Evaluation of Moringa Oleifera Extract on Demineralized Enamel of Primary Teeth

Purpose: This invitro study was designed to evaluate the remineralization potential of Moringa Oleifera (M.O) extract in comparison with Casein PhosphoPeptideAmorphous Calcium Phosphate (CPP-ACP) on enamel surface of human primary teeth using Environmental Scanning Electron Microscope (ESEM) &Analysing of mineral content. (EDAX). Material and Methods: forty samples of extracted human teeth were obtained in accordance with guidelines from Al-Azhar University. EDAX & SEM were done for all the samples after demineralization, then they were divided into two main groups, the first group was subjected to CPP-ACP application for 5 minutes twice a day for a week. The second group was soaked in an aqueous extract of M.O for 5 minutes twice a day for a week. EDAX & SEM analyses were done for all specimens after remineralization. All data showed normal (parametric) distribution. Parametric data were presented as mean and standard deviation (SD) values. One-way Analysis of Variance (ANOVA) was used to compare between the two materials and de-mineralized enamel. The significance level was set at P ≤ 0.05. Results: calcium& phosphorus levels of both groups of re-mineralization were higher than the demineralization level. Higher re-mineralization results were obtained from M. O extract. Conclusion: Moringa oleifera extract has a protective effect on enamel remineralization better than CPP-ACP.

Long-term evaluation of early-enamel lesions treated with novel experimental tricalcium silicate paste: A 2-year randomized clinical trial.

The aim of the current study is to evaluate the remineralization potential of experimental tricalcium silicate (TCS) paste in comparison with more popular remineralizing agents like silver diamine fluoride potassium iodide (SDF-KI) and casein phosphopeptide amorphous calcium phosphate (CPP-ACP) on early enamel lesions. Forty-five patients in the age range of 15-50 years had early enamel lesions on the buccal surface of molar teeth. The patients were randomly divided into three treatment groups (SDF-KI, CPP-ACP, and TCS group) with 15 patients per group. Lesions were evaluated clinically by DIAGNOdent pen immediately and after 3,6,12, and 24 months of treatment. The study was completed with 45 patients and 92 teeth. Twice-daily application of CPP-ACP and TCS paste showed a significant remineralization effect on early enamel lesions after 24 months (p< 0.001). Also, annual application of SDF-KI showed a significant remineralization effect after 24 months (p< 0.001). There was a significant difference between (SDF-KI and CPP-ACP) and (SDF-KI and TCS) at the different follow-up periods 3,6,12, and 24 months (p< 0.001). Meanwhile, there was no significant difference between CPP-ACP and TCS at the mentioned follow-up periods (p> 0.05). TCS showed potential remineralization for early enamel lesions. Experimental TCS is a promising remineralizing agent for management of early enamel lesions.

Effect of different surface treatments on the microhardness and colour change of artificial enamel lesions.

To investigate the effect of three different surface treatments on the microhardness and colour change of artificial enamel lesions. One hundred bovine teeth were randomly assigned into four groups. Artificial enamel lesions were created using demineralizing solution for all groups except the sound enamel group. Different surface treatments were then performed G1: resin-infiltrant; G2: Casein phosphopeptide-amorphous calcium phosphate (CPP-ACP); G3: artificial saliva; G4: Sound Enamel. Each group was subdivided into three subgroups, where each subgroup was subjected to a different testing method. Subgroup 1: surface microhardness; subgroup 2: cross-sectional microhardness; subgroup 3: colour measurement. Statistical analysis was performed by ANOVA, followed by Tukey's post hoc test. Sound enamel group recorded the highest surface and cross-sectional microhardness results. No significant difference was found between the resin-infiltrant group and CPP-ACP regarding surface and cross-sectional microhardness at different lesion depths. Resin-infiltrant group showed the least colour change (∆E) results compared to the other groups. Resin-infiltrant can effectively enhance surface microhardness and enamel resistance to demineralization, additionally, reduces the staining susceptibility of white spot lesions (WSLs) after treatment. CPP-ACP application for 4 weeks seems to improve surface microhardness; however, has a limited effect in resisting staining of WSLs after treatment. © 2022 Australian Dental Association.

