Oxidative Stress and Air Pollution: Its Impact on Chronic Respiratory Diseases

Redox regulation participates in the control of various aspects of metabolism. Reactive oxygen and nitrogen species participate in many reactions under physiological conditions. When these species overcome the antioxidant defense system, a distressed status emerges, increasing biomolecular damage and leading to functional alterations. Air pollution is one of the exogenous sources of reactive oxygen and nitrogen species. Ambient airborne particulate matter (PM) is important because of its complex composition, which includes transition metals and organic compounds. Once in contact with the lungs' epithelium, PM components initiate the synthesis of inflammatory mediators, macrophage activation, modulation of gene expression, and the activation of transcription factors, which are all related to the physiopathology of chronic respiratory diseases, including cancer. Even though the pathophysiological pathways that give rise to the development of distress and biological damage are not fully understood, scientific evidence indicates that redox-dependent signaling pathways are involved. This article presents an overview of the redox interaction of air pollution inside the human body and the courses related to chronic respiratory diseases.

Long-term exposure to PM2.5 aggravates pulmonary fibrosis and acute lung injury by disrupting Nrf2-mediated antioxidant function

Epidemiological studies have indicated that exposure to ambient air-borne fine particulate matter (PM2.5) is associated with many cardiopulmonary diseases; however, the underlying pathological mechanisms of PM2.5-induced lung injury remain unknown. In this study, we aimed to assess the impact of acute or prolonged exposure to water-insoluble fractions of PM2.5 (PM2.5 particulate) on lung injury and its molecular mechanisms. Balb/c mice were randomly exposed to PM2.5 once (acute exposure) or once every three days for a total of 6 times (prolonged exposure). Lung, BALF and blood samples were collected, and pulmonary pathophysiological alterations were analyzed. Nrf2 knockout mice were adapted to assess the involvement of Nrf2 in lung injury, and transcriptomic analysis was performed to delineate the mechanisms. Through transcriptomic analysis and validation of Nrf2 knockout mice, we found that acute exposure to PM2.5 insoluble particulates induced neutrophil infiltration-mediated airway inflammation, whereas prolonged exposure to PM2.5 insoluble particulate triggered lung fibrosis by decreasing the transcriptional activity of Nrf2, which resulted in the downregulated expression of antioxidant-related genes. In response to secondary LPS exposure, prolonged PM2.5 exposure induced more severe lung injury, indicating that prolonged PM2.5 exposure induced Nrf2 inhibition weakened its antioxidative defense capacity against oxidative stress injury, leading to the formation of pulmonary fibrosis and increasing its susceptibility to secondary bacterial infection.

Respiratory and cardiovascular effects of ambient particulate matter from dust storm and non-dust storm periods in Kuwait

Epidemiological studies demonstrate a positive association between daily changes in concentrations of ambient airborne particulate matter (PM) and adverse respiratory and cardiovascular health effects. However, physicochemical properties of PM can vary greatly across geographical, atmospheric, and temporal conditions and influence the relative toxicity of airborne PM. The purpose of this study was to investigate the adverse pulmonary and cardiovascular health effects of ambient PM collected from discrete sampling sites in Kuwait during dust storm (DS) and non-dust storm (NDS) conditions. Collected dust samples were characterized for their chemical composition using atomic absorption, GC–MS, and HPLC–MS analyses. Male BALB/cJ mice were exposed to 100 µg of either NDS or dust storm (DS) PM in 50 µl of PBS by oropharyngeal aspiration. Lung function was measured and bronchoalveolar lavage was conducted at 1, 7, and 14 days post-exposure. Ischemia–reperfusion injury was performed 24 h after exposures by obstructing the left main coronary artery approximately 4 mm distal to its origin for 20 min, followed by 2 h. of reperfusion. Exposure to either NDS or DS PM resulted in airway hyperresponsiveness to acetylcholine compared to PBS controls. Total protein and cells in BAL fluid were elevated in both dust groups one day after exposure; however, DS PM induced a greater increase in cell numbers than did NDS PM, particularly in neutrophils, eosinophils, and lymphocytes. Representative lung sections exhibited positive staining for mucus in large airways at 7 days which resolved by 14 days in dust storm-exposed mice but persisted in NDS-exposed animals. Our findings suggest that NDS PM may be more effective in producing an adaptive immune response, while the early inflammation induced by DS PM may better resolve. We also observed a prolonged airway mucus response after exposure to NDS PM, suggesting it may produce more asthma-like features than dust storm PM. PM-induced changes to cardiac ischemia–reperfusion injury were not observed in this study. The lack of cardiovascular response may have been due to the limited exposure and single time point used in this study.

