A Review of Antimicrobial Polymer Coatings on Steel for the Food Processing Industry.

This article will focus on the issue of protection against the pathogenic biofilm development on steel surfaces within the food sectors, highlighting steel's prominence as a material choice in these areas. Pathogenic microorganism-based biofilms present significant health hazards in the food industry. Current scientific research offers a variety of solutions to the problem of protecting metal surfaces in contact with food from the growth of pathogenic microorganisms. One promising strategy to prevent bacterial growth involves applying a polymeric layer to metal surfaces, which can function as either an antiadhesive barrier or a bactericidal agent. Thus, the review aims to thoroughly examine the application of antibacterial polymer coatings on steel, a key material in contact with food, summarizing research advancements in this field. The investigation into polymer antibacterial coatings is organized into three primary categories: antimicrobial agent-releasing coatings, contact-based antimicrobial coatings, and antifouling coatings. Antibacterial properties of the studied types of coatings are determined not only by their composition, but also by the methods for applying them to metal and coating surfaces. A review of the current literature indicates that coatings based on polymers substantially enhance the antibacterial properties of metallic surfaces. Furthermore, these coatings contribute additional benefits including improved corrosion resistance, enhanced aesthetic appeal, and the provision of unique design elements.

Water - Based Polyurethanes for Antibacterial Coatings: an Overview

The spread of bacterial infections and the resulting loss of life and material have led scientists to research ways to develop knowledge in these disease-causing microorganisms. Usage of antibacterial polymer coatings is an important part of this area. Among the polymers, water-based polyurethanes (WPU’s) have received a lot of attention in biomedical fields such as antibacterial coatings, biological products and wound dressings due to their unique properties such as reduced use of volatile organic compounds (VOC), biocompatibility, the possibility of using a variety of raw materials. In this review, the methods of creating antibacterial properties in polymers, the synthesis of WPU’s and WPU-based antibacterial coatings are reviewed. The products produced as a result of these studies have been recommended for various fields such as the dressing and packaging industries, and the coating of medical equipment.

Recent Advance in Polymer Coatings Combating Bacterial Adhesion and Biofilm Formation†

Comprehensive SummaryImplantable medical device‐associated infections (DAIs) originating from bacterial adhesion and biofilm formation have threatened to the health and life of patients. Antibacterial polymer coatings with antifouling and/or bactericidal properties have showed great potentials to combat DAI issues. In this review, we report recent advances in antibacterial polymer coatings fighting bacterial adhesion and biofilm formation on implantable biomaterial surfaces. We summarize the mechanisms of bacterial adhesion and biofilm formation, which provides guidance for the design of antibacterial coatings. We describe the polymer and coating preparation methods and discuss the structure‐property relationships of antibacterial polymer coatings. Applications of these polymer coatings in medical catheters, orthopaedic implants, and other applications are elaborated. Future challenges and prospects associated with antibacterial polymer coatings for implantable medical devices are discussed.

Comparable Studies on Nanoscale Antibacterial Polymer Coatings Based on Different Coating Procedures.

The antibacterial activity of different antibiotic and metal-free thin polymer coatings was investigated. The films comprised quaternary ammonium compounds (QAC) based on a vinyl benzyl chloride (VBC) building block. Two monomeric QAC of different alkyl chain lengths were prepared, and then polymerized by two different polymerization processes to apply them onto Ti surfaces. At first, the polymeric layer was generated directly on the surface by atom transfer radical polymerization (ATRP). For comparison purposes, in a classical route a copolymerization of the QAC-containing monomers with a metal adhesion mediating phosphonate (VBPOH) monomers was carried out and the Ti surfaces were coated via drop coating. The different coatings were characterized by X-ray photoelectron spectroscopy (XPS) illustrating a thickness in the nanomolecular range. The cytocompatibility in vitro was confirmed by both live/dead and WST-1 assay. The antimicrobial activity was evaluated by two different assays (CFU and BTG, resp.,), showing for both coating processes similar results to kill bacteria on contact. These antibacterial coatings present a simple method to protect metallic devices against microbial contamination.

Effect of titanium surface topography on plasma deposition of antibacterial polymer coatings

Plasma processing, e.g., functionalisation and deposition of antibacterial coatings, is often used to enhance surface properties of biomaterials. Plasma is, however, a non-uniform active medium, and the result of processing depends on the nature of both the plasma and the substratum. Here we show that when an antibacterial coating (i.e., polyterpenol) is plasma polymerised onto four types of titanium substrata that differ in their micro- and nano-scale topography (but not the bulk chemistry), the distribution of functional groups, e.g., OH and CO, in the polymer across the surface differs sufficiently, and so does the antibacterial activity of the resulting material system. While the addition of a coating hinders biofilm formation by Staphylococcus aureus and Pseudomonas aeruginosa, the bactericidal effect is significantly stronger in polymers deposited onto surfaces possessing lower degrees of nanoscale roughness, e.g., substrata after mechanical and chemical polishing. The reduced antibacterial efficacy of polymers on substrata with greater surface roughness (e.g., on mechanically polished or lotus leaf-like surfaces) is attributed to a greater extent of thickness non-uniformity and heterogeneity in the functional group distribution across the surface. These findings suggest that the magnitude and distribution of topographical features of the substratum should be considered when designing plasma-enabled surface modification strategies.

