Prevalence of antibiotic resistance genes in bacteria from Gomti and Ganga rivers: implications for water quality and public health.

Rivers serve as a significant habitat and water sources for diverse organisms, including humans. An important environmental and public health concern is the increase in antibiotic-resistant bacteria (ARBs) and genes (ARGs) in aquatic ecosystems brought about by excessive pollutant flow. The research highlighted that river water, which is receiving discharge from wastewater treatment plants, is harbouring multidrug-resistant bacteria. River water samples were collected in January, April, July and October 2022 from three separate locations of each Gomti and Ganga river. A total of 114 bacteria were isolated from Gomti as well as the Ganga River. All the isolates were tested for their resistance to various antibiotics by disc diffusion method. The isolated bacteria were tested for the antibiotic resistance genes using PCR and were identified by 16s rRNA sequencing. The ARBs percentages for each antibiotic were as follows: ampicillin (100%); cefotaxime (96.4, 63.1%); erythromycin (52.6, 57.8%); amikacin (68.4, 50.8%); tetracycline (47.3, 54.3%); nalidixic acid (47.3, 45.6%); streptomycin (68.4, 49.1%); gentamycin (43.8, 35%); chloramphenicol (26.3, 33.3%); neomycin (49.1, 29.8%) and ciprofloxacin (24.5, 7.01%). Further, antibiotic resistance genes in Gomti and Ganga water samples disclose distinctive patterns, including resistance to ermB (25, 40%); tetM (25, 33.3%); ampC (44.4, 40%) and cmlA1 (16.6%). Notably cmlA1 resistant genes were absent in all bacterial strains of the Gomti River. Additionally, gyrA gene was not found in both the river water samples. The presence of ARGs in the bacteria from river water shows threat of transferring these genes to native environmental bacteria. To protect the environment and public health, constant research is necessary to fully understand the extent and consequences of antibiotic resistance in these aquatic habitats.

Diversity, functions, and antibiotic resistance genes of bacteria and fungi are examined in the bamboo plant phyllosphere that serve as food for the giant pandas.

The phyllosphere of bamboo is rich in microorganisms that can disrupt the intestinal microbiota of the giant pandas that consume them, potentially leading to their death. In the present study, the abundance, diversity, biological functions (e.g., KEGG and CAZyme), and antibiotic resistance genes (ARGs) of bacteria and fungi in two bamboo species phyllosphere (Chimonobambusa szechuanensis, CS; Bashania fangiana, BF) in Daxiangling Nature Reserve (an important part of the Giant Panda National Park) were investigated respectively by amplicon sequencing of the whole 16S rRNA and ITS1-ITS2 genes on PacBio Sequel and whole-metagenome shotgun sequencing on Illumina NovaSeq 6000 platform. The results suggested that there were respectively 18 bacterial and 34 fungi biomarkers between the phyllosphere of the two species of bamboo. Beta diversity of bacteria and fungi communities exited between the two bamboos according to the (un)weighted UniFrac distance matrix. Moreover, the functional analysis showed that the largest relative abundance was found in the genes related to metabolism and global and overview maps. Glycoside hydrolases (GHs) and glycosyl transferases (GTs) have a higher abundance in two bamboo phyllospheres. Co-occurrence network modeling suggested that bacteria and fungi communities in CS phyllosphere employed a much more complex metabolic network than that in BF, and the abundance of multidrug, tetracycline, and glycopeptide resistance genes was higher and closely correlated with other ARGs. This study references the basis for protecting bamboo resources foraged by wild giant pandas and predicts the risk of antibiotic resistance in bamboo phyllosphere bacterial and fungal microbiota in the Giant Panda National Park, China.

Carbapenems and colistin resistance genes isolated in Musca domestica from a garbage dump near a hospital in Lima.

