Short-term impacts of polyethylene and polyacrylonitrile microplastics on soil physicochemical properties and microbial activity of a marine terrace environment in maritime Antarctica

Evidence of microplastic (MP) pollution in Antarctic terrestrial environments reinforces concerns about its potential impacts on soil, which plays a major role in ecological processes at ice-free areas. We investigated the effects of two common MP types on soil physicochemical properties and microbial responses of a marine terrace from Fildes Peninsula (King George Island, Antarctica). Soils were treated with polyethylene (PE) fragments and polyacrylonitrile (PAN) fibers at environmentally relevant doses (from 0.001% to 1% w w−1), in addition to a control treatment (0% w w−1), for 22 days in a pot incubation experiment under natural field conditions. The short-term impacts of MPs on soil physical, chemical and microbial attributes seem interrelated and were affected by both MP dose and type. The highest PAN fiber dose (0.1%) increased macro and total porosity, but decreased soil bulk density compared to control, whereas PE fragments treatments did not affect soil porosity. Soil respiration increased with increasing doses of PAN fibers reflecting impacts on physical properties. Both types of MPs increased microbial activity (fluorescein diacetate hydrolysis), decreased the cation exchange capacity but, especially PE fragments, increased Na+ saturation. The highest dose of PAN fibers and PE fragments increased total nitrogen and total organic carbon, respectively, and both decreased the soil pH. We discussed potential causes for our findings in this initial assessment and addressed the need for further research considering the complexity of environmental factors to better understand the cumulative impacts of MP pollution in Antarctic soil environments.

Microbial community composition of terrestrial habitats in East Antarctica with a focus on microphototrophs.

The Antarctic terrestrial environment harbors a diverse community of microorganisms, which have adapted to the extreme conditions. The aim of this study was to describe the composition of microbial communities in a diverse range of terrestrial environments (various biocrusts and soils, sands from ephemeral wetlands, biofilms, endolithic and hypolithic communities) in East Antarctica using both molecular and morphological approaches. Amplicon sequencing of the 16S rRNA gene revealed the dominance of Chloroflexi, Cyanobacteria and Firmicutes, while sequencing of the 18S rRNA gene showed the prevalence of Alveolata, Chloroplastida, Metazoa, and Rhizaria. This study also provided a comprehensive assessment of the microphototrophic community revealing a diversity of cyanobacteria and eukaryotic microalgae in various Antarctic terrestrial samples. Filamentous cyanobacteria belonging to the orders Oscillatoriales and Pseudanabaenales dominated prokaryotic community, while members of Trebouxiophyceae were the most abundant representatives of eukaryotes. In addition, the co-occurrence analysis showed a prevalence of positive correlations with bacterial taxa frequently co-occurring together.

Integrative description of a new Dactylobiotus (Eutardigrada: Parachela) from Antarctica that reveals an intraspecific variation in tardigrade egg morphology

Tardigrades constitute one of the most important group in the challenging Antarctic terrestrial ecosystem. Living in various habitats, tardigrades play major roles as consumers and decomposers in the trophic networks of Antarctic terrestrial and freshwater environments; yet we still know little about their biodiversity. The Eutardigrada is a species rich class, for which the eggshell morphology is one of the key morphological characters. Tardigrade egg morphology shows a diverse appearance, and it is known that, despite rare, intraspecific variation is caused by seasonality, epigenetics, and external environmental conditions. Here we report Dactylobiotus ovimutans sp. nov. from King George Island, Antarctica. Interestingly, we observed a range of eggshell morphologies from the new species, although the population was cultured under controlled laboratory condition. Thus, seasonality, environmental conditions, and food source are eliminated, leaving an epigenetic factor as a main cause for variability in this case.

