Chemical Composition Analysis andIn Vivo Anti-diabetic Activity of Aqueous Extract of Aerial Part ofPallenis spinosa in Diabetic Rats

Background: This work aims to confirm the anti-diabetic activity of Pallenis spinosa, as recommended in traditional medicine, and perform a chemical analysis of the aerial part of Pallenis spinosa. Methods: The isolated compounds have been identified by chemical methods and spectrometric analysis, such as UV, 1H NMR, and 13C NMR spectroscopy. With regard to the anti-diabetic activity, a series of experiments have been carried out in vivo on Westar albino rats. Diabetes has been induced in animals by an intraperitoneal injection of streptozotocine. They have been treated with an aqueous extract from the aerial part of Pallenis spinosa (250 and 500 mg/kg body weight) and glibenclamide (5 mg/kg body weight) every day for 21 days. Results: The chemical analysis of the aerial part of Pallenis spinosa led to the isolation of five known flavonoids, including patuletin 7-galactopyranoside, patuletin-3-O-α-L-rharnnopyranosyl (1-6)-β-D-galactopyranoside, tricin 7-glucopyranoside, tricin, and quercetin. The aqueous extract of both doses, 250 mg and 500 mg, has shown significant activity in reducing blood sugar, with 43.38% for the dose of 250 mg/kg and 37.76% for the dose of 500 mg/kg, as well as a significant decrease in the total fatness, triglycerides, and the total cholesterol levels in animals. The treatment was compared to the diabetes control group (p = 0.05). We used glibenclamide as a reference, and it showed similar results. Conclusion: In order to explore and develop new anti-diabetic drugs, more studies are needed on this plant for scrutinizing its mechanism of activity.

Antidiabetic Activity of Aqueous Extracts of <i>Laurus nobilis</i>, a Spice Used by Beninese Traditional Therapists

All over the world, humans developed knowledge about the use of medicinal plants to treat all kinds of diseases. Spices were included. This work aimed to evaluate the hypoglycemic property Laurus nobilis, a spice used by traditional therapists in Benin for diabetes management. The aqueous extracts of the leaves of Laurus nobilis were obtained by maceration. The phytochemical screening was carried out. Acute oral toxicity tests by single dose gavage of 2000 mg / kg body weight were performed in female Wistar rats. The weight of the animals, the serum creatinine, the ALT transaminase and the hemoglobin level were determined on day 0 and then on day 14. For the efficacy test, the extract was administered to rats at a concentration of 300 mg / Kg of body weight for 21 consecutive days. The blood tests performed were blood sugar, triglycerides and cholesterol. Histological sections were taken from the liver, kidneys and spleen. The phytochemical screening revealed the presence of tannins, mucilages and flavonoids. There were no deaths of rats and their weights, blood creatinine, ALT transaminase, and hemoglobin levels did not vary significantly during the toxicity study, indicating no toxicity. Regarding the efficacy test, Laurus nobilis extract significantly lowered the mean blood sugar level in rats on day 14 and day 21 (P Laurus nobilis displayed hypoglycemic properties in non-diabetic rats and was not acutely toxic. It could be used in the management of diabetes.

Phytochemical analysis and In vitro Antidiabetic activity of aqueous extract of Lagerstroemia speciosa and Aegle marmelos

