An electrochemiluminescence microsensor based on DNA-silver nanoclusters amplification for detecting cellular adenosine triphosphate.

Adenosine triphosphate (ATP), as the primary energy source, plays vital roles in many cellular events. Developing an efficient assay is crucial to rapidly evaluate the level of cellular ATP. A portable and integrated electrochemiluminescence (ECL) microsensor array based on a closed bipolar electrode (BPE) was presented. In the BPE unit, the ECL chemicals and oxidation/reduction were separated from the sensing chamber. The ATP aptamer was assembled with single-stranded DNA (ssDNA) in the sensing chamber. ATP capture made the aptamer disassemble from the ssDNA and facilitated DNA-templated silver nanocluster (Ag NC) generation by the target-rolling circle amplification (RCA) reaction. The guanine-rich padlock sequence produced tandem periodic cytosine-rich sequences by the RCA, inducing Ag NC generation in the cytosine-rich region of the produced DNA strands through Ag+ reduction. The in situ Ag NC generation enhanced the circuit conductivity of the BPE and promoted the ECL reaction of [Ru(bpy)2dppz]2+/tripropylamine in the anodic reservoir. On this ECL microsensor, a good linear relationship of ATP was achieved ranging from 30 to 1000 nM. The ATP content in HepG2 cells was selectively and sensitively determined without complex pretreatment. The ATP amount of 25 cells could be successfully detected when a sub-microliter sample was loaded.

Urinary ATP may be a biomarker of interstitial cystitis/bladder pain syndrome and its severity.

Urinary tract cells respond to bladder distension by releasing adenosine triphosphate (ATP). Patients with interstitial cystitis/bladder pain syndrome (IC/BPS) exhibit elevated urinary ATP levels compared to asymptomatic controls. This study aimed to evaluate the potential of urinary ATP as a non-invasive biomarker for IC/BPS and its correlation with symptom severity. We included 56 patients diagnosed with IC/BPS and 50 asymptomatic controls. Urine samples were collected from both groups. Urinary ATP levels were quantified using the luciferin-luciferase bioluminescence method. The severity of IC/BPS symptoms was assessed using the visual analogue score (VAS), Interstitial Cystitis Symptom Index (ICSI), and Interstitial Cystitis Problem Index (ICPI) from the O'Leary-Sant score. We specifically examined the correlation between symptom scores and urinary ATP levels in IC/BPS patients. Urinary ATP levels were significantly higher in IC/BPS patients compared to the control group (P < 0.0001). There was a significant positive correlation between urinary ATP concentrations and VAS, ICPI, and ICSI scores among IC/BPS patients (P < 0.0001). The threshold value for ATP concentration was set at 56.6 nM, with an area under the receiver operating characteristic (ROC) curve of 0.811 (95% CI 0.730 - 0.892). Our findings indicate that IC/BPS patients excrete elevated amounts of ATP in their urine. This suggests that urinary ATP might serve as a non-invasive biomarker for IC/BPS, with a predictive potential in terms of symptom severity.

THIAMINE AND HIGH DOSE INSULIN TREATMENT FOR SEPSIS

Sepsis is a major health problem and accounts for 20% of deaths worldwide. It is the most expensive condition treated in United States hospitals at $62 billion per year or about $46,000 per patient. Treatment consisting largely of fluid resuscitation and antibiotics has only a marginal impact. Mortality is about 27% for hospitalised patients and about 42% for patients in intensive care. There are two phases of sepsis – a hyperinflammatory phase and a subsequent hypoinflammatory phase. During the hyperinflammatory phase, the metabolic rate increases, and this is associated with an increase in body temperature and a rapid escalation of immune system functioning including increased numbers of leucocytes and their migration to infected and damaged tissues and increased supply and consumption of glucose to fuel this immune system. During the subsequent hypoinflammatory phase, the metabolic rate decreases, and this is associated with a decrease in body temperature and a generalised decrease in the physiological activity of many organs including the immune system akin to hibernation. The activated immune system has priority for the available glucose over most other organs and physiological functions during such potentially life-threatening circumstances. Thus, adenosine triphosphate (ATP) production by mitochondria (the source of energy at the cellular level for the organism as a whole) also has a lower priority for the available glucose relative to the activated immune system. If glucose availability is threatened, then the mitochondrial production of ATP is partially or substantially suppressed in favour of glycolysis because glycolysis can rapidly produce large quantities of ATP that are necessary for immune cell function in infected, anaerobic, ischaemic, or damaged tissues. However, glycolysis is only a temporary fix as it cannot produce the quantities of ATP necessary on an ongoing basis for the normal functioning of the healthy animal. Mitochondrial production of ATP must be recommenced for full recovery. It appears that the partial or substantial suppression of mitochondrial production of ATP by activation of the immune response becomes relatively fixated in some patients, leading to a substantial ATP deficit. This is the fundamental issue of sepsis. This paper reviews the metabolism of glucose and insulin during sepsis and concludes that high dose insulin with mild hyperglycaemia in conjunction with the intravenous administration of thiamine, an inhibitor of the pyruvate dehydrogenase kinase enzymes, to re-establish physiological ATP production by mitochondria, administered early in the hypometabolic (hypoinflammatory) phase of sepsis, may enhance survival relative to thiamine alone.

