Different strategies and bio-removal mechanisms of petroleum hydrocarbons from contaminated sites

PurposePetroleum hydrocarbons are naturally occurring flammable fossil fuels used as conventional energy sources. It has carcinogenic, mutagenic properties and is considered a hazardous pollutant. Soil contaminated with petroleum hydrocarbons adversely affects the properties of soil. This paper aim to remove pollutants from the environment is an urgent need of the hour to maintain the proper functioning of soil ecosystems.Design/methodology/approachThe ability of micro-organisms to degrade petroleum hydrocarbons makes it possible to use these microorganisms to clean the environment from petroleum pollution. For preparing this review, research papers and review articles related to petroleum hydrocarbons degradation by micro-organisms were collected from journals and various search engines.FindingsVarious physical and chemical methods are used for remediation of petroleum hydrocarbons contaminants. However, these methods have several disadvantages. This paper will discuss a novel understanding of petroleum hydrocarbons degradation and how micro-organisms help in petroleum-contaminated soil restoration. Bioremediation is recognized as the most environment-friendly technique for remediation. The research studies demonstrated that bacterial consortium have high biodegradation rate of petroleum hydrocarbons ranging from 83% to 89%.Social implicationsProper management of petroleum hydrocarbons pollutants from the environment is necessary because of their toxicity effects on human and environmental health.Originality/valueThis paper discussed novel mechanisms adopted by bacteria for biodegradation of petroleum hydrocarbons, aerobic and anaerobic biodegradation pathways, genes and enzymes involved in petroleum hydrocarbons biodegradation.

Anaerobic biodegradation of anthracene by oral Firmicutes isolates from smokers and its potential pathway

Exposure to polycyclic aromatic hydrocarbons (PAHs) from tobacco smoke has been linked to many negative health effects. Studies on the biodegradation of PAHs by human microbiota and detailed pathways for their anaerobic biodegradation are scarce despite their importance in getting rid of these toxic compounds. In a previous study for our group, we determined the ability of oral bacterial isolates in the anaerobic biodegradation of anthracene as a model of PAHs. Three isolates with the highest anthracene degradation ability were selected for the present study which include Limosilactobacillus fermentum, Veillonella parvula, and Streptococcus anginosus. In this study, we aimed at exploring and elucidating the anthracene anaerobic biodegradation pathways in selected Firmicutes oral isolates. Metabolites throughout the pathway were detected by gas chromatography coupled with mass spectroscopy (GC-MS) using anthracene as sole source of carbon. After incubation for 3 days, anthracene was undetected in the supernatant of L. fermentum and V. parvula, while a residual of 3% of anthracene was detected in presence of S. anginosus. Results revealed that anaerobic biodegradation by L. fermentum and V. parvula started with hydroxylation and dehydrogenation producing 9,10- anthraquinone and ended up with simpler structures such as catechol, while S. anginosus hydroxylation for anthracene resulted in the production of 1,2-anthracenediol and ended up with catechol and phthalic acid. The biodegradation of anthracene by oral bacteria could convert it to other toxic metabolites such as anthraquinone and catechol which were reported to have potential carcinogenic effects. Moreover, fatty acids detected as biodegradation metabolites could be one of the causes of smokers’ heart-related diseases. Thus, this study explored oral metabolites resulting from smoking under anaerobic conditions towards elucidating the role of oral microbiota in health and disease states.

Multiple pathways for the anaerobic biodegradation of microcystin-LR in the enriched microbial communities from Lake Taihu

Anaerobic biodegradation is a non-negligible elimination approach for microcystin (MC) pollution and exhibits important bioremediation potential for environmental problems. However, the specific anaerobic MC-degrading mechanism remains unclear and few functional bacteria have been found. In this study, three microbial communities of sludges from different locations in Lake Taihu were collected and further enriched by microcystin-LR (MC-LR) under anaerobic conditions. MC-LR (1 mg/L) could be completely degraded by these enriched microbial communities under anaerobic conditions, but their degradation rates were significantly different. In addition, two different ring-opening sites of MC-LR in Ala-Leu and Arg-Adda were observed, and three new anaerobic degradation products were first identified, including two hexapeptides (MeAsp-Arg-Adda-Glu-Mdha-Ala and Adda-Glu-Mdha-Ala-Leu-MeAsp) and one end-product pentapeptide (Glu-Mdha-Ala-Leu-MeAsp). Based on the chemical structures and temporal trends of all detected degradation products, two novel anaerobic biodegradation pathways of MC-LR were proposed. Moreover, the MC-degrading genes mlrABC were not detected among all microbial communities, which suggested that some new MC-degrading mechanisms might exist under anaerobic conditions. Finally, through the comparison of microbial community structure, Gemmatimonas and Smithella were deduced as possible anaerobic MC-degrading bacteria. These findings strongly indicate that anaerobic biodegradation is an important method of self-repair in the natural environment and provides a potential removal strategy for MC pollution.

