In this aspect, more detailed phylogeographic and population genetic studies covering a large area of the Palaearctic distributions of both species are required. Our findings constitute one of the initial efforts aimed at delineating the genetic diversity, differentiation, population structure, gene flow, and migration patterns within Melolontha spp. This exploration is especially significant for M. hippocastani, given its completely unexplored genetic population structure.

Assessing the genetic integrity of sugarcane germplasm in the USDA-ARS National Plant Germplasm System collection using single-dose SNP markers.

The World Collection of Sugarcane and Related Grasses, maintained at the USDA-ARS in Miami, FL, is one of the largest sugarcane germplasm repositories in the world. However, the genetic integrity of the Saccharum spp. germplasm in this collection has not been fully analyzed. In this study, we employed a single-dose SNP panel to genotype 901 sugarcane accessions, representing six Saccharum species and various hybrids. Our analysis uncovered a high rate of clone mislabeling in the collection. Specifically, we identified 86 groups of duplicates, characterized by identical SNP genotypes, which encompassed 211 accessions (23% of the total clones), while 135 groups, constituting 471 clones (52% of the total), exhibited near-identical genotypes. In addition, twenty-seven homonymous groups were detected, which shared the same clone name but differed in SNP genotypes. Hierarchical analysis of population structure partitioned the Saccharum germplasm into five clusters, corresponding to S. barberi, S. sinense, S. officinarum, S. spontaneum and S. robustum/S. edule. An assignment test, based on the five Saccharum species, enabled correcting 141 instances of mislabeled species memberships and inaccuracies. Moreover, we clarified the species membership and parentage of 298 clones that had ambiguous passport records (e.g., 'Saccharum spp', 'unknown', and 'hybrid'). Population structure and genetic diversity in these five species were further supported by Principal Coordinate Analysis and neighbor-joining clustering analysis. Analysis of Molecular Variance revealed that within-species genetic variations accounted for 85% of the total molecular variance, with the remaining 15% attributed to among-species genetic variations. The single-dose SNP markers developed in this study offer a robust tool for characterizing sugarcane germplasm worldwide. These findings have important implications for sugarcane genebank management, germplasm exchange, and crop genetic improvement.

Population-genetics analysis of the brown trout broodstock in the “Panjica” hatchery (Serbia) and its conservation applications

Artificial propagation and stocking of brown trout is a standard practice in recreational fishery management. In recent decades, the importance of maintaining intraspecific diversity and protecting locally adapted lineages has been recognized for the species' long-term survival. The first step in selecting donors for stocking involves distinguishing native trout from non-native and introgressed individuals. The established method for discerning Atlantic hatchery strains from the wild populations involves genetic screening of individual diagnostic SNPs and microsatellite assignment tests. This study, using Serbia's Panjica hatchery as an example, illustrates the proper conduct of routine genetic screening for identifying suitable donors for supportive stocking. The broodstock and reference populations were screened using mtDNA control region, LDH nuclear gene, and 12 microsatellite loci to assess the origin, diversity, and inbreeding levels. The analysis revealed only moderate contamination with Atlantic trout and showed the regional origin of the Danubian genes – over 50% of the broodstock was composed of non-introgressed Danubian individuals tracing their origin to the Zapadna Morava River system. Additionally, the study highlighted a considerable discordance between LDH locus and microsatellites in identifying introgressed individuals, raising concerns about the sole reliance on LDH locus for the identification of Atlantic genetic origin in nuclear DNA.

Genetic underpinning of historical afforestation with allochthonous Pinus cembra in the northwestern Swiss Alps

AbstractThroughout the last centuries, the structure and genetic composition of forests have been strongly affected by forest management. Over 30% of European forests are artificially regenerated, very often using translocated forest reproductive material, among these species the Swiss stone pine (Pinus cembra L.). In the late nineteenth and early twentieth centuries, the species was largely used for artificial afforestation in the northern Alps. However, only a few planted trees have survived. Therefore, we aimed to evaluate if the historical afforestation of P. cembra in the northwestern Swiss Alps relied on allochthonous material. We sampled 12 sites, genotyping 11 nuclear microsatellites, to infer the spatial genetic structure of regional populations, to test for genetic differences between natural and planted stands, and to infer potential source regions of planted stands using reference samples covering the entire Alps. Population genetic structure analysis allowed us to distinguish planted from natural stands and to determine that forest reproductive material used for plantations was not of regional origin. We found similar levels of genetic diversity between natural and planted stands. Assignment tests revealed that reproductive material for planting was translocated to the study area from two source regions, i.e., near the border of Switzerland and Austria, and further to the East, between Austria and Italy. Our study shows how genetic tools may inform about historical transfer of forest reproductive material, which still may affect the population genetic make-up of regional occurrences, e.g., because of reduced natural regeneration.

