Evaluation of biological activity and amino acid profile of protein from Thai edible insects

Insects are one of the sources of protein for future foods. In some cultures, insects have been used as a component of traditional medicine. Moreover, some proteins from insects have also shown biological activities related to human health, such as non-communication disease treatment. In this study, the protein content, amino acid profile, and inhibitory activity against a-glucosidase, a-amylase, acetylcholinesterase, and tyrosinase of soluble protein from six species of Thai edible insects were investigated. The protein from six Thai edible insects, including Gryllus bimaculatus De Geer (Sample 1), Omphisa fuscidentalis (Sample 2), Bombyx mori L. (Sample 3), Brachytrupes portentosus (Lichtenstein 1796) (Sample 4), Acheta domesticus L. (Sample 5), and Patanga succincta (Linnaeus) (Sample 6), was extracted and evaluated for its content and biological activity. The results indicated that sample 1 yielded the highest protein content of 84.23±5.15 g/100 g which is more than the protein content of meat and dairy products. A total of eight essential amino acid contents (EAA) were found in all six edible Thai insects. The highest total EAA was obtained from Sample 2 with 37.21±1.82 g/100 g, higher than the criteria established by FAO/WHO/UNU.1985. Sample 3 yielded the highest inhibition percentage of soluble protein from edible insects against α-glucosidase. The same was true for Sample 4 for α-amylase, Sample 1 for acetylcholinesterase, and Sample 2 for tyrosinase. These results indicated the quality and functional properties of protein from six species of Thai edible insects, which may be helpful in their future exploration and application

Bio-functional and prebiotics properties of products based on whole grain sorghum fermented with lactic acid bacteria.

Products fermented with lactic acid bacteria based on whole grain flours of red or white sorghum (Sorghum bicolor (L.) Moench) added with malted sorghum flour, or with skim milk (SM) were developed. Composition, protein amino acid profile, total acidity, pH, prebiotic potential, and bio-functional properties after simulation of gastrointestinal digestion were evaluated. In all cases, a pH of 4.5 was obtained in approximately 4.5 h. The products added with SM presented higher acidity. Products made only with sorghum presented higher total dietary fiber, but lower protein content than products with added SM, the last ones having higher lysine content. All products exhibited prebiotic potential, white sorghum being a better ingredient to promote the growth of probiotic bacteria. The addition of malted sorghum or SM significantly increased the bio-functional properties of the products: the sorghum fermented products added with SM presented the highest antioxidant (ABTS•+ inhibition, 4.7 ± 0.2 mM Trolox), antihypertensive (Angiotensin converting enzyme-I inhibition, 57.3 ± 0.5%) and antidiabetogenic (dipeptidyl-peptidase IV inhibition, 31.3 ± 2.1%) activities, while the products added with malted sorghum presented the highest antioxidant (reducing power, 1.6 ± 0.1 mg ascorbic acid equivalent/mL) and antidiabetogenic (α-amylase inhibition, 38.1 ± 0.9%) activities. The fermented whole grain sorghum-based products could be commercially exploited by the food industry to expand the offer of the three high-growth markets: gluten-free products, plant-based products (products without SM), and functional foods. © 2023 Society of Chemical Industry.

Primary exploration of mushroom protein hydrolysis and cooking impact on the protein amino acid profiles of Agaricus bisporus and Lentinula edodes mushrooms

