Anti-hyperglycemic activity of aqueous extracts of cells produced in Ibervillea sonorae(S. Watson) Green cell culture on murine model

bervillea sonorae(S. Watson) Greene, is a plant native to Mexico, where its roots have been used traditionally for treating Diabetes Mellitus. The aim of this work was to establishment of cell cultures of stem explants of I. sonoraeand evaluation of the anti-hyperglycemic activity of cell aqueous extract on a murine model of streptozotocin-induced diabetic rats. Cell extracts had 2.29 mg palmitic acid/g extracted, and other compounds with pharmacological activities like palmitoyl ethanolamide and palmitoyl tryptamine were also identified. Diabetic rats treated with aqueous cell extract decreased glucose levels from 350 mg/dL to 145 mg/dL, AST and ALT from 164 U/L to 49 U/L and 99 U/L to 53 U/L, respectively. Additionally, therewere no changes in the cellular morphology of the pancreas, liver, kidneys, and spleen. These results revealed that the cell aqueous extract from stem explants has anti-hyperglycemic activity.

Effect of Aqueous Extract of chirata in alloxan-induced diabetic rat model

The present study was performed for evaluating the anti-diabetogenic effect of chirata extract in alloxen induced rat model which has rarely been done by any researcher previously. Diabetes was induced by injecting Alloxen (ALX) @ 120 mg/kg body weight subcutaneously to swiss albino mice. The sixteen rats were divided into four groups named Group A (non-diabetic) for the negative control, Group B (diabetic) for positive control, Group C treated with aqueous extract of chirata@ 125 mg/kg body wt and Group D treated with metformin (anti-diabetic drugs) @150 mg/kg body wt.). The duration of the experiment was 16 (sixteen) days. Finally, the research showed that the final body weight of positive control (diabetes induction) Group B (43.0±0.90) was significantly decreased from the treatment of negative control Group A (43.5±1.80) and similarly followed by treatment with a synthetic anti-diabetic agent such as metformin group D (44.50±1.70) and herbal drugs as chirata treatment Group C (44.0±1.95). The creatinine level in Group A, B, C and D were 0.85±0.07 mg/dL, 1.15±0.07 mg/dL, 0.95±0.08 mg/dL and 0.9±0.14 mg/dL respectively. The SGPT level in Group A, B, C, and D was 19±1.41 U/L, 27.44±2.82 U/L, 26.5±2.12 U/L, and 20.5± 2.12 U/L respectively. The SGOT level in Group A,B,C and D were 30±2.82 g/dL, 90±3.53 g/dL, 70±1.41 g/dL and 45±2.83 g/dL respectively. In alloxen-induced mice, a fall in SGPT levels was observed in the groups treated with aqueous extract of Chirata and Metformin. Aqueous extract of Chirata decreased SGOT levels compared with a diabetic control group. The body weight of the swiss albino mice was increased significantly in those treated with chirata extraction in the diabetic group. The synthetic drug metformin-treated group improved the body weight of the swiss albino mice model. The antihyperglycemic activity of aqueous extract of Chirata is comparatively lower than the synthetic anti-diabetic drugs such as metformin. Thus the above observations suggested that extracts, from Swertia chirata possess anti-diabetic principal and can presumably be used for the treatment of diabetes mellitus.

ANTIHYPERGLYCEMIC, ANTIOXIDANT AND ANTI-INFLAMMATORY EFFECTS OF AQUEOUS EXTRACT OF MISTLETOE (Cladocolea loniceroides) IN STZ-INDUCED DIABETIC MICE

Background: Inhibition of carbohydrate hydrolyzing enzymes, such as α-amylase and α-glucosidase, is a key element in the regulation of diabetes mellitus (DM). The purpose of this work was to study the inhibition of carbohydrate hydrolyzing enzymes, and the antihyperglycemic activity of aqueous extract of Cladocolea (C. loniceroides) in streptozotocin (STZ)- induced diabetic mice. Materials and Methods: The inhibitory activities of C. loniceroides aqueous extract on α-amylase and α-glucosidase were investigated in vitro. Glucose tolerance test was performed in normoglycemic (NG) mice which were fed with starch or sucrose. The effect of mistletoe aqueous extract (ME) was measured in (STZ)-induced diabetic mice. On day 35 of the treatment, the effect of decreasing oxidative stress (lipid peroxidation, glutathione redox state, GPx and GR specific activities, cytokines and aminotransferases analysis) was assessed. Results: ME showed a competitive mode of inhibition for the carbohydrate hydrolyzing enzymes (CHE). The maximum antihyperglycemic activity in mice was observed for the unripe fruit aqueous extract (UFAE) for -amylase and stem aqueous extract (SAE); for -glucosidase due to the glycemic response reduction by 23% or 35%, respectively. UFAE decreased malondialdehyde (MDA) 1.76 times; GSH/GSSG ratio was mantained (3.08 ± 0.66); GPx activity was reduced (24%); IL-6 was reduced (18%) and the concentration of TNF- (37%) was leveled with respect to the (STZ)-induced diabetic mice; ALT and AST (liver transaminases) levels were nearly the same compared with those found in the NG mice. Conclusion: UFAE of C. loniceroides exhibited the highest antidiabetic activity in (STZ)-induced diabetic mice.