Motivation for the study. The presence of antibiotic resistance genes in bacteria isolated from common flies is a potential public health hazard because it facilitates the presence and spread of antibiotic resistance genes in the environment. Main findings. Thirty-eight bacterial strains identified in 14 species were isolated from within the fly bodies, of which 31 strains showed resistance to carbapenems and 26 strains showed resistance to colistin. Seven bacterial strains showed carbapenem resistance genes and one Escherichia coli strain had resistance to KPC, OXA-48 and mcr-1. Implications. This is the first report of antibiotic resistance genes in bacteria carried by common flies in Peru. The objective was to determine the presence of carbapenem resistance genes and plasmid resistance to colistin (mcr-1) in bacteria isolated from Musca domestica in a garbage dump near a hospital in Lima, Peru. Bacteria with phenotypic resistance to carbapenemics were isolated on CHROMagar mSuperCARBATM medium and colistin resistance profiling was performed using the colistin disk elution method. Detection of blaKPC, blaNDM, blaIMP, blaOXA-48, blaVIM and mcr-1 genes was performed by conventional PCR. The antimicrobial susceptibility profile was determined using the automated MicroScan system. We found that 31/38 strains had phenotypic resistance to carbapenemics and 26/38 strains had phenotypic resistance to colistin with a minimum inhibitory concentration ≥ 4 µg/ml. Finally, we identified seven bacterial strains with carbapenem resistance genes (OXA-48 and KPC) and one bacterial strain with plasmid resistance to colistin (mcr-1). One Escherichia coli strain had three resistance genes: KPC, OXA-48 and mcr-1.

Filamentous bacteria-induced sludge bulking can alter antibiotic resistance gene profiles and increase potential risks in wastewater treatment systems

Sludge bulking caused by filamentous bacteria is a prevalent issue in wastewater treatment systems. While previous studies have primarily concentrated on controlling sludge bulking, the biological risks associated with it have been overlooked. This study demonstrates that excessive growth of filamentous bacteria during sludge bulking can significantly increase the abundance of antibiotic resistance genes (ARGs) in activated sludge. Through metagenomic analysis, we identified specific ARGs carried by filamentous bacteria, such as Sphaerotilus and Thiothrix, which are responsible for bulking. Additionally, by examining over 1,000 filamentous bacterial genomes, we discovered a diverse array of ARGs across different filamentous bacteria derived from wastewater treatment systems. Our findings indicate that 74.84% of the filamentous bacteria harbor at least one ARG, with the occurrence frequency of ARGs in these bacteria being approximately 1.5 times higher than that in the overall bacterial population in activated sludge. Furthermore, genomic and metagenomic analyses have shown that the ARGs in filamentous bacteria are closely linked to mobile genetic elements and are frequently found in potentially pathogenic bacteria, highlighting potential risks posed by these filamentous bacteria. These insights enhance our understanding of ARGs in activated sludge and underscore the importance of risk management in wastewater treatment systems.

Occurrence and dissemination of antibiotics and antibiotic resistance in aquatic environment and its ecological implications: a review.

The occurrence of antibiotics and antibiotic-resistant bacteria (ARBs), genes (ARGs), and mobile genetic elements (MGEs) in aquatic systems is growing global public health concern. These emerging micropollutants, stemming from improper wastewater treatment and disposal, highlight the complex and evolving nature of environmental pollution. Current literature reveals potential biases, such as a geographical focus on specific regions, leading to an insufficient understanding of the global distribution and dynamics of antibiotic resistance in aquatic systems. There is methodological inconsistency across studies, making it challenging to compare findings. Potential biases include sample collection inconsistencies, detection sensitivity variances, and data interpretation variability. Gaps in understanding include the need for comprehensive, standardized long-term monitoring programs, elucidating the environmental fate and transformation of antibiotics and resistance genes. This review summarizes current knowledge on the occurrence and dissemination of emerging micropollutants, their ecological impacts, and the global health implications of antimicrobial resistance. It highlights the need for interdisciplinary collaborations among researchers, policymakers, and stakeholders to address the challenges posed by antibiotic resistance in aquatic resistance in aquatic systems effectively. This review highlights widespread antibiotic and antibiotic resistance in aquatic environment, driven by human and agricultural activities. It underscores the ecological consequences, including disrupted microbial communities and altered ecosystem functions. The findings call for urgent measures to mitigate antibiotics pollution and manage antibiotic resistance spread in water bodies.

Screening for Antibiotic Resistance Genes in Bacteria and the Presence of Heavy Metals in the Upstream and Downstream Areas of the Wadi Hanifah Valley in Riyadh, Saudi Arabia.