Compared to free amino acids, protein amino acids in mushroom have gained little attention. No studies have investigated mushroom protein hydrolysis kinetics; moreover, investigation into cooking impact on protein amino acids is scarce. This study aimed to 1) examine the hydrolysis time-scheme of mushroom protein amino acids in order to investigate the hydrolysis method using a mushroom matrix; and 2) investigate the impact of cooking on protein amino acids within two groups: white and crimini mushrooms with a roasting cooking method and portobello and shiitake mushrooms with sautéing and roasting methods. For Aim 1, mushroom powder was hydrolyzed at 2, 4, 12, 24, 48, 72, and 96 h. The results showed logical hydrolysis progression. For Aim 2, whole mushroom samples were hydrolyzed with the traditional 24 h 6N acid hydrolysis. Protein amino acids in the hydrolysates were then quantified via EZ:Faast derivatization and ensuing GC-MS analysis. Significant cooking method and mushroom type interactions were observed for 66.7–81.8% of the 21–22 amino acids detected, confounding direct comparison of the two cooking methods. All total and most (66.7–100%) individual protein amino acids significantly decreased upon cooking for all samples. Met was the most limiting essential amino acid in all samples, but it was subject to oxidation-induced variance. The second most limiting amino acid, His, did not significantly differ between sautéing and roasting cooking methods. This study adds knowledge regarding mushroom's protein amino acid profile and how cooking methods affect the profile.

Effect of Production Technology Intensity on the Grain Yield, Protein Content and Amino Acid Profile in Common and Durum Wheat Grain

Products derived from wheat grains are an important source of protein in the daily diet of people in many parts of the world. The biological value of protein is determined by its amino acid composition and the proportions of the individual amino acids. Synthesis of these compounds in wheat grains is influenced by genetic factors, as well as habitat conditions and the agrotechnology applied in cultivation. The aim of this study was to assess the effect of production technology (integrated, intensive) on the grain yield and the content amino acid profile of protein in common and durum wheat grain. Field research was conducted at the Experimental Station IUNG-PIB in Osiny (Poland) in two growing seasons. It was found that grain yield significantly depended on the weather conditions in the years of harvesting and genotype, but did not depend on the production technology. On the other hand, the protein content and their amino acid composition depended significantly on the production technology and genotype. A significantly higher content of protein substances was found in durum wheat grain. Increasing the intensity of production technology had a positive effect on the total protein content and the content of individual amino acids, both exogenous and endogenous. The amino acid limiting the biological value of protein contained in grains of both wheat species was lysine, and the deficiency of this amino acid was significantly lower in grain protein from intensive than integrated cultivation technology.

Alternative protein sources: bacteria Methylococcus capsulatus concentrate, characteristic of composition and biological value

A promising growth vector of food protein production in the context of the Russian Federation's food sovereignty security is the use of microbial synthesis. Taking into consideration the proven promising use of biotechnological processes in the production of alternative protein sources, modern scientific research is focused, among other issues, on improving the technology of obtaining food microbial protein using a variety of substrates and strains-producers, as well as evaluating the consumer properties, food, biological value and safety of such products. The purpose of the research was to study and comparatively evaluate protein concentrate (PC) from bacteria Methylococcus capsulatus and basic food of animal and plant origin within the development of the technology of optimal in nutritional and biological value PC production. Material and methods. Analysis of the nutritional and biological value of PC obtained from denucleinized and purified from cell walls biomass of methanoxidizing bacteria Methylococcus capsulatus (strain GSB-15) was carried out on 46 indicators, including estimation of protein content and amino acid composition, fat content and fatty acid composition, ash and moisture. Biological studies based on measuring of net protein ratio / net protein utilization were performed on 28 growing (between 25-50 days of life) male Wistar rats. Rats in the control group (n=14) received a semi-synthetic casein diet with a protein content of ~12% in calories, the test group (n=14) received a diet including an equivalent amount of PC protein. Body weight, feed intake, and fecal and urine nitrogen losses were measured during the experiment. The biological value and digestibility of protein were judged by coefficients of protein efficiency ratio, net protein ratio, true protein digestibility, true protein biological value, true net protein utilization. Results. The nutritional value study of PC showed high protein content - 69.0%, the share of fat, moisture and ash, accounted for 0.17, 9.5 and 14.4%, respectively. The carbohydrate content was 7.0% (of which mono- and disaccharides were <0.1%). The results of a comparative assessment of Methylococcus capsulatus protein amino acid profile and basic food of animal and plant origin showed a balanced content of the most amino acids, the level of which is comparable with the protein of chicken egg, which is traditionally a standard of quality of complete protein. At the same time, the content of the essential amino acid tryptophan in PC was an order of magnitude lower than in chicken egg protein; the content of this amino acid in PC is comparable with incomplete plant proteins (sunflower, flax, rapeseed). The results of the biological value evaluation of the Methylococcus capsulatus protein in the experiment on rats indicate a relatively low biological value of the microbial synthesis protein, that is caused, most likely, by tryptophan deficiency. Rats of the test group had a significant decrease in body weight gain, feed/protein intake, coefficient of protein efficiency ratio, coefficient of net protein ratio, true protein biological value, true net protein utilization. Conclusion. The results of a comparative evaluation of PC from methanotrophic bacteria Methylococcus capsulatus denucleinized biomass and basic food of animal and plant origin indicate its relatively high nutritional value. However, the characteristics of this PC sample were not optimal in regard of protein biological value by reason of tryptophan deficiency. A single amino acid deficiency is not a valid argument for not using microbially synthesized protein in human nutrition, considering the capabilities of the modern food industry, including ways to enrich foodstuffs with missing components. In addition, there is every cause to believe that adjusting the hydrolysis technology used in the production of PC will allow to eliminate the essential amino acid loss, thereby increasing the biological value of this product.

