Global deficiency in the inflammatory chemokine receptors 1,2,3 and 5 prevents vascular dysfunction in angiotensin II-induced hypertension and selectively modulates aortic myeloid cell phenotype

Abstract Background Leukocyte migration to the vasculature and the heart plays a critical role in hypertensive immune responses and is a carefully orchestrated process, regulated by the interactions of inflammatory chemokines with their receptors, notably CCR1, CCR2, CCR3, and CCR5 (collectively termed inflammatory chemokine receptors (iCCRs)). However, the inflammatory chemokine/receptor system is characterised by redundancy, ligand sharing and overlapping expression patterns. Consequently, our understanding of the specific and combinatory roles of chemokine receptors in cardiovascular inflammation remains incomplete, thus hindering the development of targeted therapies. To address this challenge, we have utilised two novel mouse models: iREP mice, carrying fluorescent reporters of inflammatory chemokine receptor expression, and TACKO mice, in which the entire iCcr locus containing Ccr1, Ccr2, Ccr3, and Ccr5 has been deleted. Purpose To evaluate the expression pattern of iCCRs in experimental hypertension and understand how the global deletion of iCCRs impacts disease progression. Methods We induced hypertension in mice by subcutaneously implanting osmotic minipumps administering angiotensin II (Ang II) for a duration of 14 days. Single-cell RNA sequencing was utilised for investigating iCCR expression in aortas during hypertension. Additionally, we employed flow cytometry and confocal microscopy to track iCCR expression and localization in aortas and hearts obtained from iREP mice. TACKO mice were utilised to assess the effect of global iCCr deficiency on hypertensive phenotypes. Blood pressure was measured via telemetry and vascular function was assessed using myography. Vascular and heart remodelling were evaluated through histology. Molecular mechanisms were analysed using Western Blotting and RT-qPCR, while immune cell changes in the aorta were evaluated through cytometry of time-of-flight (CyTOF). Results Ang II infusion induced higher iCCRs expression levels in both the aorta and the heart in comparison with sham treatment, particularly in fibrotic areas. After Ang II-treatment, global deletion of iCCRs in TACKO mice resulted in the prevention of endothelial dysfunction (n=5/group, P<0.05) and perivascular fibrosis (n=6-9, P<0.01), and a reduction in cardiomyocyte size (n=12-16, P<0.01) compared to wild type mice. However, no significant improvements were observed in other hypertensive phenotypes, including blood pressure, heart fibrosis, vascular NO synthase, and oxidative stress. CyTOF analysis revealed reductions in inflammatory monocytes and conventional type 2 dendritic cells in TACKO aorta, accompanied by a phenotypic switch in macrophages towards a less inflammatory phenotype. Conclusion Deficiency in iCCRs confers protective effects against vascular dysfunction in experimental hypertension, primarily by selectively reducing inflammatory myeloid cell homing to the aorta.

α1C S1928 Phosphorylation of CaV1.2 Channel Controls Vascular Reactivity and Blood Pressure.

Increased vascular CaV1.2 channel function causes enhanced arterial tone during hypertension. This is mediated by elevations in angiotensin II/protein kinase C signaling. Yet, the mechanisms underlying these changes are unclear. We hypothesize that α1C phosphorylation at serine 1928 (S1928) is a key event mediating increased CaV1.2 channel function and vascular reactivity during angiotensin II signaling and hypertension. The hypothesis was examined in freshly isolated mesenteric arteries and arterial myocytes from control and angiotensin II-infused mice. Specific techniques include superresolution imaging, proximity ligation assay, patch-clamp electrophysiology, Ca2+ imaging, pressure myography, laser speckle imaging, and blood pressure telemetry. Hierarchical "nested" and appropriate parametric or nonparametric t test and ANOVAs were used to assess statistical differences. We found that angiotensin II redistributed the CaV1.2 pore-forming α1C subunit into larger clusters. This was correlated with elevated CaV1.2 channel activity and cooperativity, global intracellular Ca2+ and contraction of arterial myocytes, enhanced myogenic tone, and altered blood flow in wild-type mice. These angiotensin II-induced changes were prevented/ameliorated in cells/arteries from S1928 mutated to alanine knockin mice, which contain a negative modulation of the α1C S1928 phosphorylation site. In angiotensin II-induced hypertension, increased α1C clustering, CaV1.2 activity and cooperativity, myogenic tone, and blood pressure in wild-type cells/tissue/mice were averted/reduced in S1928 mutated to alanine samples. Results suggest an essential role for α1C S1928 phosphorylation in regulating channel distribution, activity and gating modality, and vascular function during angiotensin II signaling and hypertension. Phosphorylation of this single vascular α1C amino acid could be a risk factor for hypertension that may be targeted for therapeutic intervention.

Gastrodin Alleviates Angiotensin II-Induced Hypertension and Myocardial Apoptosis via Inhibition of the PRDX2/p53 Pathway In Vivo and In Vitro.

Gastrodin, a highly potent compound found in the traditional Chinese medicine Gastrodia elata Blume, exhibits significant antihypertensive properties. However, its role and the mechanism behind its protective effects on hypertensive cardiac conditions are not well understood. This study aims to investigate the cardiac protective effects and underlying mechanisms of gastrodin in angiotensin II (Ang II)-induced hypertensive models, both in vivo and in vitro. Treatment with gastrodin significantly decreased blood pressure and the heart weight/tibial length (HW/TL) ratio and attenuated cardiac dysfunction and pathological damage in Ang II-infused C57BL/6 mice. RNA sequencing analysis (RNA-seq) revealed 697 up-regulated and 714 down-regulated transcripts, along with 1105 signaling pathways, in Ang II-infused C57BL/6 mice following gastrodin treatment, compared to Ang II-induced hypertensive mice. Furthermore, the analyses of the top 30 Gene Ontology (GO) terms and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway indicated significant enrichment in apoptosis and the peroxiredoxin 2 (PRDX2)/p53 pathway. Consistently, gastrodin treatment significantly reduced myocardial apoptosis in both the cardiac tissues of Ang II-induced hypertensive mice and Ang II-stimulated H9c2 cells. Additionally, gastrodin treatment significantly decreased the protein levels of PRDX2, p53, cleaved caspase-3, cleaved caspase-9, and Bax/Bcl-2 ratio in the cardiac tissues of Ang II-infused mice and H9c2 cells stimulated with Ang II. In conclusion, gastrodin treatment can mitigate hypertension-induced myocardial apoptosis in hypertensive mice by inhibiting the PRDX2/p53 pathway.