Advances in mitophagy and mitochondrial apoptosis pathway-related drugs in glioblastoma treatment.

Glioblastoma (GBM) is the most common malignant tumor of the central nervous system (CNS). It is a leading cause of death among patients with intracranial malignant tumors. GBM exhibits intra- and inter-tumor heterogeneity, leading to drug resistance and eventual tumor recurrence. Conventional treatments for GBM include maximum surgical resection of glioma tissue, temozolomide administration, and radiotherapy, but these methods do not effectively halt cancer progression. Therefore, development of novel methods for the treatment of GBM and identification of new therapeutic targets are urgently required. In recent years, studies have shown that drugs related to mitophagy and mitochondrial apoptosis pathways can promote the death of glioblastoma cells by inducing mitochondrial damage, impairing adenosine triphosphate (ATP) synthesis, and depleting large amounts of ATP. Some studies have also shown that modern nano-drug delivery technology targeting mitochondria can achieve better drug release and deeper tissue penetration, suggesting that mitochondria could be a new target for intervention and therapy. The combination of drugs targeting mitochondrial apoptosis and autophagy pathways with nanotechnology is a promising novel approach for treating GBM.This article reviews the current status of drug therapy for GBM, drugs targeting mitophagy and mitochondrial apoptosis pathways, the potential of mitochondria as a new target for GBM treatment, the latest developments pertaining to GBM treatment, and promising directions for future research.

Structures of water on the surface of anatase TiO2 studied by diffuse reflectance near-infrared spectroscopy

Investigating the structures of water on metal oxides is helpful for understanding the mechanism of the adsorptions in the presence of water. In this work, the structures of adsorbed water molecules on anatase TiO2 (101) were studied by diffuse reflectance near-infrared spectroscopy (DR-NIRS). With resolution enhanced spectrum by continuous wavelet transform (CWT), the spectral features of adsorbed water at different sites were found. In the spectrum of dried TiO2 powder, there is only the spectral feature of the water adsorbed at 5-coordinated titanium atoms (Ti5c). With the increase of the adsorbed water, the spectral feature of the water at 2-coordinated oxygen atoms (O2c) emerges first, and then that of the water interacting with the adsorbed water can be observed. When adenosine triphosphate (ATP) was adsorbed on TiO2, the intensity of the peaks related to the adsorbed water decreases, indicating that the adsorbed water is replaced by ATP due to the strong affinity to Ti5c. Therefore, there is a clear correlation between the peak intensity of the adsorbed water and the adsorbed quantity of ATP. Water can be a NIR spectroscopic probe to detect the quantity of the adsorbed ATP. A partial least squares (PLS) model was established to predict the content of adsorbed ATP by the spectral peaks of water. The recoveries of validation samples are in the range of 92.00–114.96% with the relative standard deviations (RSDs) in a range of 2.13–5.82%.

The ATP-dependent Pathways and Human Diseases.

Adenosine triphosphate (ATP) is one of the most important molecules of life, present both inside the cells and extracellularly. It is an essential building block for nucleic acids biosynthesis and crucial intracellular energy storage. However, one of the most interesting functions of ATP is the role of a signaling molecule. Numerous studies indicate the involvement of ATP-dependent pathways in maintaining the proper functioning of individual tissues and organs. Herein, the latest data indicating the ATP function in the network of intra- and extracellular signaling pathways including purinergic signaling, MAP kinase pathway, mTOR and calcium signaling are collected. The main ATP-dependent processes maintaining the proper functioning of the nervous, cardiovascular and immune systems, as well as skin and bones, are summarized. The disturbances in the ATP amount, its cellular localization, or interaction with target elements may induce pathological changes in signaling pathways leading to the development of serious diseases. The impact of an ATP imbalance on the development of dangerous health dysfunctions such as neurodegeneration diseases, cardiovascular diseases (CVDs), diabetes mellitus, obesity, cancers and immune pathogenesis are discussed here.