Valley surface water is considered a focal public health concern owing to the presence of multi-drug-resistant bacteria. The distribution of antimicrobial resistance (AMR) bacteria in the surface water is affected by the presence of multiple factors, including antibiotics coming from wastewater discharge or other contaminant sources such as pharmaceuticals, biocides, and heavy metals. Furthermore, there is evidence suggesting that high levels of antibiotic resistance genes (ARGs) can be transferred within bacterial communities under the influence of heavy metal stress. Hence, the primary aim of this study is to investigate the presence of heavy metals and bacterial ARGs in upstream as well as downstream locations of Wadi Hanifah Valley in Riyadh, Saudi Arabia. Sample collection was conducted at eighteen surface water sites within the valley in total. The selection of ARGs was associated with the most common antibiotics, including β-lactam, tetracycline, erythromycin, gentamicin, sulphonamide, chloramphenicol, vancomycin, trimethoprim, and colistin antibiotics, which were detected qualitatively using polymerase chain reaction (PCR) technology. The tested antibiotic resistance genes (ARGs) included (blaNDM-1 (for the antibiotic class Beta-lactamases), mecA (methicillin-resistant Staphylococcus aureus), tet(M) and tet(B) (for the antibiotic class Tetracycline), ampC (for the antibiotic class Beta-lactamases), vanA (for the antibiotic class vancomycin), mcr-1 (for the antibiotic class colistin), erm(B) (for the antibiotic class erythromycin), aac6'-Ie-aph2-Ia (for the antibiotic class Gentamicin), sulII (for the antibiotic class sulphonamide), catII (for the antibiotic class Chlorophincol), and dfrA1 (for the antibiotic class trimethoprim). Moreover, an assessment of the levels of heavy metals such as lithium (Li), beryllium (Be), chromium (Cr), cobalt (Co), arsenic (As), cadmium (Cd), tin (Sn), mercury (Hg), and lead (Pb) was conducted by using inductively coupled plasma mass spectrometry (ICPMS). According to our findings, the concentrations of sulphonamide, erythromycin, and chloramphenicol ARGs (erm(B), sulII, and catII) were observed to be the most elevated. Conversely, two ARGs, namely mecA and mcr-1, were not detected in the samples. Moreover, our data illustrated a significant rise in ARGs in the bacteria of water samples from the upstream sites as compared with the water samples from the downstream sites of Wadi Hanifah Valley. The mean concentration of Li, Be, Cr, Co, As, Cd, Sn, Hg, and Pb in the water samples was estimated to be 37.25 µg/L, 0.02 µg/L, 0.56 µg/L,0.32 µg/L, 0.93 µg/L, 0.01 µg/L, 200.4 µg/L, 0.027 µg/L, and 0.26 µg/L, respectively, for the selected 18 sites. Furthermore, it was revealed that the concentrations of the screened heavy metals in the water samples collected from various sites did not surpass the maximum limits set by the World Health Organization (WHO). In conclusion, this study offers a concise overview of the presence of heavy metals and ARGs in water samples obtained from the Wadi Hanifah Valley in Riyadh, KSA. Such findings will contribute to the ongoing monitoring and future risk assessment of ARGs spread in surface water.

Crucifer Lesion-Associated Xanthomonas Strains Show Multi-Resistance to Heavy Metals and Antibiotics.

Copper resistance in phytopathogens is a major challenge to crop production globally and is known to be driven by excessive use of copper-based pesticides. However, recent studies have shown co-selection of multiple heavy metal and antibiotic resistance genes in bacteria exposed to heavy metal and xenobiotics, which may impact the epidemiology of plant, animal, and human diseases. In this study, multi-resistance to heavy metals and antibiotics were evaluated in local Xanthomonas campestris pv. campestris (Xcc) and co-isolated Xanthomonas melonis (Xmel) strains from infected crucifer plants in Trinidad. Resistance to cobalt, cadmium, zinc, copper, and arsenic (V) was observed in both Xanthomonas species up to 25 mM. Heavy metal resistance (HMR) genes were found on a small plasmid-derived locus with ~ 90% similarity to a Stenotrophomonas spp. chromosomal locus and a X. perforans pLH3.1 plasmid. The co-occurrence of mobile elements in these regions implies their organization on a composite transposon-like structure. HMR genes in Xcc strains showed the lowest similarity to references, and the cus and ars operons appear to be unique among Xanthomonads. Overall, the similarity of HMR genes to Stenotrophomonas sp. chromosomal genomes suggest their origin in this genus or a related organism and subsequent spread through lateral gene transfer events. Further resistome characterization revealed the presence of small multidrug resistance (SMR), multidrug resistance (MDR) efflux pumps, and bla (Xcc) genes for broad biocide resistance in both species. Concurrently, resistance to antibiotics (streptomycin, kanamycin, tetracycline, chloramphenicol, and ampicillin) up to 1000 µg/mL was confirmed.