Spatial Extent and Distribution of Ambient Airborne Particulate Matter (PM2.5) in Selected Land Use Sites in Nairobi, Kenya.

Air pollution is one of the most important environmental and public health concerns worldwide. Urban air pollution has been increasing since the industrial revolution due to rapid industrialization, mushrooming of cities, and greater dependence on fossil fuels in urban centers. Particulate matter (PM) is considered to be one of the main aerosol pollutants that causes a significant adverse impact on human health. Low-cost air quality sensors have attracted attention recently to curb the lack of air quality data which is essential in assessing the health impacts of air pollutants and evaluating land use policies. This is mainly due to their lower cost in comparison to the conventional methods. The aim of this study was to assess the spatial extent and distribution of ambient airborne particulate matter with an aerodynamic diameter less than 2.5 μm (PM2.5) in Nairobi City County. Seven sites were selected for monitoring based on the land use type: high- and low-density residential, industrial, agricultural, commercial, road transport, and forest reserve areas. Calibrated low-cost sensors and cyclone samplers were used to monitor PM2.5 concentration levels and gravimetric measurements for elemental composition of PM2.5, respectively. The sensor percentage accuracy for calibration ranged from 81.47% to 98.60%. The highest 24-hour average concentration of PM2.5 was observed in Viwandani, an industrial area (111.87 μg/m³), and the lowest concentration at Karura (21.25 μg/m³), a forested area. The results showed a daily variation in PM2.5 concentration levels with the peaks occurring in the morning and the evening due to variation in anthropogenic activities and the depth of the atmospheric boundary layer. Therefore, the study suggests that residents in different selected land use sites are exposed to varying levels of PM2.5 pollution on a regular basis, hence increasing the potential of causing long-term health effects.

Fine particulate matter-induced lung inflammation is mediated by pyroptosis in mice

BackgroundExposure to ambient air-borne fine particulate matter (PM2.5) increases respiratory morbidity and mortality. The mechanisms underlying PM2.5-induced adverse effects remain unclear. This study aimed to uncover the molecular mechanisms of PM2.5-induced lung toxicity using a mouse model. MethodsScanning electron microscopy and inductively coupled plasma mass spectrometry were used to examine and analyze PM2.5 morphology and element compositions, respectively. Twenty four male mice were randomly divided into three groups: control (PBS), PM2.5 (4.0 mg/kg b.w.), and PM2.5 + Z-YVAD-FMK. In the latter group, the pan-caspase inhibitor (Z-YVAD-FMK) was intraperitoneally injected into mice at a dose of 12.5 mg/kg body weight prior to intratracheal instillation of PM2.5 (4.0 mg/kg b.w.) every other day for a total of 3 times (n = 8 in each group). Bronchoalveolar lavage fluids (BALFs) were collected 24 h after the last instillation of PM2.5. Levels of total proteins (TP), lactate dehydrogenase (LDH), IL-1β and IL-18 were analyzed for biomarkers of cell injury and inflammation. Additionally, histological alterations of lung tissues were assessed by hematoxylin-eosin staining. mRNA and protein expression of Caspase1, NLRP3 and GSDMD were examined by real-time fluorescent quantitative PCR and immunohistochemical staining. ResultsExposure to PM2.5 increased levels of TP, LDH, IL-1β, IL-18 and inflammatory cell counts in lung. The mRNA and protein expression of Caspase1, NLRP3 and GSDMD were increased. Inhibition of the NALRP3/Caspase-1 signaling pathway ameliorated PM2.5-induced lung injury and inflammation, partially through suppressing pyroptosis in lung. ConclusionPM2.5 exposure induces lung injury and inflammation, which is mediated by the NALRP3/Caspase-1 signaling pathway.

