Regulation of crucial enzymes and transcription factors on 2-phenylethanol biosynthesis via Ehrlich pathway in Saccharomyces cerevisiae.

2-Phenylethanol (2-PE) is widely used in food, perfume and pharmaceutical industry, but lower production in microbes and less known regulatory mechanisms of 2-PE make further study necessary. In this study, crucial genes like ARO8 and ARO10 of Ehrlich pathway for 2-PE synthesis and key transcription factor ARO80 in Saccharomyces cerevisiae were re-regulated using constitutive promoter; in the meantime, the effect of nitrogen source in synthetic complete (SC) medium with L-phenylalanine (L-Phe) on Aro8/Aro9 and Aro10 was investigated. The results showed that aromatic aminotransferase activities of ARO8 over-expressing strains were seriously inhibited by ammonia sulfate in SC+Phe medium. Flask fermentation test demonstrated that over-expressing ARO8 or ARO10 led to about 42% increase in 2-PE production when compared with the control strain. Furthermore, influence of transcription factors Cat8 and Mig1 on 2-PE biosynthesis was explored. CAT8 over-expression or MIG1 deletion increased in the transcription of ARO9 and ARO10. 2-PE production of CAT8 over-expressing strain was 62% higher than that of control strain. Deletion of MIG1 also led to 2-PE biosynthesis enhancement. The strain of CAT8 over-expression and MIG1 deletion was most effective in regulating expression of ARO9 and ARO10. Analysis of mRNA levels and enzyme activities indicates that transaminase in Ehrlich pathway is the crucial target of Nitrogen Catabolize Repression (NCR). Among the engineering strains, the higher 3.73g/L 2-PE production in CAT8 over-expressing strain without in situ product recovery suggests that the robust strain has potentiality for commercial exploitation.

Aspartate aminotransferase activity is required for aspartate catabolism and symbiotic nitrogen fixation in Rhizobium meliloti

A mutant of Rhizobium meliloti, 4R3, which is unable to grow on aspartate has been isolated. The defect is specific to aspartate utilization, since 4R3 is not an auxotroph and grows as well as its parent strain on other carbon and nitrogen sources. The defect was correlated with an inability to fix nitrogen within nodules formed on alfalfa. Transport of aspartate into the mutant cells was found to be normal. Analysis of enzymes involved in aspartate catabolism showed a significantly lower level of aspartate aminotransferase activity in cell extracts of 4R3 than in the wild type. Two unrelated regions identified from a genomic cosmid bank each complemented the aspartate catabolism and symbiotic defects in 4R3. One of the cosmids was found to encode an aspartate aminotransferase enzyme and resulted in restoration of aspartate aminotransferase activity in the mutant. Analysis of the region cloned in this cosmid by transposon mutagenesis showed that mutations within this region generate the original mutant phenotypes. The second type of cosmid was found to encode an aromatic aminotransferase enzyme and resulted in highly elevated levels of aromatic aminotransferase activity. This enzyme apparently compensated for the mutation by its ability to partially utilize aspartate as a substrate. These findings demonstrate that R. meliloti contains an aspartate aminotransferase activity required for symbiotic nitrogen fixation and implicate aspartate as an essential substrate for bacteria in the nodule.

Aromatic aminotransferase activity and indoleacetic acid production in Rhizobium meliloti

Bacterial indoleacetic acid (IAA) production, which has been proposed to play a role in the Rhizobium-legume symbiosis, is a poorly understood process. Previous data have suggested that IAA biosynthesis in Rhizobium meliloti can occur through an indolepyruvate intermediate derived from tryptophan by an aminotransferase activity. To further examine this biosynthetic pathway, the aromatic aminotransferase (AAT) activity of Rhizobium meliloti 102F34 (F34) was characterized. At least four proteins were detected on nondenaturing gels of F34 protein extracts that exhibited AAT activity. All four of these AATs were constitutively produced and utilized the aromatic amino acids tryptophan, phenylalanine, and tyrosine as amino substrates. Two AATs were also capable of using aspartate. Plasmids from an F34 gene bank were identified that coded for the synthesis of at least three of these proteins, and the respective gene sequences were localized by transposon mutagenesis. Selected transposon insertions were recombined into the F34 genome to produce strains defective in two of these proteins (AAT1 and AAT2). Characterization of the mutants revealed that neither was essential for the biosynthesis of IAA in the absence of exogenous tryptophan, but that both contributed to IAA biosynthesis when high levels of exogenous tryptophan were present. AAT1 and AAT2 were also not required for the production of a minimal level of aromatic amino acids, but both were able to scavenge nitrogen from the aromatic amino acids during nitrogen deprivation. Neither AAT1 nor AAT2 was essential for symbiosis with alfalfa.