The dissolution and microbial degradation of mobile aromatic hydrocarbons from a Pintsch gas tar DNAPL source zone

Source zones containing tar, a dense non-aqueous phase liquid (DNAPL), can contaminate groundwater for centuries. A common occurrence of tar is at former Pintsch gas factories. Little is known about the composition and fate of contaminants dissolving from Pintsch gas tar DNAPL. In this study, we determined the composition and water-soluble characteristics of mobile aromatic hydrocarbons and their biodegradation metabolites in the DNAPL contaminated groundwater at a former Pintsch gas tar plant. We assessed the factors that determine the fate of observed groundwater contaminants. Measured values of density (1.03–1.06 kg/m3) and viscosity (18.6–39.4 cP) were found to be relatively low compared to common coal tars. Analysis showed that unlike common coal tars phenanthrene is the primary component rather than naphthalene. Moreover, it was found that Pintsch gas tar contains a relatively high amount of light molecular aromatic hydrocarbon compounds, such as benzene, toluene, ethylbenzene and xylenes (BTEX). Less commonly reported components, such as styrene, ethyltoluenes, di-ethylbenzene, 1,2,4,5-tetramethylbenzene, were also detected in water extracts from Pintsch gas tar. Moreover, 46 relatively hydrophilic metabolites were found within the tar samples. Metabolites present within the tar suggest biodegradation of mobile aromatic Pintsch gas tar compounds occurred near the DNAPL. Based on eleven detected suspect metabolites, a novel anaerobic biodegradation pathway is proposed for indene. Overall, our findings indicate that Pintsch gas tar has higher invasive and higher flux properties than most coal tars due to its relatively low density, low viscosity and, high content of water-soluble compounds. The partitioning of contaminants from multi-component DNAPL into the aqueous phase and re-dissolution of their slightly less hydrophobic metabolites back from the aqueous phase into the DNAPL is feasible and demonstrates the complexity of assessing degradation processes within a source zone.

Anaerobic biodegradation of 8:2 fluorotelomer alcohol in anaerobic activated sludge: Metabolic products and pathways

The anaerobic biodegradability and metabolic pathways of 8:2 fluorotelomer alcohol (8:2 FTOH) were investigated in anaerobic activated sludge. The biodegradation was well described by a double exponential decay model. 8:2 FTOH was biodegraded to poly- and perfluorinated metabolites with the release of fluoride ion. All polyfluorinated metabolites were intermediate metabolic products and could be further transformed to other metabolites, while perfluorinated metabolites were terminal products. 2H-perfluoro-2-decenoic acid (8:2 FTUA) and perfluorooctanoic acid (PFOA) were verified as the most abundant poly- and perfluorinated metabolites, respectively. Two shorter-chain perfluorinated metabolites, perfluoropentanoic acid (PFPeA) and perfluorobutyric acid (PFBA), were first reported in the biodegradation of 8:2 FTOH. However, the total molar recovery of 8:2 FTOH decreased with increasing incubation time, indicating that there might be some unknown metabolites. Thus, the anaerobic biodegradation pathways were proposed as follows: 8:2 FTOH was oxidized to 8:2 FTUA and 2-perfluorooctyl ethanoic acid (8:2 FTCA) via 2-perfluorooctyl acetaldehyde (8:2 FTAL), and then 8:2 FTUA and 8:2 FTCA were further transformed to 1-perfluoroheptyl ethanol (7:2 sFTOH) via 3-perfluoroheptyl propionic acid (7:3 acid) or/and 3-perfluoroheptyl acrylic acid (7:3 Uacid), and eventually 7:2 sFTOH was further biodegraded to PFOA and other perfluorocarboxylates containing less than eight carbons.

