Alpha-tocopherol concentrations and case life of lamb muscle as influenced by concentrate or pasture finishing

Two experiments were conducted to evaluate alpha-tocopherol accumulation in muscle of lambs finished on pasture or concentrates. The objective for Exp. 1 was to compare accumulation of alpha-tocopherol in the longissimus muscle of pasture-fed lambs to that of lambs fed three concentrations (15, 150, and 300 IU/kg of DM) of supplemental vitamin E (all rac alpha-tocopheryl acetate) in all-concentrate diets. The objective in Exp. 2 was to investigate the effect of duration of supplemental vitamin E feeding on alpha-tocopherol content and color change during display case storage of lamb muscle. Treatments evaluated in Exp. 2 were: 15 IU of supplemental vitamin E/kg DM fed to finish; 15 IU/kg followed by 300 IU/kg of DM during the last 21 d; and 15 IU/kg DM until 7 d prior to finish, then 300 IU/kg DM. In Exp. 1, alpha-tocopherol concentration of rotational grazed alfalfa and perennial ryegrass averaged 137 and 169 mg/kg of DM. Vitamin E treatments for lambs fed concentrate diets did not affect ADG (P > 0.15), but ADG was greater (P < 0.01) for concentrate-fed lambs than for grazing lambs. For the concentrate-fed lambs, alpha-tocopherol in longissimus muscle increased quadratically (P < 0.05) as dietary concentrations of vitamin E increased. Predicted maximum alpha-tocopherol concentration in muscle occurred at about 400 IU/kg of diet DM. Longissimus muscle from lambs grazing alfalfa or ryegrass had similar (P > 0.50) alpha-tocopherol concentrations, and those concentrations were similar to values obtained when the concentrate diet supplemented with 150 IU of vitamin E/kg was fed. In Exp. 2, no differences (P > 0.10) in ADG were observed. Concentrations of longissimus alpha-tocopherol were highest when 300 IU supplemental vitamin E was fed for 21 d prior to slaughter. During a 6-d display period, semimembranosus steaks from lambs fed 300 IU of supplemental vitamin E/kg for either 7 or 21 d had higher a* and b* color readings than steaks from lambs fed 15 IU/kg of supplemental vitamin E. Increased consumption of vitamin E either via pasture or supplementation results in higher alpha-tocopherol concentrations in meat.

Effect of dietary oils and alpha-tocopheryl acetate supplementation on lipid (TBARS) and cholesterol oxidation in cooked pork.

The effect of n-3 fatty acid-enriched diets (in the form of 0.5% linseed oil with either 1.5% sunflower oil or 1.5% olive oil) and alpha-tocopheryl acetate supplementation (200 mg/kg feed) on lipid oxidation (thiobarbituric acid-reactive substances, TBARS) and cholesterol oxide products (COPS) in cooked pork was investigated. Longissimus muscle was studied. Meat from pigs fed 0.5% linseed oil-enriched diets had a higher proportion of n-3 fatty acid than meat from pigs in other dietary groups in neutral (P < 0.0001) and polar lipids (P < 0.0001), and a 20% reduction in the n-6:n-3 ratio was observed. Alpha-tocopheryl acetate supplementation increased (P < 0.05) monounsaturated fatty acids in polar lipids and increased (P = 0.0001) alpha-tocopherol levels in muscle. Alpha-tocopherol concentration in muscle was affected by dietary fat (P < 0.05). Groups receiving diets enriched with sunflower oil had significantly higher alpha-tocopherol levels (P < 0.05) in muscle than those groups receiving olive oil-enriched diets. Numbers of TBARS were significantly (P < 0.05) lower in the group fed supplemental olive oil than in those fed sunflower oil. Dietary linseed oil increased (P < 0.05) lipid oxidation principally at the initial period of storage in cooked pork. Overall, dietary alpha-tocopheryl acetate supplementation significantly increased (P < 0.001) lipid stability and decreased (P < 0.05) total COP production across the dietary groups. Alpha-tocopherol was a more effective antioxidant for decreasing TBARS values in cooked meat when adding sunflower oil to the diets instead of olive oil.

Antioxidative and oxidative status in muscles of pigs fed rapeseed oil, vitamin E, and copper.

