Phytochemical profile and antifungal activity of essential oils obtained from different Mentha longifolia L. accessions growing wild in Iran and Iraq

BackgroundMentha longifolia L. is a perennial plant belonging to the Lamiaceae family that has a wide distribution in the world. M. longifolia has many applications in the food and pharmaceutical industries due to its terpenoid and phenolic compounds. The phytochemical profile and biological activity of plants are affected by their genetics and habitat conditions. In the present study, the content, constituents and antifungal activity of the essential oil extracted from 20 accessions of M. longifolia collected from different regions of Iran and Iraq countries were evaluated.ResultsThe essential oil content of the accessions varied between 1.54 ± 0.09% (in the Divandarreh accession) to 5.49 ± 0.12% (in the Khabat accession). Twenty-seven compounds were identified in the essential oils of the studied accessions, which accounted for 85.5-99.61% of the essential oil. The type and amount of dominant compounds in the essential oil were different depending on the accession. Cluster analysis of accessions based on essential oil compounds grouped them into three clusters. The first cluster included Baziyan, Boukan, Sarouchavah, Taghtagh, Darbandikhan, Isiveh and Harir. The second cluster included Khabat, Kounamasi, Soni and Mahabad, and other accessions were included in the third cluster. Significant correlations were observed between the essential oil content and components with the climatic and soil conditions of the habitats. The M. longifolia essential oil indicated antifungal activity against Fusarium solani in both methods used. In all studied accessions, the fumigation method compared to the contact method was more able to control mycelia growth. In both methods, the inhibition percentage of essential oil on mycelia growth increased with an increase in essential oil concentration. Significant correlations were found between the essential oil components and the inhibition percentage of mycelium growth.ConclusionThe studied M. longifolia accessions showed significant differences in terms of the essential oil content and components. Differences in phytochemical profile of accessions can be due to their genetic or habitat conditions. The distance of the accessions in the cluster was not in accordance with their geographical distance, which indicates the more important role of genetic factors compared to habitat conditions in separating accessions. The antifungal activity of essential oils was strongly influenced by the essential oil quality and concentration, as well as the application method. Determining and introducing the elite accession in this study can be different depending on the breeder’s aims, such as essential oil content, desired chemical composition, or antifungal activity.

Extraction Kinetics and Antifungal Activities of Essential Oil from the Aerial Parts of Gros Baume (Hyptis suaveolens) Against Three Fungal Strains in Côte d'Ivoire

Introduction: Microscopic fungi colonize many food substrates resulting in considerable economic losses. Their development leads to an alteration in the organoleptic quality of the food, and produces mycotoxins. The aim of the present work was to evaluate the extraction kinetics and in vitro antifungal activity of the essential oil of Hyptis suaveolens on the growth of three toxigenic fungal strains. Methodology: The essential oil was extracted by steam distillation using a Clevenger-type device. It was tested for in vitro antifungal activity against three mold strains (Aspergillus flavus, Aspergillus niger and Fusarium sp) using the agar dilution method. Results: The results of the extraction kinetics evaluation show that in fifty minutes of distillation, almost all the essential oil contained in the aerial parts of H. suaveolens is recovered. In vitro antifungal tests showed a slight slowdown in mycelial growth for the three strains studied. However, the antifungal parameters determined show that these strains are sensitive to 50 μL/mL with a CMF of 100 μL/mL. This is just a simple fungistatic effect of this extract on the mycelial growth of the strains tested. Aspergillus strains seem to be slightly more affected than Fusarium strains. Conclusion: The antifungal potential of H. suaveolens essential oil does not offer a novel approach to the control of these three fungal strains.

Chemical profiling and in-vitro anti-oxidant, anti-diabetic, anti-inflammatory, anti-bacterial and anti-fungal activities of essential oil from Rosmarinus officinalis L.

