Correction of peroxidation of lipids with upadacitinib, a Janus kinase 1 inhibitor, in patients with true eczema

Objective. To study the effect of upadacitinib (UPA), a Janus kinase inhibitor 1 kinase, on the status of peroxidation of lipids (POL) and the antioxidant system in patients with true eczema. Subjects and methods. The investigation involved 64 patiens (43 males and 21 females) with true eczema (the mean age was 58.5±3.4 years). The patients were randomized into 2 groups: 1) 33 patients who received basic drug therapy (BDT); 2) 31 patients who received the Janus kinase 1 inhibitor UPA 15 mg/day during 2 months. The components of the antioxidant system and the state of POL were determined by enzyme immunoassay. Results. After 2 months of therapy, Group 1 showed no statistically significant changes in the analyzed parameters from the baseline level (p>0.05). In Group 2, the total oxidative capacity of blood reduced by 1.27 tines; the level of oxidized low-density lipoproteins decreased by 1.28 times; the total antioxidant activity of blood increased by 1.24 times; the level of superoxidase dismutase increased by 1.43 times (p < 0.001 for all values). Conclusion. The combined use of BDT and the JAK1 inhibitor UPA in patients with true eczema contributed to a statistically significant decline in the oxidative capacity of blood and POL products along with the enhanced antioxidant activity of blood.

PECULIARITIES OF OXIDATIVE STRESS IN PATIENTS WITH ACUTE MYOCARDIAL INFARCTION WITH IRON DEFICIENCY ANEMIA

The article presents changes in hemogram and oxidative stress in patients with acute myocardial infarction (AMI) on the background of iron deficiency anemia (IDA). Iron deficiency anemia is an additional factor that contributes to the deepening of myocardial ischemia and deepens the processes of peroxidation and damage to cardiomyocytes, which is an important factor in the unfavorable course, both acute period and recovery processes in myocardial necrosis. In order to study the indicators of the general analysis of blood and functional state of the antioxidant system in patients with acute myocardial infarction with IDA, 36 patients with AMI with IDA were examined, including 39% men and 61% women. The first group consisted of 10 patients with AMI without IDA, the second 26 patients with AMI and IDA of varying severity, the control group consisted of 30 healthy individuals. Hemogram parameters, glutathione system function, oxyproline, arginase and magnesium levels in the blood were determined. It has been established that women with mild and moderate anemia predominate among patients with acute myocardial infarction with concomitant iron deficiency anemia. Among men, half of the patients have severe anemia. In addition to a decrease in Hb levels, patients with ACS with IDA have the following laboratory signs of anemia: a tendency to increase the average concentration of hemoglobin in one erythrocyte, low serum iron levels and an increase in serum transferrin levels. In the presence of IDA in patients with ACS there are changes in the antioxidant system. Anemic syndrome in such patients is accompanied by increased concentrations of glutathione transferase and peroxidase, as well as decreased concentrations of oxyproline.
 Anemia can trigger oxidative stress, and an increase in OS may be associated with changes in cardiac function. Possible cardiovascular effects in patients with comorbid conditions should also be considered.
 Iron deficiency is the most prevalent micronutrient disorder globally. When severe, iron deficiency leads to anemia, which can be deleterious to cardiac function. Given the central role of iron and oxygen in cardiac biology, multiple pathways are expected to be altered in iron-deficiency anemia, and identifying these requires an unbiased approach.
 Oxidative stress is an imbalance between free radicals and antioxidant molecules that can play an important role in the pathogenesis of iron-deficiency anemia (IDA).
 Maintenance of the iron homeostasis is essential for metabolic and physiological processes. Iron plays a critical role in erythropoiesis, is incorporated into erythroblasts and reticulocytes, and has a crucial role in oxygen transport and oxygen storage. Moreover, iron is essential for cardiac and skeletal muscle metabolism, the synthesis and degradation of proteins, lipids, ribonucleic acids, and mitochondrial function.
 Research has now focused on non-traditional cardiovascular risk factors and the roles of iron and iron deficiency (ID) in patients with cardiovascular disease.
 The Iron required for immune response, hormonal balance, and plays an important role in regulating oxidative stress and aerobic metabolism. The myocardial tissue has a mainly aerobic cellular metabolism, which depends of mitochondria's Krebs cycle enzymes that need iron as an essential cofactor. In this regard, there is some evidence that myocardial iron deficiency is highly prevalent in HF and may play a role in the progression of the disease.

