Microplastics in anoxic/aerobic membrane bioreactor (A/O-MBR): Characteristics, biofilms, degradation and carrier for antibiotic resistance genes

Wastewater treatment plants (WWTPs) are proposed as one of the ways for microplastics (MPs) to enter the water environment. MPs as the carrier of other pollutants, the inevitable aging process will change the characteristics and fate of MPs. Herein, the physicochemical property alteration, microbial community evolution and antibiotic resistance genes (ARGs) occurrence of the aged MPs surface were analyzed in A/O-MBR system. Remarkable alteration in MPs surface properties, which included the decrease in hydrophobicity and the increase in oxygen-containing functional groups, led to aging of MPs. Aging MPs are more likely to grow biofilms, which further increased the probability of microbial colonization. The genera Hydrogenophaga, Exiguobacterium, Enterobacter, Vibrio, Flavobacterium and Bacillus on MPs surface showed higher degradation potential. ARGs determination results indicated that MPs could enrich ARGs on their surface, and the relative abundance of β-lactam ARGs on different MPs surfaces was the highest. The above research results indicated that MPs in sewage treatment systems not only undergo aging, but also carry ARGs, causing further harm to the environment.

Investigation of the vaccine potential of an in silico designed FepA peptide vaccine against Shigella flexneri in mice model

BackgroundShigellosis is one of the significant causes of diarrhea in Bangladesh. It is a global health problem; approximately 1.3 million people die yearly from Shigellosis. The current treatment method, using different antibiotics against Shigellosis is ineffective. Moreover, it becomes a worrying situation due to the emergence of antibiotic-resistant pathogenic microbes responsible for these diarrheal diseases. MethodologyPrevious immunoinformatics study predicted a potential peptide from the Ferric enterobactin protein (FepA) of Shigella spp. In this study, we have chemically synthesized the FepA peptide. As a highly immunogenic, FepA peptide conjugated with KLH has been tested in mice model with complete and incomplete adjuvants as a vaccine candidate. ResultsImmunological analysis showed that all vaccinated mice were immunologically boosted, which was statistically significant (P-value 0.0325) compared to control mice. Immunological analysis for bacterial neutralization test result was also statistically significant (P-value 0.0468) where each ELISA plate was coated with 1 × 107S. flexneri cells. The Challenge test with 1 × 1012S. flexneri cells to each vaccinated and controlled mice showed that 37.5 % of control (non-vaccinated) mice died within seven days after the challenge was given while 100 % of vaccinated mice remained strong and stout. The analyses of the post-challenge weight loss of the mice were also significant (P-value 0.0367) as the weight loss percentage in control mice was much higher than in the vaccinated mice. The pathological and phenotypic appearances of vaccinated mice were also clearly differentiable compared with control mice. Thus all these immunological analysis and pathological appearances directly supported our FepA peptide as a potential immune booster. ConclusionThis study provides evidence that the FepA peptide is a highly immunogenic vaccine candidate against S. flexneri. Therefore, these findings inspire future trials for the evaluation of the suitability of this vaccine candidate against Shigellosis.

Microbial Contamination and Antibiotic Resistance in Food and Water: Assessing the Threat of Staphylococcus aureus in Lahore Metropolitan

Background: The research project in Lahore, Pakistan, seeks to detect S. aureus in drinking water, raw milk, and yogurt samples due to health concerns. S. aureus is a pathogenic bacterium with potential risks if present in food and water sources.Method: In this study, 300 samples of raw milk, yogurt, and drinking water were collected in Lahore. The presence of S. aureus was determined through morphological, microscopic, and biochemical methods. The biochemical analysis included testing for specific features of S. aureus. Disk diffusion technology was employed to assess the antimicrobial susceptibility of the isolates, following the recommendations of the Clinical Laboratory Standards Institute. Molecular confirmation was achieved through 16S rRNA sequence analysis using universal and specific primers.Results: The investigation uncovered that 6% of drinking water samples, 9% of yogurt samples and 58% of raw milk samples were tainted with S. aureus. These findings were further validated through 16S rRNA sequence analysis, affirming their reliability. S. aureus exhibited notable resistance rates, with 100% resistance to penicillin and 95% resistance to erythromycin. Conversely, resistance to ciprofloxacin and gentamicin was lower, at 10% and 5% each, indicating the potential efficacy of these antibiotics in treating S. aureus infections.Conclusion: The study emphasizes the risk of S. aureus infection from raw milk consumption in Lahore, Pakistan, due to inadequate sanitary practices. It stresses the necessity of implementing stricter measures in dairy production and water treatment to ensure public safety and reduce multidrug-resistant bacteria prevalence. The antibacterial potential and effects of Rhodiola sp. on gut microbiota