Abstract MP02: Inhibition of RORγt Ameliorates Angiotensin II-Induced Blood-Brain Barrier Disruption and Microglia Activation by Reducing Interleukin-17A Production in Rats

Inflammation-induced blood-brain barrier (BBB) disruption and microglial activation in the autonomic and cardiovascular brain centers contribute largely to sympathetic excitation and hypertension. We recently reported that interleukin (IL)-17A, a major mediator of tissue inflammation, promotes BBB disruption, neuroinflammation, and sympathetic activation in angiotensin (ANG) II-induced hypertension and myocardial infarction-induced heart failure. The nuclear receptor retinoid-related orphan receptor γt (RORγt) has been identified as a master transcription factor regulating the differentiation of T helper 17 cells, the primary producer of IL-17A. The present study sought to determine whether inhibition of RORγt to suppress IL-17A production can ameliorate BBB disruption and attenuate microglia activation in ANG II-induced hypertension model. SD rats were treated with a 2-week subcutaneous (SC) infusion of ANG II (150 ng/kg/min) via mini-pumps, combined with daily SC injections of RORγt inhibitor digoxin or vehicle (VEH). Compared to the VEH control, IL-17A levels in plasma and cerebrospinal fluid were significantly (*P<0.05) elevated in ANG II+VEH rats and were reduced in ANG II + digoxin rats. In ANG II+VEH rats, BBB permeability, measured by fluorescein isothiocyanate-dextran (FITC) 10 kDa, was elevated (6.82±0.61%* vs. 2.36±0.28% FITC10 extravasation), along with increased mRNA levels of caveolin-1 (a transcytosis marker, 2.26±0.39* vs. 1.07±0.17) and decreased mRNA levels of mfsd2a (a BBB-specific lipid transporter and inhibitor of transcytosis, 0.55±0.04* vs. 1.06±0.09) in the hypothalamic paraventricular nucleus (PVN). However, BBB permeability and mRNA levels of caveolin-1 and mfsd2a in the PVN were reversed in ANG II + digoxin rats. Immunostaining revealed that both caveolin-1 and mfsd2a were primarily located in the endothelial BBB within the PVN. Microglia activation, assessed by the number of branches (26±2* vs. 37±4) and total branch length (184±15* vs. 250±23), as well as the mRNA levels of tumor necrosis factor-α (2.78±0.45* vs. 1.04±0.12) and IL-1β (2.57±0.53* vs. 1.03±0.14) in the PVN were augmented in ANG II+VEH rats but were attenuated (40-58%*) in ANG II + digoxin rats. Additionally, ANG II-induced hypertension and sympathetic tone were attenuated by digoxin. These data suggest that inhibiting RORγt diminishes BBB disruption, microglial activation and sympathetic excitation induced by ANG II, likely via reducing IL-17A production.

Abstract P358: Interaction of Cytochrome P450 (CYP) 4A10 and CYP1B1 in the Paraventricular Nucleus in the Angiotensin II-induced Hypertension and Neuroinflammation in Female Mice

We have shown that 17β-estradiol (E2)-cytochrome P450 (CYP) 1B1 and catechol-ο-methyltrasnferase-generated metabolite 2-methoxyestradiol (2-ME) in the paraventricular nucleus (PVN) protects against angiotensin (Ang) II-induced hypertension and neuroinflammation. This study was conducted to determine the relationship between CYP4A10 that is expressed in the PVN and metabolizes arachidonic acid (AA) into prohypertensive eicosanoid, 20-hydroxyeicosatetraenoic acid (20-HETE), and CYP1B1 in Ang II-induced hypertension, neuroinflammation, and reactive oxygen species (ROS) production in female mice. Systemic Ang II infusion (700 ng/kg/min, s.c., two weeks) and transduction in the PVN with adenovirus (Ad)-GFP scrambled (Scr)-short hairpin (sh) RNA (200 nL bilaterally, 6.4×10 11 pfu/mL, single injection) produced a significant increase in systolic blood pressure (SBP, tail-cuff) at Day 12 compared to Day 0 (mmHg, 114±3 vs 151±3 respectively, P<0.05, n=6/group) in wild-type (WT) and a much larger increase in CYP1B1 knockout (KO) female mice having E2 but not 2-ME (119±4 vs 173±6 respectively, P<0.05, n=5/group). Ad-GFP-CYP4A10 shRNA (200 nL bilaterally, 1.0×10 12 pfu/mL, single injection) but not Ad-GFP Scr-shRNA transduced in the PVN diminished Ang II-induced increase in SBP at Day 12 in WT mice (mmHg, 119±2 vs 122±3 respectively, P<0.05, n=5/group), and CYP1B1KO mice (mmHg, 115±1 vs 133±2 respectively, P<0.05, n=5/group). Similar results were obtained with Ad-GFP-CYP4A10 shRNA compared to Ad-GFP Scr-shRNA transduced in the PVN on mean arterial pressure (MAP) measured by radiotelemetry (Day 12: mmHg, 121±4 vs 152±4 respectively, P<0.05, n=4/group) in CYP1B1 KO mice. The number of TMEM119 + microglia and GFAP + astrocytes, markers of neuroinflammation determined by immunofluorescence, and ROS production by dihydroethidium staining followed the changes in blood pressure produced by Ang II in the above groups. To determine the contribution of central AA-CYP4A10-generated 20-HETE to Ang II-induced hypertension, the effect of 20-HETE receptor GPR75 siRNA administered intracerebroventricularly (Dharmacon, SMARTpool: 0.4 nmol 2 μ l, single injection ) was examined in CYP1B1KO mice. GPR75 siRNA but not its non-target siRNA minimized Ang II-induced increase in SBP (Day 12: mmHg, 132±4 vs 171±3, P<0.05, n=6/group). These data suggest that the 20-HETE receptor antagonist could be useful in treating hypertension and associated neuroinflammation in females with ovarian dysfunction.