Physicochemical and functional properties of pigeon pea (Cajanus cajan) protein and non-starch polysaccharides

Pigeon pea (Cajanus cajan L. Millsp) has been commonly consumed in human diet, and it is also a promising source of protein and dietary fibers. In this study, pigeon pea protein and non-starch polysaccharides (NSP) were sequentially extracted and fractionated. The amino acid profile of protein was characterized, and total sugars and uronic acid of non-starch polysaccharides were identified. Physicochemical properties of pigeon pea protein were compared with commercial soy protein isolates; meanwhile, pigeon pea NSP was compared with commercial citrus pectin. The results indicated that the viscosity of protein solution from pigeon pea is higher than that from soy protein isolates at all concentrations and exhibited shear-thinning behavior at shear rate range 0.1–500 s−1. The emulsions prepared from pigeon pea protein at the concentrations of 0.5% and 2% (w/w) displayed significant higher emulsifying stability (p < 0.05) than those of soy protein. Pigeon pea polysaccharide exhibited higher surface activity than citrus pectin, with the 0.5% pigeon pea polysaccharide solution comparable with 1.5% citrus pectin solution. The results of this study have implied great potentials of application of pigeon pea protein and non-starch polysaccharide as novel ingredients for food industries.

Growth performance and metabolic responses of Nile tilapia fed diets with different protein to energy ratios

The effect of dietary digestible protein to digestible energy (DP/DE) ratio on the growth performance and metabolic responses of Nile tilapia juveniles was evaluated. Five experimental diets were formulated with increasing DP/DE ratio (13.2, 15.5, 18.4, 20.0, and 22.6 mg kJ−1) and fed to eight replicates of Nile tilapia (initial body weight of 9.3 g) for 110 days. The apparent digestibility coefficients of the diets were determined in a parallel trial. Digestibility coefficients of dry matter, crude protein, and crude energy linearly decreased with increasing dietary P/E ratios. A quadratic effect of dietary DP/DE ratio on growth performance was observed, and maximum weight gain (g kg−1 day−1) or daily growth increment was attained with a dietary DP/DE ratio of 16.5 and 17.2 (mg kJ−1), respectively. Feed intake and energy retention (kJ ABW kg−1 day−1) linearly decreased, and feed efficiency linearly increased with increased dietary DP/DE ratio. The increase of dietary DP/DE ratio decreased the whole body, muscle, hepatic, and visceral lipid content and increased the fillet yield. The decrease of dietary DP/DE linearly increased nitrogen retention (% N intake) and whole-body lipid deposition through the reduction of amino acid catabolism (GDH and ALAT) and gluconeogenesis (FBPase) related enzymes and the increase of glycolysis (GK and HK) and lipogenesis- (G6PDH and ME) related enzymes.In conclusion, a dietary DP/DE ratio of 16.5–17.2 mg kJ−1 promoted the maximum growth performance of Nile tilapia juveniles. However, further studies are required to evaluate if a fine-tuning of the protein amino acid profile would allow a further reduction of the optimum dietary DP/DE ratio. Metabolic response to the decrease in dietary DP/DE levels suggested activation of glycolysis and lipogenesis and inhibition of amino acid catabolism and gluconeogenesis, highlighting the high capacity of Nile tilapia for metabolic utilization of dietary carbohydrates.