Simple Model for Predicting Time Series Soiling of Photovoltaic Panels

Results from a model to predict soiling losses of photovoltaic (PV) arrays are presented. The model uses ambient airborne particulate matter concentrations, both PM $_{10}$ and PM $_{2.5}$ , the tilt of the PV array including if the array is tracking, and rain data to estimate soiling losses over time. The model uses relationships between average airborne PM concentrations and dust accumulation, rain removal of accumulated dust, and the accumulated dust and transmission loss that have been developed in the literature to estimate soiling losses in a time series. Multiple model runs were performed and were compared with measured soiling data at seven locations. The model was run with both typical Meteorological Year rain data and with recorded rain data from Oregon State University's PRISM database. Each model was run with variable deposition velocities, static deposition velocities, and static settling velocities. When the model was run with recorded rain data and static settling velocities the results matched the general slopes, frequencies, and magnitudes of soiling losses when compared with measured soiling data. Overall, the simple model demonstrates the ability at accurately simulating soiling.

Air pollution, oxidative stress, and exacerbation of autoimmune diseases.

A number of epidemiological studies have shown a strong association between exposure to ambient airborne particulate matter (PM 2.5, PM < 1.0) and lung or cardiovascular diseases characterised by high mortality and morbidity. However, much less is known about the role of air pollution in the pathogenesis of autoimmune diseases, which constitutes a significant problem in modern society.This paper summarises the state of current research regarding the influence of PM on the development and/or progression of autoimmune diseases. A brief review of the great body of research concerning pathogenesis of autoimmune disorders is presented. Then, the scope of our review is narrowed to the research related to the impact of particulate matter on oxidative and nitrosative stress, as well as exacerbation of chronic inflammation, because they can contribute to the development of autoimmune diseases. Moreover, we discuss the impact of various components of PM (metal, organic compounds) on PM toxicity and the ability to generate oxidants.

DDAH1 plays dual roles in PM2.5 induced cell death in A549 cells

BackgroundDimethylarginine dimethylaminohydrolase 1 (DDAH1) is an enzyme that can degrade asymmetric dimethylarginine (ADMA), an endogenous nitric oxide synthase (NOS) inhibitor. Emerging evidence suggests that alterations in the ADMA–DDAH1 pathway are involved in environmental pollution induced airway inflammation. However, the role of DDAH1 in protection against cytotoxicity of ambient airborne particulate matter is unclear. MethodsWe examined the influence of DDAH1 expression on oxidative stress and cell apoptosis in human type II alveolar epithelial A549 cells exposed to PM2.5 (particulate matter with an aerodynamic diameter less than 2.5μM). ResultsWe found that PM2.5 exposure for 48h significantly decreased DDAH1 expression. However, knockdown of DDAH1 prior to PM2.5 exposure actually attenuated the cytotoxicity of PM2.5. Cytoprotection in DDAH1 deficient cells was due to increased reactive oxygen species, activation of PI3K–AKT and mitogen-activated protein kinase (MAPK) pathways, subsequent activation of nuclear factor erythroid-2-related factor 2 (Nrf2) and this caused a subsequent reduction in PM2.5 induced oxidative stress relative to control. DDAH1 depletion also repressed the induction of inducible NOS (iNOS) in PM2.5-exposed cells and knockdown of iNOS protected cells against PM2.5 induced cell death. Interestingly, overexpression of DDAH1 also exerted a protective effect against the cytotoxicity of PM2.5 and this was associated with a reduction in oxidative stress and upregulation of the anti-apoptotic protein Bcl-2. ConclusionsOur data indicate that DDAH1 plays dual roles in protection against cytotoxicity of PM2.5 exposure, apparently by limiting PM2.5 induced oxidative stress. General significanceOur findings reveal new insights into the role(s) of the DDAH1/ADMA in pulmonary protection against airborne pollutants. Accuracy and precision of &amp;lt;sup&amp;gt;14&amp;lt;/sup&amp;gt;C-based source apportionment of organic and elemental carbon in aerosols using the Swiss_4S protocol

