Nutmeg is known as a spice plant that has economic value and multipurpose because every part of the plant can be used in various industries. Compounds contained in nutmeg leaves include alkaloids, triterpenoids, tannins and 2 flavonoids. The purpose of this study was to determine the antidiabetic effect of ethanolic extract of nutmeg leaf (myristica frangrans) against male wistar white rats (Rattusnorvegicus) induced by alloxan administered intraperitoneally and compare its effectiveness with glibenclamide. This study used five treatment groups, namely the administration of aquadest as a negative control, glibenclamide as a positive control and ethanol extract of nutmeg leaves at a dose of 100 mg/kg BW, 200 mg/kg BW and 300 mg/kg BW as control treatments. Blood glucose measurements were carried out on day 0 (three days after induction), day 3, day 7 and day 10. The results showed that the ethanol extract of nutmeg leaf (myristica frangrans) TEST ACTIVITY can reduce blood glucose levels. At a dose of 300 mg/kg but not exceeding the decrease in blood sugar in the administration of glibenclamide. Giving ethanol extract of nutmeg leaves (myristica frangrans) also has activity to repair damage to pancreatic -cells in white rats (Rattus norvegicus) induced by alloxan.

Anti-Diabetic Effects of Ethanol Extract from Sanghuangporous vaninii in High-Fat/Sucrose Diet and Streptozotocin-Induced Diabetic Mice by Modulating Gut Microbiota.

Type 2 diabetes mellitus (T2DM) may lead to abnormally elevated blood glucose, lipid metabolism disorder, and low-grade inflammation. Besides, the development of T2DM is always accompanied by gut microbiota dysbiosis and metabolic dysfunction. In this study, the T2DM mice model was established by feeding a high-fat/sucrose diet combined with injecting a low dose of streptozotocin. Additionally, the effects of oral administration of ethanol extract from Sanghuangporous vaninii (SVE) on T2DM and its complications (including hypoglycemia, hyperlipidemia, inflammation, and gut microbiota dysbiosis) were investigated. The results showed SVE could improve body weight, glycolipid metabolism, and inflammation-related parameters. Besides, SVE intervention effectively ameliorated the diabetes-induced pancreas and jejunum injury. Furthermore, SVE intervention significantly increased the relative abundances of Akkermansia, Dubosiella, Bacteroides, and Parabacteroides, and decreased the levels of Lactobacillus, Flavonifractor, Odoribacter, and Desulfovibrio compared to the model group (LDA > 3.0, p < 0.05). Metabolic function prediction of the intestinal microbiota by PICRUSt revealed that glycerolipid metabolism, insulin signaling pathway, PI3K-Akt signaling pathway, and fatty acid degradation were enriched in the diabetic mice treated with SVE. Moreover, the integrative analysis indicated that the key intestinal microbial phylotypes in response to SVE intervention were strongly correlated with glucose and lipid metabolism-associated biochemical parameters. These findings demonstrated that SVE has the potential to alleviate T2DM and its complications by modulating the gut microbiota imbalance.

In-vivo Antidiabetic Activity of Saccharum Spontaneum on STZ- Induced Diabetic Mice

Objectives: Saccharum spontaneum is traditionally used in Arunachal Pradesh, India for the treatment of diabetes mellitus. Review of existing literature, found no scientific investigation on this claim. Therefore, this study was aim to evaluate the antidiabetic effect of ethanolic extract of S. spontaneum on STZ induced diabetic mice.&#x0D; Materials and Methods: Diabetes was induced in male albino mice by single intraperitoneal injection of streptozotocin (100mgkg-1 BW). Initially Starch tolerance test, blood glucose level and area under the curve (AUC) were evaluated after S. spontaneum treatment in diabetic mice. The test extract was administered for three weeks and the antidiabetic activity was evaluated by means of monitoring the change in blood glucose level, body weight and biochemical parameters.&#x0D; Results: The extract showed a significant decrease in the blood glucose level, slight enhancement of body weight after the treatment. Daily oral treatment of extract also showed a significant reduction in the Serum ALT, creatinine and triglyceride when compared to the diabetic control.&#x0D; Conclusion: The results provide supports to the traditional claim made on Saccharum spontaneumand provide evidence that this plant might be a potential source of antidiabetic agent.

