Determination of Sulfites in Dried Fruits by Paper Spray Ionization Tandem Mass Spectrometry.

Sulfite, a widely used food additive, is subject to regulated labeling. The extraction of sulfite as the stable hydroxymethylsulfonate (HMS) form and its quantitative analysis by liquid chromatography-tandem mass spectrometry (LC-MS/MS) has been recognized for their good sensitivity, selectivity, and versatility across various food materials. This study aimed to develop a cost-effective and simpler method for sulfite quantitation, while maintaining the superior sensitivity and selectivity of mass spectrometry (MS). To achieve this, we introduced paper spray ionization (PSI), an ambient desorption ionization technique that could achieve the direct measurement of analytes without employing separation. We also employed a novel internal standard (IS) structurally similar to the analyte, replacing the more expensive isotopically labeled IS. Although the PSI-MS/MS method developed in this study exhibited slightly lower analytical performance compared to the conventional LC-MS/MS, it remained effective for sulfite analysis in dried fruits.

Sample Injection for Real-Time Analysis (SIRTA) Using GC-MS with Cold EI.

There is a continual demand for advanced methods and instruments for real-time analysis (RTA). Most of the current RTA techniques based on MS involve ambient desorption ionization technology. However, flow injection of liquid extracted samples is another option without added modifications or cost to existing LC-MS instruments. In this work, we introduce a new RTA approach named sample injection for real-time analysis (SIRTA) using GC-MS with Cold EI. In SIRTA, the standard GC column is replaced with a 1 m long 0.1 mm I.D. fused silica capillary that connects the GC injector to the MS transfer-line of Cold EI. Thus, SIRTA with Cold EI imposes no need for any additional instrumentation; hence, it is characterized by zero added cost. Like in flow injection in MS of LC-MS, the sample is dissolved in ∼1 mL methanol or another solvent. Subsequently, the vial is placed in the GC-MS autosampler while using a standard syringe for injection without any GC separation. The analysis takes merely 0.2-0.7 min, ensuring rapid and consecutive analyses. Unlike standard EI, Cold EI enables SIRTA by taking advantage of its fly through open ion source to avoid overwhelming the ion source during the elution of solvents while still providing enhanced molecular ions for nearly all analytes. In this study, we demonstrated SIRTA Cold EI analysis of 12 compounds and 7 mixtures, including various prescription and illicit drugs, cannabis and petroleum samples, and other synthetic organic compounds including those with molecular weight up to 800 g/mol.

Molecular ion formation on activated field emitters in atmospheric pressure field desorption mass spectrometry

Atmospheric pressure field desorption (APFD) mass spectrometry (MS) has recently been explored as a new contribution to the field of ambient desorption/ionization (ADI). Depending on the selected polarity applied to the field emitter, ionic and polar analytes were demonstrated to deliver positive as well as negative ions. Whereas this recent study solely reported on the formation of even-electron ions of either polarity, the present work on APFD-MS demonstrates the abundant formation of positive molecular ions, M+•, from polycyclic aromatic compounds. Molecular ions were formed on and desorbed from standard 13-µm activated tungsten wire emitters at atmospheric pressure. The commercial field emitters were positioned at about 2 mm distance in front of the atmospheric pressure interface of a Fourier transform-ion cyclotron resonance (FT-ICR) mass spectrometer and the entrance electrode of the interface was set to –4.5 to –5.5 kV with respect to the emitter. Emitter-disrupting electric discharges did normally not occur under these conditions. The electric field strengths achieved at the dendritic microneedles were sufficient to allow for the abundant formation of M+• ions of various polycyclic aromatic compounds such as benzo[a]pyrene, anthracene, fluoranthene, 1,1,4,4-tetraphenyl-butadiene, and 1-aza-[6]helicene. In case of the extremely basic 1-aza-[6]helicene protonation strongly competed with molecular ion formation and tended to suppress the field ionization process. Toward Depth-Resolved Analysis of Plant Metabolites by Nanospray Desorption Electrospray Ionization Mass Spectrometry.