AbstractThe challenge of antimicrobial resistance requires new solutions, and Rhodiola sp. is a promising candidate due to its rich medicinal history. This review focuses on Rhodiola sp., especially R. rosea, highlighting its phytochemical constituents, such as salidroside, rosavins, and other phenolics, and their antimicrobial potential. We investigate the efficacy of Rhodiola sp. against various pathogenic bacteria, suggesting their value in combating antimicrobial resistance. Additionally, Rhodiola compounds are being investigated both as direct antimicrobials and as resistance modifiers that increase the efficacy of antibiotics. Rhodiola extracts and salidroside modulate gut microbiota, thereby affecting health and physiological and metabolic disorders. This illustrates the comprehensive therapeutic potential of Rhodiola sp. We emphasize the importance of understanding the complex interactions between Rhodiola phytochemicals, their antimicrobial effects, and their effects on gut microbiota as well as the need for in-depth research. Further studies will be crucial for the development of holistic treatment approaches. Finally, Rhodiola sp. is highlighted as an important natural resource in the fight against microbial pathogens and antibiotic resistance. Research on Rhodiola sp. should continue to further unravel pharmacological and health-promoting potential and thus address an important public health challenge.

Antibiotic resistance of microorganisms isolated from the urine of patients before surgical treatment for benign prostatic hyperplasia: results from a 7-year single-center study

Introduction. The tendency of microorganisms to develop resistance mechanisms is a widely discussed and significant problem worldwide. Studying regional differences in the qualitative characteristics of microorganisms provides valuable information for empirically preventing and treating infectious complications, as well as providing an enhanced understanding of the variability in microbial community properties within the clinical context of diseases and patients' comorbidity status.Objective. To assess the antibiotic resistance of microorganisms isolated in high titers from the urine samples of patients with benign prostate hyperplasia (BPH) prior to surgery.Materials & Methods. This single-center, retrospective study conducted from March 2016 to February 2023 included 59 suprapubic-draining BPH-patients (Group I), 46 drainage-free BPH-patients with leukocyturia (Group II), and 44 drainage-free BPH-patients and no leukocyturia (Group III). Inclusion criteria: indications for BPH surgery, no history of sexually transmitted diseases, no symptoms of urinary tract infection, and no prostate cancer. The patient's voluntary informed consents to participate were also obtained.Results. The rates of resistance to ciprofloxacin in patients of Group I were statistically significantly higher compared to Group II (85.7% vs 55.6%, p = 0.002). Resistance of gram-negative microorganisms to meropenem and imipenem was higher in Group III compared to Group I (31.1% vs 13.7%, p = 0.006 for meropenem and 44.5% vs 9.8%, p = 0.001 for imipenem, respectively). Resistance of Gram-positive microorganisms to ampicillin in Groups I to III was 13.6%,6.3% and 20.0%, respectively with no significant difference between groups (p > 0.05). However, there was extremely high resistance among verified Gram-positive organisms to all the drugs in the fluoroquinolone class (ciprofloxacin, norfloxacin, levofloxacin) ranged from 63.6% to 80.0%.Conclusion. The present study demonstrates that the isolation frequency of antibiotic-resistant microorganisms from the urine sample of drainage-free BPH-patients or no clinical and laboratory signs of inflammation in the urinary tract is high. The presence of antibiotic resistance provides risks for developing difficult-to-control infectious complications. Currently, assessment of urine-derived microbial antibiotic resistance should be considered in every BPH-patient with indications for surgical management of bladder outlet obstruction, regardless of the presence of risk factors.

Antimicrobial and Antiviral Nanofibers Halt Co-Infection Spread via Nuclease-Mimicry and Photocatalysis.

