Abstract The objective of the present study was to test the hypothesis that the apparent ileal digestibility (AID) of amino acids (AA) in feed ingredients is not constant in pigs with 3 different body weights (BW). Eight pigs with initial BW of 22.2 ± 1.2 kg cannulated at the distal ileum were used. Two experimental diets contained corn distillers dried grains with solubles (DDGS) and soybean meal-wheat mixture (SBM-W; 70% soybean meal + 30% wheat) as the sole source of nitrogen, respectively. The diets were assigned to the animals in a 2-period cross-over design. Each period consisted of a 5-d adaptation period and a 2-d ileal digesta collection period. The animals, diets, and experimental design for 50 kg pigs (initial BW = 55.6 ± 3.3 kg) and 80 kg pigs (initial BW = 86.9 ± 5.1 kg) were identical to those used for 22 kg pigs. Orthogonal polynomial contrasts were used to determine the effect of ingredient type (corn DDGS or SBMM) and BW of pigs (from 22 to 87 kg) and the interaction between these 2 main effects. The AID of crude protein, lysine, and methionine in both corn DDGS and SBM-W linearly or quadratically increased (P < 0.05) with an increase in pig BW. An interaction between ingredient type and linear effect of BW was observed (P < 0.05) in histidine, isoleucine, leucine, phenylalanine, threonine, and valine. Specifically, a linear increase in AID of these AA with increasing BW of pigs was greater in SBM-W compared with corn DDGS. In conclusion, the AID values of most AA differed between the BW range of 22 to 87 kg, and these patterns varied among different ingredients.

Effects of dietary xylanase supplementation on growth performance, intestinal health, and immune response of nursery pigs fed diets with reduced metabolizable energy.

This study aimed to investigate the effects of xylanase on growth performance and intestinal health of nursery pigs fed diets with reduced metabolizable energy (ME). One hundred ninety-two pigs at 8.7 kg ± 0.7 body weight (BW) after 7 d of weaning were allotted in a randomized complete block design with initial BW and sex as blocks. Eight dietary treatments consisted of 5 ME levels (3,400, 3,375, 3,350, 3,325, and 3,300 kcal ME/kg) below the NRC (2012) requirement and 4 levels of xylanase (0, 1,200, 2,400, and 3,600 XU/kg) to a diet with 3,300 kcal ME/kg. All pigs received their respective treatments for 35 d in 2 phases, pre-starter (14 d) and starter (21 d). On day 35, eight pigs in 3,400 kcal/kg (CON), 3,300 kcal/kg (LE), and 3,300 kcal/kg + 3,600 XU xylanase/kg (LEX) were euthanized to collect jejunal tissues and digesta for the evaluation of mucosa-associated microbiota, intestinal immune response, oxidative stress status, intestinal morphology, crypt cell proliferation, and digesta viscosity as well as ileal digesta to measure apparent ileal digestibility. Data were analyzed using the MIXED procedure on SAS 9.4. The LE increased (P < 0.05) jejunal digesta viscosity, tended to have decreased (P = 0.053) relative abundance of Prevotella, and tended to increase (P = 0.055) Lactobacillus. The LE also increased (P < 0.05) the concentration of protein carbonyl whereas malondialdehyde, villus height (VH), villus height to crypt depth ratio (VH:CD), apparent ileal digestibility (AID) of nutrients, and finally average daily feed intake were decreased (P < 0.05). The LE did not affect average daily gain (ADG). The LEX decreased (P < 0.05) digesta viscosity, increased (P < 0.05) the relative abundance of Prevotella, decreased (P < 0.05) Helicobacter, decreased (P < 0.05) the concentration of protein carbonyl, tended to increase (P = 0.065) VH, and decreased (P < 0.05) VH:CD and crypt cell proliferation. Moreover, LEX increased (P < 0.05) the AID of dry matter and gross energy and tended to increase (P = 0.099; P = 0.076) AID of crude protein, and ether extract. The LEX did not affect ADG but did tend to decrease (P = 0.070) fecal score during the starter phase. Overall, reducing ME negatively affected intestinal health parameters and nutrient digestibility without affecting growth. Supplementation of xylanase mitigated some of the negative effects observed by ME reduction on intestinal health and digestibility of nutrients without affecting growth.

Approaches to determine the efficiency of novel 3-phytase from Klebsiella pneumoniae and commercial phytase in broilers from 1 to 14 d of age

This study aimed to evaluate the effects of a laboratory 3-phytase (the expression of the phyK gene, Lab-Phy) and a commercial 6-phytase (Quantum Blue 40 P, Com-Phy) alone and in combination (corn-soy-based diets) in broilers. A total of 400, day-old Ross 308 male broilers were randomly assigned to 5 treatments with 10 replicate cages (8 chicks/cage) for a 14-day trial. Experimental treatments included the positive control (0.95% Ca and 0.48% nonphytate phosphorus (nPP), PC), negative control (0.90% Ca and 0.22% nPP, NC), and NC which was supplemented with Lab-Phy 250 FTY/kg and Com-Phy 250 FTY/kg alone or in combination of Lab-Phy 125 FTY/kg and Com-Phy 125 FTY/kg. The inclusion of Lab-Phy in the NC diet significantly improved the P and Ca content in the tibia compared to the NC group. Moreover, the inclusion of Com-Phy alone and in combination with Lab-Phy in the NC diet significantly increased the P and Ca content in the tibia compared to the Lab-Phy. The mRNA expression of NaPi-IIb was upregulated in the duodenum by the reduction of nPP and downregulated by the inclusion of any phytase, whereas other nutrient transporters were not influenced by the reduction of nPP or the addition of phytase in the small intestine mucosa. Broilers receiving the NC diet obtained the lowest body weight (BW) and body weight gain (BWG) at 8 to 14 and 1 to 14 d of age. The NC group showed the lowest villi height and surface area, Newcastle disease (ND) antibody titer, and digestibility of nutrients compared to the PC group at 14 d of age. Supplementing the NC diet with the Lab-Phy and Com-Phy individually, or in combination tended to improve BW, BWG, tibia characteristics, villi characteristics, ND, and retained CP and P, and apparent ileal digestibility of CP, P, methionine, and threonine. The present research indicated that the studied traits by the combination of phytases were slightly better than the average of the 2 individually, suggesting there might be some value in combining the laboratory and commercial phytases.

