Clinical evaluation of AYUSH-SL in patients receiving mass drug administration for the treatment of chronic inflammatory lymphedema, a double blind, placebo controlled, multicentric study.

A 2.5-year placebo controlled double blind trial was conducted to investigate the safety and efficacy of AYUSH- SL, a poly- herbal Ayurvedic formulation on filarial lymphedema in different endemic areas of India. Lymphatic filariasis (LF) is caused by parasitic nematodes from Wuchereria bancrofti, Brugia malayi, or B. timori that are transmitted through mosquitoes. In Ayurveda, LF has been elaborately narrated under the heading Shlipada, and the literature also includes detailed therapeutic measures. The multicenter, prospective, parallel group interventional study of a total of 180 participants were recruited within the duration through competitive enrollment. Diagnosed cases of grade I and grade II filarial lymphedema of Grade I and Grade II (lower extremities) were included in the trial. The study group was treated with MDA and AYUSH-SL or its placebo tablet. The control group had received MDA and a matching placebo. The primary outcome measure was the reduction in edema calculated for analysis by percentage reduction from baseline at the 4th, 12th and 24th week. Secondary outcome measures included improved quality of life (QoL), which was assessed using the Lymphatic Filariasis Specific QoL Questionnaire, and prevention of recurrence of acute episodes, which was evaluated by investigating the presence and severity of episodes in the past 4 weeks. The water displacement method for the evaluation of the efficacy of the intervention was shown to be highly significant compared to the baseline value (right leg; 3071.60 ± 970.482, 2828.40±829.339, p<0.001and Left leg; 3158.69 ± 1136.391, 2890.73 ±1077.475, p<0.001). There was significant improvement of Quality of Life in the LF QoL Questionnaire (p <0.001) at each follow-up visit in both groups. Safety estimations on hematologic and biochemical parameters were within limits and or changes were not significant. The results revealed that AYUSH- SL is safe and effective for FL due to its comprehensive anti-inflammatory, antimicrobial, and anti-allergic activities.

Anti-viral Application of Ganoderma lucidum in COVID-19

Abstract: Ganoderma lucidum (Lingzhi), a common medicinal fungus, has been utilized in China since ancient times for health promotion and longevity. It has anti-inflammatory, antiviral, antitumor, antioxidant, and antiallergic activities. The antioxidant and immunomodulatory effects of this mushroom have shown promising impacts in the treatment of some viral diseases. COVID-19 is a life-threatening disease caused by SARS-CoV-2 that has recently been pandemic. Modern medicines have not been very efficient in the direct treatment of these viral infections, and although vaccination is effective in preventing viral infections, the incidence of acute respiratory infection can be reduced only slightly. Thus, alternative therapies are required, and in this regard, stimulating the immune system can be an effective way to prevent these infections. This study reports the potential of natural fungus-derived compounds that can possibly be used to fight SARS-CoV-2 infections.

Localization, aggregation, and interaction of glycyrrhizic acid with the plasma membrane

Glycyrrhizic acid (GLA) is the most important bioactive constituent of licorize root and exhibits antiviral, antimicrobial, anti-oxidant, anti-inflammatory, anti-allergic, and antitumor activities. GLA has an amphiphilic nature consisting of two hydrophilic and one hydrophobic part, and its mechanism of action could be mediated by its incorporation into the membrane. Furthermore, GLA presents two different forms, protonated (GLA) and deprotonated (GLAD), and has been suggested that their location inside the membrane could be different. Since GLA could be a source against many types of diseases, we have localized the GLA molecule in the presence of a complex membrane and established the detailed interactions of GLA with lipids using all-atom molecular dynamics. Our outcomes sustain that GLA/GLAD tend to locate amid the CHOL oxygen atom and the phospholipid phosphates, preferably perpendicular to the membrane surface, increasing membrane fluidity. Interestingly, GLA and GLAD tend to be surrounded by specific phospholipids, different for each type of molecule. Outstandingly, both GLA and GLAD tend to spontaneously associate in solution forming aggregates, precluding them from inserting into the membrane and, therefore, interacting with it. Consequently, some of the biological properties of GLA/GLAD could be credited to the alteration of the membrane biophysical properties by interacting with specific lipids. However, the formation of an aggregate in solution could hinder its bioactive properties and should be considered a suited vehicle when prepared to be used in biological or clinical assays.