Eradication of the non-native Poa pratensis colony at Cierva Point, Antarctica: A case study of international cooperation and practical management in an area under multi-party governance

The on-going introduction of non-native species to Antarctica due to expanding human activity presents an increasing threat to biodiversity. Under the Protocol on Environmental Protection to the Antarctic Treaty, all introduced non-native species should be removed from the Antarctic Treaty area. The non-native grass Poa pratensis was first introduced to Cierva Point (Danco Coast, Antarctic Peninsula), along with substantial quantities of non-Antarctic soil, in the mid-1950s. Consistent with the Protocol, in January 2015 an internationally coordinated team undertook the eradication of the grass. Immediately prior to removal of P. pratensis, factors affecting its establishment, persistence and impacts upon local indigenous species was examined within the international management framework of the Antarctic Treaty System. The underlying soil had a high organic content of 15.5%, which may have contributed to the successful establishment of P. pratensis and restricted, at least initially, its vegetative growth to the enriched area. Examination of P. pratensis expansion from the original introduction sites showed that the plant colony intricate root system facilitated little or no coexistence of other native plants within its extent. The non-native plant colony also constituted a novel habitat for soil fauna within Antarctic terrestrial environments. The P. pratensis plant colony provided an unfavorable habitat for two of the locally endemic soil invertebrates, Cryptopygus antarcticus and Belgica antarctica. These observations led to the selection of an appropriate eradication approach, where the plants were targeted for physical extraction along with all underlying soil. During the eradication, c. 500kg of soil and plant material from the P. pratensis colony was removed from the site. Monitoring one year later showed no evidence of re-establishment. Consistent with the Committee for Environmental Protection ‘Non-native Species Manual’, we recommend development and implementation of rapid response protocols following the discovery of a non-native plant colony to limit future impacts on indigenous species and local habitats.

Metabolic response to desiccation stress in strains of green algal photobionts (Trebouxia) from two Antarctic lichens of southern habitats

:Desiccation tolerance is a feature of most lichens. These symbiotic associations of a fungal partner (mycobiont) and a photosynthetic partner (photobiont) experience severe desiccation on a regular basis. Many lichen species colonise extreme habitats, such as the cold deserts of Antarctica. Although the stress physiology of lichens has been studied extensively, little is known about the photobionts' physiological potential. To assess the contribution of photobionts to lichen success in Antarctic terrestrial environments, the photosynthetic partners of Umbilicaria decussata (Umbilicariaceae, lichenised ascomycetes) and Usnea lambii (Parmeliaceae, lichenised ascomycetes) from extreme habitats in southern Antarctica were isolated. Significant differences regarding stress tolerance of the two green algae had been shown previously despite their close phylogenetic relationship. A deeper understanding and explanation of these differences were obtained by examination of the metabolite profiles and dynamics, especially the role of sugars, polyols and amino acids. High concentrations of ribitol and sucrose, among other metabolites, characterised the photobiont of Usnea lambii's mainly constitutive strategy of desiccation tolerance. A high degree of specialisation to dry environments can be assumed for this photobiont.

Penguin activity modify the thermal regime of active layer in Antarctica: A case study from Hope Bay

Monitoring permafrost and active layer is essential for prognostic research of future climate scenarios. Although biotic factors have a prominent role in active layer behavior, apart from vegetation effects, their influence remains little investigated. It is generally assumed that frozen ground exerts influence on nesting fauna, whereas, on the other hand, land colonization by birds, such as penguins can also interfere with the soil thermal regime. However, to our knowledge, no studies report on permafrost and active layer regime changes under penguin activity. We present a comparative study of two adjacent sites located in Hope Bay, one representing an active penguin rookery-S1, and another, an adjacent abandoned site currently vegetated-S2. Soil temperature and water content, and air temperature were monitored hourly from 2009 to 2011. Current penguin activity and the resulting deposition of guano during spring and summer in S1 is an important factor for explaining the higher number of thaw degree days due to direct physical impact and chemical reactions caused by rapid guano decomposition. In the vegetated S2 site, the snow pack lasted longer, showing the highest mean minimum temperature and larger thermal insulation, as well as larger FDD than those found in the bare soil of the active rookery (S1), and lower n-F, due to greater thermal insulation. Penguins played a significant role in changing the active layer depth and thermal regime, and represent a neglected actor on the ground thermal regime in Antarctic terrestrial environments.

