Vitamin B-6 Status Indices Are Lower in Pregnant than in Nonpregnant Women but Urinary Excretion of 4-Pyridoxic Acid Does Not Differ1–4

The cause of reduced plasma pyridoxal-5'-phosphate (PLP) concentration and the validity of plasma PLP concentration as a vitamin B-6 status indicator during pregnancy are not well understood. In this study, pregnant and nonpregnant women consumed a controlled diet for 5 d to exclude dietary intake as a factor in altered vitamin B-6 metabolic utilization. Plasma PLP and pyridoxal (PL) concentrations were significantly lower and higher, respectively, and plasma PL+PLP concentration was significantly lower during pregnancy. When plasma PLP and PL concentrations were normalized based on plasma albumin concentration, the differences were less marked but were still significant. The erythrocyte aspartate aminotransferase activity coefficient was significantly greater in pregnant women which is a further indication of reduced vitamin B-6 concentrations in the blood. No difference was observed in the urinary excretion of 4-pyridoxic acid between the pregnant and nonpregnant women. Results of this study demonstrate that vitamin B-6 status indices are lower during pregnancy without differences in the urinary excretion of 4-pyridoxic acid.

The effects of pyridoxine deficiency and supplementation on hematological profiles, lymphocyte function, and hepatic cytochrome P450 in B6C3F1 mice

Pyridoxine, a B6 vitamin, is a co-factor in a variety of enzymatic reactions involved in intermediary metabolism. The effects of pyridoxine deficiency and supplementation on hematological profiles, lymphocyte function, and hepatic CYP1A1 and CYP2E1 were investigated in B6C3F1 mice fed a diet containing either 0 (i.e., pyridoxine-deficient diet, [PD]); or 7 mg pyridoxine-HCl/kg (i.e., control diet, [CD]) for 8 or 13 weeks followed by administration of 500 μg pyridoxine-HCl (IP) daily for either 2 (PD-S2 and CD-S2) or 3 (PD-S3 and CD-S3) consecutive days. Results demonstrated that erythrocyte aspartate aminotransferase activity coefficient (EAST-AC) values, which reflect host pyridoxine status, were significantly higher in PD mice than in CD mice, and dropped to control levels after supplementation. PD mice had significantly reduced weight gains, mean corpuscular volume (MCV), hemoglobin (HGB), and hematocrit (HCT) levels compared to CD mice after 8 and 13 weeks on the prescribed diet. In addition, PD mice had significantly lower circulating levels of total white blood cells, but higher red blood cell numbers after 8 weeks (compared to CD mice). Pyridoxine supplementation for 3 days restored HGB levels in PD mice to that of the unsupplemented CD controls; HCT, MCV and MCH levels were also increased in PD-S3 mice compared to their unsupplemented PD counterparts, but failed to reach comparable levels to those seen in mice fed a control diet. The pyridoxine-deficient diet also resulted in decreased mitogen stimulated T-lymphocyte proliferation after a 13-week feeding regimen and increased hepatic CYP1A1 activity that was reversed by pyridoxine supplementation. These studies demonstrate in a murine model that pyridoxine deficiency can cause multiple alterations that, in many cases, can be reversed by supplementation.

Vitamin B6status assessment in relation to dietary intake in high school students aged 16–18 years

The vitamin B6 status of high school students and its relationship with dietary intake were investigated in this cross-sectional study by face-to-face interview. A total of 157 healthy students aged 16-18 years (eighty-three boys and seventy-four girls) were randomly recruited from two out of nineteen senior high schools in Tainan, Taiwan. Vitamin B6 intakes were calculated from three 24-h dietary records. Direct and indirect vitamin B6 status indicators were measured in plasma, erythrocytes and urine. The anthropometric data, being similar to those of the first Nutrition and Health Survey in Taiwan (1993-1996), showed the normal growth and development of these students. All students except one girl (28.7 nmol/l) had plasma pyridoxal-5'-phosphate (PLP) levels >35 nmol/l, indicating an adequate vitamin B6 status. The mean dietary vitamin B6 intakes of boys and girls were 1.04 (sd 0.29) and 0.96 (sd 0.27) mg/d, respectively. Vitamin B6 status indicators, including plasma PLP, erythrocyte alanine aminotransferase activity coefficient (EALT-AC), aspartate aminotransferase activity coefficient (EAST-AC) and urinary 4-pyridoxic acid (4-PA), were correlated with vitamin B6 intake. Students with adequate values of plasma PLP (>35 nmol/l), EALT-AC ( < 1.25), EAST-AC ( < 1.8) and urinary 4-PA (>3.0 micromol/d) had median intakes of 1.08 and 1.01 mg/d, respectively, for boys and girls. This study suggests that vitamin B6 requirements for boys and girls aged 16-18 years were approximately 1.1 and 1.0 mg/d, respectively.