Diabetes Mellitus is a metabolic disorder characterized by hypoglycemia, resulting from absolute or relative deficiency of insulin. Worldwide about 220million people affected. Low-cost herbal treatment is recommended due to their lesser side effect. The aim of the current study was to determine the antidiabetic activity of aqueous leaf extracts of Lagerstroemia speciosa (Lythraceae) and Aegle marmelos (Rutaceae) using alpha amylase inhibition assay and glucose diffusion inhibition assay. In alpha amylase inhibition assay, the leaf extracts of A.marmelos (at a concentration 100µg/ml) exhibited 54.68% of α amylase inhibitory activity with an IC 50 values 92.04µg/ml whereas the leaf extracts of L. speciosa (at a concentration of 100µg/ml) exhibited 60.36% of α amylase inhibitory activity with an IC50 value of 68.19µg/ml .However the mixture of plant extracts (at a concentration of 100µg/ml) exhibited 94.87% inhibition with an IC50 value of 13.145µg/ml. Both plant extracts showed appreciable α amylase inhibitory effects, but the mixture showed more inhibitory effects than the individual plant extracts. In glucose diffusion inhibition assay, the aqueous extract of A. marmelos leaves exhibited maximum glucose diffusion inhibition (76.886%) at 150 minutes as well as L. Speciosa leaf extract showed the maximum inhibition of 79.5357% at the same time interval. The mixture of plant extracts exhibited 87.4274% inhibition at 150 minutes which produces more effects than the two plants.

Antidiabetic activity of aqueous extract of leaves of cnestis ferruginea vahl ex DC. (Connaraceae) on alloxan-diabetic induced mice.

Abstract Background: Cnestis ferruginea is a plant species widely used in the traditional african medicine to treat diabetes mellitus. The present study aimed to evaluate the antidiabetic activities of the aqueous extract of the leaves of Cnestis ferruginea (AECF) in diabetic mice. Methods: The mice were grouped into five groups of five animals per group: Group A was not induced with alloxan, Group B animals were induced but not treated, Group C animals were treated with 500 mg / kg of BW of metformin, Group D and E animals were treated with 100, 200 mg/kg BW of AECF respectively. The extracts were administered to the animals orally for 14 days. Fasting blood sugar was measured by a glucometer. Serum concentrations of hematological and biochemical parameters were measured by standard methods. Results: The animals administered with 100 and 200 mg/Kg B.W of extract showed highly significant decrease (P ˂ 0.0001) in blood sugar level compared to the untreated animals. The 100 mg / kg BW dose of AECF produce low significant decrease (P <0.05) of total cholesterol, HDL Cholesterol, AST, ALT, urea, levels of white blood cells, platelets, hematocrit. The 200 mg / kg BW dose of AECF produce in addition significant decrease (P <0.01) of AST and urea. Conclusion: The 200 mg / kg dose of BW of AECF had a greater antidiabetic activity than the dose of 100 mg / kg BW. Keywords: Cnestis ferruginea; antidiabetic activity; alloxan; diabetes mellitus.

Evaluation of antidiabetic activity of aqueous extract of <em>Origanum floribundum</em> Munby leaves in alloxan induced diabetic rats wistar

The present study aimed to evaluate the phytochemicals and the antidiabetic properties of Origanum floribundum aqueous extract. Phytochemical screening was conducted by using various standard procedures. Acute toxicity of the extract was determined by OECD guidelines 423. The antidiabetic activity of the plant was determined by alloxan-induced diabetes in Wistar albino rats. Data obtained showed the detection of various secondary metabolites such as alkaloids, flavonoids, saponins, terpenoids in the extract. No mortality was observed during acute toxicity studies up to the dose of 2000 mg/kg. The aqueous extract presented the ability of reducing blood glucose levels. These findings indicated that Origanum floribundum aqueous extract had various phytopharmacological activities and thus it would be useful to isolate and characterize the compounds responsible for these bioactivities in the future.

Anti-diabetic Activity of Chrysophyllum albidum (G. Don) Stem Bark in Alloxan-induced Type 1 Diabetic Female Wistar Rats