For the assessment of knowledge includes: (1) written tests in which there are multiple-choice tests, description tests, and false correct tests, (2) oral tests in which there are quizzes and questions and answers, (3) assignment tests in which there are group and individual tests both in school and outside of school, (4) portfolio and observation tests. For skills, the assessment includes (1) an assessment of practice in which there is reading, writing practice, and listening practice, (2) a project-based assessment in which there is interest and creativity, (3) a portfolio assessment, and (4) an Instrument assessment

Spread of the Zoonotic Nematode Baylisascaris procyonis into a Naive Raccoon Population

The raccoon roundworm (Baylisascaris procyonis), a gastrointestinal nematode of the raccoon (Procyon lotor), may cause a severe form of larva migrans in humans, which can lead to death or permanent neurological damage. Although roundworms were inadvertently introduced to Europe alongside their raccoon hosts, the parasite is not present in every raccoon population. It is important to understand the geographic distribution of B. procyonis, as early and rapid treatment can prevent severe pathologies in humans. We present evidence for the roundworm spreading into a naive raccoon population through natural dispersal of infected raccoons. We sampled 181 raccoons from Saxony-Anhalt, a German federal state containing contact zones of different raccoon populations, two of which were previously free of the parasite. We screened the raccoons for roundworms and used microsatellite-based assignment tests to determine the genetic origin of the raccoons and their parasites. We detected roundworms in 16 of 45 raccoons sampled in a previously roundworm-free area in the northern part of the state. The largest proportion of the genetic ancestry (≥ 0.5) of the 16 raccoon hosts was assigned to the previously naive raccoon population. Conversely, the genetic ancestry of almost all the roundworms was assigned to the nearest roundworm population in the southern part of the state. Infected raccoons have, therefore, spread to the north of the state, where they interbred with and infected local raccoons. It seems likely that the roundworms will continue to spread. Health authorities should consider continuous surveillance programmes of naive populations and raise public awareness.

Genetic insights into pepper-bark tree (Warburgia salutaris) reproduction in South Africa

Genetic diversity in over-exploited and threatened species has important, but often neglected, implications for conservation. In South Africa, the bark of the regionally endangered pepper-bark tree, Warburgia salutaris, is extensively harvested for subsistence medicine. We developed 11 polymorphic microsatellite markers to examine the genetic diversity and structure among 227 W. salutaris individuals sampled from across much of its range in South Africa’s Kruger National Park (KNP) and 31 trees growing in local nurseries. We also tested the cross-amplification success of these markers in two closely related species, Warburgia elongata (IUCN listed as Endangered) and W. stuhlmannii (IUCN listed as Vulnerable). Contrary to our expectations, we found evidence of clonality and for high genetic diversity in all wild KNP sub-populations of W. salutaris. We identified between 114 and 149 unique multi-locus genotypes in the 258 individuals included in our analyses. Bayesian assignment tests identified three distinct genetic clusters that are represented by the sampled KNP sub-population of W. salutaris, with minimal geographic structure and some admixture. Three genetic clusters were also identified among all sampled individuals, i.e., KNP and nursery trees. Nursery W. salutaris germplasm comprised unique genotypes with similar levels of observed heterozygosity to the wild population. High levels of clonality and reported self-incompatibility, along with short distance pollination, likely explain the lack of recent sexual recruitment (i.e., reduced fruit set and lack of seedlings) in the wild population. These findings will be useful to identify sub-populations where genets may be sampled to maximize genetic diversity within subpopulations for restoration throughout KNP.

Isolation and characterization of 17 polymorphic microsatellite loci with tri- and tetra-nucleotide repeat motifs for Ayu (Plecoglossus altivelis) using next-generation sequencing.