Maternal plasma pyridoxal‐5′‐phosphate concentrations and risk of isolated oral clefts in the Philippines

We report that inadequate vitamin B-6 status of Filipino mothers, assessed by erythrocyte aspartate aminotransferase activity coefficient (EAST-AC), is associated with an increased risk for isolated cleft lip with or without cleft palate (CL/P) in their children. Its association with the status assessed by plasma pyridoxal-5'-phosphate (PLP) concentrations is unknown. In a case-control study in the Philippines including 46 cases (mothers of a child with CL/P) and 392 controls (mothers of an unaffected child), we evaluated the association between the risk for CL/P and maternal vitamin B-6 status assessed by PLP and EAST-AC. The ORs of CL/P were estimated by classifying mothers by PLP (>30, 20-30, and <20 nmol/L). Using the highest PLP group as the reference, ORs (95% CIs) were 1.03 (0.45-2.37) and 2.66 (1.30-5.50) for the middle and lowest groups, respectively (p trend = .01). In multivariate models controlling for various covariates including folate, the risk for CL/P was approximately 12 times higher in mothers with inadequate vitamin B-6 status, assessed by both PLP and EAST-AC values, compared to those with adequate status by both values. Inadequate vitamin B-6 status assessed by maternal PLP and EAST-AC values independently and both combined was associated with an increased risk for CL/P. The association was highest when both values were considered, suggesting that the measurement of both PLP and EAST-AC provides better assessment of vitamin B-6 status than either measurement alone.

Dietary intake of vitamin B6and concentration of vitamin B6in blood samples of German vegans

The study aimed to evaluate the dietary vitamin B6 intake and determine the vitamin B6 concentration in blood samples of German vegans. Cross-sectional study with 33 examination sites all over Germany. Subjects Ninety-three vegans (50 females) with a mean (+/- standard deviation (SD)) age of 43.7 +/- 15.7 years who took no vitamin supplements. Dietary intake was assed using a semi-quantitative food-frequency questionnaire. Erythrocyte aspartate aminotransferase activity coefficient (EAST-AC) was calculated as the ratio of stimulated (pyridoxal 5'-phosphate added) to unstimulated activity in blood samples that were provided after an overnight fast. Mean +/- SD vitamin B6 intake was 2.83 +/- 0.98 mg day(-1) and mean +/- SD protein intake was 56.6 +/- 21.7 g day(-1). Of the participants 4% showed vitamin B6 intakes lower than daily recommended intakes for Germany, 16% showed EAST-AC > 1.85, and a further 58% showed EAST-AC of 1.5-1-85. Moderate vegans were affected to a lesser extent than strict vegans. None of the established confounders was a significant predictor of EAST-AC. In logistic regression analyses the contribution of nutriments and cereals to pyridoxine intake was the only predictor of EAST-AC classified as < or = 1.85 and > 1.85, respectively. In spite of the high total intake of vitamin B6, an adequate concentration in blood samples could not be realised for a majority of the participants. Due to the health implications of a marginal pyridoxine status, vegans should be encouraged to include foods with a high bioavailability of pyridoxine, such as beans, lentils and bananas, in the daily diet.