Bioactive remineralization of dentin surface with calcium phosphate-based agents: An in vitro analysis.

Background:With the increasing prevalence of erosive tooth wear affecting both adults and children; designing optimum protocols of management in a noninvasive manner is gaining precedence.Aim:Comparative evaluation of topically applied calcium phosphate-based agents casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) and beta tricalcium phosphate (β-TCP) on the surface of eroded dentin.Materials and Methods:Dentin blocks from human third molars in four groups were subjected to the surface treatment: (G1) sound dentin (G2) demineralized dentin (G3) CPP-ACP (G4) β-TCP. All the samples except control (G1) were immersed in an acidic solution incubated at 37°C for 96 h. The samples in G3 and G4 were topically treated with CPP-ACP and β-TCP for 4 min twice daily for 14 days; followed by pH-cycling for 21 days. Surface hardness testing and surface morphology were observed using the scanning electron microscopy. Data were analyzed using the Statistical Package for the Social Sciences (SPSS) software with Kruskal-Wallis test and post hoc test.Results:Dentin treated with both CPP-ACP (37.25) and β-TCP (32.05) recorded significantly higher VHN than demineralized (G2-23.51) but lower compared to sound control (G1-57.06).Conclusion:The topical application of CPP-ACP and β-TCP agents shows definite potential in promoting the hardening of demineralized dentin surface.

SALIVA ACIDITY LEVEL TOWARD APPLICATION CASEIN PHOSPHOPEPTIDE - AMORPHOUS CALCIUM PHOSPHATE FOR CHILDREN AGED 8-9 YEARS

Background: Dental caries is a disease of dental hard tissue that is often found in children. The etiology of dental caries is very diverse and multifactorial, one of which is the decreased acidity of saliva. There are many efforts and ways to reduce the incidence of dental caries, one of which is the application of topical flour to the teeth. Casein phosphopeptide–amorphous calcium phosphate (CPP-ACP) as a type of topical flour used has been shown to reduce demineralization and increase tooth remineralization. This study aimed to determine the acidity level of saliva before and after application of CPP-ACP in children aged 8-9 years. Methods: Measurement of salivary acidity was measured using a digital pH meter in the study sample of 42 children consisting of 23 boys and 19 girls. Subjects were applied CPP-ACP for 1 month with the routine application once a week, then re-examined the salivary acidity level after 1 month. Result : The average result of salivary acidity before CPP-ACP application was 6.928 and after CPP-ACP application was 7.0611. The T-test resulted in a very significant difference in the level of salivary acidity before and after the application of CPP-ACP. Conclusion : is that Tthere are differences in the level of salivary acidity in children aged 8-9 years before and after topical application of CPP-ACP flour.

The color improvement of postdebonding white spot lesions after fluoride and casein phosphopeptide- amorphous calcium phosphate application.

White spots are common side effects of orthodontic treatment, and their presence after debonding appears unesthetic. This study aimed to quantify and compare the visual improvement in postdebonding white spot lesions following fluoride and casein phosphopeptide–amorphous calcium phosphate (CPP–ACP) application. The sample included 42 upper premolars extracted for orthodontic reasons. Universal premolar brackets were bonded to the facial surfaces of the teeth that were exposed to a demineralization solution to create artificial white spot lesions, after which the brackets were debonded. The specimens were randomly allocated to three treatment groups (n = 14, each): acidulated phosphate fluoride (APF) gel application group; CPP–ACP paste application group; and control group. pH cycling was conducted to all groups for 14 days. Quantitative measurements were carried out using a spectrophotometer at the following times: before and after the white spot lesions artificially formed and after treating them. All groups showed significant differences in color change before and after treatment. However, there was no significant difference in the color improvement between the APF gel and CPP–ACP paste application groups. Although CPP–ACP application improved the color of the white spot lesions, it did not differ significantly from that of fluoride application.

