Green synthesis of spermine coated iron nanoparticles and its effect on biochemical properties of Rosmarinus officinalis

In this study, aqueous spinach extract was used for the green synthesis of iron nanoparticles. The surface of iron oxide nanoparticles was coated with spermine. The physicochemical properties of nanoparticles were investigated using UV-Vis, TGA, FTIR, VSM, TEM, and DLS. The results showed that the nanoparticles had a spherical structure. The surface charge of the Fe3O4-NPs increased from −3.2 to 18.42 (mV) after Fe3O4 coating by spermine. In order to investigate the effect of nanoparticles on physicochemical properties of rosemary under drought stress conditions, an experiment was carried out in a completely randomized design. The results showed that the amount of antioxidant enzymes and secondary metabolites increased significantly under drought stress. Moreover, the use of spermine-coated iron nanoparticles can be useful in increasing resistance to drought stress in plants by increasing the activity of some antioxidant enzymes and secondary metabolites. The biocompatibility of Nanoparticles in cell suspension was investigated. the ability of Fe3O4-SM NPs to interact with DNA and protect it against DNaseI and ultrasonic waves using agarose gel electrophoresis was studied. The ability of Fe3O4-SM to neutralize the negative charge of DNA and protect it against DNaseΙ and ultrasonic waves was confirmed using an agarose gel electrophoresis assay.

Effects of adenosine N1-Oxide and pioglitazone on inflammatory and antioxidant state in sepsis caused by experimental cecal puncture in rat.

Sepsis is an acute condition caused by the systemic inflammatory response syndrome to an infection. There are very few drugs that could improve the severe conditions in patients with sepsis. Hence, it is important to consider different treatment options. In this survey, we studied the effect of adenosine N1-oxide (ANO) and pioglitazone on rats with cecal ligation and perforation (CLP). They were randomly divided to four groups (n = 10) including Group A: as control group receiving normal saline, Group B: the rats with CLP as surgical control group, Group C: the rats receiving ANO, and Group D: the rats receiving pioglitazone. Interleukin (IL) -6, IL-1β, tumor necrosis factor alpha (TNF-α), nitric-oxide (NO) in serum blood and superoxide dismutase (SOD), catalase (CAT) malondialdehyde, (MDA) and myeloperoxidase (MPO) in liver and spleen homogenates were measured. The amount of antioxidant enzymes in the spleen and liver, and finally cell viability and rats' survival were investigated. The measurement of blood serum nitric-oxide and survival of all groups of rats were also performed. The results indicated that both drugs could cause a decrease in IL-1β, IL-6, TNF-α and NO in rat blood serum and MDA and MPO in the liver and spleen homogenates, however, a significant increase in SOD and CAT in the liver and spleen homogenates in rats that received ANO and pioglitazone was observed compared to rats with CLP group. Cell viability and rats' survival were significantly improved in rats that received ANO and pioglitazone compared to rats with CLP group. Adenosine N1-oxide showed stronger and more effective effects.

Salt stress triggers augmented levels of Na+, K+ and ROS alters salt-related gene expression in leaves and roots of tall wheatgrass (Agropyron elongatum)