Pyridoxine supplementation corrects vitamin B6 deficiency but does not improve inflammation in patients with rheumatoid arthritis

Patients with rheumatoid arthritis have subnormal vitamin B6 status, both quantitatively and functionally. Abnormal vitamin B6 status in rheumatoid arthritis has been associated with spontaneous tumor necrosis factor (TNF)-α production and markers of inflammation, including C-reactive protein and erythrocyte sedimentation rate. Impaired vitamin B6 status could be a result of inflammation, and these patients may have higher demand for vitamin B6. The aim of this study was to determine if daily supplementation with 50 mg of pyridoxine for 30 days can correct the static and/or the functional abnormalities of vitamin B6 status seen in patients with rheumatoid arthritis, and further investigate if pyridoxine supplementation has any effects on the pro-inflammatory cytokine TNF-α or IL-6 production of arthritis. This was a double-blinded, placebo-controlled study involving patients with rheumatoid arthritis with plasma pyridoxal 5'-phosphate below the 25th percentile of the Framingham Heart Cohort Study. Vitamin B6 status was assessed via plasma and erythrocyte pyridoxal 5'-phosphate concentrations, the erythrocyte aspartate aminotransferase activity coefficient (αEAST), net homocysteine increase in response to a methionine load test (ΔtHcy), and 24 h urinary xanthurenic acid (XA) excretion in response to a tryptophan load test. Urinary 4-pyridoxic acid (4-PA) was measured to examine the impact of pyridoxine treatment on vitamin B6 excretion in these patients. Pro-inflammatory cytokine (TNF-α and IL-6) production, C-reactive protein levels and the erythrocyte sedimentation rate before and after supplementation were also examined. Pyridoxine supplementation significantly improved plasma and erythrocyte pyridoxal 5'-phosphate concentrations, erythrocyte αEAST, urinary 4-PA, and XA excretion. These improvements were apparent regardless of baseline B6 levels. Pyridoxine supplementation also showed a trend (p < 0.09) towards a reduction in post-methionine load ΔtHcy. Supplementation did not affect pro-inflammatory cytokine production. Although pyridoxine supplementation did not suppress pro-inflammatory cytokine production in patients with rheumatoid arthritis, the suboptimal vitamin B6 status seen in rheumatoid arthritis can be corrected by 50 mg pyridoxine supplementation for 30 days. Data from the present study suggest that patients with rheumatoid arthritis may have higher requirements for vitamin B6 than those in a normal healthy population.

Assessment of Vitamin B-6 Estimated Average Requirement and Recommended Dietary Allowance for Adolescents Aged 13–15 Years Using Vitamin B-6 Intake, Nutritional Status and Anthropometry

Because of limited available information, the Estimated Average Requirement (EAR) and Recommended Dietary Allowance (RDA) of vitamin B-6 for adolescents were recently estimated by extrapolation from adult values. To determine vitamin B-6 requirements and to provide recommendations for intakes, vitamin B-6 intake, nutritional status and anthropometry were studied in 134 healthy adolescents (63 boys and 71 girls) aged 13-15 y in Tainan, Taiwan. Direct and indirect vitamin B-6 indicators were measured in plasma, erythrocytes and urine. The anthropometric data of the adolescents in this study were similar to those of the first Nutrition and Health Survey in Taiwan (NAHSIT), conducted from 1993 to 1996, showing the normal growth and development of this adolescent group. All subjects had plasma pyridoxal-5'-phosphate (PLP) concentrations > or = 20 nmol/L, indicating an adequate vitamin B-6 status. The mean dietary vitamin B-6 intakes of boys and girls were 1.04 +/- 0.24 and 0.83 +/- 0.26 mg/d, respectively. Vitamin B-6 status indicators, including plasma PLP, erythrocyte alanine activity coefficient (EALT-AC), aspartate aminotransferase activity coefficient (EAST-AC) and urinary 4-pyridoxic acid (4-PA), were correlated with vitamin B-6 intake (r = 0.84, -0.84, -0.77 and 0.86, respectively, P < 0.01). Adequate values of plasma PLP (> or = 20 nmol/L), EALT-AC (<1.25), EAST-AC (<1.8) and urinary 4-PA (>3.0 micromol/d) were used to determine the EAR according to the Dietary Reference Intake committee methodology. The present study suggests that vitamin B-6 EAR (RDA) for adolescent boys and girls aged 13-15 y are 1.07 (1.28) and 0.90 (1.08) mg/d, respectively.