Abstract MP18: Evidence for An Important Physiological Role of Bradykinin B 2 Receptors in The Renomedullary Interstitial cells of The Kidney in Angiotensin II-induced Hypertension in Mice

Bradykinin is a potent vasoactive peptide of the kallikrein-kinin system and activates two different classes of G protein-coupled receptors, B 1 and B 2 , in the kidney. In the present study, we tested the hypothesis that deletion of B 2 receptors selectively in renomemdullary interstitial cells (RMICs) of the kidney medulla lowers basal blood pressure and attenuates angiotensin II (Ang II)-induced hypertension. To test the hypothesis, we generated a novel mouse model with deletion of B 2 receptors selectively in RMICs of the medulla (RMIC- Bdkrb2 -/- ) using inducible Tenascin-C-CreER2/Bdkrb2 fl/fl recombination approach. Mice with deletion of B 2 receptors selectively in RMICs developed and grew normally without structural abnormalities in the kidney medulla. Basal telemetry systolic (SBP), diastolic, and mean arterial blood pressure were significantly lower in RMIC- Bdkrb2 -/- than WT mice (WT: 123 ± 3 mmHg vs. RMIC- Bdkrb2 -/- : 106 ± 3 mmHg, P <0.01) (n=12-15). The hypotensive phenotype in RMIC- Bdkrb2 -/- mice was associated with significant decreases in 24 h urine output (WT: 1.86 ± 0.15 mL/24 h vs. RMIC- Bdkrb2 -/- : 1.09 ± 0.15 mL/24 h, P <0.05) and urine osmolality (WT: 1893 ± 441 mOsm/Kg H 2 O vs. RMIC- Bdkrb2 -/- : 1311 ± 78 mOsm/Kg H 2 O, P <0.01) (n=12=15). Deletion of Bdkrb2 receptors in RMICs significantly decreases COX-2, eNOS, ENaC, and AQP2 mRNA expression ( P <0.01 vs. WT), but not PGE 2 or prorenin receptor mRNA expression in the kidney medulla (n.s.) (n=6-10). In response to Ang II, SBP increased to 166 ± 6 mmHg in WT mice ( P <0.01 vs. Basal), but it increased only to 139 ± 5 mmHg in RMIC- Bdkrb2 -/- mice ( P <0.01 vs. WT+Ang II) (n=10-12). Ang II-induced hypertension was completely blocked by oral administration of AT 1 receptor blocker losartan (20 mg/kg/day, p.o.) in WT and RMIC- Bdkrb2 -/- mice (n=10). By comparison, infusion of DDAVP (1-deamino-8-D-arginine vasopressin, ~0.5 mg/kg/day, i.p., for 2 weeks) had no effect on blood pressure in WT and RMIC- Bdkrb2 -/- mice (n.s.) (n=8). Similarly, infusion of Ang II or DDAVP for 2 weeks significantly increased urine osmolality in WT mice ( P <0.01), and restored urine osmolality in RMIC- Bdkrb2 -/- mice to the WT basal level (1995 ± 315 mOsm/Kg H 2 O, P <0.01) (n=8-12). In conclusion, the present study demonstrates for the 1 st time that bradykinin B 2 receptors in RMICs of the kidney medulla plays an important role in maintaining basal blood pressure and urine osmolality homeostasis and the development of Ang II-induced hypertension.

Sex differences and role of lysyl oxidase-like 2 in angiotensin II-induced hypertension in mice.

Hypertension, a disease with known sexual dimorphism, accelerates aging-associated arterial stiffening, partly because of the activation of matrix remodeling caused by increased biomechanical load. In this study, we tested the effect of biological sex and the role of the matrix remodeling enzyme lysyl oxidase-like 2 (LOXL2) in hypertension-induced arterial stiffening. Hypertension was induced by angiotensin II (ANG II) infusion via osmotic minipumps in 12- to 14-wk-old male and female mice. Blood pressure and pulse wave velocity (PWV) were measured noninvasively. Wire myography and uniaxial tensile testing were used to test aortic vasoreactivity and mechanical properties. Aortic wall composition was examined by histology and Western blotting. Uniaxial stretch of cultured cells was used to evaluate the effect of biomechanical strain. LOXL2's catalytic function was examined using knockout and inhibition. ANG II infusion-induced hypertension in both genotypes and sexes. Wild-type (WT) males exhibited arterial stiffening in vivo and ex vivo. Aortic remodeling with increased wall thickness, intralamellar distance, higher LOXL2, and collagen I and IV content was noted in WT males. Female mice did not exhibit increased PWV despite the onset of hypertension. LOXL2 depletion improved vascular reactivity and mechanics in hypertensive males. LOXL2 depletion improved aortic mechanics but worsened hypercontractility in females. Hypertensive cyclic strain contributed to LOXL2 upregulation in the cell-derived matrix in vascular smooth muscle cells (VSMCs) but not endothelial cells. LOXL2's catalytic function facilitated VSMC alignment in response to biomechanical strain. In conclusion, in males, arterial stiffening in hypertension is driven both by VSMC response and matrix remodeling. Females are protected from PWV elevation in hypertension. LOXL2 depletion is protective in males with improved mechanical and functional aortic properties. VSMCs are the primary source of LOXL2 in the aorta, and hypertension increases LOXL2 processing and shifts to collagen I accumulation. Overall, LOXL2 depletion offers protection in young hypertensive males and females.NEW & NOTEWORTHY We examined the effect of sex on the evolution of angiotensin II (ANG II)-induced hypertension and the role of lysyl oxidase-like 2 (LOXL2), an enzyme that catalyzes matrix cross linking. While ANG II led to hypertension and worsening vascular reactivity in both sexes, aortic remodeling and stiffening occurred only in males. LOXL2 depletion improved outcomes in males but not females. Thus males and females exhibit a distinct etiology of hypertension and LOXL2 is an effective target in males.

Abstract 32: Vγ6 + γδ T cells protect against angiotensin II-induced hypertension and vascular injury in male mice