Synthesis of bactericidal polymer coatings by sequential plasma-induced polymerization of 4-vinyl pyridine and gas-phase quaternization of poly-4-vinyl pyridine

Plasma-based technology is an alternative to produce universal polymer coatings with the appropriate requirements of robustness and stability for antibacterial applications. Here, we proposed a sequential two-step alternative to synthesize antibacterial polymer coatings. A non-isothermal plasma reactor, operated at atmospheric pressure (Patm) and room temperature (Troom), was used to induce free radical polymerization of 4-vinyl pyridine (4VP) on high-density polyethylene (PE). In a subsequent step, the poly-4VP (P4VP) films were treated with a bromoethane/He gas stream to produce quaternized P4VP (P4VPQ) films. Chemical structure of polymer films was validated by infrared and UV–visible spectroscopy, and morphology was evaluated by optical and atomic force microscopy; scanning electron microscopy was used to determine films thickness, which was then used to estimate the surface charge density. The bactericidal capacity was determined with a standard test by using Escherichia coli. Both types of films had an estimated charge density in the order of 1016 positive charges per cm2; P4VP films removed about 95–99% of bacteria, whereas 4PVPQ films eliminated 100%. The methodology proposed for the synthesis of antibacterial polymer coatings is simpler, faster, and more environmentally friendly than other plasma-based methods; operation at Troom and Patm may also have a significant effect on the economics and the ease of implementation of the process at commercial scale. The suggested approach may facilitate the development of new universal coatings, and operating plasma conditions could be extrapolated for engineering antibacterial coatings in industrial areas where bacterial attachment is of concern.

Nanomechanics of pH-Responsive, Drug-Loaded, Bilayered Polymer Grafts.

Stimuli-responsive polymer films play an important role in the development of smart antibacterial coatings. In this study, we consider complementary architectures of polyelectrolyte films, including a thin chitosan layer (CH), poly(acrylic acid) (PAA) brushes, and a bilayer structure of CH grafted to PAA brushes (CH/PAA) as possible candidates for targeted drug delivery platforms. Atomic force microscopy (AFM) was employed to study the structure-mechanical property relationship for these mono- and bi-layered polymer grafts at pH 7.4 and 4.0, corresponding to physiological and biofilm formation conditions, respectively. Herein, the surface interactions between polymer grafts and the negatively charged silica colloid attached to an AFM lever are considered as representative interactions between the antibacterial coating and a bacteria/biofilm. The bilayered structure of CH/PAA showed significantly reduced adhesive interactions in comparison to pure CH but slightly higher interactions in comparison to PAA films. Among PAA and CH/PAA films, upon grafting CH over the PAA brushes, the normal stiffness increased by 10-fold at pH 7.4 and 20-fold at pH 4.0. Notably, the study also showed that the addition of an antibiotic drug such as multicationic Tobramycin (TOB) impacts the mechanical properties of the antibacterial coatings. Competition between TOB and water molecules for the PAA chains is shown to determine the structural properties of PAA and CH/PAA films loaded with TOB. At high pH (7.4), the TOB molecules, which remain multicationic, strongly interact with polyanionic PAA, thereby reducing the film's compressibility. On the contrary, at low pH (4.0), the water molecules preferentially interact with TOB in comparison to uncharged PAA chains and, upon TOB release, results in a stronger film collapse together with an increase in adhesive interactions between the probe, the surface, and the elastic modulus of the film. The bacterial proliferation on these platforms when compared to the measured mechanical properties shows a direct correlation; hence, understanding nanomechanical properties can provide insights into designing new antibacterial polymer coatings.

Competitive Adsorption of Polyelectrolytes onto and into Pellicle-Coated Hydroxyapatite Investigated by QCM-D and Force Spectroscopy