Relationship between Serum Gamma-Glutamyltransferase and Glutathione Antioxidant System in Patients with Liver Cirrhosis of Various Etiology.

Oxidative stress involvement in liver diseases has been extensively studied. A direct assessment of the reactive species incriminated is avoided due to their short lifespan and high cost. For these reasons an inexpensive and easy to perform test for whole body oxidative stress is highly desired. This pilot study was conducted to assess the relationship between γ-glutamyl transferase (GGT) activity and markers of oxidative stress: reduced glutathione (GSH), glutathione peroxidase (GPx) activity and lipid peroxidation in patients with liver cirrhosis due to chronic ethanol consumption and viral hepatitis. Forty-eight patients with alcoholic liver cirrhosis and cirrhosis developed after HBV and HCV infection were included in this study.Blood GSH andGPxand serum GGT and MDAwere assayed and the results were statistically analyzed. The activity of serum GGT was significantly higher in the alcoholic group. The relationship between GGT activity, GSH and MDA levels was different between groups.A strong significant positive correlation between GGT and GSH was noticed for the patients from GGT Q3 and Q4 quartiles in the group of viral liver cirrhosis, while for alcoholics the relationship between GGT and GSH showed the trend for a negative correlation.The values of serum MDA differ significantly between groups (p<0.015); a very significant variation was observed at low levels of GGT activity (p<0.006). Our study demonstrates that the GSH antioxidant defense system is more compromisedin alcoholic cirrhosisand tends to correlate negatively with GGT. Even in its normal range GGT might be an early and sensitive marker of oxidative stress.

Assessment of the state of the oxidant-antioxidant system and lipid peroxidation in patients with chronic pancreatitis combined with hypothyroidism

Changes in oxidative-antioxidant homeostasis is one of the pathogenetic links in the onset and progression of many nosologies, including chronic pancreatitis (CP) and hypothyroidism (HТ), however, data on the development of oxidative stress (OS) with a combination of CP and HP are insufficient, therefore, determining the role of these systems with a comorbid course of these pathologies is an urgent issue of our time.The aim of the work is to study the state of the oxidative and antioxidant system in patients with CP and CP combined with HT.Material and methods. 96 people were examined, including 26 patients with chronic pancreatitis (group 1), 24 patients with hypothyroidism (group 2), 26 patients with chronic pancreatitis on the background of hypothyroidism (group 3), 20 almost healthy individuals (group 4). Evaluation of the oxidative-antioxidant system and lipid peroxidation was assessed by determining the content of malonic aldehyde in blood plasma (MA pl.) And erythrocytes (MA er.), The level of reduced glutathione, glutathione peroxidase (GP) and glutathione-S-trans-ion (GT). Enzyme activity was calculated per 1 g of hemoglobin (HB).Research results. The analysis of the obtained results revealed clear patterns of changes in the state of the oxidative-antioxidant system and lipid peroxidation in patients with chronic pancreatitis on the background of hypothyroidism. Namely, violation of lipid peroxidation in patients with comorbid pathology, accompanied by an increase in the content of MA er. by 44.87% and the content of MA in blood plasma by 46.42%; oxidative-antioxidant system: decrease in HF in the blood by 51,61%, combined of a compensatory increase in BP - by 48.03% and HT - by 34.76%.Conclusion. Under the conditions of comorbidity of CP and HT, there is a violation of the oxidative-antioxidant system and the state of lipid peroxidation, which is characterized by a significant increase in MA in erythrocytes and MA in plasma against a decrease in glutathione and which complicates the course of the underlying disease, its diagnosis and treatment.