In turfgrass breeding, competent grass ecotypes are preferably identified for their resistance to salinity condition. This research was designed to explore genes that induce salt resistance (NHX1, NHX2, HKT1;4, SnRK2.4 and NAC9) and their role in physiological modifications of six tall wheatgrass ecotypes (Agropyron elongatum L.). The sites of sample collection were characterized by different levels of salinity, i.e. low (EC: 4 dS m−1 and pH: 6.5), moderate (EC: 7 dS m−1 and pH: 6.5) and high (EC: 12 dS m−1 and pH: 7.5). This study was designed as a split-plot in a randomized complete block where salinity treatments served as the whole-plot factor and ecotypes served as the subplot factor. The ecotypes were screened for their resistance to salinity, based on visual symptoms, salt injury index, physiological features and biochemical parameters. The results revealed that ecotype ‘AE5’ was most resistant to salinity than other ecotypes, whereas ‘AE3’ was the most susceptible. To understand why these differences occurred, measurements were aimed at revealing mRNA levels that resulted from genes responsible for salt resistance. Our results demonstrated that salinity-resistant ecotypes showed high expression levels of several genes, i.e. NHX1, NHX2, HKT1;4, SnRK2.4 and NAC9 in the leaves and roots. These results were corroborated by a decrease (by 1.5–2.5 times) in stress markers, namely, superoxide anion (O2−), hydrogen peroxide (H2O2) and malondialdehyde (MDA), as well as an increase (by 0.5–7 times) in enzymatic and non-enzymatic antioxidant activity in salinity-resistant ecotypes when the plants were exposed to salinity. We observed higher values of initial root length and lateral root density (21% and 18%, respectively) in salinity-resistant ecotypes under salinity condition, compared to other ecotypes. There were lower expression levels of NHX1 and NHX2 in the roots, which were 3.2 and 2.1 times less, respectively, compared to the leaves. This implied that NHX1 and NHX2 expressions can lead to the sequestration of Na+ in the leaves during salinity condition. The current research revealed that HKT1;4 was more able to restrict Na + accumulation, compared to the actions of NHX1 and NHX2 genes. The over-expression of HKT1;4 in ‘AE5’ allowed a better maintenance of root growth during salinity condition. The expression of NAC9 had an increase of 2.1-fold which correlated with an increase in the amount of antioxidant enzymes. In general, the location of sample collection explained the differences in gene expression, especially regarding the extent to which plants respond to salinity condition. Ultimately, these differences can define physiological features in salinity-resistant and salinity-susceptible ecotypes of tall wheatgrass.

Biosynthesis of metal nanoparticles and metal oxidesand their use as components of fertilizers and preparations for plant growing (literature review)

A promising direction in agriculture is the use of metal nanoparticles as nanofertilizers, which can increase the yield of agricultural crops and, at the same time, minimize the frequency of fertilization due to the long-term release of nutrients. One of the environmentally safest and cheapest methods of synthesizing metal nanoparticles is biosynthesis using plant extracts. During the redox reaction, proteins, carbohydrates, organic acids, phenols and other metabolites are able to transfer electrons to metal cations, restoring their charge to zero on the nanometer scale. This article, based on publications on the issue under study by authors from the United States of America, Europe and the Middle East, China and India, describes the biosynthesis of nanoparticles of zinc oxide, copper and copper oxide, iron and iron oxide, as well as manganese and manganese oxide using the formation of plant extracts, and data on the use of these metals and their oxides as nanofertilizers and preparations for plant growing are presented. It has been shown that the use of metal nanoparticles and their oxides as fertilizers is more effective than conventional compounds used as fertilizers. This is probably due to the fact that it is easier for nanoparticles to penetrate through the plant membrane, as well as to pass into a form accessible to plants in comparison with conventional analogs. The positive effect of the influence of nanoparticles on plants is expressed in the elongation of the roots and shoots of model plants and an increase in the biomass of seedlings. In addition, the amount of chlorophyll in the leaves increases, and some biochemical processes also change, for example, the amount of antioxidant enzymes increases, which makes it possible to increase the stress resistance of plants.

Effect of potassium fertilizer on the growth, physiological parameters, and water status of Brassica juncea cultivars under different irrigation regimes.