α2,6-linked sialylated oligosaccharides riched in goat milk alleviate food allergy by regulating the gut flora and mucin O-glycosylation

The nutritious goat milk has low allergenicity. Oligosaccharides represent one of the crucial functional constituents in goat milk, which are structurally similar to human milk oligosaccharides (HMOs). Currently, the anti-allergic activity of GMOs has not been reported. In this study, GMOs were efficiently separated into neutral (NGMOs) and sialylated (SGMOs) fractions, following by qualitative and quantitative analysis at the isomer level using online LC-MS/MS. Fifteen NGMOs and 28 SGMOs were detected in goat milk, with 10 SGMOs reported for the first time. Distinctly, α2,6-linked SGMOs were 3.9 times more abundant in goat milk than in bovine milk, with the total relative content of 6′SL, 3′SLN and 6′NGL in SGMOs approach to 60%, which is more similar to HMOs. Orally administering GMOs, especially α2,6-linked sialylated oligosaccharides, significantly alleviated food allergy in ovalbumin-induced BALB/c mice. SGMOs restored the balance of Lachnospiraceae, Erysipelotrichaceae, and Bacteroidaceae, reconstructed the intestinal mucosal barrier, especially restored the levels of fucosylation, sialylation, and sulfation of mucin O-glycans, increased the expression of four core type 2 O-glycans (F1H2N2, F2H2N2, S1F2H2N2, and A1F1H2N2) significantly. This is the first comprehensive study of the anti-allergic activity of GMOs, and the results lay the foundation for the development of GMOs-based natural anti-allergic components.

Research Progress with Atractylone as an Antitumor Agent.

Atractylone is a sesquiterpenoid compound extracted from Rhizoma Atractylodis. As one of the main active components in the volatile oil of the Atractylodes genus, it has exhibited certain therapeutic effects, including anti-inflammatory, antiviral, antioxidant, antiallergic, antiangiogenic, and neuroprotective activities, among others. With further research on the chemical constituents and pharmacology of sesquiterpenes, research on the antitumor activity of Atractylone has also been further expanded. Much of the current literature pays particular attention to the antitumor activity of Atractylone, which was found to inhibit the apoptosis of tumor cells and prevent growth, invasion, and migration through different apoptosis pathways and signaling pathways. Due to its promising potential for cancer prevention, it may play a role in reducing the incidence of malignant tumors. In this paper, the antitumor activity and mechanism of Atractylone are reviewed, providing a reference to inform future research on the tumor treatment, clinical application, and further development and utilization of this plant genus.

Evaluation of Olea europaea L. and Tamarix aphylla leaves extracts in the management of pain, pyrexia and allergy in mice and rats

A huge amount of information, related to herbals is based exclusively on historical beliefs. Therefore, we aim to provide scientific validation for some of the traditional uses attributed to Olea europaea (OE) and Tamarix aphylla (TA). To achieve this, we conducted experiments to ensure the safety of the aqueous extract of OE and the methanolic extract of TA. Subsequently, we investigated the peripheral and central antinociceptive activity, antipyretic properties, and anti-allergic activities using albino mice and Wistar rats. Both plant extracts at the doses of 100, 200 and 400 mg produced potent antinociceptive activity (p < 0.05) in a dose-dependent manner using acetic acid-induced writhing reflex and tail immersion assay. Additionally, both plants produced remarkable antipyretic activity comparable with paracetamol (100 mg) manifested by a significant (p < 0.05) reduction of brewer’s yeast-induced pyrexia in rat. Both OE and TA resulted in a potent reduction in leukocytosis and eosinophilia induced by milk in mice.

Targeting Diabetes with Azole-derived Medicinal Agents.

Azoles have long been regarded as an ideal scaffold for the development of numerous innovative therapeutic agents as well as other incredibly adaptable and beneficial chemicals with prospective uses in a variety of fields, including materials, energetics (explosophores), and catalysis (azole organocatalytic arbitration). Azoles exhibit promising pharmacological activities, including antimicrobial, antidiabetic, antiviral, antidepressant, antihistaminic, antitumor, antioxidant, antiallergic, antihelmintic, and antihypertensive activity. According to a database analysis of U.S. FDAapproved medications, 59% of specific medications are connected to small molecules that have heterocycles having nitrogen atoms. The azole moiety has impressive electron abundance. Azoles promptly attach to various receptors as well as enzymes in the physiological environment via distinct specialized interactions, contributing to their anti-diabetic potential. This review encompasses the recent research progress on potent azole-derived antidiabetic agents that can be used as an alternative for the management of type-2 diabetes.

The antisickling agent, 5-hydroxymethyl-2-furfural: Other potential pharmacological applications.