Enamel Hardness Differences After Topical Application Of Theobromine Gel And Casein Phosphopeptide-Amorphous Calcium Phosphate

Background: Most soft drinks contain citric acid / phosphoric acid with pH &lt;3.5. Strong acids in the oral cavity cause tooth erosion. Tooth erosion leads to demineralization enamel. Remineralization can prevent demineralization process. One of the commercial remineralization agents is Casein Phosphopeptide-amorphous Calcium Phosphate(CPP-ACP) but it has low solubility and less affordable price. Therefore, a natural ingredients such as theobromine from cocoa extract need to be developed as remineralization agent. Objective: To determine enamel hardness differences due to topical application of theobromine gel and CPP-ACP. Method: incisive crowns were cut off (1x1) cm then planted in cylindrical acrylic resin. All samples were immersed with citric acid for 90 minutes then divided into 3 groups. Group I (control) was immersed with aquades (96 min), group II was applied with theobromin gel 200 mg/l (96 min), group III was applied with CPP-ACP (96 min). By using vickers hardness tester, enamel hardness be measured (15 sec) at 3 points. Results: There were significant differences between the control group, CPP-ACP group, and theobromine gel group. Conclusion: Enamel hardnesse with theobromine gel application is higher than CPP-ACP application..

Effect of casein phosphopeptide-amorphous calcium phosphate and erbium:Yttrium–Aluminium–Garnet laser on shear bond strength to demineralized dentin

Abstract Objective: The aim of this in vitro study was to investigate the effect of casein phosphopeptide–amorphous calcium phosphate (CPP-ACP)-containing paste (MI Paste) and erbium:yttrium–aluminum–garnet (Er:YAG) laser radiation on the shear bond strength (SBS) of an etch-and-rinse (E and R) adhesives to demineralized dentin. Materials and Methods: Forty-eight carries-free human dentin surfaces were prepared and demineralized using acidic solutions. Then, the samples were randomly divided into four groups (n = 12) receiving different surface treatments. Group A (control group): no additional treatment, Group B: 3 min application of a CPP-ACP-containing cream (MI Paste), Group C: irradiation of Er:YAG laser, and Group D: irradiation of Er:YAG laser combined with 3 min application of CPP-ACP-containing cream (MI paste). Then, a composite cylinder (Filtek™ Ultimate, 3M ESPE) bonded to the surfaces using E and R adhesives (single bond) and the SBS was measured. The SBS data were analyzed using the one-way analysis of variance test followed by Tukey post hoc by SPSS software. Results: The highest SBS to demineralized dentin was observed after the application of CPP-ACP-containing paste (MI paste) without laser radiation (17.14 ± 2.07). The second highest SBS value showed in control group (11.21 ± 1.65) in which demineralized dentin received no additional treatment. However, the application of MI paste combined with Er:YAG laser irradiation resulted in the higher SBS (8.23 ± 1.02) than laser irradiation alone (5.26 ± 1.02), even though both were lower than control group. Conclusions: The application of CPP-ACP-containing paste (MI paste) could increase the SBS of E and R adhesives to demineralized dentin. Furthermore, laser irradiation with and without CPP-ACP application has an adverse effect on SBS.

The Effect of the Application Sequence of Casein Phosphopeptide-Amorphous Calcium Phosphate Paste on the Shear Bond Strength of Etch-and-Rinse Resin Cements to Dentin.

To evaluate the effect of the application sequence of casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) paste (MI paste) on shear bond strength (SBS) of 2 etch-and-rinse resin cements to dentin. Sixty extracted human maxillary premolars were cut to expose the mid-coronal dentin surfaces. The teeth were randomly divided into 6 groups (n = 10), based on the resin cement used and CPP-ACP application. In groups 1-3, Excite DSC/Variolink II and in groups 2-4, One-Step Plus/Duolink was bonded to dentin. There were 3 groups for each cement as follows: with no treatment as a control; 3-minute application of CPP-ACP before acid etching (CPP-ACP/Etching); 3-minute application of CPP-ACP after acid etching (Etching/CPP-ACP). After storage in distilled water at 37°C for 24 hours, SBS test was performed. Data were analyzed with 2-way ANOVA and post-hoc Tukey test (α = 0.05). CPP-ACP application on the dentin before and after acid etching significantly decreased the SBS of OS/Duolink (13.60 and 14.71 MPa, respectively vs. control; 21.21 MPa, p < 0.001); however, it did not influence the SBS of Excite DSC/Variolink II (25.14 MPa) before acid etching (27.37 MPa) or after acid etching (26.41 MPa, p > 0.388). The application sequence of CPP-ACP did not influence the SBS of etch-and-rinse resin cements evaluated. CPP-ACP applications may lower the SBS in some etch-and-rinse resin cements, and timing of applications makes no difference.

