The study showcases the utility of Laplace transformations for analyzing systems described by ordinary differential equations with Caputo derivatives. This approach facilitates both analytical and numerical methods for system analysis and parameter estimation. Development and Validation of a Viable and User-Friendly Spectrophotometric Analytical Method for Estimation of Assay of Formoterol and Beclomethasone in Metered Dose Inhaler Formulation

The primary aim of this research endeavor is to innovate a viable and user-friendly UV spectroscopic approach for quantifying formoterol and beclomethasone within metered dose inhalers (MDIs). These formulations, amalgamating Beclomethasone Dipropionate (BDP) and Formoterol Fumarate (FF), are extensively employed in managing bronchial asthma and chronic obstructive pulmonary diseases. Although analytical techniques already exist for assessing formoterol and beclomethasone in combination dry powder inhalers, this study concentrates on devising and validating a UV spectrophotometric method for simultaneously quantifying formoterol fumarate and Anhydrous Beclomethasone Dipropionate in Pressurized Metered Dose Inhalers (pMDIs). The estimation is performed at a wavelength of 240 nm for Beclomethasone Dipropionate and 216 nm for Formoterol Fumarate. Both drugs demonstrate satisfactory linearity within the concentration range of 10-30 μg/ml for BDP and 0.3 to 0.9 μg/ml for FF at wavelengths of 240 nm (r^2=0.999) and 216 nm (r^2=0.999), respectively. The Beer-Lambert law is adhered to within the concentration range of 10-30 μg/ml for BDP and 0.3-0.9 μg/ml for FF. The proposed methods are subjected to validation following ICH guidelines to assess accuracy, method precision, specificity, among other parameters. As a result, the proposed method can be effectively employed for the simultaneous determination of Beclomethasone Dipropionate and Formoterol Fumarate in routine analytical applications, offering a rapid and reliable solution for pharmaceutical analysis in the field.

A green perspective on simultaneous HPLC and UV spectrophotometric estimation of Udenafil and Dapoxetine hydrochloride in pharmaceutical formulations

Clinical research has affirmed that the combination of Udenafil and Dapoxetine hydrochloride yields greater improvement in erectile dysfunction compared to the use of individual medications. There is a requirement of precise quantitative method for accurate determination of Udenafil and Dapoxetine when taken together. Consequently, our interest lies in developing and validating analytical methods for the simultaneous estimation of Udenafil and Dapoxetine - Various strategies, such as UV spectrophotometric approaches and an innovative, straightforward, accurate, sensitive, and robust High-Performance Liquid Chromatography (HPLC) procedure, have been explored. An Isocratic HPLC approach was employed using reversed-phase CHEMSIL ODS C18 column (4.6 mm x 250 mm, 5 µm) as a stationary phase and detection wavelength (λ max) 230 nm for the two analytes in the chromatographic column. The mobile phase comprised of (70:30, v/v) mixture of Acetonitrile and 10 mM Ammonium formate buffer having pH 5, adjusted with formic acid. The elution was carried out at a constant flow rate of 1 mL/min. The UV simultaneous equation technique estimated the medicine's quantities and derived a simultaneous equation for DAP and UDN using 2 wavelengths, 230 nm and 298 nm. Furthermore, the two advanced methods underwent rigorous statistical validation, guided by the valuable ICH Q2R1 guidelines, affirming their effectiveness as a successful analytical tool for the concurrent analysis of both tablets in bulk and the synthetic mixture fabricated in the laboratory. To assess the environmental impact of contemporary UV spectrophotometric and HPLC techniques, the Green Analytical Greenness Metric (AGREE) software and the Green Analytical System Index (GAPI) were employed to evaluate the greenness profile. This examination confirmed the method's environmental friendliness concerning solvent usage, chemical ingredients, energy consumption, and waste production. The developed UV method is more sustainable and greener compare to HPLC method for simultaneous estimation of compound, which can be utilised by industry for quantification of each individual drug in their combination products.