Study of the role of lipid peroxidation and antioxidant system in patients with infectious diseases is of great interest to researchers. Numerous studies have led to a common understanding of their contribution to the pathogenesis of infectious diseases, as well as to the complications development. However, the state of lipid peroxidation and antioxidant protection in patients with chickenpox (varicella) has not been sufficiently studied which is important for the development of new targeted treatments.&#x0D; The aim of the research - to assess the state of lipid peroxidation (LPO) and the antioxidant system (AOS) in the dynamics in patients with chickenpox, depending on the severity of the disease.&#x0D; Materials and methods. We selected for participating in the study 240 chickenpox patients (142 women and 98 men) aged 18-40 years. Chickenpox was diagnosed using clinical, serological, and molecular genetic methods. The state assessment of LPO in chickenpox patients was based on the determination of serum diene conjugates (DC), malondialdehyde (MDA) levels and the total oxidative activity (TOA) of blood plasma.&#x0D; Results. Study showed that the process of lipid peroxidation increases and the activity of the antioxidant system decreases in patients with chickenpox during the acute period of the disease. The extent of these processes becomes greater as the severity of chickenpox increases. A decrease in the activity of antioxidant enzymes and a disruption in the functioning of the glutathione system in patients with moderate to severe hypertension leads to disruption of protective mechanisms and, as a result, to an increase in the free radical chain reactions, the uncontrolled growth of which causes irreversible damage to the membranes of various cells, which underlies visceropathy in chickenpox patients.&#x0D; Conclusions. Antioxidant system is activated in the acute period in patients with mild chickenpox. It is evidenced by an increase in the overall antioxidant activity of blood plasma (p=0.045) and red blood cells (p=0.00087), in the activity of catalase (p=0.001), superoxide dismutase (p=0.0093), glutathione peroxidase (p=0.036), glutathione reductase of plasma and red blood cells, and an increase in the concentration of oxidized and reduced glutathione in blood. Lipid peroxidation is hyperactivated in the acute period in patients with a moderate and severe course of the disease and an excess amount of primary and secondary hydroperoxides of fatty acids accumulates in the blood. This is accompanied by indicates a developing imbalance between the oxidative and antioxidant systems in group of patients with moderate and severe chickenpox. Our findings confirm the usefulness of antioxidants in the treatment of chickenpox.

Dietary and Lifestyle Factors Modulate the Activity of the Endogenous Antioxidant System in Patients with Age-Related Macular Degeneration: Correlations with Disease Severity.

Age-related macular degeneration (AMD) is a common cause of blindness in the elderly population, but the pathogenesis of this disease remains largely unknown. Since oxidative stress is suggested to play a major role in AMD, we aimed to assess the activity levels of components of the antioxidant system in patients with AMD. We also investigated whether lifestyle and dietary factors modulate the activity of these endogenous antioxidants and clinical parameters of disease severity. We recruited 330 patients with AMD (39 with early, 100 with intermediate and 191 with late form of AMD) and 121 controls in this study. At enrolment, patients’ dietary habits and physical activity were assessed, and each study participant underwent a thorough ophthalmologic examination. The activity of several components of the antioxidant system were measured in red blood cells and platelets using both kinetic and spectrophotometric methods. Patients with AMD consumed much lower levels of fatty fish and eggs than the control group (p = 0.008 and p = 0.04, respectively). In the nAMD group, visual acuity (VA) correlated positively with green vegetable consumption (Rs = +0.24, p = 0.004) and omega-3-rich oil intake (Rs = +0.17, p = 0.03). In the AMD group, the total physical activity MET score correlated positively with VA (Rs = +0.17, p = 0.003) and correlated negatively with the severity of AMD (Rs = −0.14, p = 0.01). A multivariate analysis of patients and controls adjusted for age, sex, and smoking status (pack-years) revealed that AMD was an independent variable associated with a lower RBC catalase (β = −0.37, p < 0.001) and higher PLT catalase (β = +0.25, p < 0.001), RBC GPx (β = +0.26, p < 0.001), PLT GPx (β = +0.16, p = 0.001), RBC R-GSSG (β = +0.13, p = 0.009), PLT R-GSSG (β = +0.12, p = 0.02) and RBC GSH transferase (β = +0.23, p < 0.001) activity. The activities of components of the antioxidant system were associated with disease severity and depended on dietary habits. The observed substantial increase in the activity of many critical endogenous antioxidants in patients with AMD further indicates that the required equilibrium in the antioxidant system is disturbed throughout the course of the disease. Our findings explicitly show that a diet rich in green vegetables, fish and omega-3-rich oils, supplemented by physical exercise, is beneficial for patients with AMD, as it might delay disease progression and help retain better visual function.