Abstract. Aerosol source apportionment remains a critical challenge for understanding the transport and aging of aerosols, as well as for developing successful air pollution mitigation strategies. The contributions of fossil and non-fossil sources to organic carbon (OC) and elemental carbon (EC) in carbonaceous aerosols can be quantified by measuring the radiocarbon (14C) content of each carbon fraction. However, the use of 14C in studying OC and EC has been limited by technical challenges related to the physical separation of the two fractions and small sample sizes. There is no common procedure for OC/EC 14C analysis, and uncertainty studies have largely focused on the precision of yields. Here, we quantified the uncertainty in 14C measurement of aerosols associated with the isolation and analysis of each carbon fraction with the Swiss_4S thermal–optical analysis (TOA) protocol. We used an OC/EC analyzer (Sunset Laboratory Inc., OR, USA) coupled to a vacuum line to separate the two components. Each fraction was thermally desorbed and converted to carbon dioxide (CO2) in pure oxygen (O2). On average, 91 % of the evolving CO2 was then cryogenically trapped on the vacuum line, reduced to filamentous graphite, and measured for its 14C content via accelerator mass spectrometry (AMS). To test the accuracy of our setup, we quantified the total amount of extraneous carbon introduced during the TOA sample processing and graphitization as the sum of modern and fossil (14C-depleted) carbon introduced during the analysis of fossil reference materials (adipic acid for OC and coal for EC) and contemporary standards (oxalic acid for OC and rice char for EC) as a function of sample size. We further tested our methodology by analyzing five ambient airborne particulate matter (PM2.5) samples with a range of OC and EC concentrations and 14C contents in an interlaboratory comparison. The total modern and fossil carbon blanks of our setup were 0.8 ± 0.4 and 0.67 ± 0.34 μg C, respectively, based on multiple measurements of ultra-small samples. The extraction procedure (Swiss_4S protocol and cryo-trapping only) contributed 0.37 ± 0.18 μg of modern carbon and 0.13 ± 0.07 μg of fossil carbon to the total blank of our system, with consistent estimates obtained for the two laboratories. There was no difference in the background correction between the OC and EC fractions. Our setup allowed us to efficiently isolate and trap each carbon fraction with the Swiss_4S protocol and to perform 14C analysis of ultra-small OC and EC samples with high accuracy and low 14C blanks.

Characteristics of airborne bacteria in Mumbai urban environment

Components of biological origin constitute small but a significant proportion of the ambient airborne particulate matter (PM). However, their diversity and role in proinflammatory responses of PM are not well understood. The present study characterizes airborne bacterial species diversity in Mumbai City and elucidates the role of bacterial endotoxin in PM induced proinflammatory response in ex vivo. Airborne bacteria and endotoxin samples were collected during April–May 2010 in Mumbai using six stage microbial impactor and biosampler. The culturable bacterial species concentration was measured and factors influencing the composition were identified by principal component analysis (PCA). The biosampler samples were used to stimulate immune cells in whole blood assay. A total of 28 species belonging to 17 genera were identified. Gram positive and spore forming groups of bacteria dominated the airborne culturable bacterial concentration. The study indicated the dominance of spore forming and human or animal flora derived pathogenic/opportunistic bacteria in the ambient air environment. Pathogenic and opportunistic species of bacteria were also present in the samples. TNF-α induction by PM was reduced (35%) by polymyxin B pretreatment and this result was corroborated with the results of blocking endotoxin receptor cluster differentiation (CD14). The study highlights the importance of airborne biological particles and suggests need of further studies on biological characterization of ambient PM.

Measurement of Soluble and Total Hexavalent Chromium in the Ambient Airborne Particles in New Jersey.