Development of an effective cleaning technique and ancillary analytical method for estimation of residues of selected kinase inhibitors from stainless steel and glass surfaces by swab sampling

Importance of cleaning validation in the pharmaceutical industry cannot be overstated. It is essential for preventing cross-contamination, ensuring product quality & safety, and upholding regulatory standards. The present study involved development of an effective cleaning method for five selected kinase inhibitors binimetinib (BMT), selumetinib (SMT), brigatinib (BGT), capmatinib (CPT), and baricitinib (BRT). For checking the effectiveness of the developed cleaning technique, a sensitive and specific RP-HPLC based analytical method employing a diode array detector has been established to quantitate drug residue on glass and stainless steel surfaces. A reproducible swab sampling protocol utilizing TX714A Alpha swabs wetted with an extracting solvent has been developed to collect representative samples from both surfaces. Chromatographic separation of selected kinase inhibitors was achieved in gradient mode using an Agilent Zorbax eclipsed C18 column with acetonitrile and 10 mM ammonium formate as the mobile phase. The analytes were chromatographically separated in a 12 min run time. The mean swab recovery for each drug from glass and stainless steel surfaces exceeded 90%. Cleaning with IPA (70%) and acetone (70%) effectively removed residues for all five drugs. A solution comprising 10 mM SDS with 20% IPA demonstrated good efficacy in cleaning residues of BGT, BRT, and CPT, but exhibited lower efficacy for SMT and BMT.

ECO-FRIENDLY METHOD DEVELOPMENT AND VALIDATION FOR THE ESTIMATION OF FENOFIBRATE AND ATORVASTATIN IN MARKETED FORMULATION

This study focuses on the development and validation of an eco-friendly analytical method for the simultaneous estimation of Fenofibrate (FNF) and Atorvastatin (ATV) in a marketed formulation. The stability of both drugs was evaluated using a mixed hydrotropic solution of 2M Ammonium Acetate and 2M Sodium Citrate (1:1). This work focuses on the development and validation of an eco-friendly method for the simultaneous estimation of Fenofibrate and Atorvastatin in a marketed formulation. The stability of both drugs in a mixed hydrotropic solution was confirmed, and the method exhibited good linearity, recovery, and precision. The validated method was successfully applied to analyze a tablet formulation, demonstrating its applicability in routine quality control. The use of environmentally friendly practices in analytical methods contributes to sustainable pharmaceutical analysis. Development and Validation of an Analytical RP-HPLC Method for Simultaneous Estimation of Losartan and its Active Metabolite (EXP-3174) in Isolated Perfused Rat Liver

Background: The liver perfusion method is frequently used in drug pharmacokinetic studies and the various effects of drugs on liver tissue. The aim of this study was to establish and validate an analytical method using high-performance liquid chromatography to determine the simultaneous concentration of losartan and its active metabolite, EXP-3174, in an isolated perfused rat liver study. Method: An HPLC system with isocratic mode was used. Various chromatographic parameters were adjusted to develop and validate a method for determination of losartan and its active metabolite in liver perfusion media. Results: In this study, losartan and its active metabolite, EXP-3174, were separated using a C18 stationary phase, a mobile phase consisting of acetonitrile: phosphate buffer at a flow rate of 1 mL.min-1, and UV detection at 254 nm. Retention times for losartan and the metabolite were 10 and 16 minutes, respectively. Linearity from 25-250 ng.ml-1 was validated with acceptable accuracy and precision. The LOD and LOQ for losartan were 7.0 and 21.1 ng.ml-1, respectively. The LOD and LOQ for metabolite were 7.4 and 22.4 ng.ml-1, respectively. ChromGate® software was used to acquire and process the data. Conclusions: The optimized and validated technique was effectively used to analyze losartan and its active metabolite in isolated perfused rat liver.