Measures against Infection in Facilities Using Air Catalysts

Background: The global outbreak of COVID-19 has affected the operations of healthcare field and various infection control measures are being carried out at each hospital and clinic.&#x0D; Aim: We have administered infection control measures using air catalysts which is a new antiviral and antibacterial technology being employed at our healthcare center.&#x0D; Methodology: We used a product called Health Bright® and this liquid substance was sprayed to coat every part of the facility. After that, we measured the amount of adenosine triphosphate (ATP) in coated places, including the waiting room, examination room and endoscopy room.&#x0D; Results: The amount of ATP decreased the day after the treatment in each room, and of note is that the amount of ATP was still low after 6 months and 12 months.&#x0D; Conclusion: Our current data showed the possible efficacy of air catalysts against bacteria and virus in the healthcare facility.

Double manual versus automated cleaning of loaner depth gauges used in clinical practice

Background: Automated cleaning is recommended for reprocessing complex design surgical instruments, as it is reproducible and cleaning parameters can be controlled. However, automated equipment may not be a reality for many hospitals, particularly in lower-middle income countries. Objective: The aim of this study was to compare the effectiveness of double manual cleaning and automated cleaning of depth gauges in use in clinical practice and supplied in a loaner system. Design: Twenty four depth gauges available for use in a loaner system were evaluated before double manual cleaning (Group 1) or immediately after double manual cleaning (Group 2), or automated thermal disinfector cleaning (Group 3) or automated ultrasonic cleaning (Group 4). Thereafter, the depth gauges in each group were analysed by visual inspection (n = 24), bacterial culture (n = 12), and adenosine triphosphate (ATP) test (n = 12). Results: Stains, grooves, oxidation or visible debris were detected on at least one of the depth gauges from each group, and most were positive for bacterial growth (n = 11/12). Cleaning methods significantly reduced the amount of ATP (P &lt; 0.05), except for automated ultrasonic cleaning. Conclusions: Double manual cleaning of depth gauges was similar to automated cleaning in a thermal disinfector, suggesting the possibility for implementing double manual cleaning as an alternative in sterilising service units where automated cleaning equipment is not avaliable.

Zeolitic imidazolate framework-8 encapsulating gold nanoclusters and carbon dots for ratiometric fluorescent detection of adenosine triphosphate and cellular imaging

A novel nanoprobe was prepared by encapsulating carbon dots (CDs) and gold nanoclusters (AuNCs) into zeolitic imidazolate framework-8 (ZIF-8) for sensitive detecting adenosine triphosphate (ATP). Under excitation at 360 nm, the obtained CDs/AuNCs@ZIF-8 nanoprobe exhibits dual-emissions at 469 nm and 660 nm, respectively, corresponding to the fluorescence emission of CDs and the aggregation-induced emission enhancement (AIEE) of AuNCs. The framework of ZIF-8 in this probe can be degraded by ATP due to the coordination competition of ATP and 2-Methylimidazole towards zinc ion (Zn2+), resulting in the release of CDs and AuNCs. The following dispersion of CDs would improve efficiencies of the fluorescence excitation and the consequent emission of CDs. On the contrary, the AIEE of AuNCs would be decreased spontaneously after the AuNCs originally restricted in ZIF-8 were allowed to escape. The intensity ratio of fluorescence at 469 nm to that at 660 nm (I469/I660) was conveniently employed as the response signal for representing the amount of ATP. This nanoprobe exhibits excellent sensitivity and selectivity toward ATP, with a limit of detection (LOD) of 0.061 μM. Besides, low cytotoxicity of this nanoprobe facilitates its application as a fluorescent indicator in fluorescence imaging of living cells. Encapsulating two types of fluorescent nanomaterials by a degradable ZIF-8 structure makes the ratiometric fluorescence response of the nanocomposite probe towards the target analyte that destroys the ZIF-8 structure possible, and simplifies the application of the probe.