Rosmarinus officinalis L. commonly known as Rosemary is a well-known genus belongs to Lamiaceae family, which is one of the major plant families with a high concentration of essential oils. Rosemary is well known to produce herbal essential oils owing to the presence of therapeutic and medicinal elements in the plant’s by-products. In the present study, rosemary essential oil (REO) was extracted from Rosmarinus officinalis and it was evaluated for its phytochemicals, antioxidant, antibacterial, antifungal, anti-diabetic and anti-inflammatory activities. GC-FID analysis and fingerprint analysis was performed. Different antioxidant assays were performed like DPPH (1,1-diphenyl-2-picrylhydrazyl), ABTS (2,2-azinobis-3-ethylbenzothiazoline-6-sulphonic acid), nitric oxide radical, hydroxyl radical, iron reducing potential, iron chelating activity. Anti-bacterial activity was observed by the disk diffusion method against Gram-positive (G+), Gram-negative (G−) bacteria. Anti-diabetic and anti-inflammatory activities were also performed. Antifungal activity against Aspergillosis and Mucormycosis causing fungal strains was also evaluated. GC-FID revealed the presence of vertocitral, α-pinene andhumulene oxidemajor component of REO along with other bioactive components. The IC50 value (µl/ml) for various assays was observed in the following order: OH- (16.6118), iron chelating (32.0115), iron reducing (92.534), DPPH (101.929), and ABTS (172.2765). The best antimicrobial activity was against Bacillus subtilis (MTCC 121), with an inhibition zone of 2.5 cm. REO showed strong anti-inflammatory and anti-diabetic potential and can be employed as an anti-diabetic agent due to its inhibitory effect on α-amylase activity. Based on these findings it was concluded that REO can be widely employed across a variety of sectors, including food, aromatherapy, herbal and allopathic medicine, cosmetics, and as a natural biocide, fungicide, and preservative.

Assessing the efficacy of essential oil fumigation in mitigating Botrytis cinerea infection in cherry tomato

Cherry Tomato (Solanum lycopersicum L. var. cerasiforme) believed to be the progenitor of modern cultivated tomatoes, is one of the world’s most cultivated and widely consumed vegetables. However, it is very susceptible to fungal (Botrytis cinerea) infection, which decreases shelf life, and increases post-harvest loss. Postharvest loss and lower quality of fruit decrease the economic value of the crop or may make it unsuitable for human consumption and affect farmer profit. Therefore, an effective method is needed to reduce tomato’s post-harvest loss. This study aims to assess the efficacy of Zingiber officinale oil and Piper nigrum oil in controlling fungal disease in tomato fruit. The antifungal activity of essential oil was evaluated using the essential oil fumigation method. The result showed that essential oil significantly inhibits Botrytis cinerea growth as compared to the control. Therefore, essential oil can effectively control the fungal diseases of cherry tomatoes against Botrytis cinerea fruit rot and reduce the post-harvest loss of tomatoes.

Antifungal Activity of Essential Oils on Helminthosporium solani Causing Potato Silver Scurf under In Vitro and In Vivo Conditions

Potato silver scurf, caused by the fungus Helminthosporium solani, is an important storage disease of potato (Solanum tuberosum L.), reducing the market value of tubers. Using in vitro and in vivo assays, the presented experiments aimed to determine the effect of selected essential oils (EOs: α-pinene, carvacrol, cinnamaldehyde, D-carvone, eucalyptol, L-linalool, L-menthol, L-menthone, (R)-(+)-limonene, and thymol) on H. solani growth. All EOs inhibited pathogen growth, but their antifungal activities differed significantly. Thymol, carvacrol, and cinnamaldehyde had the strongest inhibitory effects on mycelial growth under in vitro conditions. (R)-(+)-limonene displayed the weakest inhibition. The effectiveness of those EOs with the greatest antifungal activity was confirmed by in vivo experiments. EOs were applied through dressing and fumigation, with EOs bound to a biopolymer for dressing. Dressing and fumigation brought a highly statistically significant reduction in H. solani infection intensity and sporulation intensity on tubers. Although EOs had an insignificant effect on potato cooking quality, the taste of EO-dressed tubers was degraded by an off-odor and off-taste. EOs could provide an ecological alternative for reducing H. solani tuber infection during storage.