Geospatial variability of soil CO2–C exchange in the main terrestrial ecosystems of Keller Peninsula, Maritime Antarctica

Soils and vegetation play an important role in the carbon exchange in Maritime Antarctica but little is known on the spatial variability of carbon processes in Antarctic terrestrial environments. The objective of the current study was to investigate (i) the soil development and (ii) spatial variability of ecosystem respiration (ER), net ecosystem CO2 exchange (NEE), gross primary production (GPP), soil temperature (ST) and soil moisture (SM) under four distinct vegetation types and a bare soil in Keller Peninsula, King George Island, Maritime Antarctica, as follows: site 1: moss-turf community; site 2: moss-carpet community; site 3: phanerogamic antarctic community; site 4: moss-carpet community (predominantly colonized by Sanionia uncinata); site 5: bare soil. Soils were sampled at different layers. A regular 40-point (5×8 m) grid, with a minimum separation distance of 1m, was installed at each site to quantify the spatial variability of carbon exchange, soil moisture and temperature. Vegetation characteristics showed closer relation with soil development across the studied sites. ER reached 2.26μmolCO2m−2s−1 in site 3, where ST was higher (7.53°C). A greater sink effect was revealed in site 4 (net uptake of 1.54μmolCO2m−2s−1) associated with higher SM (0.32m3m−3). Spherical models were fitted to describe all experimental semivariograms. Results indicate that ST and SM are directly related to the spatial variability of CO2 exchange. Heterogeneous vegetation patches showed smaller range values. Overall, poorly drained terrestrial ecosystems act as CO2 sink. Conversely, where ER is more pronounced, they are associated with intense soil carbon mineralization. The formations of new ice-free areas, depending on the local soil drainage condition, have an important effect on CO2 exchange. With increasing ice/snow melting, and resulting widespread waterlogging, increasing CO2 sink in terrestrial ecosystems is expected for Maritime Antarctica.

Antarctic eukaryotic soil diversity of the Prince Charles Mountains revealed by high-throughput sequencing

Studies of Antarctic eukaryotes have been hampered by their morphological conservatism, small size and the logistical constraints of remote field work, resulting in a deficiency of baseline biodiversity information about Antarctic terrestrial environments. The application of high throughput sequencing (HTS) in metataxonomic approaches is a promising alternative. Here, we apply such HTS approaches to the hitherto largely unsurveyed micro-eukaryote fauna of the Prince Charles Mountains, East Antarctica. We sequenced 18S rDNA amplicons of twelve Antarctic bulk-soil DNA extracts, retrieved from three sampling regions (four bulk-soil extracts per sampling region). After isolating eukaryotic phylotypes with a stringent filtering approach and initial network visualization, we firstly used rarefied data to compare four α diversity metrics between the three regions. Weighted and unweighted inter-sample UniFrac distances were then used for β diversity comparisons among rarefied data. Furthermore, we analysed the distribution of the most abundant phylotypes and phylotype groups. Lastly, we checked the validity of species-level taxonomic assignments using different sets of reference data in conjunction with two different taxonomy assignment approaches. Phylotype numbers in un-rarefied data compared across regions were lowest for Mount Menzies (73°S; 3330 m), intermediate at Mawson Escarpment (73°S; 807 m) and highest at Lake Terrasovoje (70°S; 173 m), likely due to low biological load at the higher latitude and elevation inland sites. Analysis of rarefied data was difficult due to low sequence coverage particularly from Mount Menzies, but indicated differences in Shannon diversity between Mawson Escarpment and Lake Terrasovoje. PCoA of weighted UniFrac distances between samples from Mawson Escarpment and Lake Terrasovoje indicated changes in community composition in relation to elevation of the sampling locations. The most widespread phylotypes were fungal, followed by non-algal protists. Species-level assignments included known Antarctic taxa in all sampling regions. We show that HTS can provide a rapid survey of the micro-eukaryote fauna to provide baseline biodiversity information for remote, harsh, and hitherto largely unsurveyed environments in the Prince Charles Mountains.