The escalating spread of drug-resistant bacteria and viruses is a grave concern for global health. Nucleic acids dominate the drug-resistance and transmission of pathogenic microbes. Here, imidazolium-type poly(ionic liquid)/porphyrin (PIL-P) based electrospun nanofibrous membrane and its cerium (IV) ion complex (PIL-P-Ce) are developed. The obtained PIL-P-Ce membrane exhibits high and stable efficiency in eradicating various microorganisms (bacteria, fungi, and viruses) and decomposing microbial antibiotic resistance genes and viral nucleic acids under light. The nuclease-mimetic and photocatalytic mechanisms of the PIL-P-Ce are elucidated. Co-infection wound models in mice with methicillin-resistant S. aureus and hepatitis B virus demonstrate that PIL-P-Ce integrate the triple effects of cationic polymer, photocatalysis, and nuclease-mimetic activities. As revealed by proteomic analysis, PIL-P-Ce shows minimal phototoxicity to normal tissues. Hence, PIL-P-Ce has potential as a "green" wound dressing to curb the spread of drug-resistant bacteria and viruses in clinical settings.

Antimicrobial Activity of Individual Volatile Compounds from Various Essential Oils.

Interest in natural remedies has grown recently due to a variety of public health concerns such as microbial antibiotic resistance. This global health concern necessitates innovative approaches to combat bacterial infections. Building upon established therapeutic uses of essential oils, this research focused on the volatile constituents of essential oils. The volatile antimicrobial activity of these constituents was studied by employing a derivative of a modified disk diffusion assay for quantitative comparisons. This study emphasizes the significance and value of exploring natural compounds as alternatives to traditional antibiotics and provides insights into their mechanisms and applications in contending with bacterial pathogens.

Anti-biofilm activity of marine algae-derived bioactive compounds.

A large number of microbial species tend to communicate and produce biofilm which causes numerous microbial infections, antibiotic resistance, and economic problems across different industries. Therefore, advanced anti-biofilms are required with novel attributes and targets, such as quorum sensing communication system. Meanwhile, quorum sensing inhibitors as promising anti-biofilm molecules result in the inhibition of particular phenotype expression blocking of cell-to-cell communication, which would be more acceptable than conventional strategies. Many natural products are identified as anti-biofilm agents from different plants, microorganisms, and marine extracts. Marine algae are promising sources of broadly novel compounds with anti-biofilm activity. Algae extracts and their metabolites such as sulfated polysaccharides (fucoidan), carotenoids (zeaxanthin and lutein), lipid and fatty acids (γ-linolenic acid and linoleic acid), and phlorotannins can inhibit the cell attachment, reduce the cell growth, interfere in quorum sensing pathway by blocking related enzymes, and disrupt extracellular polymeric substances. In this review, the mechanisms of biofilm formation, quorum sensing pathway, and recently identified marine algae natural products as anti-biofilm agents will be discussed.

Evaluation of antimicrobial activity and bioactive compound analysis of Verbascum thapsus L. A folklore medicinal plant

BackgroundThe prevalence of microbial infectious diseases is a major contributor to morbidity and mortality on a global scale due to the emergence of antibiotic-resistant microbes, despite the fact that many new antibiotics have been introduced for several decades. In present scenario, the potential utilization of natural plant derived extracts for medicinal and therapeutic purposes has increased remarkably due to adverse effects of synthetic drugs and resistance among pathogenic microbes. Aim of the studyVerbascum thapsus L. (Common mullein) has been traditionally used as a medicine for lung, skin and throat and many other disorders since time immemorial, therefore, the current study was aimed to evaluate the antimicrobial activity of leaf and root extracts of Verbascum thapsus against pathogenic bacterial and fungal strains and its phytochemical composition by spectral studies. MethodsSoxhlet extraction method was employed for extraction process by using varying polarity solvents like hexane, ethyl acetate and methanol. Phytochemical analysis of methanol extract was determined by Fourier Transform-Infrared Spectroscopy (FTIR) and Gas Chromatography-Mass Spectrometry (GC–MS). Antimicrobial activity was evaluated by determining the MIC and MBC/MFC of each extract by broth dilution method. ResultsThe antimicrobial competence of each extract sample was evaluated against six pathogenic bacterial strains and three fungal strains. Methanolic extract of leaves and the ethyl acetate extract of roots of Verbascum thapsus exhibited significant zones of inhibition against all the tested microorganisms. The highest inhibition zone was observed against the bacterial strain Bacillus subtilis (24.1±0.18 mm) followed by Streptococcus pyogenes (22.9±0.64 mm) and the fungal strain Aspergillus niger (22.8±0.55 mm) at a concentration of 500 μg/ml. Methanolic leaf extract strongly inhibited the growth of tested microbial strains with MIC ranged from 15.625 to 125 μg/ml. The FTIR and GCMS analysis confirms the presence of bioactive compound, possessing broad range of therapeutic activities. ConclusionThis Presence of various important phytoconstituents and broad-spectrum antimicrobial activity points Verbascum thapsus as a promising source for the isolations of molecules responsible for antimicrobial activities.