Anti-diabetic activity of aqueous extract of Chrysophyllum albidum stem bark (AECASB) in female Wistar rats was investigated to confirm or refute the purported use and the underlying mechanism of action of C. albidum stem bark in the management of type 1 diabetes in Nigeria traditional medicine. Seventy female rats (180.60 ± 8.50 g) were assigned into seven groups. Rats in Group 1 (non-diabetic) were orally administered 1 mL of distilled water, while animals in Groups 2-7 were made diabetic and orally administered 1 mL of distilled water, 2.5 mg/kg body weight (bwt) of glibenclamide, 25, 50, 100 and 200 mg/kg bwt of AECASB, once daily for 14 days, respectively. Fasting blood glucose (FBG) levels of the rats were determined on days 0, 1, 4, 7, 10 and 14. Other biochemical, hematological parameters and pancreas histology of the rats were also determined/examined. The 25 mg/kg bwt of AECASB produced the most significant (p < 0.05) reversal on the alloxan treatment-induced increases in FBG, biochemical and hematological parameters and regenerated the pancreas. In conclusion, the 25 mg/kg bwt of aqueous extract of C. albidum stem was the effective in the management of diabetes and might have acted via regeneration of the pancreas, enhancement of glucose utilization and reduction of blood glucose.

Can Juniperus communis L. oil improve nephropathy in diabetic rats

Objective: Juniperus communis L. is a shrub belonging to family Cupressaceae L. mainly growth in Eurasia. The antioxidant and antidiabetic activity of aqueous extract of J. communis L. berries indicated benefits as a potent antidiabetic in streptozotocin induced diabetic albino rats. This study was carried out to determine whether J. communis L. oil supplement will effectively manage renal dysfunction in diabetic rats.Methods: Twenty eight rats were divided into 4 equal groups as follows; control group, diabetic group, J. communis L. oil (200 mg/kg) treated group, and diabetic+J. communis L. oil (200 mg/kg) treated group. At the end of the experimental period, all rats were sacrificed and renal function parameters such as kidney antioxidant and lipid peroxidation markers and serum creatinine, serum urea, blood urea nitrogen (BUN), and serum total protein levels were measured in all groups. Results: HbA1c, serum glucose, serum urea, serum creatinine, BUN, and kidney lipid peroxidation levels increased but serum total protein and antioxidant levels decreased in diabetic group comparing with control group (p<0.05). Furthermore, HbA1c, serum glucose, serum urea, serum creatinine, BUN, and kidney lipid peroxidation levels decreased and also, serum total protein and antioxidant levels increased in diabetic group treated with J. communis L. oil comparing with diabetic group (p<0.05). Conclusion: This study has provided direct evidence of a link between diabetes mellitus and diabetic nephropathy and demonstrated that J. communis L. oil provide a protective effect on the kidney as evidenced by an improvement of the renal function tests as well as reduction in oxidative stress parameters.

Antihyperglycemic and Antihyperlipidemic Effects of Lippia citriodora in Rats.

The study was conducted to assess the antidiabetic and antihyperlipidemic effects of Lippia citriodora. Lippia citriodora Kunth (Verbenaceae) is a medicinal plant with many traditional uses. The goal of this study was to investigate the antidiabetic activity of aqueous extract of Lippia citriodora and its capacity to improve the lipid profile in diabetic rats. Normal and diabetic rats were treated orally for 15 days by the leaves aqueous extract of Lippia citriodora (LCLAE) (100 mg/kg). The whole glucose utilization and liver architecture were also evaluated. The in vitro antioxidant activity of LCLAE was also assessed. LCLAE lowered blood glucose and lipid levels in all groups. Moreover, LCLAE ameliorated the liver structure and exhibited antioxidant activity. In conclusion, this study demonstrates the hypoglycemic and antihyperlipidemic effects of LCLAE in rats.

Phytochemical Screening and Establishment of the Antidiabetic Potential of Aqueous Leaf Extract of the Endangered Plant Decalepis nervosa in Rats with Alloxan-induced Diabetes.