The susceptibility of a given muscle tissue to lipid oxidation may not only depend on the presence of unsaturated fatty acids and the balance between antioxidants and prooxidants, but also on the composition of the skeletal muscle. In the present study, the effects of dietary supplementation of vitamin E (dl-alpha-tocopheryl acetate) and copper in combination with a high level of monounsaturated fatty acids were examined with regard to the antioxidant concentration and the susceptibility to lipid oxidation of two muscles, longissimus (LD) and psoas major (PM), representing different oxidative capacity. In addition, fatty acid profiles of the backfat and the intramuscular lipids, as well as fresh meat quality traits, were studied. Pigs were allotted to a 3x3 factorial experiment with three levels of dl-alpha-tocopheryl acetate (0, 100, and 200 mg/kg of feed) and three levels of copper (0, 35, and 175 mg/kg of feed) added to a diet containing 6% rapeseed oil. A basal diet (without rapeseed oil) was added to the experimental design, giving a total of 10 dietary treatments. Muscle alpha-tocopherol concentrations increased (P<.001) with increasing dl-alpha-tocopheryl acetate in the feed. The antioxidative status was higher in PM than in LD, when considering the concentration of alpha-tocopherol (P<.001) and the activity of antioxidant enzymes (superoxide dismutase, P<.001; glutathione peroxidase, P = .06). Supplemental copper did not give rise to any deposition of copper in muscle tissue or backfat, but the antioxidant status of PM increased. The susceptibility to lipid oxidation was reduced in LD with increasing dietary dl-alpha-tocopheryl acetate and in PM with increasing dietary copper. Supplemental dl-alpha-tocopherol acetate improved the water-holding capacity of LD (P = .005) and PM (P = .003). The fatty acid composition of the backfat and the triglyceride fraction of the intramuscular fat became more unsaturated with the addition of rapeseed oil to the feed. Higher intakes of monounsaturated fatty acids due to the rapeseed oil were also reflected in the phospholipid fraction of the intramuscular fat, but no influence on the proportion of saturated fatty acids was seen. The susceptibility to lipid oxidation of PM was lower for pigs on the rapeseed oil-based diet than for those on the basal diet. The energy metabolic status of the muscles and the accumulation of calcium by the sarcoplasmic reticulum were not influenced by the dietary treatments, but there were differences between muscle types. The addition of rapeseed oil to the diet reduced the muscular content of glycogen (LD, P = .02; PM, P = .06) and elevated the plasma concentration of free fatty acids (P = .05). Overall, dietary fat, dl-alpha-tocopherol acetate, and copper affected the oxidative status of pig muscles, and the results differed depending on muscle type.

Case life of seven retail products from beef cattle supplemented with alpha-tocopheryl acetate.

Packages (n = 660) of clod, strip loin, tenderloin, bottom round, eye-of-round, and top round steaks, as well as ground beef, from beef cattle fed diets supplemented with vitamin E (500 IU/d for 100 d preharvest) were compared with packages of products derived from carcasses of beef cattle fed control diets to determine the duration of acceptable muscle color during simulated retail display. In addition, the effect of storage before steak fabrication, simulating distribution time, on the acceptability of each treatment and each cut during retail display was investigated. Dietary supplementation of animals with vitamin E increased muscle alpha-tocopherol concentrations (P < .05) in all cuts. The acceptable retail display time of ground beef from vitamin E-supplemented cattle was longer (P < .05) compared to ground beef from unsupplemented cattle by 10.2, 15.6, and 17.6 h following 7, 14, and 21 d of storage in chub packages prior to final grinding and retail display, respectively. However, length of storage in a vacuum package before retail display did not have a consistent effect on retail case life of steaks. Product drip loss during retail display was not affected by storage time or vitamin E treatment. The trained panel color score during retail display of high vitamin E strip loin, bottom round, inside round, and eye-of-round steaks remained acceptable longer (P < .05) by 11.5, 25.2, 8.4, and 29.4 h, respectively, than that of control steaks. The results demonstrated that vitamin E supplementation of cattle feed can be used as a management tool to reduce the economic losses associated with beef muscle color deterioration during retail display of products.

A review of dietary vitamin E supplementation for improvement of beef quality.

Color is a primary factor used by consumers to judge beef quality, especially freshness. Recent studies indicate that dietary supplementation of vitamin E to beef cattle increases the alpha-tocopherol concentration in muscle and its membranous subcellular fractions. The increased tissue alpha-tocopherol concentration protects not only membranal lipids but also myoglobin from oxidation. This results in delayed onset of discoloration in fresh, ground, and frozen beef and in suppression of lipid rancidity, especially in fresh, ground, and frozen beef and less so in cooked beef. Extension of beef color display life depends on dose level and duration of dietary vitamin E, muscle, and aging period. Cumulative results of experiments conducted to date indicate that beef from animals that receive 500 IU/steer daily of vitamin E for 126 d could assuredly benefit the domestic retail market by extending color display life. Implementation of this technology by the beef industry requires development of a method for rapid determination of alpha-tocopherol concentration in muscle samples collected on the day of harvest.