Anti-diabetic effect of Stachytarpheta jamaicensis on low dose streptozotocin-induced diabetic rats fed on a high-fat diet

IntroductionThis study evaluates the anti-diabetic effect of ethanol extract of Stachytarpheta jamaicensis leaf on streptozotocin (STZ) - induced diabetic rats fed on high-fat diet (HFD).MethodsSets of male albino rats of the Wistar strain weighing between 180 and 250 g were exposed to high fat diet (margarine and oil from vegetable sources in a ratio of 2:1 w/v) for 3 weeks. Then the animals were fasted overnight; hyperglycemic state was induced using reduced dose of streptozotocin (35 mg/kg) and animals were randomly divided into five groups (n = 7); group A received the HFD + STZ (35 mg/kg i.p.); group B received HFD + STZ + gliberclamide (10 mg/kg; i.p); groups C, D and E were administered the HFD + streptozotocin with different doses of the ethanol extract (30, 35 and 100 mg/kg p.o., respectively).ResultsResults showed significant (p < 0.05) decrease in blood glucose concentration of the rats treated with different doses of S. jamaicensis extract and those treated with gliberclamide compared to the untreated diabetic rats (negative control). Significant (p < 0.05) reductions in activities of serum AST, ALP, total protein and bilirubin were noticed in the groups in contrast to the control. Levels of urea, creatinine, potassium and chloride were considerably (p < 0.05) low while sodium and bicarbonate levels were high in the groups except the control. Lipid profile revealed significant (p < 0.05) reduction in total cholesterol, triacylglycerol, LDL, VLDL while HDL levels were high in the groups compared to the control. The extract significantly (p < 0.05) ameliorated weight loss. Histopathology of the liver, kidney and pancreas showed ameliorative effect of the extract against the deleterious changes occasioned by the HFD and STZ induced diabetic state.ConclusionThese findings have provided scientific basis for the use of S. jamaicensis in the treatment of diabetes mellitus in ethnomedicinal practices in Nigeria.

Trace Mineral, Qualitative Phytochemical Composition and Antidiabetic Effect of Ethanol Extract of Citrullus lanatus Seeds on Diabetic Rats

The trace mineral concentration, phytochemical composition and antidiabetic effect of the ethanol extract of C. lanatus seeds were carried out using standard methods. The preclinical experimental model was 120 mgkg-1 b.w. (via intraperitoneal) Alloxan induced diabetic rat model, with ethanol extract of C. lanatus seeds administered orally at 500mgkg-1 b.w. Qualitative phytochemical screening showed presence of alkaloids, flavonoids, saponins, cardiac glycosides, steroids/triterpenoids, tannins, carbohydrates and oils. Atomic Absorption Spectrophotometric analysis revealed that iron had the highest value (8.31mgkg-1), followed by zinc (5.78 mgkg-1), then manganese (4.28 mgkg-1). Selenium concentration was appreciable (0.85 mgkg-1). Ethanol extract of C. lanatus seeds significantly (p &lt; 0.05), dose and time dependently reduced blood glucose level, increased body weights of rats and had positive effect on organ weights and organ volume. The results in this study showed that Citrullus lanatus seeds are rich in zinc, selenium, manganese, iron, alkaloids, flavonoids, saponins, cardiac glycosides, steroids/triterpenoids, and tannins. They may therefore serve as good sources of these trace mineral nutrients and bio actives for nutritional and medicinal purposes relating to diabetes management.

Phytochemical Studies and Blood Glucose Lowering Effect of Zambian Cultivars of Piliostigma thonningii in Alloxan - Induced Diabetic Rats

Background: There is anecdotal evidence among some Zambian traditional medicine practitioners that Piliostigma thonningii possesses antidiabetic effects. This study assessed the in-vivo antidiabetic effects of ethanol extract of the plant in alloxaninduced diabetic wistar rats.Study design: A laboratory-based experimental study involving 24 rats divided into 4 groups of 6 and administered the extract, glibenclamide and placebo.Methods: Diabetes was induced by a single intraperitoneal injection of 150mg/kg alloxan monohydrate. The in-vivo glucose lowering effect of ethanol extract of stem bark of Piliostigma thonningii was evaluated in comparison to normal saline as negative control and glibenclamide as positive control. Standard methods for identification of alkaloids, flavonoids, saponins, tannins, glycosides, terpenoids and anthraquinones were employed in the phytochemical analysis of the extract.Results: Piliostigma thonningii ethanol stem bark extract showed presence of alkaloids, flavonoids, saponins, tannins, glycosides and terpenoids. The extract caused a 55.3% (P=0.002) reduction in blood glucose of the experimental animals over the treatment period from the initial 19.7±7.0 mmol/l to 8.8±2.5 mmol/l, compared with glibenclamide which caused 68.3 % (P=0.001) lowering blood glucose levels from 24.0±4.5 mmol/l to 7.6±5.9 mmol/l.Conclusions: Stem bark extract of Piliostigma thonningii showed a significant glucose lowering effect in diabetic rats. The study also demonstrated presence of secondary plant metabolites which could be responsible for some of the medicinal properties of the plant. Further studies are required to purify the plant and identify active fractions responsible for the anti-diabetic effect observed.