Investigating Biofilms: Advanced Methods for Comprehending Microbial Behavior and Antibiotic Resistance.

Biofilms, which consist of microorganisms enclosed in an extracellular polymeric material (EPS), hold immense importance in the fields of environmental research, industry, and medicine. They play a significant role in ecosystem dynamics and stability, but they also pose issues such as biofouling, corrosion, and pollution. Biofilms in medical environments are linked to persistent infections and elevated healthcare expenses. The EPS matrix plays a crucial role in maintaining the structural integrity and antibiotic resistance of these structures. The research primarily investigates the role of the EPS matrix in facilitating horizontal gene transfer among biofilm communities, with a particular emphasis on EPS and its impact on this process. The process is recognized as a pivotal mechanism in the emergence of antibiotic resistance, underscoring the crucial function of EPS in the dynamics of biofilms. The analysis also highlights the significant financial constraints caused by biofilms in several industries. Biofilm-associated infections in the healthcare sector result in escalated treatment expenses and extended hospitalization periods. In an industrial context, biofilms have a role in increasing maintenance expenses and product contamination, emphasizing the need for efficient management solutions. This review presents the most recent progress in biofilm research, emphasizing the utilization of sophisticated imaging tools and molecular methodologies. In addition to conventional imaging techniques, the research explores the utilization of sophisticated molecular tools, such as DNA and RNA sequencing, in conjunction with proteomics. These approaches are essential for assessing the genetic and metabolic mechanisms that regulate biofilm development and antibiotic resistance. The review underscores the significance of employing an interdisciplinary methodology in the study of biofilms. By incorporating a range of approaches, such as sophisticated imaging and molecular analysis, a comprehensive understanding of biofilm dynamics may be achieved. This approach also opens up possibilities for developing novel solutions to address the negative impacts of biofilms on health, industry, and the environment.

Using UV/peracetic acid as pretreatment for subsequent bio-treatment of antibiotic-containing wastewater treatment: Mitigating microbial inhibition and antibiotic resistance genes proliferation

UV/peracetic acid (PAA) treatment presents a promising approach for antibiotic removal, but its effects on microbial community and proliferation of antibiotic resistance genes (ARGs) during the subsequent bio-treatment remain unclear. Thus, we evaluated the effects of the UV/PAA on tetracycline (TTC) degradation, followed by introduction of the treated wastewater into the bio-treatment system to monitor changes in ARG expression and biodegradability. Results demonstrated effective TTC elimination by the UV/PAA system, with carbon-centered radicals playing a significant role. Crucially, the UV/PAA system not only eliminated antibacterial activity but also inhibited potential ARG host growth, thereby minimizing the emergence and dissemination of ARGs during subsequent bio-treatment. Additionally, the UV/PAA system efficiently removed multi-antibiotic resistant bacteria and ARGs from the bio-treatment effluent, preventing ARGs from being released into the environment. Hence, we propose a multi-barrier strategy for treating antibiotic-containing wastewater, integrating UV/PAA pre-treatment and post-disinfection with bio-treatment. The inhibition of ARGs transmission by the integrated system was verified through actual soil testing, confirming its effectiveness in preventing ARGs dissemination in the surrounding natural ecosystem. Overall, the UV/PAA treatment system offers a promising solution for tackling ARGs challenges by controlling ARGs proliferation at the source and minimizing their release at the end of the treatment process.