Degradation of aromatic amino acids in Candida maltosa

AbstractThe degradation of l‐ and d‐aromatic amino acids has been studied in Candida maltosa. The following main metabolites were indentified: indolelactate was the degradation product of tryptophan, phenylpyruvate and phenyllactate were that of phenylalanine and p‐hydroxyphenylpyruvate and p‐hydroxyphenyllactate resulted from degradation of tyrosine. The first step of utilization of l‐aromatic amino acids was found to be a transamination. The cells contained increased aromatic aminotransferase activity if grown on medium supplemented with one of the three l‐aromatic amino acids. Three aromatic aminotransferases were separated by DEAE‐cellulose chromatography. The pathway for degradation of d‐aromatic amino acids involves deamination as the initial step. The enzyme was characterized to be a general d‐amino acid oxidase capable of utilizing various d‐amino acids. Synthesis of this enzyme was constitutive. Common products of both enzymatic reactions with the aromatic amino acids were the corresponding aromatic pyruvates. Formation of the aromatic lactate derivatives was catalyzed by an aromatic lactate dehydrogenase. This enzyme possessed a high substrate specificity for p‐hydroxyphenylpyruvate, phenylpyruvate, and indolepyruvate and was synthesized constitutively.

Novel features of prephenate aminotransferase from cell cultures of Nicotiana silvestris

A prephenate aminotransferase enzyme that produces l-arogenate was demonstrated in extracts from cultured-cell populations of Nicotiana silvestris. The enzyme was very active with low concentrations of prephenate, but required high concentrations of phenylpyruvate or 4-hydroxyphenylpyruvate to produce activity levels that were detectable. It is the most specific prephenate aminotransferase described to date from any source. Only l-glutamate and l-aspartate were effective amino-donor substrates. Prephenate concentrations greater than 1 m m produced substrate inhibition, an effect antagonized by increasing concentrations of l-glutamate cosubstrate. The enzyme was stable to storage for at least a month in the presence of pyridoxal 5′-phosphate, EDTA, and glycerol, and exhibited an unusually high temperature optimum of 70 °C. The identity of l-arogenate formed during catalysis was verified by high-performance liquid chromatography. DEAE-cellulose chromatography revealed two aromatic aminotransferase activities that were distinct from prephenate aminotransferase and which did not require the three protectants for stability. The aromatic aminotransferases were active with phenylpyruvate or 4-hydroxyphenylpyruvate as substrates, but not with prephenate. Both of the latter enzymes were similar in substrate specificity, and each exhibited a temperature optimum of 50 °C for catalysis. The primary in vivo function of the two aromatic aminotransferases is probably to transaminate between the aspartate/2-ketoglutarate and glutamate/oxaloacetate couples, since activities with the latter substrate combinations were an order of magnitude greater than with aromatic substrates. The demonstrated existence of a specific prephenate aminotransferase in N. silvestris meshes with other evidence supporting an important role for l-arogenate in tyrosine and phenylalanine biosynthesis in higher plants.

Tryptophan metabolism in Klebsiella aerogenes: regulation of the utilization of aromatic amino acids as sources of nitrogen.

Klebsiella aerogenes utilized aromatic amino acids as sole sources of nitrogen but not as sole sources of carbon. K. aerogenes abstracted the alpha-amino group of these compounds by transamination and excreted the arylpyruvate portions into the medium. When tryptophan was utilized as the sole source of nitrogen by K. aerogenes, indolepyruvate was excreted into the medium, where it polymerized non-enzymatically to form a brick red pigment. At least four separate aromatic aminotransferase activities were found in K. aerogenes. One activity (aromatic aminotransferase I) appeared to be solely responsible for the aminotransferase reaction necessary for the growth of K. aerogenes when tryptophan was the source of nitrogen; the loss of this activity by mutation (tut) prevented the growth of cells on media containing this and other aromatic amino acids. None of the other aminotransferase activities in the cells could substitute for aromatic aminotransferase in this regard. Tryptophan-dependent pigment formation in K. aerogenes was positively controlled by the intracellular level of glutamine synthetase. Nevertheless, the aromatic aminotransferase activity in cells varied less than 2-fold in response to 10-fold or greater changes in the levels of glutamine synthetase. Glutamine synthetase affected the ability of the cells to take up tryptophan from the medium.