For the last two decades, the aromatic aldehyde 5-hydroxymethyl-furfural (5-HMF) has been the subject of several investigations for its pharmacologic potential. In 2004, the Safo group reported that 5-HMF has potent antisickling activity by targeting and ameliorating the primary pathophysiology of hypoxia-induced sickling of erythrocytes (red blood cells [RBC]). Following the encouraging outcome of the preclinical and phase I/II clinical studies of 5-HMF for the treatment of sickle cell disease (SCD), there have been multiple studies suggesting 5-HMF has several other biological or pharmacologic activities, including anti-allergic, antioxidant, anti-hypoxic, anti-ischemic, cognitive improvement, anti-tyrosinase, anti-proliferation, cytoprotective, and anti-inflammatory activities. The wide range of its effects makes 5-HMF a potential candidate for treating a variety of diseases including cognitive disorders, gout, allergic disorders, anemia, hypoxia, cancers, ischemia, hemorrhagic shock, liver fibrosis, and oxidative injury. Several of these therapeutic claims are currently under investigation and, while promising, vary in terms of the strength of their evidence. This review presents the research regarding the therapeutic potential of 5-HMF in addition to its sources, physicochemical properties, safety, absorption, distribution, metabolism, and excretion (ADME) profiles.

Antioxidant potential and fatty acid composition of Lactarius sanguifluus

Edible mushrooms have been valued as a source of food and traditional medicines since ancient times. Mushrooms have drawn the attention of numerous researchers in recent decades since they contain a variety of bioactive compounds with antibacterial, antiallergic, antidiabetic, antitumoral, and antioxidant activities. As a member of the Russulaceae family Lactarius sanguifluus (Paulet) Fr. (1838) (also known as bloody milk cap) is an ectomycorrhizal edible mushroom distributed in Asia, the Mediterranean shores and mid-Europe. In this study it was aimed to identify the mushroom sample both by physical traits according to the current literature and by molecular techniques. The antioxidant capability of L. sanguifluus methanol extract was evaluated by stable radical scavenging activity and metal ion reducing capability methods. The content of total phenolic compounds and the fatty acid composition of the mushroom sample were also investigated. The mushroom sample was identified both by comparing the physical traits with the current literature and by comparing the sequence data of the ITS region of rDNA with GenBank. The methanol extract of the dried mushroom sample was used to perform the analysis to determine the antioxidant activity and content of total phenolic compounds. It was found that L. sanguifluus methanol extract contained 3.677±0.207 mg GAE/g DW phenolic compounds and exhibited radical scavenging activities as 211.652±0.537 mg TE/g DW against DPPH and 73.389±1.003 mg TE/g DW against ABTS. L. sanguifluus methanol extract also had metal ion reducing capabilities with ferric reducing and cupric reducing antioxidant capacities of 191.903±0.659 mg AAE/g DW and 379.598±7.527 mg TE/g DW, respectively. The GC-MS analysis of the hexane extract of the mushroom sample revealed that palmitic acid, linoleic acid, oleic acid and stearic acid were the predominant fatty acids found in L. sanguifluus.

Persulfated Ascorbic Acid Glycoside as a Safe and Stable Derivative of Ascorbic Acid for Skin Care Application.

The pursuit of cosmetic ingredients with proven efficacy and safety that meet consumer needs drives the advancement of new products. Ascorbic acid (AA) is utilized in cosmetic products, predominantly for its potent antioxidant properties. Nonetheless, its instability compromises its efficacy. In this work, ascorbyl 2-O-glucoside persulfate (AAGS) was synthesized, characterized, and evaluated regarding its safety profile and potential bioactivities and the results were compared to AA and its glycoside AAG. Pre-formulation studies were performed to assess the stability of the compounds and their compatibility with typical excipients commonly used in topical formulations. AAGS did not affect the metabolic activity of keratinocyte, macrophage, and monocyte cell lines, up to 500 µM. AAGS also exhibited a non-prooxidant and non-sensitizing profile and anti-allergic activity by impeding the allergen-induced maturation of THP-1 cells. When compared to AA and AAG, AAGS was shown to be more stable at pH values between 5 and 7, as well as superior thermostability and photostability. AAGS demonstrated higher stability in metal solutions of Fe(II) and Mg(II) than AA. AAGS demonstrated similar DPPH radical scavenging activity compared to AA. These results provide useful information for the development of new AA derivatives, highlighting AAGS as a novel cosmetic ingredient.