Flavonoids are an important class of phytochemicals found to be present in plants and their derived products. Belamcanda chinensis (L.) DC has been used in Traditional Chinese medicine for multiple therapeutic purposes for a very long time. Belamcanda chinensis have been reported to have antipyretic, antidote, expectorant, antiphlogistic, and analgesic activity in the scientific fields. Scientific information on irisflorentin for its medicinal importance and pharmacological activity has been collected through electronic search engines, including Science Direct, Google, Google Scholar, PubMed, and Scopus, and analyzed in the present investigation in order to know the biological potential of irisflorentin. However, analytical aspects of irisflorentin are also discussed in the present work. Further detailed pharmacological activity data of irisflorentin have been collected and analyzed in the present work in order to know the therapeutic potential of irisflorentin in medicine. Scientific data analysis of different research work signified the biological importance and therapeutic benefit of Belamcanda chinensis and irisflorentin in medicine. Scientific data analysis revealed the biological effectiveness of irisflorentin against Parkinson's disease, inflammation, cancer, and brain disorders. Further scientific data analysis also signified the biological potential of irisflorentin for its α-glucosidase inhibitory potential and immunotherapeutic adjuvant in medicine. Metabolism and pharmacokinetic parameters have also been discussed in the present work. Analytical data showed the importance of various analytical methods for separation, identification and estimation of irisflorentin in different biological samples. Scientific data analysis of various research works signified the biological potential and therapeutic effectiveness of irisflorentin in medicine. However, clinical data of irisflorentin should be also investigated in order to know their pharmacokinetic parameters and safety issue in medicine.

Facile synthesis and eco-friendly analytical methods for concurrent estimation of selected pharmaceutical drugs in their solutions: application to quality by design, lean six sigma, and stability studies

Economical, highly robust, selective, precise, and eco-friendly RP-UPLC and spectrophotometric methods were developed and validated for the concurrent estimation of selected pharmaceutical drugs represented in ceftazidime (CFZ) and pyridine (PYD) in their solutions using Agilent Zorbax SB-C18 RRHD (50 × 2.1 mm, 1.8 μm) column at flow rate 0.3 mL/min with wavelength 254 nm. Box-Behnken design (BBD) established Response surface methodology (RSM) to achieve the optimum chromatographic condition with minimal trials conducted. Three independent variables specifically acetonitrile ratio 60–70%, pH 3–7, and temperature 25–35 °C were implemented to evaluate the influences of these variables on the responses as resolution and retention time. Desirability and overlay plots were carried out to adjust the optimal condition that achieved the shortest retention time of less than 2 min and desired resolution of more than 1.5 using a mobile phase consisting of acetonitrile: purified water (70:30, v/v) at pH 5.0 adjusted by 0.1% orthophosphoric acid with the column oven temperature 30 °C and column void volume 0.46 mL. Mean centering of ratio spectra (MCR) and ratio subtraction (RS) methods were effectively applied to resolve drugs' spectral superposition at 220 nm, 255.4 nm, 260.3 nm, and 254.6 nm for CFZ and PYD, respectively. Linearity range was accomplished for UPLC, MCR, and RS methods over the concentration range of 2–100, 1–50,3–30 and 5–30 µg/mL for CFZ and PYD, respectively with correlation coefficient > 0.999 and good recovery results within 98–102%. Six Sigma methodology was achieved using the process capability index (Cpk) to compare the suggested and USP methods showing that both are highly capable with Cpk > 1.33. Application of Quality Risk Assessment and Design of Experiment in Optimizing Chromatographic Method for Estimation of Total Sarsasapogenin from the Roots of <i>Asparagus racemosus</i> Wild