Mechanical properties and compositional characteristics of beet (Beta vulgaris L.) varieties and their response to nitrogen application

Mechanical properties of sugar beet are important during harvesting and processing. To potentially correlate mechanical properties with structural features of cell wall polymers, four different Beta varieties (Beta vulgaris L.) were characterized for their mechanical properties and cell wall composition. In addition, the influence of nitrogen fertilization was analyzed. Additional nitrogen fertilization only slightly influenced mechanical properties and cell wall composition. Hardly any structural differences of cell wall polysaccharides were observed for all Beta varieties. Slight differences in alcohol insoluble residue and protein contents, in amino acid profiles of proteins, and in esterification degree of pectins were found. The Beta varieties differed in their contents of cell wall bound phenolic components, and particularly in their dry matter contents. Analyses of mechanical properties of the Beta varieties demonstrated differences in tissue firmness and compressive strength. However, no evidence was found that the observed differences of cell wall composition are responsible for the divergent mechanical properties, with the exception of water contents that were higher in samples with less compressive strength.

A favourable protein amino acid profile and the absence of gluten, being a strong allergen, are just some of the advantages of the quinoa. Besides to protein, other important component of quinoa seeds of is starch. This polysaccharide, due to its origin, can also have interesting properties. The aim of the study was to compare starch isolated from the Faro cultivar of quinoa grown in Poland with commercial starches of various botanical origins (potato, wheat and maize). The analysis included the colour, paste clarity and thermodynamic and rheological pasting properties of the starches. Our research has demonstrated that the paste clarity of quinoa starch was 1.26%, which means that the paste was the cloudiest, of all those tasted. Quinoa starch differed from the other starches in terms of thermodynamic and rheological pasting characteristics. It had the lowest temperature at the onset of transition (To) and the lowest transition enthalpy ΔHG. Additionally, quinoa starch demonstrated the lowest pasting temperature and the paste had the highest final viscosity (FV). The highest gelatinization temperature of was noted for maize starch, whose paste had the lowest viscosity.

Nutritional value of proteins and lipids in breast muscle of geese from four different Polish genotypes

Summary This research aimed at determining and comparing the fatty acid lipid and amino acid profiles of protein from breast muscles of 17-week-old geese native to northern Poland – Kartuska (KA) and Suwalska (SU) as well as native to southern Poland – Lubelska (LU) and Kielecka (KI). There were 18 birds from each genotype (n = 18). During the experimental period, geese were fed ad libitum on the same complete feed.

Chemical composition and nutritional quality of short-ripened rennet cheeses produced by traditional methods