A critical appraisal of geroprotective activities of flavonoids in terms of their bio-accessibility and polypharmacology

Flavonoids, a commonly consumed natural product, elicit health-benefits such as antioxidant, anti-inflammatory, antiviral, anti-allergic, hepatoprotective, anti-carcinogenic and neuroprotective activities. Several studies have reported the beneficial role of flavonoids in improving memory, learning, and cognition in clinical settings. Their mechanism of action is mediated through the modulation of multiple signalling cascades. This polypharmacology makes them an attractive natural scaffold for designing and developing new effective therapeutics for complex neurological disorders like Alzheimer's disease and Parkinson's disease. Flavonoids are shown to inhibit crucial targets related to neurodegenerative disorders (NDDs), including acetylcholinesterase, butyrylcholinesterase, β-secretase, γ-secretase, α-synuclein, Aβ protein aggregation and neurofibrillary tangles formation. Conserved neuro-signalling pathways related to neurotransmitter biogenesis and inactivation, ease of genetic manipulation and tractability, cost-effectiveness, and their short lifespan make Caenorhabditis elegans one of the most frequently used models in neuroscience research and high-throughput drug screening for neurodegenerative disorders. Here, we critically appraise the neuroprotective activities of different flavonoids based on clinical trials and epidemiological data. This review provides critical insights into the absorption, metabolism, and tissue distribution of various classes of flavonoids, as well as detailed mechanisms of the observed neuroprotective activities at the molecular level, to rationalize the clinical data. We further extend the review to critically evaluate the scope of flavonoids in the disease management of neurodegenerative disorders and review the suitability of C. elegans as a model organism to study the neuroprotective efficacy of flavonoids and natural products.

Potential Antiallergic Activity of Two Chemically/Enzymatically-Modified Natural Products Against Active Atopic and Systemic Anaphylaxes in CD1 Mice Models

ABSTRACT Introduction Anaphylaxis is a globally increasing allergic reaction that is often fatal. Recently, our previous study reported the possibility of using the modified natural products “sodium R-lipoate (NaRLA) and enzymatically modified isoquercitrin (EMIQ)“ as potential novel safe agents against the non-immunological-degranulation of mast cells. Methods Here, we extended our previous findings by determining the antianaphylactic activity of 50 and 100 mg/kg body weight of NaRLA and EMIQ (given orally and prior to local or systemic challenge) in mice models of ovalbumin (OVA)-induced IgE-dependent active cutaneous anaphylaxis (ACA) and active systemic anaphylaxis (ASA) in comparison with sulfasalazine (SSZ, amast cell stabilizer). Results The pre-treatment of mice with NaRLA or EMIQ completely succeeded, as SSZ, in suppression of the increased vascular permeability associated with IgE-dependent ACA and protected the OVA-sensitized mice from fatal ASA by reducing (p < .001) the skin mast cell degranulation, the elevated peritoneal histamine and interleukin-4 levels, along with decreasing the associated sever gastrointestinal and lung histopathological alterations and inflammation. The high dose of EMIQ prevented death in 70% of mice with anaphylactic shock, better than SSZ. Discussion Our data indicated that NaRLA and EMIQ may be potential prophylactic and therapeutic candidates for the alleviation of atopic and systemic anaphylaxis.

Solid-State Fermentation of Grain-Derived By-Products by Aspergillus kawachii and Rhizopus oryzae: Preparation and Evaluation of Anti-Allergic Activity

Grain processing produces many by-products, including wheat bran, wheat germ and rice bran, which are rich in carbohydrates, proteins and trace elements. In this study, these grain-derived by-products were used as raw materials to conduct solid-state fermentation using mixed strains of Aspergillus kawachii and Rhizopus oryzae, and the potential immunomodulatory and anti-allergic properties of fermented product were evaluated. Solid-state fermentation of a grain by-product mixture, consisting of rice bran, wheat bran, and wheat germ in a 2:1:1 weight ratio, using both A. kawachii L1 and R. oryzae L1 at 26 °C for 5 days, significantly increased the total phenolic, flavonoid, and amino acid contents. The anti-allergic activity of aqueous extract of the fermented product was evaluated in murine models of food allergy and delayed-type hypersensitivity. Oral administration of the fermented product extract (100–200 mg/kg) notably alleviated allergic symptoms such as diarrhea and histopathological changes in the intestines. Moreover, the extract effectively reduced allergen-specific serum antibodies, suppressed splenic cytokine secretion, and mitigated tissue edema and inflammation induced by allergens. Importantly, the extract induced the production of IL-10 and TGF-β, which are well-known cytokines primarily secreted by regulatory T cells. These results underscore the promising immunomodulatory effects of A. kawachii and R. oryzae fermented grain product, suggesting their potential as functional foods or additives for managing allergic disorders, with implications for future therapeutic and dietary applications.