Vitamins B6 and B12 and folate status in an adult Mediterranean population.

To assess the nutritional status for vitamins B(6) and B(12) and folate in an adult Mediterranean population, in order to identify patterns of intake, groups at risk for deficiency, and factors that might influence this risk. A cross-sectional epidemiological survey. Andalusia, a western Mediterranean region in southern Spain. The study was carried out with a random sample of 3528 subjects (1813 men, 1715 women) who were between 25 and 60 y of age. Blood samples were obtained for biochemical assays in a random subsample of 384 subjects (183 men, 201 women). Food consumption was assessed by 48-h recall. Vitamin B(6) was measured as alpha erythrocyte aspartate aminotransferase activation coefficient (alphaEAST); vitamin B(12) and folate concentrations were measured in plasma. Energy and vitamin intakes were significantly higher in men than in women. In men, intakes were below two-thirds of the RDA in 10.8, 2.9 and 22.6% for B(6), B(12) and folate, respectively. The corresponding figures in women were 16.7, 5.1 and 23.5% for vitamins B(6), B(12) and folate. Age, place of residence and educational level, alcohol use and smoking were also associated with differences in the intake of these nutrients. Biochemical analyses showed that vitamins B(6), B(12) and folate status was acceptable in 75.7, 89.1 and 57.6% of the population, respectively. Plasma concentration of folate was significantly higher in women. Our results provide a precise estimate of the nutritional status for vitamins B(6), B(12) and folate in the adult population of southern Spain. Factors such as age, place of residence, level of education and smoking can increase the risk of inadequate intake of some nutrients. Plasma Pyridoxal 5′-Phosphate Concentration Is Correlated with Functional Vitamin B-6 Indices in Patients with Rheumatoid Arthritis and Marginal Vitamin B-6 Status

Many patients with rheumatoid arthritis (RA) have low plasma pyridoxal-phosphate (PLP) but a normal erythrocyte aspartate aminotransferase activity coefficient (alpha EAST), a measure of vitamin B-6 status in the erythrocytes, compared with healthy subjects. The goal of the present study was to examine the correlations of PLP levels in these two compartments (plasma and erythrocytes) with other established indices of vitamin B-6 status, and to determine which indicator better reflects functional status of vitamin B-6 in patients with RA. Multiple indices of vitamin B-6 status were measured in 33 patients with RA. Plasma PLP, urinary 4-pyridoxic acid (4-PA), net increase in plasma total homocysteine after a methionine load (DeltatHcy) and net increase in urinary xanthurenic acid after a tryptophan load (DeltaXA) were log-transformed to reach normality for statistical analyses. We found that log-plasma PLP levels were inversely correlated with both log-DeltatHcy (r = -0.368, P = 0.035) and log-DeltaXA (r = -0.333, P = 0.05). Plasma PLP was not correlated with alpha EAST or urinary 4-PA excretion. In contrast, erythrocyte PLP was inversely correlated with alpha EAST (r = -0.431, P = 0.012) and positively correlated with log-4-PA (r = 0.475, P = 0.005), but erythrocyte PLP was not correlated with the outcomes of a methionine or tryptophan load test. Erythrocyte PLP and log-4-PA, but not plasma PLP, were correlated with dietary intake of vitamin B-6 after adjusting for protein intake (r = 0.420, P = 0.015 and r = 0.333, P = 0.05, respectively). We suggest that in patients with RA, plasma PLP levels are a better diagnostic indicator of functional vitamin B-6 status than erythrocyte PLP levels.