To evaluate the presence of phytochemicals in and the antidiabetic activity of aqueous extract of Decalepis nervosa (AEDN) leaf. Either sex rats were grouped into 5 classes. Alloxan monohydrate and glibenclamide were used as diabetes induction drug and standard drug, respectively. Aqueous extract of the endangered medicinal plant DN was used in two different doses. Diabetes was induced with alloxan monohydrate at 150 mg/kg b.w. The AEDN was standardized with pharmacognostic and phytochemical screening and a chemical test confirmed the presence of phytoconstituents like glycoside, alkaloid, phenols, and flavonoids. Acute toxicity was evaluated for dose selection in an antidiabetic study. Glibenclamide (5 mg/kg b.w.) and AEDN (200 and 400 mg) were given to all rats with induced diabetes. The reduced blood glucose level may be correlated with the presence of plant secondary metabolites (phenolic compounds), which was identified by thin layer chromatography and confirmed by high performance liquid chromatography studies. The decreased levels of serum total cholesterol, triglyceride, and liver enzyme activity showed the dose dependency of AEDN extract. An oral glucose tolerance test was performed after administration of 200 and 400 mg of AEDN and 5 mg of glibenclamide to different groups, which showed significantly lower oral glucose load during blood sample collection. Animal body weight and dose of AEDN extract had a significant effect on the glucose level in blood (p<0.01). The first report on the phytochemicals and therapeutic activity of AEDN leaf showed potential antidiabetic activity by increased insulin secretion via enhanced peripheral glucose utilization mechanism.

Antidiabetic activity of Musa x paradisiaca extracts in streptozotocin-induced diabetic rats and chemical characterization by HPLC-DAD-MS

Ethnopharmacological relevanceThe Musa x paradisiaca L. inflorescence, known as banana blossom or banana heart, is used in traditional medicine for the treatment of diabetes mellitus.Aim of the study: The aim of the study was to investigate the antidiabetic activity of aqueous extracts and fractions prepared from the bracts and flowers of Musa x paradisiaca in streptozotocin (STZ)-induced diabetic rats and to chemically characterize the extracts. Materials and methodsStandard aqueous extracts of the flowers, bracts, and their fractions were prepared and their chemical composition was determined tentatively by high-performance liquid chromatography coupled to diode-array detection and mass spectrometry (HPLC-DAD-MS). Changes in fasting glycemia and oral glucose tolerance were evaluated in STZ-induced diabetic rats (n = 8) treated with aqueous extracts of Musa x paradisiaca (200 mg/kg) for 20 days. ResultsChemical analyses detected 21 compounds and 17 metabolites were identified, among which were glycosylated and acetylated phenylpropanoids of p-coumaric acid and caffeic acid, as well as a glycosylated flavonol and anthocyanins. Following 15 days of treatment, the bract aqueous extracts and the methanolic fraction of the flower had significant effects on the glycemic profile after glucose load in diabetic rats as compared with the untreated diabetic group. ConclusionsThe results of the present study show the antidiabetic potential of extracts of the flowers and bracts of M. x paradisiaca.

Experimental evidences of antidiabetic activity of aqueous extract of Cressa cretica L. on streptozotocin induced diabetes in rats

With the emerging worldwide interest in adopting and studying traditional systems of medication and exploiting their potential based on different health care systems, the evaluation of the rich heritage of traditional medicine is essential. From the innumerable plants being researched since time immemorial, Cressa cretica is an important one. The present study discusses the antidiabetic activity of the entire plant of Cressa cretica L. belonging to Convolvulaceae family.From the thorough investigation of the available literature, it was evident that the plant drug possessed traditional anti-diabetic activity. The aqueous extract of the whole plant of Cressa cretica was subjected to streptozotocin induced diabetes model. The physico-metabolic parameters measured were body weight, Triglycerides, Cholesterol, LDL, HDL, and histopathological studies of pancreas were also observed. The obtained results indicate the anti-diabetic activity of Cressa cretica whole plant extract (200 mg/kg and 400 mg/kg) were increase in body weight, HDL level and reductions in levels of TG, TCH, LDL respectively

Aqueous extract of Digitaria exilis grains ameliorate diabetes in streptozotocin-induced diabetic male Wistar rats