Changes in performance and apparent ileal digestibility of broiler chickens fed diets containing electron-irradiated full-fat canola seed

Two experiments were conducted to evaluate the effect of electron-beam irradiation of canola seed on the performance and nutrient digestibility in broiler chickens. In the first experiment, the chemical composition of raw and electron-beam irradiated canola seed at doses of 10, 20, 30, and 40 kGy was characterized in order to determine the appropriate dose for the in vivo experiment. The results indicated that applied doses of electron irradiation did not affect dry matter (DM), crude fiber (CF), ash, neutral (NDF) and acid detergent fiber (ADF), crude protein (CP), and ether extract (EE) content of canola seed. Irradiation, however, decreased tannin (P < 0.01) content. Also, the glucosinolate of samples decreased by increasing irradiation dose (P < 0.01). In the second experiment, seven dietary treatments included canola seed at 5%, 10%, and 15% (raw and irradiated at 40 kGy), and a control diet was used in a completely randomized design. Birds fed 5% of irradiated canola seed had the highest feed intake during the grower (P < 0.05), finisher, and the total experiment period (P < 0.01). In the starter period, birds fed 10% of irradiated canola seed had the highest body weight gain (BWG) (P < 0.01). Birds that consumed the control diet had the highest BWG compared with the other treatments (P < 0.05) during the total experiment period. In the starter period, birds consumed the control diet, 5%, 10%, and 15% of irradiated canola seed had a better feed conversion ratio (FCR) compared to those fed raw canola seed (P < 0.01). Apparent ileal digestibility of CP, EE, and DM significantly decreased in birds fed 15% canola seed (raw or irradiated) compared to those fed other treatments (P < 0.01). In conclusion, radiation reduced glucosinolate and tannin and improved FCR in the starter period. Therefore, if high levels of canola seed (e.g., 15%) are used in the diet of broiler chickens, then radiation can be helpful.

Evaluation of a dacitic (rhyolitic) tuff breccia use on performance, inflammatory, and antioxidant responses in broilers mildly challenged with Eimeria spp.

The objective of the study was to investigate the effects of a dacitic tuff breccia (DTB) on Eimeria-infected broilers. A total of 600 one-day-old Cobb 500 male chickens were randomly assigned to 5 treatments with 10 replicates of 12 birds. Treatments were: an unchallenged control (UC), a challenged (CC) control (0% DTB), and 3 challenged groups with 0.125, 0.25, or 0.5% DTB. At d 14, birds in the CC and DTB groups were orally gavaged with mixed Eimeria spp., while the UC received water. Growth performance was evaluated during prechallenge, challenge, and postchallenge periods (0-14 d; 14-20 d; and 20-26 d, respectively). Gastrointestinal permeability was measured at 5 days postinfection (dpi). Intestinal histology and nutrient digestibility of dry matter (DM), crude protein (CP), and ileal digestible energy (IDE) were measured at 6 dpi. Liver activity of glutathione peroxidase (GSH-Px) was determined at 6 dpi, and concentrations of reduced (GSH) and oxidized glutathione (GSSG) were analyzed at 6 and 12 dpi. Data were analyzed using a linear mixed model analysis and Tukey's test (P ≤ 0.05). From 0 to 14 d, similar average daily gain (ADG) and average daily feed intake (ADFI, P > 0.05) were observed. Gain:feed ratio (GF) was higher in 0.125, 0.25, and 0.5% of DTB than the CC and UC (P < 0.001). From 14 to 20 d, the UC had the highest ADG, ADFI, and GF (P < 0.001). At 5 dpi, intestinal permeability was higher in the challenged groups than the UC. Additionally, the UC showed the highest apparent ileal digestibility of CP, whereas 0.125% DTB had higher CP digestibility than the CC and 0.5% DTB (P < 0.001). At 6 dpi, 0.125% DTB increased GSH-Px activity compared to the CC, 0.5% DTB, and UC (P < 0.001). At 12 dpi, 0.125% DTB showed increased GSH concentration compared to the CC, 0.25% DTB, and 0.5% DTB (P < 0.01). The mild coccidia infection negatively impacted growth performance, apparent ileal nutrient digestibility, intestinal histology, and gastrointestinal integrity in broilers. The use of 0.125% DTB exhibited potential in improving antioxidant responses, apparent ileal digestibility of CP, and growth performance.

Efficacy of a bacterial 6-phytase supplemented beyond traditional dose levels on jejunal mucosa-associated microbiota, ileal nutrient digestibility, bone parameters, and intestinal health, and growth performance of nursery pigs.