COLOR CHANGES OF POST-DEBONDING WHITE SPOT LESION AFTER MICROABRASION TECHNIQUE WITH FLUORIDE AND CASEIN PHOSPHOPEPTIDE-AMORPHOUS CALCIUM PHOSPHATE APPLICATION

Objective: The prevalence of enamel demineralization among patients after orthodontic treatment is about 50%, which begins with the formation of white spot lesions. The presence of these lesions causes esthetic problems. The aim of this study was to quantify the color changes in post-debonding white spot lesions after microabrasion technique with fluoride and casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) application.Materials and Methods: This study used 42 first premolar teeth that had been extracted, then were soaked for 96 h in a demineralization solution (pH 4, 37°C) to form an artificial white spot lesions. Samples were randomly divided into three treatment groups (n=14): (A) Microabrasion technique only; (B) microabrasion technique with 10% CPP-ACP paste application; and (C) microabrasion technique with 1.23% APF gel application. All groups were assigned to pH cycling for 10 days. The color change was determined using spectrophotometer at 3 different time points, which were measured before and after production of the artificial white spot lesions, and after the artificial white spot lesions were treated.Results: This study showed that there was a significant difference in the color of the artificial white spot lesions after microabrasion technique with CPP-ACP application.Conclusion: Microabrasion technique with CPP-ACP application was giving better result in changing the color of white spot lesions than only microabrasion technique and microabrasion technique with fluoride application.

Micro-CT and FE-SEM enamel analyses of calcium-based agent application after bleaching.

The objective of the present study is to evaluate the effects of casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) on bleached enamel. A bleaching agent (35% hydrogen peroxide) was applied, 4×8min on premolar teeth (n=8). A CPP-ACP paste was applied for 7days. Prior and post-treatment, microtomography images were obtained and 3D regions of interest (ROIs) were selected, from outer enamel, extending to 110.2-μm depth. CT parameters of structure: thickness (St.Th), separation (St.Sp), and fragmentation index (Fr.I.) were calculated for each (ROI). Data was submitted to paired t tests at a 95% confidence level. The samples were evaluated at 3000 to 100,000 magnification. Quantitative analysis of enamel mineral content was also determined by SEM EDX. There was a significant increase in structure thickness and calcium content. The phosphorus content increased after bleaching. There was also a decreased separation and fragmentation index on the outer enamel to a depth of 56.2μm (p<0.05). There were no changes at 110.2-μm depth for the bleaching CPP-ACP association. A covering layer and decreased spaces between the hydroxyapatite crystals appeared around the enamel prisms, 7days after the CPP-ACP application. The application of a CPP-ACP provides a compact structure on the enamel's outer surface, for 7days, due to calcium deposition. CT parameters seem to be a useful tool for mineralizing and remineralizing future studies. CPP-ACP neutralizes any adverse effects on enamel surface when applied during a week after bleaching and minimizes any side effects of the bleaching treatment due to a more compact structure.

Casein phosphopeptide- amorphous calcium phosphate effects on brackets shear bond strength and enamel damage.

BackgroundThe aim of study was to evaluate the application of casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) and fluoride regarding their effect on the shear bond strength (SBS), bond failure pattern of brackets using the adhesive remnant index (ARI) and assessing the quality of enamel surface using the enamel damage index (EDI).Material and MethodsSixty extracted premolar teeth were randomly divided into four groups regarding pretreatment application of CPP-ACP and fluoride. Brackets were bonded using the conventional method. Specimens were thermocycled for 1000 cycles and were subsequently tested for the SBS in a universal testing machine. After debonding, the teeth were examined under a stereomicroscope to evaluate the ARI. Then, The EDI was assessed using a scanning electron microscope (SEM). Data were analyzed using ANOVA and the Chi-square test.ResultsEvaluation of SBS, ARI and EDI scores showed no significant difference among the study groups. However, a correlation was found between the ARI and EDI scores, indicating that with more adhesive remnants on enamel surface, enamel damage was lower.ConclusionsThe use of CPP-ACP and fluoride can be considered a prophylactic application since these agents did not compromise bracket bond strength although they did not reduce iatrogenic damage to the enamel. Effects of casein phosphopeptide-amorphous calcium phosphate application after interproximal stripping on enamel surface: An in-vivo study.