The roots of the plant Asparagus racemosus (Wild) (family: Liliaceae) are used alone as a lactogenic and general tonic or incorporated as a major ingredient in many herbal and Ayurveda formulations. Shatavarin glycosides containing sarsasapogenin as aglycone were found bioactive, and present in predominantly higher amount. The experiments were planned to develop an analytical method for estimation of total sarsasapogenin from the extract using HPTLC, through a systematic approach after identifying the potential failure modes, termed as critical process parameters to achieve the analytical target profile. The associated risks were mitigated by studying the combined effect of the critical process parameters on area, Rf, and tailing factor corresponding to the sarsasapogenin peak, assigned as critical method attributes. The optimization studies were carried out through Design of Experiment approach and process parameters were finalized to achieve analytical target profile. The developed method could resolve the peak of sarsasapogenin from the hydrolyzed methanolic extract prepared from the root powder using silica gel-backed pre-coated aluminum plates. The optimized mobile phase was Hexane:Ethyl acetate:Formic acid (7.0:1.9:0.35, v/v/v); the plates were subjected to post-chromatographic derivatization using 5% v/v hydrochloric acid solution, and heating of the plats for 3 min at 110˚C. The chromatogram was recorded by scanning the plates in TLC scanner using fluorescence mode at 366 nm. The developed analytical method was found to yield linear response in the range of 40-280 ng/spot. The limit of quantification was determined to be 35.88 ng/spot. The method was found selective, accurate, and robust. The amount of sarsasapogenin was found to be determined using the developed method and found to be 2.31% w/w on dried weight basis from the dried root powder. This is the first report of a systematic approach in estimating sarsasapogenin from Asparagus sp.

Development and validation of a stability indicating green analytical method for the simultaneous estimation of l-glutathione, n-acetyl l-cysteine and Vitamin C in marketed formulation using UV–visible spectroscopy

BackgroundAn efficient ultraviolet spectrophotometric method has been devised for the simultaneous analysis of l-glutathione, n-acetyl l-cysteine and Vitamin C in combined tablet dosage forms. This method is characterized by its simplicity, rapidity, precision, accuracy and cost-effectiveness. It employs the simultaneous equation method to determine the concentrations of l-glutathione, n-acetyl l-cysteine and Vitamin C.ResultsThe concentration range of 50–250 μg/mL for l-glutathione, 15–75 μg/mL for n-acetyl l-cysteine and 2–10 μg/mL for Vitamin C demonstrated linearity in this study. The simultaneous equation method was employed to determine the drug concentrations. The average recovery rates were found to be 99.94 ± 0.61% for l-glutathione, 100.90 ± 0.96 for n-acetyl l-cysteine and 99.81 ± 0.58% for Vitamin C. Stability studies were performed under various stress conditions.ConclusionThe method employed in this study was observed to possess qualities of simplicity, accuracy and precision, making it suitable for the simultaneous determination of l-glutathione, n-acetyl l-cysteine and Vitamin C in pharmaceutical tablet dosage forms. The analysis results were statistically validated and supported by recovery studies and stability studies, further enhancing the credibility of the method.

Development and validation of simultaneous quantification method for gemcitabine and betulinic acid: augmenting industrial application.

Combinatorial treatment utilizing a nucleoside analogue gemcitabine (GEM), with a characteristic pentacyclic triterpenoid betulinic acid (BET), has exhibited empowering adequacy in the therapy of cancer. It lessens the advancement of collagen and upgrades the saturation of tumour medicines. With the advancement in nanotechnology, the co-loaded formulation urges for a validated method of estimation. The purposed work entails a robust, simple, and economical analytical method for the simultaneous estimation of GEM and BET through RP-HPLC. Orthophosphoric acid (0.1%)-acetonitrile was considered as the mobile phase for the detection of GEM and BET at 248nm and 210nm with retention times of 5min and 13min, respectively. The method was further validated as per the regulatory guidelines with all the parameters found within the limit. The developed method with adequate resolution and quantification was found to be linear, accurate, precise, robust, and stable with an intra- and inter-day variability of less than 2%. The method was found specific for GEM and BET with no matrix interference of drug-spiked FBS samples. To demonstrate the applicability of the developed method, a nano-formulation containing GEM and BET was prepared and assessed for various parameters including encapsulation efficiency, loading efficiency, drug release, and drug stability. The method developed can be a possible tool for the simultaneous quantification of GEM-BET in analytical and biological samples.