Using transcriptome analysis to reveal the effects of bromoacetamide at drinking water ambient concentrations on the transformation of eDNA carrying ARGs

Antibiotic resistance genes (ARGs) are frequently detected in drinking water. Transformation of extracellular antibiotic resistance genes (eARGs) is an important pathway for horizontal gene transfer, which facilitates the spread of ARGs among different species. Although most studies have pointed out that the presence of some common carbon-containing disinfection by-products, such as trihalomethanes, can increase the frequency of transformation by increasing the content of reactive oxygen species (ROS), nitrogen-containing disinfection by-products (N-DBPs), such as bromoacetamide (BAcAm), can also induce bacterial resistance to antibiotics by stimulating ROS. However, it is unclear whether such N-DBPs can similarly promote the transformation of ARGs in bacteria. In this study, the effect of BAcAm on the transformation of eDNA carrying ARGs was investigated, and found that DNA integration and repair (ruv, rec), efflux pumps (emr), and genes related to membrane proteins (bam, opm) were up-regulated in E. coli after addition of BAcAm. Furthermore, in addition to analyzing common functional genes, in terms of metabolic gene expression, which is usually overlooked, the genes related to amino acid biosynthesis (pot, arg, lys) were up-regulated while genes related to sulfur metabolism, which has a higher metabolic cost (thi, sssu) were down-regulated, which suggests that E. coli is able to effectively defend itself against the threat posed by BAcAm to its survival. In conclusion, the factors affecting the conversion of eDNA carrying ARGs into E. coli cells by BAcAm were suggested and elaborated that E. coli carrying ARGs can adapt to various unfavorable environments by regulating relevant metabolic mechanisms and survive in drinking water environment for a long period of time.

High prevalence of antibiotic-resistant and metal-tolerant cultivable bacteria in remote glacier environment

Studies of antibiotic-resistant bacteria (ARB) have mainly originated from anthropic-influenced environments, with limited information from pristine environments. Remote cold environments are major reservoirs of ARB and have been determined in polar regions; however, their abundance in non-polar cold habitats is underexplored. This study evaluated antibiotics and metals resistance profiles, prevalence of antibiotic resistance genes (ARGs) and metals tolerance genes (MTGs) in 38 ARB isolated from the glacier debris and meltwater from Baishui Glacier No 1, China. Molecular identification displayed Proteobacteria (39.3%) predominant in debris, while meltwater was dominated by Actinobacteria (30%) and Proteobacteria (30%). Bacterial isolates exhibited multiple antibiotic resistance index values > 0.2. Gram-negative bacteria displayed higher resistance to antibiotics and metals than Gram-positive. PCR amplification exhibited distinct ARGs in bacteria dominated by β-lactam genes blaCTX-M (21.1–71.1%), blaACC (21.1–60.5%), tetracycline-resistant gene tetA (21.1–60.5%), and sulfonamide-resistant gene sulI (18.4–52.6%). Moreover, different MTGs were reported in bacterial isolates, including mercury-resistant merA (21.1–63.2%), copper-resistant copB (18.4–57.9%), chromium-resistant chrA (15.8–44.7%) and arsenic-resistant arsB (10.5–44.7%). This highlights the co-selection and co-occurrence of MTGs and ARGs in remote glacier environments. Different bacteria shared same ARGs, signifying horizontal gene transfer between species. Strong positive correlation among ARGs and MTGs was reported. Metals tolerance range exhibited that Gram-negative and Gram-positive bacteria clustered distinctly. Gram-negative bacteria were significantly tolerant to metals. Amino acid sequences of blaACC,blaCTX-M,blaSHV,blaampC,qnrA, sulI, tetA and blaTEM revealed variations. This study presents promising ARB, harboring ARGs with variations in amino acid sequences, highlighting the need to assess the transcriptome study of glacier bacteria conferring ARGs and MTGs.