The aim of the study was to determine the variability in the chemical composition and nutritive value parameters of smoked and unsmoked short-ripened rennet cheeses and unsmoked long-ripened rennet cheeses produced by traditional methods. The raw material for the production of short-ripened cheeses was pasteurized cows’ milk obtained from a dairy, whereas the long-ripened cheeses were manufactured from raw cows’ milk obtained from the producer’s farm. All three varieties of cheese examined were produced with commercial dairy starter cultures. The material for the study was collected in winter, directly at the producers’ retail outlets in southern and eastern Poland. The basic chemical composition was determined according to the Polish Standards, whereas the amino acid profiles of proteins from the cheeses were determined by ion-exchange chromatography. The result analysis revealed significant differences between the different varieties of cheese in terms of their water content, ranging from 32.2% to 42.1%, as well as protein content, which varied from 25.6% to 31.6%. Fat levels ranged between 22.2% and 24%, whereas total ash content amounted to 5.1-5.8%. The significantly highest salt content was found in unsmoked short-ripened cheeses. In all three cheese varieties, the total exogenous amino acid content was comparable, ranging from 46.17 g to 47.36 g/100 g protein, and that of endogenous amino acids varied from 52 g to 53 g/100 g protein. The biological value of proteins was determined by calculating to the chemical score (CS), as described by Mitchell and Block, and the essential amino acid index (EAAI), as described by Oser. A comparison of the results with the standard chicken egg white proteins showed that the limiting amino acids for all varieties of cheese were methionine and cysteine. On the other hand, a comparison with the FAO/WHO-suggested pattern of amino acid requirements (1991) for all age groups over 1 year of age showed that the limiting amino acids were methionine and cysteine in smoked and unsmoked short-ripened cheeses, and treonine in long-ripened cheeses. Considering the chemical indices, such as CS and EAAI, it may be concluded that the traditional rennet cheeses produced in southern and eastern Poland have a favourable amino acid composition of proteins and a high nutritive value..

An Effective Method of Continuous Production of Erythritol from Glycerol by Yarrowia lipolytica MK1.

This study demonstrates the potential applicability of the UV mutant Yarrowia lipolytica MK1 for the valorisation of glycerol and erythritol production in a chemostat culture. The aim of this research is to investigate the optimal C:N ratio in the feeding medium in order to enhance erythritol production. The highest erythritol concentration, at 113.1 g/L with a volumetric erythritol production rate of 1.1 g/(L·h) and a yield of 0.57 g/g, was obtained in the feeding medium with a C:N ratio of 80:1. Moreover, no residual glycerol was observed in the culture broth during cultivation. The chemical composition of the biomass was analysed. The contents of lysine and threonine in the biomass protein amino acid profile were higher than those required by the FAO/WHO for fodder yeast.

Optimizing fish meal-free commercial diets for Nile tilapia, Oreochromisniloticus

A feeding trial was conducted in a closed recirculating aquaculture system with Nile tilapia Oreochromisniloticus juveniles (mean weight, 6.81g) to examine the response to a practical diet containing protein primarily from menhaden fish meal (FM) and soybean meal (SBM) (control, Diet 1) or to diets with decreasing ratios of PBM to SBM (Diets 2–7; dose–response) as a total replacement for digestible protein from FM, and the efficacy of 1% supplemental taurine (Tau) at the highest level of plant protein inclusion by removing Tau (Diet 8). To the extent possible, the replacement diets were formulated using currently published amino acid availabilities for the ingredients of interest in order to estimate and supplement the first two limiting amino acids (Met and Lys) to match levels in the FM control diet. The test diets were formulated to contain 35% digestible protein. Fish were fed three times daily all they would consume in 30min. All performance measures were quadratic with respect to PBM:SBM ratio in the diet. The highest weight gain, lowest average daily feed intake, lowest feed conversion, and greatest specific growth rate coincided with a dietary PBM:SBM ratio of 1.22 to 1.35 suggesting that the best tilapia performance in the current trial was achieved with replacement formula D3 that contained approximately 20% SBM, 30% PBM, and supplemental Lys, Met, and Tau. However, all growth performance measures were significantly linear and decreased with respect to increasing distance from the ideal protein amino acid profile for tilapia. Positive effects of taurine supplementation at the highest level of dietary plant protein inclusion were not observed and may have been overwhelmed by imbalances in other amino acids in the test diets. The current results provide evidence that total deviation from the ideal protein profile in tilapia is an important consideration for diet formulation when combinations of diet ingredients are used. Hence, the essential amino acid content of a fish meal control diet may be an inadequate target for optimizing fish meal replacement diets for tilapia; whereas the whole body or muscle amino acid pattern may be a more useful formulation target. Finally, while the database of ingredients that have been evaluated in tilapia is growing, the industry will benefit from more efficient diets as long-term averages of amino acid composition and digestibility accrue for a variety of traditional and novel ingredients.Statement of Relevance: The current results provide evidence that it is the total deviations from a postulated ideal protein profile that is a more important consideration for diet formulation than the combination of diet ingredients used to meet that profile. Therefore, it should be possible to formulate least-cost fish meal replacement diets for tilapia, irrespective of ingredient combinations, and diet intact protein level, as long as a reasonable amino acid model is chosen and a fairly robust set of ingredient composition and digestibility data are available.