Dynamic Ag-mediated electron transfer confined ZnO nanorods for boosted photocatalytic bacterial disinfection

Infectious diseases, particularly bacterial infections, pose a global crisis with significant impacts on public health and economic stability due to the emergence of multi-drug resistance and limitations in existing therapeutic options. The present study focused on the development of photocatalytic disinfectant and silver-decorated hexagonal zinc oxide nanorods (Ag/ZnO NRs) were fabricated through an ultrasonic-assisted solvothermal method, and investigated for the photocatalytic inactivation on E. coli. The morphological, structural, and optical characteristics of the fabricated Ag/ZnO NRs were systematically studied through various analytical techniques. Here, 3%Ag/ZnO NRs showed 96.9% inactivation of E. coli within 120 min of visible light irradiation. The excellent inactivation efficiency of the photocatalyst could be attributed to the higher charge carrier mobility, efficient electron-hole pair separation, and highvisible light harvesting ability. The Schottky heterojunction was proposed as a possible mechanism for the photoinactivation of E. coli. Further, the impacts of different dosages of nanocomposites (NCs) and varying pH on bacterial inactivation were studied under visible light exposure. The reusability and stability of the NCs were investigatedfor 6 consecutive cycles of photoinactivation and it showed excellent efficiency (91.16%) of the photocatalyst for prolonged usage. The real-time application was assessed using reclaimed wastewaterand the influence on the organic matter was determined using COD analysis. The proposed study offers a significant point reference for the practical application of photocatalysis forthe inactivation of antibiotic-resistant microbes present in water bodies and environmental wastewater.

Comparison of tet(X4)-containing contigs assembled from metagenomic sequencing data with plasmid sequences of isolates from a cohort of healthy subjects.

Recently discovered tet(X) gene variants have provided new insights into microbial antibiotic resistance mechanisms and their potential consequences for public health. This study focused on detection, analysis, and characterization of Tet(X4)-positive Enterobacterales from the gut microbiota of a healthy cohort of individuals in Singapore using cultivation-dependent and cultivation-independent approaches. Twelve Tet(X4)-positive Enterobacterales strains that were previously obtained from the cohort were fully genome-sequenced and comparatively analyzed. A metagenomic sequencing (MS) data set of the same samples was mined for contigs that harbored the tet(X4) resistance gene. The sequences of tet(X4)-containing contigs and plasmids sequences were compared. The presence of the resistance genes floR and estT (previously annotated as catD) was detected in the same cassette in 10 and 12 out of the 12 tet(X4)-carrying plasmids, respectively. MS detected tet(X4)-containing contigs in 2 out of the 109 subjects, while cultivation-dependent analysis previously reported a prevalence of 10.1%. The tet(X4)-containing sequences assembled from MS data are relatively short (~14 to 33 kb) but show high similarity to the respective plasmid sequences of the isolates. Our findings show that MS can complement efforts in the surveillance of antibiotic resistance genes for clinical samples, while it has a lower sensitivity than a cultivation-based method when the target organism has a low abundance. Further optimization is required if MS is to be utilized in antibiotic resistance surveillance.IMPORTANCEThe global rise in antibiotic resistance makes it necessary to develop and apply new approaches to detect and monitor the prevalence of antibiotic resistance genes in human populations. In this regard, of particular interest are resistances against last-resort antibiotics, such as tigecycline. In this study, we show that metagenomic sequencing can help to detect high abundance of the tigecycline resistance gene tet(X4) in fecal samples from a cohort of healthy human subjects. However, cultivation-based approaches currently remain the most reliable and cost-effective method for detection of antibiotic-resistant bacteria.

Characteristics of microbial communities and antibiotic resistance genes in typical rivers of the western Qinghai Lake basin

Environmental factors and anthropogenic activity are key factors that shape the distribution of bacteria and antibiotic resistance genes (ARGs) in natural environments. However, few studies have focused on the occurrence of bacteria and ARGs in remote and pristine environments. In this study, the distribution of bacteria and ARGs in two typical rivers of the western Qinghai Lake basin was investigated. Results showed that cold-resistant Planomicrobium sp. was the predominant genus due to the low temperature, followed by unclassified_f_Planococcaceae. High nitrogen nutrients increased the abundance and diversity of the bacteria community and denitrification was the dominant means of bacterial nitrate reduction. Tetracycline resistance genes including tetA, tetB, and tetC were the dominant ARGs in the western Qinghai Lake basin, ranging from 2.30 ​× ​103 to 1.91 ​× ​108 copies/L, while the low abundances of intI1, and ARGs such as sul1 indicated low anthropogenic activity in the western Qinghai Lake basin. Finally, the strong positive correlation between ARGs and intI1 highlights the potential transmission risk of ARGs through cross- or co-selection by horizontal gene transfer. Our study emphasized the adaptation of bacteria to the environment and the facilitation of anthropogenic activity in the propagation of ARGs in natural environments.