Hexavalent chromium (Cr(VI)) in ambient airborne particulate matter (PM) is a known pulmonary carcinogen and may have both soluble and insoluble forms. The sum of the two forms is defined as total Cr(VI). Currently, there were no methods suitable for large-scale monitoring of total Cr(VI) in ambient PM. This study developed a method to measure total Cr(VI) in ambient PM. This method includes PM collection using a Teflon filter, microwave extraction with 3% Na2CO3-2% NaOH at 95°C for 60 minutes, and Cr(VI) analysis by 1,5-diphenylcarbazide colorimetry at 540 nm. The recoveries of total Cr(VI) were 119.5 ± 10.4% and 106.3 ± 16.7% for the Cr(VI)-certified reference materials, SQC 012 and SRM 2700, respectively. Total Cr(VI) in the reference urban PM (NIST 1648a) was 26.0 ± 3.1 mg/kg (%CV = 11.9%) determined by this method. The method detection limit was 0.33 ng/m3. This method and the one previously developed to measure ambient Cr(VI), which is soluble in pH ~9.0 aqueous solution, were applied to measure Cr(VI) in ambient PM10 collected from three urban areas and one suburban area in New Jersey. The total Cr(VI) concentrations were 1.05-1.41 ng/m3 in the winter and 0.99-1.56 ng/m3 in the summer. The soluble Cr(VI) concentrations were 0.03-0.19 ng/m3 in the winter and 0.12-0.37 ng/m3 in the summer. The summer mean ratios of soluble to total Cr(VI) were 14.3-43.7%, significantly higher than 4.2-14.4% in the winter. The winter concentrations of soluble and total Cr(VI) in the suburban area were significantly lower than in the three urban areas. The results suggested that formation of Cr(VI) via atmospheric chemistry may contribute to the higher soluble Cr(VI) concentrations in the summer.

PM2.5 induces Nrf2-mediated defense mechanisms against oxidative stress by activating PIK3/AKT signaling pathway in human lung alveolar epithelial A549 cells

It has been well documented in in vitro studies that ambient airborne particulate matter (PM) with an aerodynamic diameter less than 2.5 μm (PM(2.5)) is capable of inducing oxidative stress, which plays a key role in PM(2.5)-mediated cytotoxicity. Although nuclear factor erythroid-2-related factor 2 (Nrf2) has been shown to regulate the intracellular defense mechanisms against oxidative stress, a potential of the Nrf2-mediated cellular defense against oxidative stress induced by PM(2.5) remains to be determined. This study was aimed to explore the potential signaling pathway of Nrf2-mediated defense mechanisms against PM(2.5)-induced oxidative stress in human type II alveolar epithelial A549 cells. We exposed A549 cells to PM(2.5) particles collected from Beijing at a concentration of 16 μg/cm(2). We observed that PM(2.5) triggered an increase of intracellular reactive oxygen species (ROS) in a time-dependent manner during a period of 2 h exposure. We also found that Nrf2 overexpression suppressed and Nrf2 knockdown increased PM(2.5)-induced ROS generation. Using Western blot and confocal microscopy, we found that PM(2.5) exposure triggered significant translocation of Nrf2 into nucleus, resulting in AKT phosphorylation and significant transcription of ARE-driven phases II enzyme genes, such as NAD(P)H:quinone oxidoreductase (NQO-1), heme oxygenase-1 (HO-1), and glutamate-cysteine ligase catalytic subunit (GCLC) in A549 cells. Evaluation of signaling pathways showed that a phosphatidylinositol 3-kinase (PI3K) inhibitor (LY294002), but not an ERK 1/2 inhibitor (PD98059) or a p38 MAPK (SB203580), significantly down-regulated PM(2.5)-induced Nrf2 nuclear translocation and HO-1 mRNA expression, indicating PI3K/AKT is involved in the signaling pathway leads to the PM(2.5)-induced nuclear translocation of Nrf2 and subsequent Nrf2-mediated HO-1 transcription. Taken together, our results suggest that PM(2.5)-induced ROS may function as signaling molecules to activate Nrf2-mediated defenses, such as HO-1 expression, against oxidative stress induced by PM(2.5) through the PI3K/AKT signaling pathway.

Ginsenoside Rg1 reduces toxicity of PM2.5 on human umbilical vein endothelial cells by upregulating intracellular antioxidative state