Development and Validation of Analytical Method for Estimation of Berberine in Herbal Extract of Momordica Dioica

Background:While several anti-diabetic medications have been found to lower blood glucose levels, there are numerous negative effects connected with the use of currently marketed anti-diabetic medications.However, there are medicinal plants with anti-diabetic effects with low or no side effects. Among the Cucurbitaceae family of plants, Momordica dioica is a dioecious climbing herb. It contains many phytoconstituents.One of it is a quaternary ammonium salt of benzyl-isoquinoline alkaloid called as berberine exhibiting a wide variety of pharmaceutical properties.In the current investigation, the activities of berberine for its anti-diabetic potential in the Momorica Dioica plant.The goal of the recent effort is to establish and validate an HPTLC method that is quick, accurate, exact, and specific for determining berberine from herbal extract of Momordica dioica. Methodology/Conclusions/significance: For a quick examination to determine the amount of berberine, a (HPTLC) process was created as well as validated.On HPTLC aluminium plate 60 F254,precoated with silica gel chromatographic separation was accomplished using the solvents: methanol ,ethyl acetate, toluene and formic acid (2:4:3:0.5). A wavelength of 348 nm was used for detection. Rf was found to be 0.44±5% for berberine. There was linearity for berberine in the 400ng/band concentration range.The LOD and LOQ was found to be0.0096ng/band and 0.0293ng/ band for Berberine.The average berberine recovery rate was (0.041).The method's precision, accuracy, linearity, robustness, and specificity were all authenticated in accordance with ICH guidelines. The discovered approach can be used to regularly analyse Berberine in herbal formulations for quality control.

An Adaptive and Robust Deep Learning Framework for THz Ultra-Massive MIMO Channel Estimation

Terahertz ultra-massive MIMO (THz UM-MIMO) is envisioned as one of the key enablers of 6G wireless networks, for which channel estimation is highly challenging. Traditional analytical estimation methods are no longer effective, as the enlarged array aperture and the small wavelength result in a mixture of far-field and near-field paths, constituting a hybrid-field channel. Deep learning (DL)-based methods, despite the competitive performance, generally lack theoretical guarantees and scale poorly with the size of the array. In this paper, we propose a general DL framework for THz UM-MIMO channel estimation, which leverages existing iterative channel estimators and is with provable guarantees. Each iteration is implemented by a fixed point network (FPN), consisting of a closed-form linear estimator and a DL-based non-linear estimator. The proposed method perfectly matches the THz UM-MIMO channel estimation due to several unique advantages. First, the complexity is low and adaptive. It enjoys provable linear convergence with a low per-iteration cost and monotonically increasing accuracy, which enables an adaptive accuracy-complexity tradeoff. Second, it is robust to practical distribution shifts and can directly generalize to a variety of heavily out-of-distribution scenarios with almost no performance loss, which is suitable for the complicated THz channel conditions. For practical usage, the proposed framework is further extended to wideband THz UM-MIMO systems with beam squint effect. Theoretical analysis and extensive simulation results are provided to illustrate the advantages over the state-of-the-art methods in estimation accuracy, convergence rate, complexity, and robustness.

Exploring Energy Retrofitting Strategies and Their Effect on Comfort in a Vernacular Building in a Dry Mediterranean Climate

This research explores the energy behaviour of a traditional house on the Mediterranean coast of south-eastern Spain. The objective of the work is to determine the optimal passive strategies for rehabilitating a traditional house, improving its energy savings and comfort, considering the characteristics of the warm semi-arid Mediterranean climate. The main novelty of this article is that it demonstrates that the limits imposed by current regulations, based on globalised climate strategy approaches, undermine the energy efficiency capacity that passive solutions in vernacular architecture already employed. The methodology used consists of a systematised multi-objective study of various energy rehabilitation strategies. Four strategies were studied: raising the thermal insulation of enclosures, improving thermal insulation and solar control glazing with movable shading devices, increasing the size of windows and introducing the use of natural ventilation enhanced by ceiling fans. The results show that simultaneous improvement of these parameters reduces cooling and heating requirements by up to 87%, reducing the energy consumption of air conditioning systems. Indoor temperatures are also maintained within the comfort limits set by regulations for 91% of hours per year without the need for air conditioning systems. This results in a passive energy-efficient and comfortable house almost all year round. This work offers an alternative solution to the comfort standards of current Spanish regulations and demonstrates the need to adapt Fanger’s analytical method for comfort estimation. The research concludes that the comfort criteria of current energy regulations should be modified to better adapt the design criteria to the dry Mediterranean climate.