Background: γδ T cells mediate angiotensin II (AngII)-induced blood pressure (BP) elevation, vascular injury and activated T cell infiltration in mesenteric vessels with adherent perivascular adipose tissue (MV/PVAT). γδ T cells expressing specific T cell receptor (TCR) variable (V) chain γ develop in several waves in the thymus and migrate to diverse tissues. We hypothesized that γδ T cells expressing specific Vγ subtypes in perivascular tissue are implicated in AngII detrimental hypertensive effects. Methods: C57BL/6J male mice were infused or not with AngII (490 ng/kg/min, SC) for 14 days. Vγ1 and Vγ2 (Vγ1/2) + , Vγ4 + , Vγ5 + , Vγ6 + , and Vγ7 + γδ T cell frequencies and T cell phenotypes were determined by flow cytometry in the spleen, descending thoracic aorta (DTAo)/PVAT, and MV/PVAT (n=7-13). Other sets of AngII-infused mice were injected with control or anti-Vγ6 (n=7-12) or Vγ4 antibodies (n=5-8). BP was determined by telemetry, mesenteric artery (MA) function and remodeling by pressurized myography, and T cell phenotypes by flow cytometry. Results: In DTAo/PVAT and MV/PVAT, respectively, 43±5% and 51%±6% of γδ T cells were Vγ6 + , and 17±2% and 23±4% Vγ4 + , whereas in the spleen, 36±2% of the γδ T cells were Vγ1/2 + , 32±1% Vγ4 + and 24±1% Vγ6 + . AngII increased the frequency of Vγ6 + γδ T cells in the spleen (0.8±0.1 vs 0.5±0.0% of CD3, P <0.001) and DTAo/PVAT (5.9±0.8 vs 3.6±0.5% of CD3, P <0.05), and tended to increase their frequency in MV/PVAT. The majority of Vγ6 + γδ T cells were activated (CD69 + ) in DTAo/PVAT (75±5%) and MV/PVAT (84±2%). Vγ6 + γδ T cell depletion caused a steeper BP elevation ( P <0.05) and worsened MA endothelial dysfunction (maximal acetylcholine relaxation response: 28±3 vs 55±8%, P <0.05) in mice infused with AngII. This was associated with increased CD69 + non-Vγ6 + γδ T cells in DTAo/PVAT (3.0±0.7 vs 0.8±0.4% of CD3, P <0.01) and MV/PVAT (15.2±2.6 vs 4.9±1.0% of CD3, P <0.01). Depletion of Vγ4 + γδ T cells in perivascular tissues, did not alter the AngII detrimental effects. Conclusions: Vγ6 + γδ T cells are the most abundant γδ T cell Vγ subtype in perivascular tissues. AngII increased Vγ6 + γδ T cells in the spleen and perivascular tissue. Vγ6 + γδ T cell depletion in AngII-infused mice induced a steeper BP elevation, worsened MA endothelial dysfunction, and activated non-Vγ6 + γδ T cells. Vg6 + γδ T cells may play a protective role in AngII-induced hypertension and vascular injury.

Abstract P421: Differential Osmolyte Profiles in Hypertensive and Normotensive Rats.

Hypertension is known to be associated with water-electrolyte balance disturbances that lead to changes in osmotic pressure. Osmolytes are small organic molecules essential for cellular adaptation to changes in osmotic pressure. This study investigates the levels of osmolytes in serum and tissues using two well-established rat models of hypertension: spontaneously hypertensive rats (SHRs) and Wistar-Kyoto rats with angiotensin II-induced hypertension (WKY-ANG). Experiments were conducted on male Wistar-Kyoto (WKY) rats, SHRs, and WKY-ANG rats. Blood and tissue samples were collected from all groups for liquid chromatography with mass spectrometry (LC-MS) analysis of selected osmolytes. Anesthetized SHR and WKY-ANG rats displayed significantly elevated mean arterial blood pressures compared to WKY rats (118.4±1.3, 110.2±0.9, and 76.5±1.2 mmHg, respectively). The serum and tissue levels of sarcosine were significantly decreased in SHRs compared to WKY rats (1.90±0.21 vs. 3.80±0.80 for serum, 55.55±9.79 vs 92.37±10.57 for liver, 6.06±0.78 vs. 10.55±1.19 for renal cortex, 9.14±0.45 vs.13.83±1.14 for renal medulla, 11.59±1.17 vs. 22.34±3.11 for lungs, 1.80±0.14 vs. 3.30±0.39 for heart, respectively). Reduced levels of beta-alanine were observed only in the serum of SHRs compared to WKY and WKY-ANG rats (6.48±0.93 vs. 59.43±9.47, 41.32±3.13 respectively). Additionally, higher levels of L-alanine were noted in the renal cortex and medulla of WKY than in SHRs and WKY-ANG rats (19685±592.3 vs. 14510±456.8, 14289±719.4 for cortex and 14043±560.3 vs. 11520±206.8, 10651±292.4 for medulla, respectively). Conversely, SHR rats exhibited significantly higher levels of serum glycerophosphocholine (GPC) than WKY-ANG rats (38.21±10.02 vs. 12.47±2.25). Simultaneously, lower levels of betaine were observed in the lungs of SHR rats when compared to WKY and WKY-ANG rats (1 and 351.20±36.02, 607.10±64.20, 606.70±55.88 for lungs respectively). All results are indicated in μmol/L and means±SE. In conclusion significant alterations in osmolyte concentrations are evident in hypertensive rats. These findings suggest that hypertension may be associated with disruptions in osmolyte balance. Further studies are needed to assess the potential clinical significance of osmolyte disturbances in hypertension.

Abstract 63: Acetylation of Mitochondrial Cyclophilin D Increases vascular Oxidative Stress, Induces Glycolitic Switch, Promotes Endothelial Dysfunction and Hypertension

Background: Nearly half of adults have hypertension, a major risk factor for cardiovascular disease. Mitochondrial hyperacetylation is linked to hypertension, but the role of acetylation of specific proteins is not clear. We hypothesized that acetylation of mitochondrial Cyclophilin D (CypD) at K166 contributes to endothelial dysfunction and hypertension. Methods: To test this hypothesis, we studied CypD acetylation in patients with essential hypertension, defined a pathogenic role of CypD acetylation in deacetylation mimetic CypD-K166R mutant mice and endothelial specific general control of amino acid synthesis 5 like 1 (GCN5L1) deficient mice using an angiotensin II model of hypertension. Results: Arterioles from hypertensive patients had 280% higher CypD acetylation coupled with reduced Sirt3 and increased GCN5L1 levels. GCN5L1 regulates mitochondrial protein acetylation and promotes CypD acetylation which is counteracted by mitochondrial deacetylase Sirt3. In human aortic endothelial cells GCN5L1 depletion prevents superoxide overproduction. Deacetylation mimetic CypD-K166R mice were protected from vascular oxidative stress, endothelial dysfunction and angiotensin II-induced hypertension. Angiotensin II-induced hypertension increased mitochondrial GCN5L1 and reduced Sirt3 levels resulting in 250% increase in GCN5L1/Sirt3 ratio leading to imbalance between GCN5L1-mediated acetylation and Sirt3-mediated deacetylation and promoting CypD acetylation. Treatment with mitochondria-targeted scavenger of cytotoxic isolevuglandins, mito2HOBA, normalized GCN5L1/Sirt3 ratio, reduced CypD acetylation, and attenuated hypertension. The role of mitochondrial acetyltransferase GCN5L1 in endothelial function was tested in endothelial specific GCN5L1 knockout mice. Depletion of endothelial GCN5L1 prevented angiotensin II-induced mitochondrial oxidative stress, reduced maladaptive switch of vascular metabolism to glycolysis, prevented inactivation of endothelial nitric oxide, preserved endothelial-dependent relaxation, and attenuated hypertension. Conclusions: These data support the pathogenic role of CypD acetylation in endothelial dysfunction and hypertension. We suggest that targeting cytotoxic mitochondrial isolevuglandins and GCN5L1 reduces CypD acetylation which may be beneficial in cardiovascular disease.