Introduction to Biological Dark Energy: Experimental Evidence of Information Transfer under the Influence of Taheri Consciousness Fields and Investigating the Effects of the Fields on the Properties/Behavior of Water and a Biological Cell

The effects of Taheri Consciousness Fields (TCFs) on living organisms and non-living materials have been investigated. Since there is no physical intervention in applying TCFs, the findings suggested that the information transmitted from the Cosmic Consciousness Network is crucial in affecting the subjects under the study. The authors performed two experimental studies prior to this research. In the first experiment, the pH of pure water decreased significantly under the influence of TCFs. In the second experiment, the effect of TCFs on the Adenosine Triphosphate (ATP) production in the human HEK 293 cell line was investigated. It was found that the treated cells were able to generate a much greater amount of ATP compared to the control. This energy was released within a short period of TCF treatment (one hour) without sufficient glucose as the predominant fuel for cells. This study discusses the effects of TCFs by calculating the amount of information exchanged in the previous two experiments. According to Taheri, receiving information from TCFs requires a mental body. In other words, the behavior of water, and the human cell line alters under the influence of these fields because of receiving information through their mental body. Moreover, the shortage of time and resources to produce ATP in the TCFs treated cell strongly suggests that regardless of biological pathways, there is an alternative way to increase ATP in the immediate time, which is associated with an increase in information and a decrease in entropy of the system. This kind of energy is introduced and named “Biological Dark Energy” by Taheri.

Moderate activity of RNA chaperone maximizes the yield of self-spliced pre-RNA in vivo

CYT-19 is a DEAD-box protein whose adenosine-triphosphate (ATP)-dependent helicase activity facilitates the folding of group I introns in precursor RNA (pre-RNA) of Neurospora crassa (N. crassa). In the process, they consume a substantial amount of ATP. While much of the mechanistic insight into CYT-19 activity has been gained through the studies on the folding of Tetrahymena group I intron ribozyme, the more biologically relevant issue, namely the effect of CYT-19 on the self-splicing of pre-RNA, remains largely unexplored. Here, we employ a kinetic network model, based on the generalized iterative annealing mechanism (IAM), to investigate the relation between CYT-19 activity, rate of ribozyme folding, and the kinetics of the self-splicing reaction. The network rate parameters are extracted by analyzing the recent biochemical data for CYT-19-facilitated folding of Tetrahymena ribozyme. We then build extended models to explore the metabolism of pre-RNA. We show that the timescales of chaperone-mediated folding of group I ribozyme and self-splicing reaction compete with each other. As a consequence, in order to maximize the self-splicing yield of group I introns in pre-RNA, the chaperone activity must be sufficiently large to unfold the misfolded structures, but not too large to unfold the native structures prior to the self-splicing event. We discover that despite the promiscuous action on structured RNAs, the helicase activity of CYT-19 on group I ribozyme gives rise to self-splicing yields that are close to the maximum.

Effects of dietary fishmeal levels on adenosine triphosphate-related compounds and freshness of raw and cooked muscle in large yellow croaker Larimichthys crocea

A comparative study on the adenosine triphosphate (ATP)-related compounds and freshness of raw and cooked muscle in wild and farmed large yellow croaker (Larimichthys crocea) was performed. The two groups of farmed fish (189.87 ± 0.89 g) were fed two diets containing 44% (CF) and 25% (LF) of fish meal (FM) for 82 days, respectively. Results showed that the contents of ATP, adenosine diphosphate (ADP), adenosine monophosphate (AMP) and inosine 5′-monophosphate (IMP) were highest in raw and cooked muscle of wild fish, while these values were lowest in raw and cooked muscle of fish fed LF. Conversely, wild fish had the lowest inosine (Ino) and hypoxanthine (Hx) contents in raw and cooked flesh, while the LF group had the highest levels of Ino and Hx in raw and cooked flesh. Fish fed CF had significantly higher amounts of ATP, ADP and IMP in raw muscle, ATP and AMP in cooked muscle than those in the LF group (P < 0.05). The above results allowed wild fish to have the lowest K and Ki values, closely followed by fish fed CF, and the LF group have the highest values in raw and cooked muscle (P < 0.05). Meanwhile, cooking significantly decreased ATP, ADP and IMP contents, while increased AMP and Ino levels, inducing significantly higher K and Ki values in cooked muscle of all the treatments (P < 0.05). In conclusion, lower dietary FM level could decrease the contents of IMP and its precursors, and increase the degradation products of IMP in muscle, leading to the declines in umami and freshness. The stronger umami of wild large yellow croaker could be related to the higher precursors and lower degradation products of IMP in fillets.