The absence of scientific data on the age long folkloric use of Digitaria exilis grains by sufferers of diabetes prompted the present investigation. This study was aimed at evaluating the antidiabetic activity of aqueous extract of Digitaria exilis grains in streptozotocin (STZ)-induced diabetic rats. Forty two male rats (166.43 ± 3.32 g) were completely randomized into six groups (A-F) of 7 animals each. Animals in group A (control) were administered 0.5 ml of distilled water while those in groups B, C, D, E and F which were induced with diabetes mellitus (by intraperitoneal administration of 60 mg/kg body weight of STZ) were also administered distilled water, 50 mg/kg body weight of metformin (a reference antidiabetic drug), 50, 100 and 200 mg/kg body weight of aqueous extract of D. exilis grains respectively, twice daily for 14 days. Blood glucose levels and some relevant biomolecules were determined 14 days post-administration. Alkaloids, flavonoids, saponins, tannins, anthraquinones, terpenoids, cardiac glycosides, phlobatannins, phenolics and cardenolides were detected in the extract with alkaloids (30.20 mg/ml) occurring the most and phlobatannins (0.22 mg/ml) the least. Streptozotocin significantly (p < 0.05) increased the levels of blood glucose, serum albumin, urea, creatinine and cholesterol, activities of glucose-6-phosphatase and fructose-1,6-bisphosphatase in the liver and intake of feed and water. Body weight, weight of pancreas, pancreatic insulin, liver glycogen content, red blood cell and white blood cell and their related indices, liver hexokinase and phosphofructokinase activities were significantly reduced by STZ. In contrast, the extract significantly reversed all those STZ-treatment induced changes with the 200 mg/kg body weight of the extract producing profound values that compared favourably with the distilled water treated non-diabetic animals and metformin treated diabetic animals. Overall, this study revealed that Digitaria exilis grains possess antidiabetic activity via increased insulin secretion, as plasma concentrations of insulin were not determined, enhanced activities of hexokinase and phosphofuctokinase and repletion of hepatic glycogen content.

Antidiabetic properties of aqueous and methanol extracts from the trunk bark of Ceiba pentandra in type 2 diabetic rat.

The type 2 diabetes is one of the major global health issues that affects millions of people. This study evaluated the antidiabetic activity of aqueous extracts (AECP) and methanol extracts (MECP) fromCeiba pentandra trunk bark on an experimental model of type 2 diabetes (T2D). This model was induced in rats by the combination of a high-fat diet (HFD) and a single dose of streptozotocin (40 mg/kg, intraperitoneal) at the seventh day of experimentation. Diabetes was confirmed on day 10 by fasting blood glucose more than or equal to 200 mg/dL. Diabetic animals still under HFD were treated orally and twice daily, with MECP and AECP (75 and 150 mg/kg) or metformin (40 mg/kg) for 14 days. During the experiment, blood glucose and animal weights were determined. Oral glucose tolerance test was performed on day 15, followed by animals sacrifice for blood, liver, and pancreas collection. Total cholesterol and triglyceride levels were evaluated in plasma, whereas malondialdehyde (MDA), glutathione (GSH), superoxide dismutase, and catalase were quantified in tissue homogenates. AECP and MECP significantly reduced the hyperglycemia by up to 62% and significantly improved the oral glucose tolerance test. The impaired levels of cholesterol and triglycerides registered in diabetic control were significantly reversed by both extracts at all the doses used. Alterations in diabetic pancreas weight, GSH, and MDA were also significantly reversed by plant extracts. AECP and MECP possess type 2 antidiabetic effects that could result from their ability to improve the peripheral use of glucose, lipid metabolism or from their capacity to reduce oxidative stress. These finding provide a new avenue for better control and management of early or advanced T2D.

Antidiabetic activity of aqueous extract of Sigesbeckia orientalis (St. Paul’s Wort) in alloxan-induced diabetes model