EVALUATION OF ANTIDIABETIC ACTIVITY OF FRUITS OF WITHAHNIA COAGULANS IN STREPTOZOCIN INDUCED DIABETIC RATS

In past there have been many medicinal plants, which have been used in traditional medicines for their antihyperglycemic properties without any scientific support and pharmacological evidence. The aqueous extract of Withania coagulans dunal fruit has been taken to evaluate the antihyperglycemic activity in normal and streptozotocin-induced diabetic rats. The plant is known as ‘the cheese maker’ or ‘vegetable rennet’ because fruits and leaves of the plant are used as a coagulant. The milk coagulating property of the fruit is attributed to the pulp and husk berries which posses an enzyme which has milk coagulating activity an antihyperglycemic activity of aqueous extract has not been yet done, as no such reports are available in the literature through the activity is reported. The scientific evaluation of its antihyperglycemic activity was, therefore, explored and also compared with the effect of a standard hypoglycemic drug, glibenclamide. A single oral administration of variable dose levels (100, 200 and 300 mg/kg b.wt) of aqueous extract led to lowering of blood glucose level in normal as well as in diabetic rats. In acute treatment maximum % reduction in glucose level (57.02%) was seen with 300mg/kg b.wt WCD at 6 hr while with standard drug maximum % reduction (62.25%) at 6 hr was found. In subacute study the maximum % reduction in blood glucose level was found (57.22%) with WCD aq. extract in dose 300 mg/kg b.wt which is slightly lesser then the % reduction seen with standard drug (64.52%) respectively. Both the doses 100 & 200 mg/kg b.wt of WCD showed significant effect (p<.05) at 4 hr and highly significant (p<0.01) at 6 hr while in chronic treatment both the doses showed significant reduction in blood glucose from 14th day to 28th days. The findings from this study suggest that the aqueous extract of these fruits may be prescribed as adjunct to dietary therapy and drug treatment for controlling diabetes mellitus. Effect of aqueous bark extract of Garuga pinnata Roxb. in streptozotocin-nicotinamide induced type-II diabetes mellitus

A study was undertaken to evaluate the antihyperglycemic activity of aqueous extract of bark of Garuga pinnata Roxb. (Burseraceae). The various parameters studied included fasting blood sugar levels, serum lipid levels, liver glycogen content, serum insulin level and glycated hemoglobin in diabetic and normal rats. Streptozotocin-nicotinamide was used to induce type-II diabetes mellitus. Treatment with the extract at two dose levels showed a significant increase in the liver glycogen and serum insulin level and a significant decrease in fasting blood glucose and glycated hemoglobin levels. The total cholesterol and serum triglycerides levels were also significantly reduced and the HDL cholesterol levels were significantly increased upon treatment with the extract thus proving the potent antidiabetic property of the plant.

Antihyperglycemic activity of aqueous extract of leaves of Cocculus hirsutus (L.) Diels in alloxan-induced diabetic mice.

<b>Objective: </b> To evaluate the antihyperglycemic activity of aqueous extract of leaves of <i> Cocculus hirsutus</i> (L.) Diels in alloxan-induced diabetic mice.<b> Materials and Methods:</b> Alloxan-induced (70 mg/kg, i.v.) diabetic mice were given aqueous leaf extract (250, 500, and 1000 mg/kg, p.o., <i> n=</i> 6) of <i> C. hirsutus</i> or vehicle (distilled water, 10 ml/kg, p.o.) or standard drug glyburide (10 mg/kg, p.o.) for 28 days. Blood samples were withdrawn by retro-orbital puncture and were analyzed for serum glucose on 0th, 7th, 14th, 21st, and 28th days by glucose oxidase/peroxidase method. In oral glucose tolerance test, glucose (2.5 g/kg, p.o.) was administered to nondiabetic control, glyburide (10 mg/kg, p.o.), and aqueous extract of <i> C. hirsutus</i> (1000 mg/kg, p.o.) treated mice. The serum glucose level was analyzed at 0, 30, 60, and 120 min after drug administration. <b> Results:</b> The aqueous leaf extract of <i> C. hirsutus</i> (250, 500, and 1000 mg/kg, p.o.) showed significant (<i> P<</i> 0.01) reduction of serum glucose level in alloxan-induced diabetic mice at 28th day. In oral glucose tolerance test, aqueous extract of <i> C. hirsutus</i> increased the glucose tolerance. <b> Conclusion:</b> It is concluded that <i> C. hirsutus</i> has significant antihyperglycemic activity as it lowers serum glucose level in diabetic mice and significantly increases glucose tolerance.