Ayu or sweetfish, Plecoglossus altivelis, an amphidromous fish ranging in the northwestern Pacific, is economically important inland fisheries and aquaculture resources. Genetic characterization of wild Ayu and derived culture seeds with competent molecular genetic markers is still insufficient for their sustainable use. Microsatellite DNA markers with larger repeat motifs (e.g. tri- and tetra-nucleotide motifs) are convenient and accurate compared with those having mono- and di-nucleotide motifs, but the latter motifs characterized most Ayu microsatellite markers developed previously. Here, we isolated and characterized 17 polymorphic microsatellite DNA markers with tri- and tetra-nucleotide repeat motif using next-generation sequencing. Alleles per locus varied from 6 to 23. The observed and expected heterozygosities ranged from 0.542 to1.000and 0.709to0.951, respectively. Polymorphic information content (PIC) of 15 out of the 17 loci were high (≧ 0.700), suggesting them to be highly informative. Twelve of the 17 loci were used for preliminary assignment test among three collections, and successfully allocated the examined fish to the original populations. The novel polymorphic microsatellite markers developed herein will be useful to examine the genetic diversity and population structure of wild Ayu and the effect of seed transplantation on native populations, providing a tool for conservation and sustainable adaptive management of this species.

Genetic structure of an endangered species Ormosia henryi in southern China, and implications for conservation

BackgroundThe evergreen broadleaved forest (EBLF) is an iconic vegetation type of East Asia, and it contributes fundamentally to biodiversity-based ecosystem functioning and services. However, the native habitat of EBLFs keeps on decreasing due to anthropogenic activities. Ormosia henryi is a valuable rare woody species in EBLFs that is particularly sensitive to habitat loss. In this study, ten natural populations of O. henryi in southern China were sampled, and then genotyping by sequencing (GBS) was applied to elucidate the standing genetic variation and population structure of this endangered species.ResultsIn ten O. henryi populations, 64,158 high-quality SNPs were generated by GBS. Based on these markers, a relatively low level of genetic diversity was found with the expected heterozygosity (He) ranging from 0.2371 to 0.2901. Pairwise FST between populations varied from 0.0213 to 0.1652, indicating a moderate level of genetic differentiation. However, contemporary gene flow between populations were rare. Assignment test and principal component analysis (PCA) both supported that O. henryi populations in southern China could be divided into four genetic groups, and prominent genetic admixture was found in those populations located in southern Jiangxi Province. Mantel tests and multiple matrix regression with randomization (MMRR) analyses suggested that isolation by distance (IBD) could be the possible reason for describing the current population genetic structure. In addition, the effective population size (Ne) of O. henryi was extremely small, and showed a continuous declining trend since the Last Glacial Period.ConclusionsOur results indicate that the endangered status of O. henryi is seriously underestimated. Artificial conservation measures should be applied as soon as possible to prevent O. henryi from the fate of extinction. Further studies are needed to elucidate the mechanism that leading to the continuous loss of genetic diversity in O. henryi and help to develop a better conservation strategy.

Efforts to achieve maximum Indonesian learning outcomes in the learning process must be in accordance with adequate situations and conditions, as well as appropriate learning strategies so that they can have a positive effect on the effectiveness of the learning process in achieving learning objectives, such as by using the application of teaching variations. This study uses a quantitative descriptive method. The tools and techniques for collecting data are assignment test techniques, while for analyzing the data used descriptive quantitative. Based on the results of data analysis, it was found that students who reached the excellent category were 6 people or 24%, those who reached the good category were 5 people or 20%, those who reached the moderate category were 11 people or 44%, and students who reached the less category were 3 people or 12%. The average value of the ability to develop main sentences into student paragraphs is 72.4%.

Reference Standards to Support Quality of Synthetic Peptide Therapeutics

PurposePeptides are an important class of therapeutics. Their quality is evaluated using a series of analytical tests, many of which depend on well-characterized reference standards to determine identity, purity, and strength.ObjectiveDiscuss approaches to producing peptide reference standards, including vialing, lyophilization, analytical testing and stability studies.MethodsCase studies are used to illustrate analytical approaches to characterize reference standards, including methods for value assignment, content uniformity, and identity testing. Methods described include NMR, mass spectrometry, and chromatography techniques for identity testing and HPLC and GC methods for assessing peptide content and impurities.ResultsThis report describes the analytical strategy used to establish peptide reference standard and illustrates how results from multiple labs are integrated to assign a value to the final lyophilized vial. A two-step process for value assignment is described, which uses a mass balance approach to assign a quantitative value to a bulk peptide material. The bulk material is then used as a standard to assign a final value to the vialed material. Testing to confirm peptide identity and to ensure consistency of the vialed material is also described. Considerations for addressing variability, identifying outliers, and implementing stability studies are also presented.ConclusionThe methods and case studies described provide a benchmark for best practices in establishing the preparation, analytical testing, handling, and storage of peptide reference standards for the pharmaceutical industry. Some peptide features, such as chiral or isobaric amino acids, may require additional techniques to ensure a full characterization of the peptide reference standard.