Abstract P171: 2-Methoxyestradiol By Inhibiting Central Action of 12S-Hydroxyeicosatetraenoic Acid Protects Ovariectomized Mice From Hypertension

Previously, we reported that 17β-estradiol (E2)- cytochrome P450 (CYP) 1B1-generated metabolite 2-methoxyestradiol (2-ME) in the paraventricular nucleus (PVN) protects from angiotensin (Ang) II-induced hypertension. Recently, we showed that intracerebroventricular (ICV) administration of 12(S)-hydroxyeicosatetraenoic acid (12S-HETE), an arachidonic acid-12/15-lipoxygenase (LO) metabolite produces a greater increase in blood pressure (BP) in ovariectomized (OVX) than intact LO knockout (KO) mice. These observations led to the hypothesis that 2-ME protects from Ang II-induced hypertension by inhibiting the production and action of 12S-HETE in the brain in female mice. ICV 2-ME (1.5 µg/2 µL/ alternate day) but not its vehicle (20% w/v hydroxypropyl-β-cyclodextrin) minimized the effect of Ang II (700 ng/kg/min, subcutaneous, osmotic pump, two weeks) to increase mean arterial blood pressure (MAP: Day 0 vs Day 12; 105±3 to 121±2 vs 105±3 to 153±8, mmHg) measured by radiotelemetry, urinary norepinephrine level (ELISA, 445±27 vs 898±20, ng/mL), lo mRNA expression (RT-PCR, 1.5±0 vs 2.0±1, arbitrary unit), 12S-HETE level (ELISA, 136±26 vs 285±44, pg/mg protein), and GFAP+ (immunostaining, astrocyte marker, 35±3 vs 67±3, number of cells per PVN section) cells in PVN and kidney fibrosis (histochemical-staining, 4.0±0.3 vs 9.8±0.7, arbitrary unit) in OVX-LOKO mice transduced with ICV-adenovirus-LO-DNA (1.28X10 11 pfu/ml, 2 µL, single injection) that have 12S-HETE in the brain (PVN) but not in plasma (n=4/group; P<0.05). ICV siRNA (0.4 nM, 2 µL, single injection) against 12S-HETE receptors LTB4 and GPR31 minimized Ang II-induced increased systolic BP measured by tail-cuff in intact CYP1B1 KO mice that have E2 but lacks 2-ME (mmHg, control: 108±3 to 175±6, siRNA-LTB4r2: 110±5 to 141±3, siRNA-GPR31: 114±1 to 145±2; n=8 per group). Moreover, the actions of ICV 12S-HETE (50 ng/2 µL every 3rd day) to enhance the effect of Ang II-induced increase in MAP were also minimized by ICV siRNA-LTB4 and GPR31 respectively in OXV-LOKO mice (mmHg, control: 102±2 to 168±1, siRNA-LTB4r2: 101±2 to 122±6, siRNA-GPR31: 101±3 to 129±3; n=4 per group). These data suggest that E2-CYP1B1 metabolite 2-ME, by inhibiting the production and action of 12S-HETE in the brain via LTB4r2 and GPR31 receptors protect from Ang II-induced hypertension in female mice. Thus, 2-ME and/or 12S-HETE receptor blockers could be useful for treating hypertension and its pathogenesis in females with ovarian failure.

Abstract P354: Endothelial Sirt3 deficiency and SOD2 acetylation voids female protection from hypertension

The prevalence of hypertension in early adulthood among women is 3-times lower than in men; but with aging hypertension rate increases in women much steeper, and hypertensive vascular and kidney damage is significantly higher in women. Gender-specific aspects of hypertension are poorly understood and clinical studies did not show protection by estrogen hormone therapy. We found that human hypertension is linked to hyperacetylation of critical antioxidant and metabolic mitochondrial enzymes superoxide dismutase (SOD2) and long-chain acyl dehydrogenase (LCAD) due to vascular deficiency of mitochondrial deacetylase Sirt3. We hypothesized that endothelial Sirt3 depletion leads to the loss of female protective cardiovascular phenotype and exacerbates hypertension in female compared with male mice due to hyperacetylation of mitochondrial proteins. Indeed, infusion of angiotensin II in wild-type female mice showed protection of vascular metabolism, endothelial nitric oxide and lower blood pressure compared with male littermates which was coupled with reduced acetylation of mitochondrial LCAD and SOD2 in female mice. Endothelial specific Sirt3 knockout female mice showed increased mitochondrial acetylation, exacerbated angiotensin II-induced hypertension and higher blood pressure compared with male mice which models human hypertension. Hypertension is linked to SOD2 acetylation (Circ Res. 2020;126(4):439-452) which leads to SOD2 inactivation and vascular oxidative stress. We found that endothelial specific acetylation mimetic SOD2-K68Q knock-in female mice lost antihypertensive phenotype and had higher blood pressure compared with angiotensin II-infused male mice. These studies demonstrate critical role of mitochondrial acetylation in endothelial dysfunction and hypertension suggesting that female antihypertensive phenotype can be associated with reduced mitochondrial acetylation protecting mitochondrial and endothelial functions. It is conceivable that pharmacological targeting of mitochondrial metabolism and acetylation can rescue female cardiovascular protection and improve treatment of endothelial dysfunction and hypertension.