Assessment of vitamin E nutritional status in sheep.

The effects of supplemental dietary vitamin E (as DL-alpha-tocopheryl acetate; 0, 15, 30, and 60 IU/d) on serum, platelet, and muscle tocopherol and lipid (cholesterol and triglycerides) concentrations in 32 sheep were investigated in a 60-d trial. Serum, platelet, and muscle alpha-tocopherol concentrations increased linearly (P < .05) with treatment. Platelet tocopherol concentrations were more sensitive to vitamin E intake than either serum or muscle tocopherol. There were no effects on serum lipid concentrations. There were low correlations (P > .05) between serum or platelet tocopherol and either cholesterol or triglycerides or the sum of the two lipid fractions. Correlations between serum or platelet tocopherol and muscle tocopherol were also low (P > .05). Although platelet tocopherol was more sensitive to vitamin E intake than serum tocopherol, serum tocopherol concentrations can be reliably used to estimate vitamin E status. Expressing serum tocopherol relative to blood lipids did not improve the relationship between serum tocopherol and vitamin E intake.

Endogenous skeletal muscle antioxidants.

Skeletal muscle is susceptible to oxidative deterioration due to a combination of lipid oxidation catalysts and membrane lipid systems that are high in unsaturated fatty acids. To prevent or delay oxidation reactions, several endogenous antioxidant systems are found in muscle tissue. These include alpha-tocopherol, histidine-containing dipeptides, and antioxidant enzymes such as glutathione peroxidase, superoxide dismutase, and catalase. The contribution of alpha-tocopherol to the oxidative stability of skeletal muscle is largely influenced by diet. Dietary supplementation of tocopherol has been shown to increase muscle alpha-tocopherol concentrations and inhibit both lipid oxidation and color deterioration. Dietary selenium supplementation has also been shown to increase the oxidative stability of muscle presumably by increasing the activity of glutathione peroxidase. The oxidative stability of skeletal muscle is also influenced by the histidine-containing dipeptides, carnosine and anserine. Whereas carnosine and anserine are affected by diet less than alpha-tocopherol and glutathione peroxidase, their concentrations vary widely with species and muscle type. In pigs, beef, and turkey muscle, carnosine concentrations are greater than anserine, while the opposite is true in rabbit, salmon, and chicken muscle. Anserine and carnosine are found in greater concentrations in muscle high in white fibers, with chicken white muscle containing over fivefold more anserine and carnosine than red muscle. Anserine and carnosine are thought to inhibit lipid oxidation by a combination of free radical scavenging and metal chelation.

Serum and tissue tocopherol in sheep after intramuscular injection and(or) dietary vitamin E supplementation

In a 28-d trial, 35 wethers weighing 35 to 60 kg were used to evaluate serum, liver, pancreas, kidney, diaphragm, heart, and neck muscle alpha-tocopherol concentrations after vitamin E administration as i.m. injections and(or) oral supplementation. Treatments (T) were as follows: 1) i.m. DL-alpha-tocopherol(DL-alpha-ol) d 1, 2) i.m. DL-alpha-tocopheryl acetate(DL-alpha-ac) d 1, 3) i.m. DL-alpha-ol d 1, 7, 14, and 21, 4) T 1 and oral supplementation, 5) T 3 and oral supplementation, 6) oral supplementation, and 7) control, no E supplementation. Supplemented wethers received 500 IU via injection and(or) 30 IU/kg of diet (acetate form). In a second 10-d trial, blood was periodically collected from 20 wethers weighing 32 to 47 kg that had been assigned to T as follows: 1) control, 2) i.m. DL-alpha-ol d 1, 3) i.m. DL-alpha-ac d 1, 4) T 2 d 1 and 7, and 5) T 3 d 1 and 7. Results from repeated measures ANOVA in both trials indicated that there were day x T effects (P < .0001) on serum alpha-tocopherol. Pretreatment serum tocopherol concentrations were .58 and .66 microgram/mL for Trials 1 and 2, respectively. In Trial 1, wethers receiving the four alcohol vitamin E forms showed a rapid increase in serum tocopherol between d 1 and 3, followed by a rapid decline to pretreatment concentrations for T 1, whereas T 3, 4, and 5 maintained higher than pretreatment concentrations after the decline.(ABSTRACT TRUNCATED AT 250 WORDS)