Nanospray desorption electrospray ionization (nano-DESI) is one of the ambient desorption ionization methods for mass spectrometry (MS), and it utilizes a steady-state liquid junction formed between two microcapillaries to directly extract analytes from sample surfaces with minimal sample damage. In this study, we employed nano-DESI MS to perform a metabolite fingerprinting analysis directly from a Hypericum leaf surface. Moreover, we investigated whether changes in metabolite fingerprints with time can be related to metabolite distribution according to depth. From a raw Hypericum leaf, the mass spectral fingerprints of key metabolites, including flavonoids and prenylated phloroglucinols, were successfully obtained using ethanol as a nano-DESI solvent, and the changes in their intensities were observed with time via full mass scan experiments. In addition, the differential extraction patterns of the obtained mass spectral fingerprints were clearly visualized over time through selected ion monitoring and pseudo-selected reaction monitoring experiments. To examine the correlation between the time-dependent changes in the metabolite fingerprints and depth-wise metabolite distribution, we performed a nano-DESI MS analysis against leaves whose surface layers were removed multiple times by forming polymeric gum Arabic films on their surfaces, followed by detaching. The preliminary results showed that the changes in the metabolite fingerprints according to the number of peelings showed a similar pattern with those obtained from the raw leaves over time.

Optical and mass-spectral characterization of mixed-gas flowing atmospheric-pressure afterglow sources

Plasma-based ambient desorption/ionization (ADI) sources for mass spectrometry (MS) often use helium as the primary discharge gas due to the large reaction cross section of excited helium species with atmospheric gases, which ultimately leads to efficient reagent-ion formation. However, some studies have shown that mixed-gas plasmas provide unique advantages. For instance, our group showed that a helium‑oxygen flowing atmospheric-pressure afterglow (He:O2-FAPA) source yielded enhanced ion signals for small polar analytes, but compounds with aromatic rings underwent chemical modification to produce pyrylium species. Here, it is shown that the addition of H2 to the helium FAPA gas significantly decreased the reagent-ion signals, but analyte-ion signal was not affected to the same degree. In addition, mass spectra obtained with He:H2-FAPA were chemically cleaner with fewer ions stemming from analyte species and produced less oxidation of analytes. Addition of nitrogen increased the abundance of negative reagent ions (e.g., NO2− and NO3−) which led to enhanced ion signal for RDX. To better understand these unique advantages of a mixed-gas FAPA, the impact of molecular-gas (O2, N2, or H2) addition on the optical-emission spectra from the discharge was also measured. Spatially resolved emission was obtained from the anode- and negative-glow regions. In general, addition of small fractions (~0.1%) of molecular gases decreased helium and OH emission intensities. Atomic oxygen and hydrogen emission increased with the addition of O2 and H2, respectively. Furthermore, emission characteristics of N2, N2+, and NO with respect to molecular gas composition on He-FAPA were measured. Fundamental plasma parameters, such as OH rotational temperature (Trot), were also calculated for these mixed-gas systems.

Inexpensive Ultrasonic Nebulization Coupled with Direct Current Corona Discharge Ionization Mass Spectrometry for Liquid Samples and Its Fundamental Investigations.

The concept of direct mass-spectrometric analysis, especially exploited by ambient desorption/ionization (ADI) methods, provides numerous means for convenient sample analysis. While many simple and versatile ionization sources have been developed, challenges lay in achieving efficient sample introduction. In previous work, a sample introduction method employing direct current corona discharge (CD) coupled to a surface acoustic wave nebulization (SAWN) device enhanced sampling performance for both polar and nonpolar analytes by up to 4 orders of magnitude. In fact, the SAWN-CD method generated a multiply charged peptide ion signal comparable to that of conventional ESI. Unfortunately, the high cost of the SAWN devices themselves limits their accessibility. Herein, we report on an analogous implementation of CD with an inexpensive ultrasonic nebulizer (USN) on the basis of a commercial room humidifier demonstrating equivalent exemplary performance. We subsequently compare the two methods of SAWN-CD and USN-CD in a screening application of milk for the detection of two antibiotic drugs, ciprofloxacin and ampicillin. Finally, we further investigate the relative softness of these CD-coupled acoustic nebulization methods in comparison to that of ESI using a survival yield study of the thermometer ion nitrobenzylpyridinium.