Ambient airborne particulate matter (PM) is an important environmental pollutant responsible for many human diseases. Oxidative stress is suggested to be involved in PM-induced cell injury. The present study is designed to study unsalutary effects of the organic extracts of PM with an aerodynamic diameter of less than 2.5μm (PM2.5) and protective effect of Ginsenoside Rg1 (Rg1) against PM2.5 on human umbilical vein endothelial cells (HUVECs) in vitro. Cytotoxic effects of the organic extract PM2.5 on HUVECs were measured by means of HUVEC cell viability and the generation of intracellular reactive oxygen species (ROS). Expression of heme oxygenase-1(HO-1) and Nuclear factor-erythroid 2-related factor 2 (Nrf2) and Nrf2 cytoplasm–nucleus location were assayed. The present results showed that PM2.5 (50–800μg/ml) decreased HUVEC viability and increased intracellular generation of ROS and malondialdehyde (MDA) in a concentration dependent manner, but increased HO-1 expression without concentration dependence. Rg1 (10 and 40μg/ml) diminished PM2.5-induced HUVEC viability, decrease ROS and MDA generation, increased HO-1 and Nrf2 expression and promoted Nrf2 translocation to nucleus in a concentration dependent manner. These results suggested that organic extracts of PM2.5 increase oxidative stress and decrease cell viability; Rg1 antagonize PM2.5-induced excess oxidative stress; HO-1 expression increase and Nrf2 translocation to nucleus may be involved in the effects of both PM2.5 and Rg1 on HUVECs.

Cellular Uptake and Toxic Effects of Fine and Ultrafine Metal-Sulfate Particles in Human A549 Lung Epithelial Cells

Ambient airborne particulate matter is known to cause various adverse health effects in humans. In a recent study on the environmental impacts of coal and tire combustion in a thermal power station, fine crystals of PbSO(4) (anglesite), ZnSO(4)·H(2)O (gunningite), and CaSO(4) (anhydrite) were identified in the stack emissions. Here, we have studied the toxic potential of these sulfate phases as particulates and their uptake in human alveolar epithelial cells (A549). Both PbSO(4) and CaSO(4) yielded no loss of cell viability, as determined by the WST-1 and NR assays. In contrast, a concentration-dependent increase in cytotoxicity was observed for Zn sulfate. For all analyzed sulfates, an increase in the production of reactive oxygen species (ROS), assessed by the DCFH-DA assay and EPR, was observed, although to a varying extent. Again, Zn sulfate was the most active compound. Genotoxicity assays revealed concentration-dependent DNA damage and induction of micronuclei for Zn sulfate and, to a lower extent, for CaSO(4), whereas only slight effects could be found for PbSO(4). Moreover, changes of the cell cycle were observed for Zn sulfate and PbSO(4). It could be shown further that Zn sulfate increased the nuclear factor kappa-B (NF-κB) DNA binding activity and activated JNK. During our TEM investigations, no effect on the appearance of the A549 cells exposed to CaSO(4) compared to the nonexposed cells was observed, and in our experiments, only one CaSO(4) particle was detected in the cytoplasm. In the case of exposure to Zn sulfate, no particles were found in the cytoplasm of A549 cells, but we observed a concentration-dependent increase in the number and size of dark vesicles (presumably zincosomes). After exposure to PbSO(4), the A549 cells contained isolated particles as well as agglomerates both in vesicles and in the cytoplasm. Since these metal-sulfate particles are emitted into the atmosphere via the flue gas of coal-fired power stations, they may be globally abundant. Therefore, our study is of direct relevance to populations living near such power plants.

Water-insoluble fraction of airborne particulate matter (PM10) induces oxidative stress in human lung epithelial A549 cells

Exposure to ambient airborne particulate matter (PM) with an aerodynamic diameter less than 10 μm (PM10 ) links with public health hazards and increases risk for lung cancer and other diseases. Recent studies have suggested that oxidative stress is a key mechanism underlying the toxic effects of exposure to PM10 . Several components of water-soluble fraction of PM10 (sPM10 ) have been known to be capable of inducing oxidative stress in in vitro studies. In this study, we investigated if water-insoluble fraction of PM10 (iPM10 ) could be also capable of inducing oxidative stress and oxidative damage. Human lung epithelial A549 cells were exposed to 10 μg/mL of sPM10 , iPM10 or total PM10 (tPM10 ) preparation for 24 h. Here, we observed that all three PM10 preparations reduced cell viability and induced apoptotic cell death in A549 cells. We further found that, similar to the exposure to sPM10 and tPM10 , the intracellular level of hydrogen peroxide (H2 O2 ) in the iPM10 -exposed cells was increased significantly; meanwhile the activity of catalase was decreased significantly as compared with the unexposed control cells, resulting in significant DNA damage. Our data obtained from inductively coupled plasma-mass spectrometry (ICP-MS) assays showed that iron is the most abundant metal in all three PM10 preparations. Thus, we have demonstrated that, similar to sPM10 , iPM10 is also capable of inducing oxidative stress by probably inducing generation of H2 O2 and impairing enzymatic antioxidant defense, resulting in oxidative DNA damage and even apoptotic cell death through the iron-catalyzed Fenton reaction.