This study aimed to determine the efficacy of a bacterial 6-phytase (Buttiauxella spp.) supplemented beyond traditional dose levels based on jejunal mucosa-associated microbiota, apparent ileal digestibility (AID), intestinal health and bone parameters, and growth performance of nursery pigs. Seventy-two weaned pigs (36 barrows and 36 gilts at 21 d of age with 5.8 ± 0.5kg BW) were allotted to six treatments based on randomized complete block design with sex and initial BW as blocks and fed in three dietary phases (phase 1 for 14 d, phase 2 for 10 d and phase 3 for 14 d). The treatments included a negative control (NC) diet without phytase formulated meeting nutrient requirements by NRC and the other five treatments deficient in calcium (Ca) and phosphorus (P) by 0.12% with increasing levels of a bacterial 6-phytase (0, 500, 1,000, 2,000, and 5,000 FTU/kg feed). Titanium dioxide (0.4%) was added to phase 3 diets as an indigestible marker to measure AID of nutrients. On d 45, all pigs were euthanized to collect ileal digesta to measure AID, the third metacarpus to measure bone parameters, and jejunal mucosa to evaluate intestinal health and microbiota. Data were analyzed using the MIXED procedure for polynomial contrasts and the NLMIXED procedure for broken line analysis using the SAS 9.4. Broken line analysis demonstrated that 948 FTU/kg feed increased (P < 0.05) the ADG and the bone P content. Increasing phytase supplementation increased (linear, P < 0.05) AID of CP, bone P and ash content. Increasing phytase supplementation reduced (P < 0.05) the fecal score during phases 2 and 3. Broken line analysis demonstrated that 1,889 FTU/kg feed increased (P < 0.05) bone breaking strength. Increasing phytase supplementation (PC vs. Phy) increased (P < 0.05) AID of ether extract (EE) and P. The supplementation of phytase at 2,000 FTU/ kg feed tended (P = 0.087) to reduce the relative abundance of Prevotellaceae. In conclusion, the supplementation of a bacterial 6-phytase beyond traditional dose levels improved bone breaking strength, bone ash and P content, AID of CP, EE, and P, and growth performance of nursery pigs with reduced relative abundance of Bacteroidetes specifically Prevotellaceae in the jejunal mucosa. Supplementation of a bacterial 6-phytase between 1,000 and 2,000 FTU/kg feed provided benefits associated with growth performance and bone parameters of nursery pigs.

Standardized ileal digestibility of amino acids in cottonseed meal fed to pregnant and non-pregnant sows.

This study determined the apparent ileal digestibility (AID) and standard ileal digestibility (SID) of crude protein (CP) and amino acids (AA) of six cottonseed meal (CSM) samples in pregnant and non-pregnant sows. Two CSM samples were processed by expelling with a CP level of 40.67% (ECSM41) and 44.64% (ECSM45), and four samples were processed by solvent-extracted which contained graded CP levels of 45% (SECSM45), 51.16% (SECSM51), 56.44% (SECSM56), and 59.63% (SECSM60). Landrace ×Yorkshire third parity sows, 7 at gestation and 14 non-pregnant, were fitted with T-cannula in the distal ileum. Pregnant sows were allotted to a 7 × 6 Latin square design with a cornstarch-based nitrogen-free (NF) diet and the six CSM diets, and non-pregnant sows were allotted to a replicated 7 × 3 Latin square design with seven diets and three periods, respectively, resulting in a total of six replicates per treatment. All experimental sows were fed 3.0 kg/d of the experimental diets. The AID of CP in ECSM41 (75.58%) was lower than in SECSM51 (80.42%), SECSM56 (80.50%), and SECSM60 (82.44%) diets for pregnant sows (P < 0.05). The AID of CP in ECSM41 (77.88%) was significantly lower than in SECSM60 (81.87%) diets for non-pregnant sows (P < 0.05). The physiological phase did not affect the AID of CP (P > 0.05). The SID of CP was affected by diets for both pregnant (P < 0.01) and non-pregnant sows (P = 0.06). The physiological phase also affected the SID of CP (P < 0.01). The AID of histidine, leucine, methionine, threonine, and tryptophan significantly differed between different CSM samples in both pregnant (P < 0.05) and non-pregnant sows (P < 0.05). The AID of dispensable AA aspartic acid, cysteine, glutamic acid, serine, and tyrosine differed between different CSM samples of both pregnant (P < 0.05) and non-pregnant sows (P < 0.05). For pregnant sows, the indispensable AA cysteine, glycine, proline, and tyrosine had significantly different SID between different groups (P < 0.05). For non-pregnant sows, the SID of arginine, lysine, methionine, threonine, aspartic acid, cysteine, and serine had different values among different diets (P < 0.05). In conclusion, the current study presented that the ileal AA digestibility of CSM fed to pregnant and non-pregnant sows increased with the decreased of fiber content, and the current findings can contribute to a precise formulation of diets for sows using CSM.

Ileal and total tract digestibility of energy and nutrients in pig diets supplemented with a novel consensus bacterial 6-phytase variant.