Rapid drug detection in whole blood droplets using a desorption electrospray ionization static profiling approach - a proof-of-concept.

The introduction of desorption electrospray ionization (DESI) - and ambient desorption/ionization (ADI) ion sources in general - in the 2000s has opened new possibilities for mass spectrometric (MS) analyses of biological sample surfaces. DESI allows for a rapid screening of solid samples because no sample preparation is needed and the analysis is performed at atmospheric pressure. In the present study, we used DESI as an ion source for the rapid detection of a small molecule in blood droplets deposited on glass slides. Blood was spiked with different concentrations of a model drug, mebendazole. One microliter blood droplets of each preparation were deposited on the surface of a glass slide and analyzed by DESI, either in imaging or profiling mode. The results suggested that DESI imaging mode was not appropriate for the detection of mebendazole in blood droplets as an initial solvation time was necessary before the obtention of signal. A profiling approach consisting of analyzing a single position of the blood droplet was used for further analysis and allowed mebendazole to be detected in the fg range and to monitor the volume of sample analyzed. The study suggests that profiling mode at a single position is adequate for DESI analyses in whole blood droplets. This proof-of-concept study illustrates the potential of DESI profiling as a possible alternative to liquid chromatography/MS analyses of whole blood, when analyses are needed within a restricted time. Rapid detection methods in blood at atmospheric pressure may find interesting applications in the fields of toxicology and pharmacology.

Interpretation of Ionization Mechanism Responsible for Reagent Ion and Analyte Formation in Microwave-Induced Plasma Desorption Ionization Mass Spectrometry.

Ambient desorption/ionization (ADI) sources coupled to mass spectrometer have gained increasing interest in the field of analytical chemistry for its fast and direct analysis of samples. Among many ADI sources, plasma-based ADI sources are an important branch. Despite its extensive use in mass spectrometry analysis, the ionization mechanism of these sources still remain uncertain. The study on ionization mechanism is of great significance to optimize the design of ion sources and to improve ionization efficiency. In this study, targeted research on a better understanding of afterglow distance effects on ionization process was conducted. Based on the quantified signal expression of reagent ions in mass spectrum, the concept that optimal atmospheric analysis distance of plasma ADI source is defined for the first time. From the perspective of mutual restriction effect between atmospheric components, the formation progress of reagent ions was visually revealed in detail, which involved the initial step of forming precursor reagent ions, the clusters reaction for increasing production of reagent ions, and the matrix effect results in reagent ion depletion. The formation mechanism of reagent ions further clarified the explicit reason for abundant reagent ions generated at an optimal distance. Most importantly, the analyte analysis results verified the significant impact of appropriate distance on ionization efficiency in afterglow region. It was confirmed that the quantity and type of reagent ions intimately influenced the status of analyte ions in mass spectrum.

A mechanism study of positive ionization processes in flowing atmospheric-pressure afterglow (FAPA) ambient ion source with controlled plasma and ambient conditions

Although plasma based ambient desorption/ionization (ADI) sources have been widely used for direct analysis of complex samples, mass spectrometric imaging, high throughput screening etc., the ionization mechanism of plasma-based ADI remains a mystery by now. In this report, a targeted study was conducted aiming at a better understanding of the ionization processes of plasma-based ambient desorption ionization source. As a representative of ambient desorption ionization source, an FAPA source was used and modified as a test platform to control the plasma discharge parameters and ambient ionization environment such as discharge gases, environmental gases and sampling conditions. Based on the ionization results from different ambient ionization conditions, a new mechanism was proposed to reveal the nature of regent ion production of FAPA. At the same time, the effect of buffer gas was investigated. For the first time, the multi-clustered hydronium ions formed by the massive water vapor in the air were explored to clarify reasons for the occurrence of selective ionization and the factors affecting ionization efficiency in such complex events. In addition, the formation of molecular ions and relevant reagent ions was speculated based on experimental observations.