Demonstration of “substantial research activity” to acquire consultative status under the Antarctic Treaty

Antarctic Treaty Consultative Parties are entitled to participate in consensus-based governance of the continent through the annual Antarctic Treaty Consultative Meetings. To acquire consultative status, an interested Party must demonstrate “substantial research activity,” but no agreed mechanism exists to determine whether a Party has fulfilled this criterion. Parties have generally demonstrated substantial research activity with the construction of a research station, as suggested within the Treaty itself. However, this largely demonstrates logistical capacity, rather than research activity, and often results in major and persistent impacts on Antarctic terrestrial environments. Our study found that national investment in Antarctic infrastructure, estimated by the number of bed spaces at stations, was not a reliable indicator of scientific output. Therefore, we investigated metrics to evaluate research activity directly, and identified both the overall number of Antarctic papers and the proportion of national scientific output these represented as meaningful metrics. Such metrics could (1) demonstrate a nation’s level of research activity in Antarctica or (2) help Consultative Parties assess the level of research activity undertaken by a Party seeking to acquire consultative status. Our data showed that, even without land-based Antarctic infrastructure, Canada, Denmark and Switzerland may have reasonable grounds to demonstrate “substantial research activity” on a level comparable with existing Consultative Parties. The use of these metrics may help dispel any perceived requirement for the establishment of a research station to reach consultative status, by putting a greater emphasis on generation of scientific research outputs rather than construction of Antarctic infrastructure. A method for assessing the physical recovery of Antarctic desert pavements following human-induced disturbances: A case study in the Ross Sea region of Antarctica

With increasing visitor numbers an understanding of the impacts of human activities in Antarctic terrestrial environments has become important. The objective of this study was to develop a means for assessing recovery of the ground surface desert pavement following physical disturbance. A set of 11 criteria were identified to assess desert pavement recovery. Assessed criteria were: embeddedness of surface clasts; impressions of removed clasts; degree of clast surface weathering; % overturned clasts; salt on underside of clasts; development of salt coatings; armouring per m2; colour contrast; evidence of subsidence/melt out; accumulation of salt on cut surfaces; and evidence of patterned ground development. Recovery criteria were assigned a severity/extent rating on a scale from zero to four, zero being highly disturbed, and four being undisturbed. A relative % recovery for each criteria was calculated for each site by comparison with a nearby undisturbed control area, and an overall Mean Recovery Index (MRI) was assigned to each pavement surface.To test the method, 54 sites in the Ross Sea region of Antarctica were investigated including areas disturbed by: bulldozer scraping for road-fill, contouring for infrastructure, geotechnical investigations, and experimental treading trial sites. Disturbances had occurred at timescales ranging from one week to 50 years prior to assessment. The extent of desert pavement recovery at the sites investigated in this study was higher than anticipated. Fifty of the 54 sites investigated were in an intermediate, or higher, stage of desert pavement recovery, 30 sites were in an advanced stage of recovery, and four sites were indistinguishable from adjacent control sites (MRI = 100%). It was found that active surfaces, such as the gravel beach deposits at the Greenpeace World Park Base site at Cape Evans, the aeolian sand deposits at Bull Pass, and the alluvial fan deposits of the Loop Moraine field campsite, recovered relatively quickly, whereas less active sites, such as the bulldozed tracks at Marble Point, and Williams Field to McMurdo Station pipeline site on Ross Island, showed only intermediate recovery 20–30 years after disturbance. The slabby grano-diorite surface material at the former Vanda Station site, meant that the impacts that had occurred were hard to detect following decommissioning of the station and site remediation. Desert pavements disturbed by randomly dispersed footprints, temporary field campsites at the Loop Moraine and VXE6 Pond in the Wright Valley, recovered to be undetectable (MRI = 100%) within five years, whereas track formation from repeated trampling, particularly the concentration of larger clasts along the margin of a confined track, persisted for over 15 years (MRI = 82%). The recovery assessment method developed in this study has environmental management applications and potential to advance our ability to predict the recovery of desert pavement following human impacts from activities in Antarctica.