The current study evaluated antidiabetic and antihyperlipidemic activities of aqueous extract of Sigesbeckia orientalis L. (St. Paul’s Wort) (AESO) in an alloxan-induced diabetic rat model. Following OECD guidelines safe doses of AESO were assessed in rats for the main study. Serum/bood glucose, α-amylase, and lipids levels and histopathological evaluations were conducted to assess antidiabetic and associated antihyperlipidemic efficacies of AESO. AESO was found to be safe up to the dose of 2000 mg/kg. Significant (p < 0.05) reduction in glucose and lipids (total cholesterol, triglycerides, low-density lipoproteins) levels was observed in AESO treatment groups. Serum α-amylase, high-density lipoproteins, and total body weight was increased significantly (p < 0.05) in diabetic rats treated with AESO. Histopathological data showed improvement in hepatocyte and pancreatic β-cells islets architecture. HPLC analysis identified quercetin, gallic acid, vanillic acid, p-coumaric acid, m-coumaric acid and cinnamic acid in AESO which are suggested to be responsible for observed antihyperglycemic and antihyperlipidemic attributes. Further studies to standardise the extract and evaluation of safety profile in long-term toxicity studies are recommended for safe and effective antidiabetic nutraceuticals development.

Anti-diabetic activity of aqueous extract of Fructus Ligustri Lucidi in a rat model of type 2 diabetes

Purpose : To determine the anti-diabetic activity of an aqueous extract of Fructus Ligustri Lucidi (FLL) in a rat model of type 2 diabetes (T2D). Method : Body weight (BW), food intake (FI), fasting blood glucose (FBG), glucose tolerance testing and insulin tolerance testing were used to determine the anti-diabetic activity of an aqueous extract of Fructus Ligustri Lucidi (FLL) in a rat model of streptozotocin-induced T2D. Anti-oxidant activity and oxidative stress were assessed by superoxide dismutase (SOD) and malondialdehyde (MDA) analyses, respectively. Results : Following FLL extract treatment, diabetic rat BW increased, while FI and FBG levels decreased. FLL extract increased glucose tolerance and decreased insulin tolerance. Following treatment with 300 and 600 mg/kg FLL extract, MDA levels reached 13.5 ± 0.9 nmol/ml and 13.8 ± 1.1 nmol/ml, respectively. Compared to MDA levels of 19.5 ± 1.1 nmol/ml in diabetic control group, MDA levels was decreased by 30.8 % and 29.0 % after the treatment with 300 and 600 mg/kg FLL extract, respectively, indicating alleviated oxidative stress. Conclusion : The results show that aqueous FLL extract has the potential to alleviate T2D, resulting in reduced FI and FBG, and increased BW. The anti-diabetic activity of FLL extract on T2D may be relevant to reduced oxidative stress burden. In Vitro Antidiabetic and Antioxidant Activities of Aqueous Extract from the Leaf and Fruit of Psidium guajava L.

BACKGROUND: The leaf and fruit of Psidium guajava L. are potential for neutraceutical beverage especially for antidiabetic drink. The aims of this study were to determine the antidiabetic activity of aqueous extract of leaf (LE) and fruit (FE) from P. guajava.METHODS: Both extracts were investigated for their inhibitory effect on α-glucosidase activity in vitro. Their antioxidant activities were measured by 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging, ferrous ion chelating, reducing power and phosphomolybdate methods.RESULTS: The IC50 of LE, FEandacarbose as a positive control were 5.67, 428.00 and 823.99 μg/mL, respectively. The enzyme kinetic analysis indicated that LE inhibited α-glucosidase in a competitive inhibition type, similar to that of acarbose. Both extracts showed antioxidant activities, with LE showed stronger activities than FE in all methods. In DPPH method, IC50 of LE and FE were 74.77 and 843.84 μg/mL respectively, compared to 53.24 and 21.36 μg/mL for reference antioxidants butylated hydroxytoluene (BHT) and ascorbic acid (AA), respectively. In ferrous ion chelating activity, the IC50 were 147.07 and 2105.05 μg/mL for LE and FE, whereas ethylenediaminetetraacetic acid (EDTA) as a control sample was 66.50 μg/mL. In reducing power and phosphomolybdate methods, at different concentrations, the activities of LE, FE, and standard compounds showed the following order: AA &gt; BHT &gt; LE &gt; FE.CONCLUSION: LE from P. guajava exhibited excellent inhibitory activity against α-glucosidase. In addition, LE had better antioxidant acivities than FE.