A combinatorial approach of Monarda citriodora essential oil (MEO) and linalool vapors to control fruit rot of Citrus limon caused by a new pathogen, Aspergillus foetidus, and its underlying mode of action.

Citrus limon (lemon) is a widely cultivated citrus fruit. Significant postharvest losses due to fungi plague its production. Environmental and human health hazards have made the application of synthetic fungicides unsuitable. Despite the previous reports of antifungal activities of essential oil (EO) vapors, their synergistic combinations are understudied. Synergistic vapor combinations are advantageous due to less concentration of active components. This study aimed to isolate and identify postharvest fungal pathogens lemon and to evaluate the antifungal effects of synergistic Monarda citriodora EO (MEO)-constituent vapor combinations in vivo and in vitro. Postharvest fungal pathogens of lemon (C. limon) were isolated from various infected samples. The most pathogenic isolate was identified through morphology and its ITS-based rRNA gene sequencing as Aspergillus foetidus (O4). This is the first report of A. foetidus as a postharvest pathogen of lemon. The minimum fungicidal concentrations (MFCs) of MEO vapors treatment against O4 were 1346.15µL/L air. For carvacrol, hexanal, and linalool, MFC was same (96.16µL/L air). Checkerboard assays demonstrated that 1/4 MFC of MEO (336.54µL/L air) and 1/4 MFC of linalool (24.04µL/L air) (M+L) were synergistic against O4. M+L vapors reduced the O4 growth on lemons during storage by 64%±1.50% and preserved their quality (low weight loss %, unchanged pH, increased ascorbic acid content). Propidium iodide staining, ergosterol content analysis, calcofluor white staining and chitin content analysis revealed the integrity loss of the O4 plasma membrane and cell wall. 2',7'-Dichlorofluorescin diacetate staining revealed accumulation of intracellular reactive oxygen species (ROS), and scanning electron microscopy (SEM) analysis exposed the M+L treated mycelia with malformations. M+L vapors offer protection for lemons from A. foetidus and preserve their quality during storage.

Physicochemical characterization, antioxidant and antifungal activities of essential oils of Urginea maritima and Allium sativum

Abstract This study aims to contribute to the valorizing of aromatic and medicinal plants of Morocco by studying the antioxidant and antifungal properties of the essential oils (EOs) of Allium sativum (AS) (garlic) and Urginea maritima (Syn. Drimia maritima) tested in vitro. After the hydrodistillation of EOs, we determined their physicochemical indices according to an international standard (Moroccan ISO standard), and the chemical composition of the oils was studied by gas chromatography coupled with mass spectrometry (GC-MS). Then, the antioxidant properties were determined using the free radical scavenging capacity of 2,2-diphenyl-1-picrylhydrazyl and the ferric-reducing antioxidant power of these oils. The evaluation of the antifungal activity was carried out by the direct contact method against two fungal strains, Fusarium oxysporum and Fusarium equiseti. GC-MS analysis revealed a significant percentage of terpenes in U. maritima EO, with terpinolene (21.82%) and estragole (6.24%) as the main compounds. Biological tests have demonstrated the remarkable antioxidant activities of this EO compared with AS EO, with an IC50 of 1.03 mg/mL and an RP0.5 of 1.60 mg/mL. On the other hand, while A. sativum (garlic) EO showed exceptional antifungal activities, with 100% inhibition and MICs of 0.5 µL/mL for F. equiseti and 2.5 µL/mL for F. oxysporum. Also, regardless of its antioxidant properties being moderate, this EO stands out for its efficacy against fungal infections of the Fusarium genus. In summary, the results suggest that U. maritima EO could be used as a natural source of antioxidants, while garlic EO could be considered a potentially effective treatment against fungal infections caused by Fusarium fungi.