Abstract 31: Deacetylation Mimetic Mutation of Mitochondrial Superoxide Dismutase Attenuates Angiotensin II-Induced Hypertension by Protecting Against Oxidative Stress

Almost one-half of adults have hypertension, and blood pressure is poorly controlled in a third of patients despite the use of multiple drugs, likely due to mechanisms that are not affected by current treatments. Hypertension is linked to oxidative stress; however, common antioxidants are ineffective. Hypertension is associated with inactivation of key intrinsic mitochondrial antioxidant, superoxide dismutase 2 (SOD2), due to hyperacetylation but the role of specific SOD2 lysine residues has not been defined. Hypertension is associated with SOD2 acetylation at Lysine 68 (Circ Res. 2020;126(4):439-452) and we suggested that deacetylation mimetic mutation of K68 to Arginine in SOD2 inhibits vascular oxidative stress and attenuates hypertension. To test this hypothesis, we have developed a new deacetylation mimic SOD2-K68R mice. We performed in vivo studies in SOD2-K68R mice using angiotensin II (AngII) model of vascular dysfunction and hypertension. AngII infusion in wild-type mice induced vascular inflammation and oxidative stress, and increased blood pressure to 160 mm Hg. SOD2-K68R mutation completely prevented increase in mitochondrial superoxide, abrogated vascular oxidative stress, preserved endothelial nitric oxide production, protected vasorelaxation, and attenuated AngII-induced hypertension. AngII and cytokines contribute to vascular oxidative stress and hypertension. Treatment of wild-type aortas with AngII and cytokines in organoid culture increased mitochondrial superoxide by 2-fold which was completely prevented in aortas isolated from SOD2-K68R mice. Interestingly, ex vivo treatment of aortas isolated from hypertensive mice with Sirtuin activator resveratrol reduces SOD2 acetylation, inhibits mitochondrial superoxide, and rescues endothelial nitric oxide. These data support the important role of SOD2-K68 acetylation in vascular oxidative stress and pathogenesis of hypertension. We conclude that strategies to reduce SOD2 acetylation may have therapeutic potential in the treatment of vascular dysfunction and hypertension.

Abstract MP33: A20 in the Kidney Epithelium Attenuates Angiotensin II-induced Hypertension by Constraining Renal Tubular NHE3 Expression

Whereas the ubiquitin editor A20 limits NF-κB-mediated signaling in myeloid leukocytes, the functions of A20 in renal epithelial cells during hypertension have not been described. We recently found that A20 in the kidney tubule ameliorates hypertension and cardiac hypertrophy induced by chronic angiotensin (Ang) II infusion (500ng/kg/min) in mice. Here we report that following 3 weeks of hypertension, the kidneys of mice with tubular deletion of A20 in the kidney epithelium (“A20 iKKO” = A20 flox/flox Pax8-rtTA TetO-Cre + ) have augmented protein expression of the proximal tubular sodium transporter NHE3 (1.5±0.1 vs. 1.0±0.1; p <0.01) but similar levels of ENaC subunit expression in the collecting tubule ( p =NS for all 3 subunits) vs wild-type (WT) controls. Moreover, kidneys from A20 iKKO mice have upregulated mRNA levels for the T cell chemokine CCL5 (2.4±0.3 vs. 1.0±0.3; p <0.01) and higher proportions of effector memory CD8 + T cells (51.2±2.3 vs. 39.3±2.9 % of CD8 + ; p =0.013) compared to WTs despite similar numbers of effector memory T cells in the spleen ( p =NS). To examine whether A20 in the kidney attenuates hypertension by constraining tubular NHE3 expression, we bred A20 iKKO mice with an NHE3 flox/flox line, thereby generating mice with double A20 and NHE3 deletion in the kidney tubule (“Dual KKO”) mice. At baseline, kidneys from Dual KKO mice showed robust reductions in mRNA for A20 (0.5±0.1 vs. 1.0±0.1; p =0.022) and NHE3 (0.2±0.05 vs. 1.0±0.15; p =0.002) compared to WT littermates. During the first ten days of Ang II infusion, mean arterial pressures (MAPs) in the Dual KKO mice were lower than the MAPs previously recorded in our A20 iKKO cohort (129±3 vs. 152±4 mmHg; p <0.01). Moreover, during 3 weeks of Ang II infusion, Dual KKO and WT mice maintained similar MAPs (145±7 vs. 149±13 mmHg; p =0.78), and heart weight to body weight ratios (5.9±0.3 vs. 5.5±0.2 mg/g; p =0.37) in concomitant measurements. Thus, A20 in the kidney tubule blunts the chronic hypertensive response by curtailing NHE3 expression. Our data further suggest that A20 in the nephron can blunt renal inflammation without altering systemic T cell immunity. Stimulating the A20 signaling pathway in the kidney may thus attenuate NHE3-dependent hypertension without conferring attendant risks of global immunosuppression.

Role of the Anaphylatoxin Receptor C5aR2 in Angiotensin II-Induced Hypertension and Hypertensive End-Organ Damage.

Complement activation may facilitate hypertension through its effects on immune responses. The anaphylatoxin C5a, a major inflammatory effector, binds to the C5a receptors 1 and 2 (C5aR1, C5aR2). We have recently shown that C5aR1-/- mice have reduced hypertensive renal injury. The role of C5aR2 in hypertension is unknown. For examination of C5aR2 expression on infiltrating and resident renal cells a tandem dye Tomato-C5aR2 knock-in reporter mouse was used. Human C5aR2 expression was analyzed in a single-cell RNAseq data set from the kidneys of hypertensive patients. Finally, we examined the effect of angiotensin II-induced hypertension in C5aR2-deficient mice. Flow cytometric analysis of leukocytes isolated from kidneys of the reporter mice showed that dendritic cells are the major C5aR2-expressing population (34%) followed by monocyte/macrophages (30%) and neutrophils (14%). Using confocal microscopy C5aR2 was not detected in resident renal or cardiac cells. In the human kidney, C5aR2 was also mainly found in monocytes, macrophages, and dendritic cells with a significantly higher expression in hypertension (P < 0.05). Unilateral nephrectomy was performed followed by infusion of Ang II (0.75 ng/g/min) and a high salt diet in wildtype (n = 18) and C5aR2-deficient mice (n = 14). Blood pressure, renal injury (albuminuria, glomerular filtration rate, glomerular and tubulointerstitial injury, inflammation), and cardiac injury (cardiac fibrosis, heart weight, gene expression) did not differ between hypertensive wildtype and C5aR2-/- mice. In summary, C5aR2 is mainly expressed in myeloid cells in the kidney in mice and humans but its deficiency has no effect on Ang II-induced hypertensive injury.

Sex Differences and Role of Lysyl Oxidase Like 2 (LOXL2) in Angiotensin II-Induced Hypertension in Mice.