Trigonelline prevents high-glucose-induced endothelial-to-mesenchymal transition, oxidative stress, mitochondrial dysfunction, and impaired angiogenic activity in human endothelial EA.hy926 cells

Trigonelline (TRIG) is a natural compound in an alkaloid family found in diverse plants. This compound exerts anti-inflammatory, anti-allergic, anti-oxidative and anti-fibrotic activities in several disease models. However, its beneficial role in endothelial injury, especially induced by diabetes, is unclear. We, therefore, evaluated the effects of TRIG on the cellular proteome of human endothelial (EA.hy926) cells followed by functional validation in high-glucose (HG)-induced endothelial deteriorations. Label-free quantification using nanoLC-ESI-Qq-TOF MS/MS revealed 40 downregulated and 29 upregulated proteins induced by TRIG. Functional enrichment analysis using DAVID and REVIGO tools suggested the involvement of these altered proteins in several biological processes and molecular functions, particularly cell-cell adhesion, ATP metabolic process, cell redox homeostasis, cadherin binding, and ATP hydrolysis activity. Experimental validation showed that HG triggered endothelial-to-mesenchymal transition (EndMT) (as demonstrated by increased spindle index and mesenchymal markers, i.e., fibronectin and vimentin, and decreased endothelial markers, i.e., PECAM-1 and VE-cadherin), increased oxidized proteins, and reduced intracellular ATP, active mitochondria, endothelial tube/mesh formation and VEGF secretion. However, TRIG successfully abolished all these defects induced by HG. These data indicate that TRIG prevents HG-induced EndMT, oxidative stress, mitochondrial dysfunction, and impaired angiogenic activity in human endothelial cells.

Phosphoryl oligosaccharide alleviates food allergy by suppressing IgE class switch recombination through LCN2 pathway

Specific algal polysaccharides exhibit significant anti-allergic activity. B cell class switch recombination (CSR) is a crucial step in food allergies, however, the role of polysaccharides in regulating CSR remains unclear. To investigate the regulation of B cell CSR by algal polysaccharides, an in vitro IgE-type CSR cell model was used to screen for active algal polysaccharides, and four active polysaccharides were identified. In a food allergy mouse model, Undaria pinnatifida polysaccharides (UPP) and phosphoryl oligosaccharides (POS) suppressed allergic responses and reduced the proliferation of IgE+ B cells. Transcriptome sequencing of B cells identified LCN2 as a potential target of POS, and inhibition of LCN2 blocked the activity of POS on CSR. These findings demonstrated the efficacy of POS in regulating B cell CSR and alleviating allergic responses and revealed the underlying mechanisms. This work provided data and theoretical support for the development of polysaccharide-based functional foods for individuals with allergies.

Investigation on the mechanism of hepatotoxicity of dictamnine on juvenile zebrafish by integrating metabolomics and transcriptomics

Dictamnine(DIC), as the key pharmacological component of the classical Chinese herbal medicine cortex dictamni, possesses multiple pharmacological activities such as anti-microbial, anti-allergic, anti-cancer, and anti-inflammatory activities, however it is also the main toxicant of cortex dictamni induced hepatic damage, yet the underlying molecular mechanisms causing hepatic damage are still largely unknown. With the purpose of explore possibilities hepatotoxicity of dictamnine in zebrafish and to identify the key regulators and metabolites involved in the biological process, we administered zebrafish to dictamnine at a sub-lethal dose (<LC10) for 24 h and performed biochemical index tests, pathological observations, metabolomics, and transcriptomics analyses. The results showed that the liver function indexes such as ALT and AST were affected after the exposure treatment with dictamnine, and the hepatic damage, lipid droplet formation, and increased apoptosis were observed in zebrafish by HE, oil red O(ORO), and Acridine orange hydrochloride(AO)staining. Transcriptome sequencing analysis showed that dictamnine exposure could generate 5696 down-regulated and 4936 up-regulated DEGs(Differential Expressed Genes); metabolomics analysis showed that 36 potential biomarkers were disturbed by dictamnine exposure treatment in juvenile zebrafish. Integration of metabolomics data and transcriptomics data showed that Patatin like phospholipase domain containing 3 Gene Patatin Like Phospholipase Domain Containing 3(PNPLA3), Lactase Gene(LCT), and Galactosidase Beta 1(GLB1) genes were involved in the regulation of 12 key potential biomarkers related to Galactose metabolism and Glycerophospholipid metabolism, such as LysoPC(16:0/0:0) and UDP-4-dehydro-6-deoxy-D-glucose, which in turn regulated pathways of Galactose metabolism and Glycerophospholipid metabolism and consequently induced hepatotoxicity. The comprehensive evaluation of the hepatotoxicity induced by dictamnine was realized from multiple levels, perspectives and indexes by the integrated evaluation method of zebrafish modeling, which supported the applicability of zebrafish in the evaluation of hepatotoxicity of traditional Chinese medicine, and supplied the scientific basis for elucidating the molecular mechanism of the hepatotoxicity induced by dictamnine, as well as guided the development of the toxicity-reducing therapies by dictamnine in the future.