Enzymology of l-Tyrosine Biosynthesis in Mung Bean (Vigna radiata [L.] Wilczek)

The enzymes of the 4-hydroxyphenylpyruvate (prephenate dehydrogenase and 4-hydroxyphenylpyruvate aminotransferase) and pretyrosine (prephenate aminotransferase and pretyrosine dehydrogenase) pathways of l-tyrosine biosynthesis were partially purified from mung bean (Vigna radiata [L.] Wilczek) seedlings. NADP-dependent prephenate dehydrogenase and pretyrosine dehydrogenase activities coeluted from ion exchange, adsorption, and gel-filtration columns, suggesting that a single protein (52,000 daltons) catalyzes both reactions. The ratio of the activities of partially purified prephenate to pretyrosine dehydrogenase was constant during all purification steps as well as after partial inactivation caused by p-hydroxymercuribenzoic acid or heat. The activity of prephenate dehydrogenase, but not of pretyrosine dehydrogenase, was inhibited by l-tyrosine at nonsaturating levels of substrate. The K(m) values for prephenate and pretyrosine were similar, but the specific activity with prephenate was 2.9 times greater than with pretyrosine.Two peaks of aromatic aminotransferase activity utilizing l-glutamate or l-aspartate as amino donors and 4-hydroxyphenylpyruvate, phenylpyruvate, and/or prephenate as keto acid substrates were eluted from DEAE-cellulose. Of the three keto acid substrates, 4-hydroxyphenylpyruvate was preferentially utilized by 4-hydroxyphenylpyruvate aminotransferase whereas prephenate was best utilized by prephenate aminotransferase. The identity of a product of prephenate aminotransferase as pretyrosine following reaction with prephenate was established by thin layer chromatography of the dansyl-derivative.

Aromatic aminotransferases in coryneform bacteria.

Species of coryneform bacteria (Corynebacterium glutamicum, Brevibacterium flavum, and B. ammoniagenes) are capable of transaminating all three of the aromatic pathway intermediates; prephenate, phenylpyruvate, and 4-hydroxy-phenylpyruvate. Two molecular species of aromatic aminotransferase (denoted aminotransferase I and aminotransferase II) were partially purified from C. glutamicum and B. flavum, whereas a single aromatic aminotransferase was isolated from B. ammoniagenes. In both C. glutamicum and B. flavum, aromatic aminotransferase I and aromatic aminotransferase II have molecular weights of about 155,000 and 260,000 respectively. The two aromatic aminotransferases from C. glutamicum and B. flavum, although exhibiting a similar spectrum of overlapping specificities, differ substantially in substrate preference. Pyridoxal-5'-phosphate is tightly associated with these aminotransferases, since little loss of activity was detected when partially purified enzyme preparations were assayed in the absence of exogenous pyridoxal-5'-phosphate. The aminotransferases are quite sensitive to inhibition by phenylhydrazine. This has practical application when assay of prephenate dehydratase is desired in the presence of aromatic aminotransferase activity since potentially trivial interference can be negated by selective phenylhydrazine inhibition of aromatic aminotransferase activity. At 0.1 mM concentrations of phenylhydrazine, 90% inhibitions of aminotransferase activities were achieved in partially purified preparations of B. flavum and C. glutamicum.

Amino acid metabolism in plants. IV. Kinetic studies with a multispecific aminotransferase purified from bushbean seedlings (Phaseolus vulgaris L.).

Kinetic studies were carried out on a purified multispecific aminotransferase isolated from bushbean roots. The enzyme was found to catalyze the transamination of L-aspartic and L-glutamic acids, as well as L-phenylalanine, L-tyrosine, and L-tryptophan when α-ketoglutarate or oxaloacetate was provided as the amino group acceptor. The Km values for these substrates and for the substrates of the reverse reactions were determined. Double-reciprocal plots of the data obtained for the initial relative velocities of the different aminotransferase activities indicate that all the transamination reactions catalyzed by this enzyme proceed through a binary complex mechanism. The evidence from competitive inhibition of the aromatic aminotransferase activity by the aliphatic substrate, L-aspartic acid, confirmed the view that only one enzyme is responsible for catalyzing the transamination reactions observed with this purified protein.

Nonidentity of the aspartate and the aromatic aminotransferase components of transaminase A in Escherichia coli.