An experiment was conducted to test the hypothesis that increasing levels of a novel phytase increases the apparent ileal digestibility (AID) and apparent total tract digestibility (ATTD) of nutrients in diets fed to young pigs. A negative control (NC) diet based on corn, soybean meal, and canola meal that contained approximately 0.83% phytate (i.e., 0.23% phytate-bound P) was formulated to be deficient in Ca, P, and standardized ileal digestible amino acids (AA). Five additional diets were formulated by adding 250, 500, 1,000, 2,000, or 4,000 phytase units/kg of the novel phytase to the NC diets. Eighteen ileal-cannulated pigs (17.81 ± 1.71 kg) were allotted to a 6 × 3 incomplete Latin square design with six diets and three 11-day periods. There were three pigs per diet in each period; therefore, there were nine replicate pigs per diet. The initial 5 d of each period was considered an adaptation period to the diet. For each period, fecal samples were collected via anal stimulation on days 6, 7, 8, and 9, whereas ileal digesta were collected on days 10 and 11 using standard procedures. Results indicated that the AID of crude protein, indispensable AA, and dispensable AA was increased (quadratic, P < 0.05) as the concentration of microbial phytase increased in the diets. Dietary inclusion of the novel phytase at 1,000 or 2,000 FTU/kg increased the AID of total AA from 73.7% to 79.8%. Increasing levels of microbial phytase increased (quadratic, P < 0.05) the AID of dry matter and minerals (i.e., Ca, P, K, Mg, Cu) in the diets. Likewise, a linear increase (P < 0.05) in the AID of ash and Na was observed as the inclusion level of phytase increased in the diets. Increasing levels of microbial phytase increased (linear, P < 0.01) the AID of gross energy (GE) and starch in the diets. A quadratic (P < 0.05) increase in the ATTD of ash, Ca, P, K, and Cu in experimental diets was observed as the concentration of microbial phytase increased in the diets. The ATTD of Mg and GE also increased (linear; P < 0.05) as concentration of dietary phytase increased. In conclusion, the novel microbial phytase used in this experiment was effective in increasing the AID of dry matter, GE, starch, minerals, and AA, as well as the ATTD of gross energy and minerals in diets formulated to be deficient in Ca, P, and AA.

Effects of Protein Source, Whole Wheat and Butyric Acid on Live Performance, Gut Health and Amino Acid Digestibility in Broiler Chickens.

A total of 896 1-day-old straight-run (Ross-308) broilers were used to investigate the interactive effects of protein source (PS), diet structure (DS) and butyric acid (BA) on live performance and carcass characteristics, gut development and its morphology and apparent ileal digestibility (AID) of protein and amino acids (AA). Eight experimental diets comprising 8 replicates with 14 birds each were tested in a 2 × 2 × 2 factorial arrangement with complete randomized design by two levels of BA (0 and 0.1%), two forms of DS (whole vs. ground wheat) and two PS, i.e., soybean meal and canola meal (SBM vs. CM). Throughout the entire experimental period (0 to 35 d), broilers fed SBM-based diets exhibited better (p < 0.05) growth performance (feed intake (FI), body weight gain (BWG) and feed conversion ratio (FCR)), carcass parameters (p < 0.05), gut health (p < 0.05), and nutrient digestibility (p < 0.05) than CM-fed broilers. Dietary whole wheat (WW) positively affected FI (p = 0.001), BWG (p = 0.004) and FCR (p = 0.035) during the overall experimental period. Broilers fed WW had 6, 5, 8, 11 and 10% lower empty relative weights of crop, proventriculus, jejunum, ileum and colon and 25 and 15% heavier gizzard and pancreas, respectively, with longer villus height (p < 0.001), reduced crypt depth (p = 0.031) and longer villus height-to-crypt depth ratio (p < 0.001) than those fed ground-wheat-based diets. Broilers fed WW had greater (p < 0.05) AID of CP and most of the AA. Butyric acid supplementation resulted in improved (p < 0.05) growth performance and digestibility of threonine, valine, leucine, isoleucine, phenylalanine, serine and aspartate. The broilers consuming SBM had 28% lower abdominal fat than those fed CM-based diets. In conclusion, harmful consequences of a less digestible PS can partially be compensated by the inclusion of WW, and supplementation of BA further reduces these detrimental effects.

Effect of protease supplementation on apparent ileal crude protein and amino acid digestibility of over-processed soybean meals in broilers

BackgroundNutritional value of proteins in feed ingredients can be negatively affected by hydrothermal processing, which causes large variation in the bioavailability of amino acids (AA) and negatively affects animal productive performance. Supplementation of exogenous proteases could increase the rate of digestion of damaged proteins, thereby increasing overall AA digestibility and bioavailability. The aim was to determine the effect of exogenous protease supplementation on the apparent ileal digestibility (AID) of crude protein (CP) and AA of soybean meals (SBM) with different degrees of hydrothermal processing in broilers.MethodsThe experiment involved a 3 × 2 factorial arrangement, with SBM processing time (commercial SBM or autoclaved for 30 or 60 min at 120 °C) and protease supplementation (not supplemented and supplemented) as factors. Protease was included at three times the recommended dose (0.06%) and the experimental diets were fed from 15 to 21 d.ResultsThe interaction between the effects of SBM processing and protease supplementation was significant for the AID of CP (P = 0.01), Trp (P = 0.01), Gly (P = 0.03) and Pro (P = 0.03), and also for the average daily gain (P = 0.01) and feed conversion ratio (P = 0.04). Increasing the processing time of SBM decreased (P < 0.0001) the AID of all amino acids, whilst the effect of protease supplementation was only significant for the AID of Phe (P = 0.02) and Tyr (P = 0.01).ConclusionsExogenous protease supplementation at three times the commercial dose does not seem to offset the negative effects of hydrothermal processing of SBM on the apparent ileal digestibility of CP and amino acids or performance of broilers. Whilst positive numerical improvements of digestibility and performance (ADG and FCR) were noticed with protease supplementation at relatively mild processing levels, negative results were obtained with the harsh-processed meals.

Functional roles of xylanase enhancing intestinal health and growth performance of nursery pigs by reducing the digesta viscosity and modulating the mucosa-associated microbiota in the jejunum.