Nitrogen fractions and amino acid content in alfalfa and red clover immediately after cutting and after wilting in the field

The aim of this study was to determine the composition of nitrogen fractions in alfalfa and red clover, which differ in proteolytic activity, and to evaluate the effect of wilting on changes in nitrogen fractions in alfalfa and red clover herbage. Total nitrogen was divided into protein and non-protein nitrogen, and the amino acid profile of protein was analyzed. Buffer-soluble nitrogen (BSN), including buffer-soluble protein nitrogen (BSPN) and non-protein buffer-soluble nitrogen (NPBSN), was determined. The NPBSN fraction was further subdivided into peptide nitrogen, amino acid nitrogen, neutral detergent-insoluble nitrogen (NDIN) and acid detergent-insoluble nitrogen (ADIN). Wilting in the field to 40% dry matter content (the swath was tedded once) did not reduce the total nitrogen content of alfalfa and red clover herbage, although it affected the concentration of the BSNfraction, in particular NPBSN, and free amino acid nitrogen. During alfalfa wilting, the soluble protein content decreased and the concentrations of non-protein nitrogen compounds increased, mostly due to an increase in free amino acid nitrogen. A reverse trend was observed during red clover wilting – the concentrations of non-protein nitrogen compounds decreased and soluble protein content increased. A decrease was also noted in peptide nitrogen, NDIN and ADIN. Wilting of alfalfa and red clover had no adverse effect on the amino acid profile of protein. The concentrations of essential amino acids that limit milk protein synthesis, i.e. Lys, Met, His and Arg, did not decrease. Legume wilting in the field contributes to an increase in the concentrations of soluble nitrogen in the plant material intended for ensiling.

Nutritive value of tropical forage plants fed to pigs in the Western provinces of the Democratic Republic of the Congo

The nutritive value of 20 forage plants commonly used for feeding pigs in the Democratic Republic of the Congo was studied to determine chemical composition, protein amino acid profiles, mineral content, and in vitro digestibility using a two-step method combining an enzymatic pepsin and pancreatin hydrolysis followed by a 72h gas-test fermentation. The highest protein contents (270–320g/kg DM) were obtained for Vigna unguiculata, Psophocarpus scandens, Leucaena leucocephala, Manihot esculenta, and Moringa oleifera. Grasses, Acacia mangium, and Eichhornia crassipes, showed the lowest crude protein (CP) and highest NDF contents. Cajanus cajan and Trypsacum andersonii had the most balanced amino acid profile, being deficient in lysine and slightly deficient in histidine, while Megathyrsus maximus displayed the highest number of essential amino acids deficiencies. High mineral contents were obtained from, in ascending order, with M. oleifera, V. unguiculata, E. crassipes, Ipomea batatas and Amaranthus hybridus. In vitro dry matter digestibility ranged from 0.25 to 0.52, in vitro CP digestibility from 0.23 to 0.80, in vitro energy digestibility from 0.23 to 0.52. M. esculenta, M. oleifera, I. batatas, Mucuna pruriens, V. unguiculata, P. scandens and A. hybridus showed high digestibilities for all nutrients. Gas production during fermentation of the pepsin and pancreatin-indigestible fraction of the plants varied from 42ml/g DM for A. mangium to 202ml/g DM for I. batatas (P<0.001). Short-chain fatty acid production during fermentation varied from 157 to 405mg/g of the pepsin and pancreatin indigestible fraction. It is concluded that some of these species are interesting sources of proteins and minerals with a good digestibility that might be used more economically than concentrate, especially in smallholder production systems, to improve pig feeding, mineral intake and intestinal health in pigs reared in the tropics.