High incidence of multiple intI1 genomic gene cassettes in Aeromonas strains

Antimicrobial resistance profiles of Aeromonas have been used as an effective indicator for monitoring resistances in aquatic environments. Integron gene cassettes are a major reservoir of antibiotic resistance genes in bacteria. To date, little has been reported on the antimicrobial resistance or integrons of Aeromonas in bullfrog aquaculture. In this study, 27 isolates belonging to four Aeromonas species (A. hydrophila, A. veronii, A. caviae, and A. jandaei) were isolated from farmed diseased American bullfrogs (Rana catesbeiana). all of them were multidrug-resistant, exhibiting resistance to at least seven drugs from five antibiotic classes. Class 1 integrons (intI1s), containing nine types of gene cassette arrays encoding resistance to trimethoprim (dfrA1, dfrA12, dfrA15, dfrA17), streptomycin/spectinomycin (aadA2), fluoroquinolones/aminoglycosides (aac(6′)-Ib-cr), rifampin (arr3), proline-rich antimicrobial peptides (ptrB), and multiple antimicrobial agents involved in biofilm development (dgs), were detected in 26 (26/27, 96.3%) of the isolates. Seven patterns were identified, and six of them contained two or three gene cassettes, in 27 isolates. This work highlights unusually high frequency (21/27, 77.8%) of multiple intI1 gene cassette arrays and three novel intI1 gene cassettes or arrays (dgc, ptrB, and aac(6′)-Ib-cr-arr3) in the isolates of Aeromonas spp. from American bullfrogs.

Presence of Antibiotic Resistance Genes in Bacteria Isolated from Raw Cow Milk obtained from Bowen University Dairy Farm.

Antibiotic resistance is becoming more prevalent thus posing a severe threat to global health. Bacteria were obtained from raw milk produced on the Bowen University campus using conventional methods. Morphological and biochemical testing were used to carry out phenotypic identification process. Six widely used broad-spectrum antibiotics were used in tests to determine their sensitivity. Bacteria resistant to four or fewer antibiotics were chosen for the amplification of antibiotic-resistance genes while using the 16S rRNA technique for sequencing. Eighty-one isolates were obtained, and the sensitivity patterns revealed 81.8% resistance to cefuroxime and 98.1% resistance to cefixime and amoxicillin/clavulanate (augmentin). Twelve out of the multidrug-resistant isolates were sequenced and they revealed 100% resistance to cefixime and amoxicillin/clavulanate (augmentin), 80% to cefuroxime, and 53.3% and 6.7% to gentamicin and ciprofloxacin respectively. Only ofloxacin showed a sensitivity of 100 percent. The sequenced isolates were identified as Pseudocitrobacter faecalis (-), Escherichia coli (-), Bacillus tropicus (+), Citrobacter koseri (-), Providencia burhodogranariea (-), Enterobacter hormaechei subsp. Xiangfangensis (-), Bacillus pumilus (+), Lysinibacillus boronitolerans (+), Klebsiella aerogenes (-), Klebsiella pneumoniae subsp. rhinoscleromatis (-) 2 isolates, and Acinetobacter baumannii. The TEM, SHV-1 and CTX-M genes were detected in the Citrobacter koseri isolate, while the TEM and SHV-1 genes were also found in the Escherichia coli isolate. The high rate of resistance in bacteria observed is alarming. Adequate milk storage procedures and antibiotic usage in dairy cattle farms must be regulated for safety. It is recommended that antibiotics should not be used in farm animals unless when highly necessary.

Impact of soil supplemented with pig manure on the abundance of antibiotic resistant bacteria and their associated genes.

This study was conducted to evaluate the abundance of antibiotic resistant bacteria and their resistance genes from agriculture soil supplemented with pig manure. Uncultivable soil sample was supplemented with pig manure samples under microcosm experimental conditions and plated on Luria-Bertani (LB) agar incorporated with commercial antibiotics. The supplementation of soil with 15% pig manure resulted in the highest increase in the population of antibiotic resistant bacteria (ARB)/multiple antibiotic resistant bacteria (MARB). Seven genera that included Pseudomonas, Escherichia, Providencia, Salmonella, Bacillus, Alcaligenes and Paenalcaligenes were the cultivable ARB identified. A total of ten antibiotic resistant bacteria genes (ARGs) frequently used in clinical or veterinary settings and two mobile genetic elements (MGEs) (Class 1 and Class 2 integrons) were detected. Eight heavy metal, copper, cadmium, chromium, manganese, lead, zinc, iron, and cobalt were found in all of the manure samples at different concentrations. Tetracycline resistance genes were widely distributed with prevalence of 50%, while aminoglycoside and quinolone-resistance gene had 16% and 13%, respectively. Eighteen ARB isolates carried more than two ARGs in their genome. Class 1 integron was detected among all the 18 ARB with prevalence of 90-100%, while Class 2 integron was detected among 11 ARB. The two classes of integron were found among 10 ARB. Undoubtedly, pig manure collected from farms in Akure metropolis are rich in ARB and their abundance might play a vital role in the dissemination of resistance genes among clinically-relevant pathogens.