Advanced Nanotechnological Approaches for Effective Delivery of Rutin: An Updated Review.

Polyphenols are a group of naturally occurring compounds that have intriguing biological activities. Among these compounds is rutin, a polyphenolic flavanol found in many plants, including passion flowers, buckwheat seed, fruits and fruit rinds, and citrus fruits (such as orange, grapefruit, lemon, and lime). Various studies have demonstrated rutin to possess antibacterial, antifungal, antiallergic, anti-inflammatory, anti-diabetic, anti-adipogenic, anti-carcinogenic, anti-apoptotic, anti-osteoporotic, radioprotective, gastroprotective, neuroprotective, and nephroprotective activities. Despite its benefits, rutin's therapeutic applicability is severely limited due to its low water solubility, sensitivity to oxidation, and dissolving rate. However, these problems can be overcome by employing an efficient delivery approach. An extensive number of nanocarriers can be developed for medicinal use if pre-clinical as well as human-clinical studies are completed. The current review presents an overview of effective rutin nano-formulations for targeted therapy in various health disorders. This review article discusses the clinical evidence, current status, as well as future opportunities of rutin nanocarriers for increasing rutin's bioactivity for possible medicinal uses.

Synthesis and Antiallergic Activity of Dicoumarin Derivatives.

Allergies are one of the diseases whose incidence rates have increased in recent years due to the greenhouse effect and extreme climate change. Therefore, the development of new antiallergic drugs has attracted the interest of researchers in chemistry and pharmacy fields. Dicoumarin is a coumarin derivative with various biological activities, but its antiallergic activity has not been evaluated. In this study, 14 different dicoumarin derivatives were synthesized by diethylamine-catalyzed condensation reactions of 4-hydroxycoumarin with 14 different aldehydes, and they were identified on the basis of their spectral data. The dicoumarin derivatives were subjected to studies on the degranulation of rat basophilic leukemia cells (RBL-2H3 cells) and mouse bone-marrow-derived mast cells (mBMMCs), and some of them showed good inhibitory effects on the degranulation of the two types of mast cells, demonstrating their good antiallergic activity. This study presents a new method of developing new antiallergic drugs.

Marine Bioactive Compounds with Functional Role in Immunity and Food Allergy.

Food allergy, referred to as the atypical physiological overreaction of the immune system after exposure to specific food components, is considered one of the major concerns in food safety. The prevalence of this emerging worldwide problem has been increasing during the last decades, especially in industrialized countries, being estimated to affect 6-8% of young children and about 2-4% of adults. Marine organisms are an important source of bioactive substances with the potential to functionally improve the immune system, reduce food allergy sensitization and development, and even have an anti-allergic action in food allergy. The present investigation aims to be a comprehensive report of marine bioactive compounds with verified actions to improve food allergy and identified mechanisms of actions rather than be an exhaustive compilation of all investigations searching beneficial effects of marine compounds in FA. Particularly, this research highlights the capacity of bioactive components extracted from marine microbial, animal, algae, and microalgae sources, such as n-3 long-chain polyunsaturated fatty acids (LC-PUFA), polysaccharide, oligosaccharide, chondroitin, vitamin D, peptides, pigments, and polyphenols, to regulate the immune system, epigenetic regulation, inflammation, and gut dysbiosis that are essential factors in the sensitization and effector phases of food allergy. In conclusion, the marine ecosystem is an excellent source to provide foods with the capacity to improve the hypersensitivity induced against specific food allergens and also bioactive compounds with a potential pharmacological aptitude to be applied as anti-allergenic in food allergy.