Abiotic stress, especially a lack of water, can significantly reduce crop yields. In this study, we evaluated the physiological and biochemical effects of potassium sulfate (K2SO4) fertilizer and varied irrigation regimes on the economically significant oilseed crop, Brassica juncea L, under open field conditions. Two cultivars (RH-725 and RH-749) of B. juncea were used in a randomized complete block design experiment with three replicates. Irrigation regimes consisted of a control (double irrigation: once at the 50% flowering and another at 50% fruiting stages), early irrigation (at 50% flowering only), late irrigation (at 50% fruiting only) and stress (no irrigation). The K2SO4 applications were: control (K0, no fertilization); K1, 10 kg ha−1; and K2, 20 kg ha−1. We measured growth via fresh and dry plant weight, plant height, root length, and leaf area. All the growth parameters were higher in RH-749. The physiological attributes, including the membrane stability index and relative water content, were higher at the 50% flowering stage in RH-749. The amount of antioxidant enzymes (catalase (CAT), guaiacol peroxidase (POX), ascorbate peroxidase (APX), and superoxide dismutase (SOD)) was enhanced when both plants were fertilized during water stress. All of these enzymes had higher activity in RH-749. The total chlorophyll content and photosynthesis rate were considerably higher in RH-749, which leaked fewer electrolytes and maintained a less destructive osmotic potential under limited water conditions. The results indicated that it is water-stress tolerant when given a high concentration of K2SO4, which alleviated the adverse effects of water stress on growth and physiology.

Zinc oxide nanoparticles phytotoxicity on halophyte from genus Salicornia

This study deals with the effect of zinc oxide nanoparticles (ZnO NPs) on halophyte from the genus Salicornia. The presence of ZnO nanoparticles (100 and 1000 mg/L) in the solid culture medium resulted in the negative effects on plant growth in the concentration-dependent manner. The shoot length of plant cultivated with 1000 mg/L ZnO NPs decreased by more than 50% compared to non-treated plants. The phytotoxicity was associated with the release of free zinc(II) ions, which was determined by atomic absorption spectroscopy and fluorescence microscopy. Another mechanism involved in ZnO NPs phytotoxicity was closely connected with generation of reactive oxygen species (ROS), which was accompanied by changes in activities and amounts of antioxidant enzymes. Histochemical evaluation showed that ROS were present also in the shoot of plant, which was not in direct contact with NPs. The reduction of activity and amount of antioxidant enzymes such as gamma–ESC, GR, SOD, PER, APX and higher concentration of ROS lead to lipid peroxidation, the latter being almost 3 times higher for the plant treated with 1000 mg/L NPs compared to control. The misbalance in zinc homeostasis and creation of ROS with subsequent oxidative stress led to the initiation of processes of programmed cell death, which was demonstrated by the loss of mitochondrial potential and increase of intracellular calcium (II) ions. Despite halophytes exhibit higher stress resistance than glycophytes, they are prone to negative changes if incubated in the environment containing ZnO nanoparticles.

Heavy metal tolerance in contrasting ecotypes of Alyssum montanum

The response of metallicolous (M) and nonmetallicolous (NM) Alyssum montanum ecotypes to multi-metal stress was investigated under in vitro condition and compared in this study. Shoot cultures were simultaneously treated with 0.7 mM ZnSO4, 3.0 μM Pb(NO3)2 and 16.4 μM CdCl2 for 8 weeks and evaluated for their morphogenetic and ultrastructural reaction, growth tolerance as well as ability to Zn, Pb, and Cd uptake. Moreover, tissue localization and concentrations of antioxidant compounds were determined in order to elucidate the potential role of ROS-scavenging machinery in plant tolerance to metal toxicity. The results clearly demonstrated that M specimens treated with heavy metals showed less phytotoxic symptoms and low level of lipid peroxidation than reference NM one. The enhanced tolerance of M ecotype resulted from heavy metals detoxification in trichomes and intracellular leaf compartments as well as balanced ROS accumulation. The inactivation of ROS in M plants was based on peroxidase-flavonoid system, while in NM plants such relationship was not detected and amounts of antioxidant enzymes or phenolic compounds was comparable to untreated specimens or decreased significantly. Considering the procumbent growth of such hemicryptophyte which reproduce effectively in the presence of heavy metals but is characterized by low biomass production, it is proposed to exploit M ecotype of A. montanum in revegetation schemes of polluted calamine wastes to provide the prompt stabilization of areas prone to erosion.