Background It is known that the alteration of antioxidants can been seen in early phase after traumatic brain injury (TBI) in order to block oxidative damage, but little is known about the influence of sex on antioxidant system in patients with TBI. This study investigates whether there are sex differences in these endogenous antioxidant agents during the acute phase after TBI and their association with the disease. Methods Serum levels of uric acid (UA), bilirubin, albumin and creatinine were measured in 421 individuals included 157 female TBI patients, 156 male TBI patients and 108 age- and sex-matched controls. Results The statistically significant changes were found in UA, bilirubin, albumin and creatinine for both sexes with TBI, but the trend of changes in bilirubin and creatinine was opposite for gender groups. Serum levels of UA, bilirubin, albumin and creatinine were associated with the severity of TBI patients for both sexes. Male patient subgroups with elevated UA, albumin and creatinine had higher frequency of regaining consciousness in a month. Moreover, addition of UA and creatinine to the established clinical model had significantly improved the predictive performance over using clinical model alone in male patients with TBI. However, no similar findings were observed on female TBI patients. Conclusion Our results suggest sex-based differences in the serum endogenous antioxidant response to TBI. Use of serum UA and creatinine could help in the outcome prediction of male patients with TBI in combination with other prognostic factors.

Aim: to investigate the intensity of proteins and lipids peroxidation, the state of the antioxidant system and biochemical indices of endogenous intoxication in patients with snoring. Materials and Methods: Blood serum of 20 patients with snoring and 19 healthy persons without any manifestation of respiratory complications at sleeping were studied. All patients were divided into two groups. Group I included 10 patients with moderate manifestation of snoring, and group II with the hard one. Results: An increase of the proteins’ oxidative modifications in patients with snoring without obstructive apnea at sleep were moderate and severe, that is reflected by an increase of the levels of neutral- and basiccharacter aldehyde-dinitrophenyl-hydrazones and basic-character ketone-dinitrophenyl-hydrazones has been established. The level of thiobarbiturate-positive products in patients with the moderate degree of snoring was in the value of control group. At progression of this pathology this index increased, too in compare to control group. The level of catalase activity in patients of both group was increased where as the content of free thiol groups was decreased. The content of medium weight molecules in patients of both groups was on the level of control, but at progressing of the disease it increased. The content of tyrosine-containing peptides was increased in the serum of patients in both groups. Our data prove for the intensification of the proteins’ and lipids’ peroxidation, for the changes in antioxidant system and for the development of endogenous intoxication in patients with severe level of snoring.

Antioxidant system in implementation of endotoxicosis syndrome in young persons with comorbidity of gastroesophageal reflux disease and autoimmune thyroiditis

Introduction. In recent years, there has been an increase in the prevalence of gastroesophageal reflux disease and autoimmune thyroiditis in young people. This is associated with a hereditary predisposition, viral diseases, which are often recorded in children's and adolescent groups, the unreasonable use of a large number of drugs, as well as the influence of various negative environmental factors, instability of the autonomic nervous system, eating disorders, the presence of bad habits, chronic stress and others. The aim of the work was to investigate the state of total antioxidant status and the content of biomarkers of the second phase of detoxification of xenobiotics - manganese superoxide dismutase and glutathione peroxidase in young people with gastroesophageal reflux disease and autoimmune thyroiditis. Materials and methods. To achieve this goal, 120 patients with a combined course of gastroesophageal reflux disease (GERD) and autoimmune thyroiditis (AIT) were examined, which made up the main group and two comparison groups: with isolated GERD (45 people) and AIT (42 patients). All patients were students of various universities and were between the ages of 18 and 25. Duration of the disease did not exceed 3 years. The total antioxidant status as an indicator of the first phase of defense and the levels of mitochondrial manganese superoxide dismutase and glutathione peroxidase – the second phase of antioxidant defense was determined. Results. The study showed that in all surveyed groups there was a decrease of total antioxidant status, which characterizes the first phase of the antioxidant defense system. Changes in the first phase of antioxidant defense affect the indicators of the second level of the system, which became the basis for studying the activity of mitochondrial manganese superoxide dismutase. Thus, the value of this indicator in all examined individuals exceeded the norm, which confirmed the inclusion of the second controlling phase of antioxidant system into the pathological process. At the same time, an increase inhibition of the glutathione defense link, the second phase of xenobiotic detoxification, were diagnosed. Conclusions. Negative changes occur in the enzyme system of the first line of antioxidant system in patients with GERD and AIT which may be the result of an unfavorable autoimmune background. Control over the work of antioxidant system moves to the second phase of the system, manifested by an increase in the content of mitochondrial manganese superoxide dismutase and inhibition of the activity of glutathione peroxidase in patients with a combination of GERD and AIT.