Environmental monitoring and management proposals for the Fildes Region, King George Island, Antarctica

The Antarctic terrestrial environment is under increasing pressure from human activities. The Fildes Region is characterized by high biodiversity, but is also a major logistic centre for the northern Antarctic Peninsula. Different interests, from scientific research, nature conservation, protection of geological and historical values, station operations, transport logistics and tourism, regularly overlap in space and time. This has led to increasing conflict among the multiple uses of the region and breaches of the legal requirements for environmental protection that apply in the area. The aim of this study was to assess the impacts of human activities in the Fildes Region by monitoring the distribution of bird and seal breeding sites and recording human activities and their associated environmental impacts. Data from an initial monitoring period 200306 were compared with data from 200810. We observed similar or increased levels of air, land and ship traffic, but fewer violations of overflight limits near Antarctic Specially Protected Area No. 150 Ardley Island. Open waste dumping and oil contamination are still major environmental impacts. Scientific and outdoor leisure activities undertaken by station personnel are more frequent than tourist activities and are likely to have a commensurate level of environmental impact. Despite the initial success of some existing management measures, it is essential that scientific and environmental values continue to be safeguarded, otherwise environmental impacts will increase and the habitat will be further degraded. Metagenomic Analysis of a Southern Maritime Antarctic Soil

Our current understanding of Antarctic soils is derived from direct culture on selective media, biodiversity studies based on clone library construction and analysis, quantitative PCR amplification of specific gene sequences and the application of generic microarrays for microbial community analysis. Here, we investigated the biodiversity and functional potential of a soil community at Mars Oasis on Alexander Island in the southern Maritime Antarctic, by applying 454 pyrosequencing technology to a metagenomic library constructed from soil genomic DNA. The results suggest that the commonly cited range of phylotypes used in clone library construction and analysis of 78–730 OTUs (de-replicated to 30–140) provides low coverage of the major groups present (∼5%). The vast majority of functional genes (>77%) were for structure, carbohydrate metabolism, and DNA/RNA processing and modification. This study suggests that prokaryotic diversity in Antarctic terrestrial environments appears to be limited at the generic level, with Proteobacteria, Actinobacteria being common. Cyanobacteria were surprisingly under-represented at 3.4% of sequences, although ∼1% of the genes identified were involved in CO2 fixation. At the sequence level there appeared to be much greater heterogeneity, and this might be due to high divergence within the relatively restricted lineages which have successfully colonized Antarctic terrestrial environments.