Farm Animal Welfare-From the Farmers' Perspective.

Improvements in the welfare of animals in the intensive production industries are increasingly being demanded by the public. Scientific methods of welfare improvement have been developed and are beginning to be used on farms, including those provided by precision livestock farming. The number of welfare challenges that animals are facing in the livestock production industries is growing rapidly, and farmers are a key component in attempts to improve welfare because their livelihood is at stake. The challenges include climate change, which not only exposes animals to heat stress but also potentially reduces forage and water availability for livestock production systems. Heat-stressed animals have reduced welfare, and it is important to farmers that they convert feed to products for human consumption less efficiently, their immune system is compromised, and both the quality of the products and the animals' reproduction are adversely affected. Livestock farmers are also facing escalating feed and fertiliser costs, both of which may jeopardise feed availability for the animals. The availability of skilled labour to work in livestock industries is increasingly limited, with rural migration to cities and the succession of older farmers uncertain. In future, high-energy and protein feeds are unlikely to be available in large quantities when required for the expanding human population. It is expected that livestock farming will increasingly be confined to marginal land offering low-quality pasture, which will favour ruminant livestock, at the expense of pigs and poultry unable to readily digest coarse fibre in plants. Farmers also face disease challenges to their animals' welfare, as the development of antibiotic resistance in microbes has heralded an era when we can no longer rely on antibiotics to control disease or improve the feed conversion efficiency of livestock. Farmers can use medicinal plants, pro-, pre- and synbiotics and good husbandry to help maintain a high standard of health in their animals. Loss of biodiversity in livestock breeds reduces the availability of less productive genotypes that survive better on nutrient-poor diets than animals selected for high productivity. Farmers have a range of options to help address these challenges, including changing to less intensive diets, diversification from livestock farming to other enterprises, such as cereal and pseudocereal crops, silvopastoral systems and using less highly selected breeds. These options may not always produce good animal welfare, but they will help to give farm animals a better life.

Insights into gut microbiomes in stem cell transplantation by comprehensive shotgun long-read sequencing

The gut microbiome is a diverse ecosystem, dominated by bacteria; however, fungi, phages/viruses, archaea, and protozoa are also important members of the gut microbiota. Exploration of taxonomic compositions beyond bacteria as well as an understanding of the interaction between the bacteriome with the other members is limited using 16S rDNA sequencing. Here, we developed a pipeline enabling the simultaneous interrogation of the gut microbiome (bacteriome, mycobiome, archaeome, eukaryome, DNA virome) and of antibiotic resistance genes based on optimized long-read shotgun metagenomics protocols and custom bioinformatics. Using our pipeline we investigated the longitudinal composition of the gut microbiome in an exploratory clinical study in patients undergoing allogeneic hematopoietic stem cell transplantation (alloHSCT; n = 31). Pre-transplantation microbiomes exhibited a 3-cluster structure, characterized by Bacteroides spp. /Phocaeicola spp., mixed composition and Enterococcus abundances. We revealed substantial inter-individual and temporal variabilities of microbial domain compositions, human DNA, and antibiotic resistance genes during the course of alloHSCT. Interestingly, viruses and fungi accounted for substantial proportions of microbiome content in individual samples. In the course of HSCT, bacterial strains were stable or newly acquired. Our results demonstrate the disruptive potential of alloHSCTon the gut microbiome and pave the way for future comprehensive microbiome studies based on long-read metagenomics.