A System of Rapidly Detecting Escherichia Coli in Food Based on a Nanoprobe and Improved ATP Bioluminescence Technology.

Bacterial contamination is an important factor causing food security issues. Among the bacteria, Escherichia coli is one of the main pathogens of food-borne microorganisms. However, traditional bacterial detection approaches cannot meet the requirements of real-time and on-site detection. Thus, it is of great significance to develop a rapid and accurate detection of bacteria in food to ensure food safety and safeguard human health. The pathogen heat-treatment module was designed in this paper based on the techniques including nanoprobe, pathogen heat-treatment, graphene transparent electrode (GTE), and adenosine triphosphate (ATP) bioluminescence technology. The system mainly consists of two parts: one is the optical detection unit; the other is the data processing unit. And it can quickly and automatically detect the number of bacterial colonies in food such as milk etc. The system uses not only the probe to capture and enrich E. coli by antigen-antibody interaction but also the heat treatment to increase the amount of ATP released from bacterial cells within five minutes. To enhance the detecting accuracy and sensitivity, the electric field generated by GTE is adopted in the system to enrich ATP. Compared to the other conventional methods, the linear correlation coefficient of the system can be reached 0.975, and the system meets the design requirements. Under the optimal experimental conditions, the detection can be completed within 25 min, and the detectable concentration of bacteria is in the range of 3.1 × 101–106 CFU/mL. This system satisfies the demands of a fast and on-site inspection.

Neuronal Adenosine A1 Receptor is Critical for Olfactory Function but Unable to Attenuate Olfactory Dysfunction in Neuroinflammation.

Adenine nucleotides, such as adenosine triphosphate (ATP), adenosine diphosphate (ADP), as well as the nucleoside adenosine are important modulators of neuronal function by engaging P1 and P2 purinergic receptors. In mitral cells, signaling of the G protein-coupled P1 receptor adenosine 1 receptor (A1R) affects the olfactory sensory pathway by regulating high voltage-activated calcium channels and two-pore domain potassium (K2P) channels. The inflammation of the central nervous system (CNS) impairs the olfactory function and gives rise to large amounts of extracellular ATP and adenosine, which act as pro-inflammatory and anti-inflammatory mediators, respectively. However, it is unclear whether neuronal A1R in the olfactory bulb modulates the sensory function and how this is impacted by inflammation. Here, we show that signaling via neuronal A1R is important for the physiological olfactory function, while it cannot counteract inflammation-induced hyperexcitability and olfactory deficit. Using neuron-specific A1R-deficient mice in patch-clamp recordings, we found that adenosine modulates spontaneous dendro-dendritic signaling in mitral and granule cells via A1R. Furthermore, neuronal A1R deficiency resulted in olfactory dysfunction in two separate olfactory tests. In mice with experimental autoimmune encephalomyelitis (EAE), we detected immune cell infiltration and microglia activation in the olfactory bulb as well as hyperexcitability of mitral cells and olfactory dysfunction. However, neuron-specific A1R activity was unable to attenuate glutamate excitotoxicity in the primary olfactory bulb neurons in vitro or EAE-induced olfactory dysfunction and disease severity in vivo. Together, we demonstrate that A1R modulates the dendro-dendritic inhibition (DDI) at the site of mitral and granule cells and impacts the processing of the olfactory sensory information, while A1R activity was unable to counteract inflammation-induced hyperexcitability.