Global change and Antarctic terrestrial biodiversity

Over the past 50 years, portions of Antarctica and thesub-Antarctic islands have experienced some of the mostrapid increases in mean air temperatures on Earth. Forexample, recent data released by NASA have shown thatmean annual air temperatures over the Western AntarcticIce Sheet (WAIS) have already increased by roughly 1 Cand more than 2 C in the maritime Antarctic (e.g. Turneret al. 2005). Over the same period, introductions of non-indigenous species have also continued to increase partic-ularly in the sub-Antarctic islands (Frenot et al. 2005).While ongoing debate, often politically motivated, con-tinues to dominate discussion of climate change and itsimpacts, it is virtually certain that temperature increasesand species’ introductions will intensify and continue toinfluence Antarctic terrestrial ecosystems over the next50 years. Other human activities such as increasing num-bers of tourists and scientists will affect the present ter-restrial systems as well as those newly revealed as a resultof glacial melt. Collectively, these changes will influenceAntarctica’s biodiversity and ecosystem functioning aswell as the corresponding feedbacks to glaciers, freshwatersystems and the atmosphere. Thus, it is urgent that wedevelop a strong knowledge base for Antarctic terrestrialecosystems and use this to identify ecosystem change(NAS 2011).In order to advance our current state of knowledge, theScientific Committee for Antarctic Research (SCAR),through its life sciences programme Evolution and Biodi-versity in the Antarctic (EBA, http://www.eba.aq), hasencouraged multidisciplinary and multinational researchefforts. As part of this initiative, a special session on GlobalTipping Points, ‘‘Global Change and Antarctic TerrestrialBiodiversity’’, was convened at the 2010 SCAR Open Sci-ence Conference held in Buenos Aires (August 3–6, 2010).This special issue of Polar Biology and the articles thatfollow were based on this special session and were solic-ited to cover a range of topics, viewpoints and contributingcountries. We have assembled eight such manuscripts thatcover microbiological, botanical and zoological compo-nents as well as representing studies from continental,peninsular and sub-Antarctic island locations. This specialissue also provides a useful complement to a previousspecial issue of Polar Biology devoted to polar marine andfreshwater ecosystems (Agusti´ et al. 2010).The lead article by Convey (2011, this special issue)provides an overview of the Antarctic terrestrial environ-ment and its biodiversity. It then examines the range ofenvironmental changes as well as direct and indirect humanactivities that are likely to impact on this unique environ-ment. The final section is future-focused, outlining researchneeds as well as the research programmes and initiatives,past and present, which aim to address these issues. It endswith a plea for the urgent need to plan and properly fund, atnational and international levels, appropriate monitoring,manipulations and modelling approaches across the rangeof terrestrial environments found within the Antarctic(Wall et al. 2011).Green et al. Determining the native/non-native status of newly discovered terrestrial and freshwater species in Antarctica – Current knowledge, methodology and management action

Continental Antarctic terrestrial and freshwater environments currently have few established non-native species compared to the sub-Antarctic islands and other terrestrial ecosystems on Earth. This is due to a unique combination of factors including Antarctica's remoteness, harsh climate, physical geography and brief history of human activity. However, recent increases in national operator and tourism activities increase the risk of non-native propagules reaching Antarctica, while climate change may make successful establishment more likely. The frequency and probability of human-assisted transfer mechanisms appear to far outweigh those of natural propagule introductions by wind, water, birds and marine mammals. A dilemma for scientists and environmental managers, which is exacerbated by a poor baseline knowledge of Antarctic biodiversity, is how to determine the native/non-native status of a newly discovered species which could be (a) a previously undiscovered long-term native species, (b) a recent natural colonist or (c) a human-mediated introduction. A correct diagnosis is crucial as the Protocol on Environmental Protection to the Antarctic Treaty dictates dramatically different management responses depending on native/non-native status: native species and recent natural colonists should be protected and conserved, while non-native introductions should be eradicated or controlled. We review current knowledge on how available evidence should be used to differentiate between native and non-native species, and discuss and recommend issues that should be considered by scientists and managers upon discovery of a species apparently new to the Antarctic region.