Fruits of Rosa laevigata and its bio-active principal sitostenone facilitate glucose uptake and insulin sensitivity in hepatic cells via AMPK/PPAR-γ activation

BackgroundFruits of Rosa laevigata Michx is commonly used in traditional Chinese medicine for treating of spleen deficiency, chronic diarrhea, and chronic urinary tract infection. Recently, fruits of R. laevigate have been shown to have renal protective effects in diabetic rats. However, up to now, no studies have been reported on the anti-hyperglycemic or anti-diabetic effect of R. laevigata or its derived compounds. PurposeTherefore, this study investigated the anti-diabetic effect of ethanol extract of R. laevigata (EtRL) fruits, its derivate sub-fractions, and pure compounds in terms of glucose-lowering effect on hyperglycemia-induced hepatic cells in vitro. MethodsWe investigated the anti-hyperglycemic effect of EtRL and its derivate sub-fractions (water, n-butanol, ethyl acetate, and n-hexane), and its major bioactive compound, sitostenone, in normal and high glucose-induced insulin-resistant hepatic HepG2 cells using in vitro DNS glucose uptake and Western blotting assays. ResultsTreatment with EtRL and its derivate sub-fractions significantly increased glucose uptake by hepatic cells. Besides, co-treatment with insulin further improved glucose uptake in insulin-resistant cells. Notably, compared with all soluble fractions, the n-hexane sub-fraction displayed the strongest activity in the context of glucose consumption. Furthermore, sitostenone, a primary bioactive compound was isolated from n-hexane sub-fraction by bioactivity-guided fractionation procedure. Sitostenone treatment significantly increased glucose uptake, whereas there was no further increase when co-treatment with insulin. Indeed, sitostenone significantly increased glucose uptake and promote insulin sensitivity in insulin-resistant cells. Further analysis revealed that sitostenone activates proteins involved in the insulin signal transduction pathway, including insulin receptor substrate-1 (IRS-1), AKT, and glycogen synthase kinase 3 β (GSK3β). It was also found that sitostenone provoked glucose uptake in insulin-resistant cells via peroxisome proliferator-activated receptor-γ (PPAR-γ) and AMP-activated protein kinase (AMPK) activation, which facilitates up-regulation of glucose transporters, GLUT2 and GLUT4 in the cell membrane and down-regulation of Forkhead box protein O1 (FOXO1) in the nucleus. ConclusionThis study demonstrating that sitostenone, a steroid-like compound from fruits of R. laevigata has a promising ability to promote glucose uptake and repairs insulin resistance in hepatic cells.

Effects of Carica Papaya and Psidium Guajava Extracts on Hepato-Renal Performance of Streptozotocin-Induced Diabetic Male Albino Rats

The growing number of diabetes coupled with the harsh side effects of some synthetic drugs has led to the increasing search for alternative medicine which are relatively cheap with minimum side effects. Objective: To investigate the anti-diabetic effect of ethanolic extract of Carica papaya and Psidium guajava extracts separately and in combination, and the effect of treatment on hepato-renal performance. Methods: Papaya and guava leaves were macerated in 98% ethanol for extraction. Results: The result showed that ethanolic leaf extract of P. guajava and C. papaya significantly (P&lt; 0.05) reduced blood glucose level, Total Cholesterol (TC), triglycerides (TG), low density lipoprotein (LDL) and significant increase in high density lipoprotein (HDL) as compared to diabetic control group. Serum activities of liver enzymes such as aspartate aminotransferase (AST), alkaline phosphatase (ALP) and alanine aminotransferase (ALT) was slightly elevated that indicate hepatotoxic nature of plant extracts. The level of gamma-glutamyl transferase (GGT), bilirubin and HbA1C level were restored to normal range. A slight reduction in body weight (P&gt;0.05) of treated groups was observed. Serum urea and creatinine concentration was restored to normal level in treated rats with regeneration of kidney function. Conclusions: The study concluded that individual and combined plant extract show anti-hyperglycemic effect but the combined remedy at this dose show hepatotoxicity.