Determination of Vitamin B-6 Estimated Average Requirement and Recommended Dietary Allowance for Children Aged 7–12 Years Using Vitamin B-6 Intake, Nutritional Status and Anthropometry

The Estimated Average Requirement (EAR) and Recommended Dietary Allowance (RDA) of vitamin B-6 for children were recently estimated by extrapolating from adult values because of limited available information. To determine vitamin B-6 requirements and provide recommendations for intakes, vitamin B-6 intake, nutritional status and anthropometry of 168 healthy children (79 boys and 89 girls) were studied in Tainan, Taiwan. Direct and indirect vitamin B-6 status indicators were measured in plasma, erythrocytes and urine. Anthropometric data of children in this study were similar to those of the first Nutrition and Health Survey in Taiwan (NAHSIT) conducted in 1993-1996. The plasma pyridoxal phosphate (PLP) concentration of each child was >/=30 nmol/L, indicating an adequate vitamin B-6 status. Daily dietary vitamin B-6 intakes of boys and girls were 0.80 +/- 0.16 and 0.74 +/- 0.16 mg/d, respectively. Daily dietary vitamin B-6 intakes of children who had adequate urinary 4-pyridoxic acid (4-PA) (>3.0 micro mol/L), erythrocyte alanine aminotransferase activity coefficient (EALT-AC) (<1.25) and aspartate aminotransferase activity coefficient (EAST-AC) (<1.8) were not different from those of children who had adequate plasma PLP, although the percentages of adequacy for urinary 4-PA, EALT-AC and EAST-AC ranged from 20 to 91%. Vitamin B-6 status indicators were strongly correlated with vitamin B-6 intake. Adequate values of PLP, EALT-AC, EAST-AC and urinary 4-PA were used to determine the EAR according to Dietary Reference Intake (DRI) committee methodology. We determined the vitamin B-6 EAR (RDA) for boys and girls aged 7-12 y to be 0.84 (1.01) and 0.75 (0.89) mg/d, respectively.

Low-dose vitamin B-6 effectively lowers fasting plasma homocysteine in healthy elderly persons who are folate and riboflavin replete

Current data suggest that physiologic doses of vitamin B-6 have no significant homocysteine-lowering effect. It is possible that an effect of vitamin B-6 was missed in previous trials because of a much greater effect of folic acid, vitamin B-12, or both. The aim of this study was to investigate the effect of low-dose vitamin B-6 supplementation on fasting total homocysteine (tHcy) concentrations in healthy elderly persons who were made replete with folate and riboflavin. Twenty-two healthy elderly persons aged 63-80 y were supplemented with a low dose of vitamin B-6 (1.6 mg/d) for 12 wk in a randomized, double-blind, placebo-controlled trial after repletion with folic acid (400 microg/d for 6 wk) and riboflavin (1.6 mg/d for 18 wk); none of the subjects had a vitamin B-12 deficiency. Folic acid supplementation lowered fasting tHcy by 19.6% (P < 0.001). After folic acid supplementation, baseline tHcy concentrations ranged from 6.22 to 23.52 micromol/L and 10 subjects had suboptimal vitamin B-6 status (plasma pyridoxal-P < 20 nmol/L). Two-way analysis of variance showed that the significant improvement in vitamin B-6 status in response to vitamin B-6 supplementation (on the basis of both pyridoxal-P: and the erythrocyte aspartate aminotransferase activation coefficient) was reflected in a significant reduction in plasma tHcy of 7.5%. Low-dose vitamin B-6 effectively lowers fasting plasma tHcy in healthy subjects who are both folate and riboflavin replete. This suggests that any program aimed at the treatment or prevention of hyperhomocysteinemia should include vitamin B-6 supplementation.

Vitamin B6 and vitamin C status in elderly patients with infections during hospitalization.

We have evaluated whether vitamin B6 and C metabolism may be altered by infection in the elderly. Vitamin B6 and C biochemical status has been assessed for times over a period of 21 days (days 0, 7, 14, and 21) in 18 subjects > or = 75 years. The subjects were divided into 3 groups: group I (8 subjects with acute infection), group II (4 malnourished subjects), and group III (6 control subjects). Vitamin B6 status was determined by plasma pyridoxal-5'-phosphate (PLP) and erythrocyte aspartate aminotransferase activation coefficient (alpha-EAST), and vitamin C status by plasma ascorbic acid. During the 3 weeks, vitamins B6 and C values were significantly different between groups: at days 7 and 14, PLP values were significantly higher in group III than in both groups I and II, and alpha-EAST values were significantly higher in group I than in both groups II and III. Plasma ascorbate values were significantly lower in group I than in both groups II and III. These data suggest that an acute catabolic state like infection may influence vitamin B6 and C metabolism. Nevertheless, more work is needed to assert that vitamin B6 and C supplementation may be useful during infection.