INDICATORS OF OVARIAL RESERVE, PRO- AND ANTIOXIDANT SYSTEM, CYTOKINE STATUS OF PERIPHERAL BLOOD AND PERITONEAL LIQUID IN PATIENTS WITH ENDOMETRIOID OVARIAN CYSTS

The aimof this work was to identify the characteristics of the cytokine status and state of the pro- and antioxidant systems in patients of late reproductive age with endometrioid cysts and determine their role in changing the ovarian reserve.Materials and methods.A total of 89 patients with endometrioid ovarian cysts (EOC) were examined. The control group consisted of 35 healthy patients admitted for surgical sterilisation. Peripheral blood was collected before surgery, while the peritoneal fluid collection was performed intraoperatively. In peripheral blood and peritoneal fluid, the level of cytokines was determined. The calculation of the anti-inflammatory index was performed, along with the evaluation of the content of lipid peroxidation products and the antioxidant activity. The analysis of the ovarian reserve parameters was conducted.Results.In patients of late reproductive age suffering from EOC, a decrease in the ovarian reserve is observed under the preserved hormonal background. The presence of EOC is accompanied by the activation of lipid peroxidation, both at the systemic and local levels, under a pronounced inhibition of the antioxidant defence link. At the systemic and local levels, patients with EOC experience an imbalance in the cytokine system with a significant activation of the pro-inflammatory component.Conclusion.The disruption of oxidative homeostasis and activation of immune inflammation, detected in patients of late reproductive age with EOC, may lead to the damage of the structural cell components of the functional ovarian tissue, thereby reducing the ovarian reserve.Conflict of interest: the authors declare no conflict of interest.

Aim.&nbsp;To identify infertility prognostic factors&nbsp;of thepro-/&nbsp;antioxidant system in men with left-side varicocele grade II-III. Materials and methods. The trial includes 67 patients with left-side varicocele grade II-III and concomitant changes in spermogram and / or infertility. During the follow up, three months after varicocelectomy, 62 patients were reexamined. The control group consisted of 25 virtually healthy men aged 19 to 33 years. Enzymes activity of pro-/antioxidant system in the blood plasma from periferal and from testicular veins, as well as in the spermatozoa were measured, before and 3 months after varicocelectomy. In particular, the activity of glutathione peroxidase, glutathione reductase and glutathione transferase, the concentration of reduced glutathione and malonic dialdehyde were determined. In addition, the activity of arginase, constitutive and inducible isoforms of nitric oxide synthase, the activity of ion-transport systems of sodium, potassium and calcium, magnesium adenosine triphosphatase&nbsp;in the&nbsp;spermatozoa were studied. Results and Discussion.&nbsp;The activity of glutathione antioxidant enzymes and the concentration of malonic dialdehyde as well as reduced glutathione in the serum of the internal spermatic veins in men with left side varicocele grade II-III were significantly different in comparison groups. Thus, a statistically significant reduction in the concentration&nbsp; of the glutathione-peroxidase system enzymes and of reduced glutathione in the serum of the internal&nbsp;spermaticvein were observed against the background of a reliable increase in the level of malonic dialdehyde compared with the concentrations of these parameters in serum of peripheral venous blood, and even more so with norm. Therefore, the ROC analysis of the above enzymes’&nbsp;activity of the pro-/antioxidant system in serum from the internal spermatic vein of patients with left side varicocele grade II-III revealed certain patterns. Concentrations of glutathione-peroxidase system enzymes and reduced glutathione in spermatozoa of patients with left side varicocele grade II-III by analogy with the differences observed in the serum of the internal spermatic vein of these same patients, were characterized by a significant lag behind the rates in practically healthy men. In contrast, the level of malonic dialdehyde statistically significantly exceeded the norm. The obtained results confirm the thesis about the exhaustion of the compensatory mechanisms of the glutathione antioxidant system and increase the intensity of lipoperoxidic processes in spermatozoa in men with left-side varicocele grade II-III. The reduction in serum of the internal testicular vein&nbsp;of&nbsp;glutathione peroxidase activity&nbsp;to&nbsp;£148,55 nmol GSH/min·g Hb,&nbsp;of&nbsp;glutathione reductase&nbsp;to&nbsp;£32.7 mcmol NADPH/min·g Hb,&nbsp;and glutathione transferase&nbsp;to&nbsp;£137 nmol GSH/min·g Hb, as well as reduced glutathione&nbsp;to£0.85 mmol/l with malondialdehyde growth greater than 7,9 mcmol/l should be attributed to the prognostic predictors of the probability of infertility development in patients with left-side varicocele grade II-III. Results showed, that the most significant predictors of the probability of infertility development in spermatozoa were decreased activities of arginase&nbsp;£46 pmol citrulline/min&nbsp;·1 mg protein and inducible nitric oxide synthase &gt;1,2 pmol citrulline/min·1 mg protein. Conclusions.&nbsp;The negative influence of the reactive oxygen species on the quality of the ejaculate remains a relevant issue. Therefore, the practical application of the data about antioxidant imbalance in patients with varicocele requires further, more detailed study. Changes in fertility status in patients with varicocele leads to decreased levels of antioxidant and increased levels&nbsp;of&nbsp;prooxidant enzymes in the blood and spermatozoa wich can be used as a predictors of&nbsp;maleinfertility. Indicators of the pro-/antioxidant system have proven themselves as predictors of infertility in patients with varicocele.