Dissipation kinetics and dietary risk assessment of pesticides in/on citrus (mandarin)

AbstractA supervised field trial was conducted to evaluate the dissipation pattern and dietary risk assessment of the pesticides, Solomon (recommended dose, X (63+147 g a.i./ha) and double the recommended dose, 2X (126+294 g a.i./ha)); spirotetramat (X dose: 90 g a.i./ha and 2X dose: 180 g a.i./ha) and indaziflam (X dose: 62.5 g a.i./ha and 2X dose: 125 g a.i./ha) in citrus (mandarin) fruits and cropped soil. Solomon and spirotetramet were applied as foliar spray at the fruit development stage and indaziflam was applied directly on to the weeds. Fruit and soil samples thus collected were extracted using a quick, easy, cheap, effective, rugged, and safe analytical method for residue estimation, which was carried out using Gas Chromatography‐Electron Capture Detector for betacyfluthrin and High‐Performance Liquid Chromatography for all other analytes. Residues of betacyfluthrin reached below the determination level (0.05 mg/kg) after the 3rd and 5th day of treatment at X and 2X dosage while imidacloprid and spirotetramat residues reached below the determination level on the 5th and 7th day, respectively. Residues of indaziflam and its metabolites were not detected in any citrus and soil matrices. Dietary risk assessment calculated as hazard quotient and theoretical maximum dietary intake revealed the safety of the pesticides for human consumption after application of these pesticides on citrus.

RP-HPLC Based Stability Indicating Assay of Evogliptin and Metformin: Development and Validation of the Analytical Method

Background: Several new fixed dose combinations have been recently approved by the Food and Drug Administration for treatment of disease conditions. All these combinations require specific, accurate and precise analytical methods for estimation for simultaneous estimation.
 Methods: A new RP-HPLC method was developed for the stability indication of evogliptin and metformin in tablets and it was validated as per ICH guidelines. The chromatogram for was found to be satisfactory on symmetry C-18 (4.6×150mm, 5µ Kromasil column) using phosphate buffer (pH 4.5) and acetonitrile in the ratio of 30:70 v/v at a flow rate of 0.8 ml/min.
 Results: The retention time of evogliptin and metformin were found to be 7.03 min and 9.50 min respectively. The system suitability parameters proved that the proposed method was found to be suitable for stability indication of evogliptin and metformin. Tailing factor for the peak was found to be 1.003 and 1.258. The method was found to be linear in the range of 10-50µg/ml for both the drugs. The precision of the method was good and the recovery of drugs is well within the acceptance limits of 80-120%. The LOD and LOQ was found to be 0.003 µg/ml, 0.12 µg/ml and for evogliptin and 0.09 µg/ml and 0.03 µg/ml for metformin.
 Conclusion: A simple, accurate and precise method tor routine analysis of the combination of evogliptin and metformin was developed and was applicable for the stability analysis of the combination.

Methods to reduce residual welding stresses in mining excavator metal structures

Residual stresses in metal structures of mining machinery caused by welding occur due to simultaneous uneven heating and cooling of local sections of structures, variable cooling rates in the different areas of the weld and deformations caused by metallurgical phase transformations. Residual stress in the welded joint can significantly increase the external load that can lead to structural failure. Research on the calculation, measurement and relief of residual stresses under welding is an important issue when predicting the service life of metal structure units of mining machinery and equipment. An accurate quantitative estimation of residual stresses in welded products, repair welds included, as well as the search for the fundamental principles of the elimination methods of residual deformations are of considerable practical interest, which is relevant for the authors of this paper. The study involved a general review of the latest researches in the field of estimation and measurement of residual stresses caused by electrogas welding. It has been proposed to use various techniques and analytical methods for quantitative estimation of welded joint residual stresses on the basis of fracture mechanics, which enabled to take preventative measures at early stages to reduce the cost of repair and maintenance of welded metal structures of mining excavators. The conducted study resulted in formulating the proposals on relief of residual welding stresses in the metal structures of mining excavators.