Differential effects of Bacillus species – fermented products on antibiotic resistome and virulence factor gene composition in the cecal digesta of broilers

Abstract This study investigated the differential effects of Bacillus subtilis – and Bacillus licheniformis – fermented products (defined as SFP and LFPs, respectively) on microbial antibiotic resistance gene (ARG) and virulence factor gene (VFG) composition in the cecal digesta of 35-day-old broilers by using metagenomic sequencing. First, 160 1-day-old unsexed Arbor Acres broiler chicks were randomly allocated to four treatment groups: basal diet (CON group); basal diet + enramycin (10 mg/kg, ENM group); basal diet + SFPs (108 colony-forming units (CFU) of B. subtilis spores/kg, SFP group); and basal diet + LFPs (108 CFU of B. licheniformis spores/kg, LFP group). Principal coordinate analysis of ARG and VFG composition indicated distinct clustering among the cecal samples of the groups. At the antibiotic resistance class level, LFP treatment increased the expression of peptide resistance genes and decreased the expression of aminocoumarin resistance genes compared with the other groups. Compared with the other groups, LFP treatment promoted bcrA, ugd, and efrB expression but suppressed parY expression, whereas SFP treatment inhibited efrA expression. The abundance of the peptide resistance gene bcrA in Lachnoclostridium species was higher in the CON and LFP groups than in the ENM and SFP groups, whereas the abundance of the peptide resistance gene rpoB2 in Bacteroides species was lower in the ENM and LFP groups than in the SFP group. No specific VFGs were regulated only by SFPs or LFPs. SFP and LFP treatment inhibited clpC expression compared with the other groups. clpC abundance in Bacteroides species was lower in the LFP group than in the CON group, whereas its abundance in Faecalibacterium species was lower in the SFP group than in the CON and ENM groups. These results demonstrated that SFPs and LFPs differentially regulate microbial ARG and VFG composition in the cecal digesta of broilers. LFP supplementation modulated more antibiotic resistance classes and ARGs than did SFP supplementation.

Gut resistome profiling reveals high diversity and fluctuations in pancreatic cancer cohorts.

Pancreatic cancer is one of the deadliest cancer, with a 5-year overall survival rate of 11%. Unfortunately, most patients are diagnosed with advanced stage by the time they present with symptoms. In the past decade, microbiome studies have explored the association of pancreatic cancer with the human oral and gut microbiomes. However, the gut microbial antibiotic resistance genes profiling of pancreatic cancer patients was never reported compared to that of the healthy cohort. In this study, we addressed the gut microbial antibiotic resistance genes profile using the metagenomic data from two online public pancreatic cancer cohorts. We found a high degree of data concordance between the two cohorts, which can therefore be used for cross-sectional comparisons. Meanwhile, we used two strategies to predict antibiotic resistance genes and compared the advantages and disadvantages of these two approaches. We also constructed microbe-antibiotic resistance gene networks and found that most of the hub nodes in the networks were antibiotic resistance genes. In summary, we describe the panorama of antibiotic resistance genes in the gut microbes of patients with pancreatic cancer. We hope that our study will provide new perspectives on treatment options for the disease.

Size effects of microplastics on antibiotic resistome and core microbiome in an urban river

Microplastics (MPs) in aquatic environments provide a new ecological niche that facilitates the attachment of antibiotic-resistance genes (ARGs) and pathogens. However, the effect of particle size on the colonization of antibiotic resistomes and pathogens remains poorly understood. To address this knowledge gap, this study explored the antibiotic resistome and core microbiome on three distinct types of MPs including polyethylene, polypropylene, and polystyrene (PS), with varying sizes of 30, 200, and 3000 μm by metagenomic sequencing. Our finding showed that the ARG abundances of the PS type increased by 4-folds with increasing particle size from 30 to 3000 μm, and significant differences in ARG profiles were found across the three MP types. In addition, the concentrations of ARGs and mobile genetic elements (MGEs) were markedly higher in the MPs than in the surrounding water, indicating their enrichment at these artificial interfaces. Notably, several pathogens such as Pseudomonas aeruginosa, Mycobacterium tuberculosis, and Legionella pneumophila were enriched in MP biofilms, and the co-occurrence of ARGs and virulence factor genes (VFGs)/MGEs suggested the presence of pathogenic antibiotic-resistant microbes with potential mobility. Both redundancy analysis (RDA) and structural equation modeling (SEM) demonstrated that physicochemical properties such as zeta potential, MP size, and contact angle were the most significant contributors to the antibiotic resistome. Strikingly, no significant differences were observed in the health risk scores of the ARG profiles among different sizes and types of MPs. This study expands our knowledge on the impact of MP size on microbial risks, thus enhancing our understanding of the potential health hazards they pose.