The aim of this study was to investigate the effects of casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) application after interproximal stripping on enamel surface structures invivo. Fifteen patients with a mean age of 15.8years participated in this study. For each patient, the extraction of 4 first premolars was part of the orthodontic treatment plan. The patients were randomly divided into 5 groups of 3 patients. With the exception of group 1, the mesial and distal surfaces of all first premolars were stripped with a stripping disc (Komet; Gebr Brasseler, Lemgo, Germany) under air cooling and then polished with Sof-Lex polishing discs (3M Dental Products, St Paul, Minn). In group 1, no stripping was performed, and the teeth were removed immediately. In group 2, the teeth were removed immediately after the stripping. In group 3, the stripped teeth were extracted after exposure to oral conditions for 3months. In groups 4 and 5, CPP-ACP (Recaldent Tooth Mousse; GC Europe, Leuven, Belgium) or fluoride varnish (Bifluoride 12; Voco, Cuxhaven, Germany) was applied to the stripped surfaces for 3months, respectively, before the teeth were extracted. Surface roughness and microhardness values were evaluated with 1-way analysis of variance and Tukey HSD tests. The CPP-ACP and the fluoride varnish applications increased the surface roughness and microhardness values that had been decreased by stripping. No statistically significant differences were found between groups 3, 4, and 5 for microhardness or between groups 4 and 5 for surface roughness (P>0.5). The saliva and saliva plus remineralizing agents (fluoride varnish and CPP-ACP) increased the microhardness and surface roughness values of stripped enamel surfaces that had been decreased by stripping.

Effect of erbium, chromium: yttrium, scandium, gallium, garnet laser and casein phosphopeptide-amorphous calcium phosphate on surface micro-hardness of primary tooth enamel.

Objective:The aim was to evaluate the effect of Er, Cr: YSGG laser and casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) on surface micro-hardness of primary tooth enamel.Materials and Methods:A total of 30 freshly extracted caries free primary anterior teeth were cleaned and stored in 1% thymol. Teeth were embedded in acrylic resin such that only their buccal surfaces were exposed and were divided into four groups. Group I: Five intact teeth (negative control). The remaining 25 teeth were immersed for 30 min in 1% citric acid for demineralization. Group II: Five demineralized teeth (positive control), Group III: CPP-ACP (GC tooth mousse-GC International, Itabashi-Ku, Tokyo, Japan) application and Group IV: Etching using Er, Cr: YSGG laser + CPP-ACP application. Groups III and IV were subjected to pH cycling for 5 days. Surface micro-hardness of all the teeth was measured using Brinell hardness tester (Fuel Instruments and Engineers Pvt. Ltd.). Data were analyzed using ANOVA.Results:Mean surface micro-hardness of Groups I and II were 177.43 kgf/mm2 and 164.86 kgf/mm2, respectively. Group IV showed a higher mean surface micro-hardness (230.68 kgf/mm2) compared with that of Group III (190.28 kgf/mm2). In comparison to all other groups, laser etching prior to CPP-ACP application increased surface micro-hardness significantly (P < 0.001).Conclusion:Laser irradiation of primary teeth followed by CPP-ACP application increased surface micro-hardness of enamel.

Influence of casein phosphopeptide-amorphous calcium phosphate application, smear layer removal, and storage time on resin-dentin bonding