Tyrosine, added to the growth medium of a strain of Escherichia coli K-12 lacking transaminase B, repressed the tyrosine, phenylalanine, and tryptophan aminotransferase activities while leaving the aspartate aminotransferase activity unchanged. This suggested that the aspartate and the aromatic aminotransferase activities, previously believed to reside in the same protein, viz. transaminase A, are actually nonidentical. Further experiments showed that, upon incubation at 55 C, the aspartate aminotransferase of crude extracts was almost completely stable, whereas the tyrosine and phenylalanine activities were rapidly inactivated. Apoenzyme formation was faster, and apoenzyme degradation proceeded more slowly with aspartate aminotransferase than with tyrosine aminotransferase. Electrophoresis in polyacrylamide gels separated the aminotransferases. A more rapidly moving band contained tyrosine, phenylalanine, and tryptophan aminotransferases, and a slower band contained aspartate aminotransferase. A mutant of E. coli K-12 with low levels of aspartate aminotransferase exhibited unchanged levels of tyrosine aminotransferase. Thus, transaminase A appears to be made up of at least two proteins: one of broad specificity whose synthesis is repressed by tyrosine and another, specific for aspartate, which is not subject to repression by amino acids. The apparent molecular weights of both the aspartate and the aromatic aminotransferases, determined by gel filtration, were about 100,000.

Amino acid metabolism in plants. 3. Purification and some properties of a multispecific aminotransferase isolated from bushbean seedlings (Phaseolus vulgaris L.).

The development of aromatic aminotransferase activity was examined in cotyledons, roots, and shoots of bushbean seedlings growing under light or dark conditions for the first 2 weeks after germination. All three aromatic amino acid – α-ketoglutarate aminotransferase activities were found to have similar patterns of development in comparable organs grown under the two environmental conditions, and the changes in levels of activity appeared unrelated to variations in the endogenous amounts of free aromatic amino acids in the organs of these seedlings. The highest total activity for all three transamination reactions was found in the shoots of light-grown seedlings after 14 days, whereas the aminotransferases showing highest specific activity were found in roots of both kinds of seedlings after 8 days of growth. The intracellular distribution of the three aromatic aminotransferase activities and of aspartate aminotransferase activity was investigated by differential centrifugation of root homogenates. Only a total of 10% of these two activities was found in the two particulate fractions; the soluble protein in the final supernatant fraction accounted for almost 90% of the total aromatic and aspartate aminotransferase activities.The aromatic aminotransferase in the soluble protein fraction from seedling roots was purified about 600-fold by pH precipitation, ammonium sulfate fractionation, and Sephadex chromatography, and the recovery obtained was 30–35% based on total activity. It was observed that the specific activity for aspartate–α-ketoglutarate aminotransferase increased proportionally to the increase in aromatic aminotransferase activities during the different steps of purification. Gel electrophoresis of the purified fraction revealed only one protein band which corresponded to the product-specific stained band for the three aromatic aminotransferase activities assayed on other gels. The molecular weight of the purified aminotransferase was found to be about 128 000 daltons and its Stokes radius was calculated to be 43 ± 3 Å. The pH optima for the three aromatic aminotransferase activities and for aspartate aminotransferase activity were all found to be 8.5. The purified enzyme showed no specific requirement for pyridoxal phosphate and an examination of its amino acid substrate specificity revealed that it was able to catalyze transamination of L-aspartic acid, L-phenylalanine, L-tyrosine, and L-tryptophan when α-ketoglutarate was provided as amino group acceptor. The enzyme was also found to catalyze transamination of L-glutamic acid when oxaloacetate was used as amino group acceptor, but neither pyruvate nor glyoxylate were utilized as amino acceptors for transamination of any of the amino acids examined. The enzyme was found to catalyze transamination of aspartic acid with much greater velocity than its rate of reaction with any of the three aromatic amino acids, and the inclusion of aspartic acid in a reaction medium at equimolar concentration with any one of the three aromatic amino acids resulted in strong inhibition of the aromatic aminotransferase activity of the enzyme. All the evidence indicates that the soluble protein fraction purified from bushbean roots contained only one aminotransferase which was able to catalyze the transamination of five L-amino acids. The demonstration of the substrate multispeciftcity of this pure enzyme represents the first evidence for a multispecific aminotransferase in plants.

A new radioactive assay procedure of aromatic aminotransferase

A rapid and sensitive assay of aromatic aminotransferase activity based on semimicro cation-exchange paper chromatography to separate 14C- l-glutamic acid from 14C-α-ketoglutaric acid is described. Its sensitivity, specificity, and broad flexibility constitute some of the principal advantages over the existing procedure. The stoichiometry of the reaction indicated a 1:1 relationship between the amount of phenylpyruvic and glutamic acid formed over a 2 hr interval.