A current effort in preventive dentistry is to inhibit surface attachment of bacteria using antibacterial polymer coatings on the tooth surface. For the antibacterial coatings, the physisorption of anionic and cationic polymers directly onto hydroxyapatite (HA) and saliva-treated HA surfaces was studied using quartz crystal microbalance, force spectroscopy, and atomic force microscopy. First, single species adsorption is shown to be stronger on HA surfaces than on silicon oxide surfaces for all polymers (i.e., Gantrez, sodium hyaluronate (NaHa), and poly(allylamine-co-allylguanidinium) (PAA-G75)). It is observed through pH dependence of Gantrez, NaHa, and PAA-G75 adsorption on HA surfaces that anionic polymers swell at high pH and collapse at low pH, whereas cationic polymers behave in the opposite fashion. Thicknesses of Gantrez, NaHa, and PAA-G75 are 52 nm (46 nm), 35 nm (11 nm), and 6 nm (54 nm) at pH 7 (3.5), respectively. Second, absorption of charged polymer is followed by absorption of the oppositely charged polymer. Upon exposure of the anionic polymer layers, Gantrez and NaHa, to the cationic polymer, PAA-G75, films collapse from 52 to 8 nm and 35 to 11 nm, respectively. This decrease in film thickness is attributed to the electrostatic cross-linking between anionic and cationic polymers. Third, for HA surfaces pretreated with artificial saliva (AS), the total thickness decreases from 25 to 16 nm upon exposure to PAA-G75. Force spectroscopy is used to further investigate the PAA-G75/AS coating. The results show that the interaction between a negatively charged colloidal bead and the AS surface is strongly repulsive, whereas PAA-G75/AS is attractive but varies across the surface. Additionally, AFM studies show that AS/HA is smooth with a RMS roughness of 1.7 nm, and PAA-G75-treated AS/HA is rough (RMS roughness of 5.4 nm) with patches of polymer distributed across the surface with an underlying coating. The high roughness of PAA-G75 treated AS/HA is attributed to the strong adsorption of the relatively small PAA-G75 onto the heterogeneously distributed negatively charged AS surface. In addition, uptake of PAA-G75 by pellicle layer (saliva-treated HA surface) is observed, and the adsorbed amount of PAA-G75 on/into pellicle layer is ∼2 times more than that on/into AS layer. These studies show that polymer adsorption onto HA and saliva-coated HA depends strongly on the polymer type and size and that there is an electrostatic interaction between polymer and saliva and/or oppositely charged polymers that stabilizes the coatings on HA. Lastly, assessing the viability of the adherent bacteria collected from the PAA-G75-coated surfaces showed a significant reduction (∼93%) in bacterial viability when compared to bacteria collected from untreated and Gantrez-coated HA. These results suggest the potential antimicrobial activity of PAA-G75.

Poly[(mercaptopropyl)methylsiloxane] (PMMS)-based antibacterial polymer coatings prepared by a two-step sequential thiol–ene click chemistry

Poly[(mercaptopropyl)methylsiloxane] (PMMS)-based antibacterial polymer coatings have been prepared through a two-step sequential thiol–ene click chemistry utilizing 1-allyl-3-decylimidazolium bromide (ADIm) as antibacterial monomer and triallyl cyanurate (TAC) as the crosslinker. These films with different content of ADIm were characterized by thermogravimetric analysis (TGA), dynamic mechanical analysis (DMA) and pencil hardness. It was found that the mechanical and thermal properties of these films were largely influenced by the content of ADIm in the films. Films with imidazolium bromide groups displayed excellent antimicrobial activity against Staphylococcus aureus with 100% killing efficiency.

Facile preparation of antibacterial polymer coatings via thiol-yne click photopolymerization

Abstract Novel antibacterial polymer coatings were prepared by a facile thiol-yne click photopolymerization of 1-propargyl-3-alkyl-1,3-diazanyl-2,4-cyclopentadiene bromide ([PAIM]Br) and tetra(3-mercapto-propionate)pentaerythritol (PETMP) (2:1 molar ratio) using 2,2-dimethoxy-2-phenylacetophenone (DMPA) as initiator. The antibacterial activity of the coatings was tested against Staphylococcus aureus (ATCC 292130) and Escherichia coli (ATCC 25922) by the dynamic shake method. The evaluation results revealed the antibacterial polymer coatings exhibited excellent inhibitory activity against S. aureus and E. coli, especially for S. aureus.

Allylimidazolium salt based antibacterial polymer coatings produced by thiol–ene photocuring

Photocurable formulations containing trifunctional thiol, trifunctional ene, and antibacterial allylimidazolium salts have been employed for transparent antibacterial coatings. The antibacterial component 1-allyl-3-dodecylimidazolium salt (ADIm) is prepared and chemically attached to polymer networks using a one-step thiol–ene photocuring reaction. Ultra-small (USANS) and small angle neutron scattering (SANS) measurements show that the photocured polymers are loosely networked three-dimensional structures with a mass fractal of approximately 2.7±0.2. The minimum inhibitory concentration (MIC) for the ADIm was determined to be 500μg/ml and 15.63μg/ml for Escherichiacoli (Gram negative) and Staphylococcus aureus (Gram positive) bacteria, respectively. Coating formulations containing 10mol% of the antibacterial ADIm photocured on glass substrates showed strong antibacterial activity against environmental bacteria such as E. coli and/or S. aureus.

Targeted Synthesis of Porous Aromatic Frameworks and their Composites for Versatile, Facile, Efficacious, and Durable Antibacterial Polymer Coatings

Novel quaternary pyridinium-type porous aromatic frameworks, PAF-50, and their composites, AgCl-PAF-50, have been synthesized to effectively and efficiently inhibit the growth of bacteria. Most importantly, both PAF-50 and AgCl-PAF-50 have excellent compatibility with conventional polymers, which lead to great operation flexibility and versatility for antibactrial coatings on various medical devices simply via solution or spray coating.