Distinct Dual C-Cl Isotope Fractionation Patterns during Anaerobic Biodegradation of 1,2-Dichloroethane: Potential To Characterize Microbial Degradation in the Field.

This study investigates, for the first time, dual C-Cl isotope fractionation during anaerobic biodegradation of 1,2-dichloroethane (1,2-DCA) via dihaloelimination by Dehalococcoides and Dehalogenimonas-containing enrichment cultures. Isotopic fractionation of 1,2-DCA (εbulkC and εbulkCl) for Dehalococcoides (-33.0 ± 0.4‰ and -5.1 ± 0.1‰) and Dehalogenimonas-containing microcosms (-23 ± 2‰ and -12.0 ± 0.8‰) resulted in distinctly different dual element C-Cl isotope correlations (Λ = Δδ13C/Δδ37Cl ≈ εbulkC/εbulkCl), 6.8 ± 0.2 and 1.89 ± 0.02, respectively. Determined isotope effects and detected products suggest that the difference on the obtained Λ values for biodihaloelimination could be associated with a different mode of concerted bond cleavage rather than two different reaction pathways (i.e., stepwise vs concerted). Λ values of 1,2-DCA were, for the first time, determined in two field sites under reducing conditions (2.1 ± 0.1 and 2.2 ± 2.9). They were similar to the one obtained for the Dehalogenimonas-containing microcosms (1.89 ± 0.02) and very different from those reported for aerobic degradation pathways in a previous laboratory study (7.6 ± 0.1 and 0.78 ± 0.03). Thus, this study illustrates the potential of a dual isotope analysis to differentiate between aerobic and anaerobic biodegradation pathways of 1,2-DCA in the field and suggests that this approach might also be used to characterize dihaloelimination of 1,2-DCA by different bacteria, which needs to be confirmed in future studies.

Insights into the Anaerobic Biodegradation Pathway of n-Alkanes in Oil Reservoirs by Detection of Signature Metabolites.

Anaerobic degradation of alkanes in hydrocarbon-rich environments has been documented and different degradation strategies proposed, of which the most encountered one is fumarate addition mechanism, generating alkylsuccinates as specific biomarkers. However, little is known about the mechanisms of anaerobic degradation of alkanes in oil reservoirs, due to low concentrations of signature metabolites and lack of mass spectral characteristics to allow identification. In this work, we used a multidisciplinary approach combining metabolite profiling and selective gene assays to establish the biodegradation mechanism of alkanes in oil reservoirs. A total of twelve production fluids from three different oil reservoirs were collected and treated with alkali; organic acids were extracted, derivatized with ethanol to form ethyl esters and determined using GC-MS analysis. Collectively, signature metabolite alkylsuccinates of parent compounds from C1 to C8 together with their (putative) downstream metabolites were detected from these samples. Additionally, metabolites indicative of the anaerobic degradation of mono- and poly-aromatic hydrocarbons (2-benzylsuccinate, naphthoate, 5,6,7,8-tetrahydro-naphthoate) were also observed. The detection of alkylsuccinates and genes encoding for alkylsuccinate synthase shows that anaerobic degradation of alkanes via fumarate addition occurs in oil reservoirs. This work provides strong evidence on the in situ anaerobic biodegradation mechanisms of hydrocarbons by fumarate addition.

Pathway-Dependent Isotope Fractionation during Aerobic and Anaerobic Degradation of Monochlorobenzene and 1,2,4-Trichlorobenzene

Stable carbon isotope fractionation is a valuable tool for monitoring natural attenuation and to establish the fate of groundwater contaminants. In this study, we measured carbon isotope fractionation during aerobic and anaerobic degradation of two chlorinated benzenes: monochlorobenzene (MCB) and 1,2,4-trichlorobenzene (1,2,4-TCB). MCB isotope fractionation was measured in anaerobic methanogenic microcosms, while 1,2,4-TCB isotope experiments were carried out in both aerobic and anaerobic microcosms. Large isotope fractionation was observed in both the anaerobic microcosm experiments. Enrichment factors (ε) for anaerobic reductive dechlorination of MCB and 1,2,4-TCB were -5.0‰ ± 0.2‰ and -3.0‰ ± 0.4‰, respectively. In contrast, no significant isotope fractionation was found during aerobic microbial degradation of 1,2,4-TCB. The cleavage of a C-Cl σ bond occurs during anaerobic reductive dechlorination of MCB and 1,2,4-TCB, while no σ bond cleavage is involved during aerobic degradation via dioxygenase. The difference in isotope fractionation for aerobic versus anaerobic biodegradation of MCB and 1,2,4-TCB can be explained by the difference in the initial step of aerobic versus anaerobic biodegradation pathways.