This study was conducted to investigate the functional roles of an endo-β-1,4-xylanase on the intestinal health and growth performance of nursery pigs. A total of 60 pigs (21 d old, 6.9 ± 0.8 kg body weight [BW]) were allotted based on a randomized complete block design with sex and initial BW as blocks. Dietary treatments had nutrients meeting the requirements with increasing levels of endo-β-1,4-xylanase (0, 220, 440, 880, 1,760 xylanase unit [XU] per kg feed) and fed to pigs in three phases (phases 1, 2, and 3 for 10, 14, and 14 d, respectively). Titanium dioxide (0.4%) was added to the phase 3 diets as an indigestible marker. On day 38, all pigs were euthanized to collect ileal digesta to measure apparent ileal digestibility (AID), jejunal digesta to measure viscosity, and jejunal mucosa to evaluate intestinal health. Data were analyzed using the MIXED procedure for polynomial contrasts and the NLMIXED procedure for broken line analysis of SAS. Increasing xylanase in the nursery diets reduced (linear, P < 0.05) the digesta viscosity in the jejunum. Increasing xylanase tended to reduce the relative abundance of Cupriavidus (P = 0.073) and Megasphaera (P = 0.063); tended to increase the relative abundance of Succinivibrio (P = 0.076) and Pseudomonas (P = 0.060); and had a quadratic effect (P < 0.05) on the relative abundance of Acinetobacter (maximum: 2.01% at 867 XU per kg feed). Xylanase from 0 to 1,087 XU per kg feed reduced (P < 0.05) jejunal malondialdehyde. Xylanase from 0 to 1,475 XU per kg feed increased (P < 0.05) the AID of neutral detergent fiber. Increasing xylanase increased (P < 0.05) the AID of ether extract and tended to increase (P = 0.058) the AID of crude protein. Increasing xylanase did not affect growth performance on overall period, whereas xylanase from 0 to 736 XU per kg feed increased (P < 0.05) average daily gain (ADG) during days 31 to 38. In conclusion, xylanase supplementation showed benefits on intestinal health by reducing digesta viscosity, the relative abundance of potentially harmful bacteria, and the oxidative stress in the jejunal mucosa, collectively enhancing intestinal morphology and the AID of nutrients. Xylanase supplementation at a range of 750 to 1,500 XU per kg feed provided benefits associated with reduced oxidative stress, increased nutrient digestibility, resulting in potential improvement on growth performance of nursery pigs by increasing the average daily feed intake and moderately improving the ADG throughout the last week of feeding.

It was demonstrated that the use of phytase enhanced the intestinal health of the broiler chickens by potentially increasing beneficial and reducing harmful bacteria, enhancing intestinal morphology, and consequently improving nutrient digestibility and bone parameters. Our results suggest that the use of phytase can positively modulate the jejunal mucosa-associated microbiota in the jejunum, enhance intestinal integrity, nutrient digestibility, and bone parameters of broiler chickens.This study aimed to determine supplemental effects of phytase on modulation of the mucosa-associated microbiota in the jejunum, intestinal morphology, nutrient digestibility, bone parameters, and growth performance of broiler chickens. Three hundred and sixty newly hatched broiler chickens (Ross 308) (44 ± 2 g BW) were randomly allotted in 6 treatments with 10 birds per cage based on a completely randomized design and fed for 27 d. The treatments consisted of one negative control (NC), diet formulated meeting the requirements suggested by Ross recommendations (2019), and without phytase supplementation. The other treatments consisted of a positive control diet (PC) formulated with 0.15% deficient Ca and P and split into 5 treatments with different phytase inclusion levels (0, 500, 1000, 2000, 4000 FTU/kg feed). Titanium dioxide (0.4%) was added to feeds as an indigestible marker to measure apparent ileal digestibility (AID) of nutrients. On d 27, 3 birds were randomly selected from each cage and euthanized to collect samples for analyzing the mucosa-associated microbiota in the jejunum, oxidative stress status, AID, and bone parameters. Data were analyzed using the proc Mixed of SAS 9.4. Phytase supplementation tended to have a quadratic effect (p = 0.078) on the overall ADG (maximum: 41 g/d at 2833 FTU/kg of feed). Supplementation of phytase at 2,000 FTU/kg increased (p < 0.05) the relative abundance of Lactobacillus and reduced (p < 0.05) Pelomonas. Moreover, it tended to reduce Helicobacter (p = 0.085), Pseudomonas (p = 0.090) Sphingomonas (p = 0.071). Phytase supplementation increased (p < 0.05) the villus height and the AID of CP; and tended to increase (p = 0.086) the AID of P. Phytase supplementation increased (p < 0.05) breaking strength and P content in the tibia. In conclusion, phytase supplementation showed potential benefits on the modulation of the mucosa-associated microbiota in the jejunum by tending to reduce harmful bacteria (Pelomonas, Helicobacter, and Pseudomonas) and increase beneficial bacteria (Lactobacillus). In addition, it showed positive effects increasing apparent ileal digestibility of CP and P, enhancing intestinal morphology (villus height), and improving the bone parameters (bone breaking strength, ash, and P content). Phytase supplementation at a range of 38 to 59 FTU/d or 600 to 950 FTU/kg of feed provided the most benefits related to nutrient digestibility.

Partial Replacement of Animal Fat with Full-Fat Almond in Broiler Chicken Diets: Performance, Nutrient Digestibility, Blood Profile, Cecal-Fecal Microflora Composition, and Foot-Pad Dermatitis.