Change in gene abundance in the nitrogen biogeochemical cycle with temperature and nitrogen addition in Antarctic soils

The microbial community (bacterial, archaeal, and fungi) and eight genes involved in the nitrogen biogeochemical cycle ( nifH, nitrogen fixation; bacterial and archaeal amoA, ammonia oxidation; narG, nitrate reduction; nirS, nirK, nitrite reduction; norB, nitric oxide reduction; and nosZ, nitrous oxide reduction) were quantitatively assessed in this study, via real-time PCR with DNA extracted from three Antarctic soils. Interestingly, AOB amoA was found to be more abundant than AOA amoA in Antarctic soils. The results of microcosm studies revealed that the fungal and archaeal communities were diminished in response to warming temperatures (10 °C) and that the archaeal community was less sensitive to nitrogen addition, which suggests that those two communities are well-adapted to colder temperatures. AOA amoA and norB genes were reduced with warming temperatures. The abundance of only the nifH and nirK genes increased with both warming and the addition of nitrogen. NirS-type denitrifying bacteria outnumbered NirK-type denitrifiers regardless of the treatment used. Interestingly, dramatic increases in both NirS and NirK-types denitrifiers were observed with nitrogen addition. NirK types increase with warming, but NirS-type denitrifiers tend to be less sensitive to warming. Our findings indicated that the Antarctic microbial nitrogen cycle could be dramatically altered by temperature and nitrogen, and that warming may be detrimental to the ammonia-oxidizing archaeal community. To the best of our knowledge, this is the first report to investigate genes associated with each process of the nitrogen biogeochemical cycle in an Antarctic terrestrial soil environment.

Evolution and biodiversity of Antarctic organisms: a molecular perspective

The Antarctic biota is highly endemic, and the diversity and abundance of taxonomic groups differ from elsewhere in the world. Such characteristics have resulted from evolution in isolation in an increasingly extreme environment over the last 100 Myr. Studies on Antarctic species represent some of the best examples of natural selection at the molecular, structural and physiological levels. Analyses of molecular genetics data are consistent with the diversity and distribution of marine and terrestrial taxa having been strongly influenced by geological and climatic cooling events over the last 70 Myr. Such events have resulted in vicariance driven by continental drift and thermal isolation of the Antarctic, and in pulses of species range contraction into refugia and subsequent expansion and secondary contact of genetically distinct populations or sister species during cycles of glaciation. Limited habitat availability has played a major role in structuring populations of species both in the past and in the present day. For these reasons, despite the apparent simplicity or homogeneity of Antarctic terrestrial and marine environments, populations of species are often geographically structured into genetically distinct lineages. In some cases, genetic studies have revealed that species defined by morphological characters are complexes of cryptic or sibling species. Climate change will cause changes in the distribution of many Antarctic and sub-Antarctic species through affecting population-level processes such as life history and dispersal.

Long-term survival of human faecal microorganisms on the Antarctic Peninsula

Human faecal waste has been discarded at inland Antarctic sites for over 100 years, but little is known about the long-term survival of faecal microorganisms in the Antarctic terrestrial environment or the environmental impact. This study identified viable faecal microorganisms in 30–40 year old human faeces sampled from the waste dump at Fossil Bluff Field Station, Alexander Island, Antarctic Peninsula. Viable aerobic and anaerobic bacteria were predominantly spore-forming varieties (Bacillus and Clostridium spp.). Faecal coliform bacteria were not detected, indicating that they are less able to survive Antarctic environmental conditions than spore-forming bacteria. In recent years, regional warming around the Antarctic Peninsula has caused a decrease in permanent snow cover around nunataks and coastal regions. As a result, previously buried toilet pits, depots and food dumps are now melting out and Antarctic Treaty Parties face the legacy of waste dumped in the Antarctic terrestrial environment by earlier expeditions. Previous faecal waste disposal on land may now start to produce detectable environmental pollution as well as potential health and scientific problems.

Solar UV-B radiation inhibits the growth of Antarctic terrestrial fungi.