Hypertension, a disease with known sexual dimorphism, accelerates aging associated arterial stiffening, in part due to the activation of matrix remodeling caused by increased biomechanical load. In this study, we tested the effect of biological sex and the role of the matrix remodeling enzyme lysyl oxidase like 2 (LOXL2) in hypertension induced arterial stiffening. Angiotensin II (Ang II) was delivered using osmotic pumps in Loxl2+/- and WT male and female mice. Blood pressure and pulse wave velocity (PWV) were measured noninvasively to assess hypertension and aortic stiffness. Wire myography and uniaxial tensile testing were used to test aortic vasoreactivity and mechanical properties. Aortic wall composition was examined by histology and Western blotting. The effect of biomechanical strain on LOXL2 expression and secretion by vascular smooth muscle (VSMC) and endothelial cells (EC) was evaluated by uniaxial cyclic stretching of cultured cells. The role of LOXL2 catalytic function on VSMC alignment in response to mechanical loading was determined with LOXL2 inhibition and knockout. Ang II infusion induced hypertension in WT and Loxl2+/- mice of both sexes and increased PWV in WT males but not in Loxl2+/- males, WT females, or Loxl2+/- females. LOXL2 depletion protected males from Ang II mediated potentiation of vasoconstriction but worsened in females and improved aortic mechanical properties in both sexes. Histological analysis showed increased aortic wall thickness in hypertensive WT males but not females and increased intralamellar distance in both sexes, that was ameliorated in Loxl2+/- mice. Western blotting revealed increased collagen I, decreased collagen IV, and increased LOXL2 accumulation and processing in hypertensive mice. Hypertensive cyclic strain contributed to LOXL2 upregulation in the cell-derived matrix in VSMCs but not ECs. LOXL2 catalytic function facilitated VSMC alignment in response to biomechanical strain. In males, arterial stiffening in hypertension is driven both by VSMC response and matrix remodeling. Females exhibit a delayed onset of Ang II-induced hypertension with minimal ECM remodeling but with VSMC dysfunction. LOXL2 depletion ameliorates arterial stiffening and preserves functional contractility and aortic structure in male hypertensive mice. LOXL2 depletion improves aortic mechanics but worsens aortic contractility in hypertensive females. VSMCs are the primary source of LOXL2 in the aorta and hypertension increases LOXL2 processing and shifts to collagen I accumulation. Overall, LOXL2 depletion offers protection in young hypertensive males and females.

HIF-1α and adaptor protein LIM and senescent cell antigen-like domains protein 1 axis promotes tubulointerstitial fibrosis by interacting with vimentin in angiotensin II-induced hypertension.

Activation of the renin-angiotensin system, as a hallmark of hypertension and chronic kidney diseases (CKD) is the key pathophysiological factor contributing to the progression of tubulointerstitial fibrosis. LIM and senescent cell antigen-like domains protein 1 (LIMS1) plays an essential role in controlling of cell behaviour through the formation of complexes with other proteins. Here, the function and regulation of LIMS1 in angiotensin II (Ang II)-induced hypertension and tubulointerstitial fibrosis was investigated. C57BL/6 mice were treated with Ang II to induce tubulointerstitial fibrosis. Hypoxia-inducible factor-1α (HIF-1α) renal tubular-specific knockout mice or LIMS1 knockdown AAV was used to investigate their effects on Ang II-induced renal interstitial fibrosis. In vitro, HIF-1α or LIMS1 was knocked down or overexpressed in HK2 cells after exposure to Ang II. Increased expression of tubular LIMS1 was observed in human kidney with hypertensive nephropathy and in murine kidney from Ang II-induced hypertension model. Tubular-specific knockdown of LIMS1 ameliorated Ang II-induced tubulointerstitial fibrosis in mice. Furthermore, we demonstrated that LIMS1 was transcriptionally regulated by HIF-1α in tubular cells and that tubular HIF-1α knockout ameliorates LIMS1-mediated tubulointerstitial fibrosis. In addition, LIMS1 promotes Ang II-induced tubulointerstitial fibrosis by interacting with vimentin. We conclude that HIF-1α transcriptionally regulated LIMS1 plays a central role in Ang II-induced tubulointerstitial fibrosis through interacting with vimentin. Our finding represents a new insight into the mechanism of Ang II-induced tubulointerstitial fibrosis and provides a novel therapeutic target for progression of CKD.

A novel dual inhibitor of the angiotensin and endothelin systems for the treatment of hypertension

Hypertension, affecting nearly half of the US adult population, is a primary cause of morbidity and mortality worldwide. Despite the availability of several classes of antihypertensive medications, only 25% of patients have their blood pressure under control, highlighting the need for novel antihypertensive therapeutics with better effcacy. Among hypertensive cases, salt-sensitive and drug-resistant hypertension are particularly concerning, as they are unresponsive to a combined regimen of three or more classes of medications, including angiotensin II (Ang II) receptor blockers (ARBs), angiotensin-converting enzyme inhibitors (ACEis), and diuretics. Salt-sensitive, observed in 50% of the general and 73% of black hypertensive patients, disproportionately affects the latter, leading to significantly higher mortality. Salt-sensitive hypertension in humans, as well as in various experimental models of hypertension (e.g., Ang II-induced, Dahl-salt sensitive [DSS] rats, deoxycorticosterone acetate [DOCA]-salt hypertensive rats), is characterized by significantly elevated levels of plasma endothelin-1 (ET-1), oxidative stress, and kidney dysfunction. ET-1 is often considered a final common pathway in orchestrating cardiovascular dysfunction through a range of proinflammatory mediators such as Ang II, cytokines, hypoxia, thrombin, reactive oxygen species (ROS), oxidized LDL, and vasopressin, all of which stimulate ET-1 production. In this capacity, we report the discovery of a gut microbiota-derived metabolite, indole-3-acetic acid (IAA), that simultaneously blocks ET-1 receptors (ETAR and ETBR) and the ET-biosynthetic enzyme ECE-1. Our data demonstrate that IAA inhibits ET-1-induced constriction of resistance mesenteric arteries, ROS production in human aortic smooth muscle cells, activation of cultured endothelial cells, adhesion molecule expression, and leukocyte adhesion. IAA inhibited purified human ECE-1 and suppressed Ang-II-induced and hypoxia-elicited ET-1 production from endothelial cells in vitro. Administration of a once-daily, extended-release formulation of IAA inhibits blood pressure elevation and kidney dysfunction in both DSS and Ang II-induced hypertension. IAA treatment preserved endothelial nitric oxide (NO) production and maintained smooth muscle sensitivity to a NO donor, sodium nitroprusside. Furthermore, IAA inhibited the expression of ROS-associated proteins such as Nox2 and Nox4, reduced oxidative damages, and decreased the production of ET-1 in vivo. Overall, IAA is the first compound that, through novel dual targeting of both the angiotensin and endothelin systems, demonstrates profound antihypertensive action in both salt-sensitive and angiotensin II-induced hypertension. 2-Methoxyestradiol protects from angiotensin II-induced renal CD3+/FOXP3+ T cell-dysregulation via 12/15 lipoxygenase inhibition