The aim of this study is to evaluate the influence of Tooth Mousse (TM) application, smear layer removal, and storage time on resin-dentin microtensile bond strength (µTBS). Dentin specimens were divided into two groups: (1) smear layer covered; (2) smear layer removed using 15% EDTA for 90 s. In each group, half the specimens were treated once with TM for 60 min. After bonding procedures using a two-step self-etching adhesive (Clearfil SE Bond (CSE); Kuraray Medical, Tokyo, Japan), an all-in-one adhesive (G-Bond (GB); GC Corp, Tokyo, Japan), and a total-etch adhesive (Adper Single Bond 2 (SB); 3M ESPE, St. Paul, MN, USA), the specimens were stored for 3 d or 6 months in deionized water at 37 °C, and µTBS was tested and analyzed. With the exception of SB (no TM application) and GB, the μTBS was significantly increased for CSE and SB using EDTA pre-conditioning and 3 d of storage (P≤0.001). Bond strength of GB decreased significantly when using EDTA (3 d storage, P<0.05). TM application only increased the μTBS of GB (no EDTA) and SB (with EDTA) after 3 d (P≤0.02). Comparing the adhesives after 3 d of storage, CSE exhibited the greatest μTBS values followed by GB and SB (P≤0.02). The factors of adhesive, EDTA, and TM did not show any significant impact on μTBS when specimens were stored for 6 months (P>0.05). The additional application of TM and EDTA for cavity preparation seems only to have a short-term effect, and no influence on µTBS of dentin bonds after a period of 6 months.

Can demineralized enamel surfaces be bonded safely?

Objective. To evaluate and compare the effects of enamel demineralization, microabrasion therapy and casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) application on the shear bond strength (SBS) of orthodontic brackets bonded to enamel surfaces and enamel color. Materials and methods. Eighty freshly extracted human maxillary premolar teeth were allocated to one of the four groups. Brackets were bonded directly to non-demineralized enamel surfaces in Group I (control group), directly to the demineralized enamel surfaces in Group II, to demineralized enamel surfaces after CPP-ACP application in Group III and to demineralized enamel surfaces after microabrasion therapy in Group IV. The samples were stored in water for 24 h at 37°C and then underwent thermocycling. The SBS in megapascals (MPa) was determined by a shear test with 0.5 mm/min crosshead speed and failure types were classified with modified adhesive remnant index scores. The data were analyzed with one-way analyses of variance (ANOVA), Tukey and chi-square tests at the α = 0.05 level. Results. Significant differences were found among the four groups (F = 21.57, p < 0.01). No significant difference was found between Group I and III (17.12 ± 2.84 and 15.08 ± 3.42 MPa, respectively) or between Group III and IV (12.82 ± 2.64 MPa). The lowest SBS value was determined in Group II (5.88 ± 2.12 MPa). Enamel demineralization, microabrasion therapy and CPP-ACP application affected enamel color significantly. Conclusion. CPP-ACP application and microabrasion therapy are able to increase the decreased SBS of orthodontic brackets because of enamel demineralization.

Effect of cpp-acp on the bond strength of silorane and methacrylate based restorative systems

This study evaluated the influence of the previous application of casein phosphopeptide amorphous calcium phosphate (CPP-ACP) on the bond strength of two restorative systems to dentin. Self-etching adhesive systems Clearfil SE Bond (Kuraray) and FiltekTM LS System Adhesive (3M ESPE) were used in combination with two micro-hybrid composites FiltekTM Z250 (3M ESPE) and FiltekTM LS (3M ESPE), respectively. Twenty-eight sound human third molars had the occlusal surface worn until the total exposure of dentin and were randomly divided into 4 groups (n = 7): G1 (Clearfil + Z250), G2 (CPP-ACP + Clearfil + Z250), G3 (LS system) and G4 (CPP-ACP + LS system). The application of the paste containing CPP-ACP was held in the dentin of G2 and G4 prior to adhesive protocol, which followed the manufacturer's recommendations. Two cylinders of a single increment of resin composite were built on each dentin surface using tygon`s matrix with 1.26 mm of diameter x 1 mm high and light-cured with LED Bluephase 16i (Vivadent) apparatus. Microshear bond strength test was conducted in a universal testing machine (EZ Test, Shimadzu) with a speed of 0.5 mm/min. The data were subjected to two-way ANOVA test with a significance of 5%. There was no significant difference on bond strength between restorative systems with or without the previous use of a CPP-ACP paste. It was concluded that the application of CPP-ACP in the dentin prior to adhesive protocol did not influence the bond strength of the restorative systems tested.