This study evaluated the effect of full-fat almonds (FFA) on broiler nutrition. In the study, the formulation of FFA as a raw material showed higher growth performance than the basic feed. This seems to be due to the high digestibility of protein and fat and the promotion of intestinal health through the reduction of harmful microorganisms in the intestine. Therefore, FFA can be used as a high-quality protein and fat substitute as a raw material for broiler feed.The purpose of this study was to evaluate the effect of full-fat almonds (FFA) as an alternative protein and fat source for broiler feed on broiler productivity, nutrient digestibility, blood characteristics, cecal-fecal microflora, and foot-pad dermatitis (FPD). A total of 96, one-day-old broiler chickens (Arbor Acres) with initial body weight 41.61 ± 0.36 g were placed in 16 cages. In each trial, four treatments were set up: a basal diet partially replacing animal fat with FFA 0% (Control, CON), a basal diet partially replacing animal fat with FFA 1% (T1), a basal diet partial replacing animal fat with FFA 2% (T2), a basal diet partially replacing animal fat with FFA 4% (T3). The experiment was conducted for a total of 4 weeks. Feed conversion ratio (FCR) was higher (p < 0.05) in the T3 group of broilers at weeks 0 to 1 than in the CON group of broilers. From weeks 3 to 4, and for the entire experimental period, FCR was lower (p < 0.05) in the T3 group of broilers than in the CON and T1 groups of broilers. The apparent ileal digestibility (AID) of the ether extract (EE) was higher (p < 0.05) in the T3 group than in the other treatment groups, and AID of crude protein (CP) was higher (p < 0.05) in the T3 group than in the CON group. The apparent total tract digestibility (ATTD) of EE was lower (p < 0.05) in the CON group than in the other treatment groups, and the ATTD of CP and energy was higher (p < 0.05) in the T3 group of broilers than in the CON group of broilers. The AID and ATTD of total amino acids were higher (p < 0.05) in the T3 group than in the other treatment groups. Blood cholesterol levels were lower (p < 0.05) in the T3 group of broilers than in the CON and T1 groups of broilers, and higher (p < 0.05) in the CON group of broilers than in the T2 and T3 groups of broilers. The amount of E. coli in the cecal and fecal was lower (p < 0.05) in the T3 group than in CON and T1 groups. FPD score was higher (p < 0.05) in the T3 group of broilers than in the CON group of broilers. In conclusion, replacing a partial of animal fat with at least 4% FFA in broiler diets can increase growth performance and nutrient digestibility in broiler nutrition.

Chemical Compositions of Brown and Green Seaweed, and Effects on Nutrient Digestibility in Broiler Chickens.

Proximate content, mineral and amino acid contents were analysed. In addition, the gross energy value of brown and green seaweed was measured. A digestibility trial was conducted to determine the apparent ileal digestibility of seaweed in broiler chickens. Apparent metabolisable energy was determined as well in this study. Birds were fed with 90.30% seaweed-based diet with an indigestible marker. At the end of the feeding trial, birds were euthanised and ileal digesta was collected. Nutrient contents of experimental feed and digesta were analysed, and gross energy was measured. The results revealed that there was no significant difference in the apparent ileal digestibility of dry matter, organic matter, crude lipid and ash contents among the brown and green seaweed-based diets. The findings also demonstrated that the apparent ileal digestibility of crude protein and crude fibre was significantly higher in brown seaweed compared to green seaweed. Nevertheless, no significant difference was observed in the apparent digestibility of metabolisable energy between the types of seaweed.This study aimed to analyse the nutritional properties, apparent ileal digestibility (AID) and apparent metabolisable energy (AME) of broiler chickens fed with brown seaweed (BS) and green seaweed (GS). Proximate analysis was performed to determine the nutrient composition of seaweed. The amino acids were determined using high-performance liquid chromatography (HPLC), and atomic absorption spectroscopy was used to determine the minerals content. The gross energy (GE) was determined using a fully automatic bomb calorimeter, and the AME value was calculated. Titanium dioxide (TiO2) was used as an indigestible marker to calculate the AID. A digestibility trial was conducted to investigate the effects of seaweeds on crude protein (CP), crude fibre (CF), ether extract (EE), dry matter (DM), organic matter (OM), amino acids (AA) and minerals digestibility, and AME on broiler chickens. Thirty-six broiler chickens were randomly distributed into two dietary treatment groups with six replicates and three birds per replicate. Results showed that brown and green seaweed was a source of macro and micronutrients. For the AME and AID of seaweed-based diets, the results showed that the AME value for BS and GS was 2894.13 and 2780.70 kcal/kg, respectively. The AID of BS and GS was 88.82% and 86.8% for EE, 82.03% and 80.6% for OM, 60.69% and 57.80% for CP, 48.56 and 44.02% for CF, and 17.97 and 19.40% for ash contents, respectively. Meanwhile, the AID of CP and CF was significantly higher for BS compared to the GS. Findings showed that the AID of various AA was 40.96 to 77.54%, and the AID of selected minerals (Ca, Na, K, Mg, Zn, Cu, Fe) for both BS and GS groups were above 90%.

Modulation of jejunal mucosa-associated microbiota in relation to intestinal health and nutrient digestibility in pigs by supplementation of β-glucanase to corn-soybean meal-based diets with xylanase.