Anaerobic degradation behavior of nonylphenol polyethoxylates in sludge

Anaerobic biodegradation behavior of nonylphenol polyethoxylates (NPEOs) was investigated. Results showed that terminal electron acceptors, organic matters, initial concentration, and temperature had great influence on the anaerobic biodegradation of NPEOs. Anaerobic biodegradation of NPEOs could be enhanced by adding sulfate or nitrate while this process could be inhibited by adding organic matters. The maximum removal rate increased 1.24 μM d −1 for each ten micromoles increase in initial concentration. The decrease in temperature caused a sharp decrease in the removal efficiency of NPEOs. The temperature coefficient ( Ф) for the anaerobic biodegradation of NPEOs was 0.01 °C −1. Nonylphenol (NP), the typical intermediate of NPEOs, could inhibit the anaerobic biodegradation of NPEOs only at high concentration. However, these environmental factors had no effect on the anaerobic biodegradation pathway of NPEOs. The accumulation of NP and short-chain NPEOs during NPEO biodegradation led to a significant increase in the estrogenic activity during the biodegradation period.

Anaerobic biodegradability of Tween surfactants used as a carbon source for the microbial reductive dechlorination of hexachlorobenzene

Three structurally-related, nonionic, polysorbate surfactants (Tween 60, 61, and 65) were used as the sole carbon source to sustain the microbial, sequential reductive dechlorination of hexachlorobenzene (HCB) in a mixed, methanogenic culture derived from a contaminated estuarine sediment. The surfactants were partially degraded and fermented to methane with no measurable accumulation of volatile fatty acids, indicating that methanogenesis was rapid relative to the rates of hydrolysis and acidogenesis. Addition of the methanogenesis inhibitor 2-bromoethanesulfonic acid resulted in acetate accumulation without impact on the sequential dechlorination of HCB. An anaerobic biodegradability assay was performed and the following data were obtained for the Tween 60, 61, and 65, respectively: 53, 62, and 62% COD destruction; 35, 57, and 48% COD to methane conversion; and 38, 38, and 45% COD to acetate conversion. These data suggest that the hydrophobic moiety (stearate) of the surfactants was preferentially degraded, most likely through beta-oxidation, to acetate and ultimately to methane and carbon dioxide. Between 38 and 47% of the initial surfactant COD remained after 46 d incubation, which most likely corresponds to the hydrophilic polyoxyethylene moiety. An anaerobic biodegradation pathway of the Tween surfactants is proposed.

Carbon and hydrogen isotopic fractionation during anaerobic biodegradation of benzene.

Compound-specific isotope analysis has the potential to distinguish physical from biological attenuation processes in the subsurface. In this study, carbon and hydrogen isotopic fractionation effects during biodegradation of benzene under anaerobic conditions with different terminal-electron-accepting processes are reported for the first time. Different enrichment factors (epsilon ) for carbon (range of -1.9 to -3.6 per thousand ) and hydrogen (range of -29 to -79 per thousand ) fractionation were observed during biodegradation of benzene under nitrate-reducing, sulfate-reducing, and methanogenic conditions. These differences are not related to differences in initial biomass or in rates of biodegradation. Carbon isotopic enrichment factors for anaerobic benzene biodegradation in this study are comparable to those previously published for aerobic benzene biodegradation. In contrast, hydrogen enrichment factors determined for anaerobic benzene biodegradation are significantly larger than those previously published for benzene biodegradation under aerobic conditions. A fundamental difference in the previously proposed initial step of aerobic versus proposed anaerobic biodegradation pathways may account for these differences in hydrogen isotopic fractionation. Potentially, C-H bond breakage in the initial step of the anaerobic benzene biodegradation pathway may account for the large fractionation observed compared to that in aerobic benzene biodegradation. Despite some differences in reported enrichment factors between cultures with different terminal-electron-accepting processes, carbon and hydrogen isotope analysis has the potential to provide direct evidence of anaerobic biodegradation of benzene in the field.