We tested the effects of solar radiation, and UV-B in particular, on the growth of Antarctic terrestrial fungi. The growth responses to solar radiation of five fungi, Geomyces pannorum, Phoma herbarum, Pythium sp., Verticillium sp., and Mortierella parvispora, each isolated from Antarctic terrestrial habitats, were examined on an agar medium in the natural Antarctic environment. A 3-h exposure to solar radiation of >287 nm reduced the hyphal extension rates of all species relative to controls kept in the dark. Pythium sp. cultures exposed to solar radiation for 1.5 h on five consecutive days were most sensitive to radiation of >287 nm, but radiation of >313 nm also inhibited growth to a lesser extent. Radiation of >400 nm had no effect on hyphal growth relative to controls kept in the dark. Short-wave solar UV-B radiation of between 287 and 305 nm inhibited the growth of Pythium sp. hyphae on and below the surface of the agar medium after 24 h, but radiation of > or =345 nm only reduced the growth of surface hyphae. Similar detrimental effects of UV-B on surface and, to a lesser extent, submerged hyphae of all five fungi were shown in the laboratory by using artificial UV-B from fluorescent lamps. A comparison of growth responses to solar radiation and temperature showed that the species that were most resistant to UV radiation grew fastest at higher temperatures. These data suggest that solar UV-B reduces the growth of fungi on the soil surface in the Antarctic terrestrial environment.

Glutamate dehydrogenase from two Antarctic organisms, the icefish Chaenocephalus aceratus and the bacterium Psychrobacter sp. TAD1

Glutamate dehydrogenase (GDH) was purified from the liver of the teleost Chaenocephalus aceratus (Notothenioidei: Channichthyidae) and the microorganism Psychrobacter sp. TAD1, from Antarctic marine and terrestrial environments, respectively. GDH isolated from C. aceratus liver had a hexameric molecular structure very similar to that of other vertebrates and displayed preference for NAD+, a feature shared with other fish enzymes. The bovine and fish GDH activity and stability were differently affected by temperature and hydrostatic pressure. At low temperatures, the specific activity of fish GDH was higher than that measured with the homologous bovine enzyme. Psychrobacter sp. TAD1 showed a feature quite unusual in bacteria, i.e. the presence of two distinct GDHs specific either for NADP+ or for NAD+. NADP+ ‐dependent GDH was purified and characterised. It has a hexameric structure with a subunit molecular weight similar to that described for this class of GDHs and a specific activity at low and moderate temperatures similar to that measured with the homologous enzyme from Escherichia coli. The kinetic properties of NADP+ ‐dependent GDH of Psychrobacter sp. TAD1 and the presence of another NAD+‐dependent GDH suggest that, during the cold‐adaptation process, this enzymatic function acquired a pattern of changes different from that of C. aceratus.

Diversity of Antarctic terrestrial protozoa

Heterotrophic protozoa have a global distribution in terrestrial habitats. The functional groups significantly represented are zooflagellates, cillates, gymnamoebae and testate amoebae. Their range extends into the Antarctic zone, but the species richness of the communities is rarely of the same order of magnitude as those in temperate latitudes. Species diversity is usually very low owing to dominance of the communities by single, or a few, species which are best adapted to the Antarctic terrestrial environment. This is characterized by seasonal, diurnal or unpredictable fluctuations in moisture, temperature and bacterial food supply of high amplitude. The fauna shows pauperization with latitude and climatic severity. Nearly all records of species distribution are consistent with the model that community composition is determined by local conditions. An important exception is the distribution of the testate amoeba genus Nebela whose species distribution is influenced by biogeographical factors. Successional changes in community composition in fellfield habitats are characterized by the sequence: pioneer microflagellate colonizers, larger flagellates and small ciliates, and finally testate amoebae. The succession is most closely correlated with the accumulation of organic matter. A model of the strategies of dominant microflagellate species can be constructed by ordinating them on a two-dimensional habitat template of A-r-K selection continuum. The globally ubiquitous microflagellate Heteromita globosa emerges as the most strongly A-selected and K-selected. The occurrence of terrestrial protozoa near their latitudinal limits of distribution can serve as sensitive indicators of the biological effects of climatic change. Having short generation times and effective means of cyst dispersal, changes in the gross distribution can provide rapid warning of critical changes in thermal regimes.