This study aimed to evaluate the effects of increasing levels of β-glucanase on the modulation of jejunal mucosa-associated microbiota in relation to nutrient digestibility and intestinal health of pigs fed diets with 30% corn distiller’s dried grains with solubles and xylanase. Forty pigs at 12.4 ± 0.5 kg body weight (BW) were allotted in a randomized complete block design with initial BW and sex as blocks. Dietary treatments consisted of a basal diet with xylanase (1,500 endo-pentosanase units [EPU]/kg) and increasing levels of β-glucanase (0, 200, 400, and 600 U/kg) meeting nutrient requirements and fed to pigs for 21 d. Blood samples were collected on day 19. On day 21, all pigs were euthanized to collect intestinal tissues and digesta. Tumor necrosis factor-alpha, interleukin (IL)-6, and malondialdehyde were measured in the plasma and mid-jejunal mucosa. Viscosity was determined using digesta from the distal jejunum. Ileal and rectal digesta were evaluated to determine apparent ileal digestibility (AID) and apparent total tract digestibility (ATTD) of nutrients. Mucosa samples from the mid-jejunum were utilized for microbiota sequencing. Data were analyzed using the MIXED procedure on SAS 9.4. Overall, increasing dietary β-glucanase tended to increase (linear; P = 0.077) the average daily gain of pigs. Increasing dietary β-glucanase affected (quadratic; P < 0.05) the relative abundance of Bacteroidetes, reduced (linear; P < 0.05) Helicobacter rappini, and increased (linear, P < 0.05) Faecalibacterium prausnitzii. β-Glucanase supplementation (0 vs. others) tended to increase (P = 0.096) the AID of crude protein in the diet, whereas increasing dietary β-glucanase tended to increase (linear; P = 0.097) the ATTD of gross energy in the diet and increased (linear; P < 0.05) the concentration of IL-6 in the plasma of pigs. In conclusion, increasing β-glucanase up to 600 U/kg feed in a diet containing xylanase (1,500 EPU/kg) modulated mucosa-associated microbiota by increasing the relative abundance of beneficial bacteria and reducing potentially harmful bacteria. Furthermore, increasing β-glucanase up to 600 U/kg feed in a diet containing xylanase (1,500 EPU/kg feed) enhanced the status of the intestinal environment and nutrient utilization, as well as reduced systemic inflammation of pigs, collectively resulting in moderate improvement of growth performance. Supplementing β-glucanase at a range of 312 to 410 U/kg with xylanase at 1,500 EPU/kg feed showed the most benefit on jejunal mucosa-associated microbiota and reduced systemic inflammation of pigs.

Growth performance, nutrients digestibility, caecum microbiota, antioxidant status and immunity of broilers as influenced by kombucha fermented on white sugar or sugar beet molasses

The purpose of this study was to evaluate the effects of fermented kombucha on green tea with white sugar or sugar beet molasses in broiler chickens. Birds (n = 448) were randomly allotted into seven treatments of four replicates each. Treatments were included: tap water with no kombucha (Control), water containing 3 ml/100 ml kombucha based on white sugar (SK3%), water containing 6 ml/100 ml kombucha based on white sugar (SK 6%), water containing 3 ml/100 ml kombucha based on sugar beet molasses (MK3%), water containing 6 ml/100 ml kombucha based on sugar beet molasses (MK6%), SK 3%+MK 3% and SK 6%+MK 6%. Except for SK3%, all birds received SK or MK in their drinking water had greater (p ≤ .05) BWG than control. Feed intake increased (p ≤ .05) in SK6% group in comparison to the control. All SK or MK treatments resulted in lower (p ≤ .05) FCR in comparison with the control. Birds received the MK6% and SK6%+MK6% treatments showed greater (p<.05) apparent ileal digestibility of CP. ALT levels decreased (p ≤ .05) in birds received all kombucha treatments. The SK and MK treatments decreased (p ≤ .05) total coliform counts in caecum whereas the lactobacillus population increased (p ≤ .05) in birds received 6% of SK, MK or their combination. The SK6%+MK6% and MK6% groups had greater (p ≤ .05) antibody titre against NDV. Plasma and liver activities of CAT and SOD were greater (p ≤ .05) in kombucha treatments in comparison with the control group. In conclusion, kombucha fermented on white sugar or sugar beet molasses showed growth promoting effect in broilers and addition of 6 ml fermented kombucha on sugar beet molasses per 100 ml of drinking water result in the best performance. PSIV-21 Pellet processing reduces variability of corn soybean meal-based diet digestibility in growing pigs

Abstract This study was conducted to determine the effect of pelleting on the digestibility of corn-soybean meal-based diet in growing pigs. Two trials with 6 pigs cannulated at the distal ileum were conducted. In each trial, pigs were assigned to each treatment following a crossover design. In each experiment, the same diet, composed of corn and soybean meal with 10% wheat from two different feed mills, was served in pellet or mash form. Pelleting allowed an increase in digestibility in one of the trials. Apparent ileal digestibility (AID) of dry matter (DM), crude protein (CP) and digestible energy (DE) were improved with pelleting by 8, 12 and 9% (P&amp;lt; 0.01). The AID of amino acids (AA) was also improved (P&amp;lt; 0.05). Apparent total tract digestibility (ATTD) was increased by pelleting in DM, CP and DE by 5, 7 and 6% respectively (P&amp;lt; 0.01). The digestibility of the mash diet in experiment 1 was lower than in the pelleted diet in the experiment 1 and both diets in experiment 2 as shown by the interaction Pelleting X Trial which was significant for the AID and ATTD of DM, CP and DE (P&amp;lt; 0.01). Therefore, in experiment 1, pelleting allows to improve the digestibility of diet to the same level as in experiment 2. The AID of CP was higher by 37% in the mash diet from the second experiment compared to the one in the first experiment. Even though the same ingredients were chosen in the two experiments, this shows the variability in digestibility existing between different feed mills and ingredient sources. This difference was not observed in pelleted diets. The results obtained in these two trials show that pelleting can reduce the variability of digestibility and then give a good digestibility of diets even if the ingredients are of different quality or sources.