Effects of casein phosphopeptide–amorphous calcium phosphate remineralizing paste and 8% arginine desensitizing paste on dentin permeability

To compare the effects of casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) paste and 8% arginine paste on dentin permeability. Sixteen human third molars were sectioned to remove the roots and expose occlusal dentin surfaces. The pulpal side was flattened to obtain 1 mm remaining dentin thickness. Each specimen was attached on a Perspex plate and connected to a dentin-permeability measuring apparatus. The specimens were divided into two groups according to the tested products. Dentin permeability was measured under a simulated pulpal pressure after: (a) etching with 37% phosphoric acid; (b) application of CPP-ACP or 8% arginine paste; and (c) acid challenge with 2.6 mmol/L lactic acid. Twelve specimens were prepared so that the treated dentin surfaces could be examined using a scanning electron microscope. Dentin permeability after the application of 8% arginine paste was significantly reduced and lower than that of CPP-ACP paste (P < 0.01). Acid challenge with lactic acid significantly increased dentin permeability in both experimental groups (P < 0.01). The dentin surface treated with 8% arginine paste was covered with particles, but they were removed by acid challenge. Most dentinal tubules were patent when the CPP-ACP paste was applied. The application of 8% arginine paste decreased dentin permeability, but CPP-ACP paste did not.

Surface remineralization potential of casein phosphopeptide-amorphous calcium phosphate on enamel eroded by cola-drinks: An in-situ model study.

Aim:The aim of this study was to investigate the remineralization potential of casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) on enamel eroded by cola drinks.Subjects and Methods:A total of 30 healthy subjects were selected from a random sample of 1200 children and divided into two groups of 15 each wherein calcium and phosphorus analyses and scanning electron microscope (SEM) analysis was carried out to investigate the remineralization of enamel surface. A total of 30 non-carious premolar teeth were selected from the human tooth bank (HTB) to prepare the in-situ appliance. Three enamel slabs were prepared from the same. One enamel slab was used to obtain baseline values and the other two were embedded into the upper palatal appliances prepared on the subjects’ maxillary working model. The subjects wore the appliance after which 30 ml cola drink exposure was given. After 15 days, the slabs were removed and subjected to respective analysis.Statistical Analysis Used:Means of all the readings of soluble calcium and phosphorous levels at baseline,post cola-drink exposure and post cpp-acp application were subjected to statistical analysis SPSS11.5 version. Comparison within groups and between groups was carried out using ANOVA and F-values at 1% level of significance.Results:Decrease in calcium solubility of enamel in the CPP-ACP application group as compared to post-cola drink exposure group (P < 0.05) was seen. Distinctive change in surface topography of enamel in the post-CPP-ACP application group as compared to post-cola drink exposure group was observed.Conclusion:CPP-ACP significantly promoted remineralization of enamel eroded by cola drinks as revealed by significant morphological changes seen in SEM magnification and spectrophotometric analyses.

Influence of surface treatments on enamel susceptibility to staining by cigarette smoke

Objectives: The purpose of this study was to evaluate the influence of remineralizing agents, including artificial saliva, neutral fluoride, and casein phosphopeptide-amorphous calcium phosphate (CPP-ACP), on the susceptibility of bleached enamel to staining by cigarette smoke. Study Design: Fifty bovine enamel blocks were randomly divided into five groups (n = 10): G1- bleaching; G2- bleaching and immersion in artificial saliva; G3- bleaching and application of CPP-ACP; G4- bleaching and application of neutral fluoride; and G5- untreated (Control). Teeth were bleached with 35% hydrogen peroxide and treated with the appropriate remineralizing agent. After treatment, all groups were exposed to cigarette smoke. Enamel color measurements were performed at three different times: before treatment (T1), after treatment (bleaching and remineralizing agent) (T2), and after staining (T3), by using the CIE Lab method with a spectrophotometer. The data coordinate L* was evaluated by analysis of repeated-measures PROC MIXED and Tukey-Kramer’s test, and the ΔE values were submitted to one-way ANOVA and Tukey’s test (α = 0.05). Results: The G1 group did not show any statistically significant difference for L* values between times T1 and T2. The G4 and G5 groups showed lower L* values at T3 compared to T2. No significant differences between the groups were observed for ΔE (after treatment and staining). However, G4 showed a clinically apparent color change. Conclusions: Treatment of bleached enamel with neutral fluoride can contribute to the increased staining of enamel due to cigarette smoke.