Detection of Horizontal Gene Transfer Mediated by Natural Conjugative Plasmids in E. coli.

Conjugation represents one of the main mechanisms facilitating horizontal gene transfer in Gram-negative bacteria. This work describes methods for the study of the mobilization of naturally occurring conjugative plasmids, using two naturally-occurring plasmids as an example. These protocols rely on the differential presence of selectable markers in donor, recipient, and conjugative plasmid. Specifically, the methods described include 1) the identification of natural conjugative plasmids, 2) the quantification of conjugation rates in solid culture, and 3) the diagnostic detection of the antibiotic resistance genes and plasmid replicon types in transconjugant recipients by polymerase chain reaction (PCR). The protocols described here have been developed in the context of studying the evolutionary ecology of horizontal gene transfer, to screen for the presence of conjugative plasmids carrying antibiotic-resistance genes in bacteria found in the environment. The efficient transfer of conjugative plasmids observed in these experiments in culture highlights the biological relevance of conjugation as a mechanism promoting horizontal gene transfer in general and the spread of antibiotic resistance in particular.

Isolation, characterization, and fingerprinting of some multidrug resistance clinical isolates from patients in Al-Qaseem Hospitals, Saudi Arabia

The aim of this study was to investigate the prevalence of antibiotic resistance genes in bacteria isolated from Saudi Arabian patients in the Al-Qassim region. We also examine the application of Random Amplified Polymorphic DNA- Polymerase chain reaction (RAPD-PCR) in genetic diversity of the strains in use. In this study, we have used 29 different clinical samples from Al-Qassim hospitals which consists of wound swabs, blood, sputum and urine. Clinical samples were streaked onto sterile Petri dishes containing nutrient agar media using sterile dry swabs. Additionally antibiotic sensitivity tests, micro scan walkway susceptibility tests and extraction of genomic DNA, amplification of RAPD and Inter simple sequence repeat assay were performed in this study. This study results confirmed from the clinical samples were streaked onto sterile Petri dishes containing nutrient agar media using sterile dry swabs. The PCR analysis in ISSR was confirmed that 30–70% of E. coli in Asia and Africa carried ESBLs in the GLASS 2020 study, which is on par with the 59% in VINARES 2016–2017. Comparing VINARES 2016–2017 to other nations, the prevalence of ESBL carriage among K. pneumoniae was 35%. However, RAPD results showed that all employed strains and primers could successfully fingerprint DNA. Between isolated bacterial strains, similarity dendrograms were produced. The technique generated bands with the same intensity as the typical PCR carried out using pure DNA, and it worked for all 29 bacterial strains examined. In conclusion, the results show that all of the isolates have at least one antibiotic-resistance gene, and that the PCR approach is a quick, easy, and reliable way to identify genes.

Characteristics and driving factors of antibiotic resistance genes in aquaculture products from freshwater ponds in China Yangtze River Delta

ABSTRACT Antibiotic resistance genes (ARGs) are widespread in aquaculture and pose a huge threat to aquaculture organisms and human health. In this study, occurrences and relative abundances of ARGs were analysed in the guts of products cultured in freshwater ponds in the Yangtze River Delta region in China. A total of 29 ARGs were found in the gut samples, with detection frequencies ranging from 4.8% to 81%, and the relative abundances (ARGs/16S rRNA) ranging from 10−7 to 1. In addition, the human dietary intake of ARGs via aquaculture products was assessed, where the daily intake of most ARGs via aquaculture products was higher than those via PM2.5 and drinking water, but lower than that via vegetables. The relative abundances of MGE (IS613, Tp614, tnpA and int1) were significantly correlated with those of multiple ARGs, indicating the horizontal gene transfer (HGT) of ARGs among gut microorganisms. Proteobacteria, Firmicutes and Actinobacteria were the dominated microbial communities found in the guts of aquaculture products. In addition, significant correlations were found between Cyanobacteria and int1, between Nitrospira and tetE, and between sul2 and aadA2, indicating potential same hosts of these genes. In addition, results from co-correlation indicated both HGT (dominated by MGEs) of ARGs and the enrichment of ARGs in bacteria. MGEs, mostly int1, were more effective than bacteria in increasing the ARG abundance. This study could provide a better understanding of the transmission of ARGs in the aquaculture environment and improve the quality of aquaculture products and the ecology.