Features of antioxidant system in patients with obstructive jaundice of benign and malignant origin in dynamics

Aim. Study of the features of changes of antioxidant protection in patients with mechanical jaundice of benign and malignant origin in dynamics. Methods. The author studied the role of antioxidant system in the progression of mechanical jaundice of various origins in 104 patients. Groups of patients with a syndrome of mechanical jaundice of benign (62 patients) and malignant origin (42 patients) were isolated. The material of the study was blood from the ulnar vein, which was taken in the morning on an empty stomach before surgery. In the dynamics (on the 7th day after the operation), 53 patients were examined. To assess the state of antioxidant protection, a spectrophotometric method of investigation was used. Results. A high level of malonic dialdehyde, the product of lipid peroxidation, was determined in all patients regardless of origin. There was also a depression of the enzymatic link of antioxidant protection (a decrease in the activity of superoxide dismutase and catalase). After surgical intervention in patients with mechanical jaundice of benign origin, correction of the level of reduced glutathione was observed. In patients with mechanical jaundice of malignant origin in dynamics, the activity of antioxidant enzymes did not differ significantly from the norm (p1-3 = 0,23; p1-3 = 311). Conclusion. After surgical intervention, partial improvement of the condition of patients with mechanical jaundice of benign origin was observed, and dysfunction of antioxidant protection persisted in patients with mechanical jaundice of malignant origin.

Раннее выявление нарушений антиоксидантной системы, которые появляются уже на стадии доклинических проявлений, позволят предотвратить или улучшить течение осложнений связанных с сахарным диабетом.Цель:Оценить состояние антиоксидантной системы у пациентов с различными сосудистыми осложнениями СД 2типа, такими как диабетическая ретинопатия (ДР) и ангиопатия сосудов нижних конечностей.Материалы и методы:В исследовании принимали участие три группы пациентов, всего 75 человек в возрасте 55±6,0лет. Первая группа пациентов (n=25) включала больных СД 2 типа (инсулиннезависимый) с наличием диабетической ретинопатии. Во второй группе пациентов (n=25), течение СД 2типа (инсулиннезависимый), протекало с развитием ангиопатии сосудов нижних конечностей. Третью группу составили 25 здоровых людей.Определение исследуемых показателей производили ВЭЖХ методами, разработанными в нашей лаборатории.Результаты исследований и обсуждение:Полученные данные свидетельствуют о том, что антиоксидантный потенциал плазмы (Cysfree/Cys-S-S-Cys) крови при сосудистых осложнениях СД 2типа уменьшается и окислительно-восстановительное состояние GSH/GSSG изменяется в зависимости от различных видов сосудистых осложнений СД 2типа.Выводы:При развитии сосудистых осложнениях СД 2типа страдает как антиоксидантный потенциал плазмы, так и непосредственно клетки, что выражается в значимом уменьшении восстановленных форм определяемых показателей.При прогрессировании заболевания и наличии диабетической ангиопатии коэффициенты Cysfree/Cys-S-S-Cys и GSH/GSSG достоверно отличаются от таковых в группе контроля. Опираясь на это, можно с уверенностью говорить о том, что у пациентов с СД 2типа необходима полная оценка антиоксидантной системы, с изучением различных фракций не только глутатиона, но и цистеина для своевременной фармакологической коррекции и профилактике преждевременного развития осложнений.Ключевые слова: цистеин, глутатион, антиоксидантная система, сахарный диабет.