Analysis of Antifungal Activity of Essential Oil from Orange Peels

The largest fruit in Nepal is citrus, which takes up 2684 of the country's total fruit-growing land. 71.65% of the nation's citrus-growing land is devoted to the planting of citrus called mandarin oranges. Pectin is a structural hetero-polysaccharide that is integral to the motion of terrestrial flora. The fact some components are becoming resistant to specific fungal infections and illnesses has become among the biggest obstacles in treating them at the moment. This necessitated the employment of novel medications to treat infectious disorders brought on by these microbes. Additionally, they are exceedingly low in toxicity and abundant sources of active chemicals. Soxhlet can be used to easily extract the essential oil. The end result confirms that essential oils were successfully extracted as pectin solutes, which is particularly helpful for pectin extraction in industrial sectors both economically and for environmental health.

In order to valorize Algerian aromatic and medicinal plants, this study focused on both the evaluation of the antifungal activity of Pelargonium graveolens essential oil against the citrus fungus Fusarium proliferatum and the in-silico study of its fractions. Moreover, the results showed a strong antifungal potential of all tested essential oil (EO) concentrations compared to the commercial fungicide Agriconazole. Five major molecules that, to our knowledge, have not been the subject of any theoretical study are selected to perform the in-silico study. The stability and molecular reactivity of these five molecules were computed using the HOMO-LUMO energies, energy gap, chemical potential (μ), electronegativity (χ), hardness (η), and softness (S) values. In silico analysis through molecular docking and pharmacokinetic evaluation was used to assess its antifungal activity and drug-likeness property. The in silico DFT, ADMET, molecular docking, and molecular dynamics studies confirm that the results have a greater affinity with the in vitro tests carried out for the selection of new antifungal products of natural origin.

IN VITRO ANTIFUNGAL ACTIVITY OF ESSENTIAL OILS (FAMILY LAMIACEAE) AGAINST CLADOSPORIUM SP. STRAINS – POSTHARVEST PATHOGENS OF FRUITS

Essential oils (EOs) are a suitable alternative for extending the shelf life of foods. The aim of this research was to test the effect of fifteen EOs extracted from plants of the Lamiaceae family on five strains of Cladosporium cladosporioides. Cladosporium cladosporioides is frequently found on fruit and may be involved in fruit spoilage. The strains used in the study were isolated directly from lesions on berries. The growth of the fungi on the fruit was the reason for its rejection from sale. The antifungal activity of EOs against Cladosporium cladosporioides strains was determined by the microatmospheric method (625 μl EO/L air) during 14 days of cultivation. Thirteen EOs: thyme and red thyme (from Thymus vulgaris L.), mitcham mint (Mentha x piperita L. var. Mitcham), peppermint (Mentha x piperita L.), savory (Satureja hortensis L.), sage (Salvia officinalis L.), spearmint (Mentha spicata L. var. crispa), lavender (Lavandula angustifolia Mill.), marjoram (Origanum majorana L.), bergamot-mint (Mentha citrata Erh.), wild thyme (Thymus serpyllum L.), hyssop (Hyssopus officinalis L.), and oregano (Origanum vulgare L.) were shown to inhibit growth of all strains to 100%. Basil (Ocinum basilicum L.) and rosemary (Rosmarinus officinalis L.) EOs did not have a 100% inhibitory effect on all strains throughout the cultivation period. Subsequently, minimum inhibitory concentrations (MIDs) were determined using the microatmospheric method. LD90 and LD50 values were estimated by probit analysis. Based on the MID, LD90, and LD50 results, the essential oils can be divided into three groups. The first group (most effective): thyme, red thyme, spearmint, wild thyme, and oregano EO; the second group: peppermint, lavender, mitcham mint and savory EO; the third group: sage, marjoram, bergamot-mint and hyssop EO. The most effective EOs can be used to suppress the growth of Cladosporium cladosporioides in the vapour phase.

Application of Essential Oils Extracted from Peel Wastes of Four Orange Varieties to Control Anthracnose Caused by Colletotrichum scovillei and Colletotrichum gloeosporioides on Mangoes.