Quaternary Ammonium Salts: Insights into Synthesis and New Directions in Antibacterial Applications.

The overuse of antibiotics has led to the emergence of a large number of antibiotic-resistant genes in bacteria, and increasing evidence indicates that a fungicide with an antibacterial mechanism different from that of antibiotics is needed. Quaternary ammonium salts (QASs) are a biparental substance with good antibacterial properties that kills bacteria through simple electrostatic adsorption and insertion into cell membranes/altering of cell membrane permeability. Therefore, the probability of bacteria developing drug resistance is greatly reduced. In this review, we focus on the synthesis and application of single-chain QASs, double-chain QASs, heterocyclic QASs, and gemini QASs (GQASs). Some possible structure-function relationships of QASs are also summarized. As such, we hope this review will provide insight for researchers to explore more applications of QASs in the field of antimicrobials with the aim of developing systems for clinical applications.

ASSESSMENT OF THE RELATIVE ABUNDANCE OF ANTIBIOTIC RESISTANCE GENES OF BACTERIA IN THE GUT OF PIGLETS (SUS SCROFA DOMESTICUS) IN THE EARLY NEONATAL PERIOD

The article presents the assessment of the presence and relative amount of antibiotic resistance genes of bacteria in the gut microbiome of piglets in the early neonatal period. The genes for beta-lactam antibiotics (penicillins, cephalosporins, carbapenems, monobactams) have become the dominant majority of the antibiotic resistance genes present in both healthy and sick piglets. Tetracycline resistance genes were also found in the samples of healthy and sick animals. Healthy animals had the aminoglycoside resistance gene Aph3-III (relative content - 81.15%). The quinolone antibiotic resistance genes QnrB5 and QnrB19 were present in both healthy and sick animals, and the gene QnrD was identified only in the piglets with diarrhea (relative content - 10.27%). The same relative abundance of the sulfonamide resistance gene SulII (relative content - 82%) was demonstrated in both healthy and sick piglets, while the gene SulIII was found only in the animals with diarrhea (relative content - 13.44%). The results show a wide spread of antibiotic resistance genes in the population of healthy piglets and piglets with diarrhea, and this requires special attention from pig breeding farms in order to minimize further migration of antibiotic resistance genes.

Evaluating the occurrence frequency of horizontal gene transfer induced by different degrees of heavy metal stress

Horizontal gene transfer of heavy metal resistance genes and antibiotic resistance genes in bacteria frequently occurred during composting. However, the relevancy between heavy metal pollution with different degrees and the occurrence of horizontal gene transfer during composting was unclear. Therefore, this study aimed to clarify the underlying relationship. The changes of resistance gene abundance and network analysis results showed that heavy metal pollution induced high level of resistance gene abundance in microorganisms and promoted the occurrence of horizontal gene transfer. Furthermore, regression fitting models indicated that the relevancy between resistance genes abundance and horizontal gene transfer under high pollution (HP: R = 0.90) was weakener than that under low pollution (LP: R = 0.95) and medium pollution (MP: R = 0.97). Ultimately, structural equation models further verified that the abundance of resistance genes was mainly caused by horizontal gene transfer during LP (coefficients: 0.997) and MP (coefficients: 0.993) composting. However, the high level of resistance gene abundance was caused by host bacteria (coefficients: 0.775) and horizontal gene transfer (coefficients: 0.701) during HP composting. The serious heavy metal pollution had selection pressure on host bacteria to induce them to develop resistance during HP composting, rather than only through horizontal gene transfer. Therefore, mild heavy metal pollution was more liable to cause horizontal gene transfer and increase the spread risk of resistance genes during composting. This study helped to better understand the risks caused by heavy metal pollution by evaluating the frequency of horizontal gene transfer at different pollution levels during composting.