COMPARATIVE ANALYSIS OF THE DYNAMICS OF IMMUNE-ANTIOXIDANT STATUS OF THE ANTIOXIDANT SYSTEM IN PATIENTS SUFFERING FROM HYPOTHYROIDISM AND COPD

Aim . To carry out a comparative study of the features and shift tendency of individual indicators of the antioxidant system in patients with hypothyroidism and COPD of various phenotypes. 79 patients suffering from COPD III and IV were examined at the age of 60 to 80 years (men and women) who were hospitalized in the pulmonology department of the Regional Clinical Hospital No. 2 in Krasnodar, along with 47 patients suffering from hypothyroidism. Materials and methods. The state of the blood antioxidant system was assessed by the activity of erythrocyte catalase and superoxide dismutase, the level of reduced thiol groups of erythrocytes, and the amperometric index of the total antioxidant activity of blood plasma. The activity of catalase was studied in the hemolysate of erythrocytes according to the method of Beers R. Results. It was established that there were significant changes in the functioning of individual links of the antioxidant protection system in patients with COPD and hypothyroidism, as well as the stress in the system in general. At the same time, enzyme activity of the antioxidant system of erythrocytes varies in multidirectional ways, whereas the indices of the cellular and extracellular non-enzyme links of the endogenous antioxidant system change in a unidirectional way towards the decrease, which may indicate a significant activation of free radical oxidation processes and partial depletion of the low-molecular antioxidant blood link. Conclusion. The revealed imbalance in the pro-/antioxidant system requires the introduction of drugs with antioxidant activity into therapeutic regimens. In patients with COPD, hypoxia and chronic inflammation may provoke oxidative stress, while in patients with hypothyroidism –decrease in the level of basal metabolism and anabolic processes in general.

Features of proand antioxidant systems in patients with uterine leiomyoma and its combination with adenomyosis and endometrial hyperplasia and the development of recurrence after organ-saving operations

We assessed the incidence of uterine leiomyoma recurrence after myomectomy in cases of its isolated development and combination with other hyperplastic processes of endo- and myometrium. We showed the role of pro- and antioxidant systems’ disturbances in the recurrence of combination of leiomyoma, adenomyosis and endometrial hyperplasia. We propose indole-3-carbinol and epigallocatechin administration as preventing post-operative treatment.

In this research, we discover effect ofpolymorphism rs1801282 of the PPARG gene on the formation oxidative stress in patients with type 2 diabetes, depending on the duration of the disease: till 5 years, 5-10 years and more than 10 years. Activity of lipidperoxidation was assess in terms of diene conjugates (DC) and malonic dealdehid (MDA) and antioxidant system status - the activity of superoxide dismutase (SOD), catalase and the level of a-tocopherol (a-TF). Molecular genetic studies conducted by the method ofpolymerase chain reaction (PCR) in real time. During disease 5-10 years, found increase levels of DC and MDA in patients with polymorphism Pro₁₂--Ala for 34.9 and 34.7%, in compared with ProPro₁₂ --Pro (P=O. 01). Availability ProPro₁₂ --Pro stipulated reduction of catalase activity during disease 5-10 years at 75% (P=O. 001), andfor those, who are illfor more than 10 years at 2,04 times (P=O.01), which is not different from the reference level (F=1. 19; P=O. 600), but in the case ProPro₁₂ --Ala, this figure was in 2 times higher The main conclu- sion is that the type ProPro₁₂ --+Ala ofpolymorphism rs1801282 of the PPARG gene causes the development of oxidative stress in patients with type 2 diabetes with 5-10 years durations, and Pro Pro₁₂ --Pro - deficiency the enzyme catalase level of antioxidant system in patients with durations of disease more than 5 years.