Coupling laser desorption with corona beam ionization for ambient mass spectrometric analysis of solution and powder samples

An infrared (940nm) laser desorption coupled with corona beam ionization (LD-CBI) technique was developed for expanding the application of the ambient mass spectrometry (AMS). A black ceramic sample plate was employed to facilitate the non-resonance absorption of laser energy. Solution and powder samples were immobilized in the shallow depression on ceramic plate and desorbed precisely since there was not gas disturbing. The dimer ion (m/z 1216.51) of reserpine was detected without any matrix assistance. In comparison with similar APCI-related ambient technique, the upper mass rang of analyte was broadened significantly. The feasibility of LD-CBI approach was demonstrated with 10 model pesticides. Typically, the limit of detection (LOD) of Malathion was as low as 6pg in the selected reaction monitoring (SRM) mode. The value is 166 times lower than the results from preceding technique desorption corona beam ionization (DCBI). As an APCI related ambient desorption ionization approach, LD-CBI technique could be a well complementary tool for direct detection of solution and powder samples.

The Effects of Added Hydrogen on Noble Gas Discharges Used as Ambient Desorption/Ionization Sources for Mass Spectrometry.

We demonstrate the effectiveness of using hydrogen-doped argon as the support gas for the dielectric barrier discharge (DBD) ambient desorption/ionization (ADI) source in mass spectrometry. Also, we explore the chemistry responsible for the signal enhancement observed when using both hydrogen-doped argon and hydrogen-doped helium. The hydrogen-doped argon was tested for five analytes representing different classes of molecules. Addition of hydrogen to the argon plasma gas enhanced signals for gas-phase analytes and for analytes coated onto glass slides in positive and negative ion mode. The enhancements ranged from factors of 4 to 5 for gas-phase analytes and factors of 2 to 40 for coated slides. There was no significant increase in the background. The limit of detection for caffeine was lowered by a factor of 79 using H2/Ar and 2 using H2/He. Results are shown that help explain the fundamental differences between the pure-gas discharges and those that are hydrogen-doped for both argon and helium. Experiments with different discharge geometries and grounding schemes indicate that observed signal enhancements are strongly dependent on discharge configuration. Graphical Abstract ᅟ.

Humidity Effects on Fragmentation in Plasma-Based Ambient Ionization Sources.

Post-plasma ambient desorption/ionization (ADI) sources are fundamentally dependent on surrounding water vapor to produce protonated analyte ions. There are two reports of humidity effects on ADI spectra. However, it is unclear whether humidity will affect all ADI sources and analytes, and by what mechanism humidity affects spectra. Flowing atmospheric pressure afterglow (FAPA) ionization and direct analysis in real time (DART) mass spectra of various surface-deposited and gas-phase analytes were acquired at ambient temperature and pressure across a range of observed humidity values. A controlled humidity enclosure around the ion source and mass spectrometer inlet was used to create programmed humidity and temperatures. The relative abundance and fragmentation of molecular adduct ions for several compounds consistently varied with changing ambient humidity and also were controlled with the humidity enclosure. For several compounds, increasing humidity decreased protonated molecule and other molecular adduct ion fragmentation in both FAPA and DART spectra. For others, humidity increased fragment ion ratios. The effects of humidity on molecular adduct ion fragmentation were caused by changes in the relative abundances of different reagent protonated water clusters and, thus, a change in the average difference in proton affinity between an analyte and the population of water clusters. Control of humidity in ambient post-plasma ion sources is needed to create spectral stability and reproducibility.

Effect of internal and external conditions on ionization processes in the FAPA ambient desorption/ionization source.

Ambient desorption/ionization (ADI) sources coupled to mass spectrometry (MS) offer outstanding analytical features: direct analysis of real samples without sample pretreatment, combined with the selectivity and sensitivity of MS. Since ADI sources typically work in the open atmosphere, ambient conditions can affect the desorption and ionization processes. Here, the effects of internal source parameters and ambient humidity on the ionization processes of the flowing atmospheric pressure afterglow (FAPA) source are investigated. The interaction of reagent ions with a range of analytes is studied in terms of sensitivity and based upon the processes that occur in the ionization reactions. The results show that internal parameters which lead to higher gas temperatures afforded higher sensitivities, although fragmentation is also affected. In the case of humidity, only extremely dry conditions led to higher sensitivities, while fragmentation remained unaffected.