We previously showed that 2-methoxyestradiol (2-ME), an active metabolite of 17β-estradiol (E2) and lox (12/15-lipoxygenase) gene disruption, protects against angiotensin (Ang) II-induced hypertension. Moreover, 2-ME reduces Ang II-induced increases in plasma levels of 12(S)-hydroxyeicosatetraenoic acid (HETE), a major arachidonic acid-LOX-generated metabolite in ovariectomized (OVX, endogenous E2 depleted) wild-type (WT) mice. 2-ME also exerts anti-inflammatory effects, and regulatory T cells (Tregs) have been reported to impact Ang II-induced hypertension in rodent models, with premenopausal females being protected relative to males. This study tested the hypothesis that 2-ME protects against the inflammatory effects of Ang II by upregulating Tregs in the kidney via LOX inhibition in female mice. Intact (ovaries present) or OVX WT and OVX-LOX knock-out (KO) mice (8-10 weeks, n=3-4/group) were infused with Ang II (700 ng/kg/min) or saline (vehicle) via osmotic minipumps (S.C.) with or without 2-ME (15 μg/g, I.P., every third day) treatment for 2 weeks. Renal protein expressions of CD3+ T cells and FOXP3+ Tregs were evaluated by immunofluorescence staining and mRNA expressions of tnf-α and il-10 by RT-PCR using TaqMan assay (relative to saline-infused intact WT) in the whole kidney. Results represented as mean ± SEM were analyzed using one-way ANOVA followed by Tukey's multiple comparisons test. The CD3+, tnf-α and il-10 expressions were comparable between saline-infused intact WT, OVX-WT, and OVX-KO (CD3+: 8.3±0.3, 7.0±0.5, 6.3±0.3; tnf-α: 1.0±0.0, 0.9±0.0, 0.8±0.2; il-10: 1.1±0.1, 0.9±0.1, 1.0±0.0). However, saline-infused intact WT and OVX-KO expressed higher Tregs (p&lt;0.05) as compared to OVX-WT (7.6±0.3, 7.3±0.3 vs 4.0±0.5). Ang II increased CD3+, Tregs, and il-10 (p&lt;0.05) but not tnf-α expressions as compared to its vehicle in intact WT (12.6±0.3, 15.3±0.3, 3.7±0.3,0.8±0.1). Ang II also increased CD3+ (11.0±0.6) and Tregs (11.0±0.6) in OVX-KO (p&lt;0.05), without altering mRNA expressions of tnf-α (0.9±0.3) and il-10 (1.5±0.1) . In contrast, Ang II resulted in a greater increase in CD3+ (25.0±1.1) in OVX-WT than OVX-KO and intact WT, besides increasing tnf-α expression (2.5±0.4) (p&lt;0.05). 2-ME reduced CD3+ (16.7±0.9) and tnf-α (1.4±0.4) expressions, and increased Tregs (10.0±1.0) and il-10 (2.0±0.0) expressions in Ang II-infused OVX-WT mice (p&lt;0.05). 2-ME also reduced CD3+ (6.7±0.3, p&lt;0.05) and showed a trend to increase Tregs (14.0±1.1, P=0.056), and il-10 (2.3±0.2, P=0.058) expressions without altering tnf-α (0.7±0.1) expression in Ang II-infused OVX-KO mice. In conclusion, Ang II produces a specific effect on the renal T cell profile, greater increases in proinflammatory T cells in OVX mice, and anti-inflammatory Tregs in intact female mice, most likely mediated by E2 through metabolite 2-ME via inhibition of LOX. DELETION OF BRADYKININ TYPE 2 RECEPTORS SELECTIVELY IN THE RENOMEDULLARY INTERSTITIAL CELLS OF THE KIDNEY MEDULLA ATTENUATES ANGIOTENSIN II-INDUCED HYPERTENSION IN MICE

Objective: The present study tested the hypothesis that deletion of vasoactive peptide bradykinin B2 receptors selectively in the renomemdullary interstitial cells (RMICs) of the kidney medulla attenuates angiotensin II-induced hypertension in mice. Design and method: To test the hypothesis, we employed inducible Tenascin-C-CreER2 and bradykinin B2 receptor-floxed recombination approach to generate a novel mouse model with deletion of B2 receptors selectively in the RMICs of the kidney medulla (RMIC-B2R-/-). Basal blood pressure and urine osmolality and their responses to angiotensin II (Ang II, ∼500 μg/kg/day, i.p., for 2 weeks) were determined in adult male wildtype (WT) and RMIC-B2R-/- mice (n=10 per group), respectively. Results: Mice with deletion of B2 receptors selectively in RMICs of the kidney medulla developed normally without structural abnormalities in the kidney medulla. However, basal telemetry systolic, diastolic, and mean arterial blood pressure were significantly lower in RMIC-B2R-/- than WT mice (WT: 123 ± 3 mmHg vs. RMIC-B2R-/-: 106 ± 3 mmHg, P&lt;0.01). The hypotensive phenotype in RMIC-B2R-/- mice was associated with significant decreases in 24 h urine output (WT: 1.86 ± 0.15 mL/24 h vs. RMIC-B2R-/-: 1.09 ± 0.15 mL/24 h, P&lt;0.05) and urine osmolality (WT: 1893 ± 441 mOsm/Kg H2O vs. RMIC-B2R-/-: 1311 ± 78 mOsm/Kg H2O, P&lt;0.01). In response to Ang II infusion (∼0.5 mg/kg/day, i.p., for 2 weeks), systolic blood pressure increased to 166 ± 6 mmHg in WT mice (P&lt;0.01 vs. Basal), but it increased only to 139 ± 5 mmHg in RMIC-B2R-/- mice (P&lt;0.01 vs. WT). By comparison, infusion of DDAVP (1-deamino-8-D-arginine vasopressin, ∼0.5 mg/kg/day, i.p., for 2 weeks) did not significantly alter blood pressure in WT and RMIC-B2R-/- mice (n.s.). However, infusion of Ang II for 2 weeks significantly increased urine osmolality in WT mice (P&lt;0.01), and restored urine osmolality in RMIC-B2R-/- mice to the WT basal level (1995 ± 315 mOsm/Kg H2O, P&lt;0.01). Conclusions: In conclusion, the present study demonstrates for the 1st time that bradykinin B2 receptors in the RMICs of the kidney medulla plays an important role in maintaining basal blood pressure and urine osmolality homeostasis and the development of Ang II-induced hypertension.