Tryptophan metabolism and vitamin B6 nutritional status in patients with schistosomiasis mansoni and in infected mice.

Patients infected with Schistosoma mansoni showed an abnormal response to a test dose of tryptophan, with little increase in the urinary excretion of kynurenine, hydroxykynurenine, xanthurenic and kynurenic acids, N1-methyl nicotinamide, methyl pyridone carboxamide, 5-hydroxytryptamine or 5-hydroxyindoleacetic acid. In contrast to previous reports, this is different from the pattern of tryptophan metabolism seen in vitamin B6 deficiency. Furthermore, the patients' plasma concentrations of pyridoxal phosphate were within the reference range, and supplementation for 5 days with 20 mg vitamin B6/day did not affect tryptophan metabolism. Treatment with a single dose of Praziquantel resulted in a substantial restoration of normal tryptophan metabolism. In mice infected with S. mansoni there was a similar impairment of tryptophan metabolism, as shown by considerably reduced formation of 14CO2 after the administration of a tracer dose of [14C]tryptophan. Again, the administration of vitamin B6 supplements did not correct tryptophan metabolism in the mice. Treatment with Praziquantel resulted in substantial restoration of the production of 14CO2 from [14C]tryptophan. There was no evidence of vitamin B6 deficiency (as determined by erythrocyte aspartate aminotransferase activation coefficient) associated with infection in the mice, although there was a redistribution of pyridoxal phosphate between tissues, with a reduction in the concentration of liver, spleen and kidney, and an increase in skeletal muscle.

Vitamin B-6 Requirements of Elderly Men and Women

The vitamin B-6 requirements of 12 men and women over 60 y old were studied. The protocol consisted of a 5-d baseline period and four experimental periods during which the subjects successively received 0.003, 0.015, 0.0225 and 0.03375 mg of vitamin B-6/(kg body wt.d). Dietary protein was 1.2 or 0.8 g/(kg body wt.d). At 5- or 6-d intervals, xanthurenic acid (XA) after a 5-g L-tryptophan load and 4-pyridoxic acid (4-PA) in 24-h urine, erythrocyte aspartate aminotransferase activity coefficient (EAST-AC) and plasma pyridoxal-5'-phosphate (PLP) were measured. These measurements were abnormal during vitamin B-6 depletion but returned to normal during repletion. Men who ingested approximately 120 g protein/d required 1.96 +/- 0.11 mg of vitamin B-6 to normalize XA; women who ingested 78 g protein/d required 1.90 +/- 0.18 mg of vitamin B-6 to normalize XA. To attain normal levels of EAST-AC and 4-PA in men, 2.88 +/- 0.17 mg of vitamin B-6 were needed; to normalize PLP, 1.96 +/- 0.11 mg of vitamin B-6 were required. Women required 1.90 +/- 0.18 mg or more of vitamin B-6 to normalize these measurements. Vitamin B-6 requirements were not decreased in two of three subjects who ingested 54 g of protein daily. Thus, vitamin B-6 requirements of elderly men and women are about 1.96 and 1.90 mg/d, respectively.

Hemolysate activation coefficient and its relationship to holoenzyme levels of aspartate aminotransefrase in rabbit erythrocytes

The relationship between the aspartate aminotransferase (AST) activation coefficient and the intracellular in situ proportion of the pyridoxal 5′-phosphate (PLP) form of AST (PLP-AST) in total AST was determined. Erythrocytes from Japanese-White strain rabbits had an activation coefficient of 1.85. This indicated that 54.1% of the AST in the hemolysates from these erythrocytes was the holoenzyme. Sodium borohydride reduction showed that 64.8% of the total erythrocyte AST was present as the PLP-AST holoenzyme. Increases in intracellular PLP and pyridoxamine 5′-phosphate (PMP) concentrations, generated by preincubation of the erythrocytes with pyridoxal (PL) and pyridoxamine (PM), significantly decreased the values of the activation coefficient. However, these increases did not significantly increase the intracellular proportion of PLP-AST in total AST. These results showed that the AST activation coefficient reflects the saturation level of AST in the hemolysate rather than the intracellular in situ level of the holo-form of AST (holoAST). Thus, establishment of standard conditions is required for use of the activation coefficient, which is inherently a valuable biochemical marker of the vitamin B-6 status, for accurate deduction of the nutritional status of vitamin B-6 in test organisms.