A huge amount of orange peel waste is annually discharged into the environment. Processing of this waste for the control of post-harvest fruit diseases can reduce environmental pollution. Essential oils (EOs) from fruit peels of Citrus reticulata × sinensis (Sanh cultivar) and Citrus sinensis (Xoan, Mat and Navel cultivar) were investigated for their ability to control anthracnose caused by Colletotrichum gloeosporioides and Colletotrichum scovillei on mangoes. EOs were extracted by hydro-distillation and analyzed by GC-MS and GC-FID. The antifungal activity of the EOs was determined by in vitro and in vivo assays. The Mat cultivar had the highest extraction yield of 3% FW, followed by Xoan (2.9%), Sanh (2.2%), and Navel (1%). The chemical composition of the EOs was similar, with limonene as the main compound (around 96%). The antifungal activity of EOs was not different, with a minimum fungicidal concentration of 16% for both fungi. The disease inhibition of EOs increased with their concentration. The highest inhibition of anthracnose caused by both fungi on mangoes was achieved at 16% EO. EOs had no adverse effect on mango quality (pH, total soluble solids, total acidity, color and brightness of mangoes), except firmness and weight loss at high concentrations (16%). Orange EOs can be used as bio-fungicides to control mango anthracnose at high concentrations.

Antifungal activity of essential oil and emulsifiable concentrates of citronella and lemon grass against Phomopsis vexans

The present study aims to develop eco-friendly Emulsifiable Concentrates (EC) using citronella and lemon grass essential oil (EO) and evaluate its antifungal activity against Phomopsis vexans in Solanum melongena. Among nine prepared formulations, F1 formulation with EO (40%) + vegetable oil (50%) and emulsifier blends (10%) with hydrophilic-lipophilic balance value (12.86) were found suitable for both EO formulations. These formulations (CF1 and LF1) performed well in terms of emulsion stability, cold test, accelerated storage and foam test indicating feasibility for their commercial production. In laboratory studies, at 0.1% concentration, citronella essential oil, lemon grass essential oil and their EC formulations CF1 and LF1 inhibited the growth of P. vexans with EC50 values 0.0087, 0.0098, 0.009 and 0.0126, respectively. Further, under field conditions, CF1 performed better than LF1 formulation in terms of minimum disease severity (32.22%). Hence, EC formulations can be used as natural and eco-friendly fungicide for P. vexans control.

Rapid molecular assay for the evaluation of clove essential oil antifungal activity against wheat common bunt.

Common bunt of durum wheat (DW), Triticum turgidum L. ssp. durum (Desf.) Husn., is caused by the two closely related fungal species belonging to Tilletia genus (Tilletiales, Exobasidiomycetes, Ustilaginomycotina): Tilletia laevis Kühn (syn. T. foetida (Wallr.) Liro.) and T. caries (DC) Tul. (syn. T. tritici (Bjerk.) G. Winter). This is one of the most devastating diseases in wheat growing areas worldwide, causing considerable yield loss and reduction of wheat grains and flour quality. For these reasons, a fast, specific, sensitive, and cost-effective method for an early diagnosis of common bunt in wheat seedlings is urgent. Several molecular and serological methods were developed for diagnosis of common bunt in wheat seedlings but at late phenological stages (inflorescence) or based on conventional PCR amplification, with low sensitivity. In this study, a TaqMan Real Time PCR-based assay was developed for rapid diagnosis and quantification of T. laevis in young wheat seedlings, before tillering stage. This method, along with phenotypic analysis, was used to study conditions favoring pathogen infection and to evaluate the effectiveness of clove oil-based seed dressing in controlling the disease. The overall results showed that: i) the Real Time PCR assay was able to quantify T. laevis in young wheat seedlings after seed dressing by clove oil in different formulations, greatly reducing times of analysis. It showed high sensitivity, detecting up to 10 fg of pathogen DNA, specificity and robustness, allowing to directly analyze crude plant extracts and representing a useful tool to speed up the tests of genetic breeding for disease resistance; ii) temperature was a critical point for disease development when using wheat seeds contaminated by T. laevis spores; iii) at least one of the clove oil-based formulations tested was able to efficiently control wheat common bunt, suggesting that clove oil dressing could represent a promising tool for managing the disease, especially in sustainable farming.