Biomass production by novel strains of Yarrowia lipolytica using raw glycerol, derived from biodiesel production

This study demonstrated the potential applicability of the isolated strains of Yarrowia lipolytica for the valorization of glycerol waste generated during biodiesel production, throughout biomass production. Twenty-one strains were isolated from different environments and identified as Y. lipolytica. Biomass production from pure glycerol (25 g L(-1)) was performed in the shake-flasks experiment. Eight strains with the best biomass production ability were chosen for studies in bioreactor (pH 3.5). The analysis of technological process parameters and biomass chemical composition demonstrated that S6 strain was the most suitable for biomass production. Its application allowed obtaining 11.7 and 12.3 g L(-1) of the biomass with 1.30 and 1.37 g L(-1) h(-1) productivity, respectively when pure and raw glycerol (25 g L(-1)) was used. In the yeast protein amino acid profile the contents of lysine, threonine and phenylalanine/tyrosine were higher than required by FAO/WHO. According to the EAAI, the nutritional value of the biomass reached up to 72.3%.

Determination of Soybean Oil, Protein and Amino Acid Residues in Soybean Seeds by High Resolution Nuclear Magnetic Resonance (NMRS) and Near Infrared (NIRS)

AbstractA detailed account is presented of our high resolution nuclear magnetic resonance (HR-NMR) and near infrared (NIR) calibration models, methodologies and validation procedures, together with a large number of composition analyses for soybean seeds. NIR calibrations were developed based on both HR-NMR and analytical chemistry reference data for oil and twelve amino acid residues in mature soybeans and soybean embryos. This is our first report of HR-NMR determinations of amino acid profiles of proteins from whole soybean seeds, without protein extraction from the seed. It was found that the best results for both oil and protein calibrations were obtained with a Partial Least Squares Regression (PLS-1) analysis of our extensive NIR spectral data, acquired with either a DA7000 Dual Diode Array (Si and InGaAs detectors) instrument or with several Fourier Transform NIR (FT-NIR) spectrometers equipped with an integrating sphere/InGaAs detector accessory. In order to extend the bulk soybean samples calibration models to the analysis of single soybean seeds, we have analized in detail the component NIR spectra of all major soybean constituents through spectral deconvolutions for bulk, single and powdered soybean seeds. Baseline variations and light scattering effects in the NIR spectra were corrected, respectively, by calculating the first-order derivatives of the spectra and the Multiplicative Scattering Correction (MSC). The single soybean seed NIR spectra are broadly similar to those of bulk whole soybeans, with the exception of minor peaks in single soybean NIR spectra in the region from 950 to 1,000 nm. Based on previous experience with bulk soybean NIR calibrations, the PLS-1 calibration model was selected for protein, oil and moisture calibrations that we developed for single soybean seed analysis. In order to improve the reliability and robustness of our calibrations with the PLS-1 model we employed standard samples with a wide range of soybean constituent compositions: from 34% to 55% for protein, from 11% to 22% for oil and from 2% to 16% for moisture. Such calibrations are characterized by low standard errors and high degrees of correlation for all major soybean constituents. Morever, we obtained highly resolved NIR chemical images for selected regions of mature soybean embryos that allow for the quantitation of oil and protein components. Recent developments in high-resolution FT-NIR microspectroscopy extend the NIR sensitivity range to the picogram level, with submicron spatial resolution in the component distribution throughout intact soybean seeds and embryos. Such developments are potentially important for biotechnology applications that require rapid and ultra- sensitive analyses, such as those concerned with high-content microarrays in Genomics and Proteomics research. Other important applications of FT-NIR microspectroscopy are envisaged in biomedical research aimed at cancer prevention, the early detection of tumors by NIR-fluorescence, and identification of single cancer cells, or single virus particles in vivo by super-resolution microscopy/ microspectroscopy.