Development of a Mass Spectrometry Sampling Probe for Chemical Analysis in Surgical and Endoscopic Procedures

A sampling probe based on ambient desorption ionization was designed for in vivo chemical analysis by mass spectrometry in surgical and endoscopic procedures. Sampling ionization of analytes directly from tissue was achieved by sealing the sampling tip against the tissue surface without allowing leakage of the auxiliary gas used for desorption ionization. The desorbed charged species were transferred over a long distance (up to 4 m) through a flexible tube of internal diameter as small as 1/16 in. to the inlet of the mass spectrometer used for analysis. The conditions used for desorption electrospray ionization (DESI) were optimized to achieve biocompatibility for clinical applications while obtaining adequate efficiency for the analysis. This optimization involved the removal of high voltage and use of pure water as a spray solvent instead of the organic solvents or aqueous mixtures normally used. Improved sensitivity was achieved under these conditions by increasing the gas flow rate in the transfer tube. The destructive effect on tissue surfaces associated with typical desorption ionization was avoided by altering the local gas dynamics in the sample area without compromising the overall analysis efficiency.

The Effects of Added Hydrogen on a Helium Atmospheric-Pressure Plasma Jet Ambient Desorption/Ionization Source

We present mass spectrometric data demonstrating the effect that hydrogen has on a helium-based dielectric-barrier discharge (DBD) atmospheric-pressure plasma jet used as an ambient desorption/ionization (ADI) source. The addition of 0.9% hydrogen to the helium support gas in a 35-W plasma jet increased signals for a range of test analytes, with enhancement factors of up to 68, without proportional increases in background levels. The changes in signal levels result from a combination of changes in the desorption kinetics from the surface and increased ion production in the gas phase. The enhancement in ADI-MS performance despite the quenching of key plasma species reported in earlier studies suggests that ionization with a H2/He plasma jet is the result of an alternate mechanism involving the direct generation of ionized hydrogen.

Coupling of planar chromatography with Direct Analysis in Real Time mass spectrometry

Abstract Direct Analysis in Real Time mass spectrometry (DART-MS) is an emerging and rapidly developing area of ambient desorption ionization mass spectrometric techniques. Its coupling with planar chromatography is especially promising, as compared to other ambient desorption ionization techniques, because it does not require the use of liquids that may distort the shape of a spot by diffusion effects. In the first publications on TLC/HPTLC-DART-MS, due to the fixed, horizontally aligned supply of the gas flow from the DART ionization source to the MS inlet, the introduction of HPTLC/TLC plates as cut strips was inconvenient for quantitation, and the repeatability was very low due to the manual positioning. Recently a new version of the DART ion source was suggested, which allows adjusting the angle of the DART gas stream and the use of a motorized rail, thereby, improving highly the capabilities of TLC/HPTLC-DART-MS. This comprehensive review describes the development and analytical capabilities of TLC/HPTLC-DART-MS, and the general DART-MS perspectives for surface analysis or imaging MS.

Novel approaches to analysis of 3-chloropropane-1,2-diol esters in vegetable oils

A sensitive and accurate method utilizing ultrahigh performance liquid chromatography (U-HPLC) coupled to high resolution mass spectrometry based on orbitrap technology (orbitrapMS) for the analysis of nine 3-chloropropane-1,2-diol (3-MCPD) diesters in vegetable oils was developed. To remove the interfering triacylglycerols that induce strong matrix effects, a clean-up step on silica gel column was used. The quantitative analysis was performed with the use of deuterium-labeled internal standards. The lowest calibration levels estimated for the respective analytes ranged from 2 to 5 μg kg(-1). Good recovery values (89-120%) and repeatability (RSD 5-9%) was obtained at spiking levels of 2 and 10 mg kg(-1). As an alternative, a novel ambient desorption ionization technique, direct analysis in real time (DART), hyphenated with orbitrapMS, was employed for no separation, high-throughput, semi-quantitative screening of 3-MCPD diesters in samples obtained by chromatographic fractionation. Additionally, the levels of 3-MCPD diesters measured in reallife vegetable oil samples (palm oil, sunflower oil, rapeseed oil) using both methods are reported. Relatively good agreement of the data generated by U-HPLC-orbitrapMS and DART-orbitrapMS were observed. With regard to a low ionization yield achieved for 3-MCPD monoesters, the methods presented in this paper were not yet applicable for the analysis of these contaminants at the naturally occurring levels.