Dependence of Vitamin B-6 Status Assessment on Alcohol Intake Among Elderly Men and Women (Dutch Nutrition Surveillance System)

To obtain more insight into the effect of moderate alcohol intake on vitamin B-6 status indicators, we studied the associations of alcohol intake (unadjusted and adjusted for intake of vitamin B-6 and protein) with the erythrocyte aspartate aminotransferase activation coefficient (EAST-AC) and plasma pyridoxal 5'-phosphate (PLP) level. Data obtained from men (n = 224) and women (n = 217) aged 65-79 (nationwide sample in the Netherlands) were used for this purpose. Although alcohol intake (a maximum of 21% of the energy came from alcohol) tended to be positively associated with PLP, this association never reached statistical significance (p greater than or equal to 0.05). EAST-AC was inversely associated with alcohol intake, whether or not it was adjusted for vitamin B-6 and protein intake. Similar results were found for the total EAST activity (after adding PLP) or apoenzyme activity; the basal EAST activity (before adding PLP) or holoenzyme activity was not associated with the alcohol intake. These results indicate that caution is needed in the interpretation of the specificity of EAST-AC (i.e., the degree to which EAST-AC is unaffected by other factors) as an indicator of vitamin B-6 intake.

Schistosoma mansoni: Effects on tryptophan metabolism in mice

In mice, infection with 20–30 cercariae of Schistosoma mansoni resulted in a considerable reduction in the formation of 14CO 2 from [ 14C]tryptophan. Infected animals excreted significantly lower amounts of kynurenine, kynurenic acid, and methyl pyridone carboxamide than did uninfected controls. There was no difference in the ability of hepatocytes isolated from infected or control animals to metabolise [ 14C]tryptophan. Hepatocytes from infected animals synthesized less NAD(P), but more niacin and N 1-methyl nicotinamide from tryptophan. They showed no greater accumulation of kynurenine metabolites than did cells from control animals. The hepatocyte content of pyridoxal phosphate and the erythrocyte aspartate aminotransferase activation coefficient were the same in both groups of mice, suggesting that infection with S. mansoni does not deplete vitamin B 6. The impairment of tryptophan metabolism in vivo was apparently not due to impaired hepatic metabolism. Rather, it seems likely that the parasites or their eggs take up tryptophan avidly from the host's circulation. Studies of parasite and egg metabolism of tryptophan may suggest novel approaches to the chemotherapy Of bilharzia.

Evaluation of the Erythrocyte Aspartate Aminotransferase Activity Coefficient as an Indicator of the Vitamin B-6 Status of Postpubertal Gilts

In two experiments, postpubertal gilts, individually housed in gestation stalls, were fed daily 1.9 kg of a purified diet providing 0.45, 1.5, 2.1 or 83 mg of vitamin B-6/d. The erythrocyte aspartate aminotransferase (more commonly known as glutamic-oxaloacetic transaminase) activity coefficient (EGOT-Ac) of gilts fed the low vitamin B-6 diet (0.45 mg vitamin B-6/d) for 121 d increased 200%, while the EGOT-Ac of gilts fed the high vitamin B-6 diet (83 mg vitamin B-6/d) remained unchanged. Two gilts were fed the low vitamin B-6 diet for 241 d at which time hair loss was observed. Muscle glutamic-oxaloacetic transaminase activity was reduced 75% in vitamin B-6—deficient gilts. The EGOT-Ac of gilts consuming 2.1 mg vitamin B-6/d was 100% greater than that of control animals (83 mg vitamin B-6/d). These findings indicate that the current NRC (27) estimated vitamin B-6 requirement for mature swine (1.8 mg vitamin B-6/d) is inadequate. Prior intake of excess vitamin B-6 did not prevent the subsequent development of a vitamin B-6 deficiency.