Generation, breakdown and responses of ATP in the carotid body

Purinergic signaling involving adenosine triphosphate (ATP) acting on P2X2/3 receptors modulates physiological carotid body (CB) afferent discharge and chemoreflex activation however, the upregulation of P2X3 receptors on petrosal sensory neurons is partly responsible for aberrant CB tonicity and hyperreflexia, and the increase in blood pressure in the Spontaneously Hypertensive rat (SHR). The enhanced chemoreflex occurs prior to the onset of hypertension and elevated CB activity is thought to contribute to the underlying pathophysiology of the disease but the exact mechanisms remain unclear. We hypothesize that there is greater release of ATP and/or decreased ATP breakdown in the pre‐hypertensive SHR CB compared with that in the Wistar rat, and this evokes an increase in the chemoreflex response and subsequent activation of the sympathetic nervous system. We performed three studies. (i) Using digital droplet PCR (ddPCR), we measured mRNA expression levels of enzymes involved in the breakdown of ATP in CBs extracted from male 4‐week‐old prehypertensive SHR and Wistar rats; these included: ectonucleotide pyrophosphatase/phosphodiesterase 1‐3 (Enpp1‐3), ectonucleoside triphosphate diphosphohydrolase 2‐3 (Entpd2‐3), and ecto‐5’‐nucleotidase (Nt5e). (ii) We quantified ATP release from CBs in vitro during baseline conditions and during cyanide‐evoked ATP release using a colorimetric assay (213‐579‐1 ‐ Millipore), comparing between male 4‐week‐old pre‐hypertensive SHR and Wistar rats. (iii) In an in situ working heart‐brainstem preparation of 4‐week‐old Wistar rats, simultaneous recordings were made from phrenic (PN), hypoglossal (HN), recurrent laryngeal (RLN), abdominal (ABN), and thoracic sympathetic nerves (tSNA). The peripheral chemoreceptors were stimulated with either potassium cyanide (KCN 20‐100 μL, 0.04%) or ATP at varying concentrations (10‐100 µL, 50 µM ‐ 2 mM) delivered via the internal carotid artery. In whole CB extracts, ddPCR revealed increased Enpp2‐3 and Nt5e expression (p&lt;0.05) in pre‐hypertensive SHRs relative to Wistar rats suggesting greater capacity to break down ATP in the pre‐hypertensive SHR CB. There were no differences in the expressions of Enpp1 or Entpd2‐3 between rat strains. However, the amount of ATP released from the CB in both baseline and during cyanide stimulation was two‐fold greater in the pre‐hypertensive SHR (P&lt;0.05). Stimulation of the CB with KCN evoked hyperpnoea, bradycardia and sympathoexcitation at all doses. In contrast, CB injection of low dose ATP increased PN rate with little effect on tSNA, whereas higher doses induced apnea accompanied by repetitive burst discharge of post‐inspiratory activity in the RLN with tonic elevations in the discharge of both ABN and tSNA. In sum, despite the upregulation of several ATP degrading enzymes in the CBs of the SHR, CB release of ATP is higher in this species. We hypothesize that the increase in ATP release in the SHR CB drives the CB hyperexcitability and that the shift towards an upregulation of ATP degrading enzymes even prior to the development of hypertension is compensatory to oppose the increase in ATP signaling. The dose‐dependent effect of exogenous ATP at supraphysiological levels evoking greater CB‐mediated expiratory and sympathetic outflow in the Wistar rat mimics aspects of pathophysiological CB chemoreflex signaling in the SHR.

Urinary Catheterization Induces Delirium-Like Behavior Through Glucose Metabolism Impairment in Mice.

Delirium, an acute confusion status, is associated with adverse effects, including the development of Alzheimer's disease. However, the etiology and underlying mechanisms of delirium remain largely to be determined. Many patients have urinary catheterization (UC), and UC is associated with delirium. However, the cause effects of UC-associated delirium and the underlying mechanisms remain largely unknown. We, therefore, established an animal model of UC, without urinary tract infection, in mice and determined whether UC could induce delirium-like behavior in the mice and the underlying mechanism of these effects. Adult female mice (16 weeks old) had UC placement under brief isoflurane anesthesia. The delirium-like behavior was determined using our established mice model at 3, 6, 9, and 24 hours after UC placement. We measured the amounts of glucose in both blood and brain interstitial fluid, adenosine triphosphate (ATP) concentration in the cortex, and glucose transporter 1 in the cortex of mice using western blot, immunohistochemistry imaging, reverse transcriptase-polymerase chain reaction (RT-PCR), and fluorescence at 6 hours after the UC placement. Finally, we used vascular endothelial growth factor (VEGF) in the interaction studies. We found that UC induced delirium-like behavior in mice at 3, 6, 9, but not 24 hours after the UC placement. UC decreased glucose amounts in brain interstitial fluid (86.38% ± 4.99% vs 100% ± 6.26%, P = .003), but not blood of mice and reduced ATP amounts (84.49% ± 8.85% vs 100% ± 10.64%, P = .031) in the cortex of mice. Finally, UC reduced both protein amount (85.49% ± 6.83% vs 100% ± 11.93%, P = .040) and messenger ribonucleic acid (mRNA) expression (41.95% ± 6.48% vs 100% ± 19.80%, P = .017) of glucose transporter 1 in the cortex of mice. VEGF attenuated these UC-induced changes. These data demonstrated that UC decreased brain glucose and energy amounts via impairing the glucose transport from blood to brain, leading to delirium-like behavior in mice. These findings will promote more research to identify the etiologies and underlying mechanisms of delirium.