Air pollution impairs cognition, provokes depressive-like behaviors and alters hippocampal cytokine expression and morphology

Particulate matter air pollution is a pervasive global risk factor implicated in the genesis of pulmonary and cardiovascular disease. Although the effects of prolonged exposure to air pollution are well characterized with respect to pulmonary and cardiovascular function, comparatively little is known about the impact of particulate matter on affective and cognitive processes. The central nervous system may be adversely affected by activation of reactive oxygen species and pro-inflammatory pathways that accompany particulate matter pollution. Thus, we investigated whether long-term exposure to ambient fine airborne particulate matter (<2.5 μm (PM(2.5))) affects cognition, affective responses, hippocampal inflammatory cytokines and neuronal morphology. Male mice were exposed to either PM(2.5) or filtered air (FA) for 10 months. PM(2.5) mice displayed more depressive-like responses and impairments in spatial learning and memory as compared with mice exposed to FA. Hippocampal pro-inflammatory cytokine expression was elevated among PM(2.5) mice. Apical dendritic spine density and dendritic branching were decreased in the hippocampal CA1 and CA3 regions, respectively, of PM(2.5) mice. Taken together, these data suggest that long-term exposure to particulate air pollution levels typical of exposure in major cities around the globe can alter affective responses and impair cognition.

Preliminary study of the sources of ambient air pollution in Serpong, Indonesia

There has been increasing interest regarding air pollution in the Serpong area of Indonesia, a region near Jakarta, especially with respect to high concentrations of Pb detected in early 2001. Several studies have been conducted, but the appropriate action needed to control this pollution has not been identified because of inadequate identification of the particulate matter sources. In this study, source investigation was performed using the chemical characteristics of ambient airborne particulate matter samples from several locations in Serpong and its surroundings. Sampling of airborne particulate matter was conducted using a Gent stacked filter unit sampler in Serpong between August and November 2008. Black carbon (BC) was determined by reflectance and elemental analysis were performed using particle induced X–ray emission (PIXE). Al, As, Ba, Br, Ca, Cl, Co, Cr, Cu, Fe, Hg, I, K, Mg, Mn, Na, Ni, P, Pb, S, Sc, Si, Se, Sr, Ti, V and Zn were determined. The results showed that the higher Pb concentrations in both fine and coarse particulate matter were observed in the industrial area compared to adjacent residential areas. The Pb percentages in the collected PM2.5 mass in the residential and industrial areas were 3.1% and 4.3%, respectively, while for PM2.5–10, Pb represented 1.9% and 6.0% of the PM mass, respectively. The fine fraction data from two residential areas (Setu and EMC) were analyzed using EPA PMF (version 3) for source apportionment. The source apportionment identified 5 factors, i.e., lead industry and road dust (12%), diesel vehicles (30%), oil and power plant (26%), road dust (17%) and biomass burning mixed with road dust (15%).

Chronic Fine Particulate Matter Exposure Induces Systemic Vascular Dysfunction via NADPH Oxidase and TLR4 Pathways

Chronic exposure to ambient air-borne particulate matter of < 2.5 μm (PM₂.₅) increases cardiovascular risk. The mechanisms by which inhaled ambient particles are sensed and how these effects are systemically transduced remain elusive. To investigate the molecular mechanisms by which PM₂.₅ mediates inflammatory responses in a mouse model of chronic exposure. Here, we show that chronic exposure to ambient PM₂.₅ promotes Ly6C(high) inflammatory monocyte egress from bone-marrow and mediates their entry into tissue niches where they generate reactive oxygen species via NADPH oxidase. Toll-like receptor (TLR)4 and Nox2 (gp91(phox)) deficiency prevented monocyte NADPH oxidase activation in response to PM₂.₅ and was associated with restoration of systemic vascular dysfunction. TLR4 activation appeared to be a prerequisite for NAPDH oxidase activation as evidenced by reduced p47(phox) phosphorylation in TLR4 deficient animals. PM₂.₅ exposure markedly increased oxidized phospholipid derivatives of 1-palmitoyl-2-arachidonyl-sn-glycero-3-phosphorylcholine (oxPAPC) in bronchioalveolar lavage fluid. Correspondingly, exposure of bone marrow-derived macrophages to oxPAPC but not PAPC recapitulated effects of chronic PM₂.₅ exposure, whereas TLR4 deficiency attenuated this response. Taken together, our findings suggest that PM₂.₅ triggers an increase in oxidized phospholipids in lungs that then mediates a systemic cellular inflammatory response through TLR4/NADPH oxidase-dependent mechanisms.