Ambient desorption ionization mass spectrometry (DART, DESI) and its bioanalytical applications

In recent years, ambient desorption ionization techniques for mass spectrometry were introduced. Among them, the most established techniques are Direct Analysis in Real Time (DART) and Desorption Electrospray Ionization (DESI). Therefore, the current review focuses on the bioanalytical applications of ambient desorption ionization techniques by the example of DART and DESI mass spectrometry. The potential and also limitations of both ambient mass spectrometry (MS) techniques in such areas, as identification and quantitation of small molecules, coupling DART-MS and DESI-MS with planar chromatography, protein/peptide analysis, as well as molecular imaging applications, are discussed.

Detection of Positive and Negative Ions from a Flowing Atmospheric Pressure Afterglow Using a Mattauch-Herzog Mass Spectrograph Equipped with a Faraday-Strip Array Detector

An ambient desorption/ionization (ADI) source, known as the flowing atmospheric pressure afterglow (FAPA), has been coupled to a Mattauch-Herzog mass spectrograph (MHMS) equipped with a focal plane camera (FPC) array detector. The FAPA ionization source enables direct mass spectral analysis of solids, liquids, and gases through either positive or negative ionization modes. In either case, spectra are generally simple with dominant peaks being the molecular ions or protonated molecular ions. Use of the FAPA source with the MHMS allows the FPC detector to be characterized for the determination of molecular species, whereas previously only atomic mass spectrometry (MS) has been demonstrated. Furthermore, the FPC is shown to be sensitive to negative ions without the need to change any detector parameters. The analysis of solid, liquid, and gaseous samples through positive and negative ionization is demonstrated with detection limits (1-25 fmol/s, approximately 0.3-10 pg of analyte per mL of helium) surpassing those obtained with the FAPA source coupled to a time-of-flight mass analyzer.

Low-Temperature Plasma Probe for Ambient Desorption Ionization

A low-temperature plasma (LTP) probe has been developed for ambient desorption ionization. An ac electric field is used to induce a dielectric barrier discharge through use of a specially designed electrode configuration. The low-temperature plasma is extracted from the probe where it interacts directly with the sample being analyzed, desorbing and ionizing surface molecules in the ambient environment. This allows experiments to be performed without damage to the sample or underlying substrate and, in the case of biological analysis on skin surfaces, without electrical shock or perceptible heating. Positive or negative ions are produced from a wide range of chemical compounds in the pure stateand as mixtures in the gaseous, solution, or condensed phases, using He, Ar, N2, or ambient air as the discharge gas. Limited fragmentation occurs, although it is greater in the cases of the molecular than the atomic discharge gases. The effectiveness of the LTP probe has been demonstrated by recording characteristic mass spectra and tandem mass spectra of samples containing hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX) and 2,4,6-trinitrotoluene (TNT) from poly(tetrafluoroethylene) (PTFE) surfaces where limits of detection are as low as 5 pg. Other performance characteristics, when using a commercial ion trap mass spectrometer, include 3-4 orders of magnitude linear dynamic range in favorable cases. Demonstration applications include direct analysis of cocaine from human skin, determination of active ingredients directly in drug tablets, and analysis of toxic and therapeutic compounds in complex biological samples. Ionization of chemicals directly from bulk aqueous solution has been demonstrated, where limits of detection are as low as 1 ppb. Large surface area sampling and control of fragmentation by a simple adjustment of the electrode configuration during operation are other demonstrated characteristics of the method.