P2Y12 Purinergic Receptor and Brain Tumors: Implications on Glioma Microenvironment.

Gliomas are the most common malignant brain tumors in adults, characterized by a high proliferation and invasion. The tumor microenvironment is rich in growth-promoting signals and immunomodulatory pathways, which increase the tumor’s aggressiveness. In response to hypoxia and glioma therapy, the amounts of adenosine triphosphate (ATP) and adenosine diphosphate (ADP) strongly increase in the extracellular space, and the purinergic signaling is triggered by nucleotides’ interaction in P2 receptors. Several cell types are present in the tumor microenvironment and can facilitate tumor growth. In fact, tumor cells can activate platelets by the ADP-P2Y12 engagement, which plays an essential role in the cancer context, protecting tumors from the immune attack and providing molecules that contribute to the growth and maintenance of a rich environment to sustain the protumor cycle. Besides platelets, the P2Y12 receptor is expressed by some tumors, such as renal carcinoma, colon carcinoma, and gliomas, being related to tumor progression. In this context, this review aims to depict the glioma microenvironment, focusing on the relationship between platelets and tumor malignancy.

Red blood cell ATP release correlates with red blood cell hemolysis.

The precise matching of blood flow to skeletal muscle during exercise remains an important area of investigation. Release of adenosine triphosphate (ATP) from red blood cells (RBCs) is postulated as a mediator of peripheral vascular tone in response to shear stress, hypoxia, and mechanical deformation. We tested the following hypotheses: 1) RBCs of different densities contain different quantities of ATP; 2) hypoxia is a stimulus for ATP release from RBCs; and 3) hypoxic ATP release from RBCs is related to RBC lysis. Human blood was drawn from male and female volunteers (n = 11); the RBCs were isolated and washed. A Percoll gradient was used to separate RBCs based on cellular density. Density groups were then resuspended to 4% hematocrit and exposed to normoxia or hypoxia in a tonometer. Equilibrated samples were drawn and centrifuged; paired analyses of ATP (luminescence via a luciferase-catalyzed reaction) and hemolysis (Harboe spectrophotometric absorbance assay) were measured in the supernatant. ATP release was not different among low-density cells versus middle-density versus high-density cells. Similarly, hemoglobin (Hb) release was not different among the red blood cell subsets. No difference was found for either ATP release or Hb release following matched exposure to normoxic or hypoxic gas. The concentrations of ATP and Hb for all subsets combined were linearly correlated (r = 0.59, P ≤ 0.001). With simultaneous probing for Hb and ATP in the supernatant of each sample, we conclude that ATP release from RBCs can be explained by hemolysis and that hypoxia per se does not stimulate either ATP release or Hb release from RBCs.

Monitoring the Cleanliness of Reusable Surgical Instruments in Central Sterile Supply Department by Adenosine Triphosphate Method.

Cleaning is very important in reusable surgical instruments (RSI) reprocessing. The adenosine triphosphate (ATP) method is widely used to assess cleanliness, but few studies focus on using this method on RSIs. To assess the effectiveness of the ATP Bioluminescence Assay for monitoring the cleanliness of RSIs. The study was conducted in three central sterile supply departments (CSSD) from 2015 to 2020. Surgical scissors, hemostatic forceps, and other non-lumen instruments were sampled after cleaning. The surface of each instrument was swabbed in a uniform manner, including hinged joints and teeth, and the amount of ATP (amol) was calculated. A total of 871 RSIs were tested, the mean ATP lg (amol) was 2.829 ± 0.539 and the qualified rate was 80.57%. Washer-disinfector cleaning was more efficient than manual cleaning, the mean ATP lg (amol) were 2.776 ± 0.513 and 2.948 ± 0.575, respectively; the qualified rates were 84.97% and 70.59%, respectively. With the time of bare instruments expose to environment increased, the qualified rate was decreased (Ptrend = 0.044). ATP levels are stable and repeatable for continuous monitoring of the cleanliness of RSIs. It is a rapid and viable method for assessing the cleanliness of RSIs. Washer-disinfector cleaning is recommended and cleaned instruments should be placed in sterile packaging in good time. The ATP method is viable for assessing the cleanliness of RSI in CSSDs.