Temperature-induced volatility of molecular markers in ambient airborne particulate matter

Abstract. Molecular markers are organic compounds used to represent known sources of particulate matter (PM) in statistical source apportionment studies. The utility of molecular markers depends on, among other things, their ability to represent PM volatility under realistic atmospheric conditions. We measured the particle-phase concentrations and temperature-induced volatility of commonly-used molecular markers in California's heavily polluted San Joaqin Valley. Concentrations of elemental carbon, organic carbon, levoglucosan, and polycyclic aromatic hydrocarbons were not reduced by mild (~10 K) heating. In contrast, both hopane/sterane and n-alkane concentrations were reduced, especially during the summer sampling events at the urban site. These results suggest that hopanes and steranes have effective saturation concentrations ~1 μg m−3, and therefore can be considered semi-volatile. The volatility of an individual compound depends both on its inherent properties (primarily vapour pressure) and the interactions between itself and any potential absorbing phase. The volatility behavior of n-alkanes during the urban summer is consistent with that predicted for absorption by suberic acid (a C8 diacid) using a group contribution modelling method. Observations can also be matched by an absorbent whose composition is based on recently-obtained high-resolution aerosol mass spectrometer factors (approximately 33% "hydrocarbon-like" and 67% oxygenated organic aerosol). The reduced evaporation of the n-alkanes, hopanes, and steranes with mild heating during rural and/or winter experiments could be explained by a more oxygenated absorbing phase along with a non-absorptive partitioning mechanism, such as adsorption to soot. This suggests that the temperature-induced volatility of large hydrocarbons in PM is most important if a relatively non-polar absorbing organic phase exists. While the activity coefficients of most organic aerosol compounds may be close to unity, the assumption of ideality for large hydrocarbons (e.g., hopanes) may result in large errors in partitioning calculations.

Current Trend and Regional Distribution of Lung Cancer Among Koreans: Findings From the Korea National Health Insurance Claims Data 2001–2008

PP-30-160 Background/Aims: Many studies have found a strong exposure-response relationship between particulate matter and cumulative health effects, such as lung cancer. Numerous epidemiologic studies highlighted the role of ambient airborne particulate matter as an important environmental pollutant for lung cancer. The incidence of lung cancer was 27.9 persons per 100,000, ranking the fourth place in 2007. In particular, the overall incidence was second place in men (48.1 persons per 100,000), and it was the highest among persons aged ≥65 years. Moreover the 5-year survival rate was 16.7% in 2003–2007, which was only amounting to 30% of average survival rate (57.1%). Therefore, this study aims to identify the magnitude, trend, and regional distribution of lung cancer for establishing the effective management in Korea. Methods: Study data were driven from Korean National Health Insurance that covers 96.3% of Korean people. Lung cancer was defined as an inpatient or outpatient for ICD code of C34 and so far more than 300,000 won have been spent on it. Results: The prevalence of lung cancer was 77 persons in 2006, 87 in 2007, and 85 in 2008 per 100,000 persons. Although the prevalence of men was about 2.5 times higher than that of women, the increasing rate was higher in women (14.3%) than that of men (10.1%) during 2006–2008. In addition, the prevalence of those aged 65 years and older was 16–17 times higher than that of below the age 65 years. The prevalence of Jeonnam was the highest, and Chungnam, Jeonbuk, and Gyongbuk were commonly top 4 areas. Despite the prevalence of Ulsan, Daejeon, and Seoul are relatively low, the increasing rate was fairly high during 2006–2008. Conclusion: Although Korean government has established the comprehensive planning for the control of air pollution, the higher prevalence together with the continuing increasing trend requires the more intensive management of lung cancer.