183 Isoquinoline alkaloids increase apparent ileal digestibility of amino acids and crude protein and apparent total tract digestibility of crude protein by finishing pigs fed corn-soybean meal diets

Abstract An experiment was conducted to determine if the inclusion of a preparation of isoquinoline alkaloids (IQ) in corn-soybean meal diets fed to finishing pigs increases the apparent ileal digestibility (AID) of amino acids (AA) and crude protein (CP) and the apparent total tract digestibility (ATTD) of CP and gross energy. Twelve ileal cannulated barrows (BW: 77.2 ± 6.23 kg) were allotted to a triplicated 4 x 3 incomplete Latin square design with 4 diets and 9 replicates per diet. Diets were supplemented with either 0, 40, 80, or 160 mg/kg IQ and with 0.40% titanium dioxide as an indigestible marker. Each collection period lasted 14 d with d 1 through 10 as an adaptation to the diets, fecal samples were taken using the grab sampling technique on d 11 and 12, and ileal digesta were collected for 8 h on d 13 and 14. Data were analyzed using the Proc MIXED procedure of SAS. Contrast statements were used to determine linear and quadratic effects of including IQ in the diets. Results were considered significant at P ≤ 0.05 and considered a trend at 0.05 &amp;lt; P ≤ 0.10. Inclusion of IQ resulted in a quadratic increase (P &amp;lt; 0.05) in the AID of CP and AA in corn-soybean meal diets fed to finishing pigs, with the greatest digestibility values generally observed in the 40 mg/kg IQ treatment. Additionally, IQ resulted in a quadratic increase (P = 0.009) in the ATTD of CP, with the greatest digestibility observed in the 80 mg/kg IQ treatment. Therefore, it was concluded that supplementing corn-soybean meal diets with 40 or 80 mg/kg IQ may increase the AID of CP and AA and ATTD of CP by finishing pigs.

Effect of particle size of insoluble fibre on growth performance, apparent ileal digestibility and caecal microbial population in broiler chickens fed barley-containing diets

ABSTRACT 1. This study was conducted to investigate the effect of particle size of insoluble fibre on growth performance, apparent ileal digestibility (AID) and caecal microbial population in broiler chickens fed barley-containing diets. 2. The dietary treatments included: a barley-based diet (control, CTL) or test diets which contained high-fibre ingredients, either sunflower hulls (SFH), sugarcane bagasse (SB) or wheat bran (WB) ground through a 1.0 (fine) or 3.0 mm (coarse) screen that were added to the control diet at 30 g/kg. 3. For the entire experimental period, insoluble fibre inclusion improved ADG (P < 0.05) and FCR (P < 0.05) compared to the CTL group. Broilers fed SFH had higher (P < 0.05) ADG and better (P < 0.05) FCR than broilers fed SB. 4. Fibre inclusion increased the relative weight of breast and thigh and decreased relative weight of liver compared to the CTL group, but coarse grinding of the SB decreased relative weight of abdominal fat (P < 0.05). 5. The digestibility of nutrients increased with dietary inclusion of insoluble fibre compared to the CTL group. Coarse grinding of SFH increased AID of crude protein compared to the coarse grinding of WB or SB. Fine grinding of various types of fibre improved the AID of fat (as ether extract) and organic matter (P < 0.05). 6. Fine grinding of the WB decreased caecal populations of E. coli. The inclusion of SFH increased caecal populations of Lactobacillus spp. (P < 0.05). Coarse grinding of various types of fibre decreased the caecal population of coliforms (P < 0.05). 7. The inclusion of WB increased digesta viscosity in the ileum compared to samples from the SB and SFH groups (P < 0.05). Coarse grinding of various types of fibre decreased the digesta viscosity in the ileum (P < 0.05). 8. Overall, the data showed that dietary inclusion of insoluble fibre improved growth performance, increased AID of nutrients and decreased ileal viscosity in the birds fed diets containing barley.

Short communication: A pilot study to describe duodenal and ileal flows of nutrients and to estimate small intestine endogenous protein losses in weaned calves

The aims of this pilot study were (1) to evaluate the effect of an ileal and duodenal cannulation surgery on body weight and dry matter intake, (2) to estimate endogenous losses of crude protein (CP) and AA in the small intestine, and (3) to describe duodenal and ileal flows of nutrients in weaned dairy calves. Three Holstein male calves were fitted at 7 wk of life with a T-cannula at the terminal ileum and another cannula at the proximal duodenum. On wk 14 of life, calves were randomly assigned to a single 3 × 3 Latin square with 10-d periods. The 3 diets were fed ad libitum and consisted of a control calf starter (CS) with conventional soybean meal (SBM) as the main source of protein (CTRL), an isonitrogenous (20% CP) CS with an enzyme-treated SBM as the main source of protein (ENZT), and a CS with low content of CP (10%) and no soy protein (LOCP). Flows and digestibilities of nutrients were compared between the soy-based high-protein diets (HICP) and LOCP, and between CTRL and ENZT. Final data were only available from 2 calves per diet (n = 2) because cannulas from 1 calf became inoperative after the first collection period. Duodenal flows of CP, total AA, nonprotein nitrogen, microbial N, and fatty acids, as well as apparent duodenal digestibility of starch, were greater for HICP than for LOCP, indicating a greater foregut microbial activity and digestion. The apparent ileal digestibility (AID) of organic matter, CP, and total AA were greater for HICP than for LOCP. Duodenal net flow of CP was greater for ENZT than for CTRL, but flow of AA was not different. On the other hand, duodenal flow of microbial N was greater, and flows of nonprotein N and starch were lower for the ENZT diet, suggesting a more efficient microbial activity in the rumen. Even though CTRL had a greater AID when compared with ENZT, the AID of CP and AA were greater for enzyme-treated SBM than for SBM. Endogenous losses in the small intestine per kilogram of duodenal dry matter flow were 47 ± 15 and 37 ± 12 g/d, and the true ileal digestibilities for the HICP diets were 86 ± 0.1 and 87 ± 0.1% for CP and AA, respectively. An optimal supply of CP and the inclusion of an enzyme-treated SBM improved the efficiency of microbial digestion and increased AA absorption. Although further research with greater biological replication is needed, our results indicate that there is potential to improve digestion and absorption of proteins through dietary strategies in young weaned calves.