Comparative Study on Antidiabetic Effect of Ethanolic Extract of Jute Leaf on Neonatal Streptozotocin- Induced Type-2 Diabetic Model Rat

Aim: Functional food and their bioactive compounds have been considered as a new approach for the prevention and management of type 2 diabetes and its complications. According to this approach current study was carried out as an elucidation of antidiabetic properties of Corchorus capsularis and Corchorus olitorius varieties of jute leaf (ethanolic extract) on nSTZ-induced type-2 diabetic rats.&#x0D; Methodology: The type-2 diabetic model rat was developed by a single intraperitoneal injection of freshly prepared STZ (90 mg/kg/10 ml) in sterile citrate buffer (0.1 M, pH 4.5) to rat pups (48 hour old). After three months, OGTT was performed to select diabetic (FSG &gt; 6.5mmol/L and after 90 min of glucose load &gt; 14 mmol/L) experimental rats. The rats were randomly divided into four groups [DWC, GT, Ext-1 and Ext-2 represent, diabetic water control, glybenclamide treated (20 mg/5 ml/kg body weight), C. capsularis treated and C. olitorius treated group (1.25 g/10 ml/kg body weight) respectively]. One group was kept with normal rats [normal water control, NWC]. The treatment was given once daily or 28 consecutive days. Fasting serum glucose, liver glycogen and lipid profile were estimated by using standard methods.&#x0D; Results: The results showed that Ext-1 and Ext-2 treated groups gradually decreased serum glucose level (7.15 ±0.67 to 5.94 ± 1.19 and 7.20 ± 0.93 to 5.28 ±1.03 respectively) and reducing effect by Ext-2 was significant (p=0.001). Both extract showed lower liver glycogen level compared with GT group [5.0±2.5 Vs 17.7±6.5 (Ext-1 vs GT) and 7.5±6.4 Vs 17.7±6.5 (Ext-2 vs GT)] and even Ext-1 manifested significant effect (p=0.05). Additionally, lipid profile estimation revealed no significant improvement by the consumption of both the extracts.&#x0D; Conclusion: On the basis of current investigations, it may be concluded that both variety of jute’s leaf demonstrated hypoglycemic properties in Type 2 diabetic model rats; further in-depth studies are recommended to explore the exact mechanism(s) of hypoglycemic effect.

Anti-diabetic effect of ethanol extract of Cyclolepis genistoides D. Don (Palo azul), made in Paraguay

Extract of Cyclolepis genistoides D. Don (vernacular name Palo azul; Palo) are traditionally consumed in the Republic of Paraguay in South America for the treatment of diabetes and kidney disease, and is sold in Japan as dietary supplement. This study aimed to elucidate the mechanism of anti-diabetes activity of Palo, especially focused on insulin resistance. Palo promoted adipocytes differentiation and regulated adipokine profiles in 3T3-L1 adipocytes by modulation of PPARγ, a major regulator of adipose differentiation. Human adipocyte showed almost similar profile with 3T3-L1 against Palo treatment. Furthermore, Palo treatment (250 or 1000 mg/kg) was performed with C57BL/6J mice for 14 weeks, being fed high-fat-diet (HFD60) simultaneously. Palo 250 mg/kg exhibited a tendency to decrease subcutaneous adipose volume along with increase of PPARγ and its target, adiponectin mRNA expression. In addition, as the other insulin targeted cell, effect on muscle differentiation was examined. Palo increased differentiation of C2C12 mouse muscle myoblasts by increase of IGF-1, myogenin, and myosine heavy chain (MHC) as well as 5'-AMP-activated protein kinase (AMPK) activation. Palo subsequently promoted myotube formation under differentiation condition. From the above, it was clarified that Palo acts variously on the differentiation and maturation of both adipocytes and muscle cells, and from the viewpoint of the regulatory mechanism for adipocytes, PPARγ-inducing action was shown to be a mechanism that acts across species.

Lack of antidiabetic effect of ethanolic extract of leaves of Solanum trilobatum Linn (Solanaceae)

Objective: The objective was to study the antidiabetic properties of ethanolic extract of the leaves of Solanum trilobatum (EEST) in streptozocin (STZ)-induced diabetics in Sprague-Dawley (SD) rats. Materials and Methods: EEST was prepared by using hot percolation method and the extract was used for antidiabetic screening. The SD rats were divided into six groups each of six animals, namely normal control, diabetic control, glibenclamide and EEST-treated groups at 125, 250, and 500 mg/kg. Except normal control animals, all the other animals were induced diabetes with intraperitoneal injection of STZ (55 mg/kg). The control and diabetic animals were treated with respective assigned treatment once daily for 21 consecutive days. The blood glucose was monitored at regular intervals and biochemical parameters such as aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, urea, and creatinine were measured with terminal sample. At the end of the study, the animals were sacrificed; lung, heart, stomach, liver, and kidney were harvested and absolute organ weight was measured. Results: The rats administrated with the extracts at dose of 500 mg/kg body weight (BW)/day showed significant antidiabetic activity and this effect was comparable with that of glibenclamide. The diabetic control animals showed significant increased levels of total cholesterol, high-density lipoprotein (HDL), HDL ratio, very-low-density lipoprotein, and glibenclamide, and EEST prevented STZ-induced cholesterol impairments. EEST did not show any significant antidiabetic effect at 125 and 250 mg/kg BW/day treated rats. Conclusion: EEST showed significant antidiabetic activity at 500 mg/kg and prevented STZ-induced metabolic changes in experimental animals. EEST did not show any antidiabetic activity in diabetic animals treated with 125 and 250 mg/kg of EEST.

EVALUATION OF ANTIHYPERGLYCAEMIC EFFECT OF BUTEA MONOSPERMA LEAF EXTRACT ON ADRENALIN INDUCED AND HIGH GLUCOSE FEED ANIMAL MODEL

Objective: As per the ethnopharmacological information has Butea monosperma been used to treat diabetes mellitus by the tribal people of tropical and subtropical areas. However, there is no much more scientific report available about the antidiabetic property of the leaves of the plant. Hence, the study was undertaken to evaluate the antidiabetic effect of ethanolic extract of B. monosperma on blood levels of adrenaline-induced and glucose feed diabetic rabbits.Methods: The three different doses of the extracts (100, 200, and 400 mg/kg) were administered orally to an experimental animal. The animals were induced diabetics by adrenaline and high glucose diet. Blood glucose level was measured accordingly. For antidiabetic activity, photocolorimeter was used to monitoring the blood glucose level with crest kit box (GOP-POD method).Results: The extracts showed considerable dose-dependent activity. However, the dose 400 mg/kg showed considerable lower of blood glucose level. p&lt;0.01 indicates the significance result. 8 hrs reading 182.5±3.83 for 400 mg does is most effective for reducing blood sugar.Conclusion: The study indicates that the ethanolic extract of B. monosperma leaves possesses antidiabetic properties which suggest the presence of biologically active components.

Anti-diabetic Effects of Ethanol Extract from Bitter Melon in Mice Fed a High-fat Diet.

ABSTRACTPresent study aimed to determine the effect of ‘bitter melon’, a popularly used fruit in Bangladesh and several other Asian countries, on high-fat-diet-induced type 2 diabetes. To investigate the effect, ethanol extract from bitter melon (BME) as a dietary supplement with mouse chow was used. BME was found to significantly attenuate the high-fat diet (HFD) -induced body weight and total fat mass. BME also effectively reduced the insulin resistance induced by the HFD. Furthermore, dietary supplementation of BME was highly effective in increasing insulin sensitivity and reducing hepatic fat and obesity. These results indicate that BME could be effective in attenuating type 2 diabetes and could therefore be a preventive measure against type 2 diabetes.

Antioxidant and antidiabetic activity of Phoenix pusilla Gaertn. unripe fruit extract in streptozotocin-induced sprague dawley rats

The fruit of Phoenix pusilla Garetn. has been used in herbal medicines, as it is sweet, sour, cooling and laxative, cardiotonic, aphrodisiac, carminative and roborant. The objective of the present study was to investigate the antioxidant and antidiabetic effect of ethanolic extract of unripe fruit of Phoenix pusilla in streptozotocin-induced diabetic rats. The extract was analyzed for the presence of various phytoconstituents like tannins, flavonoids, vitamin C, vitamin E, protein, carbohydrates, lipids and phenolic compounds. Streptozotocin (40 mg/kg body weight, i.p.) was administered to induce diabetes in adult rats. The extract (100 and 200 mg/kg) and glibenclamide (6 mg/kg) were administered orally for 21 days to evaluate antioxidant and antidiabetic activity. Blood glucose, serum total cholesterol and triglycerides levels were estimated. Carbohydrate and lipid metabolizing enzymes glucose-6-phosphatase, fructose-1,6-diphosphatase, glycolytic enzymes like hexokinase and liver-function enzymes superoxide dismutase (SOD), glutathione peroxidase (GPx), production of thiobarbituric acid reactive substances (TBARS), reduced glutathione (GSH), alkaline phosphatase (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALP), renal-function markers like urea and creatinine level were also measured. Histopathology of pancreas was studied. The results indicated that extract normalized the blood, liver, renal and pancreatic functions in streptozotocin-induced diabetic rats. Hence it can be concluded that the extract possesses antioxidant and antidiabetic activity. The findings support the conventional usage of Phoenix pusilla unripe fruit in treating diabetes.
